Preparation and characterization of porous starch/β-cyclodextrin microsphere for loading curcumin: Equilibrium, kinetics and mechanism of adsorption

Xinying Zheng,Chao Qiu,Jie Long,Aiquan Jiao,Xueming Xu,Zhengyu Jin,Jinpeng Wang

In this study, porous starch/β-cyclodextrin microsphere (PCS-CD) was fabricated for the first time through the crosslink reaction between corn starch (CS) and β-cyclodextrin (β-CD) combined with enzymatic hydrolysis . The structure, morphology, particle size, thermal property and pasting property were characterized by X-ray diffraction (XRD), scanning electronic microscope (SEM), laser light scattering particle size analyzer and differential scanning calorimetry (DSC), respectively. The results showed that the combination significantly increased the adsorption capacity of hydrophobic substance, which was evaluated by oil adsorption (316 g/g), and achieved an improvement of thermal stability (for an increased pasting temp at 86.5 °C) and shearing resistance (for a decreased breakdown viscosity at 148 cP). The water solubility index (WSI), swelling power index (SPI) and paste clarity were decreased after being crosslinked, but increased slightly after enzymatic hydrolysis. Furthermore, the adsorption kinetics and mechanism of curcumin on the PCS-CD were also evaluated. The results of adsorption kinetics showed that the adsorption process could be described by Pseudo-first-order model and was controlled by two or more diffusion mechanism. The free energy calculated by adsorption isotherm model was 0.1017 kJ/mol, which revealed that a physical-controlled adsorption was occurred between the PCS-CD and curcumin.

Nitrogen-doped fluorescence carbon dots as multi-mechanism detection for iodide and curcumin in biological and food samples

Xiaodan Tang,Hongmei Yu,Brian Bui,Lingyun Wang,Christina Xing,Shaoyan Wang,Mingli Chen,Zhizhi Hu,Wei Chen

Iodine ion is one of the most indispensable anions in living organisms, particularly being an important substance for the synthesis of thyroid hormones. Curcumin is a yellow-orange polyphenol compound derived from the rhizome of Curcuma longa L. , which has been commonly used as a spice and natural coloring agent, food additives, cosmetics as well as Chinese medicine. However, excess curcumin may cause DNA inactivation, lead to a decrease in intracellular ATP levels, and trigger the tissue necrosis. Therefore, quantitative detection of iodine and curcumin is of great significance in the fields of food and life sciences. Herein, we develop nitrogen-doped fluorescent carbon dots (NCDs) as a multi-mechanism detection for iodide and curcumin in actual complex biological and food samples, which was prepared by a one-step solid-phase synthesis using tartaric acid and urea as precursors without adding any other reagents. An assembled NCDs-Hg 2+ fluorescence-enhanced sensor for the quantitative detection of I − was established based on a fluorescence “turn-off-on” mechanism in a linear range of 0.3–15 μM with a detection limit of 69.4 nM and successfully quantified trace amounts of I − in water samples and urine sample. Meanwhile, the as-synthesized NCDs also can be used as a fluorescent quenched sensor for curcumin detection based on the synergistic internal filtration effect (IFE) and static quenching, achieving a good linear range of 0.1–20 μM with a satisfactory detection limit of 29.8 nM. These results indicate that carbon dots are potential sensing materials for iodine and curcumin detection for the good of our health.

Co-assembly of curcumin and a cystine bridged peptide to construct tumor-responsive nano-micelles for efficient chemotherapy

Yemei Dai,Zelei Jiang,Jingyi Li,Mengfan Wang,Chang Liu,Wei Qi,Rongxin Su,Zhimin He

The effective uptake and release of hydrophobic antitumor drugs in cancer cells is a practical challenge for tumor chemotherapy. Many methods were developed to conquer it through modifying drug molecules with hydrophilic groups, or fabricating nanodrugs based on hydrophilic materials. In recent years, peptides have attracted significant interest as part of a promising platform for fabricating nanodrugs due to their low cytotoxicity, favorable variability and self-assembly property. In this study, a cystine bridged peptide (CBP) was designed to co-assemble with a hydrophobic antitumor drug curcumin (CCM), to form a tumor-responsive nanodrug. The hydrophilicity of the peptide promotes the water-dispersity of nanodrugs, and the disulfide bond in cystine, which is cleavable by glutathione (GSH), was involved considering the overexpressed GSH in tumor microenvironments. In vitro and in vivo tests on cervical cancer cells revealed that the obtained nanodrug can rapidly dissociate at tumor sites and inhibit the tumor growth with limited side effects on healthy tissues.

