A Common Indoor Pollutant, 1,4-Dichlorobenzene, Induces Colonic Inflammation via Vitamin D Receptor-Reg3γ-Gut Microbiota Axis

Chaoyu Tong,Yang Song,Daniel Schlenk,Jing Liu

1,4-Dichlorobenzene (1,4-DCB), a widely used household insect repellent and fungicide, has been epidemiologically linked to ulcerative colitis, yet its potential mechanisms remain poorly understood. In this study, juvenile male mice developed colonic inflammation following exposure to 1,4-DCB at doses of 0.03–3 mg/kg/day, a range corresponding to the human estimated daily intake (EDI) and relevant internal exposure levels in adolescents. This was evidenced by increased fecal moisture (by 12.22%–18.21%) and colon weight (17.57%–38.33%), shortened colon length (13.51%–24.88%), crypt expansion, and connective tissue hyperplasia. 1,4-DCB exposure upregulated the colonic expression of regenerating family member 3 gamma (Reg3γ), a key antimicrobial peptide that targets Gram-positive bacteria. This upregulation was accompanied by gut microbiota dysbiosis, elevated colonic levels of lipopolysaccharide (LPS) and pro-inflammatory cytokines, as well as decreased levels of tight junction proteins. Microbiota depletion alleviated these colitis symptoms induced by 1,4-DCB. In human colonic cells, we further validated LPS-induced inflammation and impaired permeability as well as identified vitamin D receptor (VDR) as a potential primary molecular target for 1,4-DCB. Overall, this study provides the first evidence that environmentally relevant levels of 1,4-DCB induce colonic inflammation through the VDR-Reg3γ-gut microbiota axis. These findings highlight the potential colonic health risk posed by this prevalent indoor pollutant.

High Molecular Weight α-Galactosidase from the Novel Strain Aspergillus sp. D-23 and Its Hydrolysis Performance

Ke Chen,Jingyun Zhang,Xing Liu,Peiyuan Zhang,Wenlong Yue,Zhiqiang Cai

Aspergillussp. D-23 was obtained by ultraviolet-diethyl sulfate (UV-DES) compound mutagenesis fromAspergillussp. C18 that the α-galactosidase was purified from. According to sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and non-denaturing polyacrylamide gel electrophoresis (Native-PAGE), the purified enzyme demonstrated apparent homogeneity. The monomeric α-galactosidase’s native molecular weight was 125 kDa. The optimal temperature of α-galactosidase was 65 °C, and 75% of the initial enzyme activity could be maintained between 45 and 55 °C. Its optimal pH was 5.0 with good pH stability. After incubating for 2 h at pH 3.0–8.0, it could retain more than 80% of its original activity. Different concentrations of metal ions had different effects on the α-galactosidase activity. High concentrations of Cu2+could strongly inhibit enzyme activity and low concentrations of Fe2+could promote enzyme activity. Additionally, as shown by thin layer chromatography and high-performance liquid chromatography, the enzyme also had good hydrolysis ability, which could efficiently hydrolyze melibiose and raffinose by more than 95%. Therefore, these excellent characteristics could make α-galactosidase a good candidate for the food and feed industries.Keywords:α-galactosidase;enzymatic characterization;Aspergillussp.;enzymatic hydrolysis;higher molecular weight

Defect-Assisted High Anion Conductivity in Diethyldimethylammonium d-Camphorsulfonate Plastic Crystal: A Size Effect of Target Ions

Xue Lan,Wenjing Sun,Xin Wang,Wei Xu,Yoji Horii,Jiandang Liu,Hongjun Zhang,Bangjiao Ye,Tiancheng Mu,Xiaozheng Lan

We demonstrate anion (chloride and fluoride ions) transfer in an organic ionic plastic crystal (OIPC), diethyldimethylammonium d-camphorsulfonate, and the role of defects in ion conduction. The phase transitions, crystalline structures, dynamics, and ion transfer mechanisms of the pure material and the doped mixtures were investigated using a combination of differential scanning calorimetry, X-ray diffraction, solid-state nuclear magnetic resonance spectroscopy, and electrochemical impedance spectroscopy. The doped mixtures show minor modifications in thermal behaviors and solid phase structures to the host material. The ion mobility of the pure material in the plastic phase was assigned mainly to cations. The fluoride salt-doped mixture has drastically enhanced conductivity at all tested temperatures, and the chloride salt-doped mixture displays a strong temperature-dependent behavior. Ionic conductivity measurements suggest transfer mechanisms through crystalline phases. The pure material and doped mixtures exhibit similar defect volumes and concentrations, and both factors are phase-dependent, as determined using positron annihilation lifetime spectroscopy. The conductivity displays dependence on not only the defect volume but also the target ion volume. The relationships between the defect volume and the conductivity qualitatively follow the Cohen–Turnbull free volume model, while critical volumes were very large. For the first time in OIPCs, the size of the target ions was found to significantly influence ionic conductivity.

