Physicochemical properties, antioxidant and antiproliferative activities of polysaccharides from Morinda citrifolia L. (Noni) based on different extraction methods

Jian Li,Debao Niu,Ying Zhang,Xin-An Zeng

The influences of different extraction methods on the yields, physicochemical properties, antioxidant activities and antiproliferative abilities of polysaccharides from Morinda citrifolia L. (Noni) were investigated and compared. Three extraction methods were applied to extract Noni polysaccharides (NP), which include hot water extraction (HWE), pulsed electric field-assisted extraction (PEFAE) and ultrasonic-assisted extraction (UAE). The extraction yields, sugar contents, uronic acid contents, molecular weights, monosaccharide proportions, antioxidant activities and antiproliferative abilities of three NP samples were significantly different. However, their preliminary structural characteristics and monosaccharide types were similar. More specifically, UAE-NP had the highest extraction yield, the smallest molecular weight and the best antioxidant activities. Especially, UAE-NP showed excellent antiproliferative abilities. Compared with HWE-NP and PEFAE-NP, the IC 50 value of UAE-NP on HepG2 cells reduced by 45.45% and 33.14%, respectively. The highest antioxidant activities of the UAE-NP might be attributed to its smaller molecular weight, lower Gal content and higher Fuc content. The strongest antiproliferative effects of the UAE-NP might be related to the low molecular weight. Overall, UAE could be a potential technique for the extraction of high-quality NP due to its high yield, high efficiency and notable bioactivities.

Effect of pulsed electric fields processing on physiochemical properties and bioactive compounds of apricot juice

Abdul Rahaman,Xin-An Zeng,Muhammad Adil Farooq,Ankita Kumari,Mian Anjum Murtaza,Nabeel Ahmad,Muhammad Faisal Manzoor,Sadia Hassan,Zahoor Ahmad,Chen Bo-Ru,Zhan Jinjing,Azhari Siddeeg

The aim of this study was to investigate the effect of pulsed electric fields (PEF) on bioactive compounds and physiochemical properties present in apricot juice. In which 0, 7, and 14 kV/cm of PEF intensity were applied to treat the juices with a flow rate of 40 ml/min, frequency of 1 kHz at 25 ± 2°C for 500 μs. Thereafter the phenolics, flavonoids, antioxidant volatile compounds, and physiochemical properties of juice were analyzed. Furthermore, the change in functional groups of biological compounds was investigated by Fourier-transform infrared spectroscopy. Results indicated no significant change in pH, °Brix, and color in PEF treated sample even at increased PEF intensities as compared to untreated. A significant increase in cloud value, phenolic compounds, flavonoids, and antioxidant activity of the PEF treated sample was observed. A decrease in the browning index was observed as compared to unprocessed samples. The little variation in biological compounds was observed in PEF treated sample. Practical Applications Nowadays, researchers are paying more attention to non-thermal techniques for the processing of food products, which could enhance the nutritional profile and shelf life stability of food products. PEF retains the heat-sensitive bioactive constituents, extends the shelf life in order to achieve the fresh like quality of liquid products. This research proved that PEF treatment significantly increased the cloud value, phenolic compounds, flavonoids, antioxidant activity, and helps to retain the bioactive compounds present in apricot juice. Furthermore, this present research would also provide valuable data that can be helpful for food industries to design and select PEF technology to process the liquid-based food product like fruit juice.

Effect of modified atmosphere packaging of different oxygen levels on cooking qualities and phytochemicals of brown rice during accelerated aging storage at 37 °C

