Mechanical properties of β-Si3N4/SiCN ceramics formed by digital light processing

Zhongya Zhang,Lijuan Zhang,Guangwu Wen,Yongzhao Hou,Nannan Zhu,Cheng Zhong,Jing Xue,Chuncheng Wei,Shuang Li

In this paper, polysilazane photocurable slurry with low viscosity and excellent rheological properties was prepared with β-Si 3 N 4 filler. Digital light processing (DLP) technique was utilised to form green bodies through optimized printing parameters. It was found that the introduction of β-Si 3 N 4 filler reduced the shrinkage and improved the mechanical properties of the polysilazane derived SiCN ceramics. The flexural strength of the resulted ceramics reached a maximum of 253.0 ± 15.1 MPa while 20 wt% of β-Si 3 N 4 powder was added, and the fracture toughness increased to 3.1 ± 0.3 MPa m 1/2  at 30 wt% of the β-Si 3 N 4 powder addition. The shape of the introduced β-Si 3 N 4 particles were believed to be faveriable for uniform dispersion in the printing slurry without agglomeration. In the following pyrolysis treament the β-Si 3 N 4 powders promoted the release of gaseous products so that maintained the structure integrety of the printed parts and the rod-like β-Si 3 N 4 particles enhanced the energy dissipation during ceramic fracture. In this study, silicon-based composite of β-Si 3 N 4 /SiCN ceramics with excellent mechanical properties were prepared by DLP additive manufacturing technology, which confirmed the feasibility and the great potential of photo-curing additive manufacturing technology in the manufacturing of SiCN ceramic matrix composites with complex structures.

Mechanism of β-Catenin in Pulmonary Fibrosis Following SARS-CoV-2 Infection

Min Jiang,Jiaqi Hou,Qianqian Chai,Shihao Yin,Qian Liu

Pulmonary fibrosis due to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is the leading cause of death in patients with COVID-19. β-catenin, a key molecule in the Wnt/β-catenin signaling pathway, has been shown to be involved in the development of pulmonary fibrosis (e.g., idiopathic pulmonary fibrosis, silicosis). In this study, we developed a SARS-CoV-2-infected A549-hACE2 cell model to evaluate the efficacy of the A549-hACE2 monoclonal cell line against SARS-CoV-2 infection. The A549-hACE2 cells were then subjected to either knockdown or overexpression of the effector β-catenin, and the modified cells were subsequently infected with SARS-CoV-2. Additionally, we employed transcriptomics and raw letter analysis approaches to investigate other potential effects of β-catenin on SARS-CoV-2 infection. We successfully established a model of cellular fibrosis induced by SARS-CoV-2 infection in lung-derived cells. This model can be utilized to investigate the molecular biological mechanisms and cellular signaling pathways associated with virus-induced lung fibrosis. The results of our mechanistic studies indicate that β-catenin plays a significant role in lung fibrosis resulting from SARS-CoV-2 infection. Furthermore, the inhibition of β-catenin mitigated the accumulation of mesenchymal stroma in A549-hACE2 cells. Additionally, β-catenin knockdown was found to facilitate multi-pathway crosstalk following SARS-CoV-2 infection. The fact that β-catenin overexpression did not exacerbate cellular fibrosis may be attributed to the activation of PPP2R2B.

Oral Administration of Berberine Hydrochloride Based on Chitosan/Carboxymethyl-β-Cyclodextrin Hydrogel

Bukatuka Futila Clemence,Lin Xiao,Guang Yang

In this study, a novel oral formulation of berberine hydrochloride (BBH) hydrogel was successfully synthesized through physical cross-linking using chitosan (CS) and carboxymethyl-β-cyclodextrin (CMCD). The characterization results confirmed the successful synthesis of the CS/CMCD hydrogel and the subsequent loading of BBH into this composite (CS/CMCD/BBH) was effectively accomplished. The BBH was used as a model drug and the resulting hydrogel demonstrated a sustained drug release profile. In addition to its improved solubility and sustained release characteristics, the hydrogel exhibited excellent antibacterial activity against common pathogens such asEscherichia coli(E. coli),Staphylococcus aureus(S. aureus), andCandida albicans(C. albicans). Additionally, in vitro studies indicated that the hydrogel was not cytotoxic to NIH3T3 and HaCaT cells, suggesting its safety for biomedical applications. This lack of cytotoxic effects, combined with the mechanical strength, solubility improvements, and antibacterial properties of the hydrogel, positions the CS/CMCD/BBH hydrogel as a promising candidate for the effective oral delivery of BBH. By addressing the solubility and delivery challenges of BBH, this hydrogel offers a viable solution for the oral administration of BBH, with potential applications in various biomedical fields.

