Non-enzymatic electrochemiluminescence biosensor for ultrasensitive detection of ochratoxin A based on efficient DNA walker

Xiaoyu Shi,Xiaochun Zhu,Yaqin Chai,Ying Zhou,Ruo Yuan

Ochratoxin A (OTA) with high toxicity represents a serious threat to the agriculture and food chain, consequently to human health. Herein, a simple electrochemiluminescence (ECL) biosensor was constructed for ultrasensitive detection of OTA based on mercaptopropionic acid templated Au nanoclusters (Au NCs) as intensive signal probe and a non-enzymatic 2D DNA walking machine as the effective amplification strategy. Specifically, the target related bipedal DNA walker efficiently moved along 2D DNA tracks through toehold-mediated DNA strand displacement, which triggered abundant signal probes for combining to the DNA tracks. Moreover, the Au NCs could exhibit strong ECL emission due to fast electron transfer from massive Au-S electronic pathways under the electrochemical excitation. Thus, the biosensor possessed significant ECL response for achieving ultrasensitive detection toward OTA with low detection limit of 3.19 fg/mL. Impressively, the sensing platform was also applied to detect OTA from edible oils, exhibiting great application potential in food analysis.

A self-assembly aptasensor based on thick-shell quantum dots for sensing of ochratoxin A

Xianfeng Chu,Xiaowen Dou,Ruizheng Liang,Menghua Li,Weijun Kong,Xihui Yang,Jiaoyang Luo,Meihua Yang,Ming Zhao

A novel self-assembling aptasensor was fabricated by precisely attaching three phosphorothioate-modified capture aptamers onto a single thick-shell quantum dot in a controllable manner for monitoring of ochratoxin A (OTA), a poisonous contaminant widespread in foodstuffs. Herein, CdSe/CdS QDs coated in ten layer CdS shells were synthesized using a continual precursor injection method. Analysis of the prepared CdSe/CdS QDs showed a zinc-blende structure, high photoluminescence quantum yields (>80%), and a photoemission peak with a narrow full-width at half-maximum (about 29 nm), all qualities that render them as a superior choice for optical applications. By adjusting the number of phosphorothioate bases in the anchor domain, the tunable-valency aptasensor was able to self-assemble. In the sensing strategy, the thick-shell quantum dot was provided as an acceptor while OTA itself was used as a donor. In the presence of OTA, the capture aptamers drive the aptasensor function into a measurable signal through a fluorescence resonance energy transfer (FRET) system. The newly developed aptasensor had a detection limit as low as 0.5 ng mL−1, with a linear concentration in the range of 1 to 30 ng mL−1, and therefore meets the requirements for rapid, effective, and anti-interference sensors for real-world applications. Moreover, the high quality thick-shell QDs provide an ideal alternative for highly sensitive imaging and intensive illumination in the fields of biotechnology and bioengineering.

Determination, content analysis and removal efficiency of fining agents on ochratoxin A in Chinese wines

Xiangyu Sun,Yuxue Niu,Tingting Ma,Pingkang Xu,Weidong Huang,Jicheng Zhan

A simple and convenient HPLC-FD detection method for ochratoxin A (OTA) with a high detection limit and a short run time has been developed. OTA has been found in most samples of Chinese market wine, including domestic and imported wines, but the content was not very high. Only a few wines showed an OTA content that exceeded the EC and OIV limits, indicating that most Chinese market wines were safe. The OTA intake for Chinese from wine was 0.017 ng/kg (bw) per day, which was lower than the SCF and JECFA limits and also lower than in many other countries. This was mainly due to the low per capita wine consumption in China, but it is still necessary for the Chinese government and wine makers to monitor OTA levels in wine and to establish relevant regulations. An egg white treatment (0.20 mg/mL, 48 h) was the best removal method for OTA.

Affinity maturation of anti-ochratoxin a nanobody via synthetic library strategy for enhanced small molecule detection

Yulin Mao,Tieqiang Sun,Tingting Gong,Kaiyuan Chen,Cong Liu,Hao Zheng,Wenjia Niu,Jian Hou,Zhilong Lu,Chen Luo,Liangqin Li,Jiannan Feng,Jialei Bai,Longlong Luo,Ran Zhang

