Targeted Theranostic Nanoprobes Assisted In Vivo NIR-II Fluorescence Imaging-Guided Surgery Therapy for Alveolar Echinococcosis

Chi Zhang,Li Jun,Yuxiang Gao,Le Guo,Jiayi Ding,Lijun Zhu,Zhong Du,Jiabao Xiong,Xueliang Zhang,Wenbao Zhang,Biao Dong,Nuernisha Alifu

Alveolar echinococcosis (AE) is a serious parasitic infectious disease that is highly invasive and destructive to the liver and has a high mortality rate. However, currently, there is no effective targeted imaging and treatment method for the precise detection and therapy of AE. We proposed a new two-step targeting strategy (TSTS) for AE based on poly(lactic-co-glycolic acid) (PLGA). We designed and constructed a novel type of ICG@PLGA@Lips nanoprobe with integrated imaging and treatment properties. First, we used the characteristics of PLGA gluconeogenic raw material to target the liver during blood circulation. Then, we utilized the characteristics of PLGA specifically penetrating the AE shell to achieve specific identification of AE in the liver. Under 808 nm laser excitation, ICG@PLGA@Lips effectively achieved accurate imaging of AE based on near-infrared II (NIR-II, 900–1700 nm) fluorescence imaging methods and achieved AE treatment through PDT effects. PLGA improved the optical properties of ICG, while liposomes further improved the biocompatibility of the nanoprobe. As ICG@PLGA@Lips showed strong NIR-II fluorescence emission and good biocompatibility, ICG@PLGA@Lips showed advantages in the specific fluorescence navigation of AE surgical resection lesions. Thus, with the assistance of ICG@PLGA@Lips, we achieved precise targeted and real-time NIR-II fluorescence imaging of AE for the first time. We successfully obtained in vivo NIR-II fluorescence imaging-guided photodynamic/surgical therapy of AE. This TSTS-based AE imaging and treatment exploration provided a new strategy for accurate imaging and treatment of early AE, which is expected to significantly improve the prognosis of patients.

Ultrabroadband (Vis-NIR) Emission in Single-Component Perovskite LEDs via Tailored Multi-Exciton Energy Transfer Pathways

Yuxiang Xin,Jianru Wang,Xiaoli Gao,Yang Yang,Jianbing Zhang,Jiang Tang,Zhuolei Zhang

The development of ultrabroadband light sources spanning the visible (Vis) to near-infrared (NIR) range is of fundamental importance for cutting-edge applications in communication, metrology, and quantum technologies. Although phosphor-converted LEDs have extended emission, they still suffer from spectral gaps, inadequate color rendering, and constrained NIR output. Here, a promising approach is introduced by tailoring multi-exciton energy transfer pathways to realize ultrabroadband Vis-to-NIR emission in Sb 3 ⁺/Ln 3 ⁺ co-doped vacancy-ordered Cs₂HfCl₆ perovskite nanocrystals. By systematically optimizing Sb 3 ⁺ and Ln 3 ⁺ doping concentrations, excitation wavelengths, and lanthanide ion selection, the energy transfer pathways between singlet and triplet self-trapped exciton (STE) states and lanthanide energy levels are modulated. This design facilitates broad spectral tunability through energy distribution control among emission centers, improves radiative efficiency by reducing energy losses during transfer processes, and promotes stable performance by mitigating excessive energy accumulation. The resulting single-component LED based on Sb 3 ⁺/Pr 3 ⁺ co-doped NCs delivers broadband emission extending to ≈1200 nm, high spectral quality (CRI ∼ 98.1, R9 ∼ 98), strong visible/NIR PLQY (≈80%), and steady operation over 50 h. The findings provide insight into energy transfer engineering in low-dimensional perovskites and offer a viable route toward emerging ultrabroadband solid-state light sources.

Polyvalent Aptamer-Functionalized NIR-II Quantum Dots for Targeted Theranostics in High PD-L1-Expressing Tumors

Xin Huang,Jiawei Zhu,Chuhuang Dong,Yuqing Li,Qing Yu,Xuan Wang,Zhejie Chen,Jiabei Li,Yu Yang,Haifang Wang

Ag2S quantum dots (QDs) show superior optical properties in the NIR-II region and display significant clinical potential with favorable biocompatibility. However, inherent defects of low targeting and poor solubility necessitate practical modification methods to achieve the theranostics of Ag2S QDs. Herein, we used rolling circle amplification (RCA) techniques to obtain long single-stranded DNA containing the PD-L1 aptamer and C-rich DNA palindromic sequence. The C-rich DNA palindromic sequences can specifically chelate Ag2+ and thus serve as a template to result in biomimetic mineralization and formation of pApt-Ag2S QDs. These QDs enable specific targeting and illuminate hot tumors with high PD-L1 expression effectively, serving as excellent molecular targeted probes. In addition, due to the high NIR-II absorption of Ag2S QDs, pApt-Ag2S QDs exhibit remarkable photothermal properties. And besides, polyvalent PD-L1 aptamers can recognize PD-L1 protein and effectively block the inhibitory signal of PD-L1 on T cells, enabling efficient theranostics through the synergistic effect of photothermal therapy and immune checkpoint blocking therapy. Summary, we enhance the biological stability and antibleaching ability of Ag2S QDs using long single-stranded DNA as a template, thereby establishing a theranostic platform that specifically targets PD-L1 high-expressing inflamed tumors and demonstrates excellent performance both in vitro and in vivo.

