This study successfully grafted caffeic acid and 3,4-dihydroxybenzoic acid into chitosan through a coupling reaction, yielding grafting ratio of 8.93 % for caffeic acid grafted chitosan (CA-GC) and 9.15 % for 3,4-dihydroxybenzoic acid grafted chitosan (DHB-GC) at an optimal concentration of 4 mmol phenolic acids. The characterization of modified chitosans through ultraviolet visible spectrometer (UV–vis), Fourier transform infrared spectrometer (FTIR), proton nuclear magnetic resonance ( 1 H NMR), and x-ray photoelectron spectrometer (XPS) confirmed the successful grafting of phenolic acids. In the subsequent step of emulsion preparation, confocal laser scanning microscope images confirmed the formation of O/W (oil-in-water) emulsions. The phenolic acid-grafted chitosans exhibited better emulsification properties compared to native chitosan, such as reduced droplet size, more uniform emulsion droplet distribution, increased ζ-potential, and enhanced emulsifying activity and stability. Moreover, the modified chitosans demonstrated increased antioxidant activities (evidenced by DPPH and β-carotene assays) and displayed greater antimicrobial effects against E. coli and S. aureus . Its efficacy in curcumin encapsulation was also notable, with improved encapsulation efficiency, sustained release rates, and enhanced storage and photostability. These findings hint at the potential of modified chitosans as an effective emulsifier.

Recent research studies have shown that the low survival rate of liver cancer is due to drug resistance and metastasis. In the tumor microenvironment (TME), activated hepatic stellate cells (aHSCs) have been proven to favor the development of liver cancer. Hence, the combination therapy dual-targeting aHSCs and tumor cells might be an effective strategy for treatment of liver cancer. In this study, the novel multifunctional liposomes (CAPS-CUR/GA&Gal-Lip) were prepared for co-delivery of curcumin (CUR) and capsaicin (CAPS), in which glycyrrhetinic acid (GA) and galactose (Gal) were chosen as targeting ligands to modify the liposomes (Lip) for dual-targeting liver cancer. To mimic TME, a novel HSCs+HepG2 (human hepatoma cell line) cocultured model was established for the antitumor effect in vitro. The results showed that, compared to HepG2 cells alone, the cocultured model promoted drug resistance and migration by upregulating the expression of P-glycoprotein (P-gp) and Vimentin, which were effectively inhibited by CAPS-CUR/GA&Gal-Lip. The efficacy of the in vivo antitumor was evaluated by three mice models: subcutaneous H22 (mouse hepatoma cell line) tumor-bearing mice, H22+m-HSC (mouse hepatic stellate cell) tumor-bearing mice, and orthotopic H22 cells-bearing mice. The results showed that CAPS-CUR/GA&Gal-Lip exhibited lesser extracellular matrix (ECM) deposition, lesser tumor angiogenesis, and superior antitumor effect compared with the no- and/or Gal-modified Lip, which was attributed to the simultaneous blocking of the activation of HSCs and inhibition of the metastasis of tumor cells. The dual-targeting method using Lip is thus a potential strategy for liver cancer treatment.

Aim: Curcumin has shown promising anticancer activity, which relies on its inhibition on NF-κB pathway. In this study, we characterized the pharmacological profile of a novel curcumin analog P1 and elucidate the related mechanisms. Methods: HEK293/NF-κB cells, stably transfected with an NF-κB-responsive luciferase reporter plasmid, were generated for high-throughput screen (HTS). Eight cancer cell lines, including PC3, COLO 205, HeLa cells etc . were tested. Cell viability was assessed using the sulforhodamine B (SRB) assays. Cell apoptosis was evaluated using FACS, immunocytochemistry, and Western blotting. H 2 -DCFDA and MitoSOX Red were used to detect cellular and mitochondrial reactive oxygen species (ROS). The mitochondrial function was evaluated using mitochondrial oxygen consumption assay. Results: P1, a tropinone curcumin, was found in HTS targeting the NF-κB pathway. Its IC 50 value in inhibition of TNF-α-induced NF-κB activation was 0.8 μmol/L, whereas its IC 50 values in inhibiting the growth of A549 and HeLa cells were 1.24 and 0.69 μmol/L, respectively, which was 20- to 30-fold more potent than curcumin. The inhibition of P1 on the NF-κB pathway was further addressed in HeLa cells. The compound up to 10 μmol/L did not affect the binding of NF-κB to DNA, but markedly inhibited NF-κB nuclear translocation, IκB degradation and IκB kinase phosphorylation. The compound (1 and 3 μmol/L) concentration-dependently induced ROS generation, whereas curcumin up to 20 μmol/L had no effect. P1-induced ROS generation was mainly localized in mitochondria, and reversed by NAC. Moreover, the compound significantly enhanced TNF-α-induced apoptosis. Conclusion: P1 is a novel curcumin analog with potent anticancer activities, which exerts a distinct inhibition on the NF-κB pathway.

