In situ reduction synthesis of quinoline-based copper(I) complexes: “Self-activating” chemical nuclease, antioxidation and anticancer activity

Yong-Po Zhang,Qing He,Xin-Hui Zhou,Guang-Huan Liu,Ai-Qin Yue,Chun-Yan Gao,Jin-Zhong Zhao,Wei-Jun Du,Shi-Ping Yan

Two new Cu(І) complexes, [Cu 2 (L 1 ) 2 (μ-Br) 2 ] (1) and [Cu 2 (L 1 ) 2 (μ-Cl) 2 ] (2) were synthesized from Cu(II) halogen salts and the reduced Schiff base ligand. Stable Cu(І) complexes were obtained via in situ reduction of Cu(II) to Cu(I) and the C N was oxidize to C = N in the ligand. The structures of two complexes were characterized by X-ray single crystal diffraction analysis as well as IR and UV–Vis spectra. The two complexes have potential antioxidant capacity and exhibit efficiently self-activated DNA cleavage activity and hydroxyl radical ROS cleavage mechanism. MTT was used to assay cytotoxicity of the complexes toward a series of tumor cell and HUVEC cell lines. Both complexes exhibited higher selective cytotoxicity against tumor cells than normal cells, which is significantly superior to the positive control. The complexes could induce cell apoptosis via mitochondrial pathway by increase of ROS levels and decrease of mitochondrial membrane potential (Δψ m ). Furthermore, the complexes can obviously inhibit the migration and invasion of the quiz tumor cells, as well as inhibit the angiogenesis of HUVEC cells.

A digital multimeter-based portable photoelectrochemical immunoassay for the detection of cardiac troponin I with enzymatic biocatalytic precipitation

Bizhen Huang,Jing Ran,Ruishen Li,Wei Zhuang,Jiabi Chen,Haixin Guo

Early diagnosis of acute myocardial infarction (AMI) can significantly reduce mortality, which can be achieved by the highly sensitive detection of AMI-specific cardiac troponin I (cTnI) biomarkers. Herein a highly sensitive and portable photoelectrochemical (PEC) immunoassay for the detection of cTnI was innovatively fabricated based on an efficient photocurrent response of Cu2O coupling with a split-type enzymatic biocatalytic precipitation reaction with a digital multimeter readout. Initially, Cu2O cubic nanomaterials with a good photocurrent response were obtained by facile green room temperature synthesis and dropped on fluorine-doped tin oxide (FTO) plates as photoactive materials (Cu2O/FTO). In the presence of target cTnI, horseradish peroxidase (HRP) on the detection antibody catalyzes the substrate 4-chloro1-naphthol (4-CN) to produce insoluble precipitates of benzo-4-chlorohexadienone (4-CD) on Cu2O/FTO with the assistance of H2O2, resulting in a decrease in the photocurrent of the Cu2O/FTO electrode. To make the whole inspection process more concise and efficient, we used a flashlight and a digital multimeter as the excitation light source and reading device for the PEC sensing system, respectively. Under the optimal conditions of immunoreaction time, pH, and loading, the photocurrent of Cu2O/FTO decreased with increasing target cTnI concentration with a limit of detection (LOD) of 4.7 pg mL−1 in the operating range of 0.01–10 ng mL−1. It is of interest that the developed portable PEC immunoassay is also capable of detecting cTnI in human serum samples with acceptable accuracy compared to the reference cTnI enzyme-linked immunosorbent assay (ELISA) method while maintaining portability and sensitivity.

A Ratiometric Electrochemical Aptasensor with Exonuclease I-assisted Target Recycling Amplification for Highly Sensitive Detection of Carcinoembryonic Antigen

Ping Wang,Huikai Ma,Yaoyao Xie,Sanqiang Li

In this study, a ratiometric electrochemical aptasensor was proposed for the highly sensitive detection of carcinoembryonic antigen (CEA) with exonuclease I (Exo I)-assisted target recycling amplification. The thiolated methylene blue (MB)-labelled hairpin capture probe (MB-HP) was immobilized on the surface of gold nanoparticles (AuNPs) modified gold electrode (AuE) via Au-S bonding, and the hybridization between MB-HP and ferrocene (Fc)-labelled aptamer (Fc-Apt) formed a duplex structure, leading to Fc molecules approaching to the electrode surface while MB molecules were far away. In the presence of CEA, its specific recognition with the aptamer caused the release of Fc-Apt from the electrode surface and subsequently the formation of a hairpin structure for MB-HP, resulting in both “signal-on” of MB (I MB ) and “signal-off” of Fc (I Fc ). To improve the sensitivity of the aptasensor, an Exo I-assisted target recycling amplification strategy was designed to achieve an amplified ratiometric signal. Under the optimal conditions, the proposed aptasensor showed a linear detection range from 10 pg mL -1 to 100 ng mL -1 with a detection limit of 4.1 pg mL -1 . Furthermore, the stability, reproducibility, selectively and real sample detection ability of the aptasensor were investigated and satisfactory results were obtained.

