Chiral recognition of some D and L-amino acids by microcrystalline cellulose assisted diffusion-ordered NMR spectroscopy

Bowen Han,Jing He,Shaohua Huang

Chiral amino acids play an indispensable role in living organisms. Diffusion-ordered NMR spectroscopy is an effective NMR tool and a noninvasive analytical method for the analyses of mixture without the need for physical separation of the analytes. However, conventional diffusion-ordered NMR spectroscopy method usually fails to resolve the mixtures of chiral amino acids because of their same molecular masses, sizes, and shapes. Microcrystalline cellulose has been gradually gained more and more interest owing to its wide compatibility, surface area, excellent separation efficiency, non-toxicity, cost-effective, and mechanical stability in many fields. Herein we provide a fast, simple, specific and sensitive method to resolve some D and L-amino acids mixtures by using microcrystalline cellulose as a matrix. This work recognized some D and L- proteinogenic amino acid enantiomers with diffusion-ordered NMR for the first time.

Construction of Covalent Triazine Framework With D-A1-A2 Configuration via Post-Modification for Photocatalytic Hydrogen Evolution

Yong Yang,Wei He,Dengmeng Song,Fang Xie,Jiaying Li,Jun Li,Ning Wang

A D-A 1 -A 2 type covalent triazine framework (CTF) was prepared for the photocatalytic hydrogen evolution (HER). In the resulting CTF-CZ-Bpy 2+ , 9-ethyl-9 H -carbazole (CZ) group is used as the donor, triazine ring as the first acceptor, and cyclic diquat (Bpy 2+ ) moiety as the second acceptor. Both experimental and theoretical results show that the D-A 1 -A 2 structure significantly enhances the efficiency of photogenerated charge separation and transfer. As a result, CTF-CZ-Bpy 2+ demonstrated outstanding photocatalytic HER activity with a yield rate of 31.1 mmol g −1 h −1 , which is 7.8 times higher than that of nonfunctionalized CTF-CZ-Bpy.

Preparation and evaluation of a self-nanoemulsifying drug delivery system loaded with Akebia saponin D–phospholipid complex

Jinyang Shen,Jianping Bi,Hongli Tian,Ye Jin,Yuan Wang,Xiaolin Yang,Zhonglin Yang,Junping Kou,Fei Li

Background Akebia saponin D (ASD) exerts various pharmacological activities but with poor oral bioavailability. In this study, a self-nanoemulsifying drug delivery system (SNEDDS) based on the drug–phospholipid complex technique was developed to improve the oral absorption of ASD.Methods ASD–phospholipid complex (APC) was prepared using a solvent-evaporation method and characterized by infrared spectroscopy, differential scanning calorimetry, morphology observation, and solubility test. Oil and cosurfactant were selected according to their ability to dissolve APC, while surfactant was chosen based on its emulsification efficiency in SNEDDS. Pseudoternary phase diagrams were constructed to determine the optimized APC-SNEDDS formulation, which was characterized by droplet size determination, zeta potential determination, and morphology observation. Robustness to dilution and thermodynamic stability of optimized formulation were also evaluated. Subsequently, pharmacokinetic parameters and oral bioavailability of ASD, APC, and APC-SNEDDS were investigated in rats. Results The liposolubility significantly increased 11.4-fold after formation of APC, which was verified by the solubility test in n-octanol. Peceol (Glyceryl monooleate [type 40]), Cremophor® EL (Polyoxyl 35 castor oil), and Transcutol HP (Diethylene glycol monoethyl ether) were selected as oil, surfactant, and cosurfactant, respectively. The optimal formulation was composed of Glyceryl monooleate (type 40), Polyoxyl 35 castor oil, Diethylene glycol monoethyl ether, and APC (1:4.5:4.5:1.74, w/w/w/w), which showed a particle size of 148.0±2.7 nm and a zeta potential of −13.7±0.92 mV after dilution with distilled water at a ratio of 1:100 (w/w) and good colloidal stability. Pharmacokinetic studies showed that APC-SNEDDS exhibited a significantly greater Cmax1 (733.4±203.8 ng/mL) than ASD (437.2±174.2 ng/mL), and a greater Cmax2 (985.8±366.6 ng/mL) than ASD (180.5±75.1 ng/mL) and APC (549.7±113.5 ng/mL). Compared with ASD, Tmax1 and Tmax2 were both remarkably shortened by APC-SNEDDS. The oral bioavailability in rats was enhanced significantly to 183.8% and 431.8% by APC and APC-SNEDDS, respectively. Conclusion These results indicated that APC-SNEDDS was a promising drug delivery system to enhance the oral bioavailability of ASD.

