Construction and application of nano ZnO/eugenol@yam starch/microcrystalline cellulose active antibacterial film

Junfeng Cheng,Hualin Wang

The goal of this study was to develop new biocomposite films that can better protect and prolong the shelf life of food. Here, a ZnO: eugenol@yam starch/microcrystalline cellulose (ZnO:Eu@SC) antibacterial active film was constructed. Because of the advantages of metal oxides and plant essential oils, codoping with these can effectively improve the physicochemical and functional properties of composite films. The addition of an appropriate amount of nano-ZnO improved the compactness and thermostability , reduced the moisture sensitivity, and enhanced the mechanical and barrier properties of the film. ZnO:Eu@SC exhibited good controlled release of nano-ZnO and Eu in food simulants. Nano-ZnO and Eu release was controlled by two mechanisms: diffusion (primary) and swelling (secondary). After loading Eu, the antimicrobial activity of ZnO:Eu@SC was significantly enhanced, resulting in a synergistic antibacterial effect. Z 4 :Eu@SC film extended the pork shelf life by 100 % (25 °C). In humus, the ZnO:Eu@SC film was effectively degraded into fragments. Therefore, the ZnO:Eu@SC film has excellent potential in food active packaging.

Electrochemical determination of eugenol using a poly-γ-aminobutyric acid modified glassy carbon electrode

Mingjing Yin,Shanshan Liu,Xinying Ma,Xiangyan Meng,Mingyong Chao

Eugenol, an active substance widely used in the medical, food, and cosmetic industries, has garnered increasing attention due to its potential adverse effects on human health resulting from excessive use. In this study, a sensitive sensor for the determination of eugenol was developed by electropolymerizing γ-aminobutyric acid ( g ABA) onto the surface of a glassy carbon electrode (GCE). Experiments showed that the poly γ-aminobutyric acid ( pg ABA) membrane exhibited excellent electrocatalytic activity for the oxidation of eugenol, and its oxidation peak current was greatly increased. Under optimized conditions, the oxidation peak current of eugenol was linearly proportional to its concentration in a range of 0.6–100 μmol/L, with a correlation coefficient of 0.9985 and a detection limit of 0.2 μmol/L. The sensor’s ability to detect eugenol in various authentic samples, with recovery rates ranging from 98.0 % to 101.3 %, attests to its great potential for practical application.

A molecularly imprinted electrochemical sensor using copper nanoparticles as built-in probe for eugenol detection

Shuang Luo,Feiyang Shen,Junyao Li,Wenrong Cai,Yong Kong

A molecularly imprinted electrochemical sensor is facilely designed for the detection of eugenol (EU). Copper nanoparticles (CuNPs) are first electrodeposited on the surface of a glassy carbon electrode (GCE) and act as the built-in probe. By using EU as the template molecules, o -phenylenediamine (o-PD) monomers are electropolymerized on the surface of the CuNPs/GCE. After the removal of the EU template molecules with the ethanol/water mixture, the molecularly imprinted electrochemical sensor is obtained. According to the cathodic peak currents ( I pc ) of copper oxide generated from the oxidation of the CuNPs probe on the differential pulse voltammograms (DPVs), sensitive detection of EU can be achieved. The EU sensor shows a wide linear concentration range from 0.05 to 100 μM with a low limit of detection (LOD) of 0.022 μM under the optimized conditions. Additionally, the EU sensor exhibits excellent specificity, repeatability, reproducibility and stability. Finally, the developed EU sensor is used for the detection of EU in real samples, and reliable results are obtained.