Hao Huang,Tarun Belwal,Li Li,Yi Wang,Halah Aalim,Zisheng Luo

Consumption of brown rice decreased the risk of chronic diseases, which might be due to the presence of phytochemicals. However, the aging process reduced phytochemical contents of brown rice. The present work investigated modified atmosphere packaging (MAP) of different oxygen levels to preserve cooking qualities and phytochemicals of brown rice during aging storage. Compared with other treatments, 90 % N 2 + 10 % O 2 packaging showed best effect of preserving cooking qualities including texture and microstructure. The oxidation level of brown rice was also the lowest in 90 % N 2 + 10 % O 2 packaging. After storage, contents of phytosterol (campesterol, stigmasterol, β-sitosterol) and phenolic compounds (gallic acid, p-coumaric acid, ferulic acid, caffeic acid, syringic acid, catechin, quercetin) of brown rice in 90 % N 2 + 10 % O 2 packaging were significantly higher than those in other packagings. These results indicated that 90 % N 2 + 10 % O 2 packaging was a suitable MAP method for the retention of acceptable cooking qualities and phytochemical contents of brown rice.

A size-controlled green synthesis of silver nanoparticles by using the berry extract of Sea Buckthorn and their biological activities

Simin Wei,Yinghui Wang,Zhishu Tang,Jinhang Hu,Rui Su,Jingjie Lin,Tuan Zhou,Hui Guo,Nan Wang,Rongrong Xu

Green synthesis of silver nanoparticles (AgNPs) has been extensively studied by using a variety of plant extracts for applications in biomedical sciences and engineering. However, there are no reports on the synthesis of AgNPs by utilizing the berry extract of Sea Buckthorn, which is a traditional Chinese medicine and exhibits a wide spectrum of antioxidant, anti-inflammatory and anticancer activities. In this paper, we report an easy and eco-friendly technique for the preparation of AgNPs using the Sea Buckthorn berry extract under ultrasonic radiation at ambient temperature and the evaluation of both biosynthesis parameters and biological activities. The UV-visible spectrum and dynamic light scattering (DLS) analysis showed that the size of AgNPs was sensitive to the biosynthesis parameters, such as the pH of the extract, material proportion and reaction time, offering a size-controlled synthetic method for AgNPs. The X-ray diffraction (XRD), transmission electron microscopy (TEM), selected area electron diffraction (SAED) and DLS studies showed that the AgNPs (pH 10.0; material proportion 1 : 1; 4 h) had a face-centered cubic (fcc) structure and spherical shape with an average particle size of 27.3 ± 0.2 nm covered by anions, and existed in a monodispersed form with a polydispersity index (PDI) of 0.213. The biosynthesized AgNPs showed potent anticancer activity against human colorectal cancer (HCT116 and SW620), hepatoma cancer (HepG2), breast cancer (MCF-7) and cervical cancer (HeLa) cell lines as well as strong antioxidant activity. The IC50 values for these five cell lines were 8.77, 4.61, 14.59, 16.05 and 27.98 μg mL−1, respectively. However, the biosynthesized AgNPs revealed poor inhibition activities for the growth of E. coli and S. aureus. These results confirmed that the Sea Buckthorn could be a low-cost, nontoxic and eco-friendly natural resource for the synthesis of AgNPs, which might be useful for the development of new alternative antioxidant and anticancer agents in biomedicine.

Simultaneous determination of antioxidant properties and total phenolic content of Siraitia grosvenorii by near infrared spectroscopy

Yan Jun,Huang Xiao-Ping,Zhu Wei-Wei

This study was conducted to evaluate the feasibility of using near-infrared spectroscopy (NIRS) for simultaneous determination of total phenolic content (TPC) and antioxidant properties of Siraitia grosvenorii , specifically 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), ferric reducing antioxidant power (FRAP), and 2,2-diphenyl-1-picrylhydrazyl (DPPH). Partial least squares (PLS) coupled with wavelength selection was applied to develop NIRS calibration models based on a set of 80 samples. All models established were cross- and test set validated. The root mean square error of cross validation (RMSECV) and the coefficient of determination for cross validation \(\left( {{\text{R}}_{{{\text{cv}}}}^{2} } \right) \) were calculated to evaluate the performance of models. The results showed good precision of models for TPC ( \({\text{R}}_{{{\text{cv}}}}^{2}\) = 0.9829, RMSECV = 0.0426), DPPH ( \({\text{R}}_{{{\text{cv}}}}^{2}\) = 0.9477, RMSECV = 0.0279), ABTS ( \({\text{R}}_{{{\text{cv}}}}^{2}\) = 0.8958, RMSECV = 0.0154), and FRAP ( \({\text{R}}_{{{\text{cv}}}}^{2}\) = 0.9137, RMSECV = 0.0176). In addition, 20 unknown samples were used to evaluate the predictive ability of the obtained models, and the ratio performance deviation (RPD) for TPC, ABTS, DPPH, and FRAP ranged from 2.70 to 4.57, which indicate that these models had acceptable predictive ability. These results demonstrated that NIRS has the potential to be a time saving and cost-effective method for the determination of both TPC and antioxidant capacities of Siraitia grosvenorii .

