Synthesis and evaluation of Grateloupia Livida polysaccharides-functionalized selenium nanoparticles

Bilang Cao,Qiang Zhang,Juan Guo,Ruixue Guo,Xiaodan Fan,Yongguang Bi

Grateloupia Livida polysaccharides-functionalized selenium nanoparticles (GLP-SeNPs) have been successfully prepared in a simple redox system of sodium selenite and ascorbic acid. The size, morphology, structure, stability and thermal behavior were analyzed by various characterization methods. These results showed that, GLP-SeNPs (particle size of 115.54 nm) prepared in optimal synthesis conditions (temperature of 45 °C, reaction time of 3 h, GLP concentration of 1.0 mg/mL and ascorbic acid concentration of 0.04 M) obtained by orthogonal experiments were uniform spherical and could be stable for 30 days at 4 °C. GLP-SeNPs exhibited significant scavenging ability on DPPH, ABTS, hydroxyl radical and superoxide anion radical when compared to GLP and Na 2 SeO 3 . GLP-SeNPs showed selective cytotoxicity toward various human cancer cells, but not normal cells. Besides, GLP-SeNPs exhibited low oral acute toxicity. Taken together, GLP-SeNPs might be used as potential diet nutritional supplement or anticancer agent.

Rhodiola rosea Rhizome Extract-Mediated Green Synthesis of Silver Nanoparticles and Evaluation of Their Potential Antioxidant and Catalytic Reduction Activities

Daihua Hu,Xu Yang,Wang Chen,Zili Feng,Chingyuan Hu,Fei Yan,Xiaohua Chen,Dong Qu,Zhiyuan Chen

The silver nanoparticles (AgNPs) using the rhizome extract of Rhodiola rosea have been reported. However, their antioxidant activity and whether the biogenic AgNPs could be used to catalyze the reduction of hazardous dye or used as fluorescence enhancers are unknown. This study focused on the facile green synthesis of silver nanoparticles using the rhizome aqueous extract of R. rosea (G-AgNPs). We then studied their antioxidant activity and catalytic degradation of hazardous dye Direct Orange 26 (DO26) and Direct Blue 15 (DB15). Their effects on fluorescein’s fluorescent properties were also evaluated. The chemical AgNPs (C-AgNPs) were synthesized by reducing solid sodium borohydride (NaBH4), and its above activities were compared with those of G-AgNPs. The formation of G-AgNPs was confirmed by the appearance of brownish-gray color and the surface plasmon resonance (SPR) peak at 437 nm. The biogenic AgNPs were approximately 10 nm in size with a regular spherical shape identified from transmission electron microscopy (TEM) analysis. G-AgNPs exhibited significantly improved 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity than butylated hydroxytoluene (BHT) and C-AgNPs (p < 0.05). The biogenic G-AgNPs were also found to function as an effective green catalyst in reducing DO26 and DB15 by NaBH4, which is superior to C-AgNPs. Furthermore, G-AgNPs showed better fluorescence enhancement activity than C-AgNPs, and the concentration required was lower. When the concentration of the G-AgNP solution was 64 nmol/L, the fluorescence intensity reached the maximum of 5460, with the fluorescence enhancement efficiency of 3.39, and the fluorescence activity was stable within 48 h. This study shows the efficacy of biogenic AgNPs in catalyzing the reduction of hazardous dye DO26 and DB15. Biogenic AgNPs could also be used as fluorescence enhancers in low concentrations.

Facile Synthesis of Water-Soluble Fullerene (C60) Nanoparticles via Mussel-Inspired Chemistry as Efficient Antioxidants