Pellets of phospholipids and d-glucose with improved intestinal absorption and oral bioavailability of salvianolic acid B

Ziyi Li,Zhenghua Li,Jianhua He,Jianping Liu

Salvianolic acid B (SAB) is a widely used cardioprotective agent, while its clinical application was limited by poor intestinal absorption and low oral bioavailability. In this study, SAB phospholipid (SP) complex was first prepared to improve the lipophilicity of SAB and then combined with d-glucose to further enhance intestinal absorption. Compared with free SAB, SP or the mixture of SAB and d-glucose, combination of SP and d-glucose showed higher intestinal absorption evidenced by increased effective permeation coefficient (Peff) in the in situ single-pass intestinal perfusion (SPIP) assay. Subsequently, SP and d-glucose at mass ratio of 1:6, with the highest Peff of SAB, were chosen for the preparation of complexed pellets to improve oral absorption efficiency of SAB. As expected, the obtained pellets significantly enhanced oral bioavailability of SAB in the pharmacokinetic study characterized by increasing Cmax and AUC0–t of SAB by 14.88-fold and 5.02-fold than free SAB, respectively. In conclusion, combination of d-glucose in SP pellets can effectively improve the intestinal absorption and oral bioavailability of SAB.

γ-Glutamylcysteine ameliorates D-gal-induced senescence in PC12 cells and mice via activating AMPK and SIRT1

Shuai Lu,Jinyi Zhou,Chen Yang,Xiaoxue Zhang,Yingying Shi,Jie Liu,Xintong Yan,Juanjuan Liang,Xianli Liu,Lan Luo,Da Zhou,Zhimin Yin

Aging is a natural process accompanied by inflammation and oxidative stress and is closely associated with age-related diseases. As a direct precursor of glutathione, γ-glutamylcysteine (γ-GC) possesses antioxidant and anti-inflammatory properties; however, whether γ-GC plays an important role in anti-aging remains unknown. Here, we investigated the protective effects and mechanisms of γ-GC in D-galactose (D-gal)-induced senescence in PC12 cells and aging mice. Our results showed that γ-GC treatment significantly reduced the percentage of senescence-associated-β-galactosidase (SA-β-Gal)-positive cells and inhibited D-gal-induced cell cycle arrest in PC12 cells. The results of Nissl and hematoxylin and eosin (H&E) staining in mouse brain showed that γ-GC treatment markedly reversed the damage in the hippocampus of D-gal-induced aging mice. Moreover, γ-GC increased the phosphorylation of AMP-activated protein kinase (AMPK) to promote the nuclear translocation of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) while inhibiting the nuclear translocation of deleted in breast cancer 1 (DBC1), which leads to the activation of sirtuin 1 (SIRT1) and deacetylation of p53 in the nucleus. Therefore, γ-GC may be a potential therapeutic candidate compound for the prevention and treatment of age-related diseases.

Combing with redox regulation via quorum-sensing system and fermentation strategies for improving D-pantothenic acid production

Hai-Bin Qin,Jun-Ping Zhou,Bo Zhang,Zhi-Qiang Liu,Yu-Guo Zheng

Microbial production of D-pantothenic acid (D-PA) has become an important research area, as D-PA is an essential vitamin widely applied in food, feed, chemical and pharmaceutical industries. Here both dynamic regulations of redox metabolism and fermentation strategies were applied to enhancing D-PA biosynthesis in Escherichia coli W3110. Firstly, a quorum-sensing (QS) system for redox regulation developed, in which zwf and nadK overexpression were shown to be effective for enhancing D-PA production. Based on this, a strain DPA11/p LBCWK was constructed with D-PA titer at 4.19 g/L in flask. Moreover, the fermentation strategies including pH-feedback feeding and gradient β-alanine feeding were introduced, under optimal conditions, the strain DPA11/p LBCWK could produce 48.21 g/L D-PA in 5 L fermenter, with the specific productivity of 0.72 g/L·h and the yield of 0.33 g/g glucose. These modification strategies were proved effective and would provide valuable reference for biosynthesis of other chemicals.

