Combined treatment with metformin and melatonin improves placental mitochondrial function via the AMPK/SIRT3 pathway in an LPS-induced model of preeclampsia

Zhen Zhou,Yongfen Bao,Rongjie Li,Rui Yang,Qingyu Peng,Tianci Lu,Binyu Wang,Ming Zhang,Jie Xu,Ying Xiang,Shigang Shan,Liqiong Huang

Background Preeclampsia (PE) is a severe hypertensive disorder in pregnancy, and disruption of placental energy homeostasis, particularly mitochondrial dysfunction, plays a critical role in its pathogenesis. Melatonin (MEL) and Metformin (MET), an effect associated through the AMP-activated protein kinase/Sirtuin 3 (AMPK/SIRT3) signaling pathway. However, the precise mechanisms remain unclear. Materials and methods The PE rat model was established in Sprague–Dawley (SD) rats by lipopolysaccharide (LPS) injection after confirmation of pregnancy. Pathological changes, protein expression, inflammatory and angiogenic factors, and mitochondrial status in placental tissue and HTR-8/SVneo trophoblasts were assessed by histology, immunohistochemistry, Western blotting, ELISA, RT-qPCR, reactive oxygen species (ROS) detection, transmission electron microscopy, and measurements of mitochondrial membrane potential and cytosolic calcium. Results Both MEL and MET individually activated the AMPK/SIRT3 signaling pathway, thereby enhancing mitochondrial function and placental energy homeostasis, while attenuating LPS-induced oxidative stress and inflammatory damage in placental tissue and trophoblast cells. Conclusions MEL and MET, when administered together, improve placental mitochondrial dysfunction and inflammatory injury in PE, which is associated with activation of the AMPK/SIRT3 signaling pathway, suggesting a potential therapeutic strategy for preeclampsia using clinically available agents.

Mitochondrial oxidative stress inhibited Sirt3/Foxo3/PPARα pathway and aggravated copper and zinc co-deficiency-induced hepatic lipotoxicity in a fish model

Zhong Chong-Chao,Zhang Xu,Pantopoulos Kostas,Song Chang-Chun,Yang Hong,Wei Xiao-Lei,Luo Zhi

Copper (Cu) and zinc (Zn) are essential trace elements for terrestrial animals. Studies suggested that single dietary Cu- or Zn- deficiency lead to liver lipid deposition and cause metabolic dysfunction-associated steatotic liver disease (MASLD). However, the mechanisms by which dietary Cu and Zn co-deficiency promote hepatic lipid deposition and metabolism remain unknown. Our study found that, compared to single Cu- or Zn- deficiency, Cu and Zn co-deficiency significantly increased lipid deposition, induced mitochondrial oxidative stress, disrupted mitochondrial structure and function, and inhibited lipolysis in hepatocytes and whole liver tissue. Mechanistically, Cu and Zn co-deficiency-induced lipotoxicity was mediated by sirtuin 3 (Sirt3). Sirt3 inhibited the acetylation and degradation of forkhead box O3 (Foxo3), preventing its binding to the peroxisome proliferator-activated receptor alpha (PPARα) promoter, which accordingly increased lipid deposition. Overall, for the first time, we elucidated the mechanism by which Cu and Zn co-deficiency aggravates hepatic lipotoxicity and identified the critical regulatory role of the Sirt3/Foxo3/PPARα pathway during this process, with the fish as the model.

Mechanistic Study on SIRT3/RORγt/STAT3-Regulated Th17-Targeted Bionic Black Phosphorus Quantum Dot Cluster Nanozyme for Improving Sepsis-Associated Acute Kidney Injury

Qinbiao Chen,Yanli Ma,Yuewu Tang,Ni Du

Sepsis-related acute kidney injury (SA-AKI) represents a severe complication associated with elevated mortality rates, primarily driven by imbalanced Th17 immune responses and mitochondrial dysfunction. Consequently, there is an urgent need for a versatile nanomaterial that can enhance mitochondrial performance while also exerting anti-inflammatory effects. A synthetically produced nanoenzyme has the potential to address this challenge. In this study, we created a biomimetic nanozyme (BPQDs@Raw264.7@Kim-1) by engineering clusters of black phosphorus quantum dots (BPQDs) that are coated with macrophage membranes and functionalized with kidney injury molecule-1 (Kim-1) for targeted delivery. This nanozyme improves its antioxidant and anti-inflammatory properties by clustering black phosphorus quantum dots, while the stability of the drug is maintained through encapsulation within Raw264.7 cell membranes. The system effectively achieves homologous targeting via external Kim-1 molecules. The objective of this nanozyme is to modulate the SIRT3/RORγt/STAT3 pathway to reduce Th17-driven inflammation and alleviate SA-AKI. In vitro experiments demonstrated that the nanozyme possesses excellent biocompatibility, efficient cellular uptake, and the ability to scavenge reactive oxygen species (ROS) in LPS-stimulated HK-2 cells. It mitigated mitochondrial injury, decreased apoptosis, and lowered the levels of pro-inflammatory cytokines. Mechanistically, the nanozyme's overexpression of SIRT3 inhibited RORγt/STAT3 signaling, resulting in reduced Th17 differentiation and inflammatory cytokine release. Importantly, silencing SIRT3 negated these therapeutic benefits, underscoring its critical role. This research introduces a targeted nanozyme approach that alleviates SA-AKI by simultaneously restoring mitochondrial balance and suppressing Th17 inflammation through the SIRT3/RORγt/STAT3 pathway, presenting a promising therapeutic strategy for organ injury caused by sepsis.

