New application of rutin: Repair the toxicity of emerging perfluoroalkyl substance to Pseudomonas stutzeri

Yi Qian,Yunhao Zhang,Achuo Anitta Zuh,Weichuan Qiao

Per- and polyfluoroalkyl substances (PFASs) are toxic to microorganisms , thereby affecting microbial communities in sludge and soil, but how to repair the toxicity of microorganisms remains unclear. In this study, rutin, an antioxidant, was added into a culture medium with an aerobic denitrification bacteria, Pseudomonas stutzeri , under the exposure of sodium perfluorononyloxy-benzenesulfonate (OBS) to evaluate the repair mechanisms of rutin to the toxicity of OBS to the bacteria. The results showed that rutin could repair the damage of OBS to cell structures, and reduce the death rates of the bacteria under OBS exposure. The dosage of rutin reduced the effect on the inhibition of denitrification ability of P. stutzeri under OBS exposure. Compared with the bacteria exposed to single OBS, the dosage of rutin resulted in that the death rates recovered from 96.2% to 66.4%, the growth inhibition rate decreased from 46.5% to 15.8%, the total nitrogen removal rate recovered from 66.9% to 100%, and the NO 2 − content recovered from 34.5 mg/L to 0.22 mg/L. The expressions of key denitrification genes ( napA , nirS , norB , nosZ ) were recovered after adding rutin under OBS exposure. Rutin changed the positive rate of reactive oxygen species , the relative superoxide dismutase and catalase activities in the bacteria which exposed to OBS. The mechanism by which rutin repaired the toxicity of OBS to P. stutzeri related to inhibiting the activities of antioxidant and denitrification enzymes rather than affecting the expressions of genes involved in these enzymes. This study sheds light on the repair method of micro-organics and reveals the repair mechanisms under PFASs exposure.

Fluorescent Sensors Based on Cu-Doped Carbon Quantum Dots for the Detection of Rutin

Wang Cheng-ya,Shang Shanzhai,Zheng Xudong,Lei Ping,Han Jingmei,Yuan Linda,Li Zhiqiang,Wang Ru,Gong Weimin,Tang Jianguo,Yang Yaling

In this paper, Cu-doped carbon quantum dots (Cu-CQDs) were prepared by carrying out thermolysis of Na2[Cu(EDTA)] and hydroxylamine hydrochloride at 300 ºC for 2 h. The as-prepared Cu-CQDs were characterized by UV-Vis, Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD). The Cu-CQDs exhibited good fluorescence property, high stability, excitation-dependent emission behavior and a quantum yield of 9.8%. More significantly, the synthesized Cu-CQDs were developed for sensing rutin (RT) based on the fluorescence quenching of the prepared Cu-CQDs. A good linear relationship between the fluorescence quenching of Cu-CQDs and different concentrations of RT was obtained in the range of 0.1-15 µg mL-1 with a correlation coefficient of 0.9959. The detection limit was as low as 0.05 µg mL-1 (signal-to-noise ratio, S/N = 3). The possible mechanism of fluorescence quenching was explored, which was proved to be inner filter effect. Importantly, the proposed method was successfully applied in the detection of RT tablet samples and with satisfactory results, showing the practical applications.

M.A. Khaleque,M.R. Ali,M.S. Bacchu,M.R.A. Mamun,M.I. Hossain,M.S. Hossain,Mohamed Aly Saad Aly,M.Z.H. Khan

In this work, a novel electrochemical detection strategy was developed based on a metal-organic framework of zinc oxide nanorod nanoparticles and rutin for selective screening of Thiourea as toxic chemicals. The zinc oxide nanorod were synthesized by following direct chemical precipitation methods and characterized by X-ray diffraction and X-ray photoelectron spectroscopy analysis. The surface of modified electrodes was also characterized by field emission scanning electron microscopes, energy-dispersive X-ray spectroscopy, and attenuated total reflectance flourier transform infrared spectroscopy. Furthermore, the electrochemical activity of the developed sensor was tested by cyclic voltammetry, differential pulse voltammetry, and electrochemical impedance spectroscopy. The modified electrode showed outstanding electro-catalytic activity towards the detection of Thiourea in phosphate buffer saline at a high pH level of 12.0. The proposed sensor showed a linear range of linearity in a concentration ranging from 5.0 × 10 −6 - 900 × 10 −6 molL −1 and a detection limit of 2.0 × 10 −6 molL −1 . Moreover, the selectivity of the developed electrochemical sensor was investigated for the detection of Thiourea in the presence of organic compounds and a group of anions. Furthermore, the proposed strategy demonstrated an excellent recovery value in the spiked farmland water and fruit juice sample.

