In Vitro Biocompatibility of MC3T3-E1 Osteoblast-like Cells on Arg-Gly-Asp Acid Peptides Immobilized Graphite-like Carbon Coating on Carbon/Carbon Composites

Cao Sheng,Li He-Jun,Li Ke-Zhi,Lu Jin-Hua,Zhang Lei-Lei

Carbon/carbon (C/C) composites were deposited with graphite-like carbon (GLC) coating, and then, Arg-Gly-Asp acid (RGD) peptides were successfully immobilized onto the functionalized GLC coating. GLC coating was utilized to prevent carbon particles releasing and create a uniform surface condition for C/C composites. RGD peptides were utilized to improve biocompatibility of GLC coating. Surface chemical characterizations of functionalized GLC coating were detected by contact angle measurement, X-ray photoelectron spectroscopy and Raman spectra. Optical morphology of GLC coatings was observed by confocal laser scanning microscopy. In vitro biological performance was determined using samples seeded with MC3T3-E1 osteoblast-like cells and cultured for 1 week. Surface characterizations and morphological analysis indicated that C/C composites were covered by a dense and uniform GLC coating. Contact angle of GLC coating was reduced to 27.2° when it was functionalized by H 2 O 2 oxidation at 40 °C for 1 h. In vitro cytological test showed that the RGD peptides immobilized GLC coating had a significant improvement in biocompatibility. It was suggested that RGD peptides provided GLC coating with a bioactive surface to improve cell adhesion and proliferation on C/C composites.

Osteogenic Effects of Limosilactobacillus fermentum GBE18 Cell-Free Supernatant (CFS) in MC3T3-E1 Cells via the Wnt/β-Catenin and PI3K/Akt Signaling Pathways

Xingyuan Peng,Xuan Zheng,Xiyu Li,Xiaona Pang,Junhua Jin,Hui Liu,Hongxing Zhang,Yuanhong Xie

Osteoporosis is a major global health challenge, particularly among aging populations, underscoring the need for safe and effective nutritional interventions. Probiotics and their metabolites have emerged as promising candidates for modulating bone health via the gut-bone axis. In this study, we investigated the effects of a cell-free culture supernatant (CFS) from the food-grade bacteriumLimosilactobacillus fermentumGBE18 on the proliferation, differentiation, and mineralization of MC3T3-E1 pre-osteoblasts. GBE18 CFS exhibited no cytotoxicity at concentrations ranging from 1% to 4% (v/v). Notably, 2% (v/v) CFS significantly enhanced alkaline phosphatase (ALP) activity and extracellular matrix mineralization (p< 0.05). Transcriptomic profiling revealed that differentially expressed genes were enriched in osteoblast-related processes and two key signaling pathways: Wnt/β-catenin and PI3K/Akt. Subsequent qRT-PCR and Western blot analyses confirmed the upregulation of critical regulators (Rspo2,Pdpk1,Malat1) and demonstrated coordinated activation of Akt phosphorylation, β-catenin stabilization, and Runx2 protein expression. Our findings indicate that GBE18 CFS promotes osteogenic differentiation through coordinated modulation of the PI3K/Akt and Wnt/β-catenin pathways. Consequently, this study provides mechanistic evidence supporting the potential application ofL. fermentumGBE18-derived metabolites as functional food ingredients or dietary interventions for bone health and osteoporosis management.

Regulation of T Cell Responses by Nano-Hydroxyapatite to Mediate the Osteogenesis

Fangze Guo,Changqing Yuan,Hailin Huang,Xuyang Deng,Zirui Bian,Danyang Wang,Keke Dou,Li Mei,Qihui Zhou

Nano-hydroxyapatite (nHA) has been widely applied as a tissue-engineering biomaterial and interacted with osteoblasts/stem cells to repair bone defects. In addition, T cells that coexist with osteoblasts/stem cells in the bone modulate the regulation of osteoimmunology by cytokine formation. However, the effects of nHA on T cells and the following regulatory interplay on osteogenic differentiation have been rarely examined. In this work, the physicochemical properties of needle-like nHA are characterized by field emission scanning electron microscopy, zeta potential, Fourier transform-infrared and X-ray diffraction. It is found that as the concentration of nHA increases, the proliferation of T cells gradually increases, and the proportion of apoptotic T cells decreases. The percentage of CD4 + T cells is higher than that of CD8 + T cells under the regulation of needle-like nHA. Furthermore, the supernatant of T cells co-cultured with nHA significantly inhibits the osteogenic differentiation of MC3T3-E1 by downregulating the formation of alkaline phosphatase and calcium nodule compared with the supernatant of nHA. Thus, our findings provide new insight into the nHA-mediated T cell and osteoblast interactions.

