The severity of foodborne diseases caused by foods contaminated by pathogens or their toxins creates an urgent need for the development of specific and sensitive method for detection of bacteria. In this study, taking advantages of CRISPR-Cas13a system, namely, the crRNA programmability and Cas13a “collateral effect” of promiscuous RNase activity upon target RNA recognition, we developed a bacterial sensing strategy with the name of CCB-Detection ( C RISPR- C as13a based B acterial Detection ). Staphylococcus aureus ( S. aureus ) was chosen as a model bacteria for validating the performance of CCB-Detection. Specifically, four steps were carried out: 1) simple extraction of genome DNA ; 2) specific gene amplification by PCR; 3) in vitro transcription; and 4) the “collateral effect” cleavage of reporter RNA to report the analyte signal. It was observed that CCB-Detection was capable to successfully detect the target genomic DNA (gDNA) as low as 10 0 aM. The limit of detection (LOD) was 1 CFU/mL with a dynamic detection range of S. aureus from 10 0 to 10 7 CFU/mL. The entire sample-to-answer time for this biosensor was less than 4 h. CCB-Detection demonstrated satisfactory selectivity for S. aureus without interference from other bacteria. Furthermore, CCB-Detection was successfully applied for sensing S. aureus in real food samples with both known and unknown amounts bacteria (spiked ones and non-spiked ones) and its performance is comparable to the conventional culture-based counting method but with short assay time and high sensitivity. With desirable reliability, sensitivity, specificity and simplicity, herein proposed CCB-Detection could be extended and generalised for other bacterial detection, and has great potential to be used in a wide range of applications such as food safety inspection, disease diagnosis, environment monitoring, etc .

Nitric oxide (NO) is an emerging and promising antimicrobial agent for treating biofilms, yet its enrichment in biofilms has become a major challenge. In this contribution, iron oxide-polydopamine (Fe3O4-PDA) core–shell nanoparticles were fabricated and then loaded with (N,N′-di-sec-butyl-N,N′-dinitroso-1,4-phenylenediamine, BNN6), a NO donor, to yield a magnetic field-driven and near-infrared (NIR)-responsive NO delivery system for biofilm treatment. In a magnetic field, the nanoparticles were found to penetrate into and accumulate in the biofilms more effectively, resulting in a higher concentration of the nanoparticles in the biofilms, which upon exposure to NIR laser (808 nm) irradiation, heat up the local environment by the photothermal effect to trigger the release of more NO in a controlled manner. The combination of photothermal and NO gas therapies exhibited a synergistic antibacterial effect against Staphylococcus aureus biofilms in vitro, and the nanoparticles showed excellent antibacterial properties in vivo in the treatment of the mice wound infection model and accelerated the wound healing process. Overall, the Fe3O4-PDA–BNN6 nanoparticles realized spatial- and temporal-controlled release of NO in biofilms using NIR under a magnetic field, providing an effective antibiofilm strategy and taking the NO gas therapy one step closer to clinical application.

Achieving accurate detection and early sterilization of Staphylococcus aureus ( S. aureus ) is one of the most important methods to prevent further contamination and spread of pathogenic bacteria. In this study, an aptasensor based on PDA/AuPt nanocomposites was prepared for specific binding, colorimetric detection, and photothermal inactivation of S. aureus . The aptamer (Apt) was immobilized on PDA/AuPt by gold-sulfur bonding (Au-S). Utilized the excellent peroxidase mimetic activity of PDA/AuPt as a colorimetric signal probe. The detection range in colorimetric mode was 1.4 × 10 2 -1.4 × 10 7 CFU/mL and the detection limit (LOD) was 44 CFU/mL. The comprehensive inspection procedure can be fully completed within a 45-min timeframe. Meanwhile, the aptasensor was successfully applied to the detection of S. aureus in real samples with the recoveries of 99.4%–110.8%. In addition, PDA/AuPt had a photothermal conversion efficiency of 22.2%, which made it an excellent photothermal agent. After incubation of 150 μg/mL apt-PDA/AuPt with S . aureus , the solution temperature rapidly increased to 62.9 °C after 5 min of near-infrared irradiation. The bacteria were killed by high temperature, and the inactivation rate was 95%.The results confirmed the application of PDA/AuPt for detecting and killing bacteria, contributing to the establishment of a food safety control system and protecting human health.

