Kaempferol from Alpinia officinarum Hance induces G2/M cell cycle arrest in hepatocellular carcinoma cells by regulating the ATM/CHEK2/KNL1 pathway

Xiaoliang Li,Mingyan Zhou,Zhe Zhu,Zhe Wang,Xuguang Zhang,Lu Lu,Zhenrui Xie,Bingshu Wang,Yipeng Pan,Junqing Zhang,Jian Xu

Ethnopharmacological relevance Alpinia officinarum Hance ( A . officinarum ), a perennial herb known for its medicinal properties, has been used to treat various ailments, such as stomach pain, abdominal pain, emesis, and digestive system cancers. A . officinarum is extensively cultivated in the Qiongzhong and Baisha regions of Hainan, and it holds substantial therapeutic value for the local Li people of Hainan. Kaempferol, a flavonoid derived from A . officinarum , has demonstrated anticancer properties in various experimental and biological studies. Nevertheless, the precise mechanisms through which it exerts its anti-hepatocellular carcinoma (HCC) effects remain to be comprehensively delineated. Aim of the study This investigation aims to elucidate the anti-HCC effects of kaempferol derived from A . officinarum and to delve into its underlying mechanistic pathways. Materials and Methods Using ultra-high performance liquid chromatography-mass spectrometry/mass spectrometry (UPLC-MS/MS) to identify active compounds in A. officinarum . HCCLM3 and Huh7 cells were used to study the anti-HCC effect of kaempferol from A . officinarum . The cytotoxicity and proliferation of kaempferol and A . officinarum were measured using CCK-8 and EDU staining. Wound-healing assays and three-dimensional tumor spheroid models were further used to evaluate migration and the anti-HCC activity of kaempferol. The cell cycle and apoptosis were evaluated by flow cytometry. Western blot and qRT-PCR were used to detect the expression of proteins and genes associated with the cell cycle checkpoints. Finally, bioinformatics was used to analyze the relationship between the differential expression of core targets in the ATM/CHEK2/KNL1 pathway and a poor prognosis in clinical HCC samples. Results UPLC-MS/MS was employed to detect five active compounds in A . officinarum , such as kaempferol. The CCK-8 and EDU assays showed that kaempferol and A . officinarum significantly inhibited the proliferation of HCC cells. A wound-healing assay revealed that kaempferol remarkably inhibited the migration of HCC cells. Kaempferol significantly suppressed the growth of tumor spheroids. In addition, kaempferol markedly induced G2/M arrest and promoted apoptosis of HCC cells. Mechanically, kaempferol significantly reduced the protein and mRNA expression levels of ATM, CHEK2, CDC25C, CDK1, CCNB1, MPS1, KNL1, and Bub1. Additionally, the combination of kaempferol and the ATM inhibitor KU55933 had a more significant anti-HCC effect. The results of bioinformatics showed that ATM, CHEK2, CDC25C, CDK1, and KNL1 were highly expressed in patients with HCC and cancer tissues, indicating that these genes have certain value in the clinical diagnosis of HCC. Conclusions Collectively, our results revealed that kaempferol from A . officinarum inhibits the cell cycle by regulating the ATM/CHEK2/KNL1 pathway in HCC cells. In summary, our research presents an innovative supplementary strategy for HCC treatment.

Magnetic-responsive Pickering emulsions based on MFe2O4 (M = Mn, Fe, Co, Ni, Cu, Zn) for green and efficient oxidation of benzyl alcohol

Yan Wang,Xuan Wang,Yonghao Dong,Mingli Peng,Lina Guo,Mengyao Cui,Yuan He,Jiabao Yi,Huijun Ma,Huan Zhang,Haiming Fan

The selective oxidation of alcohols has drawn significant attention in heterogeneous catalysis with the increasing use of Pickering interfacial catalysis (PIC) for green transformations of alcohols. However, it is still highly desirable to design and develop a solid catalyst with easy separation and recycling, made from abundant resources through a simple synthesis route, for the selective oxidation of alcohols. In this study, ferrite nanoparticles, synthesized via a hydrothermal process, were employed to act as a stabilizer and catalyst in PIC for the oxidation of benzyl alcohol using H2O2. The ferrite nanoparticles exhibited higher catalytic performance in PIC than in acetonitrile. The turnover number (TON) of the nanocatalyst in PIC was found to be 1.5–4 times higher than that in acetonitrile, and the reaction constant in PIC (kPIC = 0.712 h−1) was almost twice that in acetonitrile (kCH3CN = 0.310 h−1) for MnFe2O4 nanoparticles. Moreover, other alcohols could also be oxidized with moderately high conversion and selectivity in PIC. Significantly, the ferrite nanocatalyst could be easily separated from PIC using a magnet for reuse, achieving 88% BzOH conversion after five cycles. The use of hazardous organic solvents was avoided in PIC, making it a more advanced, green and sustainable reaction system.