Tanshinone I alleviates steroid-induced osteonecrosis of femoral heads and promotes angiogenesis: in vivo and in vitro studies

Sun Kai,Xue Yuman,Zhang Xin,Li Xiaodong,Zhao Jun,Xu Xilin,Zhang Xiaofeng,Yang Fubiao

Background The impaired blood supply to the bones is an important pathological feature of steroid-induced osteonecrosis of the femoral head (SIONFH). Danshen is a Chinese herb that shows therapeutic effects on SIONFH, but the effects of one of its major bioactive constituents, Tanshinone I (TsI), on SIONFH remain unknown. Here, we evaluated the effects of TsI on SIONFH, particularly focusing on its effects on angiogenesis, in in vivo and in vitro research. Methods SIONFH was induced in Sprague–Dawley rats by an intramuscular injection of methylprednisolone (40 mg/kg) in combination with an intraperitoneal injection of lipopolysaccharide (20 μg/kg). Morphological alterations of the femoral head were observed by dual-energy X-ray absorptiometry and HE staining. Western blot, qRT-PCR, and immunohistochemical/immunofluorescence staining were used to determine gene expression. Results TsI (10 mg/kg) alleviated bone loss and rescued the expression of angiogenesis-related molecules (CD31, VWF, VEGF, and VEGFR2) in the femoral heads of SIONFH rats. Notably, TsI rescued the down-regulated expression of SRY-box transcription factor 11 (SOX11) in CD31 + endothelial cells in the femoral heads of SIONFH rats. In vitro studies showed that TsI preserved the dexamethasone-harmed angiogenic property (migration and tube formation) of human umbilical vein cells (EA.hy926), suppressed dexamethasone-induced cell apoptosis, reduced pro-apoptotic proteins (cytosolic cytochrome C, Bax, and caspase 3/9) and increased anti-apoptotic protein Bcl-2, whereas silencing of SOX11 reversed these beneficial effects. Conclusions This study demonstrates that TsI alleviates SIONFH and promotes angiogenesis by regulating SOX11 expression. Our work would provide new evidence for the application of TsI to treat SIONFH. Graphical Abstract

Enhanced activation of H2O2 by bimetallic Cu2SnS3: A new insight for Cu (II)/Cu (I) redox cycle promotion

Qiao Yang,Chuanhai Xia,Shuai Chen,Xuezhi Cao,Jingcheng Hao

Hypothesis Despite that the development of Cu 2 SnS 3 (CTS) catalyst has attracted increasing interests, few study has reported to investigate its heterogeneous catalytic degradation of organic pollutants in a Fenton-like process. Furthermore, the influence of Sn components towards Cu (II)/Cu (I) redox cycling in CTS catalytic systems remains a fascinating research. Experiments In this work, a series of CTS catalysts with controlled crystalline phases were prepared via a microwave-assisted pathway and applied in the H 2 O 2 activation for phenol degradation. The efficiency of phenol degradation in CTS-1/H 2 O 2 system (CTS-1: the molar ratio of Sn (copper acetate) and Cu (tin dichloride) is determined to be Sn:Cu = 1:1) was systematically investigated by controlling various reaction parameters including H 2 O 2 dosage, initial pH and reaction temperature. We discovered that Cu 2 SnS 3 exhibited superior catalytic activity to the contrast monometallic Cu or Sn sulfides and Cu (I) acted as the dominant active sites. The higher Cu (I) proportions conduce to the higher catalytic activities of CTS catalysts. Quenching experiments and electron paramagnetic resonance (EPR) further proved that the activation of H 2 O 2 by CTS catalyst produces reactive oxygen species (ROS) and subsequently leads to degradation of the contaminants. A reasonable mechanism of enhanced H 2 O 2 activation in Fenton-like reaction of CTS/H 2 O 2 system was proposed for phenol degradation by investigating the roles of copper, tin and sulfur species. Findings The developed CTS acted as a promising catalyst in Fenton-like oxidation progress for phenol degradation. Importantly, the copper and tin species contribute to a synergetic effect for the promotion of Cu (II)/Cu (I) redox cycle, which thus enhanced the activation of H 2 O 2 . Our work may offer new insight on the facilitation of Cu (II)/Cu (I) redox cycle in Cu-based Fenton-like catalytic systems.

