NIR-715 photodynamic therapy induces immunogenic cancer cell death by enhancing the endoplasmic reticulum stress response

Zheng Zhen-Yuan,Lin Wan,Su Jia-Wan,Huang Qing-Feng,Zhang Cong,Pan Wen-Xing,Li En-Min,Zhang He-Feng,Xu Li-Yan

Effectively interfering with endoplasmic reticulum (ER) function in tumor cells and simultaneously activating an anti-tumor immune microenvironment to attack the tumor cells are promising strategies for cancer treatment. However, precise ER-stress induction is still a huge challenge. In this study, we synthesized a near-infrared (NIR) probe, NIR-715, which induces tumor cell death and inhibits tumor growth without causing apparent side effects. NIR-715 triggers severe ER stress and immunogenic cell death (ICD) after visible light exposure. NIR-715 induced ICD-associated HMGB1 release in vitro and anti-tumor immune responses, including increased cytotoxic T lymphocyte (GZMB + CD8 + T cell) infiltration and decreased numbers of exhausted T lymphocytes (PD-L1 + CD8 + T cell). These findings suggest that NIR-715 may be a novel agent for “cold” tumor photodynamic therapy (PDT). Schematic illustration of NIR-715 photodynamic therapy for visible light-triggered, endoplasmic reticulum-targeting antitumor therapy.

pH-/Viscosity-Activatable NIR Fluorescent Probes via Acceptor Engineering of Hemicyanine Dyes for High-Contrast Bioimaging

Da Feng,Li Guo,Yihan Zhao,Fang Yuan,Lulu Ning,Yuan Guo,Jianjian Zhang

Real-time monitoring of the lysosomal microenvironment using small-molecule probes is critical for understanding the complex interactions between organelles and diseases associated with lysosomal dysfunction. Most traditional fluorescent-dye-based lysosome probes rely on the protonation of the nitrogen atom in the morpholine unit to visualize the lysosomal microenvironment by inhibiting the photoinduced electron transfer (PET) effect. However, these probes often face selectivity issues within the complex cellular microenvironment. For instance, classic hemicyanine dyes (HD) show nonspecific fluorescence responses in the liver due to inappropriate pKa value, leading to low imaging contrast and the risk of false positives. Herein, a series of novel pH/viscosity-activatable, lysosomal-targeted near-infrared (NIR) fluorescent probes were developed by incorporating naphthalimide dye with hemicyanine dyes. These probes exhibit no fluorescence at physiological pH and weak fluorescence under acidic conditions (key 1), with substantial fluorescence activation triggered by abnormal viscosity in pathological tissues (key 2). Notably, NpCy-4 demonstrated a superior signal-to-background ratio (SBR) for cellular imaging and proved effective for real-time in situ diagnosis of acute gastritis.

Mitochondrial SO2-Activated NIR Photodiagnostic Agent Harnessing Hydroxyl Radicals for Efficient Inflammation Eradication and Tumor Suppression

Yingcui Bu,Yingyu Zhu,Yuxin Yang,Ting Wang,Xiaojiao Zhu,Xingxing Chen,Qiong Zhang,Mingdi Yang,Hongping Zhou

At present, some progress has been made in developing NIR light-responsive free radical generators. However, the efficacy of theranostics continues to be hindered by tumor-associated inflammatory reactions. Hence, fulfilling the in situ release of free radicals upon NIR light excitation specifically activated by the inflammation microenvironment would be an ideal strategy for efficient inflammation eradication and tumor suppression but remains a challenge. Herein, a SO2 (overexpressed reactive sulfur species in inflamed site)-stimulated phototheranostic agent (CVS) is successfully developed. Through a specific response to both endogenous and exogenous SO2 with a low LOD (31.7 nM), CVS demonstrates the “switch on” two-photon activity as well as efficient OH· generation. Remarkably, in CVS-treated H22-tumor-bearing mice, the NIR light-activated accurate inflammation eradication and tumor suppression are accomplished. This reactive phototheranostic platform not only facilitates the quantification of SO2 during inflammation but also renders it a potent NIR antihypoxic tumor agent.

