Polydopamine-Mediated EGCG-Modified Polystyrene Microspheres for the Synergistic removal of Inflammatory Cytokines TNF-α and Bilirubin in liver failure

Du Yunzheng,Liu Zhuang,Chai Yamin,Wang Biao,Wang Lichun,Wang Zimeng,Guo Xiaofang,Zhou Guanlun,Yang Jingxuan,Zhu Chunling,Yang Leilei,Lv Xinyao,Ou Lailiang

Hemoperfusion has emerged as a crucial treatment for liver failure in clinics. However, the challenge remains in the incapacity to effectively and concurrently eliminate multiple harmful toxins, including inflammatory cytokines and bilirubin. This study employed molecular docking for directing the design of adsorbents. To develop a multi-target adsorbent material for liver failure, capable of absorbing both bilirubin and TNF-α, a functional platform was constructed. This platform employs a TiO2-modified polystyrene-based microsphere PSVT, which was designed for bilirubin adsorption, and coated with polydopamine and grafted with EGCG for TNF-α adsorption. The results confirm that EGCG possesses a high binding affinity for TNF-α, and the introduction of EGCG as a functional moiety markedly improves the clearance effectiveness of TNF-α. Meanwhile, PSVT served as the foundation to guarantee the effective removal of bilirubin. PSVT/P/EGCG exhibits outstanding clearance performance, with a maximum adsorption capacity of 263 ng/g for TNF-α and 23.91 mg/g for bilirubin, demonstrating its high efficiency in removing both inflammatory mediators and toxins. EGCG contributes exceptional antioxidant capabilities to PSVT/P/EGCG. The adsorbent exhibits remarkable biocompatibility and stability. The multi-target adsorbent material developed in this study has an extensive spectrum of potential applications in liver failure treatment employing an artificial liver.

Phytochemical Compositions and Antioxidant and Anti-Inflammatory Activities of Crude Extracts from Ficus pandurata H. (Moraceae)

Huiqing Lv,Xiaoping Zhang,XueZhi Chen,Zhijun Xie,Changfeng Hu,Chengping Wen,Kezhi Jiang

Background. Ficus pandurata H. (Moraceae) is widely used in traditional Chinese medicine as a healthy food condiment or a medicine for treatment of various diseases including inflammation. Objective. The purpose of the present study is to investigate the phytochemical compositions and antioxidant and anti-inflammatory activities of crude water (FPW) and ethanolic extracts (FPE) from Ficus pandurata H. Methods. Phytochemical compositions were identified by a high-performance liquid chromatography-electrospray ionization-mass spectrometry method (HPLC-ESI-MS). The antioxidant activities were evaluated by diphenylpicrylhydrazyl (DPPH) and hydroxyl radical assays, and the anti-inflammatory activities were evaluated by paw edema and levels of inflammatory mediator TNF-α and PGE2 in monosodium urate (MSU) crystal-induced rats. Results. Six compounds were identified by HPLC-MS method, and abundance of phenolics was found in FPE. The FPE showed concentration-dependent-significant scavenging of DPPH and hydroxyl radicals with IC50 values 118.4 and 192.9 μg/mL, respectively. The FPE treatment significantly inhibited the paw edema and the production of TNF-α and PGE2 in MSU crystal-induced rats. Conclusion. The FPE exerted stronger antioxidant and anti-inflammatory activities which may be attributed to its high phenolic content.

PtTe2-alginate hydrogels for combined photothermal-chemodynamic cancer therapy by inducing cell apoptosis via modulation of TNF-α-related signaling pathways

Jiayao Tong,Jiaqi Qin,Jianghao Xing,Huansuo Ren,Peng Cao,Hui Wang,Pengyu Pan,Xianwen Wang,Haowei Xue

