Antioxidant play a crucial role in the prevention and treatment of diseases associated with oxidative stress. Curcumin (CUR), as a natural antioxidant, exhibits numerous therapeutic properties, including antioxidant, anti-inflammatory, antibacterial, and antitumor activities. However, its limited bioavailability and poor water solubility hinder its application as an effective antioxidant. In this study, a series of mesoporous silica nanomaterials with distinct morphologies, i.e., mesoporous silica nanoparticles (MSN) and mesoporous silica nanorods (MSR) were synthesized by a template–sediment–etching method. CUR was selected as a model drug and encapsulated into these nanomaterials to improve its bioavailability in vivo. The morphology and size distribution of MSN and MSR were determined through transmission electron microscopy (TEM) imaging and Zetasizer analysis. Fourier transform infrared spectroscopy (FTIR) spectra confirmed the successful encapsulation of CUR within these nanomaterials. Furthermore, these CUR-loaded silica nanomaterials, denoted as CUR@MSN and CUR@MSR, exhibited excellent DPPH and ABTS free radical scavenging activity in vitro. Furthermore, CUR@MSN and CUR@MSR also exhibited obvious in vivo antioxidant activity. This study opens up new avenues for the development of enhanced antioxidants through the utilization of mesoporous silica nanomaterials.

Purpose Tumor xenograft model is an indispensable animal cancer model. In esophageal squamous cell carcinoma (ESCC) research, orthotopic tumor xenograft model establishes tumor xenograft in the animal esophagus, which allows the study of tumorigenesis in its native microenvironment. Materials and Methods In this study,we described two simple and reproducible methods to develop tumor xenograft at the cervical or the abdominal esophagus in nude mice by direct injection of ESCC cells in the esophageal wall. Results In comparing these two methods, the cervical one presented with more clinically relevant features, i.e., esophageal stricture, body weight loss and poor survival. In addition, the derived tumor xenografts accompanied a rapid growth rate and a high tendency to invade into the surrounding structures. This model was subsequently used to study the anti-tumor effect of curcumin, which is known for its potential therapeutic effects in various diseases including cancers, and its analogue SSC-5. SSC-5 was selected among the eight newly synthesized curcumin analogues based on its superior anti-tumor effect demonstrated in an MTT cell proliferation assay and its effects on apoptosis induction and cell cycle arrest in cultured ESCC cells. Treatment of orthotopic tumor-bearing mice with SSC-5 resulted in an inhibition in tumor growth and invasion. Conclusion Taken together, we have established a clinically relevant orthotopic tumor xenograft model that can serve as a preclinical tool for screening new anti-tumor compounds, e.g., SSC-5, in ESCC.

In current study, curcumin-loaded bioactive nanocomplexes (Cur NCs) (2 %, 5 %, 8 %, and 11 %) were used to prepare corn starch (CS)-based composite films (CS-Cur NCs). Fourier-transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy revealed that Cur NCs were uniformly dispersed in the polymer matrix via physical interaction. Moreover, the mechanical, gas barrier, hydrophobicity, optical, and thermal properties and the antioxidant activity of composite films were potentially improved with the addition of Cur NCs. Subsequently, CS-based film with 11 % Cur NCs exhibited high antioxidant activity (the scavenging rates of DPPH and ABTS are 50.07 % ± 0.82 % and 65.26 % ± 1.60 %, respectively) and was used for packaging blueberries. Compared with the control, the CS-Cur NCs packaging treatment effectively improved the appearance and nutrition of blueberries, and maintained the high activity of several antioxidant enzymes. Furthermore, CS-Cur NCs packaging treatment significantly improved the ascorbic acid (AsA) and glutathione (GSH) levels, thus regulating the AsA-GSH cycle system and suppressing the accumulation of reactive oxygen species (ROS). In summary, the CS-Cur NCs packaging could effectively conserve the postharvest quality of blueberries by improving antioxidant enzyme activity and suppressing excessive accumulation of ROS, which contributes to the development of bioactive packaging and provides novel insights into the preservation of blueberries. This work demonstrates that the development of active packaging is promising to absorb the oxidative radicals from food, and protect the food from inherent and external factors, thus enhancing the quality, security, and shelf-life of the food during storage.