Single-domain properties and ease of gene manipulation of the nanobody (Nb) make it suitable for affinity maturation in vitro. In this study, the anti-ochratoxin A (OTA) Nb3G was selected as a model antibody to explore feasible approach of affinity enhancement. Artificial intelligence (AI)-augmented molecular modeling tools were used to analyze the interaction between OTA and Nb3G, and nine key residues (Thr28, Tyr32, Asp54, Thr57, Trp99, Phe103, Glu106, Trp110, Tyr112) were selected to construct a site-directed mutagenesis library. After biological screening and identification, the affinity-enhanced mutants 1H and 11B were obtained, exhibiting half maximal inhibitory concentration (IC₅₀) values of 0.31 ng/mL and 0.25 ng/mL, corresponding to 1.26-fold and 1.56-fold sensitivity improvements compared with the parent Nb (IC₅₀ 0.39 ng/mL). Enzyme-linked immunosorbent assay (ELISA) and surface plasmon resonance (SPR) further confirmed 2-fold and 4-fold increases in titer, and 6.27-fold and 11.49-fold improvements in affinity, respectively. The mutants also demonstrated superior stability under diverse conditions and improved competitive inhibition performance with lower variability. Structural analysis indicated that deeper antigen-binding cavities may facilitate more efficient OTA encapsulation, enhancing detection sensitivity. Recoveries in spiked oat and coffee samples ranged from 94.20 % to 113.85 %. Overall, the synthetic nanobody library strategy developed in this work provides an effective method for the preparation of high-affinity nanobodies against small molecule antigens.

A colorimetric immunoassay based on cobalt hydroxide nanocages as oxidase mimics for detection of ochratoxin A

Hongshuai Zhu,Zhu Quan,Haoyuan Hou,Yue Cai,Weipeng Liu,Yingju Liu

Oxidase nanozymes have attracted particular attention in colorimetric immunoassay due to their high activity, stability, and biocompatibility. In this work, the well-dispersed Co(OH) 2 nanocages were synthesized by a template-assisted strategy. The prepared Co(OH) 2 nanocages exhibited an intrinsic oxidase-like activity, which can oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) without the addition of hydrogen peroxide (H 2 O 2 ). Hence, a simple and sensitive colorimetric immunoassay was established to detect ochratoxin A (OTA). In this immunosensor, the microplates were modified with dopamine as the substrate to capture antigens. Then, the uniform Co(OH) 2 nanocages were used as the vehicle to carry the secondary antibody for the recognition of immunoreaction. The sensing system can generate color responses with the OTA concentration in the range from 0.5 ng/L to 5 μg/L, and the detection limit was as low as 0.26 ng/L. Therefore, as a nano-artificial enzyme, Co(OH) 2 nanocages may show promising potential in visual detection of the real samples.

Dual-Fluorescence Ratiometric Aptasensor Using Structure-Switching Aptamer Modified With 2-Aminopurine for Detecting Ochratoxin A

Huang Weihui,Tang Jidong,Ma Wenjin,Tao Xiaoqi

Structure-switching aptamers (SSAs) generate measurable signals through target-induced conformational changes, serving as sensitive molecular recognition elements. We developed a dual-fluorescence SSA for ochratoxin A (OTA) detection by inserting 2-aminopurine (2AP) into the OTA aptamer and hybridizing a complementary DNA (cDNA) strand at the 5’ end. OTA binding induced the aptamer-cDNA duplex to form a G4, enhancing 2AP fluorescence at 370 nm. Together with the intrinsic fluorescence of OTA at 450 nm, this enabled ratiometric dual-fluorescence measurements without external quenchers or fluorophores. To elucidate the molecular mechanism, all-atom molecular dynamics (MD) simulations and spectroscopic experiments assessed the effects of cDNA hybridization sites and strand lengths on sensing performance. Simulations further examined OTA binding at various 2AP insertion sites. Apt@2AP-cDNA1, with cDNA hybridized to bases 1–12, exhibited a binding pattern closely resembling the original aptamer. Umbrella sampling simulations revealed similar binding affinities: 46.3 kJ/mol for Apt@2AP-cDNA1 and 43.2 kJ/mol for the original aptamer. Experimentally, Apt@2AP-cDNA1 converted to G-quadruplexes more efficiently than other designs. A ratiometric dual-fluorescence aptasensor using Apt@2AP-cDNA1 was developed, with a linear detection range of 10–800 nM, a detection limit of 5.9 nM. The dual-fluorescence ratiometric aptasensor has good selectivity and was proven effective for detecting OTA in beer samples.