A lipophilicity-based strategy: Curcumin-mediated photodynamic inactivation combined with eugenol for enhanced bacon preservation

Yongqi Yao,Wen Zhang,Lijun Tan,Haoran Li,Yumeng Li,Baocai Xu

Foodborne spoilage bacteria are the key factor causing food deterioration, seriously restricting the shelf life of food and threatening consumer safety. In this study, we investigated the synergistic bactericidal effect of curcumin-mediated photodynamic inactivation technology (Cur-PDI) combined with eugenol (Eug) on two typical f foodborne spoilage bacteria Leuconostoc mesenteroides and Carnobacterium maltaromaticum , and further evaluated its application in bacon preservation. The results indicated that under the combined treatment conditions (20 μΜ Cur, 25 μg/mL Eug, 20 min of light exposure), the viable bacterial counts of two bacteria decreased significantly by 7.11 log CFU/mL and 7.12 log CFU/mL compared with the blank control. As both Cur and Eug are lipophilic, the combination exhibited a stronger bactericidal effect in a lipid system than the water-soluble photosensitizer erythrosine (ERY). Mechanistic studies revealed that, compared with PDI alone, the combined treatment more significantly disrupted bacterial membrane integrity, induced higher levels of reactive oxygen species (ROS), and severely interfered with antioxidant defense and energy metabolism. In the bacon preservation experiment, the combined treatment effectively reduced the initial colony count of the product and continuously inhibited the growth of microorganisms during storage for 25 days. This study systematically clarified the synergistic antibacterial mechanism of Cur-PDI and Eug, and verified its potential in the preservation of bacon, providing a theoretical basis for the development of natural and efficient preservation strategies for foods rich in fat.

Enhanced Activity of Eugenol Pickering Emulsion Using Lactoferrin-Chlorogenic Acid Complex as a Green Stabilizer

Xiyao Dong,Tongyan Liu,Mengfan Li,Xinying Wang,Qingxiang Guan

Exploiting natural and bioactive stabilizers to fabricate green emulsions has attracted increasing attention. In this work, lactoferrin (LF) was combined with chlorogenic acid (CGA) to construct the LF-CGA complex to prepare a eugenol (EG) Pickering emulsion (ELC/PE). The surface activity and binding mechanism of LF-CGA were explored. Additionally, the antioxidant, antibacterial, and anti-inflammatory capacities of ELC/PE were compared. The contact angle of LF-CGA approached 90° (81.9°), indicating enhanced stability at the oil-water interface. The interaction between LF and CGA was primarily driven by static quenching and hydrogen bonding. The ELC/PE consisted of 1.5 wt% LF-CGA, with distilled water and EG as the aqueous and oil phases (15:1 v/v, aqueous/oil). The DPPH − , O 2 − , and ABTS + scavenging rates of ELC/PE were 86.3%, 75.3%, and 83.4%, respectively. The ELC/PE displayed superior antibacterial ability, along with inhibition rates of 99.0% and 99.5% against Candida albicans and Staphylococcus aureus , respectively. Additionally, the ELC/PE exhibited the most significant suppression of ear swelling (81.8%), confirming its anti-inflammatory ability. These values were the highest among all experimental groups. The LF-CGA complex acts as an emulsifier to enhance EG bioactivities. This novel complex holds great potential for expanding the practical applications of poorly soluble bioactive compounds. Graphical Lactoferrin (LF) and chlorogenic acid (CGA) were used to design a green Pickering emulsion (PE) emulsifier. Several chemical methods were used to characterize the complex. The binding mechanism between LF and CGA was evaluated. The complex exhibited superior antibacterial, antioxidant, and anti-inflammatory ability, which enhanced the eugenol PE effect. This emulsifier will widen the application of insoluble ingredients.

Polymethoxyflavones-eugenol nanoemulsion: Dual physiological and metabolomic insights into its antibacterial action on Escherichia coli

