Cobalt-based hybrid nanoparticles loaded with curcumin for ligand-enhanced synergistic nanocatalytic therapy/chemotherapy combined with calcium overload

Mengyang Li,Zijie Lu,Chao Fang,Bingzhu Zheng,Yike Fu,Xiang Li

The therapeutic efficacy of Fenton or Fenton-like nanocatalysts is usually restricted by the inappropriate pH value and limited concentration of hydrogen peroxide (H2O2) at the tumor site. Herein, calcium carbonate (CaCO3)-mineralized cobalt silicate hydroxide hollow nanocatalysts (CSO@CaCO3, CC) were synthesized and loaded with curcumin (CCC). This hybrid system can simultaneously realize nanocatalytic therapy, chemotherapy and calcium overload. With the stabilization of liposomes, CCC is able to reach the tumor site smoothly. The CaCO3 shell first degrades in an acidic tumor environment, releasing Cur and Ca2+, and the pH value of the tumor is increased simultaneously. Then the exposed CSO catalyzes the Fenton-like reaction to convert H2O2 into ˙OH and enhances the cytotoxicity of curcumin (Cur) by catalytically oxidizing it to a ˙Cur radical. Curcumin not only induces the chemotherapy effect but also serves as a nucleophilic ligand and an electron donor in the catalytic system, enhancing the Fenton-like activity of CCC by electron transfer. In addition, calcium overload also amplifies the efficacy of ROS-based therapy. In vitro and in vivo results show that CCC exhibited an excellent synergistic tumor inhibition effect without any clear side effect. This work proposes a novel concept of nanocatalytic therapy/chemotherapy synergistic mechanism by the ligand-induced enhancement of Fenton-like catalytic activity, and inspires the construction of combined therapeutic nanoplatforms and multifunctional nanocarriers for drug and ion delivery in the future.

A novel electrochemical sensor based on Fe3+-curcumin/multi-walled carbon nanotubes complex enabling electrochemical sensing of hydroxylamine

Weiyan Xi,Zimeng Kong,Zhaoxue Deng,Yu Chen,Hongwei Mou,Yupeng Zhang,Zhijia Zhang,Zhen Li,Xiaoxue Xu,Wei Zheng

This work reported a novel electrocatalyst , Fe 3+ -curcumin/multi-walled carbon nanotubes (MWCNTs) complex, for efficient detection of hydroxylamine by modifying the glassy carbon electrode (Fe 3+ -curcumin/MWCNTs-modified electrode). The Fe 3+ -curcumin/MWCNTs complex was prepared via a green and facile one-pot synthesis method and the as-prepared complex was characterized using TEM , EDS , XRD, XPS , and EIS. Subsequently, the Fe 3+ -curcumin/MWCNTs complex modified electrode was adopted as an electrochemical sensor for hydroxylamine detection. The catalytic peak current for sensing the hydroxylamine using the Fe 3+ -curcumin/MWCNTs-modified electrode was 9.5 times higher the catalytic peak current using the curcumin/MWCNTs-modified electrodes. The overpotential was reduced by 550 mV compared to MWCNTs-modified electrodes. The interfacial electron transfer rate was 0.37 s −1 for the Fe 3+ -curcumin/MWCNTs-modified electrode. Furthermore, the Fe 3+ -curcumin/MWCNTs-modified electrode exhibited outstanding sensing performance within analyte concentration range from 0.05 to 1100 μM (a low detection limit of 0.02 μM, S/N = 3). Finally, the constructed sensor was successfully adopted for the hydroxylamine detection from the hydrolysis of the pralidoxime iodide injections, demonstrating the practical applications.

