An in-situ polymerized PEGDA hydrogel electrolyte is developed for AZIBs. • Ether groups disrupt water's hydrogen-bond network to suppress side reactions. • Carbonyl groups regulate Zn 2+ flux and enable dendrite-free deposition. • An Ah-level pouch cell validates the practical applicability of this work. Aqueous zinc-ion batteries (AZIBs) are promising for low-cost, safe, and eco-friendly energy storage. Vanadium-based cathode materials have garnered intense research interest due to their inherently high specific capacity. However, their practical applications are severely hindered by stability issues, including cathode dissolution, hydrogen evolution reactions, and zinc dendrite formation. Here, we report a novel in-situ polymerized hydrogel electrolyte based on polyethylene glycol diacrylate (PEGDA-HGE) to tackle these challenges. The abundant ether oxygen groups preferentially coordinate with water, disrupting its intrinsic hydrogen-bond networks and thus suppress water-induced parasitic hydrogen evolution and Vanadium-based cathode dissolution. Simultaneously, the carbonyl groups served as zincophilic sites not only facilitate ordered Zn 2+ transport but also homogenize interfacial Zn 2+ flux, thereby effectively preventing dendritic zinc growth. Meanwhile, the PEGDA chain can partially anchor OTf − , thereby suppressing its migration. As a result, the PEGDA-HGE delivers a high ionic conductivity (9.95 mS cm −1 ) and a large Zn 2+ transference number of 0.69. The Zn||Zn cells demonstrate exceptional cycling stability for 4380 h at 1 mA cm −2 /1 mAh cm −2 . Zn||V 2 O 3 full cells exhibit a high specific capacity of 454 mAh g −1 at 0.1 A g −1 and maintain a reversible capacity of 279 mAh g −1 across 500 cycles at 0.5 A g −1 . Notably, an Ah-level pouch cell achieves a high reversible capacity of 2642 mAh with stable operation for 50 cycles at 0.1 A g −1 . This work introduces a novel class of polymerizable monomer and achieves a significant advance in the fabrication of dual-functional hydrogel electrolytes for long-life aqueous AZIBs and Ah-level scalability. A novel dual functional in-situ polymerized PEGDA-based hydrogel electrolyte is designed for aqueous zinc-ion batteries with stable cyclability and Ah-level scalability. Abundant ether moieties immobilize free water, reduce water activity, and break H-bond networks to impede H + ion migration, thus suppressing cathode dissolution and HER/corrosion at Zn anode. Carbonyl groups served as zincophilic sites optimize Zn 2+ ion flux and effectively inhibit dendrite formation. Download: Download high-res image (206KB) Download: Download full-size image

Eradicating cutaneous infections induced by pathogenic microorganisms, such as bacteria, fungi, and viruses, via conventional topical drug delivery is challenging owing to limited epidermal penetration. To overcome this limitation, we fabricated a poly(ethylene glycol) diacrylate (PEGDA) microneedle array coated with a carboxymethyl chitosan (CMC) film embedded within situ-synthesized polydopamine-modified silver nanoparticles (PDA@Ag NPs) for enhanced antibacterial efficacy. This design utilizes microneedle (MN)-mediated skin perforation to enhance drug permeation, combined with rapid film dissolution for the efficient release of the payload. The experimental findings revealed that the PDA@Ag-CMC/PEGDA MNs possess favorable biocompatibility along with dual photothermal and photodynamic functionalities. Mild photothermal therapy (PTT) triggered by these MNs promotes the production of antibacterial reactive oxygen species (ROS), resulting in a high antibacterial rate of up to 98% against both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) under NIR irradiation at a PDA@Ag concentration of 40 μg mL−1. To assess transdermal delivery performance, lidocaine hydrochloride (LiH) (78 μg per MN array) was incorporated into the CMC film as a model drug. Physiological conditions confirmed accelerated LiH release from the PDA@Ag-LiH-CMC/PEGDA system upon aqueous exposure. This minimally invasive approach demonstrated promising broad-spectrum antibacterial activity, suggesting that the PDA@Ag-LiH-CMC/PEGDA platform could serve as a viable clinical solution for managing polymicrobial skin infections and alleviating wound pain.

