Lysosomal-targeted near-infrared phototherapy agent with type-I photodynamic activity for high-performance synergistic therapy under hypoxia

Xianjiao Tang,Xiaojing Yang,Yuhan Wang,Liping Chen,Pengwei Chen,Mei-Xia Zhao

The development of near-infrared (NIR) multifunctional phototherapy agents is important for expanding the application of photodynamic therapy (PDT)/photothermal therapy (PTT) for tumor diagnosis and treatment. Here, we constructed a phototherapeutic agent, CYPT nanoparticles (NPs), based on a cyanine dye (Cy7) for synergistic NIR image-guided tumor-specific PDT/PTT. CYPT NPs produced large amounts of type I O 2 •− and type II 1 O 2 under 808-nm laser irradiation with a high photothermal conversion efficiency (33.5%). Moreover, CYPT NPs targeted the lysosomes of cancer cells and reduced GSH, contributing to the high efficiency of PDT. CYPT NPs had similar inhibitory effects on 4T1 cells under hypoxic and normoxic conditions. Experiments in tumor-bearing mice confirmed that CYPT NPs can be enriched in tumor tissues, have good NIR fluorescence imaging performance, and exhibit efficient synergistic PDT/PTT, thus providing an attractive method for accurate tumor targeting and efficient treatment.

Win-win integration: A mitochondria targeted AIE photosensitizer for hypochlorite detection and type I & type II photodynamic therapy

Jiahao Ni,Lichao Yu,Yixuan Wang,Tong Yang,Yueqi Bai,Bowen Zheng,Manshan Liang,Xiaoxia Ye,Yun-Yun Quan,Feifei Lin,Zu-Sheng Huang

Background Photodynamic therapy (PDT) is a pioneering and effective anticancer modality with low adverse effects and high selectivity. Hypochlorous acid or hypochlorite (HClO/ClO − ) is a type of inflammatory cytokine. The abnormal increase of ClO − in tumor cells is related to tumor pathogenesis and may be a “friend” for the design and synthesis of responsive phototherapy agents. However, preparing responsive phototherapy agents for all-in-one noninvasive diagnosis and simultaneous in situ therapy in a complex tumor environment is highly desirable but still remains an enormously demanding task. Results An acceptor-π bridge-donor-π bridge-acceptor (A-π-D-π-A) type photosensitizer TPTPy was designed and synthesized based on the phenothiazine structure which was used as the donor moiety as well as a ClO − responsive group. TPTPy was a multifunctional mitochondria targeted aggregation-induced emission (AIE) photosensitizer which could quickly and sensitively respond to ClO − with fluorescence “turn on” performance (19-fold fluorescence enhancement) and enhanced type I reactive oxygen species (ROS) generation to effectively ablate hypoxic tumor cells. The detection limit of TPTPy to ClO − was calculated to be 185.38 nM. The well-tailored TPTPy anchoring to mitochondria and producing ROS in situ could disrupt mitochondria and promote cell apoptosis. TPTPy was able to image inflammatory cells and tumor cells through ClO − response. In vivo results revealed that TPTPy was successfully utilized for PDT in tumor bearing nude mice and exhibited excellent biological safety for major organs. Significance and novelty A win-win integration strategy was proposed to design a tumor intracellular ClO − responsive photosensitizer TPTPy capable of both type I and type II ROS production to achieve photodynamic therapy of tumor. This work sheds light on the win-win integration design by taking full advantage of the characteristics of tumor microenvironment to build up responsive photosensitizer for in situ PDT of tumor.

Ultrasensitive Fluorescent Microsensors Based on Aptamers Modified with SYBR Green I for Visual Quantitative Detection of Organophosphate Pesticides

Qianru Zhang,Anqi Liu,Xin Song,Shihao Xu,Liangguo Da,Dan Lin,Changlong Jiang

Organophosphate pesticides (OPs) are widely utilized in agricultural production, and the residues threaten public health and environmental safety due to their toxicity. Herein, a novel and simple DNA aptamer-based sensor has been fabricated for the rapid, visual, and quantitative detection of profenofos and isocarbophos. The proposed DNA aptamers with a G-quadruplex spatial structure could be recognized by SYBR Green I (SG-I), resulting in strong green fluorescence emitted by SG-I. The DNA aptamers exhibit a higher specific binding ability to target OP molecules through aromatic ring stacking, disrupting the interaction between SG-I and DNA aptamers to induce green fluorescence quenching. Meanwhile, the fluorescence wavelength of G-quadruplex fluorescence emission peaks changes, accompanied by an obvious fluorescence variation from green to blue. SG-I-modified aptasensor without any additive reference fluorescence units for use in multicolor fluorescence assay for selective monitoring of OPs was first developed. The developed aptasensor provides a favorable linear range from 0 to 200 nM, with a low detection limit of 2.48 and 3.01 nM for profenofos and isocarbophos, respectively. Moreover, it offers high selectivity and stability in real sample detection with high recoveries. Then, a self-designed portable smartphone sensing platform was successfully used for quantitative result outputs, demonstrating experience in designing a neotype sensing strategy for point-of-care pesticide monitoring.

