Naringenin loaded fucoidan/polyvinylpyrrolidone nanoparticles protect against folic acid induced acute kidney injury in vitro and in vivo

Tao Jiang,Feikai Zhu,Xintao Gao,Xiaochen Wu,Wenyong Zhu,Chuanlong Guo

Acute kidney injury (AKI) is a common clinical problem with no effective treatment. Excessive folic acid (FA) induced kidney tubular injury is characterized by oxidative stress and inflammation, and is a common model of AKI. The excellent pharmacological activity of naringenin (NAR) makes it a potential agent for treating AKI, but its poor solubility limits its application. This study prepared NAR loaded nanoparticles (FU/PVP-NAR) using fucoidan (FU) and polyvinylpyrrolidone (PVP) as carriers, with a particle size of 23.96 ± 2.77 nm. In vitro studies showed that FU/PVP-NAR inhibited excessive FA induced proliferation inhibition, accumulation of reactive oxygen species (ROS), and disruption of mitochondrial membrane potential (MMP) of HK-2 cells. Further confirmed that FU/PVP-NAR inhibited FA induced DNA damage and Cyclic GMP–AMP synthase (cGAS)-stimulator of interferon genes (STING) activation. In vivo studies showed that excessive FA induced AKI features in mice, such as elevated serum creatinine (SCr) and blood urea nitrogen (BUN) levels, accompanied by pathological damage to kidney tissues. The above AKI characteristics induced by FA were alleviated by FU/PVP-NAR. FU/PVP-NAR also inhibited the decrease in antioxidant enzyme levels in kidney tissues induced by FA. Furthermore, in vivo mechanism studies indicated that FU/PVP-NAR inhibited the release of inflammatory factors by inhibiting DNA damage-cGAS-STING pathway. In summary, this study provided the possibility for FU/PVP-NAR as a potential candidate drug for treating FA induced AKI.

Sodium alginate/carboxymethyl chitosan/gelatin-naringenin pH-responsive hydrogel beads for oral delivery of traditional chinese herbal medicines

Zixing Yan,Qingling Nie,Jiajia Liu,Jingxin Chen,Youmei Liu,Yudong Lu,Mengjun Xu,Zhenwen Lin

Stomach disorders are a common category of digestive system diseases. With the increasing emphasis on traditional medicine, the use of herbal medicines in treating gastric diseases has attracted much attention. However, direct administration of herbal medicines with short half-life leads to low drug utilization. To address this problem, pH-responsive composite hydrogel beads consisting of sodium alginate/carboxymethyl chitosan/gelatin-naringenin (SA/CMCh/Gel-Nar) were designed in this study to encapsulate decocted herbal saddle decoctions (AZD) for use as sustained-release medications for gastrointestinal disorders. The SA/CMCh/Gel-Nar hydrogel beads have high drug loading capacity, strong adhesion (98.33 % retention), and excellent pH-responsive sustained-release properties. In SGF, 39 % of the drug was released within 2 h, ensuring rapid therapeutic efficacy, while in SIF, the remaining 61 % was gradually released within 23 h. With excellent antimicrobial efficacy (>93.4 %) and high biosafety (cell viability >97.21 %), these hydrogel beads provide a promising and safe platform for sustained drug delivery in the gastrointestinal tract.

Comparative study of glycyrrhizin interaction with plant-derived proteins for encapsulation and delivery of naringenin: Role of prolamins

Ruoyang Wang,Dongmei Jia,Zhanyu Zhang,Meixing Yan,Xianggen Wu

This study evaluated dipotassium glycyrrhizinate (DG) modification on four distinct plant-based alternative proteins: legume-derived faba bean protein (mainly globulins), oilseed pumpkin protein (mainly globulins), pseudocereal buckwheat protein (mainly globulins and few prolamins), and cereal zein (exclusive prolamins), to enhance delivery efficiency of naringenin. Through pH-shifting preparation, DG significantly enhanced protein solubility and naringenin encapsulation efficiency (~78.0–98.1 %), particularly in prolamin-rich zein showing 117.6 % solubility increase and 13.0-fold efficiency improvement (from 5.6 % to 78.0 %). Molecular docking revealed that hydrophobic and electrostatic interactions were the key drivers of DG's effects, with prolamins exhibiting greater structural changes under DG modification. Additionally, all DG-modified proteins enhanced naringenin bioaccessibility. These findings highlight DG's potential as a versatile modifier for plant-derived alternative proteins, particularly those rich in prolamins, with promising applications in functional foods.