In recent years, magnesium alloys (MgA) have been reckoned as the most promising material of biomedical importance on account of its excellent degradable properties and mechanical properties mimicking natural bone tissues. However, MgA are prone to rapid corrosion under physiological conditions, causing toxicity around the neighboring tissues. In addition, they are susceptible to bacterial colonization, a detrimental factor for medical causes. In this study, antibacterial material coated hydrogel-based micro-patterns were developed on MgA to achieve long-term antibacterial, antifouling, osteogenic, and cell-compatible properties. First, the Mg(OH) 2 nanosheet coating was prepared on the surface of MgA as a physical barrier to prevent the corrosion of MgA. Then the hydrogel micropatterns of poly(alendronate sodium methacrylate)/poly(dimethyldiallylammonium chloride)/poly(ethylene glycol) diacrylate (PALNMA/PDADMAC/PEGDA) of different sizes were constructed on the surface of the Mg(OH) 2 coating using the photomask method. Finally, an intelligently responsive antibacterial material hyaluronic acid/berberine (HA/BBR) was coated on MgA-Mg(OH) 2 -PALNMA/PDADMAC/PEGDA patterns via layer-by-layer self-assembly. The excellent antifouling performance of the samples is attributed to the topological structure of the pattern. Interestingly, as the pattern size of PALNMA/PDADMAC/PEGDA decreases, the antibacterial, antifouling, and cell compatibility properties of the samples gradually improve. UV–Vis spectra and bacterial plate count indicate that HA/BBR coating provide a pH and hyaluronidase (HAase) dual-responsive surface to kill the attached bacteria quickly. Finally, the in vitro experiments demonstrate excellent blood compatibility, cell compatibility and osteogenic properties of the modified MgA samples. Therefore, the intelligent multifunctional assembly of MgA presented here has a promising future in the field of metal implant materials.

Oxidative stress and inflammasome-driven inflammation are key contributors to atherosclerotic disease progression. Here, we report a ROS-responsive hyaluronic acid-PEGDA hydrogel designed for the sustained delivery of palmitoleic acid. In the study, catechol-functionalized hyaluronic acid was cross-linked with polyethylene glycol diacrylate to create an extracellular matrix mimic that possessed tunable mechanical strength and redox-sensitive degradability. It showed controlled release of palmitoleic acid under normal physiological conditions and a faster rate of release under oxidative conditions. In vitro studies using vascular endothelial cells and macrophages demonstrated good cytocompatibility and a marked reduction in intracellular reactive oxygen species, lipid peroxidation, and pro-inflammatory cytokine secretion. At the molecular level, treatment with the palmitoleic acid-loaded hydrogel resulted in activation of AMP-activated protein kinase, inhibition of NF-κB nuclear translocation, and suppression of NLRP3 inflammasome activation, as evidenced by decreased caspase-1 cleavage and IL-1β maturation. These findings indicate that ROS-responsive hydrogel-mediated delivery of palmitoleic acid effectively modulates oxidative and inflammatory signalling pathways relevant to atherosclerosis. Graphical

As one of the most frequently used chemotherapeutic drugs, doxorubicin (DOX) is accompanied by low accumulation in tumors and severe dose-limiting side effects with systemic administration, which limits its therapeutic index. In this work, a novel and injectable in situ photo-sensitive inorganic/organic hybrid hydrogel as a localized drug-delivery system was examined. It explored poly(ethylene glycol) double acrylates (PEGDA) as a polymeric matrix, DOX as a model drug, a TiO2@MWCNT nanocomposite as the photoinitiator and photosensitizer–photothermal agent for tumor therapy possessing a multi-mechanism using a single NIR laser. Briefly, a PEGDA solution containing DOX and TiO2@MWCNTs was injected into a tumor and rapidly gelled in vivo via a photo-crosslinking action triggered by a NIR laser. DOX release from the DOX/TiO2@MWCNTs/PEGDA hydrogel was sustained and long-lasting, over 10 days, indicating that the PEGDA gel acted as a drug depot. Simultaneously, a NIR laser light was adopted which can be absorbed and converted into reactive oxygen species (ROS) or local hyperthermia by TiO2@MWCNTs, leading to tumor cell death. This DOX/TiO2@MWCNTs/PEGDA hydrogel exhibited remarkable anti-proliferative activities against MCF-7 cancer cells in vitro. Experiments in vivo showed that a single intratumoral injection of this hydrogel with 808 nm laser irradiation was the most effective among all DOX formulations in the tumor-bearing mice models. There was a relatively small DOX distribution in normal tissues and much lower systemic toxicity than the control group (DOX-only). In general, it is believed that the novel photo-sensitive hybrid hydrogel system prepared in this study can afford high drug-loading, sustained and stable drug release, as well as repeated phototherapy of the tumor with the administration of a single dose.

