SERS combined with the difference in bacterial extracellular electron transfer ability to distinguish Shewanella

Mingxia Jiang,Anxun Chen,Jinghong Chen,Hui Zeng,Weikang Zhang,Yong Yuan,Lihua Zhou

Shewanella plays an important role in geochemical cycle, biological corrosion, bioremediation and bioenergy. The development of methods for identifying Shewanella can provide technical support for its rapid screening, in-depth research into its extracellular respiratory mechanism and its application in ecological environment remediation. As a tool for microbial classification, identification and detection, Surface-enhanced Raman scattering (SERS) has high feasibility and application potential. In this work, bio-synthesized silver nanoparticles (AgNPs) were used as SERS substrates to effectively distinguish different types of Shewanella bacteria based on the difference in bacterial extracellular electron transfer (EET) ability. AgNPs were combined with the analyzed bacteria to prepare “Bacteria-AgNPs” SERS samples, which can strongly enhance the Raman signal of the target bacteria and reliably obtain spatial information of different molecular functional groups of each bacteria. Our developed approach can effectively distinguish between non-metal reducing and metal-reducing bacteria, and can further distinguish the three subspecies of Shewanella ( Shewanella oneidensis MR-1, Shewanella decolorationis S12, and Shewanella putrefaciens SP200) at the genus and species level. The Raman signal enhancement is presumably caused by the excitation of local surface plasma (LSP) and the enhancement of surrounding electric field. Therefore, our developed method can achieve interspecific and intraspecies discrimination of bacteria. The proposed method can be extended to distinguish other metal-reducing bacteria, and the novel SERS active substrates can be developed for practical applications.

How iron-bearing minerals affect the biological reduction of Sb(V): A newly discovered function of nitrate reductase

Haikun Zhang,Yanyu Sun,Manman Cheng,Xiaori Sui,Yanyan Huang,Xiaoke Hu

As a toxic element of global concern, the elevated concentration of antimony (Sb) in the environment has attracted increasing attention. Microorganisms have been reported as important driving forces for Sb transformation. Iron (Fe) is the most important metal associated element of Sb, however, how Fe-bearing minerals affect the biological transformation of Sb is still unclear. In this study, the effects of Fe-bearing minerals on biological Sb(V) reduction were investigated by employing a marine Shewanella sp. CNZ-1 (CNZ-1). Our results showed that the presence of hematite, magnetite and ferrihydrite (1 g/L) resulted in a decrease in Sb(III) concentration of ~19–31 % compared to the Fe(III)-minerals free system. The calculated Sb(V) reduction rates are 0.0256 (R 2 0.71), 0.0389 (R 2 0.87), 0.0299 (R 2 0.96) and 0.0428 (R 2 0.95) h −1 in the hematite-, magnetite-, ferrihydrite-supplemented and Fe(III)-minerals free systems, respectively. The cube-shaped Sb 2 O 3 was characterized as a reductive product by using XRD, XPS, FTIR, TG and SEM approaches. Differential proteomic analysis showed that flagellar protein, cytochrome c , electron transfer flavoprotein, nitrate reductase and polysulfide reductase (up-regulation >1.5-fold, p value <0.05) were supposed to be included in the electron transport pathway of Sb(V) reduction by strain CNZ-1, and the key role of nitrate reductases was further highlighted during this reaction process based on the RT–qPCR and confirmatory experiments. Overall, these findings are beneficial to understand the environmental fate of Sb in the presence of Fe-bearing minerals and provide guidance in developing the bacteria/enzyme-mediated control strategy for Sb pollution.

Characterization of a Novel Myrosinase with High Activity from Marine Bacterium Shewanella baltica Myr-37

Qinwen Ye,Yaowei Fang,Mengjiao Li,Haoyu Mi,Shu Liu,Guang Yang,Jing Lu,Yaling Zhao,Qitong Liu,Wei Zhang,Xiaoyue Hou

Myrosinase can hydrolyze glucosinolates to generate isothiocyanates, which have cancer prevention and anti-cancer properties. The main sources of myrosinase are cruciferous plants. To further improve the efficiency of isothiocyanates preparation, it is necessary to explore novel sources of myrosinases. In this study, we described a bacterium,Shewanella balticaMyr-37, isolated from marine mud, capable of producing a novel myrosinase (Smyr37) with a molecular weight of 100 kDa. The crude enzyme of Smyr37 showed the highest activity at 50 °C and pH 8.0. The sinigrin- and glucoraphanin-hydrolyzing activities of Smyr37 were 6.95 and 5.87 U/mg, respectively. Moreover, when the reaction temperature was 40 °C and pH was 7.0, the crude enzyme of Smyr37 could efficiently degrade glucoraphanin into sulforaphane within 25 min with a yield of 0.57 mg/mL. The corresponding conversion efficiency of sulforaphane from glucoraphanin was 89%. In summary,S. balticaMyr-37 myrosinase Smyr37, a novel myrosinase, can be used in the preparation of isothiocyanates. Keywords:microbial enzyme;myrosinase;sinigrin;glucoraphanin;sulforaphane