Naringenin-Loaded Core–Shell Mg-MOF Nanocomposite Integrated Injectable Hydrogel for Bone Regeneration: Anti-inflammatory, Osteogenic, and Angiogenic

Chaonan He,Peipei Feng,Dingli Xu,Leidong Lian,Jin Li,Kaifeng Gan,Yi Gu,Tingting Pan,Yun Tang,Xiang Wu,Zhaoxiang Peng,Chunhai Ke,Jingyun Ma

Composite biomaterial systems facilitate regenerative medicine through component synergy. Herein, novel core–shell nanocomposites (N-HMMs) are prepared, with hollow mesoporous silica (HMSNs) as the core and magnesium-gallic acid metal–organic frameworks (Mg-MOFs) as the shell. N-HMMs are loaded with the small-molecule osteoinductive drug naringin (Nar) to form N-HMMs@Nar, which is integrated into methacrylate gelatin/polyethylene glycol diacrylate hydrogels (GelMA/PEGDA, GP) to construct N-HMMs@Nar@GP. N-HMMs@Nar@GP enables sustained release of Nar, bioactive Mg2+, and antioxidant gallic acid for synergistic anti-inflammation, osteogenesis, and angiogenesis of bone regeneration. GP adapts N-HMMs@Nar to the traumatic characteristics of bone defects and facilitates in situ treatments. In vitro studies have confirmed the composite materials’ biocompatibility, osteoinductive differentiation, and angiogenesis capabilities. The ability of N-HMMs@Nar@GP to enhance osteogenesis and angiogenesis is demonstrated in an in vivo tibial defect model using micro-CT and histopathological analysis. Therefore, N-HMMs@Nar@GP holds significant potential for application in the repair of bone defects.

Naringenin Microsphere as a Novel Adjuvant Reverses Colistin Resistance via Various Strategies against Multidrug-Resistant Klebsiella pneumoniae Infection

Qiushuang Sheng,Xiaoning Hou,Yang Wang,Nan Wang,Xuming Deng,Zhongmei Wen,Dan Li,Li Li,Yonglin Zhou,Jianfeng Wang

The efficacy of colistin, the last option against multidrug-resistant (MDR) Gram-negative bacteria, is severely threatened by the prevalence of plasmid- or chromosome-mediated colistin resistance genes. Herein, naringenin has dramatically restored colistin sensitivity against colistin-resistant Klebsiella pneumoniae infection without affecting bacterial viability, inducing resistance and causing obvious cell toxicity. Mechanism analysis reveals that naringenin potentiates colistin activity by multiple strategies including inhibition of mobilized colistin resistance gene activity, repression of two-component system regulation, and acceleration of reactive oxygen species-mediated oxidative damage. A lung-targeted delivery system of naringenin microspheres has been designed to facilitate naringenin bioavailability, accompanied by an effective potentiation of colistin for Klebsiella pneumoniae infection. Consequently, a new recognition of naringenin microspheres has been elucidated to restore colistin efficacy against colistin-resistant Gram-negative pathogens, which may be an effective strategy of developing potential candidates for MDR Gram-negative bacteria infection.

