Development of a Gemini Interfacial Antioxidant for Oil in Water Emulsion with Gallic Acid and Dodecyl Gemini Chains

Peifei Wang,Songbai Liu

In this study, development of the gemini gallate (GG) interfacial antioxidant for oil in water (O/W) emulsion was performed employing Steglich esterification with gallic acid and dodecyl gemini chains through a prepacked column and peristaltic pump-based purification system. The structural identity and purity of the prepared GG and monogallate (MG) were confirmed by NMR, Fourier transform infrared spectroscopy, and high-performance liquid chromatography–mass spectrometry. Further evaluation revealed that the GG possessed excellent radical scavenging activity and superior antioxidant activity in an O/W emulsion relative to the MG, especially under the condition of a reduced amount of the emulsifier. Microscopic investigation by a fluorescent probe and transmission electron microscopy (TEM) unveiled that the extraordinary antioxidant performance in the O/W emulsion was presumably attributed to preferable interfacial location because of the unique gemini molecular architecture. The superior antioxidant activity of the gemini antioxidant holds great promise for antioxidation in O/W emulsions.

Black goji berry (Lycium ruthenicum) tea has higher phytochemical contents and in vitro antioxidant properties than red goji berry (Lycium barbarum) tea

Liu Bei,Xu Qingqing,Sun Yujing

Goji berry tea, a traditional herbal tea, is the main ate mode of goji berry in Asia, yet few studies in comparison with red goji berry tea and black goji berry tea are carried out. This study investigated the effects of water temperature and soak time on the colour, phytochemicals, and the antioxidant capacity [2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl radical (DPPH), and the ferric-reducing antioxidant power (FRAP)] of two goji berry tea. A comparison of the bioactive compounds and antioxidant activities between black and red goji berry tea was conducted. Results showed that both red and black goji berry tea were rich in phytochemicals, giving high antioxidant ability. The levels of bioactive compounds and the antioxidant activity of the two goji berry tea increased as the increases in soak temperature and time. Black goji berry tea had higher phytochemicals and antioxidant property than those of red goji berry tea. Infused at 100° water for the same time, the levels of total polysaccharides (150 mg/100 ml), total polyphenols (238 mg/ml), and antioxidant capacity (550 μmol/100 ml) of black goji berry tea were 3.5, 2, and 5 times higher, respectively, in comparison with red goji berry tea. The results of this study demonstrate that hot drink of goji berry in China is a good habit and black goji berry tea may be a better choice.

The antioxidant activity and composition of the seed oil of mulberry cultivars

Xiao-Hui Yao,Yu-Shi Shen,Run-Ze Hu,Ming Xu,Jia-Xing Huang,Chen-Xi He,Fu-Liang Cao,Yu-Jie Fu,Dong-Yang Zhang,Wei-Guo Zhao,Li Liu,Tao Chen

Mulberry is a traditional crop that has been grown in China for thousands of years. While leaves and fruits of the mulberry have been consumed, mulberry seed use has been less extensive. The goal of this study was to investigate the value of the seeds. Seven cultivars of mulberry were investigated. Oil was extracted from seeds, and the oil content was determined. The primary components of the oils were analyzed using gas chromatography . It was found that all the mulberry cultivars contained high linoleic acid contents, suggesting that mulberry seed may be a potential resource of linoleic acid. The total phenolic and flavonoid contents were also compared among different mulberry seed oils . The physical and chemical parameters, as well as the antioxidant activity of the mulberry seed, were determined. Principal component analysis indicated that the ferric ion reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging activity (DPPH) of mulberry seed oil may be due to high phenolic compounds and linoleic acid. The cultivars Dongsheng, Fengtian, and Guisang had the highest antioxidant activities (FRAP, DPPH), and it was proposed that these cultivars would produce the highest quality oil for use as an ingredient for healthcare or functional food use.

