Autoimmunity remains challenging to treat without broad immunosuppression. We previously showed that anti-CD38 antibody can rapidly elevate platelet counts in refractory immune thrombocytopenia (ITP), but the underlying mechanism was unclear. Here we report that anti-CD38 antibody induces platelet recovery within 3 days, including after retreatment in relapsed cases. Mechanistically, CD38-mediated nicotinamide adenine dinucleotide (NAD + ) depletion drives M1-like macrophage polarization with increased Fc gamma receptor I (FcγRI) expression, thereby promoting macrophage phagocytosis of opsonized platelets. In mice, CD38 inhibition or nicotinamide mononucleotide (NMN) supplementation restores NAD + , reprograms macrophages, downregulates FcγRI and prevents thrombocytopenia. In an ovalbumin immunization model, NMN treatment does not impair antigen-specific antibody production, supporting preservation of humoral responses. Based on these findings, we conducted a single-arm, open-label phase 1/2 trial of low-dose oral NMN (450 mg twice daily for 2 weeks) in adults with steroid-refractory or steroid-dependent ITP. Primary endpoints were safety/tolerability and platelet response (≥50 × 10 9 per liter within 2 weeks, confirmed by two consecutive measurements one or more days apart, without rescue therapy or dose escalation of thrombopoietin receptor agonists or corticosteroids). Among 25 enrolled patients, no dose-limiting toxicities or treatment-related serious adverse events occurred; NMN was well tolerated, with only mild treatment-related adverse events in 12% and non-severe infections (grade 1) in 8% of patients while immunoglobulin levels remained stable, consistent with preserved humoral immunity. Five patients (20.0%) met the primary platelet-response endpoint. In exploratory analyses, overall, 60% of patients achieved platelet counts more than 1.5× baseline during treatment, and 52% maintained responses through week 8. Together, these data identify the CD38−NAD + axis as an immunometabolic checkpoint in ITP and support further exploration of NMN as a non-antibody-depleting metabolic strategy for antibody-mediated disease. Selective hydrogenation of cinnamaldehyde (CAL) to cinnamyl alcohol (COL) presents a significant challenge owing to the thermodynamically unfavorable hydrogenation of the carbonyl (C═O) over the olefinic (C═C) bond. Herein, we report the rational design of silicate-1 (S-1) anchored PtCo catalysts (PtCo@S-1) synthesized via a sequential hydrothermal-impregnation method. Notably, the 4Pt1Co@S-1 catalyst exhibited superior catalytic performance, achieving a high COL selectivity of 92% at a near-complete CAL conversion of 98.2%. Furthermore, the catalyst exhibits robust stability with only slight fluctuations after five cycle tests. Beyond CAL hydrogenation, the developed catalyst is also applicable to the chemo-selectivity hydrogenation of other α, β-unsaturated aldehydes to the corresponding α, β-unsaturated alcohols during the substrate-extending hydrogenation tests. Experiments and theoretical calculations reveals the presence of CoSiO 2 species critically modifies the geometric and electronic structure of the Pt metallic active sites. This modification promotes the preferential adsorption and activation of the C═O bond, thereby significantly enhancing selectivity towards COL.

DNA serves as the primary target for numerous metal complexes in suppression tumor cell proliferation, and Poly ADP ribose polymerase-1 (PARP-1) is critical for DNA repair. In this study, benzothiazole hydrazone was used as a framework to design and synthesize a series of Cu(II) complexes (Cu1-Cu3), which functions as novel PARP-1 inhibitors. These Cu(II) complexes not only effectively inhibited the proliferation of a panel of tumor cells (IC 50 range: 0.51–5.37 μM) but also exhibited potent PARP-1 inhibitory activity (IC 50 range: 0.45–1.52 μM). Notably, complex Cu3 had the most potent activities, exhibiting strong cytotoxicity against T98G cells (IC 50 = 0.51 μM) and significant PARP-1 inhibition (IC 50 = 0.45 μM). Additionally, Cu3 effectively inhibited the migration of T98G cells. Mechanistic studies revealed that Cu3 generated excessive reactive oxygen species (ROS), induced DNA damage, exacerbated DNA double-strand breaks by inhibiting PARP-1 activity, and promoted apoptosis in tumor cells via the mitochondrial pathway.