Studies on the Inclusion Complexes of Daidzein with β-Cyclodextrin and Derivatives

Shujing Li,Li Yuan,Yong Chen,Wei Zhou,Xinrui Wang

The inclusion complexes between daidzein and three cyclodextrins (CDs), namely β-cyclodextrin (β-CD), methyl-β-cyclodextrin (Me-β-CD, DS = 12.5) and (2-hydroxy)propyl-β-cyclodextrin (HP-β-CD, DS = 4.2) were prepared. The effects of the inclusion behavior of daidzein with three kinds of cyclodextrins were investigated in both solution and solid state by methods of phase-solubility, XRD, DSC, SEM,1H-NMR and 2D ROESY methods. Furthermore, the antioxidant activities of daidzein and daidzein-CDs inclusion complexes were determined by the 1,1-diphenyl-2-picryl-hydrazyl (DPPH) method. The results showed that daidzein formed a 1:1 stoichiometric inclusion complex with β-CD, Me-β-CD and HP-β-CD. The results also showed that the solubility of daidzein was improved after encapsulating by CDs.1H-NMR and 2D ROESY analyses show that the B ring of daidzein was the part of the molecule that was most likely inserted into the cavity of CDs, thus forming an inclusion complex. Antioxidant activity studies showed that the antioxidant performance of the inclusion complexes was enhanced in comparison to the native daidzein. It could be a potentially promising way to develop a new formulation of daidzein for herbal medicine or healthcare products. Keywords:daidzein;cyclodextrin;inclusion complex;antioxidant activity

Engineered cellulosic triboelectric materials for multi-modal sensing via β-phase programming

Ling-Zhi Huang,Yan Ding,Dan-Dan Li,Ming-Guo Ma

Cellulosic triboelectric materials have gained widespread attention in triboelectric nanogenerators (TENGs) recently due to their fascinating merits of renewability, biodegradability, and low cost. However, cellulose-based TENGs still are challenged by the low surface charge density of triboelectric materials. Herein, the negative charged cellulosic triboelectric materials are functionalized as triboelectric layers for self-powered sensors via an electrostatic spinning method, where the electrospun polyvinylidene fluoride (PVDF) nanofibers fill with the porous air-laid paper (AP), boosting the formation of polar β-phase crystals and a negative shift of the surface potential. The TENG constructed with PVDF/12 wt%@AP affords charge output of 60.7 μC m −2 and power output of 233.28 mW m −2 , and the as-prepared contact self-sensor presents unparalleled sensitivity of 6.20 V kPa −1 under pressures from 1.23 kPa to 5.0 kPa. In addition, we design the non-contact self-powered sensor, capable of recognizing different objects and accurately detecting human movements like walking, running, and jumping. Notably, the TENGs can also be used as a wireless sensor terminal for smart devices to provide convenient sensing functions. This β-phase cellulosic materials are a step toward wearable self-powered sensor systems that are durable to capture mechanical energy from the environment.

Engineering of β-carotene hydroxylase for enhanced astaxanthin production in Saccharomyces cerevisiae

Jiayan Du,Youtong Bao,Jingyuan Zhu,Xueqing Pang,Depeng Ren,Xinjian Yin,Pingping Zhou