Naringenin alleviates heat stress-induced thymic injury through a triple mechanism of immune modulation, antioxidant defense, and inflammation suppression

Ke Liao,Luqi Wang,Jingyan Luo,Xi Jiang,Siyan Zhou,Feiyang Li,Ping Liu,Gaofeng Cai,Zhanhong Zheng,Xiaoquan Guo,Xiaona Gao

The thymus is an important immune organ, and heat stress can significantly impact the immune function of chickens. Naringenin, as a novel immunomodulator, can restore the imbalanced immune state (pathological state) to a near-normal immune equilibrium (physiological state). This research aims to investigate the effects of naringin on heat stress-induced damage in the thymus of Ningdu Yellow chickens. Two hundred eighty-eight 28-day-old Ningdu yellow chickens were randomly assigned to four groups: control (Con), naringenin-supplemented (Con+Nar), heat stress (HS), and heat stress + naringenin (HS+Nar). HS and HS+Nar groups were subjected to cyclic temperatures daily (37±2°C, 07:00-19:00; 24±2°C, 19:00-07:00). Con+Nar and HS+Nar groups received diets supplemented with 200 mg/kg naringenin for 35 consecutive days. Results showed that naringenin significantly ameliorated heat stress-induced thymic pathological damage in Ningdu yellow chickens and reversed heat stress-reduced immune indices (IgA, IgG, IgM). Molecularly, heat stress markedly suppressed the Nrf2/Keap1 signaling axis, downregulated Nrf2, HO-1, and NQO1 expression, while activating NF-κB pathway key molecules (TLR4, MYD88), thereby upregulating pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-17) and downregulating anti-inflammatory cytokine IL-10. In contrast, naringenin enhanced antioxidant capacity via modulating the Nrf2/Keap1 pathway and inhibited excessive pro-inflammatory factor elevation, consequently alleviating thymic injury. Overall, naringenin exerts multi-targeted regulation of oxidative stress-inflammation crosstalk, maintaining thymic immune microenvironment homeostasis under heat stress. These findings lay a theoretical basis for developing naringenin-based feed additives to prevent and control poultry heat stress syndrome.

Naringenin boosts Parkin-mediated mitophagy via estrogen receptor alpha to maintain mitochondrial quality control and heal diabetic foot ulcer

Xin-Meng Zhou,Ying Yang,Dao-Jiang Yu,Teng Xie,Xi-Lu Sun,Ying-Xuan Han,Hai-Ying Tian,Qing-Qing Liao,Yu-Jie Zhao,Yih-Cherng Liou,Wei Huang,Yong Xu,Xi Kuang,Xiao-Dong Sun,Yuan-Yuan Zhang

Diabetic foot ulcer (DFU) is an increasing global burden due to the rising prevalence of diabetes, and no specific pharmacological targets or satisfactory drugs are currently available for this devastating ailment. In this study, naringenin (NAR) was found to accelerate diabetic wound healing in diabetic C57BL/6J wild-type mice by reducing oxidative stress, as assessed through histological assay. NAR also alleviated the inhibition of proliferation, inflammation, cell senescence, and apoptosis in HaCaT cells induced by high glucose (HG). Mechanistically, the beneficial effects of NAR on wound healing are dependent on the E3 ubiquitin-protein ligase parkin. NAR upregulated the expression level of Parkin and promoted its mitochondrial translocation, thereby activating Parkin-mediated mitophagy and maintaining mitochondrial quality control (MQC). Moreover, the wound healing-promoting effects of NAR were significantly diminished in Parkin knockdown HaCaT cells and Prkn knockout DFU mice. Inhibition of NAR binding to estrogen receptors (ERs) using tamoxifen abolished the protective effects of NAR in HG-induced HaCaT cells. The luciferase reporter assay confirmed that NAR enhanced ERs binding to the estrogen response element (ERE), thereby upregulating Parkin transcription. Additionally, the cellular thermal shift assay revealed that NAR specifically bound to estrogen receptor alpha (ERα). In conclusion, NAR promoted DFU wound healing by enhancing Parkin-mediated mitophagy via binding to ERα, highlighting its potential as a promising therapeutic candidate.