Phytochemicals and bioactive analysis of different sweet tea (Lithocarpus litseifolius [Hance] Chun) varieties

Mengke Wang,Xing Liu,Zejie Zhang,Jiawen Yu,Jiajia Liu,Yiqiang Wu

This paper aims to investigate the constituent phytochemicals and biological activities of six Lithocarpus litseifolius (Hance) Chun ( L. litseifolius ) cultivars. Processed by UPLC-MS/MS and untargeted metabolomics analysis software (MS-DIAL and MS-FINDER), components of different L. litseifolius cultivars were analyzed qualitatively and quantitatively. By conducting a comprehensive analysis of spectra, MS/MS fragmentation, and retention time, 40 compounds (including 6 phenolic glycosides and 34 flavonoids) were identified rapidly and accurately. Moreover, the total phenolic content (TPC), total flavonoid content (TFC), antioxidant, antimicrobial effects, and pheochromocytoma (PC12) cell viability of different L. litseifolius varieties were evaluated. Purple small leaf (PSL) was found to be the most superior variety with the highest antioxidant activities (8.27, 10.09, and 10.80 mg TE/g for DPPH, ABTS, and FRAP assays, respectively), broad-spectrum antimicrobial activity, and lowest cell cytotoxicity. The results showed that these bioactivities were closely related to phytochemicals among the six cultivars. Practical applications L. litseifolius is well known as an important folk drink in the Southern China folklore for its specific efficacy in preventing diabetes. However, there is still a paucity of information on L. litseifolius , as other active components and bioactivities of it remain to be investigated. The wide range of flavonoids discovered in L. litseifolius provides abundant natural health-promoting agents for further applications in medicine and functional food.

Investigation of Chemical Composition, Antioxidant Activity, and the Effects of Alfalfa Flavonoids on Growth Performance

Si Chen,Xiang Li,Xin Liu,Ning Wang,Qi An,Xi Mei Ye,Zi Tong Zhao,Meng Zhao,Yi Han,Ke Hui Ouyang,Wen Jun Wang

The flavonoids were extracted from alfalfa using ethanol assisted with ultrasonic extraction and purified by D101 macroporous resin column chromatography. The chemical composition and content of ethanol elution fractions (EEFs) were assessed by ultrahigh-performance liquid chromatography and hybrid quadrupole time of flight mass spectrometry (UHPLC-Q-TOF-MS) and aluminum nitrate-sodium nitrite-sodium hydroxide colorimetric method. The in vitro antioxidant activity of two EEFs was conducted by scavenging DPPH free radical, and the main antioxidants of 75% EEFs were screened using DPPH-UHPLC. Moreover, the in vivo antioxidant activity of 75% EEFs and the growth performance of broilers were studied. The results showed that the content of 30% and 75% EEFs was 26.20% and 62.57%. Fifteen compounds were identified from 75% EEFs, and five of them were reported in alfalfa for the first time. The scavenging activity of 75% and 30% EEFs (200 μg/mL) against DPPH was 95.51% and 78.85%. The peak area of 5,3,4-trihydroxyflavone and hyperoside was decreased by 82.69% and 76.04%, which exhibited strong scavenging capacities. The total antioxidant capacity (T-AOC), superoxide dismutase (SOD), and glutathione peroxidase (GSH-PX) level of three treated groups against the normal control group (NC) fed with basal diet significantly increased by 3.89-24.49%, 0.53-7.39%, and 0.79-11.79%, respectively. While the malondialdehyde (MDA) decreased by 0.47-18.27%. Compared with the NC, the feed to gain ratio (F : G) of three treated groups was lowered by 2.98-16.53% and survival rate of broilers significantly increased. Consequently, 75% EEFs extracted from alfalfa exhibited powerful antioxidant activities and might be a potential feed additive to poultry and livestock.