Copper nanoparticles modified nitrogen doped reduced graphene oxide 3-D superstructure for simultaneous determination of dihydroxybenzene isomers

Haifeng Zhou,Tingting Huang,Dejian Chen,Shunxing Li,Huiwu Yu,Yuhui Li,Qijun Song

Copper nanoparticles modified nitrogen doped reduced graphene oxide 3-D superstructure was successively prepared via a two-step reaction. Polydopamine was used as nitrogen source for the fabrication of nitrogen doped reduced graphene oxide and reducing agent for in-situ reduction of CuCl 2 . After calcination under nitrogen protection, the conductivity, electron transfer ability and electrocatalytic activity of nanocomposites were substantially improved. The enhanced redox property could be attributed to the difference in the electronegativity of carbon and nitrogen atoms of the 3-D superstructure. By using this composite material, an electrochemical sensor was constructed for the detection of dihydroxybenzene isomers, which exhibited excellent sensitivity (with limit of detection at 81•131   nmol   L ∧1 levels), wide linear ranges (0.5•720   α/4mol   L ∧1 ) and good selectivity. Hence it was directly applied for the simultaneous determination of dihydroxybenzene isomers in contaminated samples (e.g., waste waters).

β-galactosidase-catalyzed synthesis of 3-O-β-D-galactopyranosyl-sn-glycerol: Optimization by response surface methodology

Wei Wei,Danping Qi,Mingming Diao,Lu Zhaoxin,Fengxia Lv,Haizhen Zhao

In this study, the synthesis of 3-O-β-D-galactopyranosyl-sn-glycerol (GG) was performed by the reverse hydrolysis of D-galactose and glycerol using β-galactosidase from Kluyveromyces lactis. Four process variables, reaction temperature (30.0–45.0 °C), reaction time (24–48 h), enzyme concentration (150.00–350.00 U/mL), and substrate molar ratio (glycerol:D-galactose, 7.5:12.5 mmol/mmol) were investigated and optimized via response surface methodology (RSM) for optimal GG synthesis. Both quadratic equations and the optimal reaction conditions were established. Results showed that the four variables, i.e., reaction temperature, reaction time, enzyme concentration, and substrate molar ratio had significant (p < 0.0001) effects on the synthesis of GG. The optimal reaction conditions were identified as 350.00 U/mL of β-galactosidase concentration and 8.65:1.00 of substrate molar concentration ratio (glycerol: D-galactose) at 39.8 °C and 48 h of reaction. Under these conditions, the GG concentration was 140.03 g/L and GG yield was 55.71%, which both were close to the predicted values (143.26 g/L and 56.73%). This finding proves the RSM to be a useful tool in optimizing process conditions for GG synthesis.

Enzymatic synthesis and antitumor evaluation of mono- and diesters of 3-O-β-D-galactopyranosyl-sn-glycerol

Danping Qi,Wei Wei,Mingming Diao,Fengxia Lv,Zhaoxin Lu,Xiaomei Bie,Haizhen Zhao

Lipase-catalyzed synthesis of mono- and diesters of 3-O-β-D-galactopyranosyl-sn- glycerol (β-GG) with caproic acid was performed in acetone. The simultaneous production of 1(6’)-monoesters and 1,6’-diesters of β-GG was achieved in this reaction. In order to improve the yield of β-GG esters, four process parameters, enzyme concentration (15 ∼ 25 mg/mL), and substrate molar ratio (caproic acid: β-GG= 1.60 ∼ 2.00 mmol: 0.10 mmol), reaction temperature (40 ∼ 60 °C), and reaction time (8 ∼ 12 h), were optimized via response surface methodology (RSM) employing a three-level-four-variable central composite design. Results showed that enzyme concentration had the most significant (p < 0.0001) effect on the synthesis of β-GG esters. The optimal reaction conditions in acetone were given as follows: Novozyme435 concentration 18.65 mg/mL, molar rate of caproic acid to β-GG 19.46:1, reaction temperature 48 °C, and reaction time 9.83 h. The yield of β-GG esters reached 88.08% under above optimized conditions, which was very close to the predicted value 87.95%. The molar ratio of monoester to diesters was 0.39:0.61. β-GG esters with other fatty acyl chains were synthesized based on the optimized conditions. In vitro antitumor activity indicated that the antitumor activity of β-GG esters was dependent on the nature of fatty acids, such as the length of acyl chain, the degree of saturation, as well as the number of acyl chain.