Cloning and functional analysis of 1-deoxy-d-xylulose-5-phosphate synthase (DXS) in Santalum album L

Xiaohong Chen,Yueya Zhang,Haifeng Yan,Meiyun Niu,Yuping Xiong,Xinhua Zhang,Yuan Li,Jaime A. Teixeira da Silva,Guohua Ma

The commercial value of Santalum album L. lies in its aromatic heartwood and essential oil. Sesquiterpenes are the main components of sandal essential oil, and these are synthesized through the plant’s mevalonate (MVA) and methylerythritol phosphate (MEP) pathways. In this study, the first key rate-limiting enzyme, 1-deoxy- d -xylulose-5-phosphate synthase (SaDXS), was investigated to provide a theoretical molecular basis for the sandalwood MEP sesquiterpene biosynthetic pathway. The biofunctions of SaDXS were also analyzed. SaDXS promoters were successfully cloned from a seven-year-old S. album tree. SaDXS1A/1B promoter activity was verified by a β-glucuronidase (GUS) assay and by analyzing cis -acting elements of the promoters, which carried light- and methyl jasmonate (MeJA)-responsive signals. In an experiment involving yellow S. album seedlings, exposure to light upregulated SaDXS1A/1B expression and increased chlorophyll and carotenoid contents when overexpressed in Arabidopsis thaliana . Analysis of the expression of SaDXS1A/1B and SaSSy , key genes of santalol biosynthesis, revealed SaDXS1A expression in all tissues whereas SaDXS1B was expressed in tissues that contained photosynthetic pigments, such as stems, leaves and flowers. Sandal seedlings exogenously treated with two hormones, MeJA and ethylene, revealed similar expression patterns for SaDXS1A/1B and SaSSy . Sandal seedlings were treated with an inhibitor of DXS, clomazone, but showed no significant changes in the contents of α-santalene, β-santalene and α-santalol between treatment and control groups. These results suggest that SaDXS1A/1B play a role in the synthesis of sandalwood sesquiterpenes, providing carbon for downstream secondary metabolites. SaDXS1A/1B also play a role in the biosynthesis of chlorophyll, carotenoids, and primary metabolites.

Exploration on Optimized Control Way of D-Amino Acid for Efficiently Mitigating Membrane Biofouling of Membrane Bioreactor

Zhan Gao,Zhihao Yu,Xiaoli Zhang,Shougang Fan,Huiyu Gao,Caini Liu,Qixing Zhou,Huaiqi Shao,Lan Wang,Xiaoyan Guo

The thorny issue of membrane biofouling in membrane bioreactors (MBR) calls for new effective control measures. Herein, D-amino acid (DAA) was employed to mediate MBR membrane biofouling by inhibiting biofilm information and disintegrating formed biofilm. Different DAA control ways involving membrane property, DAA-adding timing, and DAA-control mode were explored through experiments and the multiple linear regression model and the response surface methodology. The optimized DAA control ways were acquired, involving DAA used as an active agent, and the DAA-adding timing of 4 h cultured before running, as well as both hydrophilic and hydrophobic membrane, resulting in an approximately 40.24% decrease in the membrane biofouling rate in comparison with the conventional MBR. DAA is an efficient membrane biofouling mediating approach for MBR under optimized control ways combination and a facile solution for solving membrane biofouling in actual membrane systems. Keywords:membrane bioreactor;membrane biofouling;D-amino acid;control way;model evaluation

Transition-metal-free decarboxylation of D-glucaric acid to furan catalyzed by SnCl4 in a biphasic system

Teng Li,Guangyu Sun,Lu Xiong,Boying Zheng,Yaqi Duan,Rui Yu,Jun Jiang,Yantao Wang,Weiran Yang

As a useful chemical, furan can be produced from bio-based chemicals, such as furfural and furoic acid (FA). There were few reports on the preparation of furan from bio-based C6 aldaric acids . Herein, a new method was developed to produce furan from D-glucaric acid (GA) without transition metals, and nearly 80% yield of furan was produced in the water/toluene biphasic system with SnCl 4 at 200 °C in 2 h. 3-Hydroxy-2-pyrone (3-HP) and FA were proved to be the key intermediates in the formation of furan from GA, and the reaction pathway was proposed to be GA→ 3-HP→ FA→ furan based on the kinetic study , model substrate reaction, and isotope labeling experiments. SnCl 4 gave the highest furan yield among the common acid catalysts, and the contrast experiments suggested that H + , Cl − and Sn 4+ all have promoting effect on GA conversion to furan. This method was also applicable with other C6 aldaric acids.

