Highly-sensitive SERS detection of tetracycline: Sub-enhancement brought by light scattering of nano-diamond

Jingxuan Pei,Zhenfei Tian,Xiang Yu,Shuting Zhang,Shiqing Ma,Yibo Sun,Rabah Boukherroub

Antibiotic residues in wastewater cause a serious threat to aquatic environment and human safety. Therefore, the development of robust strategies for detecting antibiotic residues becomes critical. In this work, a surface-enhanced Raman scattering (SERS) substrate, consisting of gold nanoparticles/nano diamond/carbon nitride (Au/ND/C 3 N 4 ) hybrid, which features enhanced light utilization and self-cleaning properties, is developed for sensitive sensing of tetracycline. The fabricated Au/ND/C 3 N 4 substrate effectively captures target molecules and enhances Raman activity, owing to abundant functional groups and light scattering effect of ND. Moreover, the heterostructure formed by Au NPs, ND and C 3 N 4 endows this SERS substrate with superior self-cleaning capability, enabling its reusability. The Au/ND/C 3 N 4 substrate exhibits high sensitivity (10 -14 M), desirable reproducibility (RSD = 13.29 %, n = 238) and favourable stability (84.8 % of initial intensity after 30 days) for crystal violet probe. Also, no significant loss of SERS or catalytic activity is observed during cyclic SERS detection. Likewise, the detection of tetracycline at nanomolar level is achieved with good recyclability. This work proposes a distinctive strategy for enhancing SERS activity, and the developed self-cleaning Au/ND/C 3 N 4 substrate represents a potential candidate for highly sensitive detection of antibiotic residues in wastewater.

Ultrasensitive Sandwich-Type SERS-Biosensor-Based Dual Plasmonic Superstructure for Detection of Tacrolimus in Patients

Xiaoyue Zheng,Jiazhou Ye,Weiwei Chen,Xiaoyuan Wang,Jianhua Li,Fengmei Su,Caiping Ding,Youju Huang

Tacrolimus (FK506) is widely used in the prevention of organ transplant rejection and the treatment of autoimmune diseases, but it is difficult to detect within the low and narrow concentration range in practical clinical fields. A magnetic plasmonic superstructure-targets-plasmonic superstructure-based sandwich-type SERS biosensor is presented here to ultrasensitively detect FK506 in the blood of organ transplant patients. The spiky Fe3O4@SiO2@Ag flower magnetic superstructure and hollow Ag@Au superstructure enhanced the SERS signals by providing rich sharp tips, cavities, and abundant hot spot gaps. And the magnetic feature makes it easy to concentrate and separate the biological target. Using the designed sandwich-type SERS biosensor, FK506 could be detected within a range of 0.5–20 ng/mL with a detection limit of 0.33 ng/mL. All results indicated that the sandwich-type SERS biosensor has good stability, sensitivity, and anti-interference properties. It is noteworthy that this allowed us to successfully analyze FK506 in the blood of transplant patients, which is in strong agreement with the clinical results. Consequently, the attractive sandwich-type SERS biosensor can be used for the detection of FK506 in real samples, which is promising for clinical diagnosis.

SERS ratiometric strategy leveraging carbon allocation trade-off enables Tremella fuciformis metabolic phenotyping and traceability

