β-Cyclodextrin and Hyaluronic Acid-Modified Targeted Nanodelivery System for Atherosclerosis Prevention

Jinjin Yu,Yajing Ma,Xinya Zhang,Siqi Wang,Lili Zhou,Xinyao Liu,Lingli Li,Lingyi Liu,Huixin Song,Yuzhi Luo,Sha Wen,Weifeng Li,Xiaofeng Niu

Natural products have been widely recognized in clinical treatment because of their low toxicity and high activity. It is worth paying attention to modifying the biopolymer into nanostructures to give natural active ingredients additional targeting effects. In this study, based on the multifunctional modification of β-cyclodextrin (β-CD), a nanoplatform encapsulating the unstable drug (−)-epicatechin gallate (ECG) was designed to deliver to atherosclerotic plaques. Acetalization cyclodextrin (PH-CD), which responds to low-pH environments, and hyaluronic acid cyclodextrin, which targets the CD44 receptor on macrophage membranes, were synthesized from β-CD and hyaluronic acid using acetalization and transesterification, respectively. The resulting dual-carrier nanoparticles (Double-NPs) loaded with ECG were prepared using a solvent evaporation method. The Double-NPs effectively scavenged reactive oxygen species, promoted macrophage migration, inhibited macrophage apoptosis, and suppressed abnormal proliferation and migration of vascular smooth muscle cells. Furthermore, the Double-NPs actively accumulated in atherosclerotic plaques in ApoE–/– mice fed with a high-fat diet, leading to a reduced plaque area, inflammatory infiltration, and plaque instability. Our findings demonstrate that the newly developed ECG nanopreparation represents an effective and safe nanotherapy for diseases such as atherosclerosis.

β-CsHg2I5, a compound with rare [Hg2I5] dimers and large optical anisotropy

Yingying Kong,Hongshan Wang,Wang Zhao,Qi Sun,Junjie Li,Shilie Pan

Hg-based compounds show abundant structural diversity and distinguished properties. Herein, a new phase transition compound CsHg2I5 was reported. The high-temperature phase β-CsHg2I5 with rare [Hg2I5] dimers was synthesized by the flux method at 573 K, and it shows a reversible phase transition at a low temperature of ∼100 K to form the low-temperature phase α-CsHg2I5. The two phases crystallize in the same P21/c space group, with different crystal structures. β-CsHg2I5 is composed of rare [Hg2I5] dimers and [CsI11] polyhedral units, while α-CsHg2I5 is composed of [Hg4I11] and [CsI10] units. The experimental band gap of β-CsHg2I5 was found to be 2.58 eV. Owing to the presence of [Hg2I5]∞ pseudo-layers, β-CsHg2I5 exhibits large optical anisotropy with a calculated birefringence of 0.132@1064 nm. Meanwhile, β-CsHg2I5 is a congruent compound and the congruent point is ∼481 K. Theoretical calculations indicate that the rare [Hg2I5] dimer is a nonlinear active unit, which can be used as a new fundamental building block for the design of advanced nonlinear optical materials. Moreover, a CsI–HgI2 pseudo-binary diagram was drawn. The results enrich the structural diversity of Hg-based halides and give some insights into the development of new functional materials based on rare [Hg2I5] dimers.

Yicai Luo,Ziwei Wu,Yingjuan Zhang,Yang Qiao,Yinge Wei,Xuan Yan,Xiangyu Ma,Xianxian Huang,Xiaoxia Zhong,Zhimao Ye,Xinping Lu,Hongbing Liao*