Effect of combination of curcumin and GLUT-1 AS-ODN on radiosensitivity of laryngeal carcinoma through regulating autophagy

Li-Bo Dai,Qi Yu,Shui-Hong Zhou,Yang-Yang Bao,Jiang-Tao Zhong,Li-Fang Shen,Zhong-Jie Lu,Jun Fan,Ya-Ping Huang

Background This study is to explore the role of curcumin and GLUT-1 antisense oligodeoxynucleotides (AS-ODN) on autophagy modulation-initiated radiosensitivity. Methods BALB/c mice were employed to establish xenograft model using Tu212 cell. The expression of autophagy- and apoptosis-related proteins was determined by WB. Autophagosome was observed under transmission electron microscope. Apoptosis of tumor tissue were detected by TUNEL staining. Results Combinations of curcumin and GLUT-1 AS-ODN with 10 Gy inhibited the tumor growth by inducing apoptosis of laryngeal cancer cells followed with the enhancement of autophagy. 3-MA also had a promotion effect on irradiation-mediated growth inhibition possibly by depressing PI3K and on curcumin/GLUT-1 AS-ODN-mediated growth inhibition potentially by regulating autophagic events. Of note, a de-escalation of radiotherapy dose (5 Gy) along with curcumin, GLUT-1 AS-ODN or 3-MA produced a stronger effect than high dosage of radiotherapy (10 Gy) alone. Conclusions Curcumin and GLUT-1 AS-ODN improve the radiosensitivity of laryngeal carcinoma through regulating autophagy and inducing apoptosis.

Glycyrrhetinic acid modified and pH-sensitive mixed micelles improve the anticancer effect of curcumin in hepatoma carcinoma cells

Jizheng Song,Yuling Liu,Longfei Lin,Ye Zhao,Xiuqing Wang,Ming Zhong,Tanggui Xie,Yuting Luo,Shaojing Li,Ruocong Yang,Hui Li

Curcumin (CUR), a natural polyphenolic compound existing in plants, exhibits anticancer potential in inhibiting the growth of various types of human cancer. However, the poor aqueous solubility and low bioavailability limit its clinical applications. pH-sensitive macromolecule F68-acetal-PCL (FAP) and active targeting macromolecule F68-glycyrrhetinic acid (FGA) were designed to fabricate mixed micelles for efficient delivery of CUR. The thin film hydration method was used to prepare CUR loaded mixed (MIX/CUR) micelles. The drug loading rate (DL) of MIX/CUR micelles was 6.31 ± 0.92%, which remained stable for 15 days at 4 °C. The particle size and zeta potential of the MIX/CUR micelles were 91.06 ± 1.37 nm and −9.79 ± 0.47 mV, respectively. The MIX/CUR micelles exhibited pH sensitivity in a weak acid environment, and showed rapid particle size variation and drug release. In addition, in vitro tests demonstrated that MIX/CUR micelles induced higher cytotoxicity and apoptosis than free CUR, non-pH-sensitive F68-PCL (FBP)/CUR micelles and pH-sensitive FAP/CUR micelles in SMMC7721 and Hepa1-6 cells. Besides, mixed micelles were more effective than FBP and FAP micelles in a cell uptake experiment, which was medicated by a GA receptor. All in all, these results indicated that MIX/CUR micelles could be regarded as an ideal drug administration strategy against hepatoma carcinoma cells.

Exudate Absorbing and Antimicrobial Hydrogel Integrated with Multifunctional Curcumin-Loaded Magnesium Polyphenol Network for Facilitating Burn Wound Healing

Yan Gong,Pei Wang,Ran Cao,Jiayingzi Wu,Haoran Ji,Mingsong Wang,Chuang Hu,Peng Huang,Xiansong Wang

Burns are among the most common causes of trauma worldwide. Reducing the healing time of deep burn wounds has always been a major challenge. Traditional dressings not only require a lengthy medical procedure but also cause unbearable pain and secondary damage to patients. In this study, we developed an exudate-absorbing and antimicrobial hydrogel with a curcumin-loaded magnesium polyphenol network (Cur-Mg@PP) to promote burn wound healing. That hydrogel was composed of an ε-poly-l-lysine (ε-PLL)/polymer poly(γ-glutamic acid) (γ-PGA) hydrogel (PP) and curcumin-loaded magnesium polyphenol network (Cur-Mg). Because of the strong water absorption property of ε-PLL and γ-PGA, Cur-Mg@PP powder can quickly absorb the wound exudate and transform into a moist and viscous hydrogel, thus releasing payloads such as magnesium ion (Mg2+) and curcumin (Cur). The released Mg2+ and Cur demonstrated good therapeutic efficacy on analgesic, antioxidant, anti-inflammation, angiogenesis, and tissue regeneration. Our findings provide a strategy for accelerating burn wound healing.