Revitalizing mitochondrial function and empowering spinal cord recovery using copper-catalyzed Pdots

Bao-Feng Zhao,Fan Wang,Jia-Chen Sun,Jin-Peng Gao,Li-Juan Zhao,Zhao-Liang Shen,Zhi-Peng Li,Yi-Xuan Wang,Yuan Wang,Dan Li,Xi-Fan Mei

Spinal cord injury (SCI) inflicts devastating consequences for health. While drugs have been employed to alleviate SCI symptoms, their complex synthesis procedures and the need for specialized drug delivery raise concerns regarding administration. Herein, we utilize a green synthesis protocol to produce polymer quantum dots (Pdots) from polyvinylpyrrolidone precursors, empowered by the catalytic prowess of copper. These Copper-catalyzed Pdots (CPdots) not only offer enhanced improved fluorescence properties but also exhibit remarkable antioxidant, anti-inflammatory, and mitochondrial function repair effects. An intriguing finding emerged from our research as CPdots were observed to activate the AMPK/PGC-1α/Sirt3 pathway-a distinctive molecular mechanism that combats mitochondrial dysfunction, and mitigates calcium overload associated with acute SCI. Furthermore, the rapid metabolism and exceptional biocompatibility of our CPdots reinforce their safety and efficacy.

Mitochondrial dysfunction-associated cellular senescence is partially involved in bleomycin-induced pulmonary fibrosis in mice

Ya-Xin Yao,Xue Lu,Zhao Li,Hao-Yu Liao,Zhi-Bing Liu,Hui Zhao,Hua Wang,De-Xiang Xu,Zhu-Xia Tan

Accumulating data demonstrate that bleomycin (BLM), a clinically used antineoplastic drug, induces pulmonary interstitial fibrosis. However, the specific mechanism remains unclear. This study was carried out for the purpose of evaluating the effect of mitochondrial dysfunction-associated senescence on BLM-induced pulmonary fibrosis. Adult C57BL/6 J mice were intratracheally instilled with BLM (2.5 mg/kg). Pulmonary α-SMA and Vimentin, two indicators of epithelial-mesenchymal transition (EMT), were increased, and pulmonary collagen deposition was shown in BLM-treated mice. Pulmonary p16 and p21, two biomarkers of cell cycle arrest, were elevated, and pulmonary SASP indicators were up-regulated in BLM-treated mice. The reduction of mitochondrial area in BLM-treated mouse lungs was revealed by transmission electron microscopy. ATP content was reduced in BLM-treated mouse lungs. Pulmonary SOD2 and IDH2, two enzymes located in mitochondria, were decreased in BLM-treated mice. Sirtuin3 (SIRT3), an NAD + -dependent deacetylase located in mitochondria, was down-regulated in BLM-treated mouse lungs. Interestingly, Sirt3 gene knockout aggravated BLM-evoked mitochondrial dysfunction-associated senescence in mouse lungs. Sirt3 gene knockout exacerbated BLM-induced lung fibrosis. Conversely, nicotinamide mononucleotide (NMN), an NAD + precursor, weakened BLM-induced down-regulation of mitochondrial SIRT3 activity in mouse lungs. NMN pretreatment attenuated BLM-induced mitochondrial dysfunction-associated senescence in mouse lungs. Finally, NMN pretreatment alleviated BLM-induced EMT and lung fibrosis. These results indicate that mitochondrial dysfunction-associated senescence partially contributed to BLM-induced pulmonary fibrosis.

