Synergistic anti-obesity therapy: ursolic acid-loaded mesoporous polydopamine nanoparticles (MPDA@UA) in an injectable OHA-CMCS hydrogel with a mild photothermal effect for adipocyte reprogramming

Li Tingting,Zhao Yueming,Lin Mian,Hu Jiajun,Duan Kairui,Zhu Hu,Zi Taotao,Lee Bae Hoon

Obesity and its metabolic complications urgently require safe, effective, and minimally invasive interventions. Here, we engineered an injectable, self-healing hydrogel system (OCMU hydrogel) based on oxidized hyaluronic acid (OHA) and carboxymethyl chitosan (CMCS) for the localized delivery of plant-based ursolic acid (UA) loaded in mesoporous polydopamine (MPDA) nanoparticles. This system synergistically induces white adipose browning through combined photothermal therapy (PTT) and pharmacological action, effectively counteracting obesity. The OCMU hydrogel exhibited excellent self-healing properties, high UA loading capacity (81.04%), efficient photothermal conversion, and favorable biocompatibility. Mechanistically, the system promotes lipolysis, mitochondrial biogenesis, and adipose browning via activation of the AMPK/SIRT1/PGC-1α pathway, with a 439% increase in UCP1 expression in vitro. In obese mice, OCMU combined with PTT reduced body weight by 10.94% and inguinal fat by 56.32%, while significantly improving metabolic profiles and alleviating inflammation. Notably, the combination of mild PTT and naturally derived UA achieved superior anti-obesity efficacy compared to monotherapies, highlighting a favorable safety and efficacy profile. This work establishes a promising platform in which UA and mild PTT act in concert to reprogram adipocyte metabolism, offering a safe and synergistic approach to combat obesity. Graphical

An inherent T cell-activating mRNA delivery carrier for in vivo CAR T generation

Cao Qiannan,Yao Yingli,Zheng Wenming,Liu Hongqian,Jiang Mingxia,Xie Dayang,Zhang Siting,Su Pijun,Yuan Huilin,Chen Xiaoyuan,Fang Huapan,Tian Huayu

The clinical success of chimeric antigen receptor (CAR) T cell therapy requires scalable, non-invasive strategies for in vivo T cell engineering. Although mRNA delivery offers a promising alternative, lipid-nanoparticle-based carriers show limited efficiency for in vivo T cell transfection and typically require antibody conjugation. Here we report an inherent T cell-activating polymer–lipid nanoparticle that enables ligand-free, efficient mRNA transfection and activation of T cells in vivo. This mRNA delivery vehicle, composed of p -toluenesulfonyl arginine (RT)-modified oligoethylenimine-based lipid nanoparticles (ERTLNPs), preferentially mediated mRNA transfection in the spleen following systemic administration. Without exogenous stimulation, ERTLNPs intrinsically activated T cells, triggering robust mRNA expression and proliferation. Mechanistically, ERTLNPs engaged the PI3K/AKT/mTOR signalling axis to reprogram T cell metabolism, promoting expansion and restraining exhaustion. The systemic delivery of mRNA encoding fibroblast activation protein CAR via ERTLNPs contributed to the in situ generation of functional CAR T cells, which efficiently eliminated pathological fibroblasts in models of cancer and fibrosis, with minimal off-target effects. This ligand-free, metabolically reprogramming mRNA delivery system provides a clinically translatable approach for in vivo CAR T cell generation.

Dihydrotanshinone I as a novel signal transducer and activator of transcription 3 inhibitor for glioblastoma treatment

Cheng Kai-wen,Zhou Shao-jun,Zeng Jie,Zhang Yang-rui,Jin Bing-qian,Wang Ping,He Ling-yan,Qi Xu-chen,Liu Xu-dong,Yu Lu-shan,Zhang Yan-xing