Spinal cord injury (SCI) often accompanies impairment of motor function, yet there is currently no highly effective treatment method specifically for this condition. Oxidative stress and inflammation are pivotal factors contributing to severe neurological deficits after SCI. In this study, a type of curcumin (Cur) nanoparticle (HA-CurNPs) was developed to address this challenge by alleviating oxidative stress and inflammation. Through non-covalent interactions, curcumin (Cur) and poly (-)-epigallocatechin-3-gallate (pEGCG) are co-encapsulated within hyaluronic acid (HA), resulting in nanoparticles termed HA-CurNPs. These nanoparticles gradually release curcumin and pEGCG at the SCI site. The released pEGCG and curcumin not only scavenge reactive oxygen species (ROS) and prevents apoptosis, thereby improving the neuronal microenvironment, but also regulate CD74 to promote microglial polarization toward an M2 phenotype, and inhibits M1 polarization, thereby suppressing the inflammatory response and fostering neuronal regeneration. Moreover, in vivo experiments on SCI mice demonstrate that HA-CurNPs effectively protect neuronal cells and myelin, reduce glial scar formation, thereby facilitating the repair of damaged spinal cord tissues, restoring electrical signaling at the injury site, and improving motor functions. Overall, this study demonstrates that HA-CurNPs significantly reduce oxidative stress and inflammation following SCI, markedly improving motor function in SCI mice. This provides a promising therapeutic approach for the treatment of SCI.

Due to the abuse of traditional antibiotics, the increasing prevalence of antibiotic resistance poses a significant challenge to the finding of novel antimicrobial materials. Curcumin, a natural polyphenolic compound, exhibits broad-spectrum antibacterial, anti-inflammatory, and antioxidant properties. However, its therapeutic utility is limited by its bioavailability. To improve the biological activity of curcumin, the highly water-soluble carbon dots ( E -CDs) were synthesized by a one-step hydrothermal method using curcumin and ethylenediaminetetraacetic acid disodium salt (Na 2 EDTA) as precursors. It was shown that the surfaces of E -CDs still remain the structural and functional groups of their precursors. The as-obtained E -CDs exhibited a higher antimicrobial efficacy against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) with the minimum inhibitory concentration (MIC) of only 40 and 150 ppm, respectively. Strikingly, the E -CDs did not induce significant bacterial resistance while exhibiting excellent biocompatibility. In a mouse model of bacterial-induced wound infection, the wound closure had approached 60 % within just four days, highlighting their significant potential for accelerating wound healing. Our experimental results convincingly demonstrated that the E -CDs are a promising candidate for clinical applications in treating infected wounds and combating drug-resistant bacterial infections.

The therapeutic efficacy of Fenton or Fenton-like nanocatalysts is usually restricted by the inappropriate pH value and limited concentration of hydrogen peroxide (H2O2) at the tumor site. Herein, calcium carbonate (CaCO3)-mineralized cobalt silicate hydroxide hollow nanocatalysts (CSO@CaCO3, CC) were synthesized and loaded with curcumin (CCC). This hybrid system can simultaneously realize nanocatalytic therapy, chemotherapy and calcium overload. With the stabilization of liposomes, CCC is able to reach the tumor site smoothly. The CaCO3 shell first degrades in an acidic tumor environment, releasing Cur and Ca2+, and the pH value of the tumor is increased simultaneously. Then the exposed CSO catalyzes the Fenton-like reaction to convert H2O2 into ˙OH and enhances the cytotoxicity of curcumin (Cur) by catalytically oxidizing it to a ˙Cur radical. Curcumin not only induces the chemotherapy effect but also serves as a nucleophilic ligand and an electron donor in the catalytic system, enhancing the Fenton-like activity of CCC by electron transfer. In addition, calcium overload also amplifies the efficacy of ROS-based therapy. In vitro and in vivo results show that CCC exhibited an excellent synergistic tumor inhibition effect without any clear side effect. This work proposes a novel concept of nanocatalytic therapy/chemotherapy synergistic mechanism by the ligand-induced enhancement of Fenton-like catalytic activity, and inspires the construction of combined therapeutic nanoplatforms and multifunctional nanocarriers for drug and ion delivery in the future.