Head and neck squamous cell carcinoma ranks as the sixth most common malignant tumor worldwide, with the majority of cases manifesting as oral squamous cell carcinoma (OSCC). The current clinical standard for treating OSCC consists of surgical resection followed by radiotherapy and chemotherapy. This multimodal approach, however, is associated with substantial morbidity, significant perioperative risk, and frequent systemic toxicities that markedly impair quality of life. Therefore, novel, more effective, and safer treatment strategies are urgently needed. Here, PtTe 2 nanorods were synthesized and encapsulated in sodium alginate (ALG) to form a novel injectable hydrogel for the precise treatment of OSCC. This ALG hydrogel prolongs the intratumoral retention of PtTe 2 , ensuring sustained local availability. PtTe 2 exhibits tumor microenvironment-responsive catalytic activity, catalyzing the conversion of hydrogen peroxide into highly toxic hydroxyl radicals via Fenton-like reactions. Moreover, PtTe 2 possesses exceptional photothermal conversion efficiency, enabling precise, light-driven hyperthermia that selectively and effectively ablates tumor cells. By synergistically accelerating the kinetics of Fenton-like reactions through photothermal effects, the generation of reactive oxygen species is substantially amplified, effectively overwhelming the antioxidant defense system and achieving significant antitumor effects. Both in vitro and in vivo experiments demonstrated that the PtTe 2 - ALG hydrogel achieved robust antitumor efficacy and excellent biosafety. Transcriptomic analyses revealed significant activation of apoptosis-related signaling cascades, elucidating the molecular mechanism of PtTe 2 - based tumor therapy. By combining chemodynamic and photothermal therapy within a single, injectable platform, this work provides a promising, low-toxicity strategy for the clinical management of OSCC.

Neferine alleviated corticosterone-induced depressive-like behaviors in mice via the inhibition of TNF/NF-κB pathway by binding to PPARγ

Zhao Chen-di,Liu Nan-feng,Samad Abdus,Tian Zhen,Xu Hong-mei,Tian Jiao

Depression is a widespread neuropsychiatric disorder with the current therapeutic approaches achieving only suboptimal efficacy. Neferine is the main bioactive component of Nelumbinis plumula with multiple pharmacological functions. This study sought to investigate the antidepressant activity of neferine and elucidate its underlying mechanisms. Network pharmacology analysis was performed to uncover the molecular targets and pathways involved in neferine’s antidepressant effects. The interaction of neferine with the core target was demonstrated by molecular docking and molecular dynamics simulation. The mouse model of depression triggered by chronic corticosterone (CORT) administration was utilized to validate the impact of neferine on core targets and pathways. We found that neferine remarkably attenuated CORT-induced depressive-like behaviors in mice. A total of 178 overlapping targets were acquired through the intersection of neferine and depression-related genes. The protein–protein interaction (PPI) network analysis revealed nine pivotal hub genes, namely AKT1, TNF, ESR1, PPARG, JUN, HIF1A, CASP3, NFKB1, and MMP9. Functional enrichment analysis indicated that these targets predominantly mapped to the TNF signaling pathway in depression. Molecular docking and molecular dynamics simulation showed that neferine has strong binding stability with PPARγ, a key molecule involved in regulating the TNF pathway. The results of animal experiments found that a PPARγ antagonist abolished neferine-induced alleviation of depressive-like behaviors. Moreover, neferine suppressed TNF/NF-κB pathway activation and attenuated neuronal loss in the hippocampus, potentially through activating PPARγ. Collectively, our study suggested that neferine produced antidepressant effects by suppressing hippocampal inflammation and neuronal loss through inhibiting the TNF/NF-κB signaling pathway. These preclinical evidence laid a foundation for further exploring the role of neferine in treating depression. Graphical

Quercetin Reverses DEHP-Induced ZBP1-Mediated PANoptosis in Thymus by Targeting NF-κB to Regulate the TNF-α/ROS Signaling Pathway