Background Among tumors affecting the central nervous system (CNS), glioblastoma multiforme (GBM) is the most aggressive and lethal form. Given the complex mechanisms of the CNS and the blood-brain barrier (BBB), effective therapies for GBM remain limited. To identify potential therapeutic candidates for glioma, we developed a small-molecule library to screen for compounds with potent anti-proliferative effects and BBB permeability. The mechanisms underlying their anti-glioma activity were further elucidated to provide insights into new treatment strategies. Methods A library of small molecules was screened to identify agents that significantly inhibit glioma cells activity. The effects of the lead compound (Dihydrotanshinone I, DHT) on glioma progression were evaluated through a series of experimental approaches, including cell counting kit-8 assays, cell cycle analysis, apoptosis detection, wound healing, transwell migration, reactive oxygen measurement, subcutaneous xenograft mouse models, and intracranial orthotopic tumor models. To elucidate the underlying mechanisms, network pharmacology and transcriptomic analyses were employed. The mechanism of DHT in glioma pathogenesis was further validated using bioinformatics analyses and clinical glioma tissue samples. Results Based on pharmacokinetic evaluation, DHT was found to traverse the BBB, indicating its capacity to reach the cerebral parench. In vitro and in vivo experiments demonstrated that DHT significantly suppresses glioma growth and progression. Mechanistic analyses using network pharmacology and RNA sequencing revealed that DHT induces glioma cell apoptosis by inhibiting the Janus kinase-signal transducer and activator of transcription 3 (STAT3) signaling pathway, increasing intracellular reactive oxygen species levels and triggering intrinsic apoptotic cascades. Furthermore, bioinformatic analyses of clinical cohorts coupled with validation in patient-derived glioma specimens confirmed that elevated STAT3 expression was correlated with unfavorable prognosis and was significantly increased in patients with high-grade glioma. Conclusions This study demonstrates that, in addition to its role as a key contributor to glioma malignancy and a prognostic marker, STAT3 is also a target of DHT, highlighting its potential as a promising therapeutic candidate for treating GBM. Conductive Network-Embedded Outer Anodic Layer Enabling Coaxial Electrochromic Fibers with Highly Homogenized Electric Field

Mu Wenhao,Zhang Zaiyuan,Fan Qingchao,Zhai Kaiyue,Wu Xilu,Zhang Qinghong,Li Yaogang,Hou Chengyi,Li Kerui,Wang Hongzhi

Electrochromic (EC) fibers, with their excellent embeddability and rapid reversible optical modulation in electronic textiles, exhibit promising applications in wearable displays and smart textile integration. However, due to nonuniform electric field distribution and limited electrochromic active regions, EC fibers exhibit issues such as insufficient optical contrast and poor cycling stability in practical applications. Herein, we develop a coaxial EC fiber featuring a conductive network-embedded outer anode and a dual-layer synergistic coloration mechanism (outer poly(3,4ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) layer and inner polyaniline (PANI) layer). Benefiting from the optimized structure, large-scale fabrication of coaxial EC fiber achieves enhanced optical modulation (32.6%), fast response time (8.15 s/6.32 s) and long cycling stability (1000 cycles, 90%). EC textiles woven therefrom, upon integration with sensing modules, offer real-time physiological sensing with dynamic color feedback, underscoring their potential for advanced smart wearable displays. Integrating liquid-liquid extraction and dispersive solid-phase extraction cleanup for high-throughput profiling of 11 neonicotinoids and 6 metabolites in human urine by HPLC-Q-Orbitrap HRMS

Liu Feng,Yu Decong,He Kun,Song Yanan,Li Yichao,Gao Haoxuan,Wu Bing,Li Xiaoyu,Zhao Ji,Xu Fei,Yang Huifang

The widespread use of neonicotinoids has raised growing concerns about their potential health risks. In this study, we developed a robust and high-throughput analytical method based on liquid-liquid extraction (LLE) and dispersive solid-phase extraction (dSPE), followed by high-performance liquid chromatography-quadrupole-Orbitrap high-resolution mass spectrometry (HPLC-Q-Orbitrap HRMS) for the simultaneous determination of 11 neonicotinoids and their 6 metabolites in human urine. After pretreatment of LLE and dSPE, qualitative and confirmatory analysis was achieved using tMS 2 acquisition of HPLC-Q-Orbitrap HRMS, and quantitative analysis was performed using isotope-labeled internal standards and matrix-matched calibration curves. The method was validated with good linearity ( R 2  > 0.9975), recoveries of 75.8%~118.9% (RSDs < 15%), and detection limits of 0.001~0.06 μg/L. The established method was successfully applied to urine samples of community-dwelling elderly population individuals ( n  = 150) and to a suspected case of acute acetamiprid poisoning in Ningxia, China. The results showed that thirteen neonicotinoids and metabolites were detected in the elderly cohort, and the poisoning incident was confirmed to be caused by acetamiprid and its metabolite N-desmethyl-acetamiprid. Meanwhile, Spearman correlation analysis indicated significant positive correlations between parent compounds and their metabolites for all analytes except acetamiprid, and a strong correlation between clothianidin and thiamethoxam, suggesting shared exposure sources or common metabolic pathways. The established method is sensitive, accurate, and high-throughput, and has proven reliable for human biomonitoring and health risk assessment of neonicotinoid exposure. Graphical

