Intracerebral Hemorrhage-Induced Brain Injury: the Role of Lysosomal-Associated Transmembrane Protein 5

Hua Wei,Ma Shuainan,Pang Yuxin,Liu Qi,Wang Yueying,Liu Zhiyi,Zhao Nan,Ren Naixin,Jin Sinan,Wang Benshuai,Song Yuejia,Qi Jiping

Intracerebral hemorrhage (ICH) is a lethal stroke with high mortality or disability. However, effective therapy for ICH damage is generally lacking. Previous investigations have suggested that lysosomal protein transmembrane 5 (LAPTM5) is involved in various pathological processes, including autophagy, apoptosis, and inflammation. In this study, we aimed to identify the expression and functions of LAPTM5 in collagenase-induced ICH mouse models and hemoglobin-induced cell models. We found that LAPTM5 was highly expressed in brain tissues around the hematoma, and double immunostaining studies showed that LAPTM5 was co-expressed with microglia cells, neurons, and astrocytes. Following ICH, the mice presented increased brain edema, blood-brain barrier permeability, and neurological deficits, while pathological symptoms were alleviated after the LAPTM5 knockdown. Adeno-associated virus 9-mediated downregulation of LAPTM5 also improves ICH-induced secondary cerebral damage, including neuronal degeneration, the polarization of M1-like microglia, and inflammatory cascades. Furthermore, LAPTM5 promoted activation of the nuclear factor kappa-B (NF-κB) pathway in response to neuroinflammation. Further investigations indicated that brain injury improved by LAPTM5 knockdown was further exacerbated after the overexpression of receptor-interacting protein kinase 1 ( RIP1 ), which is revealed to trigger the NF-κB pathway. In vitro experiments demonstrated that LAPTM5 silencing inhibited hemoglobin-induced cell function and confirmed regulation between RIP1 and LAPTM5. In conclusion, the present study indicates that LAPTM5 may act as a positive regulator in the context of ICH by modulating the RIP1/NF-κB pathway. Thus, it may be a candidate gene for further study of molecular or therapeutic targets.

Synthesis of nanosized IM-5 zeolite and its CH4/N2 adsorption and separation

Xuan Tang,Yugao Wang,Mengni Wei,Xueqin Zhang,Ye Li,Xiaomin Li,Jinping Li,Jiangfeng Yang

Reduction of zeolite crystal size from the micron scale to the nanoscale leads to a significant increase in specific surface area, favoring mass-transfer pathways. Here, we have prepared nanoscale (ca. 30–80 nm) IM-5@100 zeolite (IMF type) by a facile strategy of regulating the amount of Al source in the starting gel, and have investigated its CH 4 /N 2 adsorption and separation properties. We found that on decreasing the amount of Al source, the crystal size of the product changed significantly, decreasing from strips of length about 1 μm to irregular nanocrystals. Compared with micron-sized IM-5@50, the mesopore volume of IM-5@100 was increased fourfold from 0.12 cm 3 /g to 0.49 cm 3 /g, its external surface area was increased from 38 m 2 /g to 118 m 2 /g, and its DFT pore size distribution based on N 2 adsorption/desorption showed a mesopore distribution in the range 5–10 nm. Adsorption kinetics tests showed that nano-IM-5@100 can achieve adsorption equilibrium more swiftly and exhibits better mass-transfer performance. Moreover, IM-5@100 showed good adsorption capacity for CH 4 (17.8 cm 3 /g) and high IAST selectivity (4.7) for CH 4 /N 2 (50:50, v/v). Breakthrough experiments have indicated that IM-5@100 permits efficient practical separation of CH 4 /N 2 .

Probe-labeled electrochemical approach for highly selective detection of 5-carboxycytosine in DNA