Saikosaponin-D Reduces H2O2-Induced PC12 Cell Apoptosis by Removing ROS and Blocking MAPK-Dependent Oxidative Damage

Lin Xuemei,Wu Songdi,Wang Qing,Shi Yaling,Liu Guozheng,Zhi Jin,Wang Fang

Neuronal oxidative stress (OS) injury has been proven to be associated with many neurodegenerative diseases, and thus, antioxidation treatment is an effective method for treating these diseases. Saikosaponin-D (SSD) is a sapogenin extracted from Bupleurum falcatum and has been shown to have many pharmacological activities. The main purpose of this study was to investigate whether and how SSD protects PC12 cells from H 2 O 2 -induced apoptosis. The non-toxic level of SSD significantly mitigated the H 2 O 2 -induced decrease in cell viability, reduced the apoptosis rate, improved the nuclear morphology, and reduced caspase-3 activation and poly ADP-ribose polymerase (PARP) cleavage. Additionally, exogenous H 2 O 2 -induced apoptosis by damaging the intracellular antioxidation system. SSD significantly slowed the H 2 O 2 -induced release of malonic dialdehyde (MDA) and lactate dehydrogenase and increased the activity of superoxide dismutase (SOD) and the total antioxidant capacity, thereby reducing apoptosis. More importantly, SSD effectively blocked H 2 O 2 -induced phosphorylation of extracellular-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 mitogen-activated protein kinase (p38MAPK), and specific inhibitors of ERK, JNK, and p38-reduced OS injury and apoptosis, suggesting that SSD reduces OS injury and apoptosis via MAPK signalling pathways. Finally, we confirmed that SSD significantly reduced H 2 O 2 -induced reactive oxygen species (ROS) accumulation, and the ROS inhibitor blocked the apoptosis caused by MAPK activation and cellular oxidative damage. In short, our study confirmed that SSD reduces H 2 O 2 -induced PC12 cell apoptosis by removing ROS and blocking MAPK-dependent oxidative damage.

RGD-decorated redox-responsive D-α-tocopherol polyethylene glycol succinate–poly(lactide) nanoparticles for targeted drug delivery

Yuanyuan Guo,Boning Niu,Qingle Song,Yongdan Zhao,Yuling Bao,Songwei Tan,Luqin Si,Zhiping Zhang

Developing multifunctional nanoparticles (NPs) to improve therapeutic efficacy is highly desirable in cancer therapy. In an attempt to respond to such a challenge, a novel copolymer, D-α-tocopherol polyethylene glycol succinate-SS-poly(lactide) (TPGS-SS-PLA) with a disulfide linkage between the TPGS and PLA units, was synthesized for paclitaxel (PTX) delivery. PTX-loaded NPs were fabricated using a nanoprecipitation method to form a particle size of ∼130 nm with good in vitro stability, which can be disassociated under intracellular reductive conditions to release PTX rapidly. The detached TPGS can further promote the drug retention and cytotoxicity through its P-glycoprotein inhibiting property. Integrin-specific targeting peptide, cyclic RGD (cRGD), was further conjugated to the surface of the NPs for targeting the drug delivery. The RGD-decorated NPs exhibited enhanced cellular uptake, PTX accumulation and cell cytotoxicity as compared to non-targeted NPs on murine melanoma B16F10 cells, PTX-sensitive human ovarian A2780 cells and PTX-resistant A2780/T cells. In vivo evaluation of the targeted NPs further showed an extended half-life, increased AUC (area under the concentration–time curve), and significant tumor growth inhibition in mouse sarcoma S180- and B16F10-tumor bearing mice, with reduced side effects as compared to Taxol® and non-targeted NPs. These results indicate that the RGD decorated redox-sensitive NPs could deliver chemotherapies specifically inside the cell via receptor-mediated endocytosis with enhanced efficacy, especially in integrin αvβ3/αvβ5-rich tumor cells. Such a targeted nanocarrier against receptor clustering prepared from a non-cytotoxic and biodegradable copolymer might have great potential in cancer treatment.