Chan Li,Sheng Zhou,Chuncheng Chen,Lijun Zhu,Shijie Li,Zhuoyue Song,Jian Liang,Chunzhi Tang,Nenggui Xu,Tao Liu,Shihui Liu

Disulfiram (DSF), a drug for alcohol withdrawal , has attracted extensive scientific attention due to its potential to treat cancer. The metabolite of DSF, diethyl dithiocarbamate (DDTC), forms a Cu-DDTC complex in vivo with copper ions , which has been shown to be a proteasome inhibitor with high antitumor activity . However, the in vivo stability of Cu-DDTC complexes remains a challenge. In this study, the nanomedicine [email protected] with high antitumor activity was prepared by using the nanoscale metal-organic framework (MOF) Cu-BTC as a carrier and loading diethyldithiocarbamate (DDTC) through coordination interaction. The results showed that [email protected] had high drug loading and adequate stability, and exhibited DDTC-Cu(I) chemical valence characteristics and polycrystalline structure features. In vitro cytocompatibility investigation and animal xenograft tumor model evaluation demonstrated the anti-cancer potential of [email protected] , especially the combination of [email protected] with low-dose cisplatin showed significant antitumor effect and biosafety. This study provides a feasible protocol for developing antitumor drugs based on the drug repurposing strategy.

Preparation of polyamidoamine dendrimer-functionalized chitosan beads for the removal of Ag(I), Cu(II), and Pb(II)

Xiaoqi Liu,Yanyun Zhang,Yan Liu,Ting'an Zhang

Chitosan bead grafted by third-generation dendrimers (CB-G3) with a diameter of 1.40 mm was synthesized to investigate their performance in recovering Ag(I), Cu(II), and Pb(II) ions in aqueous media. The prepared adsorbents were characterized by XRD, FT-IR, elemental analysis, TGA , and SEM, and the effects of pH, contact time, concentration, and temperature were examined. The results showed that the adsorbents were successfully fabricated. The optimum pH value was 5, and the increased generation number contributed to adsorption capacity improvement, indicating that electrostatic interactions between amine groups and metal ions are the governing mechanism of adsorption by the CB-G3. The kinetics, isotherms, and thermodynamics of Ag(I), Cu(II), and Pb(II) adsorption onto the CB-G3 were investigated. The adsorption processes can be described using pseudo-second-order kinetic and Langmuir models. The maximum monolayer adsorption capacities were 105.62, 88.82, and 97.87 mg·g −1 for Ag(I), Cu(II), and Pb(II) at 30 °C within 210 min, respectively. Electrostatic interactions and hydrogen bonds are the main mechanisms between metal ions and N atoms. Therefore, the CB-G3 is a promising candidate for Ag(I), Cu(II), and Pb(II) adsorption owing to its splendid ability in easy separation, good adsorptivity, and reusability for efficiently adsorbing Ag(I), Cu(II), and Pb(II) ions.

Multispectral photoacoustic imaging of cancer with broadband CuS nanoparticles covering both near-infrared I and II biological windows

Yubin Liu,Duyang Gao,Mengze Xu,Zhen Yuan

In this study, CuS nanoparticles with optical absorption covering both near-infrared I (NIR-I) and NIR-II biological windows were prepared and served as the contrast agents for multispectral photoacoustic imaging. The physiological parameters including concentrations of deoxyhemoglobin and oxyhemoglobin as well as the water content in the tumor location were quantified based on the multispectral photoacoustic reconstruction method. More importantly, the concentration of CuS nanoparticles/drugs accumulated in the tumor was also recovered after intravenously injection, which are essential for image-guided cancer theranostics. In addition, phantom and in vivo experimental tests were performed to inspect and compare the imaging depth and signal-to-noise ratio (SNR) between the two NIR biological windows. Interestingly, we discovered that a higher SNR was obtained in the NIR-II window than that in the NIR-I window. Meanwhile, the multispectral imaging results also demonstrated that the imaging contrast and penetration depth in the NIR-II window were also significantly improved as compared to those from the NIR-I window.

Synthesis, characterization, and luminescent properties of three-coordinate copper(I) halide complexes containing a carbazolyl monodentate phosphine ligand

Qian Li,Qiong Wei,Pei Xie,Li Liu,Xin-Xin Zhong,Fa-Bao Li,Nian-Yong Zhu,Wai-Yeung Wong,Wesley Ting-Kwok Chan,Hai-Mei Qin,Njud S. Alharbi

A series of three-coordinate dinuclear copper halide complexes containing acarbazolyl monodentate phosphine ligand, [CuX(dppc)]2 (dppc =3,6-di-tert-butyl-9-[2-(diphenylphosphino)phenyl]carbazole, X = I (1), Br (2), and Cl (3)), were synthesized, and their molecular structures and photophysical properties were investigated. The structural analysis reveals that two copper(I) centers are bridged by two halogen ligands to form a dinuclear structure with a four-membered Cu2X2 ring. Crystal structures of 1-3 contain 1-D supramolecular arrays constructed by intermolecular C–H···π interactions. These complexes exhibit blue to green emission in the solid state at room temperature and have peak emission wavelengths at 492–495 nm with microsecond lifetimes and low emission quantum yields (< 0.01%). The emission of 1-3 mainly originates from MLCT, XLCT, and IL (intraligand) transitions. The three complexes displayed good thermal stability.