Astaxanthin, a potent antioxidant widely applied in health supplements, cosmetics, and animal feed, faces production challenges in microbial synthesis due to inefficient β-carotene hydroxylase activity. This study identified rate-limiting enzymes and demonstrated that the expressing phytoene desaturase BtCrtI from Blakeslea trispora in Saccharomyces cerevisiae increased β-carotene production 2.8-fold compared to XdCrtI from Xanthophyllomyces dendrorhous . Integrating of XdCrtYB, BtCrtI, CrtE03M, and tHMG1 achieved 208.7 mg/L β-carotene. However, introducing β-carotene ketolase and β-carotene hydroxylase into this strain unexpectedly increased lycopene accumulation, likely due to feedback inhibition by the product and enzyme imbalance. To address this, we utilized a moderate β-carotene producer YJYast-02 for astaxanthin production. Truncating the N-terminal 39 amino acids of β-carotene hydroxylase from Haematococcus pluvialis improved astaxanthin titer by 37.7 %. Rational engineering of the hydrophobicity of the substrate-binding pocket in β-carotene hydroxylase, specifically through mutations such as N183A, C191A and T213A, further increased astaxanthin titer by 15.8 %, 30.5 %, 36.8 %, respectively. The final engineered strain, YJYast-03(t5t39 T213A ), expressing t5OBKTM30 and t39OcrtZM1 T213A , achieved 26.0 mg/L (3.6 mg/g DCW) astaxanthin. This work pioneers the use of molecular docking for engineering β-carotene hydroxylase, offering potential to significantly enhance catalytic activity and optimize astaxanthin biosynthesis in yeast.

Amyloid-β Oligomer-Induced Electrophysiological Mechanisms and Electrical Impedance Changes in Neurons

Shimeng Sun,Qing Ma,Qiyu Sheng,Shangwei Huang,Chenxia Wu,Junsong Liu,Jia Xu

Amyloid plays a critical role in the pathogenesis of Alzheimer’s disease (AD) and can aggregate to form oligomers and fibrils in the brain. There is increasing evidence that highly toxic amyloid-β oligomers (AβOs) lead to tau protein aggregation, hyperphosphorylation, neuroinflammation, neuronal loss, synaptic loss, and dysfunction. Although the effects of AβOs on neurons have been investigated using conventional biochemical experiments, there are no established criteria for electrical evaluation. To this end, we explored electrophysiological changes in mouse hippocampal neurons (HT22) following exposure to AβOs and/or naringenin (Nar, a flavonoid compound) using electrical impedance spectroscopy (EIS). AβO-induced HT22 showed a decreased impedance amplitude and increased phase angle, and the addition of Nar reversed these changes. The characteristic frequency was markedly increased with AβO exposure, which was also reversed by Nar. The AβOs decreased intranuclear and cytoplasmic resistance and increased nucleus resistance and extracellular capacitance. Overall, the innovative construction of the eight-element CPE-equivalent circuit model further reflects that the pseudo-capacitance of the cell membrane and cell nucleus was increased in the AβO-induced group. This study conclusively revealed that AβOs induce cytotoxic effects by disrupting the resistance characteristics of unit membranes. The results further support that EIS is an effective technique for evaluating AβO-induced neuronal damage and microscopic electrical distinctions in the sub-microscopic structure of reactive cells.

Anti-inflammatory peptide-modified β-TCP scaffolds enhance the hyperglycemic inflammatory microenvironment

Delong Niu,Wenjie Qiu,Jiaqi Ding,Haoyu Feng,Zhen Zhang,Wenjuan Zhou,Zhonghao Liu

Chronic hyperglycemia in diabetic patients induces a sustained systemic inflammatory microenvironment that critically impairs bone regeneration and delays healing. In this study, we developed a β-tricalcium phosphate (β-TCP) scaffold functionalized with the anti-inflammatory peptide KAFAKAARLYRKALARQLGVAA (KAFAK), aiming to regulate the local hyperglycemic inflammatory milieu and enhance bone regeneration. Polydopamine (PDA) was applied as a bioadhesive coating to facilitate the efficient immobilization of KAFAK on the scaffold surface. The physicochemical properties and biocompatibility of the modified scaffolds were systematically characterized. A hyperglycemic inflammatory model was established using lipopolysaccharide (LPS) and glucose to simulate diabetic conditions. In this model, KAFAK-functionalized β-TCP scaffolds significantly downregulated the expression of key pro-inflammatory cytokines, including IL-6, TNF-α, and IL-1β, in RAW264.7 macrophages. Furthermore, the scaffold effectively modulated macrophage behavior, exhibiting notable immunomodulatory and anti-inflammatory activity. Overall, this study demonstrates that KAFAK-functionalized β-TCP scaffolds hold strong potential for bone regeneration under hyperglycemic inflammatory conditions and offer a novel strategy for immunomodulatory therapy in metabolic bone disorders.

