Delta (B.1.617.2), a highly infectious variant of SARS-CoV-2, has been sweeping the world, and threatening the safety of human life seriously. It is urgent to develop a highly selective and sensitive assay to accurately identify the SARS-CoV-2 variant Delta. In this work, we constructed a graphene/CRISPR-dCas9 electrochemical biosensor to accurately identify SARS-CoV-2 variant Delta, where the signal was further amplified by embedded electrochemical probe [Ru(phen) 2 dppz]BF 4 . This detection assay could be finished within 47 min totally, with the detection limit of 1.2 pM and good reproducibility with a C·V.% of 2.48% (n = 5). And the biosensor could selectively identify Delta among SARS-CoV-2 and other variants, including Alpha, Beta, Gamma. This assay was further validated by 26 real clinical samples, showing 100% clinical sensitivity and 100% clinical specificity, which provides a new direction for identifying other SARS-CoV-2 variants in the future.

A novel electrochemical microsensor is constructed on a traditional acupuncture needle (AN) and used to monitor a biomarker of the SARS-CoV-2-N protein. The reversible interaction of the borate bond between the cis-diol in this glycoprotein and the phenylboronic acid in 4-mercaptophenylboronic acid (4-MPBA) is effectively exerted. This interaction was applied to anchor the SARS-CoV-2-N protein onto 4-MPBA, which is covalently self-assemblied onto electrodeposited AuNPs by the S-Au bond. Meldola blue was then electropolymerized around the protein template. After the template was eluting, three-dimensional nanocavities complementary with the protein were generated within the polymelola blue (pMB) layer. Interestingly, nanocavities could play a channel role for the electron-transfer of outer [Fe(CN)6]3−/4−, and the signal of electrochemical probe could be hindered after recombining the SARS-CoV-2-N protein, which laid a platform for the detection of this biomarker. After optimizing the influencing factors, the prepared microsensor exhibits a linear range of 0.1 ~ 1000 ng mL−1 with a low detection limit of 0.01 ng mL−1 (S/N = 3). In particular, the sensing ability was dramatically affected by the thickness correlative factors for the polymer matrix. The suitable thickness is effective for the sensing signals, which corresponds to the behavior of surface-imprinted polymer. The microsensor showed comparatively high sensitivity and selectivity and practically detected the SARS-CoV-2-N protein in the serum sample, which is of scientific significance for the development of the electrochemical microsensor and acupuncture.

Viral respiratory infections are major human health concerns. The most striking epidemic disease, COVID-19 is still on going with the emergence of fast mutations and drug resistance of pathogens. A few polysaccharide macromolecules from traditional Chinese medicine (TCM) have been found to have direct anti-SARS-CoV-2 activity but the mechanism remains unclear. In this study, we evaluated the entry inhibition effect of Lycium barbarum polysaccharides (LBP) in vitro and in vivo. We found LBP effectively suppressed multiple SARS-CoV-2 variants entry and protected K18-hACE2 mice from invasion with Omicron pseudovirus (PsV). Moreover, we found LBP interfered with early entry events during infection in time-of-addition (TOA) assay and SEM observation. Further surface plasmon resonance (SPR) study revealed the dual binding of LBP with Spike protein and ACE2, which resulted in the disruption of Spike-ACE2 interaction and subsequently triggered membrane fusion . Therefore, LBP may act as broad-spectrum inhibitors of virus entry and nasal mucosal protective agent against newly emerging respiratory viruses , especially SARS-CoV-2.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) main protease (Mpro) has been regarded as one of the ideal targets for the development of antiviral drugs. The currently used methods for the probing of Mpro activity and the screening of its inhibitors require the use of a double-labeled peptide substrate. In this work, we suggested that the label-free peptide substrate could induce the aggregation of AuNPs through the electrostatic interactions, and the cleavage of the peptide by the Mpro inhibited the aggregation of AuNPs. This fact allowed for the visual analysis of Mpro activity by observing the color change of the AuNPs suspension. Furthermore, the co-assembly of AuNPs and peptide was achieved on the peptide-covered electrode surface. Cleavage of the peptide substrate by the Mpro limited the formation of AuNPs/peptide assembles, thus allowing for the development of a simple and sensitive electrochemical method for Mpro detection in serum samples. The change of the electrochemical signal was easily monitored by electrochemical impedance spectroscopy (EIS). The detection limits of the colorimetric and electrochemical methods are 10 and 0.1 pM, respectively. This work should be valuable for the development of effective antiviral drugs and the design of novel optical and electrical biosensors.Keywords:SARS-CoV-2 main protease;colorimetry;electrochemical impedance spectroscopy;gold nanoparticles

