A SERS Composite Hydrogel Device for Point-of-Care Analysis of Neurotransmitter in Whole Blood

Lei Wu,Xuefeng Liu,Shenfei Zong,Zhuyuan Wang,Yiping Cui

Point-of-care analysis of neurotransmitters in body fluids plays a significant role in healthcare improvement. Conventional approaches are limited by time-consuming procedures and usually require laboratory instruments for sample preparation. Herein, we developed a surface enhanced Raman spectroscopy (SERS) composite hydrogel device for the rapid analysis of neurotransmitters in whole blood samples. The PEGDA/SA composite hydrogel enabled fast separation of small molecules from the complex blood matrix, while the plasmonic SERS substrate allowed for the sensitive detection of target molecules. 3D printing was employed to integrate the hydrogel membrane and the SERS substrate into a systematic device. The sensor achieved highly sensitive detection of dopamine in whole blood samples with a limit of detection down to 1 nM. The whole detection process from sample preparation to SERS readout can be finished within 5 min. Due to the simple operation and rapid response, the device shows great potential in point-of-care diagnosis and the monitoring of neurological and cardiovascular diseases and disorders. Keywords:surface enhanced Raman spectroscopy (SERS);hydrogel;neurotransmitter;point-of-care test

Surface-Roughened SERS-Active Single Silver Nanowire for Simultaneous Detection of Intracellular and Extracellular pHs

Leyi Fang,Xiao-Tong Pan,Kang Liu,Dechen Jiang,Deju Ye,Li-Na Ji,Kang Wang,Xing-Hua Xia

The simultaneous and accurate detection of intracellular pH (pHi) and extracellular pH (pHe) is essential for studying the complex physiological activities of cancer cells and exploring pH-related therapeutic mechanisms. Here, we developed a super-long silver nanowire-based surface-enhanced Raman scattering (SERS) detection strategy for simultaneous sensing of pHi and pHe. A surface-roughened silver nanowire (AgNW) with a high aspect ratio is prepared at a nanoelectrode tip using a Cu-mediated oxidation process, which is then modified by pH-sensitive 4-mercaptobenzoic acid (4-MBA) to form 4-MBA@AgNW as a pH sensing probe. With the assistance of a 4D microcontroller, 4-MBA@AgNW is efficient in simultaneously detecting pHi and pHe in both 2D and 3D culture cancer cells by SERS, with minimal invasiveness, high sensitivity, and spatial resolution. Further investigation proves that the surface-roughened single AgNW can also be used in monitoring the dynamic variation of pHi and pHe of cancer cells upon stimulation with anticancer drugs or under a hypoxic environment.

Interference-free SERS nanoprobes for labeling and imaging of MT1-MMP in breast cancer cells

Dan Zhu,Anran Li,Yunsong Di,Zhuyuan Wang,Jingzhan Shi,Xiaoqi Ni,Yiping Wang

The expression of membrane type-1 matrix metalloproteinase (MT1-MMP) in cancer cells is critical for understanding the development, invasion and metastasis of cancers. In this study, we devised an interference-free surface-enhanced Raman scattering (SERS) nanoprobe with high selectivity and specificity for MT1-MMP. The nanoprobe was comprised of silver core-silica shell nanoparticle with a Raman reporter tag (4-mercaptobenzonitrile) embedded in the interface. Moreover, the nitrile group in 4-mercaptobenzonitrile shows a unique characteristic peak in the Raman-silent region (1800–2800 cm−1), which eliminates spectral overlapping or background interference in the Raman fingerprint region (500–1800 cm−1). After surface modification with a targeting peptide, the nanoprobe allowed visualization and evaluation of MT1-MMP in breast cancer cells via SERS spectrometry. This interference-free, peptide-functionalized SERS nanoprobe is supposed to be conducive to early diagnosis and invasive assessment of cancer in clinical settings.

Hydrophilic modification of PVDF-based SERS imprinted membrane for the selective detection of L-tyrosine

Hongji Li,Junfu Wang,Haoqi Fang,Hongda Xu,Haochen Yu,Tianyu Zhou,Chunbo Liu,Guangbo Che,Dandan Wang