Solvothermal Growth of Chiral β-In2S3 Nanosheet Arrays for Circularly Polarized Photodetection

Zhou Sheng,Gaoyu Chen,Xueyan Hu,Yue Chen,Dongdong Xu,Caojin Yuan,Xiangxing Xu

The circularly polarized state of light can carry encoding information through polarization modulation and structural information from the emitter or propagation medium. Therefore, the detection of the polarization state of light is a central function in various applications. One of the concise strategies is to transform left- and right-handed circularly polarized light (CPL) directly into distinguishable electronic signals using chiral structured materials. In this study, chiral films of β-In2S3 vertical nanosheet arrays are successfully grown using a facile solvothermal route. Chirality originates from the distortion of nanosheets with left or right-handedness controlled by the introduction of l-/d-cysteine as chiral inducers. Circularly polarized photodetectors based on chiral β-In2S3 films are fabricated. They exhibit good performances with a responsivity (R) of 8.0 A/W, photodetectivity (D*) of 5.0 × 1012 Jones, the photocurrent dissymmetry factor (gIph) of 0.34, and external quantum efficiency (EQE) of 2.2 × 103 (%) for a CPL of 450 nm.

β-Hydroxybutyric Acid-Induced Bovine Endometrial Epithelial Cell Pyroptosis via Activating P2X7R

Jiajia Mu,Qiannan Sun,Nanyan Lin,Chenguang Yue,Jiaqiao Zhu,Hui Zou,Hongyan Zhao,Ruilong Song,Zongping Liu,Yonggang Ma

Ketosis in dairy cows is linked to endometritis, but the molecular mechanisms are unclear. We investigated whether P2X7R is crucial in ketosis-induced endometritis. Ketotic cows had higher levels of NLRP3, caspase-1, GSDMD, GSDME, and P2X7R in the uterine tissue. β-Hydroxybutyric acid (BHBA) exposure in bovine endometrial epithelial cells induced pyroptosis, increased pyroptosis protein and gene expression, and raised pyroptosis rates. P2X7R levels rose with BHBA, and BHBA also increased the level of extracellular ATP. Inhibitors of NLRP3, caspase-1, caspase-3, and P2X7R reduced BHBA-induced pyroptosis. P2X7R inhibition suppressed NLRP3, caspase-1, and caspase-3 activation, preventing pyroptosis. In P2rx7–/– mice with a ketosis model, blood BHBA was lower, and uterine tissue showed less pyroptosis-related protein expression compared to ketotic mice. P2X7R appears to regulate BHBA-induced pyroptosis through NLRP3 and caspase-3 signaling, suggesting it is a potential therapeutic target.

β-Cyclodextrin enabled ultrahigh-permeance loose nanofiltration membrane for efficient dye separation

Changsheng Guo,Yanli Zhao,Zhaoyi Zhang,Zhichen Liang,Hui Yu,Yao Qian,Qi Zuo,Pengbi Liu,Mengchen Zhang,Zhiwei Xu,Xianhua Zeng

Traditional loose nanofiltration (LNF) membranes show great advantages in dyeing wastewater treatment, however, their applications are limited by the “trade-off” effect. β-Cyclodextrins (β-CD) demonstrate excellent hydrophilicity and hydraulic conductivity owing to the existence of the hydrophilic ends and hydrophobic inner lumen. This study reports an LNF membrane with outstanding permeance and dye rejection capabilities achieved through the immobilization of β-CD within the pores of the substrate membrane via the thermal shrinkage of sulfonated polysulfone and the electrostatic synergy of polydopamine. The introduction of β-CD and PDA changed the surface potential, hydrophilicity, and pore structure of the membrane. Furthermore, it displays specific recognition with dyes, thereby further enhancing both permeance and rejection rates. As anticipated, both permeance and dye rejection (Methylene Blue) increased to remarkable levels, reaching up to 225 L·m −2 ·h −1 ·bar −1 and 97.8 %, respectively. These values far exceeded those achieved by conventional polymeric NF membranes or hybrid NF membranes. Besides, the permance and dye rejection didn't change much with the increase of the dye feed concentration, salt concentrations or under different pH conditions. Furthermore, both permeance and rejection rates remained stable during the cyclic filtration tests, with stability further enhanced in deionized water at 40 °C. These findings highlight the potential of strategies aimed at modifying membrane pore transport channels for applications in dye separation, particularly in treating printing and dyeing wastewater.