Mei Zhao,Guangrong Zou,Jing Tang,Jingyi Guo,Fang Wang,Zilin Chen

5-carboxycytosine (5caC) plays a critical role as an intermediate form in DNA methylation and demethylation processes. Its distribution and quantity significantly influence the dynamic equilibrium of these processes, thereby impacting the normal physiological activities of organisms. However, the analysis of 5caC presents a significant challenge due to its low abundance in the genome, making it almost undetectable in most tissues. In response to this challenge, we propose a selective method for 5caC detection using differential pulse voltammetry (DPV) at glassy carbon electrode (GCE), hinging on probe labeling. The probe molecule Biotin LC-Hydrazide was introduced into the target base and the labeled DNA was immobilized onto the electrode surface with the help of T4 polynucleotide kinase (T4 PNK). Leveraging the precise and efficient recognition of streptavidin and biotin, streptavidin-horseradish peroxidase (SA-HRP) on the surface of the electrode catalyzed a redox reaction involving hydroquinone and hydrogen peroxide, resulting in an amplified current signal. This procedure allowed us to quantitatively detect 5caC based on variations in current signals. This method demonstrated good linearity ranging from 0.01 to 100 nM with a detection limit as low as 7.9 pM. We have successfully applied it to evaluate the 5caC levels in complex biological samples. The probe labeling contributes to a high selectivity for 5caC detection, while the sulfhydryl modification via T4 PNK efficiently circumvents the limitation of specific sequences. Encouragingly, no reports have been made about electrochemical methods for detecting 5caC in DNA, suggesting that our method offers a promising alternative for 5caC detection in clinical samples.

Sub-5 nm polyamide nanofilms combined with transfer-printing compositing for ultrafast nanofiltration

Jin-Bo Li,Cheng-Ye Zhu,Bian-Bian Guo,Chang Liu,Chao Zhang,Jian Wu,Lin Zhang,Hao-Cheng Yang,Zhi-Kang Xu

Synthesizing ultrathin polyamide nanofilms via interfacial polymerization (IP) at free interface offers a promising route to achieving preferential separation performance. However, one of big challenges lies in how to acquire large-scale synthesis and defect-free transfer of sub-5 nm polyamide nanofilms into robust composite membranes . Herein, we report an integrated strategy to allow for scalable synthesis and seamless compositing of sub-5 nm polyamide nanofilms that exhibit ultrafast and robust nanofiltration performance, by virtue of glycerol-regulated IP at free interface and transfer-printing compositing. Distinct from conventional hexane-water interface, our method leverages hexane-water/glycerol interface to boost interface stability of IP and confine monomer diffusion, enabling controllable synthesis of large-scale, defect-free sub-5 nm polyamide nanofilm. Meanwhile, a facile and universal transfer printing method is employed to integrate nanofilms on various substrates with high interfacial strength , which is not achievable with conventional transfer methods. The sub-5 nm polyamide composite membrane exhibits extraordinary water permeance (53.2 L⋅m −2 ⋅h −1 ·bar −1 ) and salt rejection (R Na2SO4  = 97.3%) under ultralow pressure of 1 bar, as well as superhigh water/salt permselectivity surpassing most of the polyamide composite membranes reported. The easy composite method and extraordinary performance in this work make ultrathin polyamide nanofilms a step closer to practical separation scenarios.

Preparation of ZSM-5 zeolites by sodium-free method in rotating packed bed

Xi-shuo Wang,Ying-jiao Li,Ting-ting Qi,Ji-wen Fu,Guang-Wen Chu,Bao-chang Sun

In industry, H-type zeolites are usually synthesized under hydrothermal conditions in the sodium-containing system, in which great deal of ammonia nitrogen wastewater will be produced during the transformation of zeolites from Na-type to H-type. In this work, we prepared ZSM-5 zeolites in the sodium-free system using organic amines in a rotating packed bed (RPB), which can greatly reduce the amount of wastewater. It was found that the ZSM-5 zeolites synthesized by isopropanolamine (MIPA) had the highest acid amount with a value of 2.331 mmol/g compared to that by other three amines, and exhibits the highest conversion of benzaldehyde with a value of 77.63% in the acetal reaction of benzaldehyde and ethylene glycol . More importantly, RPB premixing can improve the transfer of Al into the framework of ZSM-5 zeolites relative to conventional stirring technique. This work provided a new route to prepare ZSM-5 zeolites with high acid amount in sodium-free system.