The polyvinylidene difluoride (PVDF) membrane has received considerable attention as a flexible surface enhanced Raman scattering (SERS) substrate due to its excellent mechanical and physicochemical properties . However, the poor fouling resistance of PVDF membrane due to its intrinsic hydrophobic property limits its practical application. To address this, in this investigation, a SERS imprinted membrane is synthesized based on W 18 O 49 /Ag composites. Firstly, to promote hydrophilicity , N-vinyl-2-pyrrolidone (NVP) and triethoxyvinylsilane (VTES) are copolymerized by hydrolysis condensation and linked with engineered polyvinypyrrolidone (PVP) chains exposed on the surface of membrane. Furthermore, W 18 O 49 /Ag composites are dispersed on the membrane under the assistance of polydopamine (pDA) to promote the pollution resistance. Subsequently, in order to demonstrate the practical detection property, W 18 O 49 /Ag/PVDF membrane is selected as the SERS substrate to synthesize SERS imprinted membrane by precipitation polymerization for the selective detection of L-tyrosine. The characteristic results reveal that the SERS-imprinted membrane exhibits satisfactory hydrophilicity, and it can effectively degrade the pollutant molecules absorbed on its surface under ultraviolet light illumination. It is proved from the detection results that the LOD of WADP-MIMs for L-tyrosine reached 10 −9  mol L −1 when the concentration of L-tyrosine changed between 10 −3 –10 −9  mol L −1 . The correlation coefficient (R 2 ) is 0.994 and the limit of detection is 10 −9  mol L −1 . Meanwhile, it can be applied for the selective detection of L-tyrosine in mixture samples. Overall, this study presents a novel approach for the hydrophilic modification and pollution resistance enhancement of PVDF-based SERS imprinted membrane, which can be effectively utilized for the selective detection of practical samples.

Transparent and flexible AuNSs/PDMS-based SERS substrates for in-situ detection of pesticide residues

Xiaoyuan Ma,Jie Xie,Zhouping Wang,Yin Zhang

Herein, we developed a PDMS-based flexible SERS substrate modified with gold nanostars (AuNSs) for in-situ detection of pesticide residues on fruit surfaces. AuNSs with sharp branches were assembled on aminated PDMS membrane by chemical bonding effect. The AuNSs/PDMS substrate showed good signal uniformity, stability and sensitivity using the Raman signal molecule 4-MBA and the detection concentration of 4-MBA was as low as 10 −8  mol/L. Then the AuNSs/PDMS substrate was used to detect methyl parathion (MP) standard solution. A good linear relationship between SERS intensity at 1342 cm −1 and MP concentration was established in the range of 4 μg/mL–100 μg/mL, and the limit of detection was down to 1.946 μg/mL. Moreover, the AuNSs/PDMS substrate was directly covered on the surface of MP contaminated apple, and the Raman laser was incident from the back of substrate to achieve in-situ detection of pesticide residues. The recovery ratio indicated that the fabricated SERS substrate was realized successfully in real sample detection. AuNSs/PDMS substrate eliminates the requirement for sample pre-processing steps before testing, which can be used as a method for rapid inspection of pesticide in agricultural products on site.

Au@Ag Nanocube-Coated Polystyrene Microspheres as Hierarchical SERS Substrates for Trace Detection of As5+

Haoyu Guan,Guoqiang Fang,Xinxin Li,Haoyu Dong,Siqingaowa Han,Xiang Lin,Li Wang

The plasmon coupling properties of assembled metal nanogap structures are highly sensitive to the distance, shape, orientation, and configuration of faceted nanoparticles. However, the orientation control of nanoparticles is usually limited to in-plane configurations. In this paper, Au@Ag nanocubes (NCs) with sharp edges and corners were assembled on a polystyrene (PS) microsphere array as a hierarchical surface-enhanced Raman spectroscopy (SERS) substrate via a facile self-assembly technique. The Au@Ag NC monolayer undergoes an out-of-plane reconstruction and forms V-shaped nanogaps under the modulation of the PS microsphere array curved surface. This unique V-shaped nanogap can generate electromagnetic fields stronger than those of traditional parallel gaps and make it easier for target molecules to diffuse into the nanogaps. The three-dimensional (3D) SERS substrates have a wider spectral response and 20 times higher SERS activity than those of the Au@Ag NC monolayer films on flat substrates. The high sensitivity was demonstrated by the detection limit down to 10–9 M for 4-MBA with a substrate enhancement factor up to 3.1 × 107. Furthermore, the optimized 3D SERS substrate was applied to determine As5+ in water with detection limits as low as 10–8 M assisted by machine learning, indicating that the 3D SERS substrates have promising applications in food safety, environmental monitoring, and biological diagnosis.

High-sensitivity and throughput optical fiber SERS probes based on laser-induced fractional reaction method

Rong Huang,Shuang Lian,Jingru Li,Yinyu Feng,Sisi Bai,Tongmiao Wu,Minjian Ruan,Pinghui Wu,Xiaojie Li,Shuangshuang Cai,Peipei Jiang

Surface-enhanced Raman scattering (SERS) is widely used in many fields, such as biosensors, medical diagnostics, materials science, and food security. Here, we report a low-cost, high-throughput laser-induced fractional reaction method for optical fiber SERS probes. Under laser irradiation, the local thermal effect and the electromagnetic interaction between nanoparticles effectively contribute to the formation and growth of silver nanoparticles on the optical fiber facet. Sodium dodecyl sulfate (SDS) solution with a concentration of 2 mM is employed as a surfactant to control the shape and size of the silver nanoparticles. A detection limit of 1.0 × 10 −11 M for R6G is achieved, which is, as far as we know, the highest sensitivity that laser-induced fabricated optical fiber SERS probes have achieved. The SERS enhancement factors (EFs) are calculated to be 6.795 × 10 11 . The SERS intensity of R6G at peaks of 621 cm −1 , 1281 cm −1 , and 1359 cm −1 are measured with probes fabricated under the same condition, and showed perfect repeatability with an RSD of less than 4.5%. This new method shows effectively in fabricating optical fiber SERS probes with high sensitivity and good repeatability.