Synthesis of Carbon Quantum Dots Derived from Coral Algae Polysaccharide and Their Application for Strawberry Preservation in Gelatin Films

Wang Libo,Hu Xinli,Fu Xuan,Li Jiayu,Li Jinwei,Wei Yanhui,Cheng Kun

In the present study, N and S co-doped carbon quantum dots (CP-CQDs) were synthesized using coral algal polysaccharide (CAP) and o-phenylenediamine (OPD) as precursors. CP-CQDs with a mean size of 4.14 ± 0.70 nm emitted yellow-green fluorescence at a wavelength of 530 nm when excited at 427 nm. CP-CQDs exhibited excellent fluorescence stability after 8 h of UV irradiation, across a pH range of 5.0–13.0, and in 1.0 mol/L NaCl solution, respectively. GEL/CP-CQDs composite films were prepared by incorporating CP-CQDs into a gelatin (GEL) matrix. The mechanical properties, barrier properties, and antibacterial activities of the films were significantly correlated with the concentration of CP-CQDs. Hemolysis assays and overall migration tests confirmed that the GEL/CP-CQDs films exhibited good biocompatibility. Moreover, the GEL/CP-CQDs3% film effectively extended the shelf life of strawberries, reducing the decay rate to 20.32 ± 1.62% at the tenth day of storage. This research offered valuable perspectives for promoting the high-value utilization of marine algal resources and their application in active packaging. Graphical

Solvent-bridged electrolytes for high-energy Li-ion batteries under extreme conditions

Chen Yawei,Huang Fanyang,Zhang Qiu,Zheng Lei,Li Ai-Min,Dong Dejian,Xia Kangxuan,Lee Taeyong,Zhang Weiran,Li Zheng,Zhang Xin,Zhao Chang-Xin,Chen Fu,Rao Jiancun,Gaskell Karen,Hu Enyuan,Wang Chunsheng

High-capacity anodes are desirable for high-energy lithium-ion batteries but suffer from limited cycle life due to large volume changes during cycling. The LiF-based solid–electrolyte interphase (SEI) has led to prolonged cycling stability. However, current LiF-forming electrolyte designs require anions to enter the Li + solvation sheath, which inherently reduces the electrolyte’s ionic conductivity, limiting both fast-charging and low-temperature performance. Here we develop solvent-bridged electrolytes composed of LiPF 6 dissolved in a cosolvent system comprising a bridging cyclic ether that solvates both PF 6 − and Li + to form a LiF-rich SEI, and a structural linear ether that governs the electrolyte liquid range. By mitigating direct Li + –anion interactions, the electrolytes maintain high ionic conductivity, enabling stable cycling of micrometre-sized silicon anodes under extreme conditions involving high rates (>4 C), low temperatures (down to −55 °C) and Li plating. Solvent-bridged electrolytes address the intrinsic trade-offs between LiF-rich SEI formation and electrolyte ionic conductivity, offering a promising approach for high-capacity anodes operating under demanding conditions.