Large artificial bone from 3D printed polycaprolactone/β-tricalcium phosphate (3D PCL/β-TCP) effectively promoting MC3T3-E1 cell adhesion, proliferation, and new bone formation

Yuxin Niu,Fuxiang Song,Rong Shan,Qianjuan Zhao,Zhuang Chen,Na Wang,Weibo Xie

The use of 3D printing technology has advanced the bone tissue engineering, and constructing large artificial bones for repairing large-scale bone defects is highly significant. This study aimed to construct large artificial bones in a precise and controllable manner, focusing on repairing critical-sized bone defects. The researchers used 3D printing technology to synthesize 3D PCL/β-TCP, and then evaluated its ability to promote MC3T3-E1 cell adhesion, proliferation, and new bone formation through a series of characterizations. The results confirmed that 3D PCL/β-TCP, presented as a lattice structure similar to natural bone, could be used to prepare personalized artificial bone blocks based on the actual range of bone defects. The researchers also experimented with dipping 3D PCL/β-TCP bone blocks in different proportions of β-TCP and found that PT2 showed better mechanical and hydrophilic properties. Further experiments showed that PT2 presented excellent biocompatibility and outstanding capacity to promote MC3T3-E1 cell proliferation and adhesion, and that 3D PCL/β-TCP bone blocks possessed good osteogenic activity. Finally, it was suggested that PT2 exhibited the property of promoting new bone formation. These findings provide experimental data support for the repair of critical-sized bone defects.

Effects of a Coating of Nano Silicon Nitride on Porous Polyetheretherketone on Behaviors of MC3T3-E1 Cells in Vitro and Vascularization and Osteogenesis in Vivo

Yong Dai,Linyang Chu,Zhengliang Luo,Tingting Tang,Han Wu,Fan Wang,Shiqi Mei,Jie Wei,Xuehong Wang,Xifu Shang

To improve the bioperformances of porous polyetheretherketone (PPK) for bone repair, silicon nitride-coated PPK (CSNPPK) was prepared by a method of suspension coating and melt binding. The results revealed that, as compared with PPK, the surface roughness, compressive strength, and water absorption of CSNPPK increased, while the pore size and porosity of CSNPPK exhibited no obvious changes. In addition, the cellular responses (including attachment, proliferation, and differentiation as well as osteogenically related gene expressions) of the MC3T3-E1 cells to CSNPPK were remarkably promoted compared with PPK and dense polyetheretherketone in vitro. Moreover, in the model of rabbit femoral condyle defects, the results of micro computed tomography and histological and mechanical evaluation revealed that the ingrowth of new vessels and bone tissues into CSNPPK was significantly greater than that into PPK in vivo. Furthermore, the load–displacement and push-out loads for CSNPPK with bone tissues were higher than for PPK, indicating good osseointegration. In short, CSNPPK not only promoted vascularization but also enhanced osteogenesis as well as osseointegration in vivo. Therefore, it can be suggested that CSNPPK with good biocompatibility, osteogenic activity, and vascularization might be a promising candidate as an implant for bone substitute and repair.