Traumatic brain injury (TBI) is a common clinical disease, and the current hyperbaric oxygen and hypothermia therapy are suboptimal. Traumatic brain injuries were accompanied by acute inflammatory reactions, brain edema, neuron damage, and the destruction of blood–brain barrier. Herein, curcumin-loaded lysine/poly(ethylene glycol) diacrylate (PEGDA) hydrogels were constructed via Michael addition for traumatic brain injury treatments. The curcumin-loaded lysine/PEGDA hydrogels were injected into brain injury defect sites with complex anatomical and irregular structures. The real-time therapeutic effects of the curcumin-loaded hydrogels were evaluated by magnetic resonance imaging (MRI). Curcumin-loaded hydrogels can inhibit the inflammatory reaction and promote nerve repair. The curcumin-loaded hydrogel could provide a promising strategy for traumatic brain injury treatments.

Despite its significant potential in various disease treatments and diagnostics, microbiotherapy is consistently plagued by multiple limitations ranging from manufacturing challenges to in vivo functionality. Inspired by the strategy involving nonproliferating yet metabolically active microorganisms, we report an intracellular gelation approach that can generate a synthetic polymer network within bacterial cells to solve these challenges. Specifically, poly(ethylene glycol dimethacrylate) (PEGDA, 700 Da) monomers are introduced into the bacterial cytosol through a single cycle of freeze–thawing followed by the initiation of intracellular free radical polymerization by UV light to create a macromolecular PEGDA gel within the bacterial cytosol. The molecular crowding resulting from intracytoplasmic gelation prohibits bacterial division and confers robust resistance to simulated gastrointestinal fluids and bile acids while retaining the ability to secrete functional proteins. Biocompatibility assessments demonstrate that the nondividing gelatinized bacteria are effective in alleviating systemic inflammation triggered by intravenous Escherichia coli injection. Furthermore, the therapeutic efficacy of gelatinized Lactobacillus rhamnosus in colitis mice provides additional support for this approach. Collectively, intracellular gelation indicates a universal strategy to manufacture next-generation live biotherapeutics for advanced microbiotherapy.

Peripheral nerve injury is a major societal concern. Black phosphorus (BP) has inherent advantages over cell-based therapies in regenerative medicine. However, controlling spontaneous degradation and size-dependent cytotoxicity remains challenging and poses difficulties for clinical translation. In this study, we constructed zero-dimensional BP quantum dots (QDs) modified with antioxidant β-carotene and comprehensively investigated them in Schwann cells (SCs) to elucidate their potential for peripheral nerve repair. In vitro experiments demonstrated that BPQD@β-carotene has an inappreciable toxicity and good biocompatibility, favoring neural regrowth, angiogenesis, and inflammatory regulation of SCs. Furthermore, the PI3K/Akt and Ras/ERK1/2 signaling pathways were activated in SCs at the genetic, protein, and metabolite levels. The BPQD@β-carotene-embedded GelMA/PEGDA scaffold enhanced functional recovery by promoting axon remyelination and regeneration and facilitating intraneural angiogenesis in peripheral nerve injury models of rats and beagle dogs. These results contribute to advancing knowledge of BP nanomaterials in tissue regeneration and show significant potential for application in translational medicine.

The main aim of this study was to develop a 3D printed stem cell-responsive polyethylene glycol diacrylate (PEGDA) and sodium alginate (SA) composite hydrogel system (PEGDA/SA) loaded with basic fibroblast growth factor (bFGF) suitable for dental pulp tissue engineering. The PEGDA/SA
loaded with bFGF (PEGDA/SA-bFGF) at different concentration ratios were 3D printed via stereolithography (SLA) under optimal processing conditions, followed by ionic crosslinking. The morphological and porous structure of the 3D printed PEGDA/SA-bFGF constructs and release profile of the bFGF
were analyzed. The cellular compatibility of the hydrogel constructs was evaluated using human dental pulp stem cells (hDPSCs)in vitro. Subsequently, biocompatibility and tissue regenerative potential of the hydrogel constructs were evaluated in immunodeficient mice model. The in
vitro cell culture results of PEGDA/SA-bFGF constructs exhibited high cell compatibility and supported cell proliferation, irrespective of their concentration ratios. Thein vivo animal experiment results showed that the hDPSCs-laden PEGDA/SA (PEGDA/SA-hDPSCs) group failed to form
a good pulp structure even at various concentration ratios. When the mass ratio of PEGDA/SA was 25:1, hDPSCs-laden PEGDA/SA-bFGF (PEGDA/SA-bFGF-hDPSCs) group also failed to form dental pulp-like tissue but a partial loose connective tissue formation was observed. However, when the mass ratio
of PEGDA/SA reduces to 20:1 ~ 15:1, the PEGDA/SA-bFGF-hDPSCs group tends to form a well-organized pulp structure post 4 weeks implantation, thus highlighting the potential of this particular hydrogel scaffolding system for dental pulp tissue regenerative applications.