The triplet state of tanshinone I and its synergic effect on the phototherapy of cancer cells with curcumin

Chenchen Zhang,Shan Jiang,Kun Li,Mei Wang,Rongrong Zhu,Xiaoyu Sun,Qingxiu Wang,ShiLong Wang

The excited triplet state of tanshinone I (Tan I) extracted from the traditional Chinese medicine Salvia miltiorrhiza Bunge was characterized by laser flash photolysis. The synergic effect of Tan I on the phototherapy of cancer cells with curcumin (Cur) was also investigated by MTT assay because the excited energy transfer from the triplet state of Tan I ( 3 Tan I ∗ ) to Cur occurred. At the same time, the characteristic absorption spectra of 3 Tan I ∗ were recorded, and its molar absorption coefficient and rate constants for several excited energy transfers were obtained. The photo-therapeutic effect of Cur is enhanced by combination with Tan I.

Multifunctionality of self-assembled nanogels of curcumin-hyaluronic acid conjugates on inhibiting amyloid β-protein fibrillation and cytotoxicity

Zhiqiang Jiang,Xiaoyan Dong,Hu Liu,Yongjian Wang,Lei Zhang,Yan Sun

Aggregation of amyloid β-protein (Aβ) is a major pathological hallmark of Alzheimer's disease. Curcumin has been recognized as an inhibitor of Aβ aggregation, but its low water solubility and bioavailability limits its clinical applications. In this work, we conjugated curcumin to hyaluronic acid and drastically improved its solubility and stability. We found that the conjugates self-assembled into nanosized hydrogels that inhibited Aβ fibrillogenesis and mitigated the amyloid cytotoxicity more efficiently than free curcumin. More importantly, we found that there was an optimal curcumin substitution degree, at which the hydrogel exhibited the strongest inhibitory effect. Based on these findings, a mechanistic model was proposed. It suggested that besides the inhibitory effect of the conjugated curcumin, HA provided three synergistic functions. The first is that curcumin encapsulation into nanogels protected cells from the toxicity of free curcumin. The second was an isolation effect of the hydrogel network, which hindered the interactions between Aβ molecules. The third was the counteraction of the hydrophobic binding between Aβ and the conjugated curcumin against the electrostatic repulsion between the like-charged Aβ and HA. The two opposite forces could stretch the conformation of Aβ monomers , slowing down the aggregation and/or leading to off-pathway aggregations. This work offered new insights into the development of more potent nanoparticles for inhibiting Aβ fibrillogenesis and cytotoxicity.

Rational Design of Multifunctional Dendritic Mesoporous Silica Nanoparticles to Load Curcumin and Enhance Efficacy for Breast Cancer Therapy

Jiao Wang,Yue Wang,Qiang Liu,Linnan Yang,Rongrong Zhu,Chengzhong Yu,Shilong Wang

Breast cancer is the primary reason for cancer-related death in women worldwide and the development of new formulations to treat breast cancer patients is crucial. Curcumin (Cur), a natural product, exerts promising anticancer activities against various cancer types. However, its therapeutic efficacy is hindered as a result of poor water solubility, instability, and low bioavailability. The aim of this work is to assess the curative effect of a novel nanoformulation, i.e., Cur-loaded and calcium-doped dendritic mesoporous silica nanoparticles modified with folic acid (Cur-Ca@DMSNs-FA) for breast cancer therapy. The results manifested that Cur-Ca@DMSNs-FA dispersed very well in aqueous solution, released Cur with a pH-responsible profile, and targeted efficiently to human breast cancer MCF-7 cells. Further investigations indicated that Cur-Ca@DMSNs-FA effectively inhibited cell proliferation, increased intracellular ROS generation, decreased mitochondrial membrane potential, and enhanced cell cycle retardation at G2/M phase, leading to a higher apoptosis rate in MCF-7 compared to free Cur. Moreover, the Western blotting analysis demonstrated that Cur-Ca@DMSNs-FA were more active than free Cur through suppression of PI3K/AKT/mTOR and Wnt/β-catenin signaling, and activation of the mitochondria-mediated apoptosis pathway. In addition, hemolysis assay showed that the Ca@DMSNs-FA exhibited good biocompatibility. Last, in vivo studies indicated that when Cur was encapsulated in Ca@DMSNs-FA, the Cur concentration in blood serum and tumor tissues was increased after 1 h intraperitoneal injection. In conclusion, Cur-Ca@DMSNs-FA might act as a potential anticancer drug formulation for breast cancer therapy.

