Detoxification of ionic liquids using glutathione, cysteine, and NADH: Toxicity evaluation by Tetrahymena pyriformis

Yin Hua Cui,Qing Shan Shi,Dan Dan Zhang,Ling Ling Wang,Jin Feng,Yi Wen Chen,Xiao Bao Xie

Ionic liquids (ILs), also known as green solvents, are widely acknowledged in several fields, such as chemical separation, synthesis, and electrochemistry , owing to their excellent physiochemical properties. However, their poor biodegradability may lead to environmental and health risks, posing a severe threat to humans, thus requiring further research. In this study, the biotoxicities of the imidazolium-based ILs were evaluated in Tetrahymena pyriformis . Moreover, IL detoxification was investigated by addition of glutathione (GSH), cysteine, and nicotinamide adenine dinucleotide (NADH). Reactive oxygen species (ROS) initiated by different IL types caused damage to Tetrahymena , while glutathione, cysteine, and NADH eliminated ROS, achieving the detoxification purposes. Detoxification results showed that NADH exhibited the best detoxification ability, followed by glutathione and cysteine. Finally, RT-PCR results suggested that metallothionein might have participated in IL detoxification.

Detoxification of patulin in apple juice by enzymes and evaluation of its degradation products

Xiaoshuang Liu,Leran Wang,Saiqun Wang,Rui Cai,Tianli Yue,Yahong Yuan,Zhenpeng Gao,Zhouli Wang

The contamination of patulin is widely present in apple, hawthorn, pear, and their products. Many patulin detoxification methods have already been reported, among which enzymes are considered as one of the most promising degradation methods. However, in the reports available so far, patulin-degrading enzymes have rarely been separated and produced, making their application limited. In this study, four commercial enzymes including esterase, peroxidase, lipase A, and lipase M with the potential ability were used for patulin degradation. The results showed that four enzymes can degrade patulin with a rate of 97.8%, 53.2%, 76.3%, and 68.2%, respectively. The soluble solids content, pH value, browning degree, transmittance, organic acids and soluble sugar content, and volatile components of apple juice did not vary much or were within the acceptable range. The main degradation products of patulin were desoxypatulinic acid and hydroascladiol, the toxicity of which have been reported to be lower than that of patulin. Our discovery may provide a more convenient option for enzymes in juice detoxification.

Detoxification and resource utilization of soda ash chromite ore processing residue by copper slag

Lanbin Wang,Yangming Xu,Yanping Ma,Yu Chen,Anju Yang,Guixiang Gan,Yaguang Du,Yan Sun

Chromite ore processing residue (COPR) and copper slag (CS) are industrial waste slags that cause environmental pollution and require effective treatment. This study proposes a new strategy for the co-treatment and resource utilization of COPR and CS. Under the optimal conditions (liquid-solid ratio of 9.99; H 2 SO 4 concentration of 0.85 M; the CS/COPR mass ratio of 9.98%; temperature of 88.20 ℃; and duration of 60 min), the reduction efficiency of Cr(VI) reached 99.48%, and the total Cr leaching concentration of residue reached 1.35 mg/L, which was below the USEPA regulatory limit of 5 mg/L. In addition, the detoxicated slag containing 61.44 wt% Fe 2 O 3 can be used as a raw material in the steel industry. Through kinetic, characterization, and DFT analyses, the effective remediation of Cr(VI) in COPR by CS is a combination of adsorption and reduction. The unreacted shrinkage nuclear reaction model under the control of the surface chemical reaction is the most suitable model to describe the process, and when the apparent activation energy is 63.90 kJ/mol, the apparent rate equation is: 1-(1-x) 1/3 = 1·371×10 7 [CS/COPR] 2·35004 [H 2 SO 4 ] 2·3945 [L/S] 1·95338 exp(–63·90/RT) This is an important study for waste-waste co-treatment and resource utilization of industrial waste.