Detection of microalgae single-cell antioxidant and electrochemical potentials by gold microelectrode and Raman micro-spectroscopy combined with chemometrics

Mostafa Gouda,Kai Chen,Xiaoli Li,Yufei Liu,Yong He

Microalgae single-cell electrochemical characteristics are playing an important key role in its industrial application and functionality. This study conducted a single-cell electrochemical study to explain the influence mechanism of intracellular phenolic and flavonoid contents of microalgae on its surface electric charge. In the method, four microalgae species, Arthrospira platensis (AP), Chlorella vulgaris (CV), Phaeodactylum tricornutum (PT), and Scenedesmus obliquus (SO), were studied through single-cell electrochemical current (SCC), single-cell micro-Raman spectrometry (SCMR), and other physicochemical methods. Also, they were correlated with the chemical analytical methods. In the results, PT and AP phenolics, flavonoids, and antioxidant activities were significantly high compared to CV and SO. On the other hand, SO had the highest SCC compared to AP and PT. Also, SCMR at 1514 cm −1 was negatively correlated with antioxidant activity by nitric oxide (r 2 = -0.80). In conclusion, single-cell current measurement by gold microelectrode can verify the antioxidant activity of the different microalgae species. Besides, this work provides a simple micro-level study of the single-cell phytochemicals to better explain their exact functional impacts compared to the methods of macro-analyses that use millions of cells, which reduces their sensitivity. In which, it can be applied in the real-time functionality evaluation during microalgae cultivation.

The modifications of a fructan from Anemarrhena asphodeloides Bunge and their antioxidant activities

Jiaojiao Zhang,Zhengguo Li,Linan Zhou,Jiahe Bao,Jing Xu

Polysaccharide modification exerts important significance for enhancing biological activity. In the present study, a fructan (AAP70-1) extracted from Anemarrhena asphodeloides Bunge was modified by carboxymethylation and sulfation modifications to obtain carboxymethylated derivatives (CM-AAP70-1s) and a sulfated derivative (S-AAP70-1), which were prepared by the chloroacetic acid-isopropanol and the chlorosulfonic acid-pyridine methods, respectively. Physicochemical characteristics and antioxidant activities in vitro of modified derivatives were determined. The results of degree of substitution (DS) and FT-IR analysis indicated that the carboxymethylated and sulfated groups were introduced successfully. After modifications, the DS of CM-AAP70-1s varied from 0.18 to 0.52 and the molecular weights (Mw) were between 4.50 × 10 3 and 8.65 × 10 3  g/mol. The DS and Mw of S-AAP70-1 were calculated to be 0.61 and 9.76 × 10 3  g/mol, respectively. The assays of the antioxidant properties demonstrated that the modified derivatives exhibited stronger scavenging effects toward DPPH and hydroxyl radicals compared with AAP70-1. These results revealed that the chemical modifications could effectively improve the potential of polysaccharides as oxidation inhibitor.

Black rice (Oryza sativa L.) processing: Evaluation of physicochemical properties, in vitro starch digestibility, and phenolic functions linked to type 2 diabetes

Halah Aalim,Di Wang,Zisheng Luo

Black rice is recognized for managing diabetes in Chinese folk medicine. Therefore, the present study investigates the effect of thermal treatments and the succeeding cooking on black rice physicochemical properties, phenolic composition, total antioxidant activity (TAA), enzymes and glycation inhibition in addition to starch digestibility. Thermal decomposition of anthocyanin and cyanidin-3-glucoside was evident across all processing methods and reflected in increasing levels of protocatechuic acid, while proanthocyanidins (TPAC) were susceptible to cooking. Roasting of grains sustained total phenolics (TPC), flavonoids (TFC), TPAC, and antilipase activity. Additionally, the combined effect of frying and cooking diminished TFC, TPAC, and α-glucosidase inhibition. The thermally treated grains showed pronounced activity against α-amylase, α-glucosidase, and glycation, whereas their cooked counterparts reduced the estimated glycemic index (eGI), and enhanced resistant starch (RS). Processed grains chrominance, TAA, and apparent amylose content (AAC) showed a significant correlation with phenolics. These findings are demonstrating that black rice processing is favorable for the dietary management of metabolic disorders such as diabetes and hyperlipidemia.

