Poly(l-glutamic acid)s (PLGs) are promising bone-targeting ligands due to their high molecular weight and facile preparation. Nevertheless, the bone-targeting efficiency of PLGs is still relatively low, validating the necessity to further enhance targeting through structural optimization. Herein, we report the use of a heteropolypeptide strategy to improve the bone targeting of PLGs through the incorporation of another side-chain functionality for enhanced affinity with bone tissues. Specifically, the introduction of cationic amino or aromatic phenolic side-chain residues resulted in a ∼2.3-fold or ∼1.6-fold increase in the in vivo bone targeting, respectively. Cationic modification not only improved the affinity with bone minerals but also exhibited prolonged retention in the bone tissues for more than 60 days. This work highlights the use of a heteropolypeptide library to screen and optimize the performance of polypeptide materials, offering promising bone-targeting polymeric materials for the design of bone-related nanomedicine.

Ectoine, so-called tetrahydropyrimidine, is an important osmotic adjustment solute and widely applied in cosmetics and protein protectant. Some attempts have been made to improve the ectoine productivity. However, the strains with both high ectoine production capacity and high glucose conversion were still absent so far. Aim to construct a strain for efficiently producing ectoine, ectoine synthetic gene cluster ectABC from Pseudomonas stutzeri was overexpressed in E. coli BL21 (DE3). The ection production was improved by 382 % (ectoine titer increased from 1.73 g/L to 8.33 g/L) after the rational design of rate-limiting enzyme L-2,4-diaminobutyrate transaminase EctB ps (protein engineering) combined with the metabolic engineering that focused on the enrichment and conversion of precursors. The final strain YW20 was applied to overproduce ectoine in fed-batch fermentation and yield 68.9 g/L of ectoine with 0.88 g/L/h of space-time yield and the highest glucose conversion reported [34 % (g/g)]. From the fermentation broth, ectoine was purified with 99.7 % purity and 79.8 % yield. This study successfully provided an engineered strain as well as an efficient method for the industrial bio-synthesis and preparation of ectoine.

L-guluronic acid, a patented drug with immunomodulatory and anti-inflammatory properties, is traditionally sourced from natural seaweed species or synthesized chemically. Both methods suffer from low selectivity and environmental concerns and there is a high demand to develop a green and efficient approach for L-guluronic acid production. Herein, we successfully developed a two-step enzymatic route to synthesize L-guluronic acid from D-glucose by two oxidases, glucose oxidase (GOx) and galactose oxidase (GOase). Due to the sluggish kinetics of GOase in catalyzing the oxidation of D-gluconic acid, rational design was implemented. Through molecular docking and iterative saturation mutagenesis, a variant, GOase T406R/K330R/Y329F, was obtained. The variant exhibited a remarkable 13-fold higher oxidation activity towards D-gluconic acid, reaching 7.8 U mg-1. The substitutions synergistically switch the D-gluconic acid molecule to a favorable conformation and facilitate the specific activation of the primary alcohol group at C6 position. In a cascade reaction, complete conversion of 40 mM D-glucose to L-guluronic acid was achieved in 2 h with 100% selectivity, presenting a promising and green bio-manufacturing strategy for the scalable production of L-guluronic acid.

Traditional phase change materials (PCMs) offer broad application potential but face challenges such as environmental unfriendliness, high rigidity and poor heat transfer performance, resulting in low utilization efficiency. In this study, using biomass loofah sponge (LS) as the main framework and natural rubber latex (NRL) as the flexible modifier, the biomass-based PCMs (L-N) were successfully synthesized after encapsulating lauric acid (LA). The addition of nano-TiO 2 further enhanced thermal conductivity, ultimately leading to the development of flexible composite PCMs, L-N-Ti. In-depth characterizations revealed that the introduction of NRL significantly improved the system's strength and toughness, with tensile strength peaking at 1.161 MPa at 19.2 % NRL content, while maximum elongation at break reached 38.65 % at 26.3 % NRL. The incorporation of 7.4 wt% TiO 2 significantly boosted thermal conductivity to 0.57 W/(m·K), a 185 % increase over the unmodified material. Simultaneously, the energy storage efficiency (E m ) of L-N-Ti consistently exceeded 89.89 %, with only a 9.34 % loss in latent heat of fusion after 100 thermal cycles, indicating robust thermal stability. The successful advancement of L-N-Ti not only addresses the mechanical constraints inherent but also offers a sustainable biomass-based solution for effective thermal management within the 20°C–60 °C range.