Metal organic framework enabled “split-type” SERS immunoassay platform for highly sensitive and reproducible biosensing

Ling Huang,Sailan Wang,Li Wang,Hongwen Wu,Hongliang Tan,Fugang Xu

MOF-enabled split-type SERS platform decouples probe carrier and substrate. • ZIF-90 enables simple, high-density loading of non-thiolated Raman reporters. • Reagent-free antibody conjugation to ZIF-90 via inherent Schiff-base chemistry. • EDTA-triggered reporter release enables detection on uniform external substrate. • 100-fold lower LOD and > 50% improved reproducibility vs. conventional assays. Surface-enhanced Raman scattering (SERS) holds immense potential for ultrasensitive immunoassays, yet conventional gold nanoparticles (AuNPs) based probe and substrate “two-in-one” designs face critical limitations including low reporter loading capacity, restricted reporter selection, poor signal reproducibility and inefficient antibody conjugation. To overcome these challenges, we propose a metal organic framework (MOF) enabled “split-type” SERS immunosensor, where a MOF material ZIF-90 acts as probe carrier and a uniform silver coated filter paper (Ag@FP) composite serves as external SERS substrate. This MOF based strategy offers four distinct advantages: (i) The abundant porosity of ZIF-90 dramatically increases Raman reporter loading compared to gold nanoparticles (AuNPs) surfaces, enabling significant signal amplification. (ii) Encapsulation of reporters into ZIF frameworks is independent of specific functional groups, thereby highly active, non-thiolated Raman dyes can be used; (iii) The ethylenediaminetetraacetic acid (EDTA)-triggered release of reporters from ZIF-90, coupled with signal acquisition on a uniform external substrate, yields superior signal reproducibility. (iv) Linking antibody to ZIF-90 is achieved via Schiff-base chemistry between their inherent functional groups, eliminating the need for costly coupling reagents. For model biomarker RIgG, this “split-type” SERS immunosensor achieved a detection limit of 2 × 10 −11 g/mL, which is two orders of magnitude lower than that of conventional AuNP-based methods (3 ×10 −9 g/mL), while the relative standard deviation (RSD) was reduced by more than 50%. Excellent recovery and reproducibility in spiked and real serum samples further validate its feasibility in complex biological matrices. This versatile MOF-enabled “split-type” platform provides a compelling new paradigm for sensitive and reproducible SERS biosensing. Cefalexin fingerprinting in water using graphene oxide/gold nanoparticle-modified silica photonic microsphere SERS substrates

Ziyi Wang,Xiu Wang,Ziqiang Li,Yikun Li,Muhammad Shan,Qianjin Li,Jianlin Li

A 3D SPM@GO@AuNPs substrate was built for cefalexin SERS detection. • GO enriches cefalexin and anchors AuNPs for hotspot generation. • Cefalexin was quantified from its 1189 cm-1 Raman band in 10 min. • The assay gave 0.01–10 μg/mL linearity and a 0.41 μg/mL LOD. • Cephalosporin analogues were distinguished by SERS fingerprints. Cefalexin residues in aquatic environments require detection methods that are fast, label-free, and spectroscopically reliable. Surface-enhanced Raman spectroscopy (SERS) can provide molecular fingerprints, but its analytical performance depends on analyte adsorption and the spatial consistency of the enhanced response. Here, a three-dimensional silica photonic microsphere (SPM) substrate was modified with graphene oxide (GO) and gold nanoparticles (AuNPs) to form an SPM@GO@AuNPs substrate for cefalexin fingerprinting in water. GO served as an AuNP-anchoring layer and as an adsorption interface that enriched cefalexin near SERS-active AuNP junctions, whereas the monodisperse SPM provided a curved, readily handled three-dimensional support. Cefalexin bands at 768, 833, 996, 1022, and 1189 cm −1 were resolved, with the 1189 cm −1 band used for quantification. Under optimized conditions, the method exhibited a linear range of 0.01–10 μg/mL, a limit of detection of 0.41 μg/mL, and a 10 min binding step. Cefalexin was distinguished from four cephalosporin analogues using the full spectral fingerprint rather than a single band. Spiked tap-water analysis gave mean recoveries of 86.99–96.83%, supporting the feasibility of direct aqueous screening. The method is best suited to rapid fingerprint-based screening of cefalexin in relatively simple water matrices. Download: Download high-res image (144KB) Download: Download full-size image