Introduction: Diabetes mellitus often leads to bone metabolism disorders, hindering bone regeneration and delaying the healing of bone defects. β-Ecdysone, a plant-derived hormone known for its wide range of physiological activities, possesses hypoglycemic effects and promotes osteogenic differentiation. This study developed a composite PLGA slow-release scaffold loaded with β-ecdysone to enhance its bioavailability through topical administration and to investigate its potential to heal diabetic bone defects. Methods: The composite scaffolds were fabricated using solution casting/particle leaching and freeze-drying techniques. Then a series of characterizations were subjected to test the performance of composite scaffolds, and in vitro safety of the composite scaffolds was tested by CCK8 assay and live/dead cell staining. Further, micro-CT and histology to evaluate the effect of β-E/PLGA composite scaffolds on healing of skull defects in diabetic rats at 4 and 8 weeks after implantation. Simultaneously, the safety of the scaffolds in vivo was also evaluated. Results: The material characterization results indicated that, in comparison to the single-pore size scaffold, the composite scaffold exhibited superior porosity, swelling ratio, drug loading capacity, and mechanical properties. Additionally, the composite scaffolds showed appropriate degradation performance and sustained drug release profiles. The CCK8 cytotoxicity assay and live/dead cell staining demonstrated that BMSCs survived and proliferated on the composite scaffold under both low-glucose and high-glucose conditions. Micro-CT and histological investigation demonstrated that β-E/PLGA composite scaffolds promoted new bone growth in the skull defect region of diabetic rats. Conclusion: Overall, these findings suggest that the β-E/PLGA composite scaffolds promote the healing of bone defects in diabetic rats. The combination of β-ecdysone and tissue-engineered scaffolds presents a promising approach for treating diabetes-related bone defects.

β-sitosterol protects against ANIT-induced hepatotoxicity and cholestasis via FXR activation

Yuhui Yan,Wenyu Wang,Aiwen Yan,Haonan Zhu,Qiang Meng

Cholestasis, a condition marked by bile acid accumulation in the liver and body systems, leads to liver dysfunction and cirrhosis. Currently, ursodeoxycholic acid (UDCA) and obeticholic acid (OCA) are the only two FDA-approved drugs for Cholestasis. Thus, new therapeutic approaches need to be developed. In this study, we validated the liver-protective properties of β-sitosterol (SIT), a key bioactive element abundant in plants, against hepatic toxicity and cholestasis induced by alpha-naphthylisothiocyanate(ANIT), while elucidating its mechanisms of action both in vivo and in vitro. SIT's FXR activation was confirmed via molecular docking and dual-luciferase assays. In the mechanisms of SIT hepatoprotection, the expression levels of bile salt export pump (Bsep) and multidrug resistance protein2 (Mrp2) which are bile acid efflux transporter, and sulfate transferase 2a1 (Sult2a1) which is a bile acid metabolizing enzyme were all increased by SIT, whereas the expression of uptake transporter sodium taurocholate transporting polypeptide (Ntcp), bile acid synthesis enzyme cholesterol 7α-hydroxylase (Cyp7a1) and oxysterol 12α-hydroxylase (Cyp8b1) was decreased by SIT. In addition, SIT alleviated liver inflammation by suppressing inflammatory factor expression. However, FXR antagonist guggulsterone and FXR siRNA abolished SIT's improvements in liver histology, bile acid transporters, and enzymes. Conclusively, through activating FXR, SIT provides a protective effect against hepatotoxicity and cholestasis. SIT might serve as a new potential therapeutic strategy for the treatment of cholestatic liver diseases.

β-synuclein regulates the phase transitions and amyloid conversion of α-synuclein

Li Xi,Yu Linwei,Liu Xikai,Shi Tianyi,Zhang Yu,Xiao Yushuo,Wang Chen,Song Liangliang,Li Ning,Liu Xinran,Chen Yuchen,Petersen Robert B.,Cheng Xiang,Xue Weikang,Yu Yanxun V.,Xu Li,Zheng Ling,Chen Hong,Huang Kun

Parkinson’s disease (PD) and Dementia with Lewy Bodies (DLB) are neurodegenerative disorders characterized by the accumulation of α-synuclein aggregates. α-synuclein forms droplets via liquid-liquid phase separation (LLPS), followed by liquid-solid phase separation (LSPS) to form amyloids, how this process is physiologically-regulated remains unclear. β-synuclein colocalizes with α-synuclein in presynaptic terminals. Here, we report that β-synuclein partitions into α-synuclein condensates promotes the LLPS, and slows down LSPS of α-synuclein, while disease-associated β-synuclein mutations lose these capacities. Exogenous β-synuclein improves the movement defects and prolongs the lifespan of an α-synuclein-expressing NL5901 Caenorhabditis elegans strain, while disease-associated β-synuclein mutants aggravate the symptoms. Decapeptides targeted at the α-/β-synuclein interaction sites are rationally designed, which suppress the LSPS of α-synuclein, rescue the movement defects, and prolong the lifespan of C. elegans NL5901. Together, we unveil a Yin-Yang balance between α- and β-synuclein underlying the normal and disease states of PD and DLB with therapeutical potentials.