Lattice distortion SnS2 piezoelectric self-Fenton system for efficient degradation and detoxification of pollutants

Jiang Runren,Lu Guanghua,Wang Min,Chen Yufang,Liu Jianchao,Yan Zhenhua,Xie Haijiao

Both piezoelectricity and self-Fenton catalysis are effective ways to degrade water pollution, but little research has combined them to construct a more efficient water pollution treatment method. Here, a Fe-doped SnS 2 (Sn 1-x Fe x S 2 ) piezoelectric self-Fenton system was constructed, which shows superior water treatment performance. The best piezoelectric properties of the Sn 0.97 Fe 0.03 S 2 system were verified by degrading rhodamine B (RhB). The toxicity analysis of degradation intermediates and solutions confirmed that the toxicity of RhB decreased after degradation. In addition, Kelvin probe force microscopy and photoelectrochemical analysis confirmed the better piezoelectric properties of Sn 0.97 Fe 0.03 S 2 . It has demonstrated the enhancement of systematic piezoelectricity by Fe lattice defects and the formation of self-Fenton by Fe as an active center in the degradation of RhB. In this work, an efficient piezoelectric and self-Fenton technology is constructed to remove organic pollutants from water, which is significant for developing water treatment technology.

Application of atmospheric cold plasma for zearalenone detoxification in cereals: Kinetics, mechanisms, and cytotoxicity analysis

Mengjie Liu,Junxia Feng,Yongqin Fan,Xudong Yang,Ruike Chen,Cui Xu,Hangbo Xu,Dongjie Cui,Ruixue Wang,Zhen Jiao,Ruonan Ma

Introduction Zearalenone (ZEN) is one of the most widely contaminated mycotoxins in world, posing a severe threat to human and animal health. Atmospheric cold plasma (ACP) holds great penitential in mycotoxin degradation. Objectives This study aimed to investigate the degradation efficiency and mechanisms of ACP on ZEN as well as the cytotoxicity of ZEN degradation products by ACP. Additionally, this study also investigated the degradation efficiency of ACP on ZEN in cereals and its effect on cereal quality. Methods The degradation efficiency and products of ZEN by ACP was analyzed by HPLC and LC-MS/MS. The human normal liver cells and mice were employed to assess the cytotoxicity of ZEN degradation products. The ZEN artificially contaminated cereals were used to evaluate the feasibility of ACP detoxification in cereals. Results The results showed that the degradation rate of ZEN was 96.18 % after 30-W ACP treatment for 180 s. The degradation rate was dependent on the discharge power, and treatment time and distance. Four major ZEN degradation products were produced after ACP treatment due to the oxidative destruction of C C double bond, namely C 18 H 22 O 7 ( m / z  = 351.19), C 18 H 22 O 8 ( m / z  = 367.14), C 18 H 22 O 6 ( m / z  = 335.14), and C 17 H 20 O 6 ( m / z  = 321.19). L02 cell viability was increased from 52.4 % to 99.76 % with ACP treatment time ranging from 0 to 180 s. Mice results showed significant recovery of body weight and depth of colonic crypts as well as mitigation of glomerular and liver damage. Additionally, ACP removed up to 50.55 % and 58.07 % of ZEN from wheat and corn. Conclusions This study demonstrates that ACP could efficiently degrade ZEN in cereals and its cytotoxicity was significantly reduced. Therefore, ACP is a promising effective method for ZEN detoxification in cereals to ensure human and animal health. Future study needs to develop large-scale ACP device with high degradation efficiency.