New Type Annihilator of π-Expanded Diketopyrrolopyrrole for Robust Photostable NIR-Excitable Triplet–Triplet Annihilation Upconversion

Fang Qi,Hong-Juan Feng,Yi Peng,Lin-Han Jiang,Le Zeng,Ling Huang

Near-infrared light excitable triplet–triplet annihilation upconversion (NIR TTA-UC) materials have attracted interest in a variety of emerging applications such as photoredox catalysis, optogenetics, and stereoscopic 3D printing. Currently, the practical application of NIR TTA-UC materials requires substantial improvement in photostability. Here, we found that the new annihilator of π-expanded diketopyrrolopyrrole (π-DPP) cannot activate oxygen to generate superoxide anion via photoinduced electron transfer, and its electron-deficient characteristics prevent the singlet oxygen-mediated [2 + 2] cycloaddition reaction; thus, π-DPP exhibited superior resistance to photobleaching. In conjunction with the NIR photosensitizer PdTNP, the upconversion efficiency of π-DPP is as high as 8.9%, which is eight times of the previously reported PdPc/Furan-DPP. Importantly, after polystyrene film encapsulation, less than 10% photobleaching was observed for this PdTNP/π-DPP-based NIR TTA-UC material after four hours of intensive NIR light exposure. These findings provide a type of annihilator with extraordinary photostability, facilitating the development of NIR TTA-UC materials for practical photonics.

Real time monitoring peripheral nerve function with ICG and BDA-ICG by NIR-II fluorescence imaging

Xiaoqi Yang,Yuanyi Wang,Chunrong Qu,Boyu Tan,Minjin Wang,Senrui Li,Jinsheng Huang,Jiangnan Li,Mengyuan Fang,Zhen Cheng,Nan Zhou

Neuroanatomical tract tracers are important for studying axoplasmic transport and the complex interconnections of the nervous system. Though traditional fluorescent tracers are widely used, they have several prominent drawbacks when imaging, including low resolutions and low tissue penetrations and inability to be supervised dynamically within a long peripheral nerve during the long term. Here, we explored the potential of ICG as a neural tracer for axoplasmic transport and for the first time demonstrated that ICG could be used to detect transport function within peripheral nerve by near-infrared region II (NIR-II) imaging. On basis of this finding, a novel bi-directional neural tracer biotinylated dextran amine-indocyanine green (BDA-ICG) was prepared and characterized with better long-term stability and higher nerve-to-background ratio than ICG in vivo, and successfully imaged the injured peripheral nerve from the healthy one within 24 h. Our results show that BDA-ICG are promising neural tracers and clinically available dyes with NIR-II emission tail characteristics as ICG.

Reprogrammable soft actuators based on a photochromic organic–inorganic hybrid membrane with modulatable NIR photothermal conversion

Xingchen Cui,Zhezhi Li,Zhihui Yi,Bing Zhang,Xing Gao,Fuqiang Fan,Yu Fu,Junbiao Wu,Tieqiang Wang

Shape-transformation soft actuators that can quickly respond to external stimuli have been potential candidates in biomimetic soft robots and flexible driving devices. However, the lack of shape reprogrammability restricts their complexity of deformation as well as reusability in practical application. Herein, we present a unique reprogrammable actuator based on asymmetric bilayer compounding with UV-modulated photochromic complex (NEU20) through a sequential polymerization process. The UV-induced electron transfer photochromic behavior synergistic with NIR photothermal effect of NEU20 fillers endows the soft actuator with rapid (∼90°/s) and customizable actuation under external NIR stimuli. Through the joint modulation of UV exposure region and duration, the UV-induced photothermal conversion efficiency is also adjustable at each local, facilitating the spatio-temporally programmable actuation process of the hybrid bilayer actuator. Significantly, the programmed actuation process of the actuator also features thermal-erasing and UV-reprogramming capacities, highlighting the unique recyclability of the hybrid actuator. In addition, the proposed reprogrammable actuator also demonstrates good universality in combination with other thermal-responsive active material systems. This work offers insight into the design and fabrication of reprogrammable soft actuators, shining enlightenment for potential application in bioinspired soft robots and smart mechanical devices.