Exogenous Electron Shuttle-Mediated Extracellular Electron Transfer of Shewanella putrefaciens 200: Electrochemical Parameters and Thermodynamics

Yundang Wu,Tongxu Liu,Xiaomin Li,Fangbai Li

Despite the importance of exogenous electron shuttles (ESs) in extracellular electron transfer (EET), a lack of understanding of the key properties of ESs is a concern given their different influences on EET processes. Here, the ES-mediated EET capacity of Shewanella putrefaciens 200 (SP200) was evaluated by examining the electricity generated in a microbial fuel cell. The results indicated that all the ESs substantially accelerated the current generation compared to only SP200. The current and polarization parameters were linearly correlated with both the standard redox potential (EES0) and the electron accepting capacity (EAC) of the ESs. A thermodynamic analysis of the electron transfer from the electron donor to the electrode suggested that the EET from c-type cytochromes (c-Cyts) to ESs is a crucial step causing the differences in EET capacities among various ESs. Based on the derived equations, both EES0 and EAC can quantitatively determine potential losses (ΔE) that reflect the potential loss of the ES-mediated EET. In situ spectral kinetic analysis of ES reduction by c-Cyts in a living SP200 suspension was first investigated with the EES, Ec-Cyt, and ΔE values being calculated. This study can provide a comprehensive understanding of the role of ESs in EET.

Promoting Shewanella Bidirectional Extracellular Electron Transfer for Bioelectrocatalysis by Electropolymerized Riboflavin Interface on Carbon Electrode

Zou Long,Wu Xian,Huang Yunhong,Ni Haiyan,Long Zhong-er

The extracellular electron transfer (EET) that connects the intracellular metabolism of electroactive microorganisms to external electron donors/acceptors, is the foundation to develop diverse microbial electrochemical technologies. For a particular microbial electrochemical device, the surface chemical property of an employed electrode material plays a crucial role in the EET process owing to the direct and intimate biotic-abiotic interaction. The functional modification of an electrode surface with redox mediators has been proposed as an effectual approach to promote EET, but the underlying mechanism remains unclear. In this work, we investigated the enhancement of electrochemically polymerized riboflavin interface on the bidirectional EET of Shewanella putrefaciens CN32 for boosting bioelectrocatalytic ability. An optimal polyriboflavin functionalized carbon cloth electrode achieved about 4.3-fold output power density (∼707 mW/m2) in microbial fuel cells and 3.7-fold cathodic current density (∼0.78 A/m2) for fumarate reduction in three-electrode cells compared to the control, showing great increases in both outward and inward EET rates. Likewise, the improvement was observed for polyriboflavin-functionalized graphene electrodes. Through comparison between wild-type strain and outer-membrane cytochrome (MtrC/UndA) mutant, the significant improvements were suggested to be attributed to the fast interfacial electron exchange between the polyriboflavin interface with flexible electrochemical activity and good biocompatibility and the outer-membrane cytochromes of the Shewanella strain. This work not only provides an effective approach to boost microbial electrocatalysis for energy conversion, but also offers a new demonstration of broadening the applications of riboflavin-functionalized interface since the widespread contribution of riboflavin in various microbial EET pathways together with the facile electropolymerization approach.

Impact of Condition Variations on Bioelectrochemical System Performance: An Experimental Investigation of Sulfamethoxazole Degradation

Qun Xue,Zhihui Chen,Wenjing Xie,Shuke Zhang,Jie Jiang,Guoxin Sun

Bioelectrochemical systems (BESs) are an innovative technology for the efficient degradation of antibiotics.Shewanella oneidensis(S. oneidensis) MR-1 plays a pivotal role in degrading sulfamethoxazole (SMX) in BESs. Our study investigated the effect of BES conditions on SMX degradation, focusing on microbial activity. The results revealed that BESs operating with a 0.05 M electrolyte concentration and 2 mA/cm2current density outperformed electrolysis cells (ECs). Additionally, higher electrolyte concentrations and elevated current density reduced SMX degradation efficiency. The presence of nutrients had minimal effect on the growth ofS. oneidensisMR-1 in BESs; it indicates thatS. oneidensisMR-1 can degrade SMX without nutrients in a short period of time. We also highlighted the significance of mass transfer between the cathode and anode. Limiting mass transfer at a 10 cm electrode distance enhancedS. oneidensisMR-1 activity and BES performance. In summary, this study reveals the complex interaction of factors affecting the efficiency of BES degradation of antibiotics and provides support for environmental pollution control.