In situ formation of Ca–Ag hybrid nanoparticles for multifunctional reconstruction of the resin–dentin interface

Zhang Zhenzhen,Chen Yadong,Zhong Qi,Xu Jiajia,Cheng Yajie,Shu Chang,Luo Qiaojie

Objectives The hybrid layer formed during dentin bonding remains susceptible to degradation. This study aimed to develop a CaCl₂/AgF treatment strategy for in situ deposition of Ca–Ag hybrid nanoparticles within the demineralized dentin matrix (DDM) and to evaluate its effects on interface-confined water regulation, antibiofilm activity, mineral deposition, and ultimately bonding durability. Materials and methods DDM was sequentially treated with CaCl₂ and AgF solutions. Interface-confined water regulation was assessed by surface potential, water uptake, and thermogravimetric analysis (TGA). Antibiofilm activity against Streptococcus mutans ( S. mutans ) and saliva-derived microcosm biofilms was evaluated by confocal laser scanning microscopy (CLSM) and scanning electron microscopy (SEM). Mineral deposition was examined by transmission electron microscopy (TEM) and selected area electron diffraction (SAED). Adhesive infiltration, nanoleakage, and microtensile bond strength (1 day and 12 months) were tested. Results Ca–Ag deposition was associated with reduced interfacial hydration, as indicated by reduced surface potential, decreased water uptake, and lower mass loss below 200 °C in TGA. It inhibited biofilm formation and disrupted established biofilms. Enhanced mineral deposition occurred within 1 h in the Ca–Ag group, with SAED patterns showing crystalline mineral phases, whereas controls remained unmineralized. Enhanced adhesive infiltration, reduced nanoleakage, and improved 12-month bond strength were observed. Conclusions Sequential CaCl₂/AgF treatment induced the in situ deposition of Ca–Ag hybrid nanoparticles within the DDM, which was associated with reduced interfacial hydration, antibiofilm activity, mineral deposition, and improved bonding performance in vitro. Clinical Significance This in vitro study suggests that CaCl₂/AgF treatment may represent a potential strategy for improving resin–dentin interface stability. Further investigations are required to evaluate its clinical applicability.

Multidimensional fluorescent array-sensing of biogenic amines based on porphyrin-functionalized carbon dots

Tang Xiaolu,Li Quan,Yang Yue,Wu Meilin,Liu Qingqing,Mo Zhihong

Protein-rich foods release biogenic amines (BAs) during prolonged storage, and these compounds serve as key biomarkers of food spoilage. Current separation-based methods for detecting BAs in food require sophisticated instrumentation and laborious sample pretreatment procedures. The array-sensing strategy, which mimics biological olfaction, provides a simple and rapid approach for the simultaneous detection of multiple BAs without the need for separation. Here, we constructed a multidimensional fluorescent sensor array using porphyrin-functionalized carbon dots (P-CDs) and their composites with metal ions (M-P-CDs), thereby incorporating the excitation wavelength as an additional signal dimension. This design reduces the number of physical sensors required while maintaining high discrimination capability, thereby overcoming a key limitation of conventional sensor arrays that depend solely on increasing the number of sensors. With the support of chemometric and machine learning methods, the sensor array successfully discriminated among ten BAs. We achieved quantitative detection of putrescine, histamine, and tyramine, determining their limits of detection (LODs) to be 2.37, 2.96, and 1.55 mg/L, respectively. The sensor array exhibited excellent stability (i.e., no significant fluorescence change over 30 days) and high selectivity against common interfering substances. Validation experiments on milk samples demonstrated the array’s ability to reliably discriminate specimens at different spoilage stages. This work establishes a novel multidimensional array-sensing strategy for detecting BAs, an approach that holds great potential for food safety applications. Graphical

In Situ Transformation of Natural Bark into Additive-Free, Self-Adhesive Composites

Ren Qian,Li Chenxi,Liu Yaqun,Song Guanghui,Zhou Yutong,Du Chunhao,Liu Zifeng,Rao Jun,Kang Yuetong,Lü Baozhong,Chen Yupeng,Peng Feng,Chen Gegu