The persistent challenges of multidrug resistance and systemic toxicity in cancer chemotherapy necessitate the development of intelligent, tumor-targeted co-delivery systems. Herein we report a novel nickel-based metal-organic framework (CUR + PTX@Ni-MOF) engineered with a phosphazene-derived ligand for the pH-responsive co-delivery of paclitaxel (PTX) and curcumin (CUR). The framework integrates a hexachlorocyclotriphosphazene (HCCP)-based linker featuring an intrinsically acid-labile P3N3 core enabling controlled degradation within the acidic tumor microenvironment (TME). The Ni-MOF exhibits a high BET surface area (177.118 m 2 /g) and a mesoporous architecture facilitating exceptional dual-drug loading capacities of 56.15% for CUR (96.3% encapsulation efficiency, EE) and 26.18% for PTX (43.01% EE). In vitro release studies demonstrated minimal drug leakage at physiological pH (7.4), while triggered, sustained release was achieved under acidic conditions (pH 6.5 and 5.0) with maximum cumulative releases of 75.42% for CUR and 49.35% for PTX at pH 5.0. Cellular assays confirmed high biocompatibility with normal LO2 hepatocytes (>80% viability) and potent, synergistic cytotoxicity against HeLa cancer cells driven by enhanced cellular uptake and apoptosis induction. This work establishes CUR + PTX@Ni-MOF as a promising, intrinsically pH-responsive platform for targeted and synergistic combination chemotherapy.

Achieving stable dispersion and sustained antibacterial activity of natural bioactive compounds in bio-based packaging remains challenging. In this study, chitosan (CS) films incorporating a β-cyclodextrin–curcumin inclusion complex (Cur/β-CD) were developed to improve film properties and the refrigerated preservation of sea bass. The CS/Cur/β-CD films were prepared by one-step solution casting without intermediate isolation or purification. The inclusion conditions were optimized, and the resulting films were evaluated in terms of tensile strength (TS), elongation at break (EAB), and water vapor permeability (WVP). Among the tested formulations, the film prepared at a Cur:β-CD ratio of 1:1, 40 °C, and 1 h (1:1 40 °C 1 h) showed the best overall performance in TS, EAB, and WVP. It was therefore selected for subsequent structural characterization, antibacterial evaluation, and preservation testing. The 1:1 40 °C 1 h film exhibited a 156% increase in tensile strength and a 28.5% decrease in water vapor permeability compared with the neat CS film. The composite film exhibited measurable diffusion-based antibacterial activity againstEscherichia coli(E. coli) andStaphylococcus aureus(S. aureus). During the 8 d refrigerated storage period, the film suppressed total viable counts (TVC), slowed the increase in pH, and retarded total volatile basic nitrogen (TVB-N) accumulation, thereby maintaining acceptable microbiological quality throughout the observation period. Compared with the unwrapped, PE, and CS-film control groups, the treated samples showed better preservation performance over the tested storage period. Overall, the incorporation of Cur/β-CD provides a simple strategy for improving the mechanical strength, moisture barrier properties, antibacterial activity, and preservation performance of CS films during refrigerated storage, highlighting their potential for active packaging of chilled aquatic products.

Diabetic chronic wounds are difficult to heal due to persistent inflammation, excessive oxidative stress, and high infection risk. Herein, a multifunctional GelMA-based microsphere co-loaded with silver ions and curcumin (GelMA-MS@Ag/Cur) was developed as an advanced wound dressing for diabetic wound repair. The porous GelMA microspheres enabled efficient encapsulation and sustained release of Ag + and curcumin while maintaining good biocompatibility and controlled degradation. GelMA-MS@Ag/Cur exhibited broad-spectrum antibacterial activity against both Gram-positive and Gram-negative bacteria and effectively reduced intracellular reactive oxygen species. The microspheres further promoted macrophage polarization toward the pro-regenerative M2 phenotype. Mechanistically, GelMA-MS@Ag/Cur regulated the KEAP1/Nrf2 signaling pathway and suppressed inflammation-induced ferroptosis by modulating ACSL4, SLC7A11, and GPX4 expression, thereby preserving cellular redox homeostasis. In a diabetic rat full-thickness wound model, GelMA-MS@Ag/Cur significantly accelerated wound closure, enhanced angiogenesis, collagen deposition, and tissue remodeling without observable systemic toxicity. This work provides a promising multifunctional strategy for diabetic chronic wound management.