Ligustilide covalently binds to Cys254 of the creatine kinase, M-type protein, ameliorating acute myocardial ischemia by enhancing the creatine phosphate level

Kaixue Zhang,Guoqing Luan,Jin Zhang,Shilong Wang,Min Jiang,Gang Bai

Background Myocardial ischemia (MI) threatens the health of middle-aged and older adults by reducing cardiac oxygen supply and function. Current therapies, including vasodilation, thrombolysis, and interventions, focus on relieving symptoms and improving blood flow but do not adequately address underlying energy metabolism issues. Ligustilide exerts a protective effect on the cardiovascular system and holds the potential for ameliorating MI; however, there is currently no systematic elucidation of ligustilide's target and action mechanism for MI. Purpose This study aimed to comprehensively assess ligustilide's potential targets for improving acute MI and elucidate its underlying mechanism. Methods The therapeutic effects of ligustilide were evaluated at doses of 30, 15, and 7.5 mg/kg over 7 days in a murine model of acute MI induced by isoproterenol hydrochloride. The putative target protein was identified through target fishing, in-gel imaging, and thermal shift assay (TSA), followed by tissue and cell localization studies via a ligustilide probe. The interaction sites between ligustilide and the target protein were elucidated using protein profiling, molecular docking, and TSA at the protein level. Subsequently, knockdown and reconstruction tests were employed at the cellular level to identify the functionally active sites where ligustilide binds to the target protein. Finally, molecular docking and molecular dynamics simulations were conducted to elucidate the underlying mechanism by which ligustilide enhances creatine kinase, M-type (CKMM) protein activity. Results The covalent bonding of ligustilide in cardiac tissue enhances the therapeutic effect on acute MI in mice. For the first time, we found ligustilide specifically targets Cys254 of the CKMM protein following epoxidation. This irreversible binding effectively reduces the proximity between creatine and ATP, promoting creatine phosphorylation and ultimately increasing the creatine phosphate (CP) level by 9.50% to 19.31%. The accumulation of CP alleviates MI by enhancing energy metabolism, mitigating oxidative stress, and suppressing inflammatory responses. Conclusions Our study unveiled ligustilide as a CKMM activator, which effectively enhances the content of CP and mitigates acute MI. The findings significantly contribute to advancing our understanding of ligustilide's function for myocardial protection while proposing a novel activation mechanism of CKMM to improve MI. And the insight into the covalent regulation of the active pocket on CKMM may lead to an alternative therapeutic strategy against acute MI.

The preparation of Fe2O3-ZSM-5 catalysts by metal-organic chemical vapour deposition method for catalytic wet peroxide oxidation of m-cresol

Yi Yang,Huiping Zhang,Ying Yan

Fe2O3-ZSM-5 catalysts (0.6 wt% Fe load) prepared by metal-organic chemical vapour deposition (MOCVD) method were evaluated in the catalytic wet peroxide oxidation (CWPO) of m-cresol in a batch reactor. The catalysts have a good iron dispersion and small iron crystalline size, and exhibit high stability during reaction. In addition, the kinetics of the reaction were studied and the initial oxidation rate equation was given. Catalysts were first characterized by N2 adsorption–desorption isotherms, scanning electronic microscopy, energy-dispersive spectroscopy, X-ray diffraction and X-ray photoelectron spectroscopy. Results show that extra-framework Fe3+ species (presenting in the form of Fe2O3) are successfully loaded on ZSM-5 supports by MOCVD method. Performances of catalysts were tested and effects of different temperature, stirring rate, catalyst amount on hydrogen peroxide, m-cresol, total organic carbon (TOC) conversion and Fe leaching concentration were studied. Results reveal that catalytic activity increased with higher temperature, faster stirring rate and larger catalyst amount. In all circumstances, m-cresol conversion could reach 99% in 0.5–2.5 h, and the highest TOC removal (80.5%) is obtained after 3 h under conditions of 60°C, 400 r.p.m. and catalyst amount of 2.5 g l−1. The iron-leaching concentrations are less than 1.1 mg l−1 under all conditions. The initial oxidation rate equation −rA0=7.2×1011e−81300/RTCA is obtained for m-cresol degradation with Fe2O3-ZSM-5 catalysts.