N, S, I co-doped carbon dots for folic acid and temperature sensing and applied to cellular imaging

Zhao,Jianhao,Yaling

The application of fluorescent carbon dots in bio-imaging has huge positive significance in the field of biomedicine. By taking this advantage, herein we prepared nitrogen, sulfur and iodine doped carbon dots (N,S,I-CDs) by a facile hydrothermal reaction using C 3 N 3 S 3 , potassium iodate (KIO 3 ) and ethylenediamine (EDA), and the obtained N,S,I-CDs show bright blue fluorescence with a high fluorescence quantum yield of about 32.4%. The prepared N, S, I-CDs could interact with the folic acid (FA) with high selectivity, lead to development of a high sensitive method for the FA detection from 0.1 to 175 μM wide linear range with a detection limit of 84 nM (S/N = 3) and also applied them in U-2 OS cells imaging. Moreover, this sensor possessed a good sensitivity, linearity and reversibility in the temperature range of 10–80 °C, and successfully applied for the temperature sensing in cell HT-29 samples. This investigation illustrates that as-prepared N, S, I-CDs probe may have great potential as a high-performance platform for the accurate recognition of temperature in cells and could provide a new tool for the detection of FA in cells.

All-inorganic perovskite quantum dots CsPbX3 (Br/I) for highly sensitive and selective detection of explosive picric acid

Xinfeng Chen,Congming Sun,Yang Liu,Long Yu,Kui Zhang,Abdullah M. Asiri,Hadi M. Marwani,Hua Tan,Yuejie Ai,Xiangke Wang,Suhua Wang

As a new family of fluorescent materials, all inorganic perovskite quantum dots (PQDs) CsPbX 3 (Cl, Br, I) have attracted rising attention due to their intrinsic optical superiorities and potential applications in various fields. This work takes advantage of the bright fluorescence of PQDs and the strong electrophilic properties of picric acid (PA) to construct an analytical method for sensitive and selective discrimination of PA against other nitroaromatic explosives, and the limit of detection is as low as 0.8 nM. The quenching mechanism was elucidated as the formation of electrostatic complex and the electron transfer between PA and PQDs. To facilitate visual and on-site detection, the CsPbBr 3 PQDs were printed onto microfiltration membrane by inkjet printing to fabricate test papers for rapid and on-site detections of explosive and pollutant PA, showing a great research prospect in the detection of ultratrace PA at different sites.

Monodispersed Copper(I)-Based Nano Metal–Organic Framework as a Biodegradable Drug Carrier with Enhanced Photodynamic Therapy Efficacy

Xuechao Cai,Zhongxi Xie,Binbin Ding,Shuai Shao,Shuang Liang,Maolin Pang,Jun Lin

Photodynamic therapy (PDT) has emerged as an alternative treatment of cancers. However, the therapeutic efficiency of PDT is severely limited by the microenvironment of insufficient oxygen (O 2 ) supply and overexpression of glutathione (GSH) in the tumor. Herein, a biodegradable O 2 -loaded CuTz-1@F127 (denoted as CuTz-1-O 2 @F127) metal–organic framework (MOF) therapeutic platform is presented for enhanced PDT by simultaneously overcoming intracellular hypoxia and reducing GSH levels in the tumor. The Cu(I)-based MOF is capable of a Fenton-like reaction to generate • OH and O 2 in the presence of H 2 O 2 under NIR irradiation. Meanwhile, the CuTz-1-O 2 @F127 nanoparticles (NPs) can release adsorbed O 2 , which further alleviates intracellular hypoxia. In addition, the Cu I in CuTz-1@F127 can react with intracellular GSH to reduce the excess GSH. In this way, the efficiency of PDT is greatly enhanced. After tail intravenous injection, the NPs show high antitumor efficacy through a synergistic effect under 808 nm laser irradiation. More importantly, the NPs are biodegradable. In vivo biodistribution and excretion experiments demonstrate that a total of nearly 90% of the NPs can be excreted via feces and urine within 30 d, which indicates significant prospects in the clinical treatment of cancers.