The increasing prevalence of bacterial multidrug antibiotic resistance has led to a serious threat to public health, emphasizing the urgent need for alternative antibacterial therapeutics. Lytic phages, a class of viruses that selectively infect and kill bacteria, offer promising potential as alternatives to antibiotics. However, injectable carriers with a desired release profile remain to be developed to deliver them to infection sites. To address this challenge, phage-loaded microparticles (Phage-MPs) have been developed to deliver phages to the infection site and release phages for an optimal therapeutic effect. The Phage-MPs are synthesized by allowing phages to be electrostatically attached onto the porous polyethylenimine-modified silk fibroin microparticles (SF-MPs). The high specific surface area of SF-MPs allows them to efficiently load phages, reaching about 1.25 × 1010 pfu per mg of microparticles. The Phage-MPs could release phages in a controlled manner to achieve potent antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA). Unlike the diffuse biodistribution of free phages post-intraperitoneal injection, Phage-MPs could continuously release phages to effectively boost the local phage concentration at the bacterial infection site after they are intraperitoneally injected into an abdominal MRSA-infected mouse model. In a mouse abdominal MRSA infection model, Phage-MPs significantly reduce the bacterial load in major organs, achieving an efficient therapeutic effect. Furthermore, Phage-MPs demonstrate outstanding biocompatibility both in vitro and in vivo. Overall, our research lays the foundation for a new generation of phage-based therapies to combat antibiotic-resistant bacterial infections.

As a common opportunistic pathogen, Staphylococcus aureus ( S. aureus ) can rapidly adapt to the host immune system and cause chronic infections. Currently, such chronic infections are extremely difficult to eliminate, severely impairing the function of tissues and organs. We observed localized tissue senescence in the mammary glands of mice with chronic S. aureus infection; however, the mechanism driving this senescence remains unclear. To address this, we employed a mouse model of chronic S. aureus -induced mastitis and an in vitro model of mouse mammary epithelial cells (mMECs) to confirm the inductive effect of S. aureus on tissue and cellular senescence. Through integrated analysis of the transcriptome and metabolome of mouse mammary tissues, it was found that the ornithine cycle was significantly disordered following S. aureus infection, wherein Arginase 1 (Arg1) serves as a key metabolic regulator. Notably, stimulation by S. aureus induces mammary epithelial cells to produce cytokines, thereby promoting the massive release of Arg1 by macrophages into the microenvironment, which constitutes one of the sources of abnormally elevated Arg1 in mammary tissue. This extracellular Arg1 is subsequently taken up by mammary epithelial cells, further accelerating the intracellular conversion of arginine to ornithine. The accumulation of ornithine and its downstream metabolites drives the occurrence of senescence in epithelial cells. Importantly, supplementation with excess arginine or ornithine can mimic this senescence phenotype, while knocking down Arg1 in epithelial cells can reverse this phenomenon. In summary, this study reveals a novel immunometabolic cross-regulation mechanism, specifically that Arg1 released by macrophages—triggered by mammary epithelial cells under S. aureus stimulation—acts as a paracrine senescence-inducing factor that acts back on mammary epithelial cells. The Arg1—ornithine axis holds promise as a potential therapeutic target for chronic infection-associated tissue senescence.
This study found that S. aureus stimulation induces senescence in mouse mammary glands and mMECs. In mMECs, S. aureus directly induces senescence-associated changes, including activation of p53/p21, DNA damage, and SASP-related inflammatory responses, despite reduced endogenous Arg1 expression. Meanwhile, the compensatory upregulation of Arg2 and other ornithine-cycle enzymes suggests that arginine/ornithine metabolism is remodeled in epithelial cells. In the tissue microenvironment, epithelial-derived inflammatory mediators further stimulate macrophages to express and release Arg1. This macrophage-derived Arg1 can be taken up by mMECs and may amplify epithelial senescence by promoting arginine-to-ornithine metabolic dysregulation. Therefore, macrophage-derived Arg1 promotes epithelial senescence but is not the sole initiator of this process.

