Developing infrared nonlinear optical (NLO) crystals with practical application potential remains a great challenge. Here, by harnessing a hybrid supertetrahedral assembly strategy, we discovered a new infrared nonlinear optical material, Ba5Zn3Ga2Sn2Se15, which crystallizes in the orthorhombic space group Ama2. Its structure is based on the assembly of hybrid T2-type supertetrahedral clusters, forming a three-dimensional anionic framework built from one-dimensional hybrid [SnZn3Se9] supertetrahedral chains and two-dimensional [SnGa2Se8] anionic layers. This multicationic framework gives rise to the following properties: a phase-matchable SHG response of 1.3 × AgGaS2, an experimental band gap of 2.27 eV, and a birefringence of 0.1055 at 1064 nm, which is sufficient for phase matching. Theoretical calculations indicate that the SHG response originates from distorted hybrid supertetrahedral units and asymmetric charge transfer along the M–Se bonds (M = Zn, Ga, Sn). The compound also exhibits congruent melting behavior, a prerequisite for growing large single crystals. This work suggests that hybrid supertetrahedral architectures are a viable platform for mid-infrared NLO materials.

To realize the accurate diagnosis of tumors by magnetic resonance imaging (MRI), switchable magnetic resonance contrast agents (CAs) between T1 and T2 contrast enhancement that are constructed based on extremely small iron oxide nanoparticles (ESIONPs) have been developed in recent years. We herein report, for the first time, a novel ESIONP-based nanocluster (named EAmP), which exhibited hypoxia responsiveness to the tumor microenvironment and offered a T2-to-T1-switchable contrast enhancement function, effectively distinguishing between the normal tissue and tumor tissue. In detail, active perfluorophenyl ester-modified ESIONPs with a diameter of approximately 3.6 nm were initially synthesized, and then 4,4′-azodianiline was used as a cross-linker to facilitate the formation of nanoclusters from ESIONPs through the reaction between the active ester and amine. Finally, poly(ethylene glycol) was further modified onto nanoclusters by utilizing the remaining active ester residues. The resulting EAmP demonstrated satisfactory colloidal stability and favorable biosafety and exhibited a desired T2-to-T1-switchable function, as evidenced by conversion from nanocluster to the dispersed state and a significant decrease in the r2/r1 ratio from 14.86 to 1.61 when exposed to a mimical hypoxic environment in the solution. Moreover, EAmP could decompose into dispersed ESIONPs at the tumor region, resulting in a switch from T2 to T1 contrast enhancement. This T2-to-T1-switchable contrast agent offers high sensitivity and signal-to-noise ratio to realize the accurate diagnosis of tumors. In conclusion, hypoxia-responsive EAmP is a potential MRI nanoprobe for improving the diagnostic accuracy of solid tumors.

Single-modal magnetic resonance imaging (MRI) contrast agents sometimes cause signal confusion in clinical diagnosis. Utilizing ligands to endow iron oxide nanoparticles (IO NPs) with excellent dual-modal MRI contrast efficiency might be an effective strategy to improve diagnostic accuracy. This work presents the development of a special ligand-assisted one-pot approach for the preparation of super-hydrophilic magnetic NPs with excellent water dispersion, biocompatibility and T1–T2 dual-modal contrast enhancement properties. In addition, the strong binding capacity between the ethylenediamine tetramethylenephosphonic acid (EDTMP) ligand and water molecules induced by the presence of abundant hydrogen bonds significantly improves spin–lattice (T1) and spin–spin (T2) imaging of the IO core. After being modified with the EDTMP ligand, the T2 relaxation rate of the IO core is dramatically increased from 71.78 mM−1 s−1 to 452.38 mM−1 s−1, and a moderate T1 relaxation rate (11.61 mM−1 s−1) is observed simultaneously, implying that the NPs with an average size of 9.7 nm may be potential candidates as high-efficiency T1–T2 MRI contrast agents. This fundamental technique of using super-hydrophilicity ligands to endow IO NPs with dual-modal contrast properties without size change and damage in the T2 contrast effect may provide a useful strategy to facilitate the application of magnetic NPs in the field of medical diagnosis.

Accurate diagnosis of diseases located in deep tissues is always challenging. The “always-on” probe often leads to false-positive signals due to nonspecific interaction of nanoprobes. Thus, stimuli-responsive nanoprobes are highly desirable, which, however, require complicated surface modification so as to achieve trigger-induced signal changes. Here pH-triggered switchable magnetic resonance imaging (MRI) nanoprobes were constructed by coordination-driven self-assembly of monodispersed iron oxide nanoparticles (MIONPs) with simple amino acid derivatives, which displayed typical T2-weighted MRI features, yet, were turned into T1-weighted MRI under slightly acidic conditions at the tumor site. The dynamic assembly and disassembly properties of MIONPs afford T2/T1 switchable contrast imaging, enabling selective “turn-on” signals at the tumor site with high specificity.