Preparation, characterization and antioxidant activity of protocatechuic acid grafted carboxymethyl chitosan and its hydrogel

Chao Xu,Shui Guan,Jianqiang Xu,Weitao Gong,Tianqing Liu,Xuehu Ma,Changkai Sun

In this study, protocatechuic acid (PCA) was grafted onto carboxymethyl chitosan (CMCS) via EDC/NHS to improve the antioxidant effect. The grafting ratio of PCA-g-CMCS conjugates could be controlled by adjusting the pH value and feed ratio of raw materials. The conjugates exhibited similar pH sensitivity to CMCS and showed dramatic enhancements of DPPH and ABTS radicals scavenging activities, total antioxidant capacity, reducing power, and Fe 2+ -chelating activity. Three-dimensional porous PCA-g-CMCS hydrogel was prepared by lyophilization and secondary cross-linking. The shaped hydrogel preserved its antioxidant activity, and the sustained release of PCA-containing degraded fragment from biodegradable hydrogel could be achieved with the aid of lysozyme in vitro (15 days). PCA-g-CMCS hydrogel also showed excellent biocompatibility and protective effect on H 2 O 2 -induced oxidative damage in SH-SY5Y cells. These results suggested that PCA-g-CMCS conjugates and its hydrogel would appear to be a promising oxidation-resistant material for applications such as drug release and tissue engineering.

Stability of bioactive compounds and in vitro gastrointestinal digestion of red beetroot jam: Effect of processing and storage

Tao Wang,Lihua Liu,Aizhan Rakhmanova,Xin Wang,Yuanyuan Shan,Yanglei Yi,Bianfang Liu,Yuan Zhou,Xin Lü

The stability of total phenolics, flavonoids and betalains , as well as antioxidant capacity of red beetroot during jam processing, storage and in vitro gastrointestinal digestion were measured using spectrophotometry . Results suggested that jam processing led to a significant reduction of total phenolics (13.8%), betalains (56.3%) and antioxidant capacity (13.2–28.2%), but a significant increase of total flavonoids (99.3%). After storage at 4 °C for 90 days, the losses of total phenolics, flavonoids, betalains and antioxidant capacity were 33.9, 10.6, 7.9 and 9.7–35.7%, respectively. In addition, jam processing improved the bioaccessibility of total phenolics, flavonoids, betalains and antioxidant capacity using in vitro simulated gastrointestinal digestion analysis. The results of this study suggested that the main bioactive compounds of red beetroot including phenolics, flavonoids and betalains, could be partially preserved during jam processing and storage, with higher levels of bioaccessibility. Therefore, the red beetroot was a promising material for the development of functional foods.

Biotransformation of two citrus flavanones by lactic acid bacteria in chemical defined medium

Guo Xiao,Guo Ailing,Li Erhu

Microbial processes are being developed to transform flavonoid glycosides to varieties of metabolites with higher bioavailability. The aim of this study was to determine the metabolic activity and survival of five lactic acid bacteria (LAB) stains ( L. rhamnosus LRa05, L. casei LC89, L. plantarum N13, L. acidophilus LA85, and L. brevis LB01) in two different citrus flavanone standards (hesperetin-7-O-rutinoside and naringenin-7-O-rutinoside). The enzymatic activity, metabolites, antioxidant activities, and α-glucosidase inhibition property in the two standards were also investigated before and after incubated with LAB. The medium contained standards permitted survival of the five LAB stains. All strains exhibited β-glucosidase activity. Of the five LAB strains tested, just L. plantarum N13 and L. brevis LB01 have the ability to metabolize hesperetin-7-O-rutinoside, only L. plantarum N13, L. acidophilus LA85, and L. brevis LB01 could metabolize naringenin-7-O-rutinoside, moreover, L. acidophilus LA85l was the strain with the highest biotransformation ratio of naringenin-7-O-rutinoside. L. acidophilus LA85 and L. plantarum N13 can degrade naringenin-7-O-rutinoside into naringenin. L. brevis LB01 can degrade hesperetin-7-O-rutinoside into hesperetin, 3-(4′-hydroxyphenyl)-2-propenoic acid, 3-(3′-hydroxy-4′-methoxyphenyl)hydracrylic acid, and 3-(4′-hydroxyphenyl)propionic acid. Incubation of L. acidophilus LA85 in naringenin-7-O-rutinoside solution supposed no apparent influence in the biological activities that tested. L. acidophilus LA85 may potentially contribute to the bioavailability of citrus flavanones, and to be applied as functional cultures to obtain more bioavailable and bioactive metabolites in food products or in the human gastrointestinal tract.