Bioaccessibility and antioxidant activity of curcumin after encapsulated by nano and Pickering emulsion based on chitosan-tripolyphosphate nanoparticles

Bakht Ramin Shah,Chunlan Zhang,Yan Li,Bin Li

In the current study the influence of different lipid-based formulations (Pickering and nanoemulsions) and their droplet size on curcumin encapsulation and bioaccessibility, as well as on its anti-oxidant activity was investigated. Oil-in-water Pickering emulsion stabilized by chitosan-tripolyphosphate (CS-TPP) nanoparticles and nanoemulsions containing an organic phase (Span80:Tween80), were prepared with either medium chain triglyceride (MCT) or corn oil as long chain triglyceride (LCT). An in vitro gastrointestinal (GIT) model consisting of mouth, gastric and intestinal phases was used to characterize the rate and extent of lipid phase digestion of the ingested samples. A centrifugation method determined fraction of curcumin released into mixed micelles after digestion (bioaccessibility). These findings showed that after subjecting to simulated GIT model, all the emulsion systems experienced a progressive increase in mean particle size, due to droplet flocculation and coalescence after digestion. Electrical charge (ζ) of particles was observed to become highly negative as they passed through GIT due to accumulation of anionic bile salts, phospholipids and free fatty acids at their interfaces. The rate and extent of lipid digestion and bioaccessibility of curcumin increased with decreasing mean droplet diameter (NMCT   >   NCO   >   PMCT   >   PCO). Finally, we showed that as compared to free curcumin, the encapsulated curcumin showed higher radical scavenging activity (RSA), which confirmed the protective effect of the emulsion systems on the antioxidant activity of curcumin.

Curcumin Attenuates Colistin-Induced Neurotoxicity in N2a Cells via Anti-inflammatory Activity, Suppression of Oxidative Stress, and Apoptosis

Dai Chongshan,Ciccotosto Giuseppe D.,Cappai Roberto,Tang Shusheng,Li Daowen,Xie Sanlei,Xiao Xilong,Velkov Tony

Neurotoxicity is an unwanted side-effect seen in patients receiving therapy with the “last-line” polymyxin antibiotics. This is the first study to show that colistin-induced neurotoxicity in neuroblastoma-2a (N2a) cells gives rise to an inflammatory response involving the IL-1β/p-IκB-α/NF-κB pathway. Pretreatment with curcumin at 5, 10, and 20 μM for 2 h prior to colistin (200 μM) exposure for 24 h, produced an anti-inflammatory effect by significantly down-regulating the expression of the pro-inflammatory mediators cyclooxygenase-2 (COX-2), phosphorylation of the inhibitor of nuclear factor-kappa B (NF-κB) (p-IκB)-α, and concomitantly NF-κB levels. Moreover, curcumin significantly decreased intracellular reactive oxygen species (ROS) production and increased the activities of the anti-ROS enzymes superoxide dismutase, catalase, and the intracellular levels of glutathione. Curcumin pretreatment also protected the cells from colistin-induced mitochondrial dysfunction, caspase activation, and subsequent apoptosis. Overall, our findings demonstrate for the first time, a potential role for curcumin for treating polymyxin-induced neurotoxicity through the modulation of NF-κB signaling and its potent anti-oxidative and anti-apoptotic effects.

A novel LDL-mimic nanocarrier for the targeted delivery of curcumin into the brain to treat Alzheimer's disease

Fanfei Meng,Sajid Asghar,Shiya Gao,Zhigui Su,Jue Song,Meirong Huo,Weidong Meng,Qineng Ping,Yanyu Xiao