Coupling Ethane Dehydrogenation with Benzene Alkylation Over Bifunctional Pt Supported HZSM-5 Catalyst

Zhang Yakun,Xu Lin,Wang Wen,Wu Qing,Zhang Yi

The efficient conversion of ethane has attracted more and more attention because of the shale gas revolution. The coupling of ethane dehydrogenation (EDH) and benzene alkylation over the 1.5Pt/HZSM-5 realize high activity close to the thermodynamic equilibrium of EDH at 500 ℃ and 0.9 bar ethane partial pressure. Meanwhile, the coupled reaction shows long-term stability during 22 h with a slightly decreased ethane conversion of 0.03% per hour and as high as 42% selectivity of ethylbenzene. Based on various characterization, including XRD, HR-TEM, NH 3 -TPD, Py-IR, TGA, it is confirmed that the coupling reaction could convert ethylene, formed by EDH over Pt sites, to ethylbenzene via benzene alkylation over Brönsted acidic sites (BAS) subsequently, and suppressing the sintering of Pt species and coke deposition induced by aromatization and deep dehydrogenation, resulting in high activity and long-time steady state. Efficient conversion of fructose to produce high-purity 5-hydroxymethylfurfural under low temperature

Wu Jinsheng,Yang Ruinan,Zhao Shiqiang,Chen Wei,Chen Zhiyong,Chang Chun,Wu Haoran

The development of mild strategies to transform of fructose to prepare high-purity 5-hydroxymethylfurfural (HMF) is of great significance for biomass valorization, and the design of an efficient catalytic approach is crucial. Herein, a mild catalytic strategy was developed to achieve the preparation of high-purity HMF from fructose over Amberlyst-15 catalyst in a water-methyl isobutyl ketone (MIBK) biphasic system under atmospheric pressure and lower temperature (90 °C). The catalytic system was optimized and analyzed by response surface methodology and kinetics study. Results indicated that a 46.6% yield of HMF could be obtained under optimal conditions: 80 wt% Amberlyst-15 catalyst, the mass ratio of MIBK to H 2 O 10, and the reaction time of 7 h. Furthermore, a simple and efficient purification strategy of HMF was established. High purity (99.0%) and recovery rate (65.5%) of HMF were obtained. This work provides an efficient catalytic strategy to produce high-purity HMF from fructose under atmospheric pressure and lower temperature.

Development of biodegradable nanogels for lipase accelerated drug release of 5-aminolevulinic acid

Xiao Liu,Yuan Zhang,Peng Zhang,Kang Ge,Ruzhi Zhang,Yixin Sun,Yang Sheng,Mark Bradley,Rong Zhang

Photodynamic therapy (PDT) using 5-aminolevulinic acid (5-ALA) is an important approach for the treatment of some skin diseases and cancers. A major defect of this approach is that it is difficult for 5-ALA to accumulate around lesions in deeper regions of tissue, resulting in poor conversion to the active fluorophore and photodynamic efficiencies. Because of their targeting and controlled release abilities, nanogel carriers could solve this problem. In this paper, nanogels were prepared by using micro-emulsion polymerization with various biodegradable polyester crosslinkers ( L -lactide and ε-caprolactone). The swelling and degradation properties and entrapment efficiency, drug loading and drug release ability of the nanogels were investigated. Nanogels co-cultured with skin cancer cells (A2058) allowed the efficiency of the PDT in vitro to be demonstrated. The results showed that the swelling rate of hydrogels reduced with increasing crosslinker levels, which caused a slow-down in the release of 5-ALA, but lipase accelerated degradation of nanogels increased 5-ALA concentrations in tumor cells and leading to higher PDT efficiency. It was proved by in vivo experiment indicating that the development of skin cancer tissues were efficiently inhibited by the 5-ALA loaded nanogels.

Biomimetic oxidase based on functionalized mesoporous SiO2 and metalloporphyrin for 5-hydroxymethylfurfural conversion

Bin Zou,Pengyun Wang,Zhenjiang Liu,Jiaojiao Xia,baihan Chen,Zhiyuan Tan

2,5-Furandicarboxylic acid (FDCA) has the potential to replace terephthalic acid (TPA) as a building block in the preparation of bio-based and degradable polyester materials. However, the lack of efficient and stable catalysts limits the commercial viability of FDCA production. In this work, metalloporphyrins were immobilized on mesoporous SiO 2 and used as a novel oxidase to convert 5-hydroxymethylfurfural (HMF) to FDCA. As the catalytic active center of P450 monooxygenases and horseradish peroxidase, supported palladium, manganese, ferri, and cobalt porphyrins via ionic-liquid-functionalized SiO 2 (PdPOP-SiO 2 , MgPOP-SiO 2 , FePOP-SiO 2 and CoPOP-SiO 2 ) were able to simulate oxidase alone under mild conditions. 100% HMF (0.02 M) conversion and 94.94% FDCA yield were achieved using PdPOP-SiO 2 as a biomimetic oxidase after a 12 h reaction at 373 K in water at atmospheric pressure. This type of mimetic oxidase is stable, inexpensive, and readily available, and the method developed in this work can potentially be used to oxidize HMF to FDCA in an industrial setting.

