Oxaliplatin (OXL) is a first-line drug for the treatment of colon cancer , with excellent efficacy. Intestinal toxicity is a common side effect of OXL, with unclear pathogenesis and a lack of effective treatment strategies. Polydatin (PD) has anti-inflammatory and antioxidant activities and is a potential drug for treating intestinal diseases , but its poor water solubility limits its application. In this study, polyvinylpyrrolidone (PVP) was used as a carrier to prepare nanoparticles loaded with PD (PVP-PD), with a particle size of 92.42 nm and exhibiting sustained release properties. In vitro results showed that PVP-PD protected NCM460 cells from OXL induced injury, mitochondrial membrane potential (MMP) disruption, and accumulation of reactive oxygen species (ROS). The in vivo results demonstrated the protective effect of PVP-PD on intestinal toxicity induced by OXL, such as alleviating weight loss and colon length reduction induced by OXL. Both in vivo and in vitro mechanisms indicated that OXL induced DNA damage and activated the cGAS-STING pathway, further inducing the expression of inflammatory factors such as IL-1β and TNF-α. PVP-PD alleviated the aforementioned changes induced by OXL by inhibiting the DNA damage-cGAS-STING pathway. In summary, our study demonstrated that the DNA damage-cGAS-STING pathway was involved in OXL induced intestinal toxicity, and PVP-PD provided a potential strategy for treating OXL induced intestinal toxicity.

Internal browning (IB) is a physiological disorder without external symptoms of postharvest pineapples, but whether and how IB influences pineapple volatiles remain unknown. We examined eigenvalues of volatiles in ‘Comte de Paris’ pineapples with or without IB using electronic nose (E-nose) and gas chromatography-mass spectrometry (GC–MS). Correlation coefficients between the responses of E-nose sensors S7 and S9 and IB were 0.836 and 0.804, respectively. The multilayer perceptron neural network and radial basis function neural network models classified IB degree with accuracy of 94.77% and 91.67%. GC–MS analysis revealed 30 volatile substances upregulated in pineapple with IB compared to those without, of which 15 were esters. IB regulated volatile compound synthesis through the lipoxygenase pathway which involved lipoxygenase , pyruvate decarboxylase 1, alcohol dehydrogenases , acyl-CoA oxidase 1 , and alcohol acyltransferase genes and their related enzymes. These results suggested that volatiles regulated by IB could be used to discriminate IB severity in pineapples.

Frontal polymerization (FP) is a self-sustaining reaction that relies on polymerization exothermicity and heat transfer. This study explores the in-plane and in-depth FP mechanisms of continuous fiber-reinforced epoxy composites. First, the effects of initiator and diluent concentrations on the FP behaviors of neat epoxy resins were examined. It was found that the frontal velocity and frontal temperature of the neat resins increase with an increase of either the initiator concentration or the diluent content, depending on the polymerization enthalpy and curing kinetics. Then, the FP behaviors of continuous carbon fiber and glass fiber-reinforced epoxy woven composites were investigated. The results showed that the in-plane FP behavior of the composites was primarily controlled by the thermal conductivity and specific heat capacity of the continuous fibers, whereas the in-depth FP behavior mainly depended on the pores of the woven fabrics. Highlights Frontal velocity and temperature of neat resins are controlled by the polymerization enthalpy and curing kinetics. The in-plane FP behavior of continuous fiber reinforced composites is dependent on the thermal conductivity and specific heat capacity of the fibers. The in-depth FP behavior of composite is determined by both the thermal conductivity of fibers and the pores of fabrics.

α-Chiral amines are key intermediates for scalable preparation of bioactive compounds; herein we present a novel palladium-based nanocatalyst capable of selectively catalyzing the reductive amination of carbonyl compounds, which enables the in situ regeneration of amino donors from wasteful co-products in a one-enzyme cascade using ω-transaminase, without the requirement of the expensive coenzyme NAD(P)H. The cascade network combines a ω-transaminase-assisted transamination with a selective reductive amination reaction facilitated by a heterogeneous palladium-based nanocatalyst. Nitrogen is sourced from hydroxylamine ions to convert generated co-products back into amino donors, yielding chiral amines with exceptional yields of up to 99% and excellent enantioselectivity. This chemoenzymatic one-enzyme transamination-reductive amination cascade network is highly atom-efficient and generates H2O as its sole by-product, demonstrating its potential impact in synthetic chemistry and beyond.