Meng Wang,Chenggang Zhang,Miao Yu,Ye Sun

Carbon allocation trade-off enables SERS metabolic phenotyping of fungi. • Polysaccharides/carbohydrates ratio serves as metabolic SERS signatures. • A superior accuracy of 99.2% and an AUC value of 1.0 are achieved. • Portable device enables 15-min on-site fungal phenotyping traceability assay. • It upgrades SERS from chemical fingerprinting to bio-metabolic phenotyping. Background Accurate metabolic phenotyping and geographical traceability of gelatinous fungi are critical for uncovering fungal physiological adaptation mechanisms and advancing edible fungus quality supervision. However, high phenotypic similarity and subtle metabolic differences in homologous fungal samples pose formidable challenges to conventional physicochemical assays. Especially, relying on chemical fingerprinting without biological phenotypic implications, conventional surface-enhanced Raman scattering (SERS) analysis is unable to discern delicate metabolic discrepancies in fungal samples. Results Herein, we propose a metabolic characteristic peak ratio-based SERS strategy using Tremella fuciformis ( T. fuciformis ) as the model. By extracting intrinsic metabolic signatures represented by the carbon allocation trade-off between stress-resistant functional polysaccharides and storage-type carbohydrates, it delivers metabolic specificity and detection stability/reproducibility that are not possible with traditional fingerprint-based SERS fungal analysis. Integrated with machine learning, this method accurately differentiates six T. fuciformis samples, including specimens with distinct metabolic phenotypes as well as geographically differentiated samples with highly similar phenotypic features, achieving a superior accuracy of 99.2% and an AUC of 1.0. Combined with a custom-built portable Raman device we constructed, the entire detection procedure can be completed within 15 min, enabling efficient on-site analysis. Significance and novelty This study establishes a bio-signature-oriented SERS paradigm that upgrades chemical fingerprint identification to fungal metabolic phenotyping, providing a versatile technical platform for exploring fungal metabolic variation and phenotypic differentiation. Cefalexin fingerprinting in water using graphene oxide/gold nanoparticle-modified silica photonic microsphere SERS substrates

Ziyi Wang,Xiu Wang,Ziqiang Li,Yikun Li,Muhammad Shan,Qianjin Li,Jianlin Li

A 3D SPM@GO@AuNPs substrate was built for cefalexin SERS detection. • GO enriches cefalexin and anchors AuNPs for hotspot generation. • Cefalexin was quantified from its 1189 cm-1 Raman band in 10 min. • The assay gave 0.01–10 μg/mL linearity and a 0.41 μg/mL LOD. • Cephalosporin analogues were distinguished by SERS fingerprints. Cefalexin residues in aquatic environments require detection methods that are fast, label-free, and spectroscopically reliable. Surface-enhanced Raman spectroscopy (SERS) can provide molecular fingerprints, but its analytical performance depends on analyte adsorption and the spatial consistency of the enhanced response. Here, a three-dimensional silica photonic microsphere (SPM) substrate was modified with graphene oxide (GO) and gold nanoparticles (AuNPs) to form an SPM@GO@AuNPs substrate for cefalexin fingerprinting in water. GO served as an AuNP-anchoring layer and as an adsorption interface that enriched cefalexin near SERS-active AuNP junctions, whereas the monodisperse SPM provided a curved, readily handled three-dimensional support. Cefalexin bands at 768, 833, 996, 1022, and 1189 cm −1 were resolved, with the 1189 cm −1 band used for quantification. Under optimized conditions, the method exhibited a linear range of 0.01–10 μg/mL, a limit of detection of 0.41 μg/mL, and a 10 min binding step. Cefalexin was distinguished from four cephalosporin analogues using the full spectral fingerprint rather than a single band. Spiked tap-water analysis gave mean recoveries of 86.99–96.83%, supporting the feasibility of direct aqueous screening. The method is best suited to rapid fingerprint-based screening of cefalexin in relatively simple water matrices. Download: Download high-res image (144KB) Download: Download full-size image

Metal organic framework enabled “split-type” SERS immunoassay platform for highly sensitive and reproducible biosensing