In this study, a novel low density lipoprotein (LDL)-mimic nanostructured lipid carrier (NLC) modified with lactoferrin (Lf) and loaded with curcumin (Cur) was designed for brain-targeted delivery, and its effect on controlling the progression of Alzheimer's disease (AD) in rats was evaluated. NLC with the composition resembling the lipid portion of LDL was prepared by using solvent evaporation method. Lf was adsorbed onto the surface of NLC via electrostatic interaction to yield Lf modified-NLC (Lf-mNLC) as the LDL-mimic nanocarrier. In order to make sure more Lf was adsorbed on the surface of NLC, negatively charged carboxylated polyethylene glycol (100) monostearate (S100-COOH) was synthesized and anchored into NLC. Different levels of S100-COOH (0–0.02   mmol) and Lf modified NLC (0.5–2.5   mg/mL of Lf solution) were prepared and characterized. The uptake and potential cytotoxicities of different preparations were investigated in the brain capillary endothelial cells (BCECs). An AD model of rats was employed to evaluate the therapeutic effects of Lf-mNLC. The results indicate that Lf-mNLC with a high level of Lf showed the maximum uptake in BCECs (1.39 folds greater than NLC) as cellular uptake of Lf-mNLC by BCECs was found to be mediated by the Lf receptor. FRET studies showed Cur still wrapped inside NLC after uptake by BCECs, demonstrating stability of the carrier as it moved across the BBB. Ex vivo imaging studies exposed Lf-mNLC could effectively permeate BBB and preferentially accumulate in the brain (2.78 times greater than NLC). Histopathological evaluation confirmed superior efficacy of Lf-mNLC in controlling the damage associated with AD. In conclusion, Lf-mNLC is a promising drug delivery system for targeting therapy of brain disease.

Electrospun Photodynamic Antibacterial Konjac Glucomannan/Polyvinylpyrrolidone Nanofibers Incorporated with Lignin-Zinc Oxide Nanoparticles and Curcumin for Food Packaging

Huimin Xiao,Lin Wang,Nitong Bu,Jie Duan,Jie Pang

Due to the growing concerns surrounding microbial contamination and food safety, there has been a surge of interest in fabricating novel food packaging with highly efficient antibacterial activity. Herein, we describe novel photodynamic antibacterial konjac glucomannan (KGM)/polyvinylpyrrolidone (PVP) nanofibers incorporated with lignin-zinc oxide composite nanoparticles (L-ZnONPs) and curcumin (Cur) via electrospinning technology. The resulting KGM/PVP/Cur/L-ZnONPs nanofibers exhibited favorable hydrophobic properties (water contact angle: 118.1°), thermal stability, and flexibility (elongation at break: 241.9%). Notably, the inclusion of L-ZnONPs and Cur endowed the nanofibers with remarkable antioxidant (ABTS radical scavenging activity: 98.1%) and photodynamic antimicrobial properties, demonstrating enhanced inhibitory effect against bothStaphylococcus aureus(inhibition: 12.4 mm) andEscherichia coli(12.1 mm). As a proof-of-concept study, we evaluated the feasibility of applying nanofibers to fresh strawberries, and the findings demonstrated that our nanofibers could delay strawberry spoilage and inhibit microbial growth. This photodynamic antimicrobial approach holds promise for design of highly efficient antibacterial food packaging, thereby contributing to enhanced food safety and quality assurance.

Curcumin/pEGCG-encapsulated nanoparticles enhance spinal cord injury recovery by regulating CD74 to alleviate oxidative stress and inflammation

Chen Tianjun,Wan Li,Xiao Yongchun,Wang Ke,Wu Ping,Li Can,Huang Caiqiang,Liu Xiangge,Xue Wei,Sun Guodong,Ji Xin,Lin Hongsheng,Ji Zhisheng