We have previously demonstrated that treatment with cannabidiol (CBD) ameliorates mitochondrial dysfunction and attenuates neuronal injury in rats following cerebral ischemia . However, the role of CBD in the progression of ischemic stroke-induced inflammation and the molecules involved remain unclear. Here, we found that CBD suppressed the production of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), reduced the activation of microglia , ameliorated mitochondrial deficits, and decreased the phosphorylation of nuclear factor κ-B (NF-κB) in BV-2 cells subjected to oxygen-glucose deprivation/reoxygenation (OGD/R). Cyclin-dependent kinase regulatory subunit 1B (CKS1B) expression was decreased in BV-2 cells following OGD/R and this reduction was blocked by treatment with CBD. Knockdown of CKS1B increased the activation of microglia and enhanced the production of IL-1β and TNF-α in BV-2 cells treated with CBD. Moreover, CKS1B knockdown exacerbated mitochondrial deficits and increased NF-κB phosphorylation. CBD treatment also ameliorated brain injury, reduced neuroinflammation , and enhanced the protein levels of mitochondrial transcription factor A and CKS1B in rats following middle cerebral artery occlusion/reperfusion. These data identify CKS1B as a novel regulator of neuroinflammation; and reveals its involvement in the anti-inflammatory effects of CBD. Interventions targeting CKS1B expression are potentially promising for treating in ischemic stroke.

All-inorganic CsPbX 3 (X = Cl, Br, and I) perovskite nanocrystals (NCs) have great application prospects in many fields due to their excellent photoelectric properties. However, CsPbX 3 NCs are extremely sensitive to water, heat, light, and oxygen, which leads to dramatic photoluminescence quenching and seriously restricts their future commercial applications. Herein, a novel, simple one-step solution strategy for synthesizing high-quality CsPbBr 3 NCs was reported. By the combination of the hot-injection process with the mesoporous template confinement effect of the KIT-6 matrix, the space-confined growth of NCs was precisely achieved. The CsPbBr 3 NCs encapsulated by KIT-6 (CsPbBr 3 @KIT-6 composite) exhibited a uniform size, a high photoluminescence quantum yield (PLQY) of up to 82%, and a narrow full width at half-maximum (FWHM) of 24 nm. Most importantly, CsPbBr 3 @KIT-6 powder composites had dramatically enhanced light, thermal and water stability in comparison with colloidal CsPbBr 3 NCs due to the dual protection of organic ligands and matrix templates. Ultimately, a white light-emitting diode (LED) device was successfully fabricated by encapsulating a mixture of green-emitting CsPbBr 3 @KIT-6 composite and red-emitting K 2 SiF 6 :Mn 4+ phosphor onto a blue-emitting InGaN LED chip. A color coordinate of (0.327, 0.337) and a wide color gamut of 128% NTSC well demonstrate their potential applications in lighting and display.

Studies have suggested that the nucleus accumbens (NAc) is implicated in the pathophysiology of major depression; however, the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been elucidated. Here, we identified a specific reduction of cyclic adenosine monophosphate (cAMP) in the subset of dopamine D1 receptor medium spiny neurons (D1-MSNs) in the NAc that promoted stress susceptibility, while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like behaviors. Ketamine treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons (D2-MSNs) of depressed mice, however, the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc D1-MSNs. We discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration, but not a lower dose. The fast onset property of crocin was verified through multicenter studies. Moreover, crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on D2-MSN. These findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc, and provide a potential rapid antidepressant drug candidate, crocin.

In order to solve the short stabilization time of the water-in-oil emulsion binder system and the shortcomings of the critical blast size of the explosive ink molding sample, a water-in-oil emulsion binder was designed with polyvinyl alcohol (PVA) aqueous solution as the aqueous phase, fluorocarbon resin (FEVE) ethyl acetate solution as the oil phase, and toluene diisocyanate (TDI) added to modify the water-in-oil emulsion binder. The hexanitrohexaazaisowurtzitane(CL-20)based explosive ink was made by incorporating submicron CL-20 into the binder system, and the print-formed samples were tested for microscopic morphology, crystallographic analysis, mechanical susceptibility, and detonation transmission performance. The results showed that the explosive ink‘s viscosity was moderate and suitable for direct printing, and the formed sample‘s surface particles were uniformly dispersed and had a honeycomb structure. The crystal form of CL-20 explosive in the explosive ink did not change, the molded sample‘s impact and frictional sensitivities were 1 J and 32 N lower than those of submicron CL-20, respectively, and the detonation velocity of the molded sample was 6655 m/s. The critical detonation size and critical detonation thickness were 1×0.0078 mm and 0.1 mm, respectively, and a critical detonation corner turning of 90°–160° could be achieved. The results show that the prepared binder system is stable for a long time, and the explosive ink molding samples have excellent blast transmission performance.