Defect Engineering in β-Bi2SeO5/Bi2O2Se Heterostructures for High-Resolution Phototransistor Arrays

Yingjie Zhao,Jiaming Hu,Zhefeng Lou,Lanxin Xu,Wenbin Li,Xiao Lin,Xiaorui Zheng

Analogous to the pivotal SiO 2 /Si junction in modern electronics, the β-Bi 2 SeO 5 /Bi 2 O 2 Se architecture has been demonstrated to enhance the performance of electronic devices. However, its potential to improve the optoelectronic properties of Bi 2 O 2 Se, such as responsivity and detectivity, remains unexplored. The photodetection performance of Bi 2 O 2 Se is primarily limited by intrinsic selenium vacancies at its surface, which lead to low photocurrent and instability. To address this, a defect-engineered β-Bi 2 SeO 5 /Bi 2 O 2 Se heterojunction is constructed with an atomically sharp interface via a developed UV-assisted oxidation strategy. This heterostructure successfully suppresses surface vacancies while enabling dual functionality—surface passivation and photoactive charge separation, resulting in substantially enhanced optoelectronic performance. First-principles calculations confirm a stable type-II band alignment with interfacial transitions enabling efficient carrier dissociation. The visible-near-infrared transparency and high- k of β-Bi 2 SeO 5 further enable top-gated phototransistors with dynamically tunable photoresponse, achieving the largely improved metrics of responsivity (1.2 × 10 4 A W −1 ), detectivity (1.5 × 10 13 Jones), and on/off ratio (2.3 × 10 6 ). Additionally, by using thermal scanning probe lithography, a high-resolution (pixel pitch = 6.5 µm) β-Bi 2 SeO 5 /Bi 2 O 2 Se phototransistor array is fabricated and its imaging capabilities are demonstrated. The results establish an effective defect-engineered strategy with in situ large-area growth capability of β-Bi 2 SeO 5 and high-resolution device patterning, making β-Bi 2 SeO 5 /Bi 2 O 2 Se a promising photoresponsive platform for advanced optoelectronic devices.

Hydrophilicity-Driven Modulation of Amyloid-β(1–40) Fibrillation by Engineered Nanomaterials

Xuecheng Yang,Xiaoyu Zhang,Ge Yu,Yubin Ma,Xin Li,Yiyang Guo,Shuping Wang,Dan Ge,Changying Xue,Kun Jin,Bingbing Sun

Amyloid-β (Aβ) fibrillation is a spontaneous, thermodynamic process governed by nucleation and elongation. While many studies have explored the ability of engineered nanomaterials (ENMs) to modulate Aβ fibrillation, such as inhibitors, promoters, and dual-modulators, the key physicochemical property of ENMs that determines this behavior remains unclear. In this study, we developed a comprehensive library of ENMs with well-controlled physicochemical properties, including surface charges, morphologies, and hydrophilicity, to systematically investigate their effects on Aβ40 fibrillation. We identified hydrophilicity as the primary determinant of ENM-mediated modulation, rather than surface charge or morphology. Thioflavin T (ThT) kinetics assays indicated that hydrophilic ENMs exhibited bidirectional modulation, both promoting and inhibiting fibrillation depending on concentration. This bidirectional effect results from a competition between accelerated nucleation and decelerated elongation. While hydrophobic ENMs exhibited only unidirectional inhibition from the initial nucleation phase, two-dimensional-NMR (2D-NMR) mechanism studies indicated that this difference resulted from specific interactions with Aβ40 residues. Hydrophilic ENMs targeted hydrophilic residues involved in elongation, including Arginine R5, Glycine G9, G25, G33, G37, and G38, Lysine K28, and Alanine A30, while hydrophobic ENMs bound to hydrophobic residues critical for nucleation, such as I31. These findings provide mechanistic insight into NP–peptide interactions and lay a foundation for the rational design of nanomaterials to modulate amyloid fibrillation.