Hongyang Chen,Zimao Ye,Yanchi Liu,Zhiqin Zhou

An integrated physiological and non-targeted metabolomics approach was used to construct a eugenol-based polymethoxyflavones nanoemulsion delivery system and examine its increased antibacterial activity on E. coli . Compared with pure eugenol nanoemulsion (EG, MIC: 640 μg/mL), the polymethoxyflavones-eugenol nanoemulsion (EGP) exhibited significantly enhanced antimicrobial activity (MIC: 320 μg/mL). EGP treatment disrupted the integrity of E. coli cell membranes and walls, increased cell permeability. Non-targeted metabolomics analysis (compared with CK) revealed 685 upregulated and 642 downregulated differentially accumulating metabolites, lysine and glutamine are reduced in E. coli , and glutaric acid, aminoadipic acid, and D-lactic acid are increased. Pathway enrichment analysis showed that amino acid metabolism and energy metabolism were significantly disturbed by TCA cycle disruption. EGP exhibits superior antimicrobial efficacy against E. coli through a multi-targeted mechanism involving cell membrane disruption and metabolic pathway interference. This study provides valuable insights into the development of natural antimicrobial nanoemulsion as potential alternatives to synthetic preservatives for food safety applications.

Hydroxypropyl-β-cyclodextrin inclusion complex improves the percutaneous therapeutic effect of eugenol on psoriasis mice

Yanyan Guo,Yingxue Yue,Huanhuan Wang,Ziyi Zhong,Bin Chen,Luan Shu,Jing Wang,Zhenhai Zhang

The insolubility and volatility of eugenol (EG) limit its clinical application. In this study, cyclodextrin (CD) was used to encapsulate EG and hydroxypropyl-β-cyclodextrin (HP-β-CD) with lower binding energy was selected by comparison. The preparation process optimization and characterization showed that the EG-HP-β-CD inclusion complex could be successfully prepared. More importantly, inclusion complex can significantly increase the solubility and stability of EG. In addition, in vitro release and transdermal studies, inclusion complex gel release curve conformed to the Higuchi equation, and the cumulative release rate reached 80% at 24 h, which provided good sustained release properties. Notably, inclusion complex gel increased EG skin retention by reducing the transdermal permeation rate and cumulative permeation amount. It was verified in the mouse psoriasis model that the inclusion complex gel group had better efficacy. By comparison, the epidermal thickness and inflammatory infiltration of inclusion complex group were improved, which was closely related to drug skin retention. The anti-inflammatory effect was revealed by regulating the expression of IL-17 A, IL-1β, IL-6, IL-23, and TNF-α, participating in JAK1/STAT3 pathway. The results provide a new idea and theoretical basis for the formulation design of volatile oil transdermal therapy for psoriasis.

Enhanced microdialysis based on hydrophobic deep eutectic solvents for sampling of eugenol in Glyptocidaris crenularis

Jiaze Yan,Chao Jiang,Jiqi Zheng,Ming Guo

Background Microdialysis is an in vivo technique for dynamically analyzing biochemical components in tissues and fluids. Traditional microdialysis systems are primarily designed for low molecular weight polar compounds, resulting in lower recoveries for hydrophobic compounds. This study aimed to develop a greener, cost-effective, and enhanced microdialysis method for recovering eugenol, isoeugenol, and methyl isoeugenol, utilizing hydrophobic deep eutectic solvents as the perfusate, with the method's feasibility verified through in vivo experiments. Results Among the 19 types of deep eutectic solvents evaluated, thymol-levulinic acid was identified as the most suitable for eugenol compounds based on liquid-liquid microextraction and microdialysis experiments, showing an in vitro microdialysis recovery approximately 100 times higher than that of water. The optimization of microdialysis parameters was conducted, including molar ratio of HBA:HBD, temperature, flow rate, and semi-permeable membrane length. In the in vivo microdialysis experiments, the temporal concentration of eugenol, which was used as an anesthetic in Glyptocidaris crenularis, was detected by GC-MS/MS and calibrated using ex vivo recovery. When the concentration of eugenol in artificial seawater was maintained at 100 μg mL −1 , the eugenol levels in the coelomic fluid of Glyptocidaris crenularis rose from 0 to 6.89 ± 0.38 μg mL −1 within 60 min, then stabilized in next 80 min. Significance The findings demonstrate that deep eutectic solvents enhanced microdialysis provides high recoveries and enables real-time monitoring of drug concentration changes in biological systems, making this method a valuable tool for pharmacokinetic studies and environmental monitoring.