Enhancing Extracellular Electron Transfer of a 3D-Printed Shewanella Bioanode with Riboflavin-Modified Carbon Black Bioink

Jiawei Yang,Pengcheng Xu,Haoming Li,Haichun Gao,Shaoan Cheng,Chaofeng Shen

3D printing of a living bioanode holds the potential for the rapid and efficient production of bioelectrochemistry systems. However, the ink (such as sodium alginate, SA) that formed the matrix of the 3D-printed bioanode may hinder extracellular electron transfer (EET) between the microorganism and conductive materials. Here, we proposed a biomimetic design of a 3D-printed Shewanella bioanode, wherein riboflavin (RF) was modified on carbon black (CB) to serve as a redox substance for microbial EET. By introducing the medicated EET pathways, the 3D-printed bioanode obtained a maximum power density of 252 ± 12 mW/m2, which was 1.7 and 60.5 times higher than those of SA-CB (92 ± 10 mW/m2) and a bare carbon cloth anode (3.8 ± 0.4 mW/m2). Adding RF reduced the charge-transfer resistance of a 3D-printed bioanode by 75% (189.5 ± 18.7 vs 47.3 ± 7.8 Ω), indicating a significant acceleration in the EET efficiency within the bioanode. This work provided a fundamental and instrumental concept for constructing a 3D-printed bioanode.

Programming interfacial electron transfer at Shewanella oneidensis – Magnetic biochar hybrids for selective tetracycline–ciprofloxacin co-removal

Lei Wang,Yudong Li,Dongle Cheng,Jian Zhang,Huu Hao Ngo,Wenshan Guo,Jie Li,Huaqing Liu,Yanlong Wang,Aiyun Guo

Antibiotic residues persist in waters and require effective, low-chemical remediation strategies. Here we create a contacted system that brings Shewanella oneidensis (MR-1) into controlled association with nitrogen doped magnetic biochar to promote interfacial electron transfer. In a mixed tetracycline-ciprofloxacin matrix, the contacted system was superior to the single biochar or microbial system. Meanwhile, the MR-1 activity was maintained within the range of 3–20 mg/L and pH 5–8, with the best performance near pH 5-6. Electrochemical and spectroscopic signatures are consistent with interfacial electron transfer from MR-1 to surface Fe centers, accompanied by Fe 3 ⁺/Fe 2 ⁺ redox cycling and coupling of adsorption with reduction. Dissolved Fe 2+ after reaction followed similar trends and increased with carrier loading, and excitation-emission matrices showed humic-like extracellular polymers that may assist electron shuttling. Electronic-structure analysis is consistent with the observed differences between tetracycline and ciprofloxacin, with tetracycline more readily accepting the first electron at chelating diketone sites, whereas the fluorinated moiety and associated bond stability may constrain ciprofloxacin conversion. Overall, the results suggest an interface-controlled route to antibiotic co-removal under mild conditions and provide design insights linking pollutant electronics, Fe site density, and operating pH to interfacial electron flow.

Methylene blue enhances the anaerobic decolorization and detoxication of azo dye by Shewanella onediensis MR-1

Weijie Liu,Liang Liu,Cong Liu,Yan Hao,Hao Yang,Bo Yuan,Jihong Jiang

Azo dyes are recalcitrant pollutants that constitute a significant burden to the environment. This study focused on decoloring azo dye Congo red using Shewanella oneidensis MR-1 and exploring methylene blue as electron shuttle to enhance its decolorization and detoxication efficiency. The effects of physicochemical parameters on anaerobic decolorization were investigated. The results showed that the suitable pH, temperature and NaCl concentration range for decolorization were 6.0–8.5, 28–40   °C and 0–20   g/L, respectively. Under the optimal conditions, 99.25% decolorization of up to 2   g/L Congo red was achieved within 24   h. Nitrite, nitrate and oxygen could significantly inhibit the decolorization. Riboflavin and methylene blue were found to be able to significantly accelerate decolorization capacity of S. oneidensis MR-1, however, block of Mtr respiratory pathway depressed the promotion effects of riboflavin and methylene blue, suggesting that the promotion effect was achieved by accelerating electron transfer between cells and dyes. Furthermore, the phytotoxicity analysis revealed that the toxicity of Congo red to Triticum aestivum , Pogostemon cablin and Isatis indigotica Fort was decreased during the decolorization process.