Selective hydrogenation reactions over oxide catalysts are crucial in a variety of energy and catalytic conversion processes, in which surface hydrogen species are vital while controllable H2 activation and hydrogenation are still challenging over the complex oxide catalysts. Here, combining multiscale theoretical simulations and surface science studies, we find a series of single-site Zn (Zn1) species that are well orderly dispersed onto spinel ZnCr2O4 surfaces under syngas conversion conditions. A pair of coordinatively unsaturated Zn1 sites with specific spatial proximity (termed geminal-Zn1+1) facilitates H2 homolysis into hydrides, which even occurs below 123 K. Intriguingly, the geminal-Zn1+1 confined on ZnCr2O4(110) can stabilize both hydride species (as high as 673 K) and related formaldehyde intermediates, which enables controllable CO hydrogenation to ketene via a synergistic H- and CO-assisted CO activation mechanism instead of deep hydrogenation to methanol. Such coordinatively unsaturated geminal-atom (M1+1) confined by the oxide surface is expected to be generalized to more application scenarios of oxide-catalyzed H2 homolysis and selective hydrogenation catalysis.

Parkinson's disease (PD) is a progressive neurodegenerative disorder with no available disease-modifying therapies. Given their diverse health-promoting properties, probiotics have emerged as a potential neuroprotective strategy. Through screening in transgenic Caenorhabditis elegans ( C. elegans ) models of α-synuclein (α-syn)-induced PD, we identified the Escherichia coli ( E. coli ) strain E8 as a candidate that effectively suppressed dendritic degeneration and motor deficits. These neuroprotective improvements were attributed to the reduction of α-syn aggregation, which was mechanistically associated with E8's ability to promote autophagy. Metabolomic profiling further pinpointed 1-allylpiperazine (1-ALPP) as a bioactive metabolite responsible for rescuing PD-related impairments, which was sufficient to recapitulate the autophagy activation induced by E8. Subsequent mechanistic studies revealed that 1-ALPP activated autophagy through the AMPK/mTOR/HLH-30 pathway. Notably, 1-ALPP effectively inhibited α-syn fibrillogenesis and cytotoxicity in a dose-dependent manner, potentially via the inhibition of α-syn liquid-liquid phase separation (LLPS). Molecular simulations demonstrated that 1-ALPP binds to α-syn, thereby preventing the critical conformational changes required for fibril formation. Strikingly, the protective effects of E8 against PD phenotypes were intergenerationally inherited, persisting for two generations in an hpl-2 -dependent manner.

The interaction between 1-(2-hydroxyphenyl)piperazine (HPP) and cucurbit[6]uril (Q[6]) has been explored via 1 H NMR spectroscopy and X-ray crystallography. The results show that the guest molecule 1-(2-Hydroxyphenyl)piperazine hydrochloride is not encapsulated within the Q[6] cavity in both aqueous solution and the solid state. Instead, it resides at the portal of Q[6], forming a highly stable exclusion complex, designated HPP 2 @Q[6]. Notably, ion-dipole and hydrogen-bonding interactions play key roles in stabilizing this complex.

This study describes a monolithic silicalite-1 catalyst support designed for tritium oxidation reactions under humid conditions. Monolithic molecular sieves (sil-s) were fabricated by converting silica binders to silicalite-1 through secondary crystallization (175 °C, 24 h). In addition to the binder conversion to silicalite-1, some recrystallization of starting silicalite-1 (sil) results in higher crystallinity, lower concentration of silanol defects, and higher hydrophobicity. With the addition of 2% platinum, Pt/sil-s exhibited better stability under humid conditions, showing only 0.01%/min conversion decay over 800 min. This work has demonstrated a moisture-resistant Pt catalyst for tritium oxidation in fusion energy systems.

Copper(Cu)-based catalysts are highly promising candidates for ethanol dehydrogenation owing to their exceptional activity in cleaving O-H and C-H bonds. Nevertheless, their widespread application is hindered by challenges such as rapid deactivation and complex preparation procedures. Herein, inspired by the deactivation mechanism of Cu/MFI (silicalite-1) catalysts, two strategies, i.e., using mixed solvents for impregnation and enriching silanol nests of MFI to anchor Cu particles, were used to synthesize Cu/MFI-R catalysts with high stability and activity. The mixed solvent approach reduces the surface tension of solvents, enhancing the uniform dispersion of Cu over the hydrophobic surface of MFI. Subsequently, a MFI-R support with abundant silanol nests was synthesized via optimizing hydrothermal conditions, as confirmed by reaction results and characterizations of FT-IR, UV-Vis and solid NMR. By combining these strategies, Cu/MFI-R catalysts were synthesized for ethanol dehydrogenation, achieving > 90 % ethanol conversion and 95 % acetaldehyde selectivity over the 600 h time on stream test. This work provides a very promising scalable approach for the design of stable Cu-based catalysts in hydrogen-related reactions.