Elucidating the detoxification efficacy of Periplaneta americana delta glutathione S-transferase 1 (PaGSTd1) against organophosphates

Hong Sun,Xinyu Li,Xinyue Yuan,Zhen Tian,Yifan Li,Yalin Zhang,Jiyuan Liu

As an important class of detoxifying enzymes , glutathione S-transferases (GSTs) are pivotal in decreasing insecticide toxicity to insects. Periplaneta americana GSTd1 (PaGSTd1) has been verified as a key enzyme in detoxifying pyrethroid insecticides, but its detoxification capability against a broader spectrum of insecticides has never been investigated. It is revealed that PaGSTd1 expression showed a rapid and significant increase upon exposure to various insecticides (organophosphates, neonicotinoids , and fipronil). Subsequent in vitro metabolic assays indicated that organophosphates , particularly chlorpyrifos-methyl, can be effectively metabolized by PaGSTd1. Further knockdown of PaGSTd1 via RNA interference significantly heightened the susceptibility of P. americana to chlorpyrifos-methyl, underscoring the enzyme's key role in detoxifying chlorpyrifos-methyl. Additionally, this study confirmed that PaGSTd1 cannot mitigate insecticide toxicity through countering oxidative stress . Collectively, these findings elucidate the involvement of PaGSTd1 in the detoxification processes for organophosphates, offering a comprehensive insight into the metabolic mechanisms mediated by GSTs in P. americana . This research provides a foundational understanding for managing GSTs-mediated metabolic resistance in this species, which is crucial for effective pest control strategies.

Catalytic detoxification of mitoxantrone by graphitic carbon nitride (g-C3N4) supported Fe/Pd bimetallic nanoparticles

Qianyu Xu,Haoyang Fu,Jiyuan Gu,Liyu Lei,Lan Ling

The overuse of antibiotics and antitumor drugs has resulted in more and more extensive pollution of water bodies with organic drugs, causing detrimental ecological effects, which have attracted attention towards effective and sustainable methods for antibiotics and antitumor drug degradation . Here, the hybrid nanomaterial (g-C 3 N 4 @Fe/Pd) was synthesized and used to remove a kind of both an antibiotic and antitumor drug named mitoxantrone (MTX) with 92.0% removal efficiency, and the MTX removal capacity is 450 mg/g. After exposing to the hybrid material the MTX aqueous solution changed color from dark blue to lighter progressively, and LC-UV results of residual solutions show that a new peak at 3.0 min (MTX: 13.2 min) after removal by g-C 3 N 4 @Fe/Pd appears, with the simultaneous detection of intermediate products indicating that g-C 3 N 4 @Fe/Pd indeed degrades MTX. Detailed mass spectrometric analysis suggests that the nuclear mass ratio decreased from 445.2 (M + 1H) to 126.0 (M + 1H), 169.1 (M + 1H), 239.2 (M + 1H), 267.3 (M + 1H), 285.2 (M + 1H), 371.4 (M + 1H) and 415.2 (M + 1H), and the maximum proportion (5.63%) substance of all degradation products (126.0 (M + 1H)) is 40-100 times less toxic than MTX. A mechanism for the removal and degradation of mitoxantrone was proposed. Besides, actual water experiments confirmed that the maximum removal capacity of MTX by g-C 3 N 4 @Fe/Pd is up to 492.4 mg/g (0.02 g/L, 10 ppm).

Cadmium Exposure Impairs Development, Detoxification Mechanisms and Gene Expression of Glyphodes pyloalis Walker (Lepidoptera: Pyralidae)