Ling Huang,Sailan Wang,Li Wang,Hongwen Wu,Hongliang Tan,Fugang Xu

MOF-enabled split-type SERS platform decouples probe carrier and substrate. • ZIF-90 enables simple, high-density loading of non-thiolated Raman reporters. • Reagent-free antibody conjugation to ZIF-90 via inherent Schiff-base chemistry. • EDTA-triggered reporter release enables detection on uniform external substrate. • 100-fold lower LOD and > 50% improved reproducibility vs. conventional assays. Surface-enhanced Raman scattering (SERS) holds immense potential for ultrasensitive immunoassays, yet conventional gold nanoparticles (AuNPs) based probe and substrate “two-in-one” designs face critical limitations including low reporter loading capacity, restricted reporter selection, poor signal reproducibility and inefficient antibody conjugation. To overcome these challenges, we propose a metal organic framework (MOF) enabled “split-type” SERS immunosensor, where a MOF material ZIF-90 acts as probe carrier and a uniform silver coated filter paper (Ag@FP) composite serves as external SERS substrate. This MOF based strategy offers four distinct advantages: (i) The abundant porosity of ZIF-90 dramatically increases Raman reporter loading compared to gold nanoparticles (AuNPs) surfaces, enabling significant signal amplification. (ii) Encapsulation of reporters into ZIF frameworks is independent of specific functional groups, thereby highly active, non-thiolated Raman dyes can be used; (iii) The ethylenediaminetetraacetic acid (EDTA)-triggered release of reporters from ZIF-90, coupled with signal acquisition on a uniform external substrate, yields superior signal reproducibility. (iv) Linking antibody to ZIF-90 is achieved via Schiff-base chemistry between their inherent functional groups, eliminating the need for costly coupling reagents. For model biomarker RIgG, this “split-type” SERS immunosensor achieved a detection limit of 2 × 10 −11 g/mL, which is two orders of magnitude lower than that of conventional AuNP-based methods (3 ×10 −9 g/mL), while the relative standard deviation (RSD) was reduced by more than 50%. Excellent recovery and reproducibility in spiked and real serum samples further validate its feasibility in complex biological matrices. This versatile MOF-enabled “split-type” platform provides a compelling new paradigm for sensitive and reproducible SERS biosensing. Template-confined assembly of Ag nanocubes: An approach to fabricate SERS substrate with good performance

Jiaqi Liu,Chunning Chen,Jiaxin Lu,Yalei Wang,Jingtong Zhai,Hongkun Zhao,Nan Lu

Surface-enhanced Raman scattering (SERS) is an important analytical technique. Its detection sensitivity and reproducibility depend on the density and distribution of SERS hotspots. Self-assembly is an efficient method to produce of SERS substrates due to its easy accessibility. However, the assembled defects can hardly be avoided on large area, which could lower the density and uniformity of the hotspots, leading to poor SERS performance. Herein, we report a method to reduce the defects by taking a patterned substrate as template to confine the assembly of Ag nanocubes. The template was prepared based on the combination of photo lithography and self-assembly. Confined by the template, the Ag nanocubes were assembled closely in each dots of the pattern. The limit of detection (LOD) is down to 3.42 × 10 −17  M and the enhanced factor (EF) is up to 3.44 × 10 10 on the prepared substrate for detecting rhodamine 6G (R6G). In addition, the relative standard deviation (RSD) of the different substrates is 8.75 %. The assembled Ag nanocubes exhibits high sensitivity and reproducibility as SERS substrate, which are contributed by the formation of high-density and uniform hotspots. The prepared substrate can be used for detecting trace amounts of melamine in milk with LOD of 2.06 × 10 −7  M and RSD of 6.91 %, so the substrate is applicable for analyzing various analytes.

Serum SERS spectroscopy combined with classification algorithm in the non-destructive identification of cervical cancer

Gao Ningning,Wang Qing,Tang Jun,Lv Xiaoyi,Li Hongmei,Yue Xiaxia,Zhong Furu,Fu Jihong,Wang Tao

Gold nanoparticles/785 porous silicon photonic crystals (Au NPs/785 PSi PhCs) were used as substrates in combination with a low-concentration serum surface-enhanced Raman spectroscopy (SERS) detection scheme to obtain spectral signals from healthy individuals and cervical cancer patients. The best principal component scores were selected by principal component analysis (PCA) combined with linear discriminant analysis (LDA) and support vector machine (SVM) to analyze the spectral differences distinguishing healthy and cervical cancer patients. The accuracy of the two models was 97.9% and 96.9%, respectively. SERS technique based on Au NPs/785 PSi PhCs has great potential to improve the screening of cervical cancer.