Sun Yuchen,Guo Mingyu

In this work, we report the efficient removal of I − /IO 3 − from wastewater by tripropylamine quaternary ammonium cation based monodisperse microspheres. Benefitting from the rapid solvent exchange process and the excellent ionic selectivity of tripropylamine quaternary ammonium cation, these microspheres show capture rate of 98% for I − and 66% for IO 3 − at a concentration of 50 ppm. Meanwhile, they exhibit high adsorption capacities of 350 mg/g for I − and 370 mg/g for IO 3 − , coupling with excellent reusability. These characteristics highlight their superior practicability for removal of iodine, particularly in the context of radioactive iodine-contaminated water.

Structure characterization and gelling properties of RG-I-enriched pectins extracted from citrus peels using four different methods

Xueping Wang,Chengying Zhao,Jirong Wang,Xingmiao Lu,Yuming Bao,Deli Zhang,Jinkai Zheng

To facilitate the application of rhamnogalacturonan-I (RG-I)-enriched pectins (RGPs) as novel, healthy, and gelling food additives, this study compared the structural characteristics and gelling properties of RGPs extracted from citrus peel via four methods (alkali: AK, high-temperature/pressure: TP, citric acid: CA, and enzyme-assisted: EA extractions). AK and CA yielded pectins with the highest RG-I proportions (54.8 % and 51.9 %, respectively) by disrupting the homogalacturonan region; TP and EA increased the RG-I proportions by ~10 %. Among the four methods, AK induced the lowest degree of esterification (DE) (6.7 %) and longer side chains that form strong entanglement, contributing to its highest gel hardness. The relatively low DE (18.5 %) of CA RGP facilitated stable gel formation. Notably, its highly branched RG-I region afforded more intramolecular hydrophobic interactions, making a more highly cross-linked gel network of better gel resilience. In contrast, TP induced the highest DE (57 %) and curved molecular chains; it inhibited Ca 2+ binding, entanglement, and intramolecular hydrophobic interactions, and thus no gel formed. EA RGP was associated with the lowest molecular size, rendering it more difficult for Ca 2+ to form links, which resulted no gel. These findings offer insights into the relationship among the extraction methods, molecular structures, and gelling properties of RGPs.

Spent lithium-ion battery recycling: investigation into the inhibition and separation between Li(I) and Al(III) ions

Lou Wen-bo,Liu Dong-yan,Zhao Da,Wang Yun,Sun Zi-cheng,Zou Yi,Niu Xiao-wei,Cheng Quan-guo,Li Jian-zhong,Liu Hong-hui

The recovery of NCM (LiNi x Co y Mn 1-x-y O 2 , abbreviated as NCM) cathode materials from retired lithium-ion batteries is a research hotspot in resource recycling field. Nevertheless, due to aluminum impurities mixed in raw materials, Li(I) and Al(III) ions are easy to combine to form water-insoluble LiAlO 2 during reduction, affecting Li(I) ions’ water-leaching efficiency. In this study, the LiAlO 2 formation environment is analyzed. Thermodynamic analysis reveals that there is generally less spontaneity in the reaction of Li (I) ions with aluminum monomers compared to Al 2 O 3 , and the introduction of CO 2 can inhibit Li-Al-O substance production. Li-Al binding experiments show that there is almost no direct reaction between Li 2 CO 3 and aluminum monomers, but under Ar atmosphere, Li 2 CO 3 and Al 2 O 3 can easily lead to Li-Al binding, a kinetic analysis considers that binding is controlled by chemical reaction, with an activation energy of 57.78 kJ/mol, and the activation energy increases to 88.028 kJ/mol with CO 2 atmosphere. Under reduced optimization conditions (reduction temperature: 500 °C, time: 6 h, and m NCM : m Al  = 1: 0.25), the carbonation water-leaching efficiency can reach 95.1% for Li(I) ions and less than 1% for Ni(II), Co(II), Mn(II), and Al(III) ions. Thus, this work offers a theoretical basis for the industrial Li(I) ions’ recovery from aluminum-containing NCM materials.