Single-mode magnetic resonance imaging (MRI) contrast agents (CAs) in clinical settings are easily disturbed by calcification, bleeding, and adipose signals, which result in inaccurate diagnoses. In this study, we developed a highly efficient T1–T2 dual-mode MRI CA using an ultra-small gadolinium oxide-decorated magnetic iron oxide nanocrystal (GMIO). The gadolinium element could effectively alter the magnetic properties of the GMIO from soft-ferromagnetism to superparamagnetism. In addition, when the Gd/Fe ratio was 15% (designated as GMIO-2), the GMIO-2 possessed the best superparamagnetism and highest magnetism. Subsequently, T1 and T2 values of GMIO-2 were measured through a series of turbo spin-echo images and then multi-spin echo sequence, respectively. Based on this, T1 and T2 relaxivities of GMIO-2 were calculated and were the highest (r1: 1.306 m M−1 s−1 and r2: 234.5 m M−1 s−1) when compared to other groups. The cytotoxicity of GMIO-2 was negligible under a wide range of dosages, thus exhibiting excellent cell biocompatibility. Moreover, GMIO-2 could quickly diffuse into cells, leading to its effective accumulation. The systemic delivery of GMIO-2 resulted in an excellent T1–T2 dual-mode MRI contrast effect in kidneys, which is expected to improve the diagnosis of kidney lesions. Therefore, this work provides a promising candidate for the development of a T1–T2 dual-mode MRI CA.

The development of hollow ferrogadolinium nanonetworks has not been reported for nanomedicine application until now. In this study, we developed a hollow and porous ferrogadolinium nanonetwork structure using the one-pot solvothermal method. This nanoparticle could be simultaneously used as a T1 and T2 dual-modal magnetic resonance imaging (MRI) contrast agent. In addition, the hollow lumen and abundant pores of the nanonetworks maximized the loading capacity and conferred the nanoplatforms for suitable anticancer drug loading capacity. Using these nanonetworks, MRI and anticancer experiments were conducted in vitro and satisfactory dual-modal MRI and cancer chemotherapy results were obtained. Therefore, the nanonetworks with dual-modal MRI and drug loading abilities effectively complement the ferrogadolinium composites' library and hold great promise in nanomedicine for simultaneous cancer diagnosis and chemotherapy.

In recent years, magnetic hyperthermia therapy (MHT), recognized as a clinically promising anticancer treatment, has focused on enhancing therapeutic outcomes in tumor management, such as optimizing magnetic nanoparticle accumulation within tumor microenvironments and developing synergistic combination approaches with other therapies. Herein, we developed nanoengineered neutrophils (NE) by loading magnetic nanoparticles (FPP) onto healthy-derived NE, designated as FPPN. The combination of FPP's pronounced T 2 -weighted magnetic resonance imaging (MRI) and superior magnetothermal efficiency with the high tumor-targeting and cancer-killing activity of NE successfully achieved the synergistic implementation of MHT and NE. Under the guidance of T 2 -weighted MRI, the accumulation of FPPN within the tumor site demonstrated a significant enhancement compared with free FPP. Under exposure to alternating magnetic fields, the thermal energy produced by FPP and FPPN could ablate tumor cells directly. Intriguingly, mitochondria within NE exhibit significant reactive oxygen species (ROS) under magnetothermal activation, thereby enhancing therapeutic efficacy and enabling synergistic tumor treatment through the combination of MHT and NE-mediated ROS. This therapeutic strategy not only highlighted the significant potential of nanoengineered NE for highly efficient MHT but also established a novel therapeutic platform for precision-targeted treatment of deep-seated tumors.

Atherosclerosis (AS) is a key pathological basis causing major cardiovascular diseases. Its proinflammatory microenvironment, characterized by aggregated inflammatory macrophages, excessive reactive oxygen species, and deficient phagocytosis, plays a pivotal role in driving AS pathogenesis. However, given the complexity of the microenvironment inside atherosclerotic plaques, developing highly effective and comprehensive nanoplatforms for precise diagnosis, efficient therapy, and reliable prognostic evaluation remains a major challenge. Herein, we construct a biomimetic nanomodulator (FeMn/Cur@CM) with responsive T1-T2 dual-modal magnetic resonance imaging (MRI) and multifaceted regulation functionalities of the plaque microenvironment for visualized treatment of AS. Coated with a macrophage membrane, curcumin-loaded FeMn/Cur@CM nanoparticles achieve prolonged circulation and specific targeting of inflammatory plaques. Diagnostically, FeMn/Cur@CM functions as a microenvironment-responsive dual-modal MRI contrast agent, amplifying T1-T2 signals in plaques. Therapeutically, FeMn/Cur@CM multifacetedly modulates the plaque microenvironment by alleviating oxidative stress, neutralizing protons, promoting macrophage reprogramming, and restoring macrophage efferocytosis. As a result, the integrated theranostic platform demonstrates remarkable plaque regression and simultaneously realizes real-time visualization of lesion progression via dual-modal MRI, providing precise diagnosis of plaque milieu. Through inflammation-targeted intervention coupled with real-time imaging feedback, this biomimetic nanomodulator establishes a promising strategy for the precise treatment of inflammatory vascular diseases, offering translational potential for AS management.