Insights into palladium nanoparticles produced by Shewanella oneidensis MR-1: Roles of NADH dehydrogenases and hydrogenases

Zhen-Ni Yang,Ya-Nan Hou,Bo Zhang,Hao-Yi Cheng,Yang-Chun Yong,Wen-Zong Liu,Jing-Long Han,Shuang-Jiang Liu,Ai-Jie Wang

Biologically synthesized palladium nanoparticles (bio-Pd) have attracted considerable interest as promising green catalysts for environmental remediation. However, the mechanisms by which microorganisms produce bio-Pd remain unclear. In the present study, we investigated the roles of Shewanella oneidensis MR-1 and its NADH dehydrogenases and hydrogenases (HydA and HyaB) in bio-Pd production using formate as the electron donor. The roles of NADH dehydrogenases and hydrogenases were studied by inhibiting NADH dehydrogenases and using hydrogenase mutants (Δ hydA , Δ hyaB , and Δ hydA Δ hyaB ), respectively. The results showed ~97% reduction of palladium by S. oneidensis MR-1 after 24 h using 250 μM palladium and 500 μM formate. Electron microscopy images showed the presence of bio-Pd on both the outer and cytoplasmic membranes of S. oneidensis MR-1. However, the inhibition of NADH dehydrogenases in S. oneidensis MR-1 resulted in only ~61% reduction of palladium after 24 h, and bio-Pd were not found on the outer membrane. The mutants lacking one or two hydrogenases removed 91–96% of palladium ions after 24 h and showed more cytoplasmic bio-Pd but less periplasmic bio-Pd. To the best of our knowledge, this is the first study to demonstrate the role of NADH dehydrogenases of S. oneidensis MR-1 in the formation of bio-Pd on the outer membrane. It also demonstrates that the hydrogenases (especially HyaB) of S. oneidensis MR-1 contribute to the formation of bio-Pd in the periplasmic space. This study provides mechanistic insights into the production of biogenic metal nanoparticles towards their possible use in industrial and environmental applications.

External electricity-powered FeS/graphene/bacterial hybrid for Cr6+ removal from water lacking organic electron donors

Qi Zhu,Fei Feng,Zhong-er Long,Yunhong Huang,Haiyan Ni,Xueqin Fu,Longhai Dai,Long Zou

Microbial reduction is an economical and environmentally benign route to deal with high-toxic Cr 6+ widely present in aquatic environments, but faces major challenges such as cytotoxicity and insufficient organic electron donors. Herein, we propose a route for the removal of Cr 6+ from water using a ternary FeS/reduced graphene oxide/ Shewanella oneidensis MR-1 (FeS/rGO/MR-1) hybrid with external electricity supply at an electrode potential of −0.36 V. The biohybrid with excellent redox activity is in-situ biosynthesized by virtue of the versatile metabolic pathway and powerful extracellular electron transfer ability of MR-1 cells without the need of organic carbon sources. The biohybrid is directly used to remove Cr 6+ under the condition of continuous electricity input, the removal rate is negatively correlated with the applied electrode potential. Compared with binary FeS/MR-1 and rGO/MR-1 counterpart, the FeS/rGO/MR-1 hybrid achieves an improved ability to remove Cr 6+ at an initial concentration of 20 mg L −1 in three consecutive batches due to the synergistic effect. The FeS is the main active component for rapid chemical reduction of Cr 6+ because of its highly reducible Fe 2+ and S 2- , and rGO acts as the bridge to accelerate electron transfer from the applied electrode to MR-1 cells. Then, the MR-1 cells use the obtained electrons for FeS regeneration, which in turn reduces Cr 6+ again, resulting in the precipitation of less toxic Cr 3+ [Cr(OH) 3 and Cr 2 O 3 ] in the biohybrid. This study provides a feasible route for removal of toxic Cr 6+ from water lacking organic electron donor by in-situ synthesis of biohybrid with external electricity supply.

Modeling bacterial interactions uncovers the importance of outliers in the coastal lignin-degrading consortium

Peng Qiannan,Zhao Cheng,Wang Xiaopeng,Cheng Kelin,Wang Congcong,Xu Xihui,Lin Lu

Lignin, as the abundant carbon polymer, is essential for carbon cycle and biorefinery. Microorganisms interact to form communities for lignin biodegradation, yet it is a challenge to understand such complex interactions. Here, we develop a coastal lignin-degrading bacterial consortium (LD), through “top-down” enrichment. Sequencing and physiological analyses reveal that LD is dominated by the lignin degrader Pluralibacter gergoviae (>98%), with additional rare non-degraders. Interestingly, LD, cultured in lignin-MB medium, significantly enhances cell growth and lignin degradation as compared to P. gergoviae alone, implying a role of additional outliers. Using genome-scale metabolic models, metabolic profiling and culture experiments, modeling of inter-species interactions between P. gergoviae , Vibrio alginolyticus , Aeromonas hydrophila and Shewanella putrefaciens , unravels cross-feeding of amino acids, organic acids and alcohols between the degrader and non-degraders. Furthermore, the sub-population ratio is essential to enforce the synergy. Our study highlights the unrecognized role of outliers in lignin degradation.
