Preparation and characterization of porous starch/β-cyclodextrin microsphere for loading curcumin: Equilibrium, kinetics and mechanism of adsorption

Xinying Zheng,Chao Qiu,Jie Long,Aiquan Jiao,Xueming Xu,Zhengyu Jin,Jinpeng Wang

In this study, porous starch/β-cyclodextrin microsphere (PCS-CD) was fabricated for the first time through the crosslink reaction between corn starch (CS) and β-cyclodextrin (β-CD) combined with enzymatic hydrolysis . The structure, morphology, particle size, thermal property and pasting property were characterized by X-ray diffraction (XRD), scanning electronic microscope (SEM), laser light scattering particle size analyzer and differential scanning calorimetry (DSC), respectively. The results showed that the combination significantly increased the adsorption capacity of hydrophobic substance, which was evaluated by oil adsorption (316 g/g), and achieved an improvement of thermal stability (for an increased pasting temp at 86.5 °C) and shearing resistance (for a decreased breakdown viscosity at 148 cP). The water solubility index (WSI), swelling power index (SPI) and paste clarity were decreased after being crosslinked, but increased slightly after enzymatic hydrolysis. Furthermore, the adsorption kinetics and mechanism of curcumin on the PCS-CD were also evaluated. The results of adsorption kinetics showed that the adsorption process could be described by Pseudo-first-order model and was controlled by two or more diffusion mechanism. The free energy calculated by adsorption isotherm model was 0.1017 kJ/mol, which revealed that a physical-controlled adsorption was occurred between the PCS-CD and curcumin.

Nitrogen-doped fluorescence carbon dots as multi-mechanism detection for iodide and curcumin in biological and food samples

Xiaodan Tang,Hongmei Yu,Brian Bui,Lingyun Wang,Christina Xing,Shaoyan Wang,Mingli Chen,Zhizhi Hu,Wei Chen

Iodine ion is one of the most indispensable anions in living organisms, particularly being an important substance for the synthesis of thyroid hormones. Curcumin is a yellow-orange polyphenol compound derived from the rhizome of Curcuma longa L. , which has been commonly used as a spice and natural coloring agent, food additives, cosmetics as well as Chinese medicine. However, excess curcumin may cause DNA inactivation, lead to a decrease in intracellular ATP levels, and trigger the tissue necrosis. Therefore, quantitative detection of iodine and curcumin is of great significance in the fields of food and life sciences. Herein, we develop nitrogen-doped fluorescent carbon dots (NCDs) as a multi-mechanism detection for iodide and curcumin in actual complex biological and food samples, which was prepared by a one-step solid-phase synthesis using tartaric acid and urea as precursors without adding any other reagents. An assembled NCDs-Hg 2+ fluorescence-enhanced sensor for the quantitative detection of I − was established based on a fluorescence “turn-off-on” mechanism in a linear range of 0.3–15 μM with a detection limit of 69.4 nM and successfully quantified trace amounts of I − in water samples and urine sample. Meanwhile, the as-synthesized NCDs also can be used as a fluorescent quenched sensor for curcumin detection based on the synergistic internal filtration effect (IFE) and static quenching, achieving a good linear range of 0.1–20 μM with a satisfactory detection limit of 29.8 nM. These results indicate that carbon dots are potential sensing materials for iodine and curcumin detection for the good of our health.

Effect of combination of curcumin and GLUT-1 AS-ODN on radiosensitivity of laryngeal carcinoma through regulating autophagy

Li-Bo Dai,Qi Yu,Shui-Hong Zhou,Yang-Yang Bao,Jiang-Tao Zhong,Li-Fang Shen,Zhong-Jie Lu,Jun Fan,Ya-Ping Huang