Yuting Dong,Xiaodan Wang,Shize Wang,Kang Yang,Tong Xu,Hongjin Lin

DEHP induces thymus injury in chickens by activating the NF-κB/TNF-α pathway, triggering oxidative stress and PANoptosis. • QUE can alleviate the activation of the NF-κB/TNF-α pathway, oxidative stress and PANoptosis induced by DEHP in chickens. • QUE exerts a targeted effect on NF-κB, thereby mitigating DEHP-induced thymus injury in chickens. Quercetin (Que), a natural dietary flavonoid, possesses significant antioxidative and immunoregulatory properties. Di(2-ethylhexyl) phthalate (DEHP), a pervasive environmental endocrine-disrupting chemical, triggers oxidative stress (OS) and elicits immunotoxic effects in various species. However, whether Que can target NF-κB to alleviate DEHP-induced OS and PANoptosis in the chicken thymus remains unclear to date. To investigate the mechanism of thymus damage induced by DEHP and the intervention effect of Que, we established an in vivo chicken model treated with DEHP and/or Que. Based on the results of network pharmacology, molecular docking, molecular dynamics simulations, and Cell Thermal Shift Assay (CETSA), we predicted that NF-κB is the key target for the protective effect of Que. Subsequently, through H&E staining, CCK-8, immunofluorescence, Western blot and other techniques, we verified the effects of DEHP and/or Que on OS, the assembly of PANoptosome, and cellular PANoptosis in chicken thymus tissue and chicken lymphocyte cell line (MSB-1). Meanwhile, to clarify the key role of NF-κB in the above regulatory process, we conducted reverse validation experiments by introducing the NF-κB activator Act1 in cellular experiments. The results demonstrated that DEHP exposure markedly induces the upregulation of NF-κB and TNF-α(P<0.05), leading to a pronounced burst of reactive oxygen species (ROS) and consequent OS within the chicken thymus. This process further activates the ZBP1-mediated PANoptosis pathway, ultimately causing damage to chicken thymus tissue. Que intervention effectively suppresses NF-κB activation, blocking the transmission of the NF-κB/TNF-α/ROS signaling axis, thereby reversing DEHP-induced ZBP1-mediated PANoptosis in chicken thymus. Analgesic and anti-inflammatory effects of galangin: a potential pathway to inhibit transient receptor potential vanilloid 1 receptor activation

Kaiwen Lin,Datian Fu,Zhongtao Wang,Xueer Zhang,Canyang Zhu

Background Galangin, commonly employed in traditional Chinese medicine for its diverse medicinal properties, exhibits potential in treating inflammatory pain. Nevertheless, its mechanism of action remains unclear. Methods Mice were randomly divided into 4 groups for 7 days a normal control group, a galangin-treated (25 and 50 mg/kg), and a positive control celecoxib (20 mg/kg). Analgesic and anti-inflammatory effects were evaluated using a hot plate test, acetic acid-induced writhing test, acetic acid-induced vascular permeability test, formalin-induced paw licking test, and carrageenan-induced paw swelling test. The interplay between galangin, transient receptor potential vanilloid 1 (TRPV1), NF-κB, COX-2, and TNF-α proteins was evaluated via molecular docking. COX-2, PGE2, IL-1β, IL-6, and TNF-α levels in serum were measured using ELISA after capsaicin administration (200 nmol/L). TRPV1 expression in the dorsal root ganglion was analyzed by Western blot. The quantities of substance P (SP) and calcitonin gene-related peptide (CGRP) were assessed using qPCR. Results Galangin reduced hot plate-induced licking latency, acetic acid-induced contortions, carrageenan-triggered foot inflammation, and capillary permeability in mice. It exhibited favorable affinity towards TRPV1, NF-κB, COX-2, and TNF-α, resulting in decreased levels of COX-2, PGE2, IL-1β, IL-6, and TNF-α in serum following capsaicin stimulation. Galangin effectively suppressed the upregulation of TRPV1 protein and associated receptor neuropeptides CGRP and SP mRNA, while concurrently inhibiting the expression of NF-κB, TNF-α, COX-2, and PGE2 mRNA. Conclusions Galangin exerts its anti-inflammatory pain effects by inhibiting TRPV1 activation and regulating COX-2, NF-κB/TNF-α expression, providing evidence for the use of galangin in the management of inflammatory pain.

A New Discovery: Corydalis yanhusuo Causes Idiosyncratic Hepatotoxicity and Its Potential Mechanisms

Longxin Guo,Li Lin,Jun Ling,Shengkai Zhu,Xinyu Li,Minjuan Long,Yingjie Xu,Zhanjiang Hu,Ming Niu,Xu Zhao,Xiaohe Xiao