The superior optical properties of lead-halide perovskites (LHPs) inspired significant research in cell imaging applications; many encapsulating processes have improved perovskite stabilities with comparable biosafety. Herein, facile solvent evaporation encapsulation based on an oil-in-fluorine emulsion for aqueous-stable and extremely nontoxic LHP microcapsules is described. Perfluorooctane dispersed the emulsifier fluorocarbon surfactant to form a continuous fluorine phase, while LHPs and polymethylmethacrylate (PMMA) were dispersed in 1,2-dichloroethane, then emulsified in the fluorine phase to form an oil-in-fluorine emulsion. CsPbBr3microcapsules with a dense PMMA shell that protect fragile CsPbBr3from the external environment and inhibit lead ion release were obtained after solvent evaporation. The CsPbBr3microcapsules not only retained 91% of fluorescence intensity after exposure to water for 30 d but also possess extremely low cytotoxicity for MCF-7 cells. After exposure to 2 mg/mL of CsPbBr3microcapsules for 48 h, the cell viability remained >90%. The intracellular uptake of CsPbBr3microcapsules indicates its potential use in cell imaging.Keywords:lead-halide perovskites;microencapsulation;oil-in-fluorine emulsion;cell imaging

The knowledge of osteoarthritis (OA) has nowadays been extended from a focalized cartilage disorder to a multifactorial disease. Although recent investigations have reported that infrapatellar fat pad (IPFP) can trigger inflammation in the knee joint, the mechanisms behind the role of IPFP on knee OA progression remain to be defined. Here, dysregulated osteopontin (OPN) and integrin β 3 signaling are found in the OA specimens of both human and mice. It is further demonstrated that IPFP-derived OPN participates in OA progression, including activated matrix metallopeptidase 9 in chondrocyte hypertrophy and integrin β 3 in IPFP fibrosis. Motivated by these findings, an injectable nanogel is fabricated to provide sustained release of siRNA Cd61 ( RGD− Nanogel/siRNA Cd61 ) that targets integrins. The RGD− Nanogel possesses excellent biocompatibility and desired targeting abilities both in vitro and in vivo. Local injection of RGD− Nanogel/siRNA Cd61 robustly alleviates the cartilage degeneration, suppresses the advancement of tidemark, and reduces the subchondral trabecular bone mass in OA mice. Taken together, this study provides an avenue for developing RGD− Nanogel/siRNA Cd61 therapy to mitigate OA progression via blocking OPN-integrin β 3 signaling in IPFP.

Despite aggressive treatments, including surgery, chemo- and radio-therapy, the prognosis of glioblastoma (GBM) remains poor and tumor recurrence is inevitable. The FDA-approved CDK4/6 inhibitor palbociclib (PB) showed interesting anti-GBM effects, but its brain penetration is limited by the blood-brain barrier. The hypothesis of this project is to find whether the cellulose-based hydrogel via in situ injection could provide an alternative route of PB brain delivery and generate sufficient drug exposure in orthotopic GBM. In brief, PB was encapsulated in a cellulose nanocrystal network structure crosslinked by polydopamine via divalent Cu2+ and hexadecylamine. The formed hydrogel (PB@PH/Cu-CNCs) exhibited sustained drug retention and acid-responsive network de-polymerization for controlled release in vivo. Specifically, the released Cu2+ catalyzed a Fenton-like reaction to generate reactive oxygen species (ROS), which was further enhanced by PB, and consequently induced irreversible senescence and apoptosis in GBM cells. Finally, PB@PH/Cu-CNCs demonstrated a more potent anti-GBM effect than those treated with free PB or PH/Cu-CNCs (drug-free hydrogel) in cultured cells or in an orthotopic glioma model. These results prove that the injection of PB-loaded hydrogel in situ is an effective strategy to deliver CDK4/6 inhibitor into the brain and its anti-GBM effect can be further enhanced by combining Cu2+-mediated Fenton-like reaction.

Lignocellulose nanopaper (LNP) assembled from lignocellulose nanofibrils (LCNFs) is an emerging eco-friendly structural material applicable to a variety of fields. Lignin serves as a crucial functional component in the LNP matrix; however, it negatively affects the interfacial hydrogen-bonding behaviors among LCNFs and consequently the inferior mechanical performance of LNP. In this study, a mild ozone-oxidation strategy was used to modify lignin macromolecules in situ without significant degradation of carbohydrate polymers (i.e., cellulose and hemicellulose) in LCNFs whereupon the interfacial hydrogen-bond energy was dramatically improved in the assembly and deformation process of LNP as validated by molecular dynamics simulation. Consequently, the lignin-modified LNP exhibited significantly enhanced tensile strength (from 83 to 140 MPa) and toughness (from 1.9 to 7.1 J/m3), which even surpassed those of conventional cellulose nanopaper. Benefiting from the well-preserved lignocellulosic structure, lignin-modified LNP maintained its inherent favorable water and thermal stability and intriguing optical performance, which supported our developed LNP to be a multifunctional structural material for diversified fields, for example, flexible electronic applications. Additionally, the estimated production cost for our developed LNP was approximately half of that for conventional cellulose nanopaper due to its significantly lower resource inputs such as the material, water, and energy. Overall, our study provides a site-specific macromolecular modulation strategy for the economically and environmentally feasible fabrication of high-performance lignocellulosic nanomaterials toward advanced structural applications.