The key role of persistent free radicals on the surface of hydrochar and pyrocarbon in the removal of heavy metal-organic combined pollutants

Yanzhuo Zhang,Xuedi Sun,Wei Bian,Jianbiao Peng,Huilin Wan,Jing Zhao

We show that persistent free radicals (PFRs) on the surface of biochar can produce hydroxyl radicals (•OH) by catalyzing H 2 O 2 to facilitate the removal of the combined pollutant BPA-Cr(VI). Microstructure characterization showed that the structures of pyrocarbon and hydrochar were significantly different when prepared at different temperatures. As the preparation temperature and preparation time for biochar increased, the concentration of PFRs first increased and then decreased. When biochar, PFRs, and H 2 O 2 were present in the same solution, the single pollutants BPA and Cr(VI) as well as the combined pollutant BPA-Cr(VI) could be removed effectively, with removal rates greater than 90%. However, when PFRs, BPA, H 2 O 2 , and Cr(VI) were present in the same solution, Cr(VI) competed with H 2 O 2 for electrons and promoted the removal of BPA. The results of this study could be applied to sludge recycling and be used to develop approaches to catalytically degrade combined pollutants.

Preparation of a microfibrillated cellulose/chitosan/polypyrrole film for Active Food Packaging

Qian Gao,Min Lei,Kemeng Zhou,Xinliang Liu,Shuangfei Wang,Huimin Li

The objective of this research was to develop an effective and complete green approach for the production of composite films with enhanced antioxidant and antibacterial properties and oxygen barrier properties. This was accomplished by incorporating chitosan (CS) and polypyrrole (PPy) to microfibrillated by coasting method prepare MFC/CS/PPy film. The contact angle was used to evaluate poor water resistance, which increased from 94.1° to 122°. The contact angle of the composite film was further increased to 122° due to the chemical polymerization of pyrrole produces a layer of PPy nanoparticles on the surface of the MFC/CS composite film. The CS and PPy improved the uniformity of the surface and filled the pores, indicating that the channel has been blocked, resulting in significant reduction in oxygen transmission (32.38 cm 3 (m −2 ·day −1 ). The MFC/CS/PPy film showed bactericidal activity against Escherichia coli (5.43 log CFU mL −1 reduction) The synergistic effect of proton amino (in acidic medium) in CS with the positively charged PPy chain improves the antimicrobial performance of MFC/CS/PPy. The positive charges Ions may attach to the negatively charged bacterial cell wall, thus destroying the cell wall and eventually killing the bacteria. The composite films have good thermal stability. The ability of MFC/CS/PPy composite membrane to inhibit 2,2-diphenyl-1-picrylhydrazyl assay (DPPH) activity increases to 54 %. This property depends on the increased activity of the hydrogen atoms. Considering a series of current properties, MFC/CS/PPy composite film was successfully used in the active packing of fresh cherry tomatoes (10 days), effectively extending shelf life. Therefore, MFC/CS/PPy composite film has the potential to be used as biodegradable packaging materials for fresh fruits.

Evaluation on the physicochemical and digestive properties of melanoidin from black garlic and their antioxidant activities in vitro

Jianfu Wu,Yan Jin,Min Zhang

As the important compounds in black garlic, the physicochemical properties and bioactivities of melanoidin (MLD) were investigated in this study. The results showed that MLD possessed strong metal-ion chelating capacity and radical scavenging activities which were positively correlative with molecular weight (MW). During the simulated digestion in vitro , the ultraviolet absorption, browning degree and MW distribution of MLD remained the same as initial. It proved that the MLD from black garlic could be indigestible like the dietary fiber with little loss of volatile compounds and polysaccharides. Remarkably, the bioactivities of MLD were reduced significantly under the treatment of α-amylase or hydrochloric acid, while they were stable and retained over 60% after adding pepsin and pancreatin. This study provides fundamental evidences for further research and widely application of MLD and black garlic in the production of functional food or food additives .