Mitochondrial dynamics imbalance and energy dyshomeostasis mediated developmental toxicity of ochratoxin a in zebrafish (Danio rerio)

Meng Li,Mengai He,Zhenfeng Liao,Chenxi Li,Yingqiu Mi,Ningning Pan,Xinyi Xie,Zhiyu Liu,Jiayin Hou,Xueli Yu,Liezhong Chen

Ochratoxin A (OTA) has been increasingly detected in foodstuffs and human samples. However, the molecular mechanisms of developmental toxicity induced by OTA remain poorly explored. Our results demonstrated that oxidative stress induced by OTA caused mitochondrial dysfunction in larvae, characterized by malformed mitochondria, and decreased mitochondrial membrane potential and complex II, IV and V activities. Central carbon metabolism analysis indicated that exposure to OTA induced energy metabolism disturbance by decreasing ATP contents and altering energy-relevant metabolite contents. Energy metabolism disturbance was further confirmed by transcriptomics analysis, with differentially expressed genes significantly enriching in glycolysis and TCA cycle. Additionally, molecular docking simulations and surface plasmon resonance analysis demonstrated that OTA interacted with mfn1 and mfn2, verifying its interference effects on mitochondrial dynamics. The insufficient energy supply induced by OTA activated AMP-activated protein kinase signal pathways, which accelerated catabolic processes, accompanied by decreases in yolk sac size and yolk lipid. Finally, insufficient energy supply caused developmental retardation of zebrafish larvae. Collectively, this study systematically elucidated the roles of mitochondrial dynamics imbalance and energy dyshomeostasis in the developmental toxicity induced by OTA, providing critical insights for refining food safety standards and therapeutic interventions.

Effective multicolor visual biosensor for ochratoxin A detection enabled by DNAzyme catalysis and gold nanorod etching

Guo Zhiqiang,Zhang Mingshuo,Zhang Haiping,Ren Xinru,Xiao Yijing,Sun Weiqing,Wang Yu,Liu Su,Huang Jiadong

A novel detection technique is introduced that offers sensitive and reliable ochratoxin A (OTA) detection. The method leverages the etching of gold nanorods (AuNRs) stabilized by hexadecyl trimethyl ammonium bromide (CTAB) using the oxidized form of 3,3′,5,5′-tetramethyl benzidine sulfate (TMB 2+ ), creating a susceptible multicolor visual detection system for OTA. The visual detection is enabled by Mg 2+ -assisted DNAzyme catalysis combined with the catalytic hairpin assembly (CHA) signal amplification strategy. The presence of OTA triggers CHA and signaling responses along with the formation of G-quadruplex-hemin DNAzyme, which promotes the oxidation of TMB with H 2 O 2 , leading to the etching of AuNRs and a reduction in their aspect ratio. AuNRs experienced a blue shift in the longitudinal localized surface plasmon resonance peak, resulting in a color change. The technique has been shown to detect OTA with a low detection limit of 0.309 pg/mL, demonstrating high sensitivity and specificity. The detection technique offers versatility by enabling the detection of other pollutants through a simple replacement of the aptamer, expanding the range of detection platforms available for pollutant determinations. An ultrasensitive homogeneous electrochemical strategy for ochratoxin a sensing based on nanoscale PCN-224@MB@Apt

Xi Wu,Chenggang Geng,Weiwei Cheng,Zhenjiong Wang,Yan Zhang,Di Wu,Xiaozhi Tang

In this reasearch, a homogeneous electrochemical sensor based on PCN-224@MB@Apt was fabricated for the ultrasensitive determination of ochratoxin A (OTA). Firstly, nanoscale PCN-224 were synthesized as the nanocarrier to embed the signal probe of methylene blue (MB). Then, the OTA aptamer (Apt) was added and connected to PCN-224@MB via the Zr– O –P bond between Zr metal sites of PCN-224 and phosphate group of Apt as the biogate. When OTA exists, the Apt would preferentially bind with OTA and fall off from PCN-224@MB, leading to the release of MB and generation of differential pulse voltammetry (DPV) response. The DPV response of MB was linearly correlated with the amount of OTA. The optimized PCN-224@MB@Apt sensor showed outstanding detection performance towards OTA with a low detection limit of 2.6 × 10 −5  ng/mL (S/N = 3) and wide linear range (10 −4 -10 ng/mL). Meanwhile, the fabricated homogeneous electrochemical sensor exhibited splendid stability, reproducibility, and specificity. To assess the practical applicability, the PCN-224@MB@Apt sensor was applied to detect OTA in real corn samples and desirable recovery rates varying from 83.2 % to 109.6 % were obtained.