NIR-II aggregation-induced emission nanoparticles camouflaged with preactivated macrophage membranes for phototheranostics of pulmonary tuberculosis

Yan Dingyuan,Li Xue,Wang Huanhuan,Li Bin,Wang Wei,Liao Yuhui,Tang Ben Zhong,Wang Dong

Phototheranostics, which allows simultaneous diagnosis and therapy, offers notable advantages in terms of noninvasiveness, controllability and negligible drug resistance, presenting a promising approach for disease treatment. By integrating second near-infrared (NIR-II, 1,000–1,700 nm) phototheranostic agents characterized by aggregation-induced emission (AIE) and cell membranes with specific targeting capacity, we have developed a versatile type of biomimetic nanoparticle (NP) for precise phototheranostics of pulmonary tuberculosis (TB). Coating the phototheranostic agents with preactivated macrophage membranes results in the formation of biomimetic NPs, which exhibit specific binding to TB through a lesion–pathogen dual-targeting strategy, allowing the accurate detection of Mycobacterium tuberculosis via NIR-II fluorescence imaging and precise photothermal therapy using the irradiation of a 1,064 nm laser. In comparison with traditional treatments, small individual granulomas (0.2 mm in diameter) in TB-infected mice are visualized, and improved antibacterial effects are achieved upon NP administration. Here we present a standardized workflow for the synthesis of the NIR-II AIE agents, their use for the fabrication of the biomimetic NPs and their in vitro and in vivo applications as phototheranostics against M. tuberculosis . The preparation and characterization of the NIR-II AIE agents requires ~8 d, the synthesis and characterization of the phototheranostic NPs requires ~8 d, the validation of in vitro targeting capacity and photothermal eradication requires ~26 d, and the in vivo NIR-II fluorescence imaging and imaging-guided photothermal therapy requires ~74 d. All procedures are straightforward and suitable for clinicians or researchers with prior training in organic synthesis and biomedical engineering.

NIR responsive microneedles patch used for promoting spinal cord injury repair by modulating mitochondrial ROS microenvironment

Shuangshuang Chen,Zelin Sang,Yumei Li,Hui Li,Zhenhua Chen,Xifan Mei,Xianbing Dai,Xiuli Ren

Spinal cord injury (SCI) is a serious traumatic disease of the central nervous system. How to effectively deliver drugs to the site of spinal cord injury lesions through the blood spinal cord brain barrier (BSCB) has always been a challenge. There is an urgent need for developing a controllable and effective method to continuously release drug for SCI repair. Microneedles are a promising drug delivery method, however, how to control drug release smartly from microneedles remains a cutting-edge field. Herein, a NIR triggered photothermal responsive microneedles patch with Au@polyphenol nanospheres encapsulated was designed for SCI repair. By regulating the NIR irradiation, the thermal swelling rate of the microneedles can be precisely controlled. Therefore, the releasing of Au@polyphenol nanospheres from the microneedles could be controlled. Gold nanoparticles are known as nanozyme in photothermal therapy. Polyphenol nanospheres released from microneedles are helpful in promoting spinal cord injury repair by modulating mitochondrial ROS microenvironment. This NIR responsive microneedles array patch loaded with gold@polyphenol nanospheres has the advantages of effectively clearing reactive oxygen species, promoting mitochondrial autophagy, rebuilding redox balance, and reducing neuronal apoptosis.

Cu/Au-doped nanopolymers with multiple catalytic activities for NIR II laser-promoted nanocatalytic tumor therapy

Xixi Wu,Xiang Feng,Peng Yu,Jing Zhang,Rui Liu

Breast cancer remains a significant global health concern owing to the limitations of conventional therapies, such as side effects, drug resistance, and high costs. Nanocatalytic therapy has emerged as a promising alternative due to its tumor specificity, spatiotemporal controllability, and noninvasiveness. However, its effectiveness is limited by endogenous antioxidants like glutathione (GSH) and the low catalytic activity of nanocatalysts. Herein, an Cu/Au-doped polypyrrole nanocatalyst, Cu-AuPP, with multiple catalytic activities is developed by sequentially polymerizing pyrrole monomers using CuCl2 and HAuCl4, followed by PEGylation. The obtained Cu-AuPP catalyzes the production of hydroxyl radicals (·OH) and facilitates the oxidation of GSH to GSSG via redox reactions mediated by multivalent Cu ions, leading to oxidative damage, mitochondrial dysfunction, and tumor cell apoptosis. Upon 1064 nm laser irradiation, these catalytic activities were enhanced by elevated temperature and electron–hole separation mediated by Au nanoclusters, resulting in more intense oxidative damage to tumor cells. Collectively, the developed Cu-AuPP nanocatalytic medicine capable of simultaneously catalyzing GSH depletion and ·OH production via several improved catalytic mechanisms, has significant promise for the treatment of malignant tumors.