Curcumin-loaded liposomes prepared from bovine milk and krill phospholipids: Effects of chemical composition on storage stability, in-vitro digestibility and anti-hyperglycemic properties

Yujie Wu,Bolin Mou,Shuang Song,Chin-Ping Tan,Oi-Ming Lai,Cai Shen,Ling-Zhi Cheong

Present study prepared curcumin liposomes with high encapsulation efficiency (>70%) using bovine milk and krill phospholipids; and investigated the effects of phospholipids composition on storage stability, in-vitro bioavailability, antioxidative and anti-hyperglycemic properties of the curcumin liposomes. Curcumin liposomes prepared from bovine milk phospholipids have smaller particle sizes (163.1 ± 6.42 nm) and greater negative zeta potentials (−26.7 mv) as compared to that prepared from krill phospholipids (particle size: 212.2 ± 4.1 nm, zeta potential: −15.23 mv). In addition, curcumin liposomes from bovine milk phospholipids demonstrated better stability under harsh storage conditions (alkaline conditions, oxygen, high temperature and relative humidity). Nevertheless, curcumin-loaded liposomes prepared from bovine milk phospholipids have inferior bioavailability compared to that prepared from krill phospholipids. No significant differences can be observed in terms of anti-oxidative and anti-hyperglycemic properties of liposomes prepared from both bovine milk and krill phospholipids. Findings from present study will open up new opportunities for development of stable curcumin liposomes with good functional properties (high digestibility, bioavailability and pharmacological effects).

Peptide fraction from sturgeon muscle by pepsin hydrolysis exerts anti-inflammatory effects in LPS-stimulated RAW264.7 macrophages via MAPK and NF-κB pathways

Ruichang Gao,Wanghui Shu,Yang Shen,Quancai Sun,Wengang Jin,Dajing Li,Ying Li,Li Yuan

Previous studies have suggested that polypeptides extracted from milk, soybean, fish, eggs, and meat possess potential anti-inflammatory effects. To date, few studies have reported the anti-inflammatory function of sturgeon peptides and their underlying mechanisms are unknown. The current study was therefore to determine the anti-inflammatory potential of sturgeon peptides with lipopolysaccharide (LPS)-induced RAW264.7 inflammatory model. Pepsin hydrolysate (PeH) was purified by ultrafiltration and Sephadex G-15 gel filtration chromatography. PeH significantly reduced the inflammatory mediator (NO) and inflammatory cytokines (IL-6、TNF-α and IL-1β) expression in a dose-dependent manner. Moreover, the purified sturgeon peptide (F2) possessed strong antioxidant potential and effectively inhibited DPPH and ABTS free radicals. F2 significantly suppressed the expression of MAPKs, IκBα, and NF-κB p65, indicating that F2 exerted anti-inflammatory influence by the inhibition of MAPK and NF-κB pathways.

Effect of purity of tea polysaccharides on its antioxidant and hypoglycemic activities

Minghao Fan,Junxiang Zhu,Yilin Qian,Wei Yue,Ying Xu,Dandan Zhang,Yuqi Yang,Xiaoya Gao,Haiyue He,Dongfeng Wang