Measurement and correlation of isobaric vapour–liquid equilibrium for systems of o-cresol, m-cresol and 2, 6-dimethylphenol at 20.0 kPa

Jun Wang,Jie Ou,Xueni Sun,Chunxiang Huang,Jiarui Sun,Hui Shao,Yixin Leng

Isobaric vapour–liquid equilibrium (VLE) data for binary systems containing o-cresol (OC) + m-cresol (MC), OC + 2, 6-dimethylphenol (DMP), and MC + DMP at 20.0 kPa were measured by using a modified Othmer still. Its reliability was confirmed by measuring the saturated vapour pressure data for the components of MC and DMP. Herington area test was used to confirm the thermodynamic consistency for these binary systems, and the results showed good thermodynamic consistency. The binary VLE experimental data were correlated and predicted by Wilson, NRTL, and UNIFAC models. The results correlated by Wilson and NRTL models agree well with the experimental values. The results predicted by UNIFAC are in accordance with experimental data with rational deviations for systems OC + MC and OC + DMP but show relatively large deviations between the predicted values and experimental data for MC + DMP system. By defining the group =COH in DMP and MC as two different groups, the experimental data were correlated, and new groups with binary interaction parameters were obtained. The results calculated by UNIFAC with new defining groups are in accordance with the experimental data.

Synthesis and properties of green/sky blue high near infrared reflection coatings based on Mg2.4-xMxTiO4.4+δ (M = Cr/Co) inorganic pigments

Menghang Qi,Liangsheng Tian,Yutong Wang,Xin Xin,Hang Zhao,Theeranun Siritanon,M.A. Subramanian,Peng Jiang

To alleviate the urban heat island effect, reduce building energy consumption and improve the living environment for residents, the development of highly reflective pigments and coatings is of great significance for sustainable development. In this study, Mg 2.4- x Cr x TiO 4.4+δ (0.1 ≤ x ≤ 0.3) and Mg 2.4- x Co x TiO 4.4 (0.1 ≤ x ≤ 0.4) pigments with intense green/sky blue colors were successfully synthesized through solid state method. The crystal structure, color development mechanism and optical properties of as-synthesized samples are analyzed. The as-prepared pigments demonstrate excellent NIR reflectance with the highest solar reflectance up to 83%. Furthermore, the color coatings with high NIR solar reflectance were successfully prepared by mixing the pigments with acrylic resin. A solar reflectance of 69.81% is achieved among the green/blue coatings. The thermal shielding effect of the coatings in practical applications was tested through chamber simulation experiments. The test temperature of the prepared coating was 8.8 ℃ lower than that of the blank stainless steel plate after radiating with a 250   W infrared lamp for 30   minutes, which proved its good heat insulation performance. Through pickling and alkali washing experiments, the color coordinates of the coatings are very stable, indicating the excellent acid-base resistance in the practical applications.

Exploration of the microwave absorption mechanism of rare earth enhanced M-type strontium ferrite: synergistic effect of lattice occupation and impurity phase

Jing Xiaodong,Zhao Qianqian,Wang Qun,Xiong Xiaoqiang,Yang Xi,Huang Hai,Yuan Chenglong,Yan Junjie,Tan Guoguo,Zhao Tongyun,Gong Huayang