Background This study is to explore the role of curcumin and GLUT-1 antisense oligodeoxynucleotides (AS-ODN) on autophagy modulation-initiated radiosensitivity. Methods BALB/c mice were employed to establish xenograft model using Tu212 cell. The expression of autophagy- and apoptosis-related proteins was determined by WB. Autophagosome was observed under transmission electron microscope. Apoptosis of tumor tissue were detected by TUNEL staining. Results Combinations of curcumin and GLUT-1 AS-ODN with 10 Gy inhibited the tumor growth by inducing apoptosis of laryngeal cancer cells followed with the enhancement of autophagy. 3-MA also had a promotion effect on irradiation-mediated growth inhibition possibly by depressing PI3K and on curcumin/GLUT-1 AS-ODN-mediated growth inhibition potentially by regulating autophagic events. Of note, a de-escalation of radiotherapy dose (5 Gy) along with curcumin, GLUT-1 AS-ODN or 3-MA produced a stronger effect than high dosage of radiotherapy (10 Gy) alone. Conclusions Curcumin and GLUT-1 AS-ODN improve the radiosensitivity of laryngeal carcinoma through regulating autophagy and inducing apoptosis.

Co-assembly of curcumin and a cystine bridged peptide to construct tumor-responsive nano-micelles for efficient chemotherapy

Yemei Dai,Zelei Jiang,Jingyi Li,Mengfan Wang,Chang Liu,Wei Qi,Rongxin Su,Zhimin He

The effective uptake and release of hydrophobic antitumor drugs in cancer cells is a practical challenge for tumor chemotherapy. Many methods were developed to conquer it through modifying drug molecules with hydrophilic groups, or fabricating nanodrugs based on hydrophilic materials. In recent years, peptides have attracted significant interest as part of a promising platform for fabricating nanodrugs due to their low cytotoxicity, favorable variability and self-assembly property. In this study, a cystine bridged peptide (CBP) was designed to co-assemble with a hydrophobic antitumor drug curcumin (CCM), to form a tumor-responsive nanodrug. The hydrophilicity of the peptide promotes the water-dispersity of nanodrugs, and the disulfide bond in cystine, which is cleavable by glutathione (GSH), was involved considering the overexpressed GSH in tumor microenvironments. In vitro and in vivo tests on cervical cancer cells revealed that the obtained nanodrug can rapidly dissociate at tumor sites and inhibit the tumor growth with limited side effects on healthy tissues.

Glycyrrhetinic acid modified and pH-sensitive mixed micelles improve the anticancer effect of curcumin in hepatoma carcinoma cells

Jizheng Song,Yuling Liu,Longfei Lin,Ye Zhao,Xiuqing Wang,Ming Zhong,Tanggui Xie,Yuting Luo,Shaojing Li,Ruocong Yang,Hui Li

Curcumin (CUR), a natural polyphenolic compound existing in plants, exhibits anticancer potential in inhibiting the growth of various types of human cancer. However, the poor aqueous solubility and low bioavailability limit its clinical applications. pH-sensitive macromolecule F68-acetal-PCL (FAP) and active targeting macromolecule F68-glycyrrhetinic acid (FGA) were designed to fabricate mixed micelles for efficient delivery of CUR. The thin film hydration method was used to prepare CUR loaded mixed (MIX/CUR) micelles. The drug loading rate (DL) of MIX/CUR micelles was 6.31 ± 0.92%, which remained stable for 15 days at 4 °C. The particle size and zeta potential of the MIX/CUR micelles were 91.06 ± 1.37 nm and −9.79 ± 0.47 mV, respectively. The MIX/CUR micelles exhibited pH sensitivity in a weak acid environment, and showed rapid particle size variation and drug release. In addition, in vitro tests demonstrated that MIX/CUR micelles induced higher cytotoxicity and apoptosis than free CUR, non-pH-sensitive F68-PCL (FBP)/CUR micelles and pH-sensitive FAP/CUR micelles in SMMC7721 and Hepa1-6 cells. Besides, mixed micelles were more effective than FBP and FAP micelles in a cell uptake experiment, which was medicated by a GA receptor. All in all, these results indicated that MIX/CUR micelles could be regarded as an ideal drug administration strategy against hepatoma carcinoma cells.