Pt nanoshells with a high NIR-II photothermal conversion efficiency mediates multimodal neuromodulation against ventricular arrhythmias

Wang Chenlu,Zhou Liping,Liu Chengzhe,Qiao Jiaming,Han Xinrui,Wang Luyang,Liu Yaxi,Xu Bi,Qiu Qinfang,Zhang Zizhuo,Wang Jiale,Zhou Xiaoya,Zeng Mengqi,Yu Lilei,Fu Lei

Autonomic nervous system disorders play a pivotal role in the pathophysiology of cardiovascular diseases. Regulating it is essential for preventing and treating acute ventricular arrhythmias (VAs). Photothermal neuromodulation is a nonimplanted technique, but the response temperature ranges of transient receptor potential vanilloid 1 (TRPV1) and TWIK-related K + Channel 1 (TREK1) exhibit differences while being closely aligned, and the acute nature of VAs require that it must be rapid and precise. However, the low photothermal conversion efficiency (PCE) still poses limitations in achieving rapid and precise treatment. Here, we achieve a nearly perfect blackbody absorption and a high PCE in the second near infrared (NIR-II) window (73.7% at 1064 nm) via a Pt nanoparticle shell (PtNP-shell). By precisely manipulating the photothermal effect, we successfully achieve rapid and precise multimodal neuromodulation encompassing neural activation (41.0–42.9 °C) and inhibition (45.0–46.9 °C) in a male canine model. The NIR-II photothermal modulation additionally achieves multimodal reversible autonomic modulation and confers protection against acute VAs associated with myocardial ischemia and reperfusion injury in interventional therapy.

Heng Youqiang,Du Zhong,Chu Chenyang,Xiong Jiabao,Gao Yuxiang,Ma Rong,Wang Jing,Alifu Nuernisha,Ma Cailing

Cervical cancer remains one of the leading cause of cancer death in women. As the development of nanomedicine, multifunctional nanoplatforms which integrate imaging and therapy for effective diagnostics and therapeutics of cancer have attracted great attention in recent years. In this work, we have designed and developed a kind of multimodal imaging and treatment strategy which combined near-infrared (NIR) fluorescence imaging with magnetic resonance imaging (MRI) as well as photothermal therapy (PTT) via a multifunctional nanoplatform. The poly lactic-co-glycolic acid (PLGA) was utilized to encapsulate magnetic Fe 3 O 4 nanoparticles (NPs) and NIR fluorescent dye clinical indocyanine green (ICG), by which integrated the advantages of Fe 3 O 4 NPs and clinical ICG under 808-nm laser excitation, to achieve highly efficient dual-modal imaging-guided cervical tumor ablation. This work provides a promising approach for multimodal imaging diagnosis and PTT of cervical cancer.

A NIR-Responsive MXene-Based Hydrogel with Antibacterial and Repair-Promoting Functions for Treating Infected Wounds

Zhongjie Ji,Dong Yang,Hongru Wang,Zhenqian Sun,Linyang Song,Xin Liu,Hongliang Wang,Yunzhen Chen,Hongxin Cao,Yunhao You,Guangjun Jiao

Skin wound infection represents one of the most frequent complications in the management of surgical diseases, significantly delaying healing processes and potentially leading to life-threatening conditions. Bacterial invasion promotes biofilm formation, which increases local reactive oxygen species (ROS) levels, disrupts the wound microenvironment, and compromises regenerative capacity. Conventional treatments often fail to effectively eradicate deep biofilm-associated infections, while prolonged antibiotic use fosters multidrug-resistant bacteria and biofilm persistence, underscoring the urgent need for novel antibacterial approaches. In this study, a multifunctional hydrogel dressing is developed that integrates MXene nanosheets with a metal-phenolic network (EGCG-Cu) and the nitric oxide (NO) donor BNN6. This composite hydrogel enables near-infrared (NIR)-triggered photothermal therapy and simultaneous NO release, resulting in synergistic antibacterial activity against Staphylococcus aureus and Escherichia coli , along with effective biofilm eradication. The EGCG-Cu coating further confers remarkable antioxidant and anti-inflammatory properties by scavenging ROS and modulating cytokine expression. In a murine model of infected wound healing, the hydrogel accelerated tissue repair through enhanced collagen deposition, angiogenesis, and regulation of the immune microenvironment. This non-antibiotic strategy provides spatiotemporal control over antibacterial and healing-promoting functions, demonstrating considerable promise for the treatment of complex wound infections.