How do rare earth (RE) ions affect the microwave absorption performance of M-type hexagonal ferrites? We synthesized Sr 0.9 RE 0.1 Fe 12 O 19 and SrFe 12− x RE x O 19 ( x  = 0.05, 0.1, 0.4, 0.8, 1.6, RE = La ~ Gd) series to investigate the similarities and differences in the mechanism of RE ion substitution on the absorption properties of materials. The occupancy tendency and substitution limit of RE ions in the M phase lattice, as well as the formation of impurity phases at grain boundaries, are key factors affecting the microwave absorption performance of materials. The synthesized materials include both pure M phase and coexistence of M phase and heterophases, and the heterophases are mainly FeRE y Sr 1− y O 3− z , CeO 2 and RE 3 Fe 5 O 12 . The strontium ferrite substituted with La and Ce has better absorption effect in pure M phase. The RL min of SrFe 11.9 La 0.1 O 19 can reach − 60.61 dB and the EAB max is 6.24 GHz with a thickness of 1.96 mm. Strontium ferrite substituted with Pr and Nd can achieve good absorption effects over a large range of substitution amounts, which is not only related to their high substitution limit in the M phase, but also to the perovskite type impurities generated by excessive substitution. The RL min of the SrFe 10.4 Nd 1.6 O 19 can reach − 75.34 dB, and the EAB max is 5.16 GHz with a thickness of 1.9 mm. The distribution of strontium ions in the two phases also becomes a key factor affecting the absorption performance. The substitution limit of Eu and Gd in the M phase is small, and excessive addition can form high resistance impurities, resulting in poor absorption efficiency of the system.

Catechol derivative as a novel bio-based corrosion inhibitor for carbon steel in 1 M HCl solution: Synthesis, experiments, and theoretical studies

Zengfeng Wei,Yumeng Zhang,Hu Xia,Lijie Ni,Wenhai Wu,Xiaoning Wang,Heng Quan

A novel bio-based corrosion inhibitor, hexadecyl amine-modified catechol (CA16), was synthesized, and its molecular structure was characterized. The ability of CA16 to inhibit the corrosion of carbon steel in 1 M HCl was evaluated by potentiodynamic polarization, electrochemical impedance spectroscopy and weight loss. The highest inhibition efficiency reached 96.87% at 10 −3 M. CA16 is a mixed-type corrosion inhibitor that can adsorb on the Fe surface spontaneously through chemical adsorption following the Langmuir adsorption model. The adsorption of CA16 and carbon steel occurred between catechol groups and the Fe surface, as confirmed by quantum chemical calculations and molecular dynamic simulations. This novel bio-derived high-performance inhibitor offers a sustainable alternative to conventional petrochemical-based counterparts, demonstrating significant potential for industrial-scale applications while aligning with green chemistry principles.

Tuning the A-site cation in spinel MAl2O4 (M = Co, Mg, Zn, Cu) supports enables superior Pd-Catalyzed hydrodechlorination of 4-Chlorophenol

Zirui Qin,Jiehao Xie,Xiaofang Wen,Hongcan Lou,Zipin Peng,Xingkun Chen,Yuan Tan,Jisong Zhang

Chlorinated phenols are priority pollutants that require efficient low-temperature removal technologies. Here, we synthesize a series of spinel-type MAl 2 O 4 (M = Co, Mg, Zn, Cu) supports and deposit 1 wt% Pd via incipient-wetness impregnation to evaluate their hydrodechlorination (HDC) performance toward 4-chlorophenol under ambient aqueous conditions (30 °C, 0.2 MPa H 2 ). Catalytic screening shows that the identity of the divalent A-site cation strongly modulates activity: Pd/Co-Al achieves 91.8 % conversion with > 98.3 % selectivity to phenol, outperforming Pd/Ni-Al (84.7 %) and the remaining analogues. Pd/Cu-Al exhibits almost no conversion (<5.0 %), while Pd/Mg-Al and Pd/Zn-Al give moderate conversions (ca.55.4–58.7 %). Detailed characterization by XRD, N 2 -physisorption, H 2 -TPR, XPS and STEM reveals that the superior performance of Pd/Co-Al is rooted in (i) strong metal–support interactions that stabilize a balanced Pd 2+ /Pd 0 surface ensemble, (ii) coherent lattice matching at Pd(1   1   1)/CoAl 2 O 4 (311) interfaces that suppress particle growth, and (iii) high spinel thermostability that mitigates leaching and coking. A 30 h continuous-flow test confirms stable operation without activity loss. This work elucidates how subtle variations in spinel composition dictate Pd electronic structure and interfacial geometry, providing design principles for next-generation HDC catalysts.