Shuaiqi Zhao,Wanglong Miao,Sheng Sheng,Xin Pan,Ping Li,Weihong Zhou,Fuan Wu

Cadmium (Cd) is the most serious heavy metal pollutant in the agricultural soil of China and can transfer and accumulate through the food chain and affect the growth, development and physiological processes of phytophagous insect.Glyphodes pyloalisWalker (G. pyloalis) is one of the most important pests of mulberry, and there are few studies on the adverse effects of heavy metals on insects, especially mulberry pests. To understand the toxicology of Cd exposure onG. pyloalis, we investigated the effects of three different concentrations of Cd (0, 3.89 and 51.69 mg/kg, labeled as control check (CK), low dose (LD) and high dose (HD)) on the development and detoxification mechanism ofG. pyloalisand explored the molecular mechanism of Cd onG. pyloalislarvae using RNA-seq technology. Transcriptome analysis showed that compared with the CK, a total of 63 differentially expressed genes (DEGs) were identified in LD exposure, including 24 upregulated and 39 downregulated candidates. In CK versus HD groups, 395 upregulated DEGs and 436 downregulated DEGs were identified and the expression patterns of 12 genes related to detoxification and metabolism were verified using qPCR. These DEGs were relevant to multiple specific peroxisome and drug metabolism-cytochrome P450 pathways. Gene annotation and quantitative real-time PCR revealed that a high concentration of Cd significantly stimulated the expression of metabolic detoxification enzyme genes. The results revealed that Cd exposure changed the pupal weight, adult emergence rate and the activities of AKP, ACP and GST inG. pyloalisand induced notable adverse effects at the molecular level on detoxification and metabolism such as concentration and time differences. In addition, we silencedCYP12A2by RNA interference. Bioassays showed that after silencingCYP12A2, the survival rate ofG. pyloalisunder Cd exposure was lower than that of the control group.

Broad-Spectrum Detoxification of Snake Venoms With Supramolecular Materials Integrated via Molecular Recognition and Coassembly

Meng-Meng Chen,Han-Wen Tian,Yuan-Qiu Cheng,Yu-Xin Yue,Yu-Chen Pan,Dong-Sheng Guo

Snakebite is a medical emergency that often leads to high mortality and morbidity in tropical and subtropical regions. The diversity in venom composition between various types of snakes necessitates the use of different specific antivenom treatments, which presents clinical and financial challenges in the management of snakebites. Therefore, developing a broad-spectrum antidote to neutralize complex and diverse snake venoms has assumed critical importance. Herein, a broad-spectrum antidote is developed through molecular recognition and coassembly to neutralize four kinds of snake venom toxins, those are three-finger toxin (3FTx), snake venom metalloproteinase (SVMP), phospholipase A 2 (PLA 2 ), and snake venom serine protease (SVSP), which have high abundance and toxicity. The coassembly of sulfonate-modified amphiphilic calixarene (SCA) and amphiphilic cyclodextrin (CD) is employed to neutralize the toxicity of 3FTx, 1,2-dodecanedithiol (DDT) and thioetheramide phosphorylcholine (TEAPC) are introduced through coassembly to neutralize toxicity of SVMP and PLA 2 , respectively, and the nafamostat (NT), which can be loaded through the host-guest complexation, is employed to neutralize toxicity of SVSP. The broad-spectrum antidote, NT@SCA/CD/DDT/TEAPC, effectively neutralizes key toxins from the most medically important cobra and vipers, significantly improving survival rates of poisoned mice. This study conceptually validated the feasibility of broad-spectrum detoxification and also offers a conveniently integrated supramolecular strategy.

Antagonistic and Detoxification Potentials of Trichoderma Isolates for Control of Zearalenone (ZEN) Producing Fusarium graminearum

Tian Ye,Tan Yanglan,Yan Zheng,Liao Yucai,Chen Jie,De Boevre Marthe,De Saeger Sarah,Wu Aibo

Fungi belonging to Fusarium genus can infect crops in the field and cause subsequent mycotoxin contamination, which leads to yield and quality losses of agricultural commodities. The mycotoxin zearalenone (ZEN) produced by several Fusarium species (such as F. graminearum and F. culmorum) is a commonly-detected contaminant in foodstuffs, posing a tremendous risk to food safety. Thus, different strategies have been studied to manage toxigenic pathogens and mycotoxin contamination. In recent years, biological control of toxigenic fungi is emerging as an environment-friendly strategy, while Trichoderma is a fungal genus with great antagonistic potentials for controlling mycotoxin producing pathogens. The primary objective of this study was to explore the potentials of selected Trichoderma isolates on ZEN-producing F. graminearum, and the second aim was to investigate the metabolic activity of different Trichoderma isolates on ZEN. Three tested Trichoderma isolates were proved to be potential candidates for control of ZEN producers. In addition, we reported the capacity of Trichoderma to convert ZEN into its reduced and sulfated forms for the first time, and provided evidences that the tested Trichoderma could not detoxify ZEN via glycosylation. This provides more insight in the interaction between ZEN-producing fungi and Trichoderma isolates.