Due to the growing concerns surrounding microbial contamination and food safety, there has been a surge of interest in fabricating novel food packaging with highly efficient antibacterial activity. Herein, we describe novel photodynamic antibacterial konjac glucomannan (KGM)/polyvinylpyrrolidone (PVP) nanofibers incorporated with lignin-zinc oxide composite nanoparticles (L-ZnONPs) and curcumin (Cur) via electrospinning technology. The resulting KGM/PVP/Cur/L-ZnONPs nanofibers exhibited favorable hydrophobic properties (water contact angle: 118.1°), thermal stability, and flexibility (elongation at break: 241.9%). Notably, the inclusion of L-ZnONPs and Cur endowed the nanofibers with remarkable antioxidant (ABTS radical scavenging activity: 98.1%) and photodynamic antimicrobial properties, demonstrating enhanced inhibitory effect against bothStaphylococcus aureus(inhibition: 12.4 mm) andEscherichia coli(12.1 mm). As a proof-of-concept study, we evaluated the feasibility of applying nanofibers to fresh strawberries, and the findings demonstrated that our nanofibers could delay strawberry spoilage and inhibit microbial growth. This photodynamic antimicrobial approach holds promise for design of highly efficient antibacterial food packaging, thereby contributing to enhanced food safety and quality assurance.

This work reported a novel electrocatalyst , Fe 3+ -curcumin/multi-walled carbon nanotubes (MWCNTs) complex, for efficient detection of hydroxylamine by modifying the glassy carbon electrode (Fe 3+ -curcumin/MWCNTs-modified electrode). The Fe 3+ -curcumin/MWCNTs complex was prepared via a green and facile one-pot synthesis method and the as-prepared complex was characterized using TEM , EDS , XRD, XPS , and EIS. Subsequently, the Fe 3+ -curcumin/MWCNTs complex modified electrode was adopted as an electrochemical sensor for hydroxylamine detection. The catalytic peak current for sensing the hydroxylamine using the Fe 3+ -curcumin/MWCNTs-modified electrode was 9.5 times higher the catalytic peak current using the curcumin/MWCNTs-modified electrodes. The overpotential was reduced by 550 mV compared to MWCNTs-modified electrodes. The interfacial electron transfer rate was 0.37 s −1 for the Fe 3+ -curcumin/MWCNTs-modified electrode. Furthermore, the Fe 3+ -curcumin/MWCNTs-modified electrode exhibited outstanding sensing performance within analyte concentration range from 0.05 to 1100 μM (a low detection limit of 0.02 μM, S/N = 3). Finally, the constructed sensor was successfully adopted for the hydroxylamine detection from the hydrolysis of the pralidoxime iodide injections, demonstrating the practical applications.

Curcumin (Cur) is of interest due to its widely reported pharmacology, which is limited by its low water solubility and low bioavailability of clinical use. Here, we report the characterization and application of a multiple stimulus-responsive nanogel loaded with Cur. Sensitive P(AM-co-NIPAM) nanogels for pH, temperature, and reduction stimuli were prepared by reflux precipitation polymerization method using AM and NIPAM as raw materials. DLS and SEM analyses showed that the fabricated nanogels had a uniform size distribution with spherical and nucleated-shell structures. By adjusting the feeding amounts of reactive monomers, cross-linkers, and initiators, a series of monodisperse nanogels with different particle sizes could be obtained. The representative nanogels have different Cur-loaded behaviors at different temperatures, times, and initial concentrations of the drug, as well as a sensitive release to changes in pH, temperature, or reducing conditions. Cur-P(AM-co-NIPAM) nanogels showed superior antioxidant activity over the free Cur in vitro . Meanwhile, it inhibited LPS and H 2 O 2 -induced oxidative stress, which could effectively inhibit the cellular production of excessive reactive oxygen species and had a protective effect against LPS-induced cellular damage. This nanogel controlled release system significantly improved the safety limit of Cur without hemolytic behavior and had good cytocompatibility. The successful development of such Cur nanogels will lead to advanced materials that can be functionalized and optimized for the controlled delivery of small molecules or biomolecules for targeted therapeutic and biomedical applications.