Hierarchical core–shell structural based on WO3@ITO nanorods with boosted electrochromic performance in NIR region

Xiaoyue Hao,Yong Liu,Gaorong Han,Gaoling Zhao,Dongxue Liu,Huitao Dai,Sainan Ma,Likun Wang

Over half of the solar energy comes from the infrared region, which makes electrochromic materials that modulate infrared transmittance essential for the thermal management of smart windows. However, existing electrochromic materials do not perform well in the near-infrared region, which exhibits slow response time, weak adhesion to conductive substrates, and poor long-term cycling stability. In the present work, we successfully synthesized WO 3 @ITO nanorod samples with hierarchical core–shell structure by hydrothermal and in-situ solvothermal methods. ITO-coated WO 3 nanorods and some ITO nanocrystals were firmly integrated with WO 3 nanorods, resulting in a corn-like hybrid heterogeneous structure. The unique WO 3 @ITO hierarchical core–shell structure improves the electronic properties and the adhesion to substrates, reducing the energy barrier of ion diffusion and increasing the ion flux. Enhanced electrochromic performance in NIR range with a high optical modulation of 53.3 % in 1600 nm and long-term reversibility (maintaining 91.5 % of its original contrast after 1000 cycles) was achieved. In addition, response time (t c  = 3.5 s, t b  = 4.6 s) and coloration efficiency (112.5 cm 2 ‧C -1 ) have been significantly improved, which is almost twice that of unmodified WO 3 nanorods (t b  = 8.6 s and CE = 56.4 cm 2 ‧C -1 ), showing a good application prospect in smart windows.

Direct synthesis of highly efficient NIR-I emitting core-only InP quantum dots via multi-ligand synergy

Rui Jiang,Jie Zhao,Yifei Qiao,Bo Tan,Jialin Yin,Zhenyu Dai,Shiliang Mei,Wanlu Zhang,Ruiqian Guo

Near-infrared (NIR) emission has garnered significant attention owing to its broad application potential in next-generation optoelectronics. Among various candidates, indium phosphide (InP) quantum dots (QDs) have emerged as particularly promising candidate due to their environmentally benign composition and relatively facile synthesis. However, the synthesis of NIR-emitting InP-based QDs exhibiting high luminescence efficiency remains a critical challenge. The inherent difficulties primarily stem from insufficient core nucleation and the complex shell growth processes essential for achieving high-performance QDs. Herein, we report an expeditious synthetic strategy for achieving NIR-I emissive core-only InP QDs with tunable emission ranging from 683 to 742 nm, mediated by the synergistic effect of ZnCl 2 and NH 4 PF 6 . Under the competitive adsorption between ZnCl 2 and NH 4 PF 6 , high-covalent InP QDs with sizes of approximately 6 to 10 nm can be obtained within 5 min. During the later stages in this dual-salt system, PF 6 − effectively eliminates surface oxides while zinc ion enrichment establishes a negatively charged surface. This Zn-related configuration promotes subsequent adsorption and defect passivation by NH 4 + . The final core-only QDs exhibit bright NIR-I emission with a photoluminescence quantum yield of up to 74 % in solution. Furthermore, the application potential of these core-only QDs is demonstrated through simple patterning. This work establishes a straightforward, post-treatment-free synthesis of high-performance NIR-I core-only InP QDs, highlighting their substantial promise for next-generation optoelectronic devices.

Macrophage Membrane-Coated Carbon Nanozymes for Targeted Drug Delivery and NIR-Responsive Synergistic Therapy in Rheumatoid Arthritis

Yuhuan Li,Longying Cong,Wei Zhang,Rongrong Zhang,Yu Gao,Songyun Zheng,Daowei Li,Zhuo Zhang,Lingyu Zhang,Modi Yang,Fei Chang

Rheumatoid arthritis (RA) remains a therapeutic challenge due to the persistently dysregulated synovial microenvironment that drives chronic inflammation and treatment resistance. Herein, a biomimetic nanoplatform composed of macrophage membrane (MM)-coated porous carbon nanospheres (MM@PCNSs) is developed for synergistic immunomodulation and targeted iguratimod (IGU) delivery in RA therapy. The PCNS core integrates multi-enzyme mimetic activities for efficient reactive oxygen species (ROS) scavenging and supports high drug loading with pH/NIR-responsive release. Biomimetic macrophage membrane cloaking enables inflammation-targeted delivery and in situ neutralization of cytokines. In vitro, MM@PCNSs/IGU effectively reduced oxidative stress and inhibited pro-inflammatory cytokines. In vivo, NIR-activated MM@PCNSs/IGU significantly alleviated synovial inflammation, reduced joint destruction, and improved clinical scores in collagen-induced arthritis (CIA) mice. This multifunctional nanoplatform integrates targeted delivery, redox regulation, and immune reprogramming to overcome limitations of conventional therapies, offering a promising strategy for restoring synovial homeostasis and achieving durable remission in RA.