Herein, we report a multifunctional DNA hydrogel (TBDH) platform that integrates the specific detection and efficient inactivation of S. aureus within a single programmable architecture. The platform consists of three core components: an aptamer (Apt) for target recognition, two DNA signaling strands (SA and SB) for fluorescence transduction, and self-assembled antibacterial nanoparticles (TBNPs). Through rational structural integration, these elements are incorporated into a tunable polymeric network, enabling a unified “recognition–reporting–inactivation” function. In the presence of S. aureus, Apt binding triggers hydrogel dissociation, restoring the quenched Cy3 fluorescence and generating a dose-dependent signal with a detection limit of 11 CFU mL−1. Simultaneously, hydrogel collapse induces on-demand release of TBNPs, which exhibit potent antibacterial and anti-biofilm efficacy (>99.9% inactivation) by disrupting bacterial membrane integrity. The TBDH platform demonstrates high specificity, rapid response (90 min), and reliable performance in complex matrices. This work provides an integrated, stimuli-responsive strategy for simultaneous pathogen detection and eradication, offering promising potential for applications in food safety and clinical infection control.

A FMIP-based paper sensor was constructed for rapid detection of S. aureus. • The paper sensor can specifically recognize the target bacteria in 5 min. • The paper sensor can accurately measure S. aureus in real samples. A fluorescent molecularly imprinted polymer (FMIP)-based paper sensor was constructed for rapid detection of Staphylococcus aureus ( S. aureus ). The FMIP nanoparticles were prepared by the boronate affinity imprinting strategy based on the interactions between the cell-surface biomolecules and the functional monomer with phenylboronic acid groups. Then, such FMIP nanoparticles were modified with amino groups for subsequent covalently grafting onto the aldehyde-functionalized paper surface, enabling its specific recognition capability to S. aureus . The paper sensor can specifically recognize the target bacteria in 5 min with an imprinting factor as high as 14.0. Such paper sensor can quantitatively detect S. aureus in the range of 10 2 -10 6 CFU/mL with a detection limit of 77 CFU/mL, and gave spiked recoveries of 87%-116% in real samples. Combining the operational simplicity with rapid detection, this FMIP-based paper sensor can be coupled with a portable fluorimeter to make a point-of-care device for on-site analysis of S. aureus . FRET-based aptasensor with HCR amplification for S. aureus detection. • Integrated magnetic beads and HCR for cascaded signal amplification. • Achieved detection limit of 53 CFU/mL. • Obtained recoveries of 94–110% and RSDs of 1.07–3.89% in spiked milk. • Showed excellent specificity against non-target bacteria up to 10 6 CFU/mL. Staphylococcus aureus ( S. aureus ) is recognized as a high-risk pathogen responsible for severe food safety incidents. To address this concern, this study developed a fluorescent aptasensor based on fluorescence resonance energy transfer (FRET) for the rapid detection of S. aureus in milk. The proposed strategy employed high-specificity aptamers as recognition elements. Target bacteria were specifically captured, enriched, and isolated using aptamer-functionalized magnetic beads (MBs), thereby converting the concentration of S. aureus into a measurable fluorescence signal. To further enhance analytical sensitivity, a hybridization chain reaction (HCR)-based nucleic acid amplification strategy was integrated to achieve cascaded signal amplification. Under optimal conditions, the developed method achieved a limit of detection as low as 5.3 × 10 1 CFU/mL for S. aureus . This work provides a valuable reference for the development of biosensors with significant potential for diverse target detection in food safety and environmental monitoring.

IntroductionMethicillin resistant Staphylococcus aureus, MRSA, is a major cause of hospital acquired infections and poses a serious therapeutic challenge because of multidrug resistance and potent virulence. Targeting virulence rather than bacterial growth may provide an alternative strategy to combat MRSA infection. This study investigated polydatin, a stilbenoid glucoside from Polygonum cuspidatum, as a potential antivirulence agent targeting caseinolytic protease P, ClpP.MethodsClpP inhibitory activity was evaluated by enzymatic assay. The effects of polydatin on bacterial growth, hemolytic activity, virulence gene expression, adhesion to fibrinogen, and host cell invasion were assessed in vitro. Target engagement was examined by thermal shift assay, fluorescence quenching, and computational simulation. Therapeutic efficacy was evaluated in a murine pneumonia model.ResultsPolydatin showed limited antibacterial activity but significantly inhibited ClpP and reduced the expression of key virulence factors, including Hla, PVL, and RNAIII. It also impaired bacterial adhesion to fibrinogen and invasion of host cells. Binding studies supported the interaction between polydatin and ClpP. In vivo, polydatin markedly alleviated S. aureus induced pneumonia, as shown by reduced lung bacterial burden, lower inflammatory cytokine levels, and attenuated tissue injury.DiscussionPolydatin attenuates MRSA pathogenicity by targeting ClpP associated virulence regulation rather than bacterial viability. These findings identify polydatin as a promising antivirulence candidate and provide a basis for developing alternative therapeutic strategies against MRSA infections.