Integrated Sensing Platform Based on MOF@COF Nanocomposites for the Ultrasensitive Biosensing of Sesame Allergens Ses i 2

Pengfei Dong,Shuang Wu,Youfa Wang,Ning Yu,Vijaya Raghavan,Jin Wang

Sesame allergy has emerged as a public health issue due to its persistent occurrence, severe symptomatic manifestations, and widespread incidence globally. Among sesame allergens, 2S albumins (Ses i 2) is one of the widely recognized allergens, and its precise detection is urgently needed to prevent sesame-related allergies. In this work, a composite structure was designed and synthesized by combining metal ion self-assembled metal organic frameworks (MOFs) and imine constructed covalent organic frameworks (COFs) (represented by MOF@COF), which constructing a high-sensitivity sensing system for identification and detection of Ses i 2. A novel hybrid material, comprising a strategically assembled combination of UIO-66-NH 2 and TA-COF, was crafted to yield a functionalized UIO-66-NH 2 @TA-COF composite, demonstrating remarkable electrochemical performance. As a result, a significant quantity of aptamer strands can adhere to the UIO-66-NH 2 @TA-COF surface resulting from robust π-π stacking interactions. With the change in the relative concentration of Ses i 2 from 5 to 650 nM, the UIO-66-NH 2 @TA-COF sensor exhibited a low detection limit of 0.13 nM, which demonstrates its extremely high sensitivity. In addition, the UIO-66-NH 2 @TA-COF sensor exhibited excellent reproducibility, stability and specific recognition ability to Ses i 2. Therefore, this innovative sensor exhibits great potential for real-world applications in food safety.

An AIE cyanine photosensitizer achieving hypoxic tumor ablation through synergistic treatment of type i photodynamic and photothermal therapy

Peng-Wei Chen,Lin-Song Li,Yu-Meng Zhang,Bang-Bang Liu,Xianjiao Tang,Liping Chen,Xiaojing Yang,Mei-Xia Zhao

In recent years, most photosensitizers suffer from poor photothermal conversion efficiency (PCE), vulnerability to aggregated-caused quenching under physiological conditions, and are highly dependent on oxygen, which seriously limit their application in cancer therapy. In this study, DTIR780 was an aggregation induced emission (AIE) photosensitizer, which was synthesized through the originality of molecular engineering. Because DTIR780 had a special organelle targeting lysosomes and mitochondria, it had excellent PCE (PCE = 28.9 %), and could simultaneously generate Type I reactive oxygen species (ROS): O 2 ·- and ·OH and Type II ROS: 1 O 2 , showing significantly enhanced tumor phototoxicity in both normoxia and hypoxic tumor microenvironments. In vivo testing demonstrated that DTIR780 NPs was efficiently accumulated at the tumor site while significantly inhibiting tumor growth in a subcutaneous H22 tumor mouse model. In summary, this lysosome and mitochondria targeted DTIR780 NPs has great potential in clinically image-guided type I photodynamic therapy (PDT) and photothermal therapy (PTT), offering a novel avenue for developing high-performance type I PDT/PTT photosensitizers for the efficient treatment of hypoxia tumors.

The semi-interpenetrating network hydrogel loaded with Oridonin and DNase-I on healing of chemoradiotherapy-induced oral mucositis

Yuxue Pan,Mengyuan Wang,Peng Wang,Hongliang Wei,Xiangjuan Wei,Dongmei Wang,Yongwei Hao,Yongxue Wang,Hongli Chen

Oral mucositis (OM) is a common side effect experienced by cancer patients undergoing chemotherapy and/or radiotherapy. In this study, we developed a semi-interpenetrating network hydrogel (IPN) that is suitable for adhesion to oral mucous membranes and gradual degradation in the oral environment. This IPN was based on the combination of ε-polylysine (PLL) and Hetastarch (HES), loaded with DNase-I and Oridonin (ORI) (ORI/DNase-I/IPN) for the treatment of OM. In vitro studies were conducted to evaluate degradation, adhesion, release analysis, as well as bioactivity including cell proliferation and wound healing assays using epidermal keratinocyte and fibroblast cell lines. Furthermore, the therapeutic effects of ORI/DNase-I/IPN were investigated in vivo using Sprague-Dawley (SD) rats with chemoradiotherapy-induced OM. The results demonstrated that the IPN exhibited excellent adhesion to wet mucous membranes, and the two drugs co-encapsulated in the hydrogel were released in a controlled manner. The in vivo wound repair effect, microbiological assays, H&E and Masson staining supported the non-toxicity of ORI/DNase-I/IPN, as well as its ability to accelerate the healing of oral ulcers and reduce inflammation. Overall, ORI/DNase-I/IPN demonstrated a therapeutic effect on oral ulcers in rats by significantly accelerating the healing process and reducing inflammation. These findings provide new insights into possible therapies for OM.