Exposure to extreme heat is associated with acute kidney injury (AKI), and neutrophil extracellular traps (NETs) may contribute to the pathogenesis of AKI under heat stress (HS). Curcumin has demonstrated protective effects on renal function. However, the precise role of NETs and the underlying mechanism by which curcumin confers protection against HS-induced AKI remain unclear. An AKI rat model was established under dry/heat conditions for 1.5 h, followed by recovery periods of 2 h and 24 h at room temperature, respectively. Rats were pretreated with curcumin at doses of 50 or 100 mg/kg prior to HS exposure. Renal injury was assessed by measuring serum creatinine (CREA) and urea nitrogen (UREA) levels, H&E staining, and Tunel assays. Immunofluorescence and ELISA kits were used to quantify NETs levels and inflammation both in vitro and in vivo . The results confirmed that neutrophil infiltration and NETs formation correlated with kidney tissue damage following HS in a dry/heat environment. Treatment with curcumin attenuated HS-induced tubular injury, hemorrhage, and necrosis, and reduced serum UREA and CREA levels. Furthermore, renal levels of inflammatory factors including interleukin (IL)-1β and tumor necrosis factor (TNF)-α, hydrogen peroxide (H 2 O 2 ), myeloperoxidase (MPO) and NETs were significantly decreased after curcumin treatment. In vitro experiments showed that curcumin can inhibit neutrophil activation and suppressed NETs formation by blocking reactive oxygen species (ROS) and MPO production. In summary, curcumin ameliorates dry/heat stress-induced AKI in rats by reducing NETs formation. Moreover, inhibition of NETs may represent a potential therapeutic strategy for mitigating AKI following HS.

This study systematically compared the interactions between gelatins derived from different collagen sources (fish and bovine, corresponding to type I and type II collagen) and curcumin (Cur), focusing on the structural and interfacial properties of the resulting complexes. Four gelatin-Cur complexes (fish/bovine type I and II) were prepared via pH cycling, all achieving encapsulation efficiencies exceeding 80%. Particle size and zeta potential analysis showed that type I gelatin (FGI: 632.51 nm, −18.53 mV; BGI: 664.47 nm, −17.22 mV) had smaller particle sizes and higher absolute zeta potentials than type II gelatin (FGII: 747.13 nm, −16.69 mV; BGII: 764.13 nm, −15.02 mV). Upon complexation with Cur, these gelatin-Cur complexes showed increased interfacial contact angles (80.28°–89.26°) and equilibrium surface tensions (56.28–63.61 mN/m), while the dynamic oil–water interfacial tension continuously decreased and reached equilibrium within 300 s. Fluorescence quenching of gelatin by Cur followed a static mechanism, and complex formation was primarily driven by hydrophobic interactions, hydrogen bonding, and electrostatic forces. The binding affinity (Ka) followed the order: FGI/Cur (7.59 × 10 6 L/mol) > BGI/Cur (7.44 × 10 6 L/mol) > FGII/Cur (6.91 × 10 6 L/mol) > BGII/Cur (6.42 × 10 6 L/mol), consistent with molecular docking results. Cur preferentially localized to the Ser129–Glu135 region of type I gelatin and the Asn179–Ser183 region of type II gelatin. The resulting gelatin-Cur particles effectively stabilized Pickering emulsions. After storage, the TBARS values of gelatin-Cur stabilized emulsions were at least 50.82% lower than those of gelatin-stabilized emulsions. Rheological analysis revealed G' > G'' (elastic-dominated) and shear-thinning behavior, with Cur loading further improving the elastic modulus. These findings provide a theoretical basis for the utilization of by-product gelatin in combination with Cur.