Gelatin hydrogel/contact lens composites as rutin delivery systems for promoting corneal wound healing

Lianghui Zhao,Xia Qi,Tao Cai,Zheng Fan,Hongwei Wang,Xianli Du

Corneal wound healing is a highly regulated biological process that is of importance for reducing the risk of blinding corneal infections and inflammations. Traditional eye drop was the main approach for promoting corneal wound healing. However, its low bioavailability required a high therapeutic concentration, which can lead to ocular or even systemic side effects. To develop a safe and effective method for treating corneal injury, we fabricated rutin-encapsulated gelatin hydrogel/contact lens composites by dual crosslinking reactions including in situ free radical polymerization and carboxymethyl cellulose/N-hydroxysulfosuccinimide crosslinking. In vitro drug release results evidenced that rutin in the composites could be sustainedly released for up to 14 days. In addition, biocompatibility assay indicated nontoxicity of the composites. Finally, the effect of rutin-encapsulated composites on the healing of the corneal injury in rabbits was investigated. The injury was basically cured in corneas using rutin-encapsulated composites (healing rate, 98.3% ± 0.7%) at 48 h post-operation, while the damage was still present in corneas using the composite (healing rate, 87.0% ± 4.5%). Further proteomics analysis revealed that corneal wound healing may be promoted by the ERK/MAPK and PI3K/AKT signal pathways. These results inform a potential intervention strategy to facilitate corneal wound healing in humans.

Fluorescent sensing of rutin by carbon dots derived from the heart of cumin seeds

Xiao-Lin Xie,Zhao Zhang,Wen Xiong,Jie Wang,Wei Gong,Wen Xu,Shuang Cai,Jing Li

Carbon dots (CDs) prepared from biomass usually produce poor quantum yield and reproducibility. A fine separation of raw materials may enhance the fluorescence performance. In this work, embryos of cumin seeds were precisely chosen as the carbon source to prepare (CDs) through hydrothermal method. The obtained product (CDs-E) possessed a higher quantum yield of 10.60 % than those synthesized with testas or whole seeds, which may be due to the relatively smaller particle size and more C-N functional groups. Rutin, as a common natural active component, could quench the fluorescence of CDs-E notably through internal filtering effect. Herein, a highly selective fluorescent sensor based on CDs-E for detecting rutin was proposed with a linearity range of 0.05 ∼ 30 μM and a detection limit of 0.015 μM obtained. Additionally, the developed method was successfully applied for the detection of rutin in Flos Sophorae , rutin tablets and capsules. This work showed the significance of fine classification for natural carbon source in preparation of CDs and provided a feasible method for quality monitoring of rutin-related pharmaceuticals, health care products and traditional Chinese medicinal materials.

Multilayer Ti3C2-CNTs-Au Loaded with Cyclodextrin-MOF for Enhanced Selective Detection of Rutin

Jia Chen,Maoheng Fei,Meijun Ni,Yilin Wang,Zhifang Liu,Yixi Xie,Pengcheng Zhao,Zhiyong Zhang,Junjie Fei

In this work, multi-layer Ti 3 C 2 – carbon nanotubes – gold nanoparticles (Ti 3 C 2 -CNTs-Au) and cyclodextrin metal-organic framework – carbon nanotubes (CD-MOF-CNTs) have been prepared by in situ growth method and used to construct the ultra-sensitive rutin electrochemical sensor for the first time. Among them, the large number of metal active sites of Ti 3 C 2 , the high electron transfer efficiency of CNTS, and the good catalytic properties of AuNPs significantly enhance the electrochemical properties of the composite carbon nanomaterials. Interestingly, CD-MOF has a unique host–guest recognition and a large number of cavities, molecular gaps, and surface reactive groups, which gives the composite outstanding accumulation properties and selectivity for rutin. Under the optimized conditions, the constructed novel sensor has satisfactory detection performance for rutin in the range of 2 × 10 −9 to 8 × 10 −7 M with a limit of detection of 6.5 × 10 −10 M. In addition, the sensor exhibits amazing anti-interference performance against rutin in some flavonoid compounds and can be used to test natural plant samples (buckwheat, Cymbopogon distans , and flos sophorae immaturus). This work has promising applications in the field of environmental and food analysis, and exploring new directions for the application of Mxene-based composites.