ATP/pH Dual Responsive Nanoparticle with d-[des-Arg10]Kallidin Mediated Efficient In Vivo Targeting Drug Delivery

Yu Zhang,Yifei Lu,Feng Wang,Sai An,Yujie Zhang,Tao Sun,Jianhua Zhu,Chen Jiang

Inflammation has been reported as one significant hallmark of breast cancer in relation to tumor development, metastasis, and invasion. The bradykinin receptor 1 (B1R) is highly expressed on inflammatory breast tumor cells thus providing a promising targeting site for tumor recognition and sufficient receptor mediated endocytosis. In this study, the authors evaluate the targeting efficiency of l -form and d -form [des-Arg 10 ]kallidin both in vitro and in vivo. To further improve the drug delivery efficiency, the authors establish a dandelion like nanoparticle by combining the polymeric drug conjugates and aptamer complex together. The doxorubicin conjugated polymer is complexed with adenosine-5′-triphosphate (ATP) sensitive hybridized aptamer in self-assembly process by intercalating into the double strand scaffolds. The acid labile conjugating bond and ATP sensitive aptamer endow the nanoparticle with dual responsiveness to intracellular milieu, thus triggering a quick drug release in tumor cells. Remarkable therapeutic effects and tuned in vivo pharmacokinetics profiles are shown by the aptamer complexed drug conjugates nanoparticle with B1R active targeting modification. Therefore the strategies of B1R targeting and ATP/pH dual-responsiveness nanoparticle help achieve enhanced drug accumulation within tumor cells and efficient chemotherapy for breast cancer.

Enantioselective discrimination of L-/D-phenylalanine by bovine serum albumin and gold nanoparticles modified glassy carbon electrode

Bingdi Liu,Xin Zhang,Yaping Ding,Liqiang Luo,Fenfen Zhang

A new chiral biosensor fabricated by immobilizing bovine serum albumin (BSA) on gold-nanoparticle-modified glassy carbon electrodes, which could discriminate and detect phenylalanine (Phe) enantiomers, was proposed for the first time. The enantioselectivity was characterized by using scanning electron microscopy, cyclic voltammetry, electrochemical impedance spectroscopy and square wave voltammetry. The results showed that the BSA-modified electrode could enantioselectively recognize Phe enantiomers, and larger response signals were obtained from L-Phe. The factors influencing the performance of the biosensor were investigated. The enantiomeric composition of the L-/D-Phe mixture could be determined from the calibration curves. The modified electrodes have the advantages of simple operation, rapid detection and low cost.

Sunlight-Induced Cross-Linked Luminescent Films Based on Polysiloxanes and d-Limonene via Thiol–ene “Click” Chemistry

Yujing Zuo,Jinfeng Cao,Shengyu Feng

The increasing pursuit of biocontained elastic materials led the investigation of the potential use of the monoterpene limonene in film synthesis via thiol–ene reaction. Poly[(mercaptopropyl)methylsiloxane] (PMMS) is first synthesized. By controlling the molar ratio of PMMS and functional monomers, such as polyethylene glycol allyl methyl ether or rhodamine-B, PMMS is partially functionalized while leaving spare mercapto groups that could be further used as cross-linking sites. On the basis of the functionalized PMMS, novel transparent silicone luminescent films with hydrophilic tunable properties are prepared by natural-sunlight-triggered thiol–ene “click” chemistry by using d -limonene as a cross-linker. Their structures and properties are thoroughly characterized. Transparent luminescent films are coated on commercially available UV-light emitting diode (LED) cell from solution medium followed by an in situ cross-linking step; a colorful LED cell is obtained through this facile and efficient method. The UV-LED coated by films show very intense photoluminescence under normal visible light or the light is on, and has very high coloric purity.