N-Acetylneuraminic acid triggers endothelial pyroptosis and promotes atherosclerosis progression via GLS2-mediated glutaminolysis pathway

Liu Zhaohong,Xiang Peng,Zeng Shengmei,Weng Ping,Wen Yilin,Zhang Wanping,Hu Hui,Zhao Dezhang,Ma Limei,Yu Chao

Vascular endothelial injury initiates atherosclerosis (AS) progression. N -Acetylneuraminic acid (Neu5Ac) metabolic disorder was found to intensify endothelial mitochondrial damage. And GLS2-associated glutaminolysis disorder contributed to mitochondrial dysfunction. However, mechanisms underlying Neu5Ac-associated mitochondrial dysfunction as well as its association with GLS2 remains unclear. In this study, we constructed GLS2 −/− ApoE −/− mice by using HBLV-GLS2 shRNA injection. And methods like immunofluorescence, western blotting, transmission electron microscopy were applied to detect profiles of endothelial injury and AS progression both in vivo and in vitro. We demonstrated that Neu5Ac accumulation increased GLS2 expression and promoted glutaminolysis disorder, which further induced endothelial mitochondrial dysfunction via a pyroptosis-dependent pathway in vivo and in vitro. Mechanically, Neu5Ac interacted with SIRT3 and led to FOXO3a deacetylation and phosphorylation, further facilitated c-Myc antagonism and ultimately increased GLS2 levels. Inhibition of GLS2 could improve mitochondrial function and mitigate pyroptosis process. In addition, blocking Neu5Ac production using neuraminidases (NEUs) inhibitor could rescue endothelial damage and alleviate AS development in ApoE −/− mice. These findings proposed that Neu5Ac induced GLS2-mediated glutaminolysis disorder and then promoted mitochondrial dysfunction in a pyroptosis-dependent pathway. Targeting GLS2 or inhibiting Neu5Ac production could prevent AS progression.

Bioenergetic-active exosomes for cartilage regeneration and homeostasis maintenance

Xulong Liu,Shangtong Jiang,Ting Jiang,Ziyang Lan,Xin Zhang,Zhenyu Zhong,Xiaodan Wu,Cunjing Xu,Yingying Du,Shengmin Zhang

Cartilage regeneration relies on adequate and continuous bioenergy supply to facilitate cellular differentiation and extracellular matrix synthesis. Chondrocytes frequently undergo energy stress under pathological conditions, characterized by disrupted cellular metabolism and reduced adenosine triphosphate (ATP) levels. However, there has limited progress in modulating energy metabolism for cartilage regeneration thus far. Here, we developed bioenergetic-active exosomes (Suc-EXO) to promote cartilage regeneration and homeostasis maintenance. Suc-EXO exhibited a 5.42-fold increase in ATP content, enabling the manipulation of cellular energy metabolism by fueling the TCA cycle. With continuous energy supply, Suc-EXO promoted BMSC chondrogenic differentiation via the P2X7-mediated PI3K-AKT pathway. Moreover, Suc-EXO improved chondrocyte anabolism and mitochondrial homeostasis via the P2X7-mediated SIRT3 pathway. In a rabbit cartilage defect model, the Suc-EXO–encapsulated hydrogel notably promoted cartilage regeneration and maintained neocartilage homeostasis, leading to 2.26 and 1.53 times increase in Col2 and ACAN abundance, respectively. These findings make a remarkable breakthrough in modulating energy metabolism for cartilage regeneration, offering immense potential for clinical translation.

A novel neuroprotective peptide against cognitive disorder in mice by regulating hippocampal oxidative stress and synaptic plasticity

Zhiqiang Lu,Yuan Gao,Zhijie Bao,Songyi Lin

The peptide Ser-Phe-Gly-Asp-Ile (SFGDI), derived from sea cucumber, has demonstrated neuroprotective effects in cell models; however, the in vivo neuroprotective efficacy of SFGDI remains unexplored. In this study, we utilized a cognitive disorder mouse model induced by scopolamine and observed that daily oral administration of SFGDI at a dosage of 40 mg/kg resulted in improved behavioral performance in the Passive avoidance test, the Channel-type water maze test, and the Novel object recognition test (P < 0.05). Furthermore, our investigation into the underlying neuroprotective mechanisms of SFGDI revealed that it reduced oxidative stress levels through the activation of the Sirt3/SOD/ROS pathway, thereby balancing hippocampal unsaturated lipids (P < 0.05) in the mouse hippocampus. Moreover, SFGDI significantly decreased hippocampal neuroinflammation by modulating the activation of microglia and hypertrophy of astrocytes. Additionally, SFGDI was found to enhance synaptic plasticity in the mouse hippocampus by activating the ChAT/p-CaMKⅡ/BDNF pathway and increasing spine density in neuron cells (P < 0.05), particularly mushroom spines (P < 0.05). These experiments demonstrate that SFGDI exhibits neuroprotective effects by regulating hippocampal oxidative stress through the Sirt3/SOD/ROS pathway and synaptic plasticity via the ChAT/p-CaMKⅡ/BDNF pathway in the mouse hippocampus, thus emerging as a potential neuro-nutraceutical agent for alleviating memory impairment.