Lignocellulosic plastic composites are widely used in many industries for their excellent mechanical strength and durability owing to the synergistic effect of lignocelluloses and polymers. However, the polymer content severely limits their recyclability and environmental sustainability. Here, a self-adhesive lignocellulosic composite is designed directly from forestry residue Eucommia ulmoides bark (EUB), which is rich in Eucommia ulmoides gum (EUG) and lignin. The fabrication consists of a two-step process of in situ epoxidation and self-adhesive technique. In situ epoxidation transformed the intrinsic EUG into an excellent viscoelastic epoxidized EUG (EEUG). The self-adhesive technique relies on hydrogen bonding reinforcement promoted by pressure-assisted densification and thermal-assisted flow of EEUG, and further enhanced by epoxy ring-opening. This process converts the raw materials into a fully natural self-adhesive composite without any additional polymers. The demonstrated composite exhibits enhanced mechanical strength, notable flexibility, high hydrophobicity (contact angle at 104.55°), remarkable water resistance, improved thermal stability, and biodegradability (52.96% mass loss in 5 months). In addition, the life cycle assessment reveals a carbon emission factor of 3.28 kg CO 2 eq/kg, which can be reduced to 1.56 kg CO 2 eq/kg under renewable-energy production. This work demonstrates a practical route for transforming forestry residue into high-value lignocellulosic materials and provides an environmentally benign alternative to conventional plastic-bonded systems. Harnessing CSC recruitment: photo-triggered “self-exploding” macrophages for targeted metastasis inhibition

Wang Teng,Wang Yu,Wang Bingjie,Su Yanguo,Jiang Tianze,Xu Wenhua,Zhao Xia,Tao Jiaojiao,Shi Chenshuai,Xu Xiang,Xu Jinlong

Cancer metastasis is a serious threat to human health. Cancer stem cells (CSC) are closely related to tumor metastasis, but it is difficult to achieve accurate drug delivery and effective inhibition of CSC due to the complex tumor microenvironment. Herein, we report a living cell delivery system, C-H@M2, engineered by loading M2 macrophages with carrier-free Ce6/Honokiol self-assembled nanoparticles. This system is designed to harness the tumor’s intrinsic recruitment mechanism for targeted anti-metastasis therapy. Acting as a “Trojan Horse”, C-H@M2 exploits the active recruitment of M2 macrophages by CSC to achieve precise tumor homing. Upon laser irradiation, the macrophages undergo photo-triggered immunogenic lysis (“self-explosion”), leading to the burst release of therapeutic payloads and the induction of robust immunogenic cell death (ICD). HK as an inhibitor of Forkhead box M1 (FOXM1) inhibits CSC stemness and epithelial mesenchymal transition (EMT) in tumor tissues. In vivo experimental results demonstrate that C-H@M2 significantly inhibits tumor and lung metastasis through a triple-modal strategy: Self-explosion enhanced ICD effect, stemness and EMT inhibition, CSC killing by stemness inhibition and PDT. Our study provides a novel strategy for CSC targeted therapy and cancer metastasis inhibition. Graphical

Dual-mode electrochemiluminescence and colorimetric detection of malathion based on a bifunctional rGO@ZnO/Zn-Fe3O4 nanozyme

Wang Wenzhuo,Lu Juan,Jiang Hanyue,Han Pengfei,Ma Guangping,Li Ruixin,Mao Huiyuan,Ma Tengying,Wang Jing,Chen Ruizhan,Tian Li

A dual-mode sensing platform has been developed for ultrasensitive malathion (MAT) detection based on bifunctional rGO@ZnO/Zn-Fe 3 O 4 composites (reduced graphene oxide-supported ZnO nanorods and Zn-doped Fe 3 O 4 microspheres). The rGO@ZnO/Zn-Fe 3 O 4 nanozyme is fabricated via a one-pot hydrothermal strategy and integrates favorable electrochemiluminescence (ECL) emission and intrinsic peroxidase-mimicking activity to realize dual-signal readout. In the K 2 S 2 O 8 ECL system, Au NPs@UiO-66 serves as a coreactant booster to further magnify the ECL intensity of the composite. Meanwhile, the nanozyme catalyzes the H 2 O 2 -mediated oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to produce a colorimetric response. MAT suppresses both ECL and colorimetric responses by occupying Fe 2+ /Fe 3+ catalytic sites and scavenging of reactive radicals. The sensor achieves limits of detection as low as 2.8 × 10 –13  mol/L under ECL mode and 1.32 × 10 –6  mol/L under colorimetric mode. In practical testing of MAT-spiked vegetable samples, the sensor exhibits acceptable recoveries and low relative standard deviations, demonstrating its practicability for field food safety supervision and rapid pesticide residue screening.
