Eco-friendly simultaneous extraction of pectins and phenolics from passion fruit (Passiflora edulis Sims) peel: Process optimization, physicochemical properties, and antioxidant activity

Dongxue Huo,Jincheng Dai,Siyu Yuan,Xiaoqing Cheng,Yonggui Pan,Lu Wang,Ruimin Wang

The objective of this study was to simultaneously extract passion fruit ( Passiflora edulis ) peel pectins and phenolics using deep eutectic solvents, to evaluate their physicochemical properties and antioxidant activity. By taking L-proline: citric acid (Pro-CA) as the optimal solvent, the effect of extraction parameters on the yields of extracted passion fruit peel pectins (PFPP) and total phenolic content (TPC) was explored by response surfaces methodology (RSM). A maximum pectin yield (22.63%) and the highest TPC (9.68 mg GAE/g DW) were attained under 90 °C, extraction solvent pH = 2, extraction time of 120 min and L/S ratio of 20 mL/g. In addition, Pro-CA-extracted pectins (Pro-CA-PFPP) and HCl-extracted pectins (HCl-PFPP) were subjected to high performance gel permeation chromatography (HPGPC), Fourier transform infrared spectroscopy (FT-IR), thermogram analysis (TG/DTG) and rheological measurements. Results verified that the Mw and thermal stability of Pro-CA-PFPP were higher than those of HCl-PFPP. The PFPP solutions featured a non-Newtonian behavior, and compared with commercially pectin solution, PFPP solution exhibited a stronger antioxidant activity. Additionally, passion fruit peel extract (PFPE) exhibited stronger antioxidant effects than PFPP. The results of ultra-performance liquid chromatography hybrid triple quadrupole-linear ion trap mass spectrometry (UPLC-Qtrap-MS) and high performance liquid chromatography (HPLC) analysis showed that (−)-epigallocatechin, gallic acid, epicatechin, kaempferol-3- O -rutin and myricetin were the main phenolic compounds in PFPE and PFPP. Our results suggest that Pro-CA can be considered as an eco-friendly solvent for high-efficient extraction of high-value compounds from agricultural by-products.

Investigating of zein-gum arabic-tea polyphenols ternary complex nanoparticles for luteolin encapsulation: Fabrication, characterization, and functional performance

Renjie Li,Zhuangwei Zhang,Jin Chen,Huihui Li,Hongjin Tang

Luteolin has extensive biological effects, but its low water-solubility and oral bioavailability have restricted its application. In this study, we successfully prepared new zein-gum arabic (GA)-tea polyphenols (TP) ternary complex nanoparticles (ZGTL) as a delivery system to encapsulate luteolin using an anti-solvent precipitation method. Consequently, ZGTL nanoparticles showed negatively charged smooth spherical structures with smaller particle size and higher encapsulation ability. X-ray diffraction revealed the amorphous state of luteolin in the nanoparticles. Hydrophobic, electrostatic, and hydrogen bonding interactions contributed to the formation and stability of ZGTL nanoparticles, as indicated by fluorescence and Fourier transform infrared spectra analyses. The inclusion of TP improved the physicochemical stability and luteolin retention rate of ZGTL nanoparticles by forming more compact nanostructures under different environmental conditions, including pH, salt ion concentration, temperature, and storage. Additionally, ZGTL nanoparticles exhibited stronger antioxidant activity and better sustainable release capacity under simulated gastrointestinal conditions due to TP incorporation. These findings demonstrate that ZGT complex nanoparticles have potential applications as an effective delivery system for encapsulating bioactive substances in food and medicine fields.

Cryptotanshinone-Doped Photothermal Synergistic MXene@PDA Nanosheets with Antibacterial and Anti-Inflammatory Properties for Wound Healing

Zongjia Li,Wei Wei,Miaomiao Zhang,Xinyue Guo,Bailin Zhang,Dapeng Wang,Xiue Jiang,Fangxin Liu,Jilin Tang