Background:Staphylococcus aureusis a common pathogenic microorganism in humans and animals. Type II NADH oxidoreductase (NDH-2) is the only NADH:quinone oxidoreductase present in this organism and represents a promising target for the development of anti-staphylococcal drugs. Recently, myricetin, a natural flavonoid from vegetables and fruits, was found to be a potential inhibitor of NDH-2 ofS. aureus. The objective of this study was to evaluate the inhibitory properties of myricetin against NDH-2 and its impact on the growth and expression of virulence factors inS. aureus. Results: A screening method was established to identify effective inhibitors of NDH-2, based on heterologously expressedS. aureusNDH-2. Myricetin was found to be an effective inhibitor of NDH-2 with a half maximal inhibitory concentration (IC50) of 2 μM. In silico predictions and enzyme inhibition kinetics further characterized myricetin as a competitive inhibitor of NDH-2 with respect to the substrate menadione (MK). The minimum inhibitory concentrations (MICs) of myricetin againstS. aureusstrains ranged from 64 to 128 μg/mL. Time–kill assays showed that myricetin was a bactericidal agent againstS. aureus. In line with being a competitive inhibitor of the NDH-2 substrate MK, the anti-staphylococcal activity of myricetin was antagonized by MK-4. In addition, myricetin was found to inhibit the gene expression of enterotoxin SeA and reduce the hemolytic activity induced byS. aureusculture on rabbit erythrocytes in a dose-dependent manner. Conclusions: Myricetin was newly discovered to be a competitive inhibitor ofS. aureusNDH-2 in relation to the substrate MK. This discovery offers a fresh perspective on the anti-staphylococcal activity of myricetin.

Biofilms of the foodborne pathogen Staphylococcus aureus show improved resistance to antibiotics and are difficult to eliminate. To enhance antibacteria and biofilm dispersion via extracellular matrix diffusion, a new lipid nanoparticle was prepared, which employed a mixture of phospholipids and a 0.8% surfactin shell. In the lipid nanoparticle, 31.56 μg mL −1 of erythromycin was encapsulated. The lipid nanoparticle size was approximately 52 nm and the zeta-potential was -67 mV, which was measured using a Marvin laser particle size analyzer. In addition, lipid nanoparticles significantly dispersed the biofilms of S. aureus W1, CICC22942, and CICC 10788 on the surface of stainless steel, reducing the total viable count of bacteria in the biofilms by 10 3 CFU mL −1 . In addition, the lipid nanoparticle can remove polysaccharides and protein components from the biofilm matrix. The results of laser confocal microscopy showed that the lipid nanoparticles effectively killed residual bacteria in the biofilms. Thus, to thoroughly eliminate biofilms on material surfaces in food factories to avoid repeated contamination, drug-lipid nanoparticles present a suitable method to achieve this.

The antimicrobial mechanism of naringenin, one of the citrus antibacterial flavonoids against food-borne Staphylococcus aureus ATCC 6538, was investigated in this study. Analysis of gas chromatography-mass spectrometry (GC-MS) and fluorescence showed that relatively low concentrations of naringenin caused perturbations in the membrane fatty acid composition and the conformation of membrane proteins through changing the microenvironment of phenylalanine, tyrosine, and tryptophan residues. Exposure of naringenin at higher levels significantly increased membrane permeability and changed the morphology of S. aureus cells. The genomic DNA-binding of naringenin was also quantitatively monitored using UV-vis spectra in combination with multivariate curve resolution-alternating least squares (MCR-ALS) analysis, and the concentration and pure spectra profiles for the three reaction species (DNA, naringenin, and DNA–naringenin) were obtained. Moreover, the thermal behavior of DNA and docking studies revealed that naringenin preferentially bound to the A–T base pair regions of genomic DNA via groove binding, and atomic force microscopy and circular dichroism showed that naringenin induced mild secondary structure and obvious morphological variations of this biomacromolecule. These results suggested that naringenin exerting its antibacterial effects might be connected with disruption of the cytoplasmic membrane and DNA targeting effects in Staphylococcus aureus.