Oral delivery of pectin-chitosan hydrogels entrapping macrophage-targeted curcumin-loaded liposomes for the treatment of ulcerative colitis

Ming Wu,Hongrui Ping,Kun Wang,Hui Ding,Meng Zhang,Ziqiong Yang,Qian Du

The oral delivery of anti-inflammatory drugs has been a promising strategy for enhancing the clinical efficacy of ulcerative colitis (UC) treatment strategies. However, achieving site specific drug delivery to colon tissues and target cells is a challenging task for formulation scientists. In this study, macrophages-targeted liposome-loaded pectin-chitosan hydrogels were developed for UC treatment via oral administration. Folate-functionalized cholesterol was synthesized as lipid membrane materials for the liposomes containing curcumin (CUR). The incorporation of the liposomal CUR within pectin-chitosan hydrogels resulted in a matrix that exhibited considerable sensitivity to colonic enzymes during in vitro release. The targeted delivery of hybrids was able to effectively reach macrophages. They also showed enhanced capability to downregulate TNF-α, IL-6, and IL-1β in the lipopolysaccharide-induced Raw 264.7 cells model. DSS-induced mice model   showed improved anti-UC effects, including accelerated mucosal repair and decreased inflammation and modulate the immune balance in the intestinal tissue of mice with colitis, which may be attributable to increased drug accumulation in the colonic lumen and improved internalization to target cells. Therefore, the incorporation of folate-modified liposomes containing CUR and pectin-chitosan physical hydrogels could potentially serve as a favorable approach for treating UC through an oral colon-targeted drug delivery system.

Thiol-modified hyaluronic acid improves the physical stability of curcumin-zein nanoparticles by forming disulfide bonds with zein

Guangshuai Zhang,Mengdi Qin,Mengran Guo,Mo Li,Di Zhang,Yichi Sun,Bingyang Liu,Zhonggui He,Qiang Fu

Zein-based nanoparticles have been developed in the food industry. However, their poor pH stability and unfavorable ionic strength stability remain a challenge even with the use of polysaccharides (such as hyaluronic acid) as stabilizers. To address this shortcoming, an improved strategy based on the disulfide bonds between thiol-modified hyaluronic acid (HASH) and zein was proposed. In this study, curcumin-zein nanoparticles (ZNs-HASH) were prepared with HASH as a stabilizer. The ZNs-HASH displayed similar particle sizes and spherical structures with ZNs and ZNs-HA (HA as a stabilizer). The Fourier transform infrared spectroscopy demonstrated the formation of disulfide bonds between zein and HASH. Among the three formulations tested, ZNs-HASH exhibited the highest pH and salt ion stability and the strongest antioxidant capacity. This study provided new insights for the improvement of physical stability of zein nanoparticles and the development of oral bioactive substances by chemical modification of natural polysaccharides.

Preparation of nano-sized multi-vesicular vesicles (MVVs) and its application in co-delivery of doxorubicin and curcumin

Qiankun Sun,Ju Liang,Yang Lin,Yunyun Zhang,Fuqing Yan,Wenlan Wu

Multi-vesicular vesicles (MVVs) offer structural advantages in terms of drug encapsulation and physiological stability. In this study, we address the challenge of preparing small-sized MVVs for drug delivery. The nano-sized MVVs (~120   nm) loaded with doxorubicin (DOX) and curcumin (CUR) (DOX/ [email protected] ) were successfully prepared using a glass bead combined with a thin film dispersion method. Transmission electron microscopy (TEM) and dynamic light scattering (DLS) analysis confirmed the independent non-homocentric vesicle structures of DOX/ [email protected] with homogeneous particle sizes. The experimental results showed high encapsulation rates of DOX and CUR in DOX/ [email protected] , reaching 82.5±0.75% and 85.9±0.69%, respectively. Moreover, the MVVs exhibited good biosafety and sustained release properties. Notably, the bioavailability of DOX and CUR in DOX/ [email protected] was enhanced compared to free DOX and CUR, with increases of 4.2 and 2.1 times, respectively. And the half-life of DOX and CUR was extended by 10 times in DOX/ [email protected] In vivo antitumor experiments demonstrated that nano-sized DOX/ [email protected] significantly improved the antitumor activity while reducing the toxic side effects of DOX. Overall, the successful preparation of nano-sized DOX/ [email protected] and their potent and low-toxic antitumor effects provide a critical experimental reference for the combined antitumor therapy of MVVs and liposomes.