Emerging nanotechnologies in cancer therapy increasingly highlight the importance of integrating therapeutic and diagnostic functions into a single platform for enhanced efficacy and real-time monitoring. Here, we present a vortex‐shaped nanoring Fe 3 O 4 @MnO 2 @PDA (polydopamine) engineered for dual‐mode T 1 – T 2 magnetic resonance imaging (MRI)‐guided, dual‐thermal, and Fenton‐like chemodynamic therapy (CDT). The ring‐shaped nano Fe 3 O 4 core provides magnetothermal hyperthermia, while the MnO 2 layer, acting as a responsive nanozyme, catalyzes endogenous H 2 O 2 to generate oxygen and bolster CDT. Meanwhile, the PDA coating efficiently absorbs near‐infrared light, amplifying photothermal effects for tumor ablation. Under dual‐heating conditions, the temperature rise rate (2.05–2.92 times) and final equilibrium temperatures (1.57–2.27 times) exceed those of single‐mode heating, underscoring a robust synergistic effect. Additionally, due to the shell’s shielding effect weakening T 2 signals and the tumor microenvironment enhancing T 1 relaxation, the T 1 (5.75 Mm −1 s −1 ) to T 2 (91.56 mM −1 s −1 ) ratio is optimized specifically at tumor sites, thus achieving effective dual‐modal MRI contrast. Moreover, the smart MnO 2 design not only furnishes dynamic imaging enhancements but also enables environment‐triggered Mn 2+ release, offering further diagnostic and therapeutic benefits. In vitro and in vivo evaluations confirm significant tumor suppression with minimal systemic toxicity. These findings position nanoring Fe 3 O 4 @MnO 2 @PDA as a promising dual‐mode MRI‐guided, dual‐thermal, and chemodynamic platform for advanced cancer therapy.

The T1−T2 dual-mode probes for magnetic resonance imaging (MRI) can non-invasively acquire comprehensive information of different tissues or generate self-complementary information of the same tissue at the same time, making MRI a more flexible imaging modality for complicated applications. In this work, three Gadolinium-diethylene-triaminepentaaceticacid (Gd-DTPA) complex conjugated superparamagnetic iron oxide (SPIO) nanoparticles with different Gd/Fe molar ratio (0.94, 1.28 and 1.67) were prepared as T1–T2 dual-mode MRI probes, named as SPIO@PEG-GdDTPA0.94, SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67, respectively. All SPIO@PEG-GdDTPA nanocomposites with 8 nm spherical SPIO nanocrystals showed good Gd3+ chelate stability. SPIO@PEG-GdDTPA0.94 nanocomposites with lowest Gd/Fe molar ratio show no cytotoxicity to Raw 264.7 cells as compared to SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67. SPIO@PEG-GdDTPA0.94 nanocomposites with r1 (8.4 mM−1s−1), r2 (83.2 mM−1s−1) and relatively ideal r2/r1 ratio (9.9) were selected for T1–T2 dual-mode MRI of blood vessels and liver tissue in vivo. Good contrast images were obtained for both cardiovascular system and liver in animal studies under a clinical 3 T scanner. Importantly, one can get high-quality contrast-enhanced blood vessel images within the first 2 h after contrast agent administration and acquire liver tissue anatomy information up to 24 h. Overall, the strategy of one shot of the dual mode MRI agent could bring numerous benefits not only for patients but also to the radiologists and clinicians, e.g. saving time, lowering side effects and collecting data of different organs sequentially.

We explored the catalytic activity and magnetic resonance imaging (MRI) capacity of Cu-doped ultrasmall iron oxides with different doping ratios. Then, we screened a highly efficient ultrasmall active catalyst (UAC). Subsequently, a biodegradable magnetic nanoliposome was developed for multimodal cancer theranostics through pH-sensitive liposome coating of these UACs. Upon entering the body, the magnetic nanoliposomes significantly prolonged the metabolic time of UACs and promoted their accumulation in tumors. Then, the strong photothermal (PT) effect of the magnetic nanoliposome quickly ablated the tumor, showing promising PT therapy. Upon entering tumor cells, the magnetic nanoliposome rapidly degraded into many UACs and released chemotherapeutic drugs, contributing to chemotherapy. In addition, UACs not only catalyzed Fenton-type reaction to produce excessive reactive oxygen species (ROS) but also inhibited the synthesis of endogenous GSH by inactivating glutamyl cysteine ligase, contributing to cancer ferroptosis. Furthermore, the assembly-dissociation process of UACs showed the function of magnetic relaxation switches, significantly enhancing tumor MRI signal change, achieving a more accurate diagnosis of the tumor. Therefore, this magnetic nanoliposome splits into many UACs upon drug release and regulates the tumor microenvironment to overproduce ROS for enhanced synergistic tumor theranostics, which provides a strategy for developing next-generation magnetic catalysts with biodegradability and multimodal antitumor theranostics.