Hole-rich CoP nanosheets with an optimized d-band center for enhancing pH-universal hydrogen evolution electrocatalysis

Shuo Geng,Fenyang Tian,Menggang Li,Xin Guo,Yongsheng Yu,Weiwei Yang,Yanglong Hou

Tailoring the d-band center is an effective method to promote the hydrogen evolution reaction (HER) activity of electrocatalysts. Herein, we demonstrate that the d-band center can be tuned through a hole-creating method to enhance pH-universal HER activity using CoP as a basic platform. The density functional theory (DFT) calculation reveals that the d-band center and the valence electron number of Co sites around the holes are upshifted, which boosts H adsorption. In addition, introducing holes into the nanosheets of CoP can optimize the ΔGH* of the Co atoms around the holes. Inspired by the DFT results, a soft template method was developed to synthesize hole-rich CoP nanosheets. With the advantages of hole-rich and unique nanosheet features, the hole-rich CoP nanosheets show low HER overpotentials with only 84 and 94 mV to achieve a current density of 10 mA cm−2 in both acidic and alkaline media, better than that of the other dimensional counterparts. In addition, the hole-rich CoP nanosheets also show satisfactory stability after long-term HER tests. This strategy of regulating the d-band center is expected to be extended to other transition metal phosphide electrocatalysts for enhancing the HER performance.

Selectivity-Switchable Conversion of Chitin-Derived N-Acetyl-d-glucosamine into Commodity Organic Acids at Room Temperature

Jingwei Wu,Man Qi,Gökalp Gözaydın,Ning Yan,Yongjun Gao,Xi Chen

Chitin is an abundant and sustainable biomass from oceanic waste. Herein, a simple, mild, and effective method was developed to convert chitin biomass into valuable organic acids at room temperature using a common base and oxidant in water. In dilute sodium hydroxide (NaOH) solution, ∼72.3% yield of organic acid products including acetic acid (AA), formic acid (FA), glyceric acid (GlyA), and so forth was obtained from the chitin monomer N-acetyl-d-glucosamine under ambient air. Meanwhile, tunable product selectivity can be achieved by employing different oxidative reagents. FA was predominantly formed in 57.1% yield using hydrogen peroxide (H2O2), while AA and GlyA were the major products in 41.2% yield with oxygen (O2) gas. Divergent reaction pathways were presumably involved when using different oxidants. Compared to the conventional methods at high temperatures and/or pressures, the established approach requires minimal energy inputs, inexpensive equipment with reduced carbon emission, capital costs, and safety risks.

2-Deoxy-d-glucose Promotes Buforin IIb-Induced Cytotoxicity in Prostate Cancer DU145 Cells and Xenograft Tumors

Yangke Wanyan,Xixi Xu,Kehang Liu,Huidan Zhang,Junai Zhen,Rong Zhang,Jumei Wen,Ping Liu,Yuqing Chen

Inhibition of the glycolytic pathway is a critical strategy in anticancer therapy because of the role of aerobic glycolysis in cancer cells. The glycolytic inhibitor 2-Deoxy-d-glucose (2-DG) has shown potential in combination with other anticancer agents. Buforin IIb is an effective antimicrobial peptide (AMP) with broad-spectrum anticancer activity and selectivity. The efficacy of combination treatment with 2-DG and buforin IIb in prostate cancer remains unknown. Here, we tested the efficacy of buforin IIb as a mitochondria-targeting AMP in the androgen-independent human prostate cancer cell line DU145. Combining 2-DG with buforin IIb had a synergistic toxic effect on DU145 cells and mouse xenograft tumors. Combination treatment with 2-DG and buforin IIb caused stronger proliferation inhibition, greater G1 cell cycle arrest, and higher apoptosis than either treatment alone. Combination treatment dramatically decreased L-lactate production and intracellular ATP levels, indicating severe inhibition of glycolysis and ATP production. Flow cytometry and confocal laser scanning microscopy results indicate that 2-DG may increase buforin IIb uptake by DU145 cells, thereby increasing the mitochondria-targeting capacity of buforin IIb. This may partly explain the effect of combination treatment on enhancing buforin IIb-induced apoptosis. Consistently, 2-DG increased mitochondrial dysfunction and upregulated Bax/Bcl-2, promoting cytochrome c release to initiate procaspase 3 cleavage induced by buforin IIb. These results suggest that 2-DG sensitizes prostate cancer DU145 cells to buforin IIb. Moreover, combination treatment caused minimal hemolysis and cytotoxicity to normal WPMY-1 cells. Collectively, the current study demonstrates that dual targeting of glycolysis and mitochondria by 2-DG and buforin IIb may be an effective anticancer strategy for the treatment of some advanced prostate cancer. Keywords:buforin IIb;2-Deoxy-d-glucose;mitochondria;glycolysis;prostate cancer