Sappanwood-derived polyphenolic antidote of amyloidal toxins achieved detoxification via inhibition/reversion of amyloidal fibrillation

Qize Xuan,JinFeng Zhou,Feng Jiang,Wei Zhang,Anqi Wei,Wenxue Zhang,Qi Zhang,Hao Shen,Hui Li,Chao Chen,Ping Wang

The formidable virulence of methicillin-resistant staphylococcus aureus (MRSA) have thrown great challenges to biomedicine, which mainly derives from their autocrine phenol-soluble modulins (PSMs) toxins, especially the most toxic member termed phenol-soluble modulins α3 (PSMα3). PSMα3 cytotoxicity is attributed to its amyloidal fibrillation and subsequent formation of cross-α sheet fibrils. Inspired by the multiple biological activity of Sappanwood, herein, we adopted brazilin, a natural polyphenolic compound originated from Caesalpinia sappan , as a potential antidote of PSMα3 toxins, and attempted to prove that the regulation of PSMα3 fibrillation was an effective alexipharmic way for MRSA infections. In vitro results revealed that brazilin suppressed PSMα3 fibrillation and disassembled preformed amyloidal fibrils in a dose-dependent manner, in which molar ratio (brazilin: PSMα3) of efficient inhibition and disassembly were both 1:1. These desired regulations dominated by brazilin benefited from its bonding to core fibrils-forming residues of PSMα3 monomers urged by hydrogen bonding and pi-pi stacking, and such binding modes facilitated brazilin-mediated inhibition or disruption of interactions between neighboring PSMα3 monomers. In this context, these inhibited and disassembled PSMα3 assemblies could not easily insert into cell membrane and subsequent penetration, and thus alleviating the membrane disruption, cytoplasmic leakage, and reactive oxygen species (ROS) generation in normal cells. As such, brazilin dramatically decreased the cytotoxicity borne by toxic PSMα3 fibrils. In addition, in vivo experiments affirmed that brazilin relieved the toxicity of PSMα3 toxins and thus promoted the skin wound healing of mice. This study provides a new antidote of PSMα3 toxins, and also confirms the feasibility of the assembly-regulation strategy in development of antidotes against supramolecular fibrillation-dependent toxins.

Hepatic ZBTB22-mediated detoxification ameliorates acetaminophen-induced liver injury by inhibiting pregnane X receptor signaling

Yingjian Chen,Tianqi Cui,Shaorong Xiao,Tianyao Li,Yadi Zhong,Kaijia Tang,Jingyi Guo,Shangyi Huang,Jiabing Chen,Jiayu Li,Qi Wang,Jiawen Huang,Huafeng Pan,Yong Gao

Overdose acetaminophen (APAP) can cause acute liver injury (ALI), but the underlying mechanism remains undetermined. This study explored the role of hepatic Zinc Finger And BTB Domain Containing 22 (ZBTB22) in defense against APAP-mediated hepatotoxicity. The results showed that hepatic ZBTB22 expression was significantly reduced in patients with ALI and mice. In mouse primary hepatocytes (MPHs), ZBTB22 deletion aggravated APAP overdose-induced ALI, whereas ZBTB22 overexpression attenuated that pathological progression. The results were further verified in ZBTB22 over-express or knockout mice models. In parallel, hepatocyte-specific ZBTB22 knockout also enhanced ALI. Furthermore, ZBTB22 decreased pregnane X receptor (PXR) expression, and the PXR activator pregnane-16α-carbonitrile suppressed the protective effect of ZBTB22 in APAP-induced ZBTB22-overexpressing mice. Collectively, our findings highlight the protective effect of ZBTB22 against APAP-induced ALI and unravel PXR signaling as the potential mechanism. Strategies to increase hepatic ZBTB22 expression represent a promising therapeutic approach for APAP overdose-induced ALI.
