In tissue engineering research, there has recently been considerable interest in using electrospun biomimetic nanofibers of hybrids, in particular, from natural and synthetic polymers for engineering different tissues. However, phase separation between a pair of much dissimilar polymers might give rise to detrimental influences on both the electrospinning process and the resultant fiber performance. A representative natural-synthetic hybrid of gelatin (GT) and polycaprolactone (PCL) (50:50) was employed to study the phase separation behavior in electrospinning of the GT/PCL composite fibers. Using trifluoroethanol (TFE) as the cosolvent of the two polymers, observation of visible sedimentation and flocculation from dynamic light scattering analysis of the GT/PCL/TFE mixture both showed that phase separation does occur in just a few hours. This consequently led to gradually deteriorated fiber morphologies (e.g., splash, fiber bonding, and varied fiber size) over time during electrospinning GT/PCL. Quantitative analysis also indicated that the ratio of GT to PCL in the resultant GT/PCL fibers was altered over time. To address the phase separation related issues, a tiny amount (<0.3%) of acetic acid was introduced to improve the miscibility, which enabled the originally turbid solution to become clear immediately and to be single-phase stable for more than 1 week. Nanofibers thus obtained also appeared to be thinner, smooth, and homogeneous with enhanced performance in wettability and mechanical properties. Given the versatility and widely uses of the electrospun GT/PCL and other similar natural-synthetic hybrid systems in constructing tissue-engineered scaffolds, this work may offer a facile and effective approach to achieve finer and compositionally homogeneous hybrid nanofibers for effective applications.

Fiber-reinforced hydrogels are widely utilized in biomedical field; however, their mechanical strength and biological functionality are constrained by the poor integration between the fibrous components and the hydrogel matrix. Herein, we developed a biomimetic composite hydrogel (H-P-MA) with effective molecular integration through covalently photopolymerization between a GelMA/HAMA/DopaMA hydrogel network and reactive surface-functionalized PCLMA short fibers created by plasma technology. The H-P-MA composite hydrogel presented a micro-nano topological structure similar to that of the extracellular matrix (ECM) and exhibited superior mechanical properties as well as adhesion capabilities through strong covalent bonding between fibers and matrix, thereby facilitating better cell proliferation. This strategy provides a reliable pathway for developing composite hydrogel materials with ECM biomimicry.

Chitosan (CS), with its non-toxic and antibacterial properties, and Poly(ε-caprolactone) (PCL), known for its biodegradability and biocompatibility, are crucial materials in medical applications. This study proposed a solution utilizing electrospinning to manufacture fibers with nanometer-sized core-shell structures from these materials, with CS serving as the fiber core and PCL forming the shell. The influence of manufacturing technology parameters on fiber size and morphology was thoroughly researched and investigated. The solvent system used, Chloroform/Dimethylformamide (DMF), ensured complete solubility, viscosity control, conductivity, and proper solvent evaporation. Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM) results clearly demonstrated the morphology and internal structure of the nanofibers. The water contact angle (WCA) measurements of the fiber membrane showed almost no significant change over time, indicating strong hydrophobicity of the nanofibers. Additionally, the FTIR spectrum revealed distinct core-shell layers without any blending between them, while DSC analysis showed the thermal properties of fiber membranes. In summary, the electrospinning of nanofibers proved to be stable, producing thin fibers with an average diameter of approximately 400 nm. These findings are expected to significantly enhance the applicability of CS/PCL nanofibers across various fields, including healthcare and smart agriculture.

Dural closure after the neurosurgery can prevent postoperative complications. Although many types of dural substitute have been developed, most of them lack functional and structural characteristics compared with the natural dura mater. In this study, we used electrospinning method to fabricate a multilayer scaffold to promote dural repair. The inner layer of the scaffold that faces the brain tissue is composed of poly-lactic acid (PLA) to reduce tissue adhesion. The middle layer of the scaffold is composed of poly-ɛ-caprolactone and PLA, which provides a watertight seal. The outer layer of the scaffold contains a large amount of collagen to promote cell attachment and proliferation. The results from in vitro study and an animal model have shown that this multilayer fibrous scaffold has sufficient mechanic strength and biochemical properties to enhance dural repair. Therefore, fabrication of scaffold with multiple functional and structural layers may provide a novel approach for tissue engineering. © 2013 Wiley Periodicals, Inc. Bone defects resulting from trauma, malignant tumors, or infections are common clinical conditions. Current clinical treatments for bone defects, however, are associated with secondary injury, poor morphological matching, and immune rejection, falling short of clinical needs. Multifunctional bioscaffolds with osteogenic induction capability have emerged as a highly promising therapeutic strategy. In this study, 3D printing technology was utilized to fabricate scaffolds integrating ROS scavenging and osteogenic differentiation dual functions, and their physicochemical properties and biocompatibility were systematically investigated. Polycaprolactone (PCL) and strontium-magnesium-doped calcium silicate (SMCS) were selected to prepare PCL/SMCS scaffolds with varying SMCS ratios. SMCS incorporation effectively enhanced scaffold hydrophilicity and accelerated degradation. Cell culture experiments confirmed good biocompatibility of the PCL/SMCS scaffolds. Subsequently, cerium-doped SMCS bioceramics were prepared via a post-impregnation process. Notably, the PCL/0.1M Ce-SMCS scaffold exhibited optimal compressive performance, achieving a strength of 22.78 MPa—a 26.42% increase over the PCL/4SMCS scaffold. Compared with PCL/4SMCS, Ce doping promoted cell proliferation and adhesion, conferred ROS scavenging ability, and in vitro osteogenic assays indicated that low-content Ce-SMCS enhanced osteogenic differentiation.