Cyclin A1 (CCNA1) inhibits osteoporosis by suppressing transforming growth factor-beta (TGF-beta) pathway in osteoblasts

Du Xiao,Zang Chuanyi,Wang Qinglei

Background Osteoporosis is a genetic disease caused by the imbalance between osteoblast-led bone formation and osteoclast-induced bone resorption. However, further gene-related pathogenesis remains to be elucidated. Methods The aberrant expressed genes in osteoporosis was identified by analyzing the microarray profile GSE100609. Serum samples of patients with osteoporosis and normal group were collected, and the mRNA expression of candidate genes was detected by quantitative real-time polymerase chain reaction (qRT-PCR). The mouse cranial osteoblast MC3T3-E1 cells were treated with dexamethasone (DEX) to mimic osteoporosis in vitro. Alizarin Red staining and alkaline phosphatase (ALP) staining methods were combined to measure matrix mineralization deposition of MC3T3-E1 cells. Meanwhile, the expression of osteogenesis related genes including alkaline phosphatase (ALP), osteocalcin (OCN), osteopontin (OPN), Osterix, and bone morphogenetic protein 2 (BMP2) were evaluated by qRT-PCR and western blotting methods. Then the effects of candidate genes on regulating impede bone loss caused by ovariectomy (OVX) in mice were studied. Results Cyclin A1 (CCNA1) was found to be significantly upregulated in serum of osteoporosis patients and the osteoporosis model cells, which was in line with the bioinformatic analysis. The osteogenic differentiation ability of MC3T3-E1 cells was inhibited by DEX treatment, which was manifested by decreased Alizarin Red staining intensity, ALP staining intensity, and expression levels of ALP, OCN, OPN, Osterix, and BMP2. The effects of CCNA1 inhibition on regulating osteogenesis were opposite to that of DEX. Then, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis demonstrated that genes negatively associated with CCNA1 were enriched in the TGF-beta signaling pathway. Inhibitor of TGF-beta signaling pathway partly reversed osteogenesis induced by suppressed CCNA1. Furthermore, suppressed CCNA1 relieved bone mass of OVX mice in vivo. Conclusion Downregulation of CCNA1 could activate TGF-beta signaling pathway and promote bone formation, thus playing a role in treatment of osteoporosis.

Dimethyloxalylglycine improves angiogenesis of ZIF-8-coated implant

Xin Zhang,Jian Wang,Jiaxin Wu,Xiaoge Jiang,Xibo Pei,Junyu Chen,Qianbing Wan,Chao Huang

Nanoscale zeolitic imidazolate framework-8 coating with certain density can significantly improve the osteogenic activity of titanium implants both in vitro and in vivo. In this study, we loaded dimethyloxalylglycine into zeolitic imidazolate framework-8-modified implants (Z-AHT) to enhance the angiogenic activity, and the obtained products were denoted as D-AHT. Firstly, we characterized the specimens via scanning electron microscopy, Fourier transform infrared spectroscopy, powder X-ray diffraction, and surface contact angle. Then, the release kinetics of both zinc ions and dimethyloxalylglycine were investigated. To illustrate the combined effects of surface topography and chemistry (zeolitic imidazolate framework-8 and dimethyloxalylglycine) on osteogenic and angiogenic activity, MC3T3-E1 preosteoblast cells and human umbilical vein endothelial cells were respectively cultured on D-AHT. Pure Ti and Z-AHT were designated as control groups. As evidenced by the expression of osteogenic genes (Alp, Col1, Opg, and Runx2) and the secretion of ALP protein, D-AHT showed comparable osteogenic activity to Z-AHT which was better than that of Ti. Further angiogenesis assay revealed a better human umbilical vein endothelial cell migration on D-AHT, suggesting that D-AHT can be an attractive candidate for application in implant surface modification.

Porous cellulose microspheres coated in one step with a polydopamine suspension of hydroxyapatite for bone tissue engineering

Gao Fei,Zeng Dong,Liu Hong,Qin Rui,Zhang Jie,Chen Yingming,Wang Weiming,Peng Chuang,Li Ming,Li Qing,Wang Hong,Peng Shuai

Abstract Deacetylated porous cellulose acetate microspheres (DPCA) modified with hydroxyapatite (HA) penetrated polydopamine (PDA) coating was prepared by a one-step in-situ method and investigated as the scaffold for bone tissue regeneration. The physical and chemical properties of different scaffolds were analyzed by Scanning electron microscopy, Transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction and X-ray photoelectron spectrum. Test results proved the unique three-dimensional porous structure, homogeneous PDA coating, dispersive HA crystals of the DPCA-PDA-HA microspheres (DPPH). The excellent attachment, adhesion and proliferation of MC3T3-E1 preosteoblast on DPPH were manifested by cell adhesion and biocompatibility assessment. In vitro mineralization also shown MC3T3-E1 was capable of differentiating osteogenically. Such satisfactory biomedical performance of DPPH was ascribed to the one-step introducing process contributing to a uniform and enrich growth of HA on the microspheres surface and interior. This study provides a promising cellulose-based scaffold applied in bone regeneration field. Graphical abstract