Emerging SARS-CoV-2 variants are the most serious problem for COVID-19 prophylaxis and treatment. To determine whether the SARS-CoV-2 vaccine strain should be updated following variant emergence like seasonal flu vaccine, the changed degree on antigenicity of SARS-CoV-2 variants and H3N2 flu vaccine strains was compared. The neutralization activities of Alpha, Beta and Gamma variants’ spike protein-immunized sera were analysed against the eight current epidemic variants and 20 possible variants combining the top 10 prevalent RBD mutations based on the Delta variant, which were constructed using pseudotyped viruses. Meanwhile, the neutralization activities of convalescent sera and current inactivated and recombinant protein vaccine-elicited sera were also examined against all possible Delta variants. Eight HA protein-expressing DNAs elicited-animal sera were also tested against eight pseudotyped viruses of H3N2 flu vaccine strains from 2011–2019. Our results indicate that the antigenicity changes of possible Delta variants were mostly within four folds, whereas the antigenicity changes among different H3N2 vaccine strains were approximately 10–100-fold. Structural analysis of the antigenic characterization of the SARS-CoV-2 and H3N2 mutations supports the neutralization results. This study indicates that the antigenicity changes of the current SARS-CoV-2 may not be sufficient to require replacement of the current vaccine strain.

To tackle the demand for highly accurate and real-time diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a highly sensitive optical fiber SARS-CoV-2 antigen sensor based on triangular gold nanosheets/gold film coupling is proposed. The coupling effect between the local surface plasmon resonance effect of the triangular gold nanosheets and the surface plasmon resonance effect of the gold film increases the local electric field, resulting in higher sensitivity and lower detection limit of the sensor, and the refractive index sensitivity of the sensor is increased to 3894.3 nm/RIU. The antigen reacts specifically with the viral antibody adsorbed on the surface of the optical fiber, which leads to a small change in refractive index, which ultimately leads to a shift in the resonance peak of the optical fiber, thus enabling antigen detection. The biosensor showed good response in the range of SARS-CoV-2 antigen concentration from 0.035 to 100 ng/mL, and the limit of detection (LOD) was 0.017 ng/mL (S/N=3). The biosensor has good selectivity and stability, possesses a short response time (10 min), which provides a new idea for the detection of SARS-CoV-2 in the biomedical field.

Developing an efficient and sensitive method for detecting the coronavirus disease (COVID-19) is crucial. In this study, a phase-dependent SPR biosensor has been developed to detect the SARS-CoV-2 S protein with high sensitivity. The phase shifts of the SPR spectral interferogram are extracted using the windowed Fourier transform (WFT) method. The summation of the phase shifts of the prominent frequency, which make up the SPR spectral interferogram, is used as a numerical index to reflect the “negative” or “positive” results. The SARS-CoV-2 S protein antibody is modified onto an Au film, which serves as a sensing surface when detecting the SARS-CoV-2 S protein. The SARS-CoV-2 S protein antibody functionalized Au nanoparticles are used to form a (anti-S@AuNPs)-(SARS-CoV-2 S protein)-(Au film) sandwich structure, which reduces the SPR detection limitation to 1 ag/mL. An external laser is then used to induce the localized surface plasmon resonance (LSPR) between the AuNPs and the Au film in the sandwich structure, enhancing the detecting sensitivity by 357 %. This study provides a label-free method for detecting the SARS-CoV-2 S protein with low concentrations, indicating its potential in detecting the SARS-CoV-2 virus and viral disease diagnosis.

Given the susceptibility of Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV-2) and Norovirus (NoV) to survive in cold chain food, thereby posing significant public health risks, we present a novel approach for biosensor development utilizing a graphene field-effect transistor (GFET) modified with nucleic acid aptamers. The biosensor’s innovative design incorporates 1-pyrenebutyric acid N-hydroxysuccinimide ester (PBASE) as a coupling agent to facilitate the attachment of nucleic acid aptamers onto channel graphene. This modification induces a redistribution of charge on the graphene surface, resulting in a shift of the Dirac point upon target capture by the nucleic acid aptamer. Through this pioneering methodology, we successfully engineered SARS-CoV-2 GFET and NoV GFET biosensors capable of detecting trace amounts of SARS-CoV-2 and norovirus within a rapid 5-minute timeframe, showcasing detection limits of 33 fg/mL and 6.17 pg/mL, respectively. Subsequently, we applied these sensors to detect SARS-CoV-2 in frozen meat and norovirus in shellfish, yielding promising results with excellent specificity and stability. This groundbreaking sensing mechanism holds significant promise for the detection of foodborne viruses across a diverse range of food samples.