Engineered β-Carotene Hydroxylase with Excellent Thermostability Promotes Zeaxanthin Production in Yeast

Mengying Shan,Zhenlu Li,Herong Wang,Yi Shi,Qi Gao,Ying Wang,Wenhai Xiao,Mingdong Yao

Zeaxanthin, as an important natural pigment and nutrient, is applied in food and cosmetics industries. Zeaxanthin is converted from β-carotenoid by the β-carotene hydroxylase (CrtZ). Limited by the thermostability of CrtZ, zeaxanthin and its derivatives tend to be fermented in Saccharomyces cerevisiae at low temperatures. In this study, CrtZ mutants with improved thermostability and catalytic efficiency were designed via the position-specific scoring matrix (PSSM), and the beneficial mutants were verified in vitro. Moreover, molecular dynamics simulations revealed the thermostability mechanisms of the mutants. Subsequently, CrtZ mutant M83L was introduced into the S. cerevisiae chassis, and the zeaxanthin titer increased by 121.2% compared with that of the CrtZ, reaching 156.8 mg/L in shake flask fermentation at 30 °C. Furthermore, the oxidoreductase RFNR/FD3 system was introduced to match the overexpressed M83L, and the zeaxanthin titer further increased by 138.9% (reaching 374.6 mg/L). Ultimately, 814.6 mg/L zeaxanthin was produced in S. cerevisiae in 5.0 L fed-batch fermentation at 30 °C, which is the highest reported titer in S. cerevisiae. This study not only provides a useful strategy to increase the thermostability of key enzymes but also describes an efficient platform for the biosynthesis of zeaxanthin and its high-value derivatives.

Characterization and in vitro prebiotic activity of polydatin/β-cyclodextrin inclusion complexes

Wu Chuan Chao,Ye You Wen,Cheng Zhao Yang,Li Yu qi,Liu Yan,Wu Hai Xia,Wu Jian Rong,Liu Qing Tao,Huang Yi Na

Polydatin (PD) possesses a variety of pharmacological activities, but its poor aqueous solubility, stability, and bioavailability limit its application. To improve PD’s water solubility, PD/β-CD inclusion complexes were prepared by solution stirring method and characterized by infrared spectroscopy, X-ray diffraction spectroscopy, nuclear magnetic resonance hydrogen spectroscopy, and molecular docking. The results showed that PD and β-CD successfully generated an inclusion complex with a 1:1 host–guest ratio and a stabilization constant of 4362 M −1 . To further explore the beneficial potential, the antioxidant capacity of PD/β-CD after digestion and the pattern of change during digestion were evaluated in stages by simulated digestion and simulated fermentation in vitro. The simulated digestion process hardly changed the total sugar and reducing sugar contents ( P  > 0.05). During the simulated fermentation process, the pH value kept decreasing, and the short-chain fatty acid content in the fermentation products increased significantly ( P  < 0.05), with the acetic acid content showing the highest increment (1.90 ± 0.35 g L −1 ). In addition, at the phylum level, PD/β-CD inclusion significantly increased the relative abundance of Firmicutes and Actinomycetes and significantly decreased the relative abundance of Proteobacteria , relative to the β-CD group. At the genus level, beneficial bacteria such as Bifidobacterium and Lactobacillus proliferated in the PD/β-CD group, and the relative abundance of potentially pathogenic bacteria such as Streptococcus , and Escherichia-Shigella was significantly reduced. The experimental results indicated that PD/β-CD inclusion complexes were a potential prebiotic with high potential in the field of medicine and health care.