5-hydroxymethylfurfural-embedded poly (vinyl alcohol)/sodium alginate hybrid hydrogels accelerate wound healing

Fanhui Kong,Conghui Fan,Yulian Yang,Bae Hoon Lee,Kun Wei

Treating large acute or chronic wounds remains a challenging task because of a lack of effective methods to accelerate wound healing. Though growth factor-based wound products are found to be effective, they are costly and are potentially associated with increased cancer mortality. Effective, safe, and affordable strategies to tackle large or chronic wounds need to further be designed and developed. Here, we designed a new antioxidant-embedded hydrogel system for speeding up the wound healing process. We prepared multifunctional poly (vinyl alcohol)/sodium alginate (PVA/SA) hydrogels with the incorporation of 5-hydroxymethylfurfural (5-HMF) and silver nanoparticles (Ag-NPs), and investigated their physiochemical and biological properties in vitro and in vivo. PVA, SA, 5-HMF, and Ag-NPs were employed for good mechanical properties, good biocompatibility, anti-inflammation, and anti-bacterial activity, respectively. 5-HMF is commonly found in many food products (e.g. honey, coffee, and black garlic) and is known as an antioxidant. In our in vitro study, 5-HMF was found to effectively facilitate the proliferation and migration of human skin fibroblasts (HSF), and collagen production. Besides, 5-HMF-embedded PVA/SA hybrid hydrogels supported controlled release and good cell compatibility, and more importantly accelerated wound healing in vivo through ameliorated inflammation, enhanced angiogenesis/vascularization, increased collagen production, and promoted re-epithelialization.

Near-infrared squaraine fluorescent probe for imaging adenosine 5′-triphosphate in live cells

Guimei Wang,Xiaoxue Jiang,Nanyan Fu

Adenosine 5′-triphosphate (ATP), as a “molecular currency” for intracellular energy transfer, not only participates in various physiological activities, but also involves in various pathological processes of living cells. Therefore, the establishment of a simple, rapid, sensitive and accurate ATP analysis method is of great significance for the study of its physiological functions. In this work, we rationally design a near-infrared phenylboronic acid group-functionalized dicyanovinyl squaraine ( SQ-PBA3 ), with which a supramolecular assembly was constructed for fluorescent imaging of ATP. In PBS buffer, SQ-PBA3 changes self-assembly when adding cetyltrimethyl ammonium bromide (CTAB) deriving fluorescence response of SQ-PBA3 to ATP with the maximum emission at 700 nm, due to the specificity of phenylboronic acid to diols and the multiple electrostatic interactions between SQ-PBA3 , ATP and CTAB molecules. Meanwhile, the probe was successfully applied to monitor intracellular ATP level in MCF-7 cells with high sensitivity and selectivity.

TEXTURE AND STRUCTURE VARIATION OF PEROVSKITE LaFeO3/ZSM-5 DURING HIGH-TEMPERATURE DESULFURIZATION

DONGJING LIU,WEIGUO ZHOU,JIANG WU

Perovskite LaFeO 3 /ZSM-5 is synthesized via citrate route for H 2 S removal at high temperatures. It shows good thermal stability after heat treatment at 500–700 ∘ C with respect to slight changes in crystallographic phase and textural property. It presents the optimal desulfurization performance at 600 ∘ C with sulfur capacity of 1017 μ mol S/g and products of S, LaS 2 , and Fe 7 S 8 . Sulfidation at 500 ∘ C yields the same products as sulfidation at 600 ∘ C but displays the lowest sulfur capacity of 408 μ mol S/g. Sulfidation at 700 ∘ C produces La 2 O 2 S, Fe 3 S 4 , and unreacted LaFeO 3 . The activation energy of the sulfidation reaction over LaFeO 3 /ZSM-5 is 109.6 kJ/mol.