Blood-contacting materials with good mechanical property, excellent anticoagulant function and promoting effect on endothelialization are in great demand for clinical application such as vascular grafts in treating cardiovascular diseases. In this study, electrospinning nanofiber scaffolds of polycaprolactone (PCL) were functionalized by oxidative self-polymerization of dopamine (PDA) on the surface followed by the modification of anticoagulant recombinant hirudin (rH) molecules. The morphology, structure, mechanical property, degradation behavior, cellular compatibility and blood compatibility of the multifunctional PCL/PDA/rH nanofiber scaffolds were evaluated. The diameter of the nanofibers was between 270–1030 nm. The ultimate tensile strength of the scaffolds was around 4 MPa and the elastic modulus increased with the amount of rH. The degradation tests in vitro indicated that the nanofiber scaffolds began to crack on the 7th day, but still maintained the nanoscale architecture within a month. The cumulative release of rH from the nanofiber scaffold was up to 95.9 % at 30th day. The functionalized scaffolds promoted the adhesion and proliferation of endothelial cells, while resisting platelet adhesion and enhancing anticoagulation effects. The hemolysis ratios of all scaffolds were <2 %. The nanofiber scaffolds are promising candidates for vascular tissue engineering.

An electrical signal is the key basis of normal physiological function of the nerve, and the stimulation of the electric signal also plays a very special role in the repair process of nerve injury. Electric stimulation is shown to be effective in promoting axonal regeneration and myelination, thereby promoting nerve injury repair. At present, it is considered that electric conduction recovery is a key aspect of regeneration and repair of long nerve defects. Conductive neural scaffolds have attracted more and more attention due to their similar electrical properties and good biocompatibility with normal nerves. Herein, PCL and MXene-PCL nerve guidance conduits (NGCs) were prepared; their effect on nerve regeneration was evaluated in vitro and in vivo . The results show that the NGCs have good biocompatibility in vitro . Furthermore, a sciatic nerve defect model (15 mm) of SD rats was made, and then the fabricated NGCs were implanted. MXene-PCL NGCs show similar results with the autograft in the sciatic function index, electrophysiological examination, angiogenesis, and morphological nerve regeneration. It is possible that the conductive MXene-PCL NGC could transmit physiological neural electric signals, induce angiogenesis, and stimulate nerve regeneration. This paper presents a novel design of MXene-PCL NGC that could transmit self-originated electric stimulation. In the future, it can be combined with other features to promote nerve regeneration.

It remains a challenge to achieve satisfactory balance between biodegradability and osteogenic capacity in biosynthetic bone grafts. In this study, we aimed to address this challenge by incorporating mesoporous bioactive glass (MBG) into poly(caprolactone-co-glycolide) (PGA-PCL) at gradient ratios. MBG/PGA-PCL (PGC/M) scaffolds with MBG incorporation ratio at 0, 10%, 25% and 40% (PGC/M0-40) were synthesized using a modified solvent casting-particulate leaching method, and their physiochemical and biological properties were comprehensively evaluated. PGC/M scaffolds exhibited highly perforated porous structure with a large-pore size of 300–450 μm, with ordered MBGs of around 6.0 nm mesopores size uniformly dispersed. The increase in MBG incorporation ratio significantly improved the scaffold surface hydrophilicity, apatite-formation ability and pH stability, increased the weight loss rate while insignificantly influenced the molecular chains degradation of PGA-PCL component, and facilitated the attachment, spreading, viability and proliferation of rat bone marrow stromal cells (rBMSCs) on scaffolds. Moreover, rBMSCs cultured on PGC/M10-40 scaffolds demonstrated enhanced ALP activity and osteogenesis-related gene expression in a MBG dose-dependent manner as compared with those cultured on PGC/M0 scaffolds. When implanted to the rat cranial bone defect, PGC/M25 and PGC/M40 scaffolds induced significantly better bone repair as compared to PGC/M0 and PGC/M10 scaffolds. Besides, the biodegradability of PGC/M scaffolds correlated with the MBG incorporation ratio. These data suggested this novel PGC/M scaffolds as promising bone repair biomaterial with highly tunable hydrophilicity, bioactivity, cytocompatibility, osteogenic activity as well as biodegradability.