Construction of high internal phase Pickering emulsion using dual protein hydrolysates-naringenin supramolecules and their contribution on curcumin bioaccessibility

Ziyin Yang,Shengbin Zeng,Jiayao Tang,Linghuan Yang,Bei Jin

The potential of dual protein–polyphenol supramolecular complexes for constructing delivery systems based on high internal phase Pickering emulsions (HIPPEs) is significant, yet the underlying mechanism has not been fully revealed. This study systematically investigated hybrid supramolecular complexes fabricated through physical processing techniques such as ultrasound (US) and high pressure (HP), integrating sunflower-walnut protein hydrolysates (SW) with naringenin (Nar) as novel stabilizers for oil-in-water HIPPEs. While physical treatments effectively enhanced SW emulsification capacity through particle size reduction, interfacial tension decrease, and wettability increase, particularly under US treatment, the physical field (US and HP) has limited effectiveness in improving the emulsion’s cream stability. The synergistic application of physical processing and naringenin complexation induced SW structural unfolding, facilitating the adsorption and anchoring of SW at the oil–water interface. Compared to the SW-Nar (US), SW-Nar (HP) generated emulsions with smaller droplets, reinforced viscoelastic networks, and improved curcumin bioaccessibility. In vitro digestion induced SW-Nar complexes interfacial structural unfolding, hydrophobic exposure, and α-helix-to-β-sheet transition, enhancing emulsification and micelle formation via strengthened hydrogen bonds and hydrophobic interactions, particularly in SW-Nar (HP). These findings collectively establish SW-Nar-stabilized HIPPEs as advanced delivery platforms for lipophilic bioactives, with system performance critically dependent on supramolecular interfacial microstructure modulated by physical processing methods.

Encapsulation of Hydrophobic and Low-Soluble Polyphenols into Nanoliposomes by pH-Driven Method: Naringenin and Naringin as Model Compounds

Mianhong Chen,Ruyi Li,Yuanyuan Gao,Yeyu Zheng,Liangkun Liao,Yupo Cao,Jihua Li,Wei Zhou

Naringenin and naringin are a class of hydrophobic polyphenol compounds and both have several biological activities containing antioxidant, anti-inflammatory and anti-tumor properties. Nevertheless, they have low water solubility and bioavailability, which limits their biological activity. In this study, an easy pH-driven method was applied to load naringenin or naringin into nanoliposomes based on the gradual reduction in their water solubility after the pH changed to acidity. Thus, the naringenin or naringin can be embedded into the hydrophobic region within nanoliposomes from the aqueous phase. A series of naringenin/naringin-loaded nanoliposomes with different pH values, lecithin contents and feeding naringenin/naringin concentrations were prepared by microfluidization and a pH-driven method. The naringin-loaded nanoliposome contained some free naringin due to its higher water solubility at lower pH values and had a relatively low encapsulation efficiency. However, the naringenin-loaded nanoliposomes were predominantly nanometric (44.95–104.4 nm), negatively charged (−14.1 to −19.3 mV) and exhibited relatively high encapsulation efficiency (EE = 95.34% for 0.75 mg/mL naringenin within 1%w/vlecithin). Additionally, the naringenin-loaded nanoliposomes still maintained good stability during 31 days of storage at 4 °C. This study may help to develop novel food-grade colloidal delivery systems and apply them to introducing naringenin or other lipophilic polyphenols into foods, supplements or drugs.Keywords:naringenin;naringin;nanoliposomes;pH-driven;stability

Formulation and intestinal absorption of naringenin loaded nanostructured lipid carrier and its inhibitory effects on nonalcoholic fatty liver disease

Rui Hu,Shu Liu,Gulinigaer Anwaier,Qinyu Wang,Wanli Shen,Qiang Shen,Rong Qi

In this study, we prepared naringenin (NGN) loaded nanostructured lipid carrier (NGN-NLC) and investigated its characterizations, transepithelial transport, intestinal absorption and inhibitory effects on nonalcoholic fatty liver disease (NAFLD) induced by a methionine choline deficient (MCD) diet in mice. The NGN-NLC, prepared by a method of emulsion-evaporation plus low temperature-solidification, displayed high drug loading capacity of 22.5 ± 1.7%. Compared to the NGN crude drug , the NGN-NLC, at an equal NGN dose, improved NGN release rate by 3.5-fold and elevated NGN transepithelial transport and intestinal absorption through enhancing intracellular transport of clathrin pathway and escaping p-gp efflux; at an 8-fold lower NGN dose, showed comparable pharmacokinetic parameters, but elevated liver NGN distribution by 1.5-fold, reduced MCD diet-induced hepatic lipid deposition by 3-fold. These results suggest that the NLC formulation significantly increased the inhibitory effects of NGN on NAFLD because of the improved drug release rate, transepithelial transport and intestinal absorption, and the elevated oral bioavailability and liver NGN distribution.

