At present, NIR-II-triggered photothermal biomedical applications are limited by complex synthesis reactions , mediocre photothermal conversion efficiency, and difficult degradation. Herein, we prepared biodegradable Bi flower-like nanoparticles (phospholipid-modified Bi nanoflowers , BNFs) with high photothermal conversion efficiency (∼33.52 %) in NIR-II by a simple method and then modified them with the red blood cell membrane and dextran 40 (DRBCM) to improve their in vitro stability, to escape macrophages clearance and to enhance tumor accumulation. Dextran coating onto the surface of particles as a dispersant shell stabilizes inorganic particles by maintaining the surface charges and creating steric repulsions upon compression of neighboring polymer chains. In vitro and in vivo experiments proved that combined thermoradiotherapy of DRBCM-BNFs exhibited significantly enhanced tumor inhibition efficacy than monotherapy with good biocompatibility and low toxicity due to its biodegradability . Furthermore, the mechanism studies demonstrated that DRBCM-BNFs could serve as a nano sensitizer to promote the thermoradiotherapy under NIR-II illumination and X-ray irradiation, by downregulating heat shock protein 70 (HSP70) and phosphorylated-p65 (p-p65) to reduce the thermal resistance and radioresistance of tumor cells and increasing the expression of apoptosis-related protein cleaved caspase-3. In conclusion, DRBCM-BNFs could be a promising green delivery platform for the sensitization of synergistic thermoradiotherapy.

Although biodegradable polymer coatings can impede corrosion of magnesium (Mg)-based orthopedic implants , they are prone to excessive degradation and accidental scratching in practice. Bone implant-related infection and limited osteointegration are other factors that adversely impact clinical application of Mg-based biomedical implants. Herein, a self-healing polymeric coating is constructed on the Mg alloy together with incorporation of a stimuli-responsive drug delivery nanoplatform by a spin-spray layer-by-layer (SSLbL) assembly technique. The nanocontainers are based on simvastatin (SIM)-encapsulated hollow mesoporous silica nanoparticles (S@HMSs) modified with polydopamine (PDA) and polycaprolactone diacrylate (PCL-DA) bilayer . Owing to the dynamic reversible reactions, the hybrid coating shows a fast, stable, and cyclical water-enabled self-healing capacity. The antibacterial assay indicates good bacteria-killing properties under near infrared (NIR) irradiation due to synergistic effects of hyperthermia, reactive oxygens species (ROS), and SIM leaching. In vitro results demonstrate that NIR laser irradiation promotes the cytocompatibility, osteogenesis, and angiogenesis . The coating facilitates alkaline phosphatase activity and expedites extracellular matrix mineralization as well as expression of osteogenesis-related genes. This study reveals a useful strategy to develop multifunctional coatings on bioabsorbable Mg alloys for orthopedic implants .

Developing an effective dressing against bacterial infection and synchronously addressing wound complications, such as bleeding, long-term inflammation, and reinfection, are highly desirable in clinical practice. In this work, a second near-infrared (NIR-II) responsive nanohybrid consisting of imipenem encapsulated liposome with gold-shell and lipopolysaccharide (LPS)-targeting aptamer, namely ILGA, is constructed for bacteria elimination. Benefiting from the delicate structure, ILGA exhibits strong affinity and a reliable photothermal/antibiotic therapeutic effect toward multidrug-resistant Pseudomonas aeruginosa (MDR-PA). Furthermore, by incorporating ILGA with a thermosensitive hydrogel poly(lactic-co-glycolic acid)–polyethylene glycol–poly(lactic-co-glycolic acid) (PLGA-PEG-PLGA), a sprayable dressing ILGA@Gel was prepared, which enables a quick on-demand gelation (10 s) for wound hemostasis and offers excellent photothermal/antibiotic efficacy to sterilize the infected wound. Additionally, ILGA@Gel provides satisfactory wound-healing environments by reeducating wound-associated macrophages for inflammation alleviation and forming a gel layer to block exogenous bacterial reinfection. This biomimetic hydrogel reveals excellent bacteria eradication and wound recovery effectiveness, demonstrating its promising potential for managing complicated infected wounds.