Controlled printing of biodegradable and bioresorbable polymers at desired 3D scaffold is of great importance for cell growth and tissue regeneration. In this work, a novel electrohydrodynamic jet 3D printing technology with the resultant effect of electrohydrodynamic force and thermal convection was developed, and its feasibility to fabricate controllable filament composite scaffolds was verified. This method introduces an effective thermal field under the needle to simultaneously enhance the ink viscosity, jetting morphology controllability and printing structure solidify. The fabrication mechanisms of thermal convection on jetting morphology and printed structures feature were investigated through theoretical analysis and experimental characterization. Under optimized conditions, a stable and finer jet was formed; then with the use of this jet, various 3D structures were directly printed at a high aspect ratio ~30. Furthermore, the PCL/PVP composite scaffolds with the controllable filament diameter (~10 μm) which is closed to living cells were printed. Cell culture experiments showed that the printed scaffolds had excellent cell biocompatibility and facilitated cellular proliferation in vitro. It is a great potential that the developed electrohydrodynamic jet 3D printing technology might provide a novel approach to directly print composite synthetic biopolymers into flexibly scale structures for tissue engineering applications.

Abstract Shape memory polymers (SMPs) are a promising class of materials for biomedical applications due to their favorable mechanical properties, fast response, and good biocompatibility. However, it is difficult to achieve controllable sequential shape change for most SMPs due to their high deformation temperature and the simplex deformation process. Herein, shape memory composites based on polylactic acid (PLA) matrix and semi-crystalline linear polymer polycaprolactone (PCL) are fabricated using 4D printing technology. Compared with pure PLA, with the rise of PCL content, the 4D-printed PLA/PCL composites show decreased glass transition temperature ( T g ) from 67.2 to 55.2 °C. Through the precise control of the deformation condition, controllable sequential deformation with an outstanding shape memory effect can be achieved for the PLA/PCL shape memory composites. The response time of shape recovery is less than 1.2 s, and the shape fixation/recovery rates are above 92%. In order to simulate sequential petal opening and sequential drug releasing effects, a double-layer bionic flower and a drug release device, respectively, are presented by assembling PLA/PCL samples with different PLA/PCL ratios. The results indicate the potential applications of 4D-printed PLA/PCL composites in the field of bio-inspired robotics and biomedical devices. Graphic abstract

Superhydrophobic polycaprolactone (PCL) membranes with hierarchical structure were fabricated via alternate electrospinning/electrospraying techniques. Electrospun PCL/methyl silicone oil (PCL/MSO) nanofibers were employed as substrate. PCL/MSO-PCL microspheres (PCL/MSO-PCL MS ) hierarchical membrane was prepared via electrosprayed PCL MS as an additional layer on the substrate. Field emission scanning electron microscopy images showed the formation of hierarchical PCL/MSO-PCL MS membranes. Compared to pure PCL fibers substrate (120 ± 1.3°), the water contact angle (WCA) of MSO-modified PCL membrane was 142 ± 0.7°. The most interesting observation was that the WCA of PCL MS without any modification could be achieved to 146 ± 2.8°. On this basis, PCL/MSO-PCL MS hierarchical membrane possessed superhydrophobic surface with 150 ± 0.6° of WCA. The excellent surface roughness and air-pocket capacity of hierarchical membranes would make the membranes more hydrophobic. The maximum oil ( n -hexane) adsorption capacity of PCL/MSO-PCL MS membrane was 32.53 g g −1 . Oil–water separation efficiencies of the superhydrophobic membranes were all higher than 99.93% after 10 cycles. The hierarchically structured PCL superhydrophobic membranes indicate the potential applications of environmentally friendly biopolymers as separation membranes. © 2019 Wiley Periodicals, Inc. J. Post-surgical site infections (PSSI) pose significant challenges in clinic treatment, and the antibacterial modification of medical materials is an effective strategy to combat these issues. In this work, a monomeric N-halamines (DCDMH) was doped in PCL matrix via electrospinning process, and further processed using a conventional twisting method to yield nanofiber-based yarns. The micro/nano-structure of the PCL-based electrospun nanofibrous yarns (ENYs) enables the effective release of N-halamines and exhibits suitable storage stability after 30 days. The N-halamine modified ENYs effectively inhibited 100 % of E. coli O157:H7, S. aureus , and C. albicans within 1 min, resulting impressive antimicrobial capacities. The effect of N-halamines dosage on the morphology and mechanical properties of fabricated ENYs was assessed. Furthermore, the in vitro degradation, hemocompatibility, and protein adsorption properties of the synthesized nanofibrous yarns were also evaluated. The prepared ENYs with good hemocompatibility and antimicrobial properties, suggesting a novel approach for the development of medical materials, specifically for surgical sutures materials.