Functional ingredient bioaccessibility is limited by instability and low solubility, thus, edible macromolecules are used to enhance delivery. This study presents a safe, sustainable method to prepare dual-network emulsion gel (CSEG). The gel is formed via Ca 2+ -induced crosslinking of sodium alginate (SA) and coffee cherry-derived polysaccharide (CCP) to enhance bioavailability. CCP and SA form a stable bimolecular network through hydrogen bonds and calcium ion coordination, improving gel stability. The effects of CCP/SA ratios on CSEG properties and curcumin delivery are also investigated. At the optimal CCP: SA mass ratio of 3:2, the system shows superior crosslinking, forming a strong gel that resists gastric shear while maintaining viscoelasticity. The curcumin-loaded CSEG demonstrates excellent pH stability, preserving curcumin in gastric fluid even in highly acidic conditions. In vitro digestion studies reveal that CSEG boosts curcumin bioavailability from 29.62 % to 63.94 %. This work reveals emulsion gel structure-function links, guiding bioactive carrier design.

To meet the practical application demands of nanofiber membranes (NFMs) under humid conditions, a PVA/NC/ZnO (S) NFM was successfully fabricated by incorporating nano zinc oxide (ZnO (S) ) into a polyvinyl alcohol (PVA)/nanocellulose (NC) matrix, combined with nitrogen-assisted thermal treatment. The synergistic effect of ZnO ( S ) -induced heterogeneous nucleation and thermal processing significantly enhanced the crystallinity of the NFM (up to 49.71 %), improving its anti-swelling behavior and liquid absorption capacity—with an absorption rate of up to 1906 % for artificial blood. Building on this foundation, a dual-layer drug-loading strategy was developed by combining the PVA/NC/ZnO (S) NFM with hydroxypropyl-β-cyclodextrin (HP-β-CD) to encapsulate curcumin (Cur). The resulting drug-loaded NFM maintained excellent mechanical strength (29.17 MPa at a 1.5 % drug loading) and demonstrated favorable moisture resistance and antibacterial activity. Moreover, its in vitro drug release behavior could be finely tuned by adjusting the drug loading and external pH conditions. Overall, the synergistic enhancement and dual-encapsulation strategy proposed in this study provides an effective solution to the common challenges of swelling and instability in traditional drug carriers under wet conditions, and demonstrates promising potential in the design and application of biomedical wound dressings.

Acute kidney injury (AKI) is a common clinical syndrome characterized by a rapid decline in kidney function, with inflammation being a key driver. In this study, the hydrophobic drug Rhein was conjugated to hyaluronic acid (HA) via esterification of its carboxyl group. The HA was then modified with 4-hydroxymethylphenylboronic acid pinacol ester (PBAP), which is responsive to reactive oxygen species (ROS). Finally, curcumin was loaded onto this construct to develop a ROS-responsive nanodelivery system (RHP@Cur). In vitro, the nanoparticles showed good ROS responsiveness in H 2 O 2 medium and successfully targeted the CD44 receptors on macrophages. Compared to curcumin alone, RHP@Cur significantly inhibited the secretion of NO and TNF-α and blocked NF-κB activation, with more pronounced effects. RHP itself also exhibited some anti-inflammatory effects. In an in vivo AKI model, the anti-inflammatory effects of RHP@Cur were consistent with the in vitro results. H&E staining results showed that RHP@Cur significantly improved kidney pathological damage. Immunohistochemical results revealed that it reduced the deposition of kidney injury molecule-1 (KIM-1) in the kidneys, with better effects than curcumin and RHP. These results suggest that Rhein exerts a synergistic anti-inflammatory effect, and its potential for combined use with curcumin for kidney therapy makes RHP@Cur more effective than RHP in treating AKI. In addition, since RHP@Cur delivers Cur, it enhances the therapeutic effect of Cur. Therefore, RHP@Cur provides a potential effective means for the treatment of acute kidney injury.