A two-color fluorescence sensing strategy based on functionalized tetrahedral DNAzyme nanotweezers for ochratoxin A detection

Yingao Yang,Ruipeng Chen,Yifen Guo,Jiaxin Zhang,Shuyue Ren,Huanying Zhou,Zhixian Gao

A two-color fluorescent sensing strategy based on a functionalized tetrahedral DNAzyme nanotweezer (FTDN) was developed to detect ochratoxin A (OTA) utilizing the multifunctional properties of DNA nanotechnology. The FTDN enables rapid OTA detection directly through a Cy5 fluorescent group, modified to respond to the target signal. Additionally, FTDN exhibits DNAzyme cutting activity in the presence of Mg 2 ⁺ ions, enabling it to traverse DNA nanoflower-functionalized magnetic beads. This process results in the continuous cleavage of DNA nanoflowers labeled with numerous FAM fluorescent groups, thereby amplifying the detection signal and enhancing OTA sensitivity. The linear ranges for the Cy5 and FAM signals in response to OTA were 5–1000 ng/mL and 0.05–100 ng/mL, respectively, with corresponding limits of detection (LOD) of 1.59 ng/mL and 0.03 ng/mL. This study demonstrates that dual-color fluorescence via Cy5 and FAM can effectively verify OTA detection in food, significantly reducing false-positive and false-negative rates. The proposed platform offers sensitive and accurate detection of mycotoxins in food and can be adapted for monitoring other trace contaminants by simply altering the aptamer.

Plasmonic ELISA for naked-eye detection of ochratoxin A based on the tyramine-H2O2 amplification system

Yi Liang,Xiaolin Huang,Xirui Chen,Wenjing Zhang,Guo Ping,Yonghua Xiong

A novel direct competitive plasmonic enzyme-linked immunosorbent assay (dc-pELISA) was applied to detect ochratoxin A (OTA) with naked eyes. In this assay, horseradish peroxidase (HRP) + hydrogen peroxide (H 2 O 2 ) + tyramine (TYR)-induced gold nanoparticle (AuNP) aggregation was considered as a signal output; AuNP aggregation could be triggered through the phenol polymerization of TYR , which was induced by hydroxyl radicals from HRP-catalyzed H 2 O 2 ; OTA-labeled catalase (CAT) was used as a competing antigen to consume H 2 O 2 . Owing to the combined advantages of ultrahigh CAT catalytic activity for H 2 O 2 and dual-color responses (red and blue) generated through AuNP aggregation, the proposed method was highly sensitive and thus could be employed with naked eyes to detect OTA qualitatively with a cut-off limit of 150 pg/mL. Our method also demonstrated a good dynamic linear range (12.5–150 pg/mL) for quantitative OTA determination with a reliable correlation coefficient of R 2  = 0.992, a half-maximal inhibitory concentration of 84.75 pg/mL, and a detection limit of 17.8 pg/mL. In brief, this newly-designed technique is considerably suitable for high-throughput screening detection or point-of-care diagnostics in resource-constrained regions because of the easy readout of results by naked eyes without the use of advanced detection equipment.

An orientation-controlled molecularly imprinted paper-based fluorescent sensor for turn-on detection of ochratoxin A

Donghui Wang,Jiawei Feng,Ningning Li,Yuanyuan Liu,Qingqing Lu,Yang Li,Yiming Zhang

Detection technologies based on molecularly imprinted polymers (MIPs) have been developed for analyzing ochratoxin A (OTA), a highly toxic food contaminant. However, conventional MIPs suffer from disordered imprinting cavities, which compromise their analytical performance for OTA. Herein, we first proposed a luminescent metal organic framework (LMOF)-based paper-imprinted sensor (FP@Eu-DCBA@MIP) via a dummy template-derivatization-based oriented surface imprinting strategy for efficient OTA analysis. By incorporating a cis-dihydroxy-derived dummy template (OTA-CD) during imprinting, FP@Eu-DCBA@MIP achieved ordered recognition cavities. Compared to the non-imprinted material (FP@Eu-DCBA@NIP), FP@Eu-DCBA@MIP exhibited better adsorption capacity (58.82 mg g -1 ) and selectivity (imprinting factor: 3.6) for OTA. The LMOF (Eu-DCBA) served as the detection unit, endowing FP@Eu-DCBA@MIP with good sensitivity for OTA (limit of detection: 0.00352 mg L -1 ) through the antenna effect between Eu-DCBA and OTA. The sensor was also successfully applied to OTA analysis in food samples, demonstrating the potential of FP@Eu-DCBA@MIP for OTA detection in complex matrices.