Preparation and characterization of glutathione-responsive polymeric micelles functionalized with core cross-linked disulfide linkage for curcumin delivery

Chen Dan,Gong Feirong,Han Xiaoxiong,Chi Lili,Sun Jing,Shen Yaling

Conventional self-assembled polymeric micelles typically lose their loaded drugs after systemic administration. To resolve this issue, the aim of the present study was to develop a novel polymer, methoxy-poly (ethylene glycol)-block-poly ( d , l -lactic acid) grafted with four lipoic acids (mPEG-PLA-(LA) 4 ), to prepare curcumin-loaded core cross-linked polymeric micelles (CCM). Nitrogen and alkalescent buffer (pH 8.5) were beneficial for cross-linking, and the cross-linking was nearly complete within 30 min. The micelle preparation parameters were optimized through both a single-factor experiment and orthogonality test. The choice of cryoprotector had the most important effect on micelle solution reconstruction and almost challenged our perceptions about cryoprotector. In addition the drug loading capacity greatly affected the stability of micelles. Moreover, a method for determining the cross-linking density was established, and the result of gel permeation chromatography indirectly demonstrated that dozens of polymers had successfully formed CCM. The CCM significantly enhanced stability under non-reductive conditions and could be reversibly broken in a reductive environment. Curcumin-loaded CCM also showed glutathione-response cytotoxicity to cancer cells. Overall, this fundamental study suggests CCM as a potential carrier for achieving improved controlled drug delivery.

BSA Nanoparticles Modified with N-Acetylcysteine for Improving the Stability and Mucoadhesion of Curcumin in the Gastrointestinal Tract

Shanshan Zhang,Sajid Asghar,Feng Yu,Zhipeng Chen,Ziyi Hu,Qineng Ping,Feng Shao,Yanyu Xiao

A major obstacle to the clinical use of curcumin (CUR) is its reduced bioavailability because of the drug’s hydrophobic nature, low intestinal absorption, and rapid metabolism. In this study, a novel oral drug delivery system was constructed for improving the stability and enhancing mucoadhesion of CUR in the gastrointestinal (GI) tract. First, CUR was encapsulated in the bovine serum albumin nanoparticles (CUR-BSA-NPs). Then, N-acetyl cysteine (NAC)-modified CUR-BSA-NPs (CUR-NBSA-NPs) were obtained. The average particle size and zeta potential of CUR-NBSA-NPs were 251.6 nm and −30.66 mV, respectively; encapsulation efficiency and drug loading were 85.79 and 10.9%, respectively. CUR-NBSA-NPs exhibited a sustained release property and prominently enhanced stability in simulated GI conditions. Additionally, enhanced mucoadhesion of CUR-NBSA-NPs was also observed. An MTT study showed that the CUR-NBSA-NPs were safe for oral administration. Overall, NAC-modified BSA-NPs may potentially serve as an oral vehicle for improving CUR stability in the GI tract and enhancing mucoadhesion.

Oxidatively stable curcumin-loaded oleogels structured by β-sitosterol and lecithin: physical characteristics and release behaviour in vitro

Linlin Li,Wenbo Wan,Weiwei Cheng,Guoqin Liu,Lipeng Han

Summary In this study, oleogel structured by β-sitosterol and lecithin was prepared as a carrier for poorly water-soluble bioactive compounds, curcumin. The mechanical and microscopic properties of curcumin-loaded oleogel were determined by texture analyser and polarised light microscopy. The oxidation stability of the delivery system was determined by accelerated oxidation test and the bioaccessibility of curcumin was studied in vitro . The results showed that the existence of curcumin did not affect the mechanical strength and crystalline network of oleogel. The oxidative stability of curcumin-loaded oleogel was higher than that of curcumin-free oleogel and corn oil-curcumin mixtures. It might be attributed to not only the antioxidative property of curcumin but also crystalline network of oleogel which served as an obstructor for the development of oxidation products. Additionally, oleogel enhanced the bioaccessibility of curcumin in fasted state (up to 67.66%). These findings suggest that oleogels can be promising candidates to delivery drugs and nutrients.

Effect of curcumin derivatives on hen egg white lysozyme amyloid fibrillation and their interaction study by spectroscopic methods

Liangliang Cui,Sujuan Wang,Jian Zhang,Mengna Wang,Yan Gao,Libin Bai,Hailei Zhang,Gang Ma,Xinwu Ba