Hierarchical core–shell structural based on WO3@ITO nanorods with boosted electrochromic performance in NIR region

Xiaoyue Hao,Yong Liu,Gaorong Han,Gaoling Zhao,Dongxue Liu,Huitao Dai,Sainan Ma,Likun Wang

Over half of the solar energy comes from the infrared region, which makes electrochromic materials that modulate infrared transmittance essential for the thermal management of smart windows. However, existing electrochromic materials do not perform well in the near-infrared region, which exhibits slow response time, weak adhesion to conductive substrates, and poor long-term cycling stability. In the present work, we successfully synthesized WO 3 @ITO nanorod samples with hierarchical core–shell structure by hydrothermal and in-situ solvothermal methods. ITO-coated WO 3 nanorods and some ITO nanocrystals were firmly integrated with WO 3 nanorods, resulting in a corn-like hybrid heterogeneous structure. The unique WO 3 @ITO hierarchical core–shell structure improves the electronic properties and the adhesion to substrates, reducing the energy barrier of ion diffusion and increasing the ion flux. Enhanced electrochromic performance in NIR range with a high optical modulation of 53.3 % in 1600 nm and long-term reversibility (maintaining 91.5 % of its original contrast after 1000 cycles) was achieved. In addition, response time (t c  = 3.5 s, t b  = 4.6 s) and coloration efficiency (112.5 cm 2 ‧C -1 ) have been significantly improved, which is almost twice that of unmodified WO 3 nanorods (t b  = 8.6 s and CE = 56.4 cm 2 ‧C -1 ), showing a good application prospect in smart windows.

NIR-II aggregation-induced emission nanoparticles camouflaged with preactivated macrophage membranes for phototheranostics of pulmonary tuberculosis

Yan Dingyuan,Li Xue,Wang Huanhuan,Li Bin,Wang Wei,Liao Yuhui,Tang Ben Zhong,Wang Dong

Phototheranostics, which allows simultaneous diagnosis and therapy, offers notable advantages in terms of noninvasiveness, controllability and negligible drug resistance, presenting a promising approach for disease treatment. By integrating second near-infrared (NIR-II, 1,000–1,700 nm) phototheranostic agents characterized by aggregation-induced emission (AIE) and cell membranes with specific targeting capacity, we have developed a versatile type of biomimetic nanoparticle (NP) for precise phototheranostics of pulmonary tuberculosis (TB). Coating the phototheranostic agents with preactivated macrophage membranes results in the formation of biomimetic NPs, which exhibit specific binding to TB through a lesion–pathogen dual-targeting strategy, allowing the accurate detection of Mycobacterium tuberculosis via NIR-II fluorescence imaging and precise photothermal therapy using the irradiation of a 1,064 nm laser. In comparison with traditional treatments, small individual granulomas (0.2 mm in diameter) in TB-infected mice are visualized, and improved antibacterial effects are achieved upon NP administration. Here we present a standardized workflow for the synthesis of the NIR-II AIE agents, their use for the fabrication of the biomimetic NPs and their in vitro and in vivo applications as phototheranostics against M. tuberculosis . The preparation and characterization of the NIR-II AIE agents requires ~8 d, the synthesis and characterization of the phototheranostic NPs requires ~8 d, the validation of in vitro targeting capacity and photothermal eradication requires ~26 d, and the in vivo NIR-II fluorescence imaging and imaging-guided photothermal therapy requires ~74 d. All procedures are straightforward and suitable for clinicians or researchers with prior training in organic synthesis and biomedical engineering.

Cu/Au-doped nanopolymers with multiple catalytic activities for NIR II laser-promoted nanocatalytic tumor therapy

Xixi Wu,Xiang Feng,Peng Yu,Jing Zhang,Rui Liu

Breast cancer remains a significant global health concern owing to the limitations of conventional therapies, such as side effects, drug resistance, and high costs. Nanocatalytic therapy has emerged as a promising alternative due to its tumor specificity, spatiotemporal controllability, and noninvasiveness. However, its effectiveness is limited by endogenous antioxidants like glutathione (GSH) and the low catalytic activity of nanocatalysts. Herein, an Cu/Au-doped polypyrrole nanocatalyst, Cu-AuPP, with multiple catalytic activities is developed by sequentially polymerizing pyrrole monomers using CuCl2 and HAuCl4, followed by PEGylation. The obtained Cu-AuPP catalyzes the production of hydroxyl radicals (·OH) and facilitates the oxidation of GSH to GSSG via redox reactions mediated by multivalent Cu ions, leading to oxidative damage, mitochondrial dysfunction, and tumor cell apoptosis. Upon 1064 nm laser irradiation, these catalytic activities were enhanced by elevated temperature and electron–hole separation mediated by Au nanoclusters, resulting in more intense oxidative damage to tumor cells. Collectively, the developed Cu-AuPP nanocatalytic medicine capable of simultaneously catalyzing GSH depletion and ·OH production via several improved catalytic mechanisms, has significant promise for the treatment of malignant tumors.