Photonic crystal (PC) hydrogels with unique micro/nanostructures have excellent prospects in many fields, such as sensing, displays, anticounterfeiting, information encryption, and adaptive camouflage. Herein, magnetic core–shell Fe3O4/SiO2 nanoparticles with a controlled size were synthesized, followed by PC hydrogels of arbitrary structures that were fabricated via the magnetically inducing self-assembly of magnetic nanoparticles in the poly(ethylene glycol) diacrylate (PEGDA) pregel solution and a subsequent digital micromirror device (DMD)-based UV polymerization process. By controlling the size of nanoparticles and magnetic field intensity, various PC hydrogel patterns with tunable structural colors were fabricated. Combining magnetic field control with DMD-based light modulation, high-throughput preparation of structural color PC hydrogels was achieved. Furthermore, the Fe3O4/SiO2 nanoparticles were assembled into poly(2-hydroxyethyl methacrylate) (PHEMA) with solvent responsiveness. Interestingly, the obtained PC hydrogels could swell in ethanol, resulting in a change in the structural color. Meaningfully, a dual-component PC hydrogel consisting of a PEGDA pattern and a PHEMA background was fabricated, and the invisible pattern in the initial state could be displayed and hidden through cyclic exposure to ethanol and ethylene glycol. The presented strategy for constructing the PC hydrogels from Fe3O4/SiO2 nanoparticles could provide valuable insights for the development of intelligent structural color materials, which exhibit significant potential in applications such as information storage, visual sensing, and anticounterfeiting.

Osteoarthritis (OA) is a common age-related degenerative disease characterized by changes in the local tissue environment as inflammation progresses. Inspired by the wind-dispersal mechanism of dandelion seeds, this study develops responsive biomimetic microsphere-drug conjugate for OA therapy and protection. The conjugate integrates dibenzaldehyde polyethylene glycol (DFPEG) with chitosan and polyethylene glycol diacrylate (PEGDA) through dynamic covalent bonds to form a dual-network hydrogel microsphere. Based on the progression of OA, the conjugate with the surface-anchored cyclic peptide cortistatin-14 (CST-14) achieves targeted drug therapy and a self-regulating hydrogel network. In cases of progressing inflammation (pH < 5), CST-14 dissociates from the microsphere surface (viz. the drug release rate increased) and inhibits TNF-α signaling to suppress OA. Concurrently, the monomer DFPEG responsively detaches from the hydrogel network and scavenges reactive oxygen species (ROS) to protect the cartilage tissue. The ROS scavenging of DFPEG is comparable to that of coenzyme Q10 and vitamin C. The degraded PEGDA microspheres provide tissue lubrication through reused conjugates. The rat OA model successfully achieved a synergistic therapeutic effect greater than the additive effect (1 + 1 > 2). This strategy offers an approach for anchoring amine-containing drugs and has marked potential for OA treatment and protection.

This paper proposes a patterned material composed of two types of photonic crystals based on CdS and SiO2 nanoparticles. We combined the photonic crystals with poly(ethylene glycol) diacrylate (PEGDA) by nanoparticle self-assembly and UV curing and successfully formed a material containing the different photonic crystals. The material prepared contains two kinds of photonic crystals with different refractive indices, CdS and SiO2 nanoparticles, and due to the large difference in refractive indices of the two materials, the reflection wavelengths of the photonic crystals composed of the two materials also have disparities when the incident angle changes. Macroscopically, the structural colors of the two photonic crystals change with the change in incident angle. In response to this phenomenon, we designed a variety of applications based on a multiangle patterned photonic crystal film to achieve a variety of information storage functions by changing the incident angle and, through the angle change, to achieve lipstick logo, mobile phone bar code, and antipeeping keyboard applications.