Hierarchical metal-dielectric-metal nanostructures as SERS sensors for sensitive detection of polycyclic aromatic hydrocarbons

Shiqiang Wang,Huiyun Jiang,Junjie Feng,Yan Jin,Fei An,Liang Zhu,Anshan Xiao

Due to the wide variety, inaccessible self-degradation, high mutagenicity and carcinogenicity of polycyclic aromatic hydrocarbons (PAHs), it still remains challenging to efficiently identify them, especially in real environment. Herein, we develop a bottom-up self-assembly strategy to construct a sandwich-like hierarchical metal-dielectric-metal system composed of mercaptan-modified Au nanospheres-graphene oxide (GO)-Ag nanowires as surface-enhanced Raman spectroscopy (SERS) sensors for sensitive detection of PAHs. The synergistic integration of metallic layers and the organic mercaptan and GO layer not only provides localized enhanced electromagnetic field region which is induced by the nanogap between the top Au nanospheres and the bottom Ag nanowires, but also effectively traps PAHs in the middle layer through hydrophobic interaction of mercaptan and π-π stacking interaction of GO. By rationally optimizing the SERS substrates, the lowest detection concentrations of pyrene, anthracene, phenanthrene and benzo[a]pyrene could be reached, respectively, at 10 −8  M, 10 −8  M, 10 −8  M and 10 −7  M. The hierarchical system shows excellent uniformity, reproducibility and is able to identify various types of PAHs in multi-component mixtures as well as in real water environment. This strategy can provide a general platform to identify hazardous pollutants that are hydrophobic or contain benzene rings, such as pesticides, dyes and polychlorinated biphenyls.

Fabrication of a MoS2/g-C3N4@Au heterojunction-based recyclable SERS substrate for detecting fungicides

Wei Qingyi,Dong Qirong,Pu Hongbin,Sun Da-Wen

Aquaculture fungicide causes threats to the human community and their sensitive detection are thus essential for food safety. Herein, MoS 2 /g-C 3 N 4 @Au heterojunction-based recyclable SERS substrate was developed for fungicide detection. The optimal substrate exhibited good SERS performance with a detection limit as low as 9.908 × 10 −5 mg L −1 for malachite green (MG). Furthermore, an MCN@Au-based photodegradation of crystal violet (CV) under simulated light irradiation was realized, resulting in a degradation rate of 95.6%. With the dual function of the as-synthesized materials, good reusability of the substrates for at least six “detection-degradation” cycles was achieved. In addition, the MG in prawn extract could be sensitively determined with a satisfactory recovery from 91.60 to 124.00%. This work paves an avenue for the fabrication of heterojunction-based recyclable SERS substrates and provides a novel weapon for food safety protection.

Designing of a functional paper-tip substrate for sensitive surface-enhanced Raman spectroscopy (SERS) detection

Xiaoxiao Qi,Yongqiang Cheng,Ranran Xu,Xiaotong Li,Ziwei Zhang,Longyu Chen,Yifan Shao,Zhenhui Gao,Meijia Zhu