Humans are threatened by bacteria and other microorganisms, resulting in countless pathogen-related infections and illnesses. Accumulation of reactive oxygen species (ROS) in infected wounds activates strong inflammatory responses. The overuse of antibiotics has led to increasing bacterial resistance. Therefore, effective ROS scavenging and bactericidal capacity are essential and the advanced development of collaborative therapeutic techniques to combat bacterial infections is needed. Here, this work developes an MXene@polydopamine-cryptotanshinone (MXene@PDA-CPT) antibacterial nanosystem with excellent reactive oxygen and nitrogen species scavenging ability, which effectively inactivates drug-resistant bacteria and biofilms, thereby promoting wound healing. In this system, the adhesion of polydopamine nanoparticles to MXene produced a photothermal synergistic effect and free radical scavenging activity, presenting a promising antibacterial and anti-inflammatory strategy. This nanosystem causes fatal damage to bacterial membranes. The loading of cryptotanshinone further expanded the advantages of the system, causing a stronger bacterial killing effect and inflammation mitigatory effect with desired biosafety and biocompatibility. In addition, combining nanomaterials and active ingredients of traditional Chinese medicine, this work provides a new rationale for the future development of wound dressings, which contributes to eliminating bacterial resistance, delaying disease deterioration, and alleviating the pain of patients.

Robust microfluidic construction of polyvinyl pyrrolidone microfibers incorporated with W/O emulsions stabilized by amphiphilic konjac glucomannan

Liying Huang,Huanglong Lin,Nitong Bu,Jie Pang,Ruojun Mu

In this work, we prepared polyvinyl pyrrolidone (PVP) microfibers incorporated water-in-oil (W/O) emulsions. The W/O emulsions were fabricated by hexadecyl konjac glucomannan (HKGM, emulsifier), corn oil (oil phase) and purple corn anthocyanins (PCAs, water phase). The structures and functions of emulsions and microfibers were characterized by confocal laser scanning (CLSM) and scanning electron microscopy (SEM), Fourier transform infrared (FT-IR), Raman and nuclear magnetic resonance (NMR) spectroscopy. The results showed that W/O emulsions exhibited good storage stability for 30 d. Microfibers presented ordered and uniform arrays. Compared with pure PVP microfiber films, the addition of W/O emulsions with PCAs improved the water resistance (WVP from 1.28 to 0.76 g mm/m 2  day kPa), mechanical strength (Elongation at break from 18.35 % to 49.83 %), antioxidation (free radical scavenging rate from 2.58 % to 16.37 %), and antibacterial activity (inhibition zone against E. coli : 27.33 mm and inhibition zone against S. aureus : 28.33 mm) of microfiber films. Results showed that microfiber film exhibited controlled release of PCAs in W/O emulsions, and about 32 % of the PCAs were released from the microfiber film after 340 min. The as-prepared microfiber films exhibited potential applications for food packaging.

A Green and Effective Polyethylene Glycols-Based Microwave-Assisted Extraction of Carnosic and Rosmarinic Acids from Rosmarinus officinalis Leaves

Chunyan Zhu,Yunchang Fan,Xiujun Bai

Rosmarinus officinalisleaves (ROLs) are widely used as a popular culinary spice for flavoring food, in which carnosic acid (CA) and rosmarinic acid (RA) are the main active components. The extraction of CA and RA is limited by lowextraction efficiency and extraction rate. In this work, a microwave-assisted extraction (MAE) method using biodegradable, low-toxic and nonflammable solvents polyethylene glycols (PEGs) as extraction solvents was developed for theextraction of CA and RA from ROLs. Experimental results suggest that PEG-400 was a better choice compared with PEG-200, and the optimal extraction conditions were as follows: 45% of PEG-400, 4.3% of phosphoric acid, 20 s of microwave irradiation time at 280 W of microwave irradiation power, and a 10 mg mL−1solid–liquid ratio, respectively. The tissue structures of ROLs could be effectively disrupted by PEG-based MAE, leading to high CA and RA extraction efficiencies. The PEG-400 extract exhibited stronger 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability compared with butylated hydroxytoluene (BHT). Finally, compared with heating reflux extraction, ultrasound-assisted extraction, maceration, and MAE using ionic liquid and ethanol as extraction solvents, the developed PEG-400 based MAE exhibited the highest extraction ability and fastest extraction rate for CA and RA. These findings suggest that MAE using PEGs as extraction solvents is a promising method for the separation of bioactive compounds from natural plants. Keywords:antioxidant activity;green solvents;microwave-assisted extraction (MAE);Rosmarinus officinalisleaves (ROLs);carnosic acid (CA);rosmarinic acid (RA)