Utilizing the Photodynamic Properties of Curcumin to Disrupt Biofilms in Cutibacterium acnes: A Promising Approach for Treating Acne

Nana Zheng,Yuanyuan Xie,Meng zhou,Yuzhen Liu,Haoxiang Xu,Rong Zeng,Chunping Wan,Min Li

Background : The treatment of acne vulgaris is often challenging due to the antibiotic resistance frequently observed in Cutibacterium acnes ( C.acnes ), a prevalent bacterium linked to this condition. Objective : The objective of this research was to examine the impact of curcumin photodynamic therapy (PDT) on the survival of C.acnes and activity of biofilms produced by this microorganism. Methods : Following the Clinical and Laboratory Standards Institute (CLSI) guidelines, we assessed the drug sensitivity of 25 clinical C.acnes strains to five antibiotics (erythromycin, clindamycin, tetracycline, doxycycline, minocycline) and curcumin by implementing the broth microdilution technique. In addition, we established C.acnes biofilms in a laboratory setting and subjected them to curcumin-PDT(curcumin combined with blue light of 180 J/cm 2 ). Afterwards, we evaluated their viability using the XTT assay and observed them using confocal laser scanning microscopy. Results : The result revealed varying resistance rates among the tested antibiotics and curcumin, with erythromycin, clindamycin, tetracycline, doxycycline, minocycline, and curcumin exhibiting resistance rates of 72%, 44%, 36%, 28%, 0%, and 100%, respectively. In the curcumin-PDT inhibition tests against four representative antibiotic-resistant strains, it was found that the survival rate of all strains of planktonic C. acnes was reduced, and the higher the concentration of curcumin, the lower the survival rate. Furthermore, in the biofilm inhibition tests, the vitality and three-dimensional structure of the biofilms were disrupted, and the inhibitory effect became more significant with higher concentrations of curcumin. Conclusion : The results emphasize the possibility of using curcumin PDT as an alternative approach for the treatment of C.acnes , especially in instances of antibiotic-resistant variations and infections related to biofilms.

Curcumin-Containing polyphosphazene nanodrug for Anti-Inflammation and nerve regeneration to improve functional recovery after spinal cord injury

Xiaochen Su,Xunan Jing,Wanting Jiang,Meng Li,Kai Liu,Menghao Teng,Daquan Wang,Lingjie Meng,Yingang Zhang,Wenchen Ji

The microenvironment of excessive inflammation and the activation of apoptotic signals are primary barriers to neurological recovery following spinal cord injury (SCI). Thus, long-lasting anti-inflammation has become an effective strategy to navigate SCI. Herein, a curcumin (CUR)-containing nanosystem (FCTHPC) with high drug loading efficiency was reported via assembling hydrophobic CUR into cross-linked polyphosphazene (PPZ), and simultaneous loading and coordinating with porous bimetallic polymers for greatly enhanced the water-solubility and biocompatibility of CUR. The nanosystem is noncytotoxic when directing its biological activities. By inhibiting the expression of pro-inflammatory factors (IL-1β, TNF-α and IL-6) and apoptotic proteins (C-caspase-3 and Bax/Bcl-2), which may be accomplished by activating the Wnt/β-catenin pathway, the versatile FCTHPC can significantly alleviate the damage to tissues and cells caused by inflammation and apoptosis in the early stage of SCI. In addition, the long-term in vivo studies had demonstrated that FCTHPC could effectively inhibit the formation of glial scars, and simultaneously promote nerve regeneration and myelination, leading to significant recovery of spinal cord function. This study emphasises the promise of the biocompatible CUR-based nanosystem and provides a fresh approach to effectively treat SCI.

Photodynamic inactivation of curcumin combined with ascorbic acid against Penicillium italicum in vitro and on fresh-cut orange

Yang Wang,Yuling Zhao,Rongrong Wu,Jingwen Gao,Menghan Chen,Yue Cui,Jianxiong Hao,Junhua Han,Karl Matthews

This study investigated the antifungal effect of curcumin-mediated photodynamic inactivation (PDI) with ascorbic acid (Vc) as a synergist on Penicillium italicum and its biofilms, and evaluated the efficacy in inactivating P. italicum on fresh-cut orange. Results showed that the number of P. italicum spores decreased by 4.61 Log CFU/mL when the concentration of curcumin, Vc and illumination time was 75 μmol/L, 250 mg/L and 20 min, respectively. After PDI treatment, the cell structure of P. italicum was destroyed and leakage of cell contents was observed. Mature P. italicum biofilms were formed after 72 h of incubation, and PDI could reduce the spores in biofilms by 3.23 Log CFU/mL. In addition, under the optimal photodynamic conditions, the number of P. italicum spores in fresh-cut orange decreased by 2.48 Log CFU/g, and the sensory quality was well maintained. In conclusion, curcumin-mediated photodynamic technology could better maintain the freshness of fresh-cut orange. Therefore, this study deepened the understanding of the antimicrobial efficacy of curcumin-mediated PDI on food borne pathogens and its impact on food quality.