The effects of purity of tea polysaccharides (TPS) on its five antioxidant activities and hypoglycemic activities in vitro were studied. The results showed that the higher the purity of TPS, the lower the antioxidant capacity. The purity of FTPSI is the highest (sugar content 80.72%), but its antioxidant activities were lower than those of Fujian tea polysaccharides (FTPS) and FTPSII. The antioxidant activity of tea polysaccharide is related to its protein and polyphenol content (Pearson r > .90). The protective effect of Zhejiang tea polysaccharides and FTPS on human umbilical vein endothelial cells (HUVEC) was better than that of its purified fractions. The inhibition rates of FTPSII (5 and 2 mg/ml) on α-glucosidase (32.76%) and α-amylase (−11.93%) were higher than those of FTPS and FTPSII. Purification does not change the basic structure of TPS. This study has certain reference value for the study of the antioxidant activities of TPS. Meanwhile, TPS can be used as a potential resource with hypoglycemic function. Practical applications A large number of studies have shown that TPS have antioxidant activity. However, several studies considered that the antioxidant activity of TPS mainly comes from the residues of tea polyphenols. Therefore, the in vitro and cell antioxidant activities of TPS were studied in this paper. We believe that both glycoprotein and tea polyphenol are antioxidants of tea, and tea polysaccharide perform preferable effect on hypoglycemic. HUVEC cell model and four in vitro antioxidant test methods were used to study the antioxidant activities of TPS, and two enzyme inhibition activities were used to study the hypoglycemic effect of TPS, in order to provide a theoretical basis for the study of biological activity of TPS.

Phenylenediamine-Based Carbon Nanodots Alleviate Acute Kidney Injury via Preferential Renal Accumulation and Antioxidant Capacity

Jie Gao,Yanfeng Liu,Bo Jiang,Wenmin Cao,Yansheng Kan,Wei Chen,Meng Ding,Guiyang Zhang,Bowen Zhang,Kai Xi,Xudong Jia,Xiaozhi Zhao,Hongqian Guo

As a reactive oxygen species (ROS)-promoted disease, acute kidney injury (AKI) is associated with high mortality and morbidity, but no effective pharmacological treatment is available. Kidney-targeted and ROS-reactive antioxidants are in urgent demand for AKI treatment. A promising nanotechnology-based strategy for targeting renal tubules offers new perspectives for AKI treatment but remains challenging because of the glomerular filtration barrier, which requires ultrasmall-sized therapeutics for penetration and filtration. Here, we fabricated four potential antioxidative carbon nanodots (CNDs) with ultrasmall size. After balancing the antioxidant properties and biocompatibility, m-phenylenediamine-based CNDs (PDA-CNDs) were chosen for further research. PDA-CNDs demonstrated remarkable antioxidant properties for scavenging multiple toxic free radicals, enabling efficient protection of cells under various oxidative stresses in vitro. Moreover, fluorescence imaging revealed that PDA-CNDs preferentially accumulated in the injured kidney of mice with ischemia–reperfusion (IR)-induced AKI. Blood renal function tests and kidney tissue staining revealed the therapeutic efficacy of PDA-CNDs for AKI in both the murine IR-induced AKI model and cisplatin-induced AKI model. Collectively, this is the first study revealing that specific rationally designed CNDs could be a promising pharmacological treatment for AKI induced by ROS.

Preparation of a hordein-quercetin-chitosan antioxidant electrospun nanofibre film for food packaging and improvement of the film hydrophobic properties by heat treatment

Sen Li,Yan Yan,Xiao Guan,Kai Huang

Bio-based functional packaging materials are attracting increasing attention, but poor mechanical properties often hinder their application. This study aims to develop a novel degradable antioxidant nanomaterial with hordein, quercetin and chitosan via electrospinning. Heat treatment was applied to increase the water resistance of the blended nanofibre film. Our results showed that heat treatment significantly improved the water resistance of the blended nanofibre film without affecting its antioxidant activity. Further physiochemical analysis indicated that heat treatment might improve the water resistance of the nanomaterial by decreasing the crystallinity, reducing surface absorbed hydroxyl groups and increasing the structural stability of the blended nanofibres. Together, our results suggested that heat treatment could be an effective method to improve the water resistance of protein-based nanofibres, and the heat-treated hordein-quercetin-chitosan electrospun nanofibre film was a novel biodegradable material with excellent antioxidant activity and water resistance for food packaging.