Corydalis yanhusuo W.T.Wang (YHS) is a commonly used traditional Chinese medicine, often prescribed for treating a variety of pains. In recent years, there has been a gradual increase in the number of reports to liver injury caused by YHS and its preparations, but the exact type and mechanism of hepatotoxicity are still unclear. In the present study, we demonstrated that YHS could induce idiosyncratic drug-induced liver injury (IDILI) in the inflammatory activation models. A total of 459 differential genes and 25 differential metabolites were identified by transcriptomics and metabolomics, which were significantly enriched in the TNF and NF-κB signaling pathways as well as glycerophospholipid metabolism, sphingolipid metabolism, and arachidonic acid metabolism. In addition, YHS significantly increased the levels of TNF-α, IL-1β, and IL-6. Therefore, we believe that the mechanism of toxicity may be related to the TNF and NF-κB signaling pathways, with glycerophospholipid metabolism, sphingolipid metabolism, and arachidonic acid metabolism also playing important roles. It provides a reference for the safe and rational use of YHS in clinical practice and contributes to the precise prevention and control of the risk of liver toxicity associated with YHS.

Screening of anti-inflammatory components in composite Houttuynia mixture using comprehensive two-dimensional ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry and network pharmacology

Xinyue Lin,Luxi Zhou,Lingfang Chen,Qiyue Chen,Zhuming Jin,Peixi Zhu,Ni Yang

Houttuynia cordata Thunb. is a perennial herb with a distinct “fishy” flavour, commonly used in Asian cuisine and traditional medicine. To overcome the compound identification challenges in the composite formula of its mixture, a comprehensive two-dimensional ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry (2D UHPLC-QTOF-MS) method was developed and identified 293 chemical compounds. Given the herb’s strong anti-inflammatory properties, network pharmacology was employed to predict potential targets, revealing TNF, AKT1, MMP9, and PTGS2 as key theoretical targets for its anti-inflammatory effects. Molecular docking showed strong binding energies between these targets and the top 10 compounds (8,13-dihydroxy-9,11-octadecadienoic acid, neobaicalein, chrysin, apigenin, genistein, chrysoeriol, norwogonin, cirsiliol, luteolin and deoxyandrographolide), with genistein exhibiting the highest affinity for AKT1 (−10.4 kcal/mol). This innovative approach, combining advanced 2D UHPLC-QTOF-MS with network pharmacology, offers great potential for screening active components in herbs and their complex mixtures.

Lei Zhu,Guan Zhang,Jiasai Kang,Xinru Xu,Haoshu Zhang,Yingying Pu,Yong Huang,Dewen Tong,Qian Du

Background Folium Isatidis (Da Qing Ye), a traditional Chinese medicine, exhibits broad-spectrum antiviral and immunomodulatory properties and has been widely used in treating respiratory diseases. Porcine circovirus type 2 (PCV2), a widespread swine virus, causes respiratory disease complex in piglets and leads to substantial economic losses in the global swine industry. This study aimed to evaluate the anti-PCV2 activity of Folium Isatidis . Materials and methods The anti-PCV2 activity of Folium Isatidis was first validated through both in vivo and in vitro experiments. Potential targets of Folium Isatidis were identified using TCMSP, BATMAN-TCM, and SwissTargetPrediction databases, and intersected with PCV2-related targets. Mechanism exploration involved PPI network analysis, GO annotation, and KEGG enrichment. Molecular docking analyzed component-target interactions, followed by experimental validation of the core component. Finally, the anti-PCV2 effect of the core component was verified through in vitro and in vivo experiments. Results A total of 135 overlapping targets between Folium Isatidis components and PCV2-related disease targets were identified. Key targets, including AKT1 and TNF-α, were implicated in the therapeutic effects of Folium Isatidis against PCV2. Isovitexin was identified to be the core component that directly binds to AKT1 and TNF-α, thereby inhibiting the PI3K/AKT signaling pathway and regulating TNF-α expression and activity to suppress PCV2 replication and alleviate PCV2-induced lung injury. Conclusions Our study reveals that Isovitexin, the core anti-PCV2 component of Folium Isatidis , exerts its effect by inhibiting the PI3K/AKT pathway and modulating TNF-α, thereby outlining a novel therapeutic strategy for combating PCV2 infection and associated respiratory injuries.