A simple and flexible fabrication method of paper SERS substrate was developed by nanoparticles (NPs) droplet self-assembly at the paper tip with a temperature gradient (PTTG). We turned the drawback of the coffee ring effect into an effective way of preparing paper SERS substrate. When the NPs droplets were continuously dripped onto the PTTG, NPs were densely and uniformly distributed at the paper-tip front based on the combination of gravity and the coffee ring effect, which could achieve 91.2-fold improvement of SERS performance compared to a flat filter paper. Meanwhile, the analytes could also be enriched at the paper-tip front, which could achieve 9.3-fold signal enhancement compared to the paper-tip tail. Thus, the PTTG realized an excellent signal amplification for SERS detection. The paper-tip SERS substrate combined with a portable Raman spectrometer yielded an excellent analytical enhancement factor of 1.15 × 10 5 with the detection limit of 10 nM Rhodamine 6G (R6G). The whole fabrication procedure was completed within 2 h, and the paper-tip substrate showed a satisfactory substrate-to-substrate reproducibility with a relative standard deviation (RSD) of 5.13% (n = 10). It was successfully applied for quantitatively detecting real samples of oxytetracycline and malachite green with recoveries of 83.84–105.25% (n = 3). Meanwhile, we further evaluated the SERS performance of the PTTG using a laboratory-based Raman spectrometer, and it could realize the detection as low as 10 pM R6G. The proposed paper-tip substrate would offer a promising potential application for the on-site SERS analysis of food safety and environmental health .

Exploration of efficient SERS features extraction algorithm for rapid detection of thiabendazole residues in apples

Xiaodong Li,Yanyan Zhang,Muhammad Awais,Syed Muhammad Zaigham Abbas Naqvi,Linze Li,Hongjun Chen,Jiandong Hu

Long-term exposure to thiabendazole (TBZ) pesticide residues in fruits can have harmful effects on human health. Therefore, this paper proposes a highly sensitive and time-efficient method for the exploration of efficient SERS feature extraction algorithms to establish an optimal quantitative model for the rapid detection of TBZ residues in apples. Specifically, shape-homogeneous Au@Ag nanoparticles (NPs) were developed as label-free substrates, and the optimal detection time of obtaining SERS signals generated by the binding of TBZ to the substrate sample was determined to be 6 min. Recursive Feature Elimination (RFE) and Competitive Adaptive Reweighted Sampling (CARS) were employed for feature extraction algorithms to optimize characteristic peaks and compare them to full spectra variables and extract effective variables on linear (Ridge, PLS) and nonlinear (SVM, RF) models, respectively. Results showed that the RFE-RF (Recursive Feature Elimination-Random Forest) model was more powerful in variable selection and achieved the highest predictive result of TBZ (R P 2  = 0.992, RMSEP = 0.012 μg/mL) and the computed LOD (Limitation of Detection) was 0.1 μg/mL based on PCA (Principal Component Analysis). Comparison with ELISA verified that the designed method is feasible and potentially applicable to guarantee the safety of pesticide residues in fruits.

A SERS-based immune-nanoprobe for ultrasensitive detection of glycinin via a lateral flow assay

Pengbo Huang,Shuo Yang,Zixin Chen,Achen Shang,Deguo Liu,Jun Xi

Glycinin is one of the most nutritious ingredients in soybean, but it is also an allergen that can cause allergic reactions in humans and animals, and even endanger life in severe cases. Therefore, it is imperative to develop a rapid and ultrasensitive detection method for glycinin. In order to achieve this goal, this experiment combined surface-enhanced Raman spectroscopy (SERS) technology with an economical, simple, fast and easy-to-carry immunochromatographic test strip, and successfully constructed an efficient sandwich immunochromatographic test strip. The immunoprobe of the test strip was covalently coupled by gold nanostars (AuNSs), Raman molecule 4-aminothiophenol (4-PATP) and rabbit polyclonal antibody. In this experiment, a gold nanostar immunochromatographic test strip for detecting glycinin was established. The detection limit of the test strip was 0.23 ng / mL, the recovery rate was 91.5–96.6 %, and the coefficient of variation was 1.61–6.15 %. In addition, the test strip had no cross reaction with whey protein, wheat protein, peanut protein, sesame protein and β-conglycinin, indicating that the detection method had good specificity and the ability to avoid false positive results. Hence, this experiment successfully prepared a rapid and sensitive test strip for glycinin.