Serum SERS spectroscopy combined with classification algorithm in the non-destructive identification of cervical cancer

Gao Ningning,Wang Qing,Tang Jun,Lv Xiaoyi,Li Hongmei,Yue Xiaxia,Zhong Furu,Fu Jihong,Wang Tao

Gold nanoparticles/785 porous silicon photonic crystals (Au NPs/785 PSi PhCs) were used as substrates in combination with a low-concentration serum surface-enhanced Raman spectroscopy (SERS) detection scheme to obtain spectral signals from healthy individuals and cervical cancer patients. The best principal component scores were selected by principal component analysis (PCA) combined with linear discriminant analysis (LDA) and support vector machine (SVM) to analyze the spectral differences distinguishing healthy and cervical cancer patients. The accuracy of the two models was 97.9% and 96.9%, respectively. SERS technique based on Au NPs/785 PSi PhCs has great potential to improve the screening of cervical cancer.

SERS ratiometric strategy leveraging carbon allocation trade-off enables Tremella fuciformis metabolic phenotyping and traceability

Meng Wang,Chenggang Zhang,Miao Yu,Ye Sun

Carbon allocation trade-off enables SERS metabolic phenotyping of fungi. • Polysaccharides/carbohydrates ratio serves as metabolic SERS signatures. • A superior accuracy of 99.2% and an AUC value of 1.0 are achieved. • Portable device enables 15-min on-site fungal phenotyping traceability assay. • It upgrades SERS from chemical fingerprinting to bio-metabolic phenotyping. Background Accurate metabolic phenotyping and geographical traceability of gelatinous fungi are critical for uncovering fungal physiological adaptation mechanisms and advancing edible fungus quality supervision. However, high phenotypic similarity and subtle metabolic differences in homologous fungal samples pose formidable challenges to conventional physicochemical assays. Especially, relying on chemical fingerprinting without biological phenotypic implications, conventional surface-enhanced Raman scattering (SERS) analysis is unable to discern delicate metabolic discrepancies in fungal samples. Results Herein, we propose a metabolic characteristic peak ratio-based SERS strategy using Tremella fuciformis ( T. fuciformis ) as the model. By extracting intrinsic metabolic signatures represented by the carbon allocation trade-off between stress-resistant functional polysaccharides and storage-type carbohydrates, it delivers metabolic specificity and detection stability/reproducibility that are not possible with traditional fingerprint-based SERS fungal analysis. Integrated with machine learning, this method accurately differentiates six T. fuciformis samples, including specimens with distinct metabolic phenotypes as well as geographically differentiated samples with highly similar phenotypic features, achieving a superior accuracy of 99.2% and an AUC of 1.0. Combined with a custom-built portable Raman device we constructed, the entire detection procedure can be completed within 15 min, enabling efficient on-site analysis. Significance and novelty This study establishes a bio-signature-oriented SERS paradigm that upgrades chemical fingerprint identification to fungal metabolic phenotyping, providing a versatile technical platform for exploring fungal metabolic variation and phenotypic differentiation. Template-confined assembly of Ag nanocubes: An approach to fabricate SERS substrate with good performance

Jiaqi Liu,Chunning Chen,Jiaxin Lu,Yalei Wang,Jingtong Zhai,Hongkun Zhao,Nan Lu

Surface-enhanced Raman scattering (SERS) is an important analytical technique. Its detection sensitivity and reproducibility depend on the density and distribution of SERS hotspots. Self-assembly is an efficient method to produce of SERS substrates due to its easy accessibility. However, the assembled defects can hardly be avoided on large area, which could lower the density and uniformity of the hotspots, leading to poor SERS performance. Herein, we report a method to reduce the defects by taking a patterned substrate as template to confine the assembly of Ag nanocubes. The template was prepared based on the combination of photo lithography and self-assembly. Confined by the template, the Ag nanocubes were assembled closely in each dots of the pattern. The limit of detection (LOD) is down to 3.42 × 10 −17  M and the enhanced factor (EF) is up to 3.44 × 10 10 on the prepared substrate for detecting rhodamine 6G (R6G). In addition, the relative standard deviation (RSD) of the different substrates is 8.75 %. The assembled Ag nanocubes exhibits high sensitivity and reproducibility as SERS substrate, which are contributed by the formation of high-density and uniform hotspots. The prepared substrate can be used for detecting trace amounts of melamine in milk with LOD of 2.06 × 10 −7  M and RSD of 6.91 %, so the substrate is applicable for analyzing various analytes.