Bimetallic ScAg-MOFs as peroxide-mimicking enzymes combined with NIR spectroscopy for rapid prediction of uric acid

Xiaoyan Duan,Shiyi Chen,Feiyan Tian,Ganghui Chu

In this study, the newly synthesized ScAg-MOFs was used as a like-peroxidase. Based on the positive correlation between uric acid concentration and the (3,3,5,5-tetramethylbenzidine) TMB oxidation inhibition rate, a partial least squares (PLS) model was developed to predict the oxidative inhibition of TMB. The results indicate that the combination of standard normal variable transform and continuous wavelet transform (SNV + CWT) with competitive adaptive reweighted sampling (CARS) yielded a root-mean-square error of cross-validation (RMSECV) of 8.6460%, and a root-mean-square error of prediction (RMSEP) of 7.9731%. The correlation coefficient for the calibration set (R_CV) was 0.9430, for the validation set (R) it was 0.9459, and the prediction deviation ratio (RPD) was 3.0268. This suggests that the model is quite accurate, and it demonstrates the feasibility of this method for rapid uric acid detection.

Single-Doped W18O49-x@Au-Embedded Dual-Network Silk Fibroin Hydrogel for NIR-Responsive Phototherapeutic Wound Healing

Chaofan Yao,Yu Zhao,You Wu,Haoming Wang,Hongbin Li,Jinmei He,Li Liu,Yudong Huang,Feng Cheng

Bacterial infections remain a critical challenge in wound care, demanding advanced multifunctional materials for effective treatment. Here, this work presents a 3D-printable hydrogel (STFWA) integrating a Schottky heterojunction and dual-network architecture for synergistic photothermal and photodynamic antibacterial therapy. The hydrogel incorporates W18O49-x@Au nanostructures, where Au nanoparticles are in situ grown on oxygen-deficient W18O49-x (bandgap ≈1.5 eV), forming a Schottky junction that enhances carrier separation and reactive oxygen species (ROS) generation. The dual-network matrix, composed of silk fibroin and tannic acid-Fe3+ (TA/Fe3+), exhibits robust mechanical integrity and hemostatic capability, while silk fibroin enables high-resolution digital light processing (DLP) 3D printing. The co-existence of W18O49-x and Au induces dual localized surface plasmon resonance (LSPR), while TA/Fe3+ contributes to deep coloration, collectively boosting photothermal conversion. Upon near-infrared (NIR) irradiation, the hydrogel exhibits potent antibacterial efficacy against Staphylococcus aureus and Escherichia coli through combined photothermal and photodynamic mechanisms. In vivo studies using a rat model of infected wounds demonstrate accelerated healing, enhanced angiogenesis, and regulated inflammation. This work highlights a nanostructure-enabled hydrogel platform for NIR-activated, synergistic antibacterial therapy with clinical potential in infected wound management.

An activatable type I photosensitizer for NIR fluorescence imaging and photodynamic therapy of drug-resistant bacterial infections

Zhongkun Wang,Rui Zhu,Chenrui Li,Weiwei Xu,Liangyou Zhao,Guangjin Gao,Weihao An,Xiang Dong,Qingqing Miao,Qing Li

Bacterial infections pose a significant global health challenge, further exacerbated by the emergence of antibiotic-resistant strains, thereby necessitating the development of alternative therapeutic strategies. Type I photosensitizers, as an emerging phototheranostic modality, hold great promise for antibacterial treatment but are limited by poor specificity. Herein, we report a hydrogen sulfide (H 2 S)-activatable theranostic probe (CyIN) that integrates near-infrared (NIR) fluorescence imaging and type I photodynamic therapy (PDT) functionalities into a single platform for the precise treatment of drug-resistant bacterial infections. CyIN is rationally constructed by masking the hydroxyl group of iodine-substituted hemicyanine with a H 2 S-cleavable 2,4-dinitrophenyl group, which efficiently quenches NIR fluorescence and suppresses reactive oxygen species generation. Such molecular configuration allows CyIN to turn on its NIR fluorescence signal and type I photodynamic action upon interaction with bacterial-derived H 2 S, achieving distinguishable detection of drug-resistant bacterial infection, potent bacterial clearance (>99 %), and accelerated healing of bacteria-infected wounds. Furthermore, histological analyses confirm that the CyIN treatment can reduce inflammation, enhance collagen deposition, and promote tissue regeneration without systemic toxicity. Thus, this work presents a promising H 2 S-responsive phototheranostic platform for precise bacterial detection and efficient photodynamic eradication of drug-resistant bacterial infections.