Spinal cord injury (SCI) often accompanies impairment of motor function, yet there is currently no highly effective treatment method specifically for this condition. Oxidative stress and inflammation are pivotal factors contributing to severe neurological deficits after SCI. In this study, a type of curcumin (Cur) nanoparticle (HA-CurNPs) was developed to address this challenge by alleviating oxidative stress and inflammation. Through non-covalent interactions, curcumin (Cur) and poly (-)-epigallocatechin-3-gallate (pEGCG) are co-encapsulated within hyaluronic acid (HA), resulting in nanoparticles termed HA-CurNPs. These nanoparticles gradually release curcumin and pEGCG at the SCI site. The released pEGCG and curcumin not only scavenge reactive oxygen species (ROS) and prevents apoptosis, thereby improving the neuronal microenvironment, but also regulate CD74 to promote microglial polarization toward an M2 phenotype, and inhibits M1 polarization, thereby suppressing the inflammatory response and fostering neuronal regeneration. Moreover, in vivo experiments on SCI mice demonstrate that HA-CurNPs effectively protect neuronal cells and myelin, reduce glial scar formation, thereby facilitating the repair of damaged spinal cord tissues, restoring electrical signaling at the injury site, and improving motor functions. Overall, this study demonstrates that HA-CurNPs significantly reduce oxidative stress and inflammation following SCI, markedly improving motor function in SCI mice. This provides a promising therapeutic approach for the treatment of SCI.

Development of starch-based films reinforced with curcumin-loaded nanocomplexes: Characterization and application in the preservation of blueberries

Xiquan Li,Xinhua Zhang,Jiale Lv,Xiuling Zhang,Yingying Li,Xiaofeng Han,Wentao Zhang

In current study, curcumin-loaded bioactive nanocomplexes (Cur NCs) (2 %, 5 %, 8 %, and 11 %) were used to prepare corn starch (CS)-based composite films (CS-Cur NCs). Fourier-transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy revealed that Cur NCs were uniformly dispersed in the polymer matrix via physical interaction. Moreover, the mechanical, gas barrier, hydrophobicity, optical, and thermal properties and the antioxidant activity of composite films were potentially improved with the addition of Cur NCs. Subsequently, CS-based film with 11 % Cur NCs exhibited high antioxidant activity (the scavenging rates of DPPH and ABTS are 50.07 % ± 0.82 % and 65.26 % ± 1.60 %, respectively) and was used for packaging blueberries. Compared with the control, the CS-Cur NCs packaging treatment effectively improved the appearance and nutrition of blueberries, and maintained the high activity of several antioxidant enzymes. Furthermore, CS-Cur NCs packaging treatment significantly improved the ascorbic acid (AsA) and glutathione (GSH) levels, thus regulating the AsA-GSH cycle system and suppressing the accumulation of reactive oxygen species (ROS). In summary, the CS-Cur NCs packaging could effectively conserve the postharvest quality of blueberries by improving antioxidant enzyme activity and suppressing excessive accumulation of ROS, which contributes to the development of bioactive packaging and provides novel insights into the preservation of blueberries. This work demonstrates that the development of active packaging is promising to absorb the oxidative radicals from food, and protect the food from inherent and external factors, thus enhancing the quality, security, and shelf-life of the food during storage.

Curcumin-derived water-soluble carbon dots without detectable resistance: Dual potentials for antimicrobial activity and infected wound healing

Jinyan Du,Shiji Liu,Jinrui Hou,Xinyue Wu,Haojie Si,Xingya Guo,Shujuan Zhuo

Due to the abuse of traditional antibiotics, the increasing prevalence of antibiotic resistance poses a significant challenge to the finding of novel antimicrobial materials. Curcumin, a natural polyphenolic compound, exhibits broad-spectrum antibacterial, anti-inflammatory, and antioxidant properties. However, its therapeutic utility is limited by its bioavailability. To improve the biological activity of curcumin, the highly water-soluble carbon dots ( E -CDs) were synthesized by a one-step hydrothermal method using curcumin and ethylenediaminetetraacetic acid disodium salt (Na 2 EDTA) as precursors. It was shown that the surfaces of E -CDs still remain the structural and functional groups of their precursors. The as-obtained E -CDs exhibited a higher antimicrobial efficacy against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) with the minimum inhibitory concentration (MIC) of only 40 and 150 ppm, respectively. Strikingly, the E -CDs did not induce significant bacterial resistance while exhibiting excellent biocompatibility. In a mouse model of bacterial-induced wound infection, the wound closure had approached 60 % within just four days, highlighting their significant potential for accelerating wound healing. Our experimental results convincingly demonstrated that the E -CDs are a promising candidate for clinical applications in treating infected wounds and combating drug-resistant bacterial infections.