Flexible/Regenerative Nanosensor with Automatic Sweat Collection for Cytokine Storm Biomarker Detection

Cong Huang,Weisong Yang,Hao Wang,Suichu Huang,Shanshan Gao,Dongliang Li,Jialin Liu,Siyu Hou,Weihao Feng,Ziran Wang,Feiran Li,Zhuang Hao,Xuezeng Zhao,PingAn Hu,Yunlu Pan

The real-time monitoring of low-concentration cytokines such as TNF-α in sweat can aid clinical physicians in assessing the severity of inflammation. The challenges associated with the collection and the presence of impurities can significantly impede the detection of proteins in sweat. This issue is addressed by incorporating a nanosphere array designed for automatic sweat transportation, coupled with a reusable sensor that employs a Nafion/aptamer-modified MoS2 field-effect transistor. The nanosphere array with stepwise wettability enables automatic collection of sweat and blocks impurities from contaminating the detection zone. This device enables direct detection of TNF-α proteins in undiluted sweat, within a detection range of 10 fM to 1 nM. The use of an ultrathin, ultraflexible substrate ensures stable electrical performance, even after up to 30 extreme deformations. The findings indicate that in clinical scenarios, this device could potentially provide real-time evaluation and management of patients’ immune status via sweat testing.

Essential Oils of Cedrus deodara Leaves Exerting Anti-inflammation on TPA-induced Ear Edema by Inhibiting COX-2/TNF-α/NF-κB Activation

Yu Chen,Lejing Chi,Xiaoxin Liang,Yaohui Shi,Tongyun Wu,Mao Ye,Ping Han,Li Lin,Lanyue Zhang,Peng Xu,Zhiyun Du

Cedrus deodara is a cedar plant from the Pinaceae family. Because Cedrus is widely used in health care products, cosmetics, medicine, and insecticides, the essential oils of Cedrus deodara (CDEOs) are gaining increasing levels of attention. However, there have only been a few studies on the anti-inflammatory effects of CDEOs. This study aimed to systematically study the composition and biological effects of CDEOs, in line with this, the study assessed the chemical composition of CDEOs and evaluated their anti-inflammatory mechanisms in mice. Gas chromatography-mass spectrometry (GC-MS) was used to determine and analyze a total of 41 compounds from CDEOs. It was found that the major components of CDEOs are thujopsene (28.23%), α-cedrene (17.01%), and cedrol (15.55%); moreover, our results showed that the active ingredients in CDEOs have anti-inflammatory and antioxidant effects and that the active ingredients in CDEOs can be used as an antioxidant, as an important biological resource in functional food, and as ingredients in cosmetics and pharmaceuticals. Taken together, these CDEOs serve as ideal additives for anti-inflammatory drugs.

Rosa davurica Pall. as a Functional Food in Ameliorating Acute Myocardial Ischemia: An Integrated Multi-Omics Study on TNF-α and Oxidative Stress

Zeyu Yi,Siyu Liu,Lincheng Bai,Fan Yang,Zhenkun Wang,Xiubo Zhu,Zunpeng Shu,Hua Han,Xiaojin Yu

Rosa davurica Pall. (RDP), a rosa fruit rich in bioactive compounds, has shown potential in alleviating cardiovascular diseases, yet its chemical composition and mechanisms against acute myocardial ischemia (AMI) remain unclear. This study comprehensively characterized 99 compounds in RDP extracts using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS), including flavonoids, terpenoids, and phenolic acids. By integrating transcriptomics and network pharmacology, we identified the tumor necrosis factor-alpha (TNF-α) signaling pathway as a key target in AMI progression. In vivo experiments demonstrated that RDP significantly ameliorated isoproterenol (ISO)-induced myocardial injury by reducing oxidative stress and suppressing the release of inflammatory cytokines (IL-6, IL-2, IL-17). Molecular docking and dynamics simulations further revealed that bioactive components in RDP, such as rutin and hyperoside, effectively bind to TNF-α and its receptor TNF receptor 1 (TNFR1), thereby modulating inflammatory responses. These findings provide valuable insights into the multi-target cardioprotective effects of RDP, supporting its application as a promising functional food. Graphical Rosa davurica Pall. (RDP) effectively ameliorates acute myocardial ischemia by neutralizing oxidative stress and disrupting the pro-inflammatory TNF-α/TNFR1 signaling axis. By integrating multi-omics profiling, molecular dynamics, and in vivo models, this study uncovers the multi-target cardioprotective mechanisms of RDP, establishing a robust scientific foundation for its application as a novel cardiovascular functional food.