Ovalbumin-Rutin complexes as hypoallergenic foaming agents: A dual-effect strategy for protein modification

Zuyue Li,Ran Yang,Mahmoud Abou-Elsoud,Peng Hu,Yijie Li,Long Sheng,Dong Uk Ahn,Xi Huang

This study aimed to overcome two inherent limitations of ovalbumin (OVA), specifically its high allergenicity and limited foaming properties, through the formulation of OVA-rutin complexes. Various modification levels of OVA-Rutin complexes were successfully produced, as confirmed by polyphenol binding equivalents and amino acid residues changes. Multispectral analyses collectively demonstrated that rutin promoted structural loosening in OVA and improved its molecular flexibility. At an OVA-Rutin ratio of 1:0.4, the IgE binding capacity diminished by 35.70%. Meanwhile, the system exhibited superior Fe 2+ chelation ability (50.88±0.31%), DPPH radical scavenging capability (67.18±2.25%) and ABTS radical scavenging ability (87.23±0.50%). The combination of rutin and OVA significantly increased foaming ability by 75.56-155.24% and foaming stability by up to 1.32-fold. This improved foam performance was attributed to the desirable air-water interfacial activity and rearrangement capacity of the OVA-Rutin complexes. These findings provide an effective strategy for simultaneously enhancing foaming properties of OVA and mitigating its allergenicity.

Synergistic Effects and Mechanisms of Action of Rutin with Conventional Antibiotics Against Escherichia coli

Lankun Yi,Yubin Bai,Xu Chen,Weiwei Wang,Chao Zhang,Zixuan Shang,Zhijin Zhang,Jiajing Li,Mingze Cao,Zhen Zhu,Jiyu Zhang

Rutin is a widely known plant secondary metabolite that exhibits multiple physiological functions. The present study focused on screening for synergistic antibacterial combinations containing rutin, and further explored the mechanisms behind this synergy. In vitro antibacterial test results of rutin showed that the ranges of minimum inhibitory concentration (MIC) and Minimum bactericidal concentration (MBC) are 0.125–1 and 0.125–2 mg/mL, respectively. However, rutin and amikacin have a significant synergistic effect, with a fractional inhibitory concentration index (FICI) range of 0.1875–0.5. The time bactericidal curve proved that the combination of rutin and amikacin inhibited bacterial growth within 8 h. Scanning electron microscopy (SEM) revealed that a low-dose combination treatment could disrupt the cell membrane ofEscherichia coli(E. coli). A comprehensive analysis using alkaline phosphatase (AKP), K+, and a protein leakage assay revealed that co-treatment destroyed the cell membrane ofE. coli, resulting in the significant leakage of AKP, intracellular K+, and proteins. Moreover, confocal laser scanning microscopy (CLSM) and red–green cell ratio analysis indicated severe damage to theE. colicell membrane following the co-treatment of rutin and amikacin. This study indicates the remarkable potential of strategically selecting antibacterial agents with maximum synergistic effect, which could significantly control antibiotic resistance.

Liposome-Encapsulated Rutin Attenuates Cisplatin-Induced Ototoxicity via Suppression of P53-Associated Oxidative Injury.

Bo Liu,Yaqin Tu,Xiangrui Li,Wenting Yu,Wenqing Zou,Wei Tang,Shimin Zong,Songwei Tan,Hongjun Xiao

Cisplatin (CDDP) is a widely used chemotherapeutic agent, but its clinical applications are constrained by ototoxic side effects. Currently, few effective strategies exist to prevent or mitigate CDDP-induced ototoxicity. Rutin is known for its cell-protective effects by reducing oxidative stress and inhibiting apoptosis. However, its limited water solubility and inefficient delivery to the inner ear pose substantial challenges. To address this, rutin is encapsulated in liposomes (Lip-Rutin) for nanoscale drug delivery, leveraging its antioxidant properties. Lip-Rutin markedly attenuates CDDP-induced oxidative stress damage and apoptosis, demonstrating a protective effect on OC-1 cells. The efficacy of Lip-Rutin in safeguarding against CDDP-induced ototoxicity is further validated through in vivo studies. Consequently, Lip-Rutin emerges as a promising novel therapeutic agent for combating CDDP-induced ototoxicity.