Severe ferroptosis is a non-apoptotic iron-dependent cell death pathway, and extreme hypoxia and anomalous iron metabolism in tumors critically suppress the efficacy of ferroptosis therapy. Overcoming tumor hypoxia while facilitating ferroptosis is an extremely challenging task. Herein, the photosensitizer named BOTTQ with specific targeting of lipid droplet (LD) was synthesized by increasing the tail chain length of triphenylamine. The addition of alkoxy chains optimized the molecular structure and fostered inter-system crossing and type I reactive oxygen species (ROS) generation. Besides, BOTTQ yielded large amounts of type I ROS under light excitation oxidize efficiently polyunsaturated fatty acids in lipid monolayer of LD and contribute to the formation of ferroptosis substrate. Meanwhile, the generation of type I ROS accelerated the Fenton reaction, thus effectively boosting the occurrences of ferroptosis. Furthermore, the successful encapsulation of BOTTQ into DSPE-PEG-MAL material not only elevates the stable type of nanoparticle colloids, but the MAL moiety can deplete the functional thiols of glutathione in a mild environment, which leads to glutathione failure and facilitates ferroptosis. Thus tumor-specific ferroptosis-inducing strategy offers a universally applicable therapy with the capacity to enhance type I ROS production and effectively contain tumorigenesis.

Lanthanide-doped biocompatible nanoparticles have promising applications in near-infrared second region imaging due to their high chemical stability, enhanced photostability and sharp emission bandwidth. However, the weak light absorption capacity limits the application of rare-earth nanoparticles (RENPs) for bioimaging. We prepared a coumarin-derived dye sensitized NaYGdF 4 :Yb,Er nanoparticle probe, in which the organic dye enhances photon absorption through the sensitization process, improving the luminescence efficiency of the rare earth particles near 1000 and 1500 nm. In addition, good water solubility and stability of the probe are imparted by coating the particles with amphiphilic polymers distearoyl phosphatidylethanolamine-polyethylene glycol (DSPE-PEG) and polyacrylic acid. This composite probe with good biocompatibility and NIR II luminescence properties can be used for vascular imaging, providing a tool for the detection of hematologic-related diseases.

Gold nanorods (AuNRs) have potential application value in the field of cancer therapy. However, the poor stability, high dark toxicity, insufficient drug-carrying capacity and photothermal conversion efficiency have limited its practical applications. In this study, we encapsulated AuNRs in mesoporous polydopamine (mPDA) shells. The mPDA has exceptional adhesion, excellent biocompatibility , large specific surface area and favorable photothermal properties, making [email protected] an ideal photothermal composite and drug delivery carrier. [email protected] can generate heat under near-infrared (NIR) laser irradiation, which disrupts the intermolecular forces and hydrogen bonds between PDA shell and drug, showing a photothermal enhanced drug release. Furthermore, by using polyethyleneimine (PEI) to seal the pore channels of mPDA, the pH-/NIR-responsive nanocarrier of [email protected] @PEI was fabricated, which shows a high drug loading capacities for DOX to 121.7 mg/g. What's more, by combination of chemotherapy and photothermal therapy , the [email protected] @PEI nanocarrier exhibits significant anticancer effect in vitro . In a word, this carrier demonstrated high drug capacity, excellent photothermal conversion efficiency (55.7%) and pH/NIR stimulation-responsive delivery performance, and promising to become a nanoplatform for multifunctional cancer therapy in the future.

The widespread use of antibiotics leads to the increasing drug resistance of bacteria and poses a threat to human health. Therefore, there is an urgent need to develop new antibacterial strategies. Herein, based on the good photothermal properties of Copper sulfide (CuS) nanoparticles under near infrared (NIR) laser, we developed a NIR-Ⅱ window triple-mode synergetic antibacterial cCuS (cationic CuS) @Vancomycin (Van) nanoplatform. In the proposed nanoplatform, the positive charge on the surface makes cCuS@Van nanoplatform show better bacterial uptake and membrane damage; vancomycin induces chemical sterilization and provides a targeting effect to the nanoplatform; combined with the strong photothermal effect and deep tissue penetration at the excitation of 1064 nm laser, cCuS@Van nanoplatform can effectively kill bacterial. The photothermal conversion efficiency of the nanoplatform can reach 49.12 % and in vitro experiments show a sterilizing rate of more than 99.5 % to staphylococcus aureus ( S. aureus ) at the concentration of 3.0 μM, which also demonstrated the synergistic effect of cCuS@Van nanoplatform. In addition, low cytotoxicity to human cells conforms the good biocompatibility of the as-prepared cCuS@Van nanoplatform, which endows it a good application prospect in the field of antibacterial, such as wound healing and implant sterilization.