Polyphenol oxidase inhibited by 4-hydroxycinnamic acid and naringenin: Multi-spectroscopic analyses and molecular docking simulation at different pH

Hongwei Jiang,Lei Zhou,Yuefang Sun,Kaibo Yu,Wenzhi Yu,Yuqing Tian,Junping Liu,Liqiang Zou,Wei Liu

It is still unclear how pH affects the inhibitory effects of phenolic acids and flavonoids on polyphenol oxidase (PPO). In this study, 4-hydroxycinnamic acid and naringenin were selected to investigate their interactions with PPO from pH 6.8 to 5.0. Results showed that acidic pH could enhance the inhibitory effect of inhibitors and a greater enhancement effect was observed in 4-hydroxycinnamic acid. Fluorescence emission spectra indicated that 4-hydroxycinnamic acid and naringenin interacted with PPO and quenched its intrinsic fluorescence, which was also enhanced by acidic pH. Circular dichroism suggested that 4-hydroxycinnamic acid and naringenin could reversibly bind to PPO molecules and transform α-helix into β-sheet. Molecular docking results revealed that 4-hydroxycinnamic acid and naringenin interacted with PPO through hydrogen bond and hydrophobic interaction, and more interactions were observed near the carboxyl group. These results indicated that acidic pH could significantly enhance the inhibitory effect of phenolic acid on PPO.

Naringenin improves mitochondrial function and reduces cardiac damage following ischemia-reperfusion injury: the role of the AMPK-SIRT3 signaling pathway

Li-Ming Yu,Xue Dong,Xiao-Dong Xue,Jian Zhang,Zhi Li,Hong-Jiang Wu,Zhong-Lu Yang,Yang Yang,Hui-Shan Wang

Mitochondrial dysfunction contributed greatly to myocardial ischemia-reperfusion (MI/R)-induced cardiomyocyte apoptosis. Naringenin is a flavonoid exhibiting potential protective effects on myocardial mitochondria under stress conditions. However, the detailed down-stream signaling pathway involved remains uncovered. This study was designed to elucidate naringenin's mitochondrial protective actions during MI/R with a focus on AMPK-SIRT3 signaling. Sprague-Dawley rats were administered with naringenin (50 mg kg−1 d−1) and subjected to MI/R surgery in the presence or absence of compound C (0.25 mg kg−1, Com.C, an AMPK inhibitor) co-treatment. An in vitro study was performed on H9c2 cardiomyoblasts subjected to simulated ischemia-reperfusion treatment. Before the treatment, the cells were administered with naringenin (80 μmol L−1) with or without SIRT3 siRNA/AMPK1α siRNA transfection. Naringenin improved post-reperfusion left ventricular systolic pressure and the instantaneous first derivative of left ventricular pressure, and reduced the infarction size and myocardial apoptosis index by suppressing mitochondrial oxidative stress damage (as evidenced by decreased mitochondrial cytochrome c release and oxidative markers) and enhancing mitochondrial biogenesis [as evidenced by increased NRF1, TFAM and oxidative phosphorylation subunit complexes (II, III and IV)]. These protective actions were abolished by Com.C (in vivo) or SIRT3 siRNA (in vitro) administration. Further investigation revealed that Com.C (in vivo) or AMPK1α siRNA (in vitro) markedly suppressed PGC-1α and SIRT3 levels while SIRT3 siRNA (in vitro) inhibited SIRT3 expression without significantly changing AMPK phosphorylation and PGC-1α levels. Taken together, we found that naringenin directly inhibits mitochondrial oxidative stress damage and preserves mitochondrial biogenesis, thus attenuating MI/R injury. Importantly, AMPK-SIRT3 signaling played a key role in this process.