Lignin nanoparticles (LNPs) have attracted increasing attention as sustainable Pickering stabilizers owing to their amphiphilic structure, biocompatibility, and intrinsic antioxidant activity. In this study, LNPs with antioxidant and UV-shielding properties were prepared from bamboo lignin via a p -toluenesulfonic acid/hydrogen peroxide (TPHP) pretreatment and subsequently applied to stabilize oil-in-water (O/W) Pickering emulsions. To regulate particle interactions, NaCl was introduced to adjust the ionic strength. At low salt concentrations, partial screening of electrostatic repulsion promoted reversible association among LNPs, leading to the formation of a weak electrostatic network that enhanced interfacial anchoring and interfacial layer compactness. Under the conditions of 1.0 wt% LNPs and 100 mM NaCl, the emulsion displayed small and uniform droplet sizes, pronounced viscoelastic behavior, and excellent storage stability exceeding 60 days. Using curcumin as a model bioactive compound, the LNPs/NaCl-stabilized system demonstrated high curcumin retention together with improved thermal stability (85.09% retention at 60 °C) and strong UV resistance (88.96% after 72 h). These results highlight the role of the LNPs-mediated electrostatic network in enhancing emulsion stability and protecting sensitive bioactive compounds. This work provides new insights into the design of sustainable lignin-based Pickering emulsions for stabilizing and protecting functional ingredients. Novel camellia oil-based microemulsion for curcumin delivery. • Microemulsion had ∼12 nm droplet size, high uniformity, and superior stability. • It boosted curcumin bioaccessibility 2.6-fold vs. macroemulsion. • Its bioaccessible fraction retained higher antioxidant activity. • Molecular dispersion of curcumin in microemulsion was confirmed. This study compared a camellia oil-based microemulsion (CUR-ME) with a macroemulsion (CUR-MacE) of identical composition to enhance the delivery of curcumin. The CUR-ME exhibited a nanoscale droplet size (∼12 nm), low polydispersity (PDI < 0.1), and superior physical stability. During in vitro digestion, CUR-ME achieved a 2.6-fold higher bioaccessibility of curcumin (79.5%) compared to CUR-MacE (30.4%). The bioaccessible fraction from CUR-ME also demonstrated significantly stronger retention of antioxidant activity in ABTS and FRAP assays. Thermal analysis confirmed the molecular dispersion of curcumin within the microemulsion and revealed compatible interactions with the camellia oil phase. These findings underscore the microemulsion structure as a decisive factor for improving the bioaccessibility and preserving the antioxidant activity of curcumin, offering a superior delivery strategy for functional foods. Fluorescence methods hold great potential for arsenate (As(V)) detection in food safety. However, the development of environmentally friendly and sustainable fluorescence indicators remains at an early stage. Herein, we develop a novel fluorescence sensing platform for As(V) detection. Curcumin (Cur), a plant-derived, non-toxic, and environmentally friendly food additive. Owing to its excellent fluorescent properties, it was used as a green fluorescent indicator in this study. Cerium dioxide nanoparticles (CeO 2 NPs) were synthesized via a green hydrothermal method and then assembled with Cur to form the CeO 2 @Cur fluorescent probe. The detection of As(V) was achieved through an adsorption-desorption mechanism, where As(V) displaces Cur from the CeO 2 surface, thereby restoring its fluorescence. Based on this mechanism, the probe exhibited a linear response (R 2 = 0.991) to As(V) concentrations within the range of 0.10-30.00 μM, with a low detection limit of 25.83 nM. Furthermore, by integrating smartphone-based RGB analysis of fluorescence images, a portable platform was constructed for the on-site detection of As(V). This work thus offers a promising strategy for sustainable and practical solution for As(V) detection in food and environmental samples.

Skin and soft tissue infections (SSTIs) present a significant clinical challenge due to persistent bacterial colonization that exacerbates oxidative stress and impairs tissue repair. To address this, we developed a novel bilayer microneedle system (PI@Cur MN) combining polydopamine (PDA)-stabilized indocyanine green (ICG) at the tips with a Curcumin (Cur)-loaded backing layer, enabling synergistic photodynamic (PDT) and photothermal (PTT) antimicrobial therapy coupled with antioxidant functionality. The PDA-encapsulated ICG nanoparticles (PI) in microneedle tips demonstrated targeted near-infrared (NIR, 808 nm) responsiveness, achieving rapid bacterial eradication through concurrent PTT-induced hyperthermia and PDT-generated reactive oxygen species (ROS), with 99.89 % and 99.74 % reduction against Staphylococcus aureus and MRSA , respectively. Meanwhile, the backing layer containing curcumin remained on the skin surface as a drug reservoir. After the needle was inserted, it enabled a sustained release of Cur, which provided long-lasting antioxidant and anti-inflammatory effects. This multifunctional platform integrates multimodal antibacterial action, oxidative stress alleviation, and minimally invasive drug delivery, offering a clinically promising solution for managing SSTIs and promoting infected wound healing.