Dental caries is a common oral disease and lacks an effective recovery method. The objective of the study was to investigate the remineralization efficiency of polyelectrolyte‑calcium complexes pre-precursor (PCCP) and polymer-induced liquid-like precursors (PILP) processes on mechanical and biochemical properties of dentin artificial caries lesions (ACLs) evaluated with micro-CT, SEM, TEM, ATR-FTIR, Raman spectra and nanoindentation. The human dentin ACLs were prepared with the depth of 100–120 μm. The intact dentin served as positive control and the dentin ACLs as negative control. The dentin ACLs were either incubated in poly-L-aspartic acid‑calcium suspension (PAsp-Ca, 4 g/L-5.44 M) followed by phosphate solution (0.325 M), and further incubated in artificial saliva serving as PCCP group, or in PAsp-stabilized amorphous calcium phosphate solution serving as PILP group. The micro-CT and SEM findings with EDX indicated that mineral contents of the dentin ACLs were dramatically increased in PCCP group, but, to some degree in PILP group. TEM and nanoindentation results showed that a heavy remineralization of dentin ACLs was obtained via PCCP process with an obvious recovery of mechanical properties while a poor remineralization via PILP process. In conclusion, the PCCP process is a promising strategy for functional remineralization of dentin caries lesions.

Electrochemical treatment of saline or brackish water by capacitive deionization (CDI) is a novel resource-saving solution to ensure the availability of freshwater resources in the environment. MOF (metal–organic framework) derived carbon as a novel carbonaceous material in CDI has received considerable research attention. Among them, ZIF-L-derived carbon (ZC) suffers from problems such as self-aggregation, poor mesoporous structure, and low electrical conductivity, which lead to poor CDI performance. In this paper, a strategy of ZIF-L coated with polydopamine (PDA) is proposed to solve the above problems. When the amount of PDA was 0.5 g, ZIF-L@PDA 0.5 -derived carbon composite (ZPC 0.5 ) is obtained for efficient capacitive deionization. The ZPC 0.5 composite not only maintains the 2D leaf shaped structure consistent with ZIF-L, exposing more active sites, but also has an elevated mesopore occupancy ratio to promote solution diffusion. Meanwhile, the interaction between PDA and ZIF-L makes the ZPC 0.5 have a cavity structure, which improves the mass transfer capacity. The resulting ZPC 0.5 composite has a specific capacitance (165.8 F g −1 ) that is 3.86 times higher than that of ZC (42.9 F g −1 ). Consequently, in 1.2 V, 500 mg/L NaCl solution, the symmetric CDI cell ZPC 0.5 //ZPC 0.5 exhibits an excellent desalination capacity (29.6 mg/g), which is 2.69 times that of the ZC//ZC.

Amine-functionalized Metal-Organic Frameworks (MOFs) stand out as highly promising materials in membrane separation technology. Nevertheless, the incorporation of amine-functionalized additives may lead to uncontrollable alterations in MOF morphology. Two intact blade-like MOFs (PEI (Polyethylenimine)-ZIF-L and PAMAM (Polyamidoamine)-ZIF-L) were successfully synthesized by controlling the conditions and then introduced into Pebax 2533 to fabricate Mixed Matrix Membranes (MMMs). Hydrogen bonding and other interactions with Pebax can be formed by PEI-ZIF-L and PAMAM-ZIF-L to enhance interfacial compatibility. Simultaneously, a reversible reaction with CO 2 molecules can be undergone by the amine groups, resulting in facilitated transport in MMMs. The CO 2 separation performance of the MMMs is enhanced by this process, with the facilitation being more pronounced under humid conditions. Under humid conditions, the CO 2 permeabilities of Pebax-PEI-ZIF-L (2 wt%) and Pebax-PAMAM-ZIF-L (2 wt%) MMMs were 639.4 Barrer and 702.7 Barrer, respectively, while the CO 2 /N 2 selectivities were 26.68 and 24.35, respectively. Both permeability and selectivity exhibited a significant increase of 200% and 130%, respectively, compared to pure Pebax. Furthermore, the MMMs demonstrated excellent long-term stability during 72 h of continuous operation in a humid environment, showcasing significant potential for industrial applications, particularly in humid flue gas carbon capture.

L - Tryptophan (L-Trp), one of the essential amino acids, is crucial for human physiological homeostasis and a limiting amino acid in animal feed. In addition, L-Trp is a precursor of some important biomolecules in the body, such as pentraxin and melatonin. Disorders of L-Trp metabolism in the body may cause Alzheimer's or Parkinson's disease. Therefore, monitoring L-Trp levels in the body fluids quickly and accurately is essential. A colorimetric biosensor was developed for the rapid detection of L-Trp in solutions by coupling a novel, screened, and validated peptide aptamer with Au nanoparticles via the Au-S bond. The biosensor showed a wide linear detection range of 1 µM − 1000 µM, and in addition, it was simple to construct, and the reaction time was as short as 10 s. In the analysis of L-Trp in actual porcine serum samples, the relative standard deviations were 4.52 % to 3.01 % compared with those of the high-performance liquid chromatography method, and the spiked recoveries were 99.2 % to 100.2 %. This new type of recognition probe is likely to gain attention in the field of bioanalysis owing to its biocompatibility, ease of modification, and easy linkage to nanomaterials.