Ginsenoside Rb1 improves energy metabolism after spinal cord injury

Wen Shan,Zou Zhi-Ru,Cheng Shuai,Guo Hui,Hu Heng-Shuo,Zeng Fan-Zhuo,Mei Xi-Fan

Mitochondrial damage caused by oxidative stress and energy deficiency induced by focal ischemia and hypoxia are important factors that aggravate diseases. Studies have shown that ginsenoside Rb1 has neurotrophic and neuroprotective effects. However, whether it influences energy metabolism after spinal cord injury remains unclear. In this study, we treated mouse and cell models of spinal cord injury with ginsenoside Rb1 . We found that ginsenoside Rb1 remarkably inhibited neuronal oxidative stress , protected mitochondria, promoted neuronal metabolic reprogramming , increased glycolytic activity and ATP production, and promoted the survival of motor neurons in the anterior horn and the recovery of motor function in the hind limb. Because sirtuin 3 regulates glycolysis and oxidative stress , mouse and cell models of spinal cord injury were treated with the sirtuin 3 inhibitor 3-TYP. When Sirt3 expression was suppressed, we found that the therapeutic effects of ginsenoside Rb1 on spinal cord injury were remarkably inhibited. Therefore, ginsenoside Rb1 is considered a potential drug for the treatment of spinal cord injury , and its therapeutic effects are closely related to sirtuin 3.

Photothermal Microneedle Hydrogel Patch for Refractory Soft Tissue Injuries through Thermosensitized Anti-Inflammaging Modulation

Wanbo Zhu,Quan Liu,Ziheng Zhang,Yingjie Wang,Jiawei Mei,Dongdong Xu,Jun Zhou,Zheng Su,Xianzuo Zhang,Chen Zhu,Jiaxing Wang,Junchen Zhu,Xiaochun Peng,Xianlong Zhang

Soft tissue injuries (STIs) are the most common cause of extremity pain and motion dysfunction. Persistent inflammatory activation of immune cells characterized by senescence-associated secretory phenotype (SASP) and mitochondrial stress are considered the primary causes of STIs, a pathological process also termed inflammaging. Meanwhile, scavenging excessive “cellular waste” in the inflammaging microenvironment and further activating tissue repair processes remain elusive. Herein, an anti-inflammaging photothermal hydrogel microneedle patch for treating STIs is developed. Taurine-loaded Prussian blue nanoparticles (Taurine@PB) are encapsulated in a methacrylate-based hyaluronic acid hydrogel (HAMA) and further fabricated into taurine@PB@HAMA@microneedles (TPH@MN) patches. The acidic microenvironment of chronic inflammation and mild photothermal effects promote taurine release and anti-inflammaging immunomodulation, inhibiting mitochondrial stress via the SIRT3-NF-κB axis to promote glycolytic metabolic microenvironment of neutrophils reprogramming toward oxidative phosphorylation metabolism. Furthermore, TPH@MN activates macrophage efferocytosis and initiates the process of tissue repair. In mouse models of chronic diabetic wounds and tibialis anterior (TA) muscle injury, TPH@MN inhibits SASP expression and promotes STIs healing through thermosensitized anti-inflammaging immunomodulation. In summary, TPH@MN circumvents the side effects of systemic administration, providing new translatable options in the treatment modalities for patients suffering from STIs worldwide.

Therapeutic Effects of Huazhuojiedu Decoction on Precancerous Lesions of Gastric Cancer by Regulating Mitophagy

Tianxiao Yang,Pingping Zhou,Ziwei Zhang,Xinyu Hao,Shaowei Liu,Haoyu Chen,Yuhua Wang,Yangang Wang

This research aims to explore the therapeutic effect and potential mechanisms of Huazhuojiedu decoction (HZJD) for alleviating precancerous lesions of gastric cancer (PLGC) both in vivo and in vitro. HZJD is a traditional Chinese herbal formula consisting of 11 herbs. Sprague-Dawley (SD) rats were randomly divided into four subgroups: control group, model group, positive drug group, and HZJD group. Hematoxylin-eosin (H&E) staining, high iron diamine-alcian blue (HID-AB) staining, alcian blue-periodic acid Schiff (AB-PAS) staining, immunohistochemistry, immunofluorescence, RT-qPCR, and Western blot assays were performed after 10 weeks of HZJD treatment. In vitro, the cell counting kit-8 (CCK-8) and 5-ethynyl-2'-deoxyuridine (EdU) assays were used to detect cell proliferation. RT-qPCR and Western blot assays were performed to evaluate mitophagy levels. The results indicated that HZJD could retard the pathological progression in PLGC rats and reduce PLGC cell proliferation. Treatment with HZJD significantly increased the mRNA and protein expression levels of Sirt3, Foxo3a, Parkin, and LC3 II/I, while decreasing the mRNA and protein expression levels of p62 and Tomm20. HZJD was found to have the ability to reverse the decline in mitophagy activity both in vivo and in vitro. In conclusion, the study assessed the impact of HZJD and provided evidence regarding its potential molecular mechanism.