Highly Selective and Sensitive Sensor Based IL and CMC-MWCNTs Nanocomposite for Rutin Determination

Xin Meng,Bao-Lin Xiao,Xin-Yan Song,Xin-Xin Ma,Yang-Yang Li,Lin-Lin Ma,Yu-Jie Chen,Yu-Ying Li,Ke-Xin Xu,Jian-She Wei,Tao Hong,Ali Akbar Moosavi-Movahedi,Jun Hong

Rutin is a natural antioxidant flavonoid compound with anti-inflammatory, antioxidant, and antiviral effects that is used to prepare drugs with wide application in clinical treatment. Therefore, the quantitative detection of rutin has important practical significance. In this work, a novel electrochemical sensor based on glassy carbon electrodes (GCEs) modified with sodium carboxymethylcellulose (CMC), multi-walled carbon nanotubes (MWCNTs), and 1-butyl-3-methylimid (ionic liquid, IL) was developed for the super-sensitive detection of the flavonoid rutin. The properties of these modified materials were analyzed by transmission electron microscope (TEM), cyclic voltammograms (CVs), and electrochemical-impedance spectroscopy (EIS). CMC was used to disperse MWCNTs to further enhance their hydrophilicity and biocompatibility. The modified MWCNTs improved the sensitivity of rutin detection. The square-wave voltammetry (SWV) technique showed that the linear range of rutin concentration determination was 0.01 μM to 1 µM and 1 µM to 10 µM. The minimum concentration detection of rutin was 0.83 nM and 6.6 nM, respectively. The proposed sensor presented good selectivity for rutin and successfully analyzed rutin content in the pharmaceutical rutin tablets. These results are consistent with those measured by ultra-high-performance liquid chromatography (UHPLC). Therefore, this sensor has latent application value in the analysis of rutin in food and drug tablets and nutraceutical samples.Keywords:sensor;multi-walled carbon nanotubes;ionic liquid;sodium carboxymethylcellulose;rutin

The facile activation of graphite for the improved determination of dopamine, rutin and acetamidophenol

Yanxue Xu,Guihua Chen,Yunting Qin,Dan Xiao

A glassy carbon electrode (GCE) was modified with graphite powder (G/GCE), and then treated with a solution of sodium peroxide for several minutes to prepare an oxidized G/GCE (OG/GCE). The OG/GCE has prominently improved responses toward dopamine (DA), rutin (RT) and acetamidophenol (APAP), of which the anodic peak current was increased by 2.4, 4.0 and 2.6 fold compared with that obtained using the G/GCE. The redox peaks of DA, RT and APAP on the OG/GCE could be separated sufficiently. The corresponding redox processes were confirmed as diffusion-controlled and parameters such as the charge transfer coefficients (α), saturating adsorption capacity (Γ*) and catalytic rate constant (kcat) were estimated. In individual detection, the linear ranges for DA, RT and APAP were 10 nM–10 μM, 1.00 nM–150 nM and 20 nM–30 μM, respectively, and the LODs for DA, RT and APAP were estimated as 6.23 nM, 0.36 nM and 13.1 nM with 3σ/S, respectively. The contents of RT and APAP in drugs were determined and agreed with the labeled contents. The recoveries of DA in serum and sweat were in the range of 91–107%, indicating that the determination results achieved by the OG/GCE are reliable. The practical use of the method was verified with a graphite-modified screen-printed carbon electrode (G/SPCE), which was also activated by Na2O2 to prepare OG/SPCE. The recovery of DA in sweat achieved using the OG/SPCE was 91.26%.

Degradation kinetics of rutin encapsulated in oil-in-water emulsions: impact of particle size

Yuteng Fu,David Julian McClements,Shunjing Luo,Jiangping Ye,Chengmei Liu

BACKGROUND Rutin is a natural bioactive flavonoid that is poor in water solubility and chemical stability. Encapsulation can be used to protect bioactive molecules from chemical or physical decomposition during food processing and storage. Thus, the effect of initial particle size on the ability of oil-in-water emulsions to retain rutin during storage was investigated. RESULTS Rutin was encapsulated in oil-in-water emulsions with different mean surface-weighted diameters: d 3,2 = 0.56 μm (small), 0.73 μm (medium), and 2.32 μm (large). As expected, the resistance of the emulsions to coalescence and creaming during storage increased as the particle size decreased due to weakening of the colloidal and gravitational forces acting on the droplets. The concentration of rutin in the emulsions decreased during storage (28 days), which was mainly attributed to photodegradation of the flavonoid. The loss of rutin from the emulsions during storage was fitted using a second-order equation. The rutin degradation rate constant k decreased and the half-life t 1/2 increased with decreasing droplet size, which was attributed to the stronger encapsulation and light scattering by smaller oil droplets reducing the amount of light that can penetrate into the emulsions. CONCLUSION This study has important implications for the design of more efficacious emulsion-based delivery systems for incorporating health-promoting nutraceuticals into foods. © 2022 Society of Chemical Industry.