TS-1 zeolite demonstrates notable activity and selectivity in cycloolefin epoxidation when employing H₂O₂ as the oxidant. However, the practical application of conventional TS-1 is significantly limited by factors including surface hydrophilic structures and mass transfer constraints. To address these challenges, a carbon-encapsulated hierarchical TS-1 (3DC_TS-1) has been synthesized through the confined growth approach, in which 3DOm carbon is used as a confined space provider for TS-1 crystallization and further acts as a hydrophobic wrapper to modulate the surface microenvironment of TS-1 crystals. In the epoxidation of cyclopentene and cyclohexene with using H₂O₂ as the oxidant, the hydrophobic 3DC_TS-1 catalyst exhibits a superior selectivity to epoxide products by suppressing epoxide hydrolysis and allyl oxidation side reactions, comparing with the 3D_TS-1 catalyst without carbon wrapper and the conventionally microporous TS-1 catalyst. It is expected that this study will provide an effective strategy for designing zeolite catalysts applying to selective oxidation reactions.

Two-dimensional (2D) 2H-1 T′ heterojunctions based on transition-metal dichalcogenides have attracted great attention due to their special interface structures and novel properties. Understanding the growth mechanism and the interface structures is essential for their controllable growth. In this work, we successfully synthesize lateral and vertical MoS 2 /ReS 2 heterojunctions by using a two-step chemical vapor deposition (CVD) method, and deeply explore the evolution of the structure. It is found that the growth mode of heterojunction undergoes a transition from lateral epitaxy to vertical stacking under a high Re/S ratio during the growth. Furthermore, the interface structure is greatly determined by the edge shape of MoS 2 core. Two kinds of structures, with the b-axis of 1 T′ ReS 2 shell parallel to the (1 0 0) and (0 1 0) plane of MoS 2 core respectively, are observed. Meanwhile, by comparing with MoS 2 monolayer, MoS 2 /ReS 2 heterostructures possess a reduced emission intensity due to the carrier transfer. This work provides a robust and controllable strategy for the synthesis of heterostructures with different phase structures.

Introduction 3D-printing implants have attracted increasing attention for enhancing the regeneration of various wound types. The rigidity of traditional 3D printing materials limits their applicability in skin wound repair. In addition, improving the bioactivity of polymeric materials remains an urgent challenge to be addressed. Insulin-like growth factor-1 (IGF-1) is widely recognized for its efficacy in skin wound repair. However, the potency of IGF-1 in wound regeneration is limited by its unstable binding to the implants. Methods In this study, A phase inversion-based deposition modeling 3D printing technique was employed to construct flexible poly(lactic-co-glycolic acid) (PLGA) scaffold. Additionally, a novel 3D-printing flexible skin implantation functionalized with bioorthogonal IGF-1 (DA-IGF-1) was fabricated. The adhesion, proliferation and relative mRNA expression of cells on PLGA scaffold were assessed. Meanwhile, a full-thickness skin defect regeneration model was established on the rat back to evaluate the wound repair efficacy of the implanted scaffolds. Results The DA-IGF-1 exhibited outstanding binding ability on the scaffold. The resulting PLGA flexible scaffold modified with DA-IGF-1 showed significantly enhanced hydrophilicity and biocompatibility compared with the control group materials ( p < 0.05). The expression of wound regeneration related genes was also upregulated significantly in the DA-IGF-1 group ( p < 0.05). In vivo study indicated that bioactive implants effectively promoted healing and neovascularization of full-thickness skin defects in rats. Conclusion These results demonstrated that the 3D-printing flexible PLGA scaffold modified with bioorthogonal IGF-1 holds great potential for application as a novel therapeutic strategy for wound regeneration.

Electrochemical synthesis of polymer/MOF composite membranes holds great potential for practical applications, yet the preparation of flexible and dense MOF layers remains a challenge, which is crucial for achieving practicality. Herein, a novel ultra-flexible polyethersulfone (PES)/Cu-based metal–organic framework (HKUST-1) composite membrane has been fabricated via an electrochemical synthesis method. Various reaction conditions, including voltage, stirring speed, and reaction time, were systematically investigated to elucidate their influence on crystal growth and the resulting membrane uniformity. The characterization results showed that high voltage and moderate stirring as well as are conducive to obtaining uniform fine crystals. Additionally, a relatively extended reaction time is advantageous for achieving a more continuous and denser MOFs membrane with outstanding flexibility. The optimized membrane possesses an excellent separation flux (2.81 × 10 3 ±32 L m −2 h −1 ·bar −1 ) and efficiency (99.7 ± 0.3%) for n -hexane/water mixtures, as well as excellent photodegradation efficiency (>99%) for methylene blue (MB). Remarkably, the optimized membrane exhibits remarkable scalability and stability, maintaining superior separation performance even after being rolled up 10 times with a curvature of 200 m −1 . This suggests that it has great potential for practical applications in complex environments.