Design and synthesis of a novel class of PDE4 inhibitors with antioxidant properties as bifunctional agents for the potential treatment of COPD

Youzhi Wang,Huifang Wang,Guoqing Yang,Qingjing Hao,Kan Yang,Huizhen Shen,Yulong Wang,Jinxin Wang

It is well known that chronic obstructive pulmonary disease (COPD) patients are always trapped in the vicious circle of inflammation and oxidative stress , therefore anti-inflammatory and antioxidant bifunctional agents may interrupt this vicious cycle in COPD. Phosphodiesterase 4 (PDE4) inhibitors, as anti-inflammatory drugs, have been used for COPD treatment in clinical, and the PDE4 inhibitors with antioxidant properties may be a good strategy to design bifunctional agents for COPD. Sappanone A was the first PDE4 inhibitor with antioxidant properties we identified from natural products in our previous study, which was used by us as a hit compound to design new bifunctional agents for COPD in this study. 27 derivatives of sappanone A including homoisoflavonoids , aurones and chalcones were designed and synthesized by innovatively fusing the antioxidant pharmacophore of catechol from polyphenols and the pharmacophore of catechol ether abstracted from the PDE4 inhibitors of the catechol ether class such as rolipram , roflumilast and apremilast respectively. All the compounds were assayed for the PDE4 inhibitory and radical scavenging against 2, 2-diphenyl-1-picrylhydrazyl (DPPH) activities in vitro. Herein we obtained a series of bifunctional compounds with better PDE4 inhibitory activity than sappanone A, and their free radical scavenging activities were superior to edaravone in vitro. In addition, they can reduce tumour necrosis factor-alpha (TNF-α) production induced by lipopolysaccharide (LPS) in RAW264.7 macrophages and malondialdehyde (MDA) production induced by Fe 2+ in mouse lung homogenate . Meanwhile, it showed outstanding abilities in reducing Fe 3+ and complexing Fe 2+ . 6o , as the candidate anti-inflammatory and antioxidant bifunctional compound, exhibited good drug-likeness, ADME (Absorption, Distribution, Metabolism, Excretion) properties and human liver microsomal stability. In vivo, 6o (50 mg/kg and 100 mg/kg, i. p.) distinctly prevented LPS-induced serum levels of TNF-α in mice. In conclusion, the preliminary investigation provided a novel class of PDE4 inhibitors with antioxidant properties as bifunctional agents for the potential treatment of COPD, which can interrupt the vicious cycle of chronic inflammation and oxidative stress in COPD.

The fiber-bound polyphenols from highland barley with inhibitory effects against carbonyls during in vitro digestion of cookies and French fries

Jinxin Li,Hao Zhang,Xijuan Yang,Ling Zhu,Gangcheng Wu,Hui Zhang

The inhibition capacity of bound-polyphenol rich insoluble dietary fiber (BP-IDF) from highland barley to carbonyl compounds was studied during in vitro gastrointestinal (GI) digestion. More than 97% of fiber-bound polyphenols were still presented in highland barley BP-IDF (HBBP-IDF) after GI digestion. The fiber-bound polyphenols in HBBP-IDF effectively inhibit carbonyls when simultaneously digested with cookies and French fries and exhibited more pronounced effect during gastric digestion stage, with inhibition of 63.4% for methylglyoxal , 54.3% for glyoxal, 47.6% for malondialdehyde and 42.4% for 4-hydroxy-2-nonenel, respectively. These findings suggest that HBBP-IDF could be use as functional ingredients able to inhibit the formation of carbonyls during digestion process and maintain healthful digestive tract environment.