Synthesis and characterization of topology-varied organic electroluminescent blue-light emitters based on carbazole and triphenylphosphine oxide derivatives

Chenglian Hu,Chunchun Fei,Hongji Jiang

Two blue light emitters M-2EtCz and P-2EtCz based on carbazole and triphenylphosphine oxide were synthesized and characterized. The 5% weight loss temperatures of emitters M-2EtCz and P-2EtCz solid powder were 437 and 495 °C, and their photoluminescent quantum yield in solution were 53.93 and 75.78%, which were both significantly improved to those of parent EtCz-Ph . The introduction of triphenylphosphine oxide into EtCz-Ph can lower the lowest unoccupied molecular orbital levels of emitters M-2EtCz and P-2EtCz , while keeping their highest occupied molecular orbital levels unchanged. All the emitters exhibited blue light emission with aggregation-induced enhanced emission characteristic. The non-doped deep blue light-emitting diodes with maximum external quantum efficiencies of 2.68% were obtained for M-2EtCz , and the electroluminescent color purity of P-2EtCz was better than that of M-2EtCz . The doped device hosted by P-2EtCz achieved a maximum brightness of 10,891 cd/m 2 , external quantum efficiency of 2.04% and current efficiency of 5.65 cd/A.

Hydrogen production via catalytic propane partial oxidation over Ce1-xMxNiO3-λ (M=Al, Ti and Ca) towards solid oxide fuel cell (SOFC) applications

Ji Qi,Mingzheng Liao,Chao Wang,Zhiqiang Jiang,Ying Chen,Bo Liang,Qingbin Song

Indirect hydrogen production routes presented extensive compatibilities for distributed solid oxide fuel cell systems . Catalyst activity directly determines hydrogen production and profoundly affects the power generation . In this work, Ce 1-x M x NiO 3-λ (M = Al, Ti and Ca) were synthesized for hydrogen production from propane partial oxidation (POx). From the results, it indicated that the choice of catalyst compositions could directly affect the microstructure during the preparation process. Also, different Ce 1-x M x NiO 3-λ catalysts displayed distinctive carbon tolerance performances due to the different active particle sizes and surface properties of the catalysts. In general, the synthesized Ce 1-x M x NiO 3-λ catalysts showed higher hydrogen production than a commercial nickel cerium catalyst. From the observation of a SOFC test system powered by injecting the generated H 2 from catalytic propane POx reaction, the obtained power density of CNO–Al displayed an increase of 22.6% compared to the commercial nickel cerium catalyst. It was quite impressive that the equivalent hydrogen (160 ml/min) produced over CNO–Al and the obtained power density (482.6 mW/cm 2 ) of SOFCs were competitive while CNO–Ca presented an excellent stability with a better carbon tolerance performance for the long-term tests. The achievements of this work might offer a novel point of view for developing low-cost catalyst towards indirect hydrogen production for SOFC.

Bamboo-like N-doped carbon nanotubes encapsulating M(Co, Fe)-Ni alloy for electrochemical production of syngas with potential-independent CO/H2 ratios

Bo Jinxiao,Li Mei,Zhu Xinli,Ge Qingfeng,Han Jinyu,Wang Hua

The electrochemical conversion of CO 2 -H 2 O into CO-H 2 using renewable energy is a promising technique for clean syngas production. Low-cost electrocatalysts to produce tunable syngas with a potential-independent CO/H 2 ratio are highly desired. Herein, a series of N -doped carbon nanotubes encapsulating binary alloy nanoparticles (M x Ni-NCNT, M= Fe, Co) were successfully fabricated through the co-pyrolysis of mela-mine and metal precursors. The M x Ni-NCNT samples exhibited bamboo-like nanotubular structures with a large specific surface area and high degree of graphitization. Their electrocatalytic performance for syngas production can be tuned by changing the alloy compositions and modifying the electronic structure of the carbon nanotube through the encapsulated metal nanoparticles. Consequently, syngas with a wide range of CO/H 2 ratios, from 0.5:1 to 3.4:1, can be produced on M x Ni-NCNT. More importantly, stable CO/H 2 ratios of 2:1 and 1.5:1, corresponding to the ratio to produce biofuels by syngas fermentation, could be realized on Co 1 Ni-NCNT and Co 2 Ni-NCNT, respectively, over a potential window of −0.8 to −1.2 V versus the reversible hydrogen electrode. Our work provides an approach to develop low-cost and potential-independent electrocatalysts to effectively produce syngas with an adjustable CO/H 2 ratio from electrochemical CO 2 reduction.