Upconversion nanoparticles (UCNPs) are ideal donors for luminescence resonance energy transfer (LRET)-based biosensors due to their excellent upconversion luminescence properties . However, the relatively large size of antibodies and proteins limits the application of UCNPs-based LRET biosensors in protein detection because the large steric hindrance of proteins leads to low energy transfer efficiency between UCNPs and receptors. Herein, we developed a magnetic responsive UCNPs-based LRET biosensor to control the coupling distance between antibody-functionalized UCNPs (Ab-UCNPs) as donors and antibody-PEG linker-magnetic gold nanoparticles (Ab-PEG-MGNs) as acceptors for ultrasensitive and highly selective detection of SARS-CoV-2 spike proteins . Our results showed that this platform reversibly shortened the coupling distance between UCNPs and MGNs and enhanced the LRET signal with a 10-fold increase in the limit of detection (LOD) from 20.6 pg/mL without magnetic modulation to 2.1 pg/mL with magnetic modulation within 1 h. The finite-difference time-domain (FDTD) simulation with cyclic distance change confirmed the distance-dependent LRET efficiency under magnetic modulation, which supported the experimental results. Moreover, the applications of this magnetic-responsive UCNP-based LRET biosensor could be extended to other large-size biomolecule detection.

In this work, we designed an ECL ratiometric biosensor with a three-stranded Y-type DNA (Y-DNA) probe and induced a hybridization chain reaction (HCR) for the highly sensitive detection of SARS-CoV-2 nucleic acid. The important component of this system is the self-assembled Y-Shaped probe based on three nucleic acids. Y1, Y2, and Y3 can be linked by complementary base pairing to Hairpin1 (H1), Hairpin2 (H2), and Ru modified DNA (Ru1), respectively. H1 and H2 can trigger the HCR reaction when activated by the SARS-CoV-2 RdRp gene and the 5′ end of Ru1. The 5′ end of Ru1 is modified with the Ru complex, which can produce a strong electrochemiluminescence luminescence signal at 620 nm under an applied voltage. Through the amplification of Y-DNA-induced HCR reaction, Ru1 on the electrode surface gradually increased, the ECL signal at 460 nm was gradually quenched, and the signal at 620 nm was steadily generated. The SARS-CoV-2 RdRp gene can be quantified according to the degree of decrease of ECL signal at 460 nm and the increase of ECL signal at 620 nm. Combining the two signal amplification strategies, this ratiometric ECL biosensor can accurately and efficiently detect the target gene with a detection limit of 59 aM.

Quick and highly sensitive detection of the SARS-CoV-2 virus can efficiently reduce the possibility of viral spreading. In this study, a differential-phase surface plasmon resonance (SPR) biosensor is constructed to directly detect the SARS-CoV-2 virus. The SARS-CoV-2 spike protein antibodies are applied as bioprobes and functionalized on Au film, forming a sensing surface. A numerical analysis of the phase shifts of the SPR interferogram, termed the index of phase shifts (IoPS), is presented, which does not depend on a reference beam or an s-component beam. The IoPS can sensitively reflect small changes in the refractive index (as low as 3.75 × 10−10 RIU) when NaCl solution is used as an analyte. The limit of detection (LOD) of the proposed SPR biosensor in detecting the SARS-CoV-2 S protein is 1 ag mL−1. In comparison with a SARS-CoV-2 negative control (NC) sample collected from healthy people, which has an IoPS of ΔϕNC = 39°, the SPR biosensor shows a very strong response to the SARS-CoV-2 virus collected from infected people with a phase shift of ΔϕIoPS = 566°. To further improve the detection sensitivity, a sandwich structure is designed by applying SARS-CoV-2 spike protein antibody functionalized gold nanoparticles (anti-S@AuNPs). This reduces the LOD for the detection of the SARS-CoV-2 S protein to 0.1 ag mL−1.

Clinical studies have demonstrated that many traditional Chinese medicines (TCM) exhibit not only antiviral effects but also efficacy in alleviating clinical symptoms of Coronavirus Disease 2019 (COVID-19). However, the pharmacologically active constituents responsible for their anti-COVID-19 efficacy remain unclear. This study aimed to establish a novel strategy for identifying active ingredients from herbs clinically used for COVID-19. An integrated approach combining data mining with Affinity Ultrafiltration (AUF) was developed. Initially, using a data mining strategy, high-frequency herbs were selected from the herbs clinically used in COVID-19. Furthermore, AUF technology was used to screen for potential bioactive components from the high-frequency herbs. The anti-COVID-19 potential of active compounds was assessed through enzyme activity assays and molecular docking, followed by validation in cellular and animal models. Data mining revealed that Glycyrrhiza uralensis Fisch., Lonicera japonica Thunb. and Forsythia suspensa (Thunb.) Vahl were the high-frequency herbs used in COVID-19. Five potential 3-Chymotrypsin-like protease (3CL pro ) inhibitors were screened from three herbs via AUF and identified using high-resolution mass spectrometry. Further enzymatic and cellular assays demonstrated that Licochalcone C (LCC) and Forsythiaside A (FTA) inhibited Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) replication at micromolar concentrations. Notably, FTA treatment significantly suppressed the elevation of pulmonary parameters and inflammatory mediators induced by SARS-CoV-2 nucleocapsid protein in mice. In summary, this study proposes a novel strategy integrating data mining with AUF to discover active compounds from TCM. Two components, LCC and FTA, were identified as dose-dependent inhibitors of SARS-CoV-2 Omicron strain replication in vitro.