Characterization of an active film prepared with Lentinus edodes (shiitake) polysaccharide and its effect on post-harvest quality and storage of shiitake

Yuxi Guo,Xuefeng Chen,Pin Gong,Hui Long,Jiating Wang,Zhenfang Deng,Ruotong Wang,Aoyang Han,Zhuoya Qi,Wenbo Yao,Wenjuan Yang,Jing Wang,Nan Li,Fuxin Chen

The aim of this study was to prepare a film based on shiitake ( Lentinus edodes ) stalk polysaccharides (LEP) for mushroom preservation. The effects of different LEP concentrations on physical, mechanical, antioxidant, and antimicrobial properties of the prepared film were evaluated. Using scanning electron microscopy, it was revealed that the addition of 1.5 % LEP resulted in homogeneous distribution in the prepared film, as well as greatly improved its antimicrobial properties. Moreover, LEP film resulted in superior mushroom preservation by regulating enzyme activities related to mushroom browning and softening, thereby decaying these processes. In addition, the prepared film maintained mushroom quality by reducing the accumulation of H 2 O 2 and activating the regulatory system against oxidative stress. Collectively, the findings of the present study highlight the potential benefits of LEP films as a strategy to improve mushroom quality and prevent post-harvest spoilage, hence constituting a novel prospect for the development of shiitake by-products.

Purification and Identification of Novel Antioxidant Peptides from Hydrolysates of Peanuts (Arachis hypogaea) and Their Neuroprotective Activities

Rui Ma,Jinrong Bai,Yuting Huang,Zhiqiu Wang,Yongju Xu,Yan Huang,Kai Zhong,Yina Huang,Hong Gao,Qian Bu

Peanut (Arachis hypogaea) peptides have various functional activities and a high utilization value. This study aims to isolate and characterize antioxidant peptides from peanut protein hydrolysates and further evaluate their neuroprotection against oxidative damage to PC12 cells induced by 6-hydroxydopamine (6-OHDA). After the peanut protein was hydrolyzed with pepsin and purified using ultrafiltration and gel chromatography, six peptides were identified and sequenced by high-performance liquid chromatography (HPLC) and liquid chromatography–tandem mass spectrometry (LC–MS/MS). Out of these six peptides, Pro-Gly-Cys-Pro-Ser-Thr (PGCPST) exhibited a desirable antioxidant capacity, as determined using the 1,1-diphenyl-2-picrylhydrazyl, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt, and hydroxyl radical scavenging assays. Moreover, our results indicated that the peptide PGCPST effectively increased the cell viability and reduced the cell apoptosis in 6-OHDA-induced PC12. RNA sequencing further showed that the neuroprotective effect of the peptide PGCPST was mediated via sphingolipid metabolism-related pathways. With further research efforts, the peptide PGCPST was expected to develop into a new neuroprotective agent.

Chemical composition and bioactivities of the essential oil of Coreopsis tinctoria and Chrysanthemum morifolium

Zhong-Rong Jiang,Ting Zhang,Ling-Bo Ji,Lei Wu,Hong-Hui Dong,Guo-Liang Zhao,Hong-Qian Shentu,Bao-Jiang He,Tian-Xiao Li

This study was aiming at the differences in the aroma, flavor compounds, and bioactivities of essential oil of Coreopsis tinctoria flower (CTFEO) and essential oil of Chrysanthemum morifolium flower (CMFEO). Both essential oils were extracted by steam distillation. Their flavor compositions were determined by GC-MS and the antioxidant, antibacterial, and anti-inflammatory activities were also evaluated. It was found that CTFEO possessed a special medicinal herb odor, which was obviously stronger than those of CMFEO. A preferred condition for steam distillation was flower powder–water ratio of 7.5:100 and extraction for more than 3.0 h. Interestingly, the main flavor compounds of CTFEO were determined as D-limonene (34.54%), cis-carveol (5.49%), trans-α-bergamotene (4.44%), and α-pinene (3.90%), which were quite different from those of CMFEO. CTFEO showed better antioxidant activities with IC50 value of 121.4 ± 9.8 μg/mL than those of CMFEO. It also exhibited stronger antibacterial effects than those of CMFEO, with minimal inhibitory concentrations (MICs) ranging from 0.183 to 1.412 mg/mL against six strains of bacteria. Moreover, CTFEO was also found to exhibit strong anti-inflammatory effects that reduced the LPS-induced RAW 264.7 cell mRNA expression of the cytokines IL-1β, IL-6, and TNF-α at a concentration of 10.0 μg/mL. All these results suggest that the aroma, flavor compounds, and bioactivities of the two essential oils were quite different and they could be used as tea, drink, and food additive in the food industry for further development.