Nano-hydroxyapatite mineralized silk fibroin porous scaffold for tooth extraction site preservation

Ling Nie,He Zhang,Aishu Ren,Yuzhou Li,Gang Fu,Richard David Cannon,Ping Ji,Xiaohong Wu,Sheng Yang

Objective To fabricate a novel nano-hydroxyapatite mineralized silk fibroin (MSF) scaffold in order to diminish the resorption of alveolar ridge and accelerate new bone formation within tooth sockets. Also, to investigate the biocompatibility and osteogenic ability of the MSF in vitro , and the effect of site preservation of the MSF graft in post-extractive sockets in vivo . Methods SEM, EDX, FTIR and XRD were used to analyze the mineral crystals deposited on the silk fibroin (SF) surface. Pre-osteoblasts (MC3T3-E1) were seeded on SF and MSF scaffolds. Cell viability, distribution and differentiation were examined using a live-dead assay, histological analysis and Alizarin Red S staining. Furthermore, prepared grafts (SF or MSF scaffold) were implanted into the maxillary right first molar sockets of Sprague Dawley rats for 6 weeks and newly formed bone tissue was analyzed by micro-CT and histological examination. Results The SEM, EDX, FTIR and XRD analysis demonstrated that granulate nano-hydroxyapatite (nHA) crystals were uniformly distributed on the SF scaffold. In addition, the MSF hydrophilicity measured by water contact angle and swelling ratio was superior to plain SF scaffold. The effect of nHA inorganic crystals on osteogenic differentiation of MC3T3-E1 cells indicated the MSF scaffolds improved osteogenesis. Furthermore, MSF grafts induced more bone formation and reduced the height of alveolar bone resorption after tooth extraction. Significance The MSF scaffold partially simulated the structure and composition of natural bone matrix. It induced osteogenic differentiation of MC3T3-E1 cells in vitro , and also promoted new bone regeneration in tooth extraction sockets in vivo , indicating it is a biomaterial with great potential for tooth extraction site preservation.

Nanoporous tantalum coated zirconia implant improves osseointegration

Lianjun Wu,Yiwen Dong,Litao Yao,Chuantong Liu,Abdullrahman M Al-Bishari,Kendrick Hii Ru Yie,Hualin Zhang,Jinsong Liu,Gang Wu

Zirconia has become a new dental implant material because of its excellent esthetics, biocompatibility, and minimal ion release. However, biological inertness of zirconia surface results in weak integration with bone tissue in the early stage of osseointegration. To enhance the early osseointegration of zirconia implant, a homogeneous nanoporous tantalum (TaNS) coating (~30 nm in diameter and 1 μm in length) with good adhesion strength was prepared on zirconia surface via magnetron sputtering combined with anodization. The roughness and hydrophilicity in ZrO 2 /TaNS groups were significantly improved as compared to other groups. The protein adsorption, initial adhesion, spreading and proliferation of MC3T3-E1 cells were significantly enhanced in ZrO 2 /TaNS group as compared to ZrO 2 and ZrO 2 /Ta group. Moreover, the osteogenic differentiation of MC3T3-E1 cells was significantly improved in ZrO 2 /TaNS groups than other groups by up-regulating the expression of the osteogenic-related genes (RunX2, ALP , COL-1, OSX, OCN and OPG). Histological analysis displayed that more newly regenerated bone was deposited around ZrO 2 /TaNS group than other groups, and the bone-implant contact in ZrO 2 /TaNS group was enhanced correspondingly. In conclusion, the TaNS coating with good adhesion strength can be successfully prepared on zirconia surface by magnetron sputtering combined with anodization. This novel coating will be a promising clinical application in improving osseointegration of the zirconia implant.