Regio-isomerization Optimization Strategy for Photosensitizers: Achieving Ultrahigh Type I Reactive Oxygen Species Generation to Enhance Cancer Photoimmunotherapy

Quan Wang,Lili Yang,Xiaoyu Xing,Wenjie Liang,Renzhi Wu,Chen Xiong,Meng Wu,Cheng Zhong,Haoke Zhang,Shixuan Wang,Fan Xia,Xiaoding Lou,Dugang Chen,Jun Dai

Phototherapy, renowned for its noninvasiveness, is widely employed in tumor treatment. However, the tumor microenvironment is usually hypoxic, with insufficient reactive oxygen species (ROS) production, severely limiting its application. Herein, we introduce a regio-isomerization optimization strategy and have synthesized four regio-isomeric photosensitizers featuring a donor–acceptor (D–A) configuration by tactically varying the linkage sites between D and A. Among them, TAF-3 with excellent photostability has an ultrahigh type I ROS production efficiency (4.79 times that of methylene blue) and a photothermal conversion efficiency of 41.7%. TAF-3 improves the conjugation degree; produces an appropriate intramolecular charge transfer effect, which enhances its optical properties and phototherapeutic efficiency; and promotes a stronger immune cell death effect, reducing postoperative melanoma recurrence by 60%. Overall, the optical attributes of D–A type photosensitizers can be tailored through the precision modulation of regio-isomerization, offering a promising avenue for the advancement of clinical photosensitizers suitable for phototherapy.

Reversible multielectron transfer I−/IO3− cathode enabled by a hetero-halogen electrolyte for high-energy-density aqueous batteries

Xie Congxin,Wang Chao,Xu Yue,Li Tianyu,Fu Qiang,Li Xianfeng

The ever-increasing need for energy-dense batteries with high safety is fuelling global research and innovations in new redox chemistry and device design. Here we show an aqueous battery employing highly concentrated hetero-halogen electrolytes that contain I − and Br - , resulting in a multielectron transfer process of I − /IO 3 − . The intermediate bromide species IBr and Br 2 , generated during the electrochemical process, enhances the reaction kinetics and alleviates the potential gap between oxidation and reduction. When using a 6 M I − electrolyte to achieve over 30 M electron transfers, the I − /IO 3 − cathode displayed a high specific capacity of over 840 Ah l catholyte −1 . A battery with Cd/Cd 2+ as the anode demonstrated a high energy density of over 1,200 Wh l catholyte −1 . Even at an exceptionally high current density of 120 mA cm −2 , an energy efficiency of 72% was obtained. Our work demonstrates that safe aqueous batteries with high energy density are possible, offering a development option for grid-scale energy storage and even electric vehicles.

RG-I pectic polysaccharides and hesperidin synergistically modulate gut microbiota: An in vitro study targeting the proportional relationship

Jiaxiong Wu,Sihuan Shen,Huan Cheng,Haibo Pan,Xingqian Ye,Shiguo Chen,Jianle Chen

In this study, we investigated how different proportions blends of Rhamnogalacturonan-I pectic polysaccharides and hesperidin impact the gut microbiota and metabolites using an in vitro simulated digestion and fermentation model. The results indicated that both of them could modulate the gut microbiota and produce beneficial metabolites. However, their blends in particular proportions (such as 1:1) exhibited remarkable synergistic effects on modulating the intestinal microenvironment, surpassing the effects observed with individual components. Specifically, these blends could benefit the host by increasing short-chain fatty acids production (such as acetate), improving hesperidin bioavailability, producing more metabolites (such as hesperetin, phenolic acids), and promoting the growth of beneficial bacteria. This synergistic and additive effect was inseparable from the role of gut microbiota. Certain beneficial bacteria, such as Blautia , Faecalibacterium , and Prevotella , exhibited strong preferences for those blends, thereby contributing to host health through participating in carbohydrate and flavonoid metabolism.