Two novel Boc- L -isoleucine-functionalized curcumin derivatives have been synthesized and characterized, which exhibited enhanced solubility in water compared with the natural curcumin. The solubility could reach 2.12   mg/mL for the monosubstituted compound and 3.05   mg/mL for the disubstituted compound, respectively. Their anti-amyloidogenic capacity on the model protein, hen egg white lysozyme (HEWL), was examined in aqueous solution. ThT fluorescence assay showed that the operation concentration was only 0.5   mM when the inhibition ratio was above 70%. Meanwhile, the inhibitory capacity of monosubstituted curcumin derivative on the formation of HEWL amyloid fibrils was found to be superior to that of disubstituted derivative, suggesting that the phenolic hydroxyl group might contribute to the anti-amyloidogenic activity. Interaction study showed that both curcumin derivatives could bind with HEWL near tryptophan residues and form new ground-state complex before HEWL self-assemblies into amyloid fibrils and thus inhibits the formation of amyloid fibrils. Both of the two cucumin derivatives have displayed low cytotoxicity with HeLa cell.

Effect of β-sitosterol on the curcumin-loaded liposomes: Vesicle characteristics, physicochemical stability, in vitro release and bioavailability

Kedong Tai,Michael Rappolt,Xiaoye He,Yang Wei,Shaoxin Zhu,Jingbo Zhang,Like Mao,Yanxiang Gao,Fang Yuan

In this work, the effect of β-sitosterol (Sito) on vesicle characteristics, physicochemical stability as well as the in vitro release and bioavailability of curcumin-loaded liposomes (Cur-LP) was studied. When 20–33 mol% of Sito was incorporated, encapsulation efficiency of curcumin was improved due to the high amount of liquid-ordered domains in membranes. At 50 mol% Sito a lower encapsulation efficiency was observed possibly due to membrane defects. The physical, thermal and photo stability of curcumin in liposomes were markedly improved with increasing the amount of Sito. First-order kinetics fitted best the curcumin release dynamics of Sito containing liposomes, clearly showing that sustained release improved with increasing amounts of Sito in liposomes. Simulated digestion studies suggested that Sito concentration of about 20–33 mol% improved the bioavailability of curcumin in liposomes. These study shows that Sito is an applicable and potential route in forming healthier cholesterol-free curcumin-loaded liposomes for functional supplements.

Tween 80-modified hyaluronic acid-ss-curcumin micelles for targeting glioma: Synthesis, characterization and their in vitro evaluation

Cihui Tian,Sajid Asghar,Yurui Xu,Zhipeng Chen,Jingwei Zhang,Qineng Ping,Yanyu Xiao

Redox-sensitive micelles based onhydrophilic hyaluronic acid-ss-hydrophobic curcumin conjugate were designed as a novel delivery system for gliomas targeting. Furthermore, the obtained micelles were further functionalized with Tween 80 (CUR-THSC) for better brain penetration. Dynamic light scattering experiment and in vitro release study showed that the synthetic disulfide-linked conjugate possessed redox-sensitivity under high glutathione conditions. Spherical micelles with a mean particle size of 74.2 nm, negative zeta potential (−30.25 mV), high entrapment efficiency (94.12%) and drug loading (8.9%) were obtained. XRD analysis of micelles revealed amorphous form of the encapsulated drug. CUR-THSC micelles showed good plasma stability and did not induce any hemolysis in erythrocytes. In addition, highest cytotoxicity in G422 cells was observed compared to the free curcumin group and non-sensitive micelles group. These results indicate that the Tween 80-modified hyaluronic acid-ss-curcumin micelles could emerge as a promising platform for the delivery of curcumin in the treatment of gliomas.

The triplet state of tanshinone I and its synergic effect on the phototherapy of cancer cells with curcumin

Chenchen Zhang,Shan Jiang,Kun Li,Mei Wang,Rongrong Zhu,Xiaoyu Sun,Qingxiu Wang,ShiLong Wang

The excited triplet state of tanshinone I (Tan I) extracted from the traditional Chinese medicine Salvia miltiorrhiza Bunge was characterized by laser flash photolysis. The synergic effect of Tan I on the phototherapy of cancer cells with curcumin (Cur) was also investigated by MTT assay because the excited energy transfer from the triplet state of Tan I ( 3 Tan I ∗ ) to Cur occurred. At the same time, the characteristic absorption spectra of 3 Tan I ∗ were recorded, and its molar absorption coefficient and rate constants for several excited energy transfers were obtained. The photo-therapeutic effect of Cur is enhanced by combination with Tan I.