Topical effectiveness of eugenol phytosome / chitosome hydrogels on the healing process of infected excision wounds

Andishe Dashti,Amir Reza Karamibonari,Mohammad Reza Farahpour,Zohreh Ghazi Tabatabaei

The fabrication of a safe antibacterial wound dressing lack of antibiotics is essential for the wound healing process. Hydrogels with appropriate mechanical properties containing antibacterial agents are good candidates for this purpose. This novel study was conducted to evaluate the effectiveness of hydrogels prepared from eugenol encapsulated into phytosome and chitosomes (HG-Phyt/Chit@Eu) on the wound healing process of infected wounds. The physicochemical properties, toxicity, eugenol release and antibacterial activities of formulations were assessed under in vitro condition. The HG-Eu, HG-Phyt@Eu, and HG-Phyt/Chit@Eu were formulated into hydrogel form and administrated for the treatment of infected wounds. The physicochemical results confirmed successful synthesis of formulations. The results showed that formulations were safe and exhibited antibacterial activity against positive and/or negative-Gram bacteria. The addition of phytosome and chitosomes reduced the rapid release of Eu. Topical administration of hydrogels significantly accelerated the wound healing process by decreasing total bacterial count and the expressions of COX-2, 4-HNE, NF-κB, Nrf2, iNOS-positive cell and increasing the expression of COL-1A positive staining compared with control mice. In conclusion, this preliminary study showed that HG-Phyt/Chit@Eu could compete with mupirocin and can be used for the treatment of infected wounds.

Huilu Sun,Ruxue Wei,Genqiang Chen,Zhiping Che,Xiaobo Huang,Yingwu Chen,Yuee Tian

Eugenol is the main component of clove basil oil extracted from plants such as Syzygium aromaticum and Ocimum gratissimum . To discover biorational natural product-based pesticides, a series of eugenol/dihydroeugenol derivatives containing carbamate moiety were prepared by introducing an active aminooyloxy group at its C1 hydroxy position, and their structures were well characterized by proton nuclear magnetic resonance ( 1 H NMR), carbon-13 NMR ( 13 C NMR), high-resolution mass spectrometry, and melting point. Bioactivities of these target compounds as anti-oomycete and anti-fungal agents against two serious agricultural pests, Phytophthora capsici and Fusarium graminearum were evaluated. Among assessed compounds: (1) Title compounds 1b and 2b showed significant anti-fungal against F. graminearum , with half maximal effective concentration values of 29.13 and 35.04 mg/L, respectively. (2) The title compounds have obvious selectivity, and their anti-fungal activity is significantly better than their anti-oomycete activity. (3) The anti-oomycete and anti-fungal activities of introducing the aminoalkyloyloxy group at the C1 hydroxyl position of eugenol or dihydroeugenol are significantly better than that of introducing the aminoaryloyloxy group. (4) The presence of a double bond is beneficial for increasing anti-fungal activity, and the reduction of a double bond is beneficial for increasing anti-oomycete activity. Graphical

Shiqing Li,Wenhui Shi,Xiaoyan Wang,Xiaona Hu,Shujing Li,Yuxi Zhang

In this study, a multifunctional food packaging material (G-Eug-NPs-NF) based on gelatin nanofiber membranes (G-NF) containing eugenol nanoparticles (Eug-NPs) were prepared. The Eug-NPs were fabricated through the electrostatic self-assembly by using two ionic polysaccharides chitosan and sulfobutylether-β-cyclodextrin (SCD). Compared with G-NF, the G-Eug-NPs-NF exhibited better antioxidant capacity , which positively correlated with Eug-IC concentration, up to 84.17%. In addition, the latter had a good inhibitory effect on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) , and was more conducive to inhibit the growth of S. aureus . At the same time, the sustained release experiment showed that the incorporation of Eug-NPs in nanofiber membranes improved the sustained release ability of G-Eug-NPs-NF. G-Eug-NPs-NF exhibits the promising application of Eug in food, medicine and other fields.