Preparation and characterization of porous hydroxyapatite/β-cyclodextrin-based polyurethane composite scaffolds for bone tissue engineering

Jingjing Du,Shuchun Gan,Qihao Bian,Duhan Fu,Yan Wei,Kaiqun Wang,Qiaoxia Lin,Weiyi Chen,Di Huang

In this study, novel porous scaffolds containing hydroxyapatite and β-cyclodextrin-based polyurethane were first successfully fabricated by polymerizing β-cyclodextrin with hexamethylene diisocyanate and hydroxyapatite in situ for bone tissue engineering. The physicochemical and mechanical properties as well as cytocompatibility of porous scaffolds were investigated. The results showed that polyurethane reinforced with hydroxyapatite composites had cancellous bone-like porous structure. The mechanical strength of the scaffolds increased with increasing the hydroxyapatite content in scaffolds. Synthesized scaffolds (PU1, PUHA1, PU2, and PUHA2) presented compressive strength values of 0.87 ± 0.24 MPa, 1.81 ± 0.10 MPa, 6.16 ± 0.89 MPa, and 12.95 ± 2.05 MPa, respectively. The pore size and porosity of these scaffolds were suitable for bone regeneration. Cytocompatibility of composite scaffolds was proven via favorable interactions with MC3T3-E1 cells. The addition of hydroxyapatite into CD-based polyurethane scaffolds improved cell attachment, well-spread morphology, and higher proliferation. The hydroxyapatite-polyurethane scaffolds have the potential to be applied in bone repair and regeneration.

Biomimetic Domain-Active Electrospun Scaffolds Facilitating Bone Regeneration Synergistically with Antibacterial Efficacy for Bone Defects

Yunzhu Qian,Xuefeng Zhou,Hong Sun,Jianxin Yang,Yi Chen,Chao Li,Hongjin Wang,Tong Xing,Feimin Zhang,Ning Gu

To improve bone regeneration in oral microenvironment, we generated a novel biodegradable, antibacterial, and osteoconductive electrospun PLGA/PCL membrane as an ideal osteogenic scaffold. The novel three-layer membranes were structured with serial layers of electrospun chlorhexidine-doped-PLGA/PCL (PPC), PLGA/PCL (PP), and β-tricalcium phosphate-doped-PLGA/PCL (PPβ). To characterize osteoconductive properties of these membranes, MC3T3-E1 (MC) cultures were seeded onto the membranes for 14 days for evaluation of cell proliferation, morphology and gene/protein expression. In addition, MC cells were cultured onto different surfaces of the three-layer membranes, PPC layer facing MC cells (PPβ-PP-PPC) and PPβ layer facing MC cells (PPC-PP-PPβ) to evaluate surface–material effects. Membrane properties and structures were evaluated. Antibacterial properties against Streptococcus mutans and Staphylococcus aureus were determined. Scanning electron microscope demonstrated smaller interfiber spaces of PPC and PPβ-PP-PPC compared to PPβ, PPC-PP-PPβ, and PP. PPC and PPβ-PP-PPC exhibited hydrophilic property. The three-layer membranes (PPC-PP-PPβ and PPβ-PP-PPC) demonstrated significantly higher Young’s modulus (94.99 ± 4.03 MPa and 92.88 ± 4.03 MPa) compared to PP (48.76 ± 18.15 MPa) or PPC (7.92 ± 3.97 MPa) (p < 0.05). No significant difference of cell proliferation was found among any groups at any time point (p > 0.05). Higher expression of integrins were detected at 12 h of cultures on PPC-PP-PPβ compared to the controls. Promoted osteoconductive effects of PPC-PP-PPβ were revealed by alkaline phosphatase assays and Western blot compared with the controls at 7 and 14 days. PPC, PPC-PP-PPβ and PPβ-PP-PPC exhibited a significantly wider antibacterial zone against the tested bacteria compared to PP and PPβ (p < 0.05). These results suggested that the three-layer electrospun membranes demonstrated superior properties: higher strength, better cell adhesion, and promoted osteoconductive properties compared to single-layer membrane: however, antibacterial properties were exhibited in three-layer electrospun membranes and chlorhexidine-doped single-layer membrane. We concluded that the novel three-layer membranes could be used as a biocompatible scaffold for intraoral bone regeneration due to its enhanced osteoconductive activity and antibacterial effect.