In this work, we introduced the acrylate recognition group into dicyanoisophorone derivative DCI-C–OH to construct the NIR fluorescent probe DCI-C-Cys with a large Stokes shift (240 nm). DCI-C-Cys could specifically respond to Cys, resulting in a 22-fold increase in fluorescence intensity at 702 nm. Meanwhile, the probe has the advantages of good water solubility, high sensitivity (93 nM), and excellent biocompatibility. Moreover, DCI-C-Cys successfully monitored endogenous and exogenous Cys in HepG2 cells and zebrafish. Most importantly, we found that balsam pear polysaccharide could lead to the increase of intracellular Cys levels, which might be conducive to the further study of the antioxidant mechanism of balsam pear polysaccharide. Low concentrations of endogenous hydrogen peroxide (H2O2) and intrinsic oxidative stress defense system greatly limited the efficiency of Fenton and Fenton-like reactions driven by a single metal element. Moreover, the complex physiological environment and inherent self-healing function of tumors make it difficult for chemodynamic therapy (CDT) alone, driven by Fenton and Fenton-like reactions, to eliminate malignant tumors. Herein, a tumor microenvironment (TME) remodeling cobalt-stannous sulfide CoSnS2 (CSS) hollow nanocubes are presented for photothermal therapy (PTT)-improved CDT, in which two redox pairs (Co2+/Co3+ and Sn2+/Sn4+) are involved in catalyzing H2O2 to generate a large number of toxic hydroxyl radicals (˙OH) resulting in massive apoptosis of tumor cells. Importantly, two redox pairs (Co2+/Co3+ and Sn2+/Sn4+) promoted the depletion of glutathione (GSH), which reduced tumor antioxidant capacity and thus enhanced the CDT therapeutic impact of CDT. Furthermore, the CDT efficiency was greatly improved by the excellent photothermal effect (η = 47.23%). The combination of PTT and CDT achieved good therapeutic effects, which may provide a new perspective for the development of other anti-tumor strategies on the basis of improved CDT and modulation of TME.

Antibacterial upconversion nanoparticles (UCNP)-based photodynamic-chemical combined therapy (UCNP-aPCCT) provides an ideal method to solve the problem of antibiotic-resistant bacteria in deep-tissue infections. Saponin is a kind of natural product exhibiting promising antibacterial activity. Curcumin is also a natural product that could be used as photosensitizer for antibacterial photodynamic therapy (aPDT). Herein, we have designed the dual-layer silica-coated upconversion nanoparticles combined with UCNP-photodynamic therapy (PDT) and chemotherapy using curcumin as a photosensitizer and the Camellia sapogenin derivative (CSD) as an antibacterial agent. It turned out that the prepared UCNP@mSiO2(Cur)@DmSiO2(CSD)@PVP nanoparticles displayed high antibacterial activity against Escherichia coli and Staphylococcus aureus under near-infrared (NIR) irradiation at 808 nm. Its antibacterial activity is stronger than that of the simple sum of saponin and curcumin aPDT, indicating the existence of a synergetic photodynamic-chemical effect. The current nanoparticle drug system may have potential applications for deep-tissue anti-infection treatment.

Nanozymes mediated catalytic therapy can produce toxic reactive oxygen species (ROS) and destroy the metabolic balance of tumor cells, providing a new direction for cancer treatment. However, the catalytic efficiency of a single nanozyme is limited by the complexity of the tumor microenvironment (hypoxia, GSH overexpression, etc.). In order to overcome these problems, we designed flower-like Co-doped FeSe2 (Co–FeSe2) nanozymes by a simple wet chemistry method. Co–FeSe2 nanozymes not only exhibit high POD and OXD-mimicking activities for facile kinetics, but also effectively consume over-expressed glutathione (GSH), inhibiting the consumption of generated ROS and destroying the metabolic balance of the tumor microenvironment. These catalytic reactions trigger cell death through apoptosis and ferroptosis dual pathways. More importantly, under the NIR II laser irradiation, the catalytic activities of Co–FeSe2 nanozymes are boosted, confirming the photothermal and catalytic synergistic tumor therapy. This study takes advantage of self-cascading engineering that offers new ideas for designing efficient redox nanozymes and promoting their clinical translation.

In this work, near-infrared (NIR) light driven concave octopod PtCu alloy nanocages (OPtCu-NCs) were fabricated and first developed as photothermal synergetic Fenton catalyst for degradation of pollutant dyes. Moreover, by irradiating with NIR laser (808 nm), the obtained OPtCu-NCs exhibit excellent NIR absorption capacity and thus produce hyperpyrexia over 50 °C. Concomitantly, by the photothermal effect, the OPtCu-NCs nanocatalysts was used for elimination of organic dyes such as Methylene blue (MB) and Rhodamine B (RhB). The NIR-driven photothermal degradation mechanism under NIR irradiation can be attributed to the Fenton-like catalytic performance of Cu ions , then -N C- and/or C-C bonds in the molecular skeleton were fractured and electrons were donated intensively. As confirmed by a typical radical trapping experiment, electrons trigger the formation of miscellaneous reactive species (O 2 - , HO 2 , especially •OH) subsequently . Moreover, the photothermal Fenton-catalytic activity of the as-prepared OPtCu-NCs had maintained at 93.15% (for MB) and 91.87% (for RhB) after ten circulating degradation processes. All acquired results reveal that the OPtCu-NCs have the potential applied value in dyestuff-wastewater treatment under neutral pH.