Emulsifying and β-carotene encapsulation properties of natural rosin-based phosphate ester

Xinyi Guan,Yufei Wang,Liuxin Yan,Xinyu Jiang,Tong Liu,Jie Song,Chunrui Han

The advancement of functional emulsions and emulsion gels that incorporate natural chemical constituents is a significant area of research. The molecular architecture of natural lecithin is characterized by a combination of hydrophilic and hydrophobic segments, which facilitates its solubility in both aqueous and organic solvents, thereby enhancing its efficacy as an emulsifying agent. The study focused on the structural modification of lecithin by incorporation of natural hard tricyclic diterpene molecules, and the resulting compounds (DDPT) could form oil-in-water emulsions with well-dispersed, regular, rounded droplets in soybean oil. It is noteworthy that stable water-in-oil (W/O) emulsion gels containing 50% dichloromethane can be successfully prepared when the emulsifier concentration exceeds 5 wt%, and the frequency scanning results of the rheological tests show that the energy storage modulus G’ is larger than the loss modulus G". Encapsulation studies involving β-carotene revealed that the emulsions achieved a high encapsulation efficiency of up to 94.7% ± 1.5%. The results indicate that the synergistic interplay between the terpene moiety and lecithin structure in the emulsifier significantly enhances encapsulation of β-carotene. The work presents a viable pathway for a promising avenue for the application of emulsion systems in the encapsulation and stabilization of bioactive compounds.

Structural and Gelation Characteristics of Alkali-Soluble β-Glucan from Poria cocos

Zhixing Li,Chenglei Sun,Fan Wang,Zhaofei Xia

Alkali-soluble polysaccharides fromPoria cocos(APCP) are typically discarded due to poor water solubility and limited bioavailability, despite their β-(1→3)-glucan backbone suggesting potential for functional applications. This study aimed to explore the structural characteristics, gelation behavior, and the capacity of APCP to reduce silver ions. Structural analysis confirmed that APCP is a homogenous β-(1→3)-D-glucan with a molecular weight of 314.2 kDa and a PDI of 1.32. A pH-mediated strategy enabled the formation of stable single-component APCP hydrogel (APCPH) with tunable mechanical strength, high swelling capacity (>590%), and thermal stability. The APCPH further acted as both a reducing and stabilizing matrix for in situ AgNP formation. Notably, the Ag-APCP hydrogel exhibited distinct antibacterial activity, with inhibition zones reaching 5.31 mm againstStaphylococcus pseudintermedius. These findings demonstrate the feasibility of transforming underutilized APCP into multifunctional hydrogel platforms for green nanomaterial synthesis and biomedical applications. Future studies will focus on optimizing AgNP synthesis parameters and evaluating long-term stability and biocompatibility for translational use in antimicrobial therapies.

Photocatalytic degradation of perfluorooctanoic acid with β-Ga2O3 in anoxic aqueous solution

Baoxiu Zhao,Mou Lv,Li Zhou

Perfluorooctanoic acid (PFOA) is a new-found hazardous persistent organic pollutant, and it is resistant to decomposition by hydroxyl radical (HO) due to its stable chemical structure and the high electronegativity of fluorine. Photocatalytic reduction of PFOA with β-Ga 2 O 3 in anoxic aqueous solution was investigated for the first time, and the results showed that the photoinduced electron (e − cb ) coming from the β-Ga 2 O 3 conduction band was the major degradation substance for PFOA, and shorter-chain perfluorinated carboxylic acids (PFCAs, C n F 2n+1 COOH, 1 ≤ n ≤ 6) were the dominant products. Furthermore, the concentration of F − was measured by the IC technique and defluorination efficiency was calculated. After 3 hr, the photocatalytic degradation efficiency was 98.8% and defluorination efficiency was 31.6% in the presence of thiosulfate and bubbling N 2 . The degradation reaction followed first-order kinetics ( k = 0.0239 min −1 , t 1/2 = 0.48 hr). PFCAs (C n F 2n+1 COOH, 1 ≤ n ≤ 7) were detected and measured by LC-MS and LC-MS/MS methods. It was deduced that the probable photocatalytic degradation mechanism involves e − cb attacking the carboxyl of C n F 2n + 1 COOH, resulting in decarboxylation and the generation of C n F 2n + 1 . The produced C n F 2n + 1 reacted with H 2 O, forming C n F 2n + 1 OH, then C n F 2n + 1 OH underwent HF loss and hydrolysis to form C n F 2n+1 COOH.