2,3,5,6-Tetramethylpyrazine (TMP), an organic compound with numerous potential health benefits, is extensively used in medical and food products. This research introduces a novel approach for the precise quantification of TMP by employing a poly(L-lysine) modified glassy carbon electrode (pLL/GCE). The pLL/GCE is fabricated through the electrochemical polymerization of L-lysine onto a glassy carbon electrode (GCE) surface. This modification enhances the electrode's electrocatalytic efficiency, resulting in a significant increase in the reduction peak current of TMP. Exploiting this distinctive property, we establish an electrochemical method for quantifying TMP using cyclic voltammetry (CV). Optimized conditions reveal a robust linear correlation between the reduction peak current of TMP and its concentration within two adjacent linearity ranges: 40.0 nmol/L to 1.0 μmol/L and 1.0 μmol/L to 100.0 μmol/L. Notably, this method achieves an impressive detection limit of 20.0 nmol/L. To validate the practicality of this approach, we analyzed various real-world samples, obtaining recovery rates ranging from 98.2% to 100.8%. The developed method holds significant promise for the determination of TMP in medical and food products, underscoring its potential for widespread application.

Metal-organic framework (MOF) membranes demonstrate significant potential for industrial gas separations, yet fabricating vertically aligned two-dimensional (2D) MOF membranes on polymer substrates remains a considerable challenge due to the weak adhesion and difficulty in achieving precise alignment. This study presents a one-step method for the rapid synthesis of vertically aligned ZIF-L membranes, using a polydopamine (PDA)-assisted approach. This method enables the rapid formation of homogenous, thin but defect-free membranes on polymer substrates within 5 min, drastically reducing the preparation time by over 200 times. The vertically aligned ZIF-L membranes, characterized by the interlayer galleries facing the gas mixtures, exhibit an excellent CO 2 permeance exceeding 1200 GPU and a CO 2 /N 2 selectivity of 14.5. The versatility of this approach allows for its application on various flat-sheet or even hollow fiber supports. Our findings highlight the potential of PDA-assisted synthesis for efficient, scalable preparation of high-performance MOF membranes for industrial gas separations.

Arsenic (As) contamination severely limits plant growth and poses a significant threat to human health due to its accumulation in crops like rice. Phytochrome B (PhyB) regulates plant development and responses to abiotic stress, but its role in modulating As stress and regulating As accumulation in rice remains unclear. In this study, we examined the physiological and molecular responses of rice phytochrome B ( OsPHYB ) knockout ( OsPHYB -KO) lines compared with wild-type (WT) plants under arsenite [As(III)] stress. Under As(III) exposure for 7 days, OsPHYB -KO seedlings exhibited significantly increased root length (40.42 %-58.08 %), shoot height (7.41 %-9.22 %), and dry biomass (18.88 %-37.41 %) compared with WT. Chlorophyll fluorescence analysis showed enhanced photosynthetic efficiency in OsPHYB -KO lines, with significant increases in maximum photochemical efficiency of PSII (Fv/Fm) and effective photochemical quantum yield of PSII (φPSII). Notably, As concentrations in both shoots and roots of OsPHYB -KO plants were reduced by 39.78 %-51.89 % and 28.26 %-33.79 %, respectively, compared with WT. OsPHYB -KO lines had 25.56 %-55.63 % higher antioxidant enzyme activities and 43.10 %-54.60 % lower malondialdehyde levels, indicating improved tolerance to oxidative stress. Transcriptomic profiling and qRT-PCR revealed upregulation of genes involved in melatonin (MT) biosynthesis and glutathione (GSH) metabolism, with MT content increased by 16.20 %-67.96 % and GSH content elevated by 38.51 %-117.12 % in both roots and shoots of OsPHYB -KO plants. These results demonstrate that OsPHYB plays a crucial role in As(III) detoxification and provide a foundation for developing low-As rice cultivars.

This paper reports a one-step hydrothermal carbonation method for the synthesis of yellow-green carbon dots from Solanum nigrum L, with phosphoric acid as dopant. The phosphorus-doped carbon dots ( P-CDs) were characterized with TEM, XRD, FT-IR, XPS, and UV–Visible. P-CDs provided good fluorescence enhancement of ciprofloxacin with excellent selectivity and sensitivity, while maintaining robust performance against ionic interference and pH variations. The P-CDs demonstrated high sensitivity toward (ciprofloxacin) CIP, with a wide linear detection range of 0.01–0.1 μM and an ultralow detection limit of 6.37 × 10 −3 μM. Moreover, the P-CDs effectively inhibited S. aureus and E. coli growth at a MIC of 0.27 mg/mL. The P-CDs enable rapid antibiotic detection within 5 min while demonstrating potent antibacterial efficacy with a remarkably low propensity for inducing drug resistance, thereby opening new avenues for combating drug-resistant bacterial infections. Notably, the P-CDs offer a simple, rapid, and highly sensitive detection method for CIP analysis in milk samples, making them a promising tool for food safety monitoring. This finding will support the future development of carbon dots for food chemistry applications.