Xiaoyan Zhang,Yihan Ma,Sheng Fu,Aiqing Zhang

Rational design and modification of the all-carbon fullerene cages to meliorate their nature of hydrophobicity is critical for biomedical applications. The outstanding electron affinity of fullerenes enables them to effectively eliminate reactive oxygen species (ROS), the excess of which may lead to health hazards or biological dysfunction. Herein reported is a facile, mild, and green approach to synthesizing the favorable water-soluble C60nanoparticles capable of ROS-scavenging by combining the mussel-inspired chemistry with the Michael addition reaction. Various characterization techniques, including Fourier-transform infrared spectroscopy (FTIR), X-ray photoelectron spectra (XPS), thermogravimetric analysis (TGA), transmission electron cryomicroscopy (Cryo-TEM), and dynamic laser scattering (DLS) were carried out to confirm the satisfactory preparation of the hybrid C60-PDA-GSH nanoparticles, which exhibited apparent scavenging capacity of DPPH and hydroxyl radicals in vitro. Additionally, the biocompatible C60-PDA-GSH nanoparticles entered into cells and displayed a universal cytoprotective effect against oxidative press induced by H2O2in four kinds of human cells at a low concentration of 2 μg/mL. The ease and versatility of the strategy present in this work will not only trigger more fullerene-based materials by the immobilization of diverse functional molecules, but will also extend their possible applications.Keywords:water-soluble fullerene nanoparticles;mussel-inspired chemistry;reactive oxygen species scavenging;cytoprotective effect

Crystal-Seeded Growth of pH-Responsive Metal–Organic Frameworks for Enhancing Encapsulation, Stability, and Bioactivity of Hydrophobicity Compounds

Zexun Liu,Qiao Wu,Jie He,Frank Vriesekoop,Hao Liang

Zeolitic imidazolate framework-L (ZIF-L) could effectively improve the stability, controlled release, and anticancer activity of natural hydrophobicity drugs in drug delivery systems (DDSs). A simple and universal strategy was developed to prepare the curcumin-loaded ZIF-L (CCM@ZIF-L) by the antisolvent coprecipitation method, which was different from the traditional approaches. The microcrystal molecules of curcumin were used as the core of ZIF-L growth to form CCM@ZIF-L, which has a very high drug encapsulation efficiency of 98.21% and a regular leaf or cruciate flower-like structure. The formation of CCM@ZIF-L with a distinct composite structure was supported by scanning electron microscopy, transmission electron microscopy, Fourier-transform infrared, powder X-ray diffraction, and zeta-potential. Because of the protective effect of ZIF-L, CCM@ZIF-L exhibited excellent stability and about a 5-fold increase in temperature stability over free curcumin. CCM@ZIF-L exhibited controlled drug release behavior in simulated in vitro tumor microenvironments (almost 81.2% drug release over a period of 72 h). Furthermore, confocal laser scanning microscopy results and cytotoxicity experiments confirmed that the encapsulated curcumin showed a significant improvement in cellular uptake and anticancer activity against A549 cancer cells. Moreover, the curcumin encapsulated in ZIF-L exhibited remarkable cellular antioxidant activity based on MGC-803 cell models. This work presents a novel approach to solve the drug loading problem by employing ZIF-L and exhibits enormous potential of ZIF-L as an effective DDS in cancer treatments.

The extraction and characterization of bioactive compounds from bamboo shoot shell was performed under ultrasonic-assisted extraction (UAE) along with UPLC-Triple-TOF/MS analysis. Under optimized UAE conditions (ethanol concentration (58%), liquid to solid ratio (24 mL/g), extraction temperature (59 °C), and sonication time (29 min)), total phenolic content (TPC) was recorded highest as 85.3 mg GAE/g DW, while DPPH and FRAP antioxidant activities were 77 μmol AAE/g DW and 33 μmol AAE/g DW, respectively. Using UPLC-Triple-TOF/MS analysis, fifteen phenolic acids, seven flavonoids, nineteen organic acids, two iridoid glucosides, and one neoglinan were identified. Among them, p -coumaric acid (119 μg/g DW), chlorogenic acid (87 μg/g DW), rutin (39 μg/g DW), and ferulic acid (17 μg/g DW) were found to be the most abundant phenolic compounds. Furthermore, compounds including phenolic acids (3 -p- coumaroylquinic acid, cryptochlorogenic acid, 3 -O- feruloylquinic acid, 5 -O- feruloylquinic acid, 3 -O- caffeoylshikimic acid, 5 -p- coumaroylquinic acid, 1,3-dicaffeoyl quinic acid, 3,5-dicaffeoyl quinic acid), flavonoids (schaftoside/c-hexosyl-c-pentosylapigenin, apigenin 6,8-di-C-α- l -arabinopyranoside, kaempferide 3 -O- α- l -rhamnopyranosyl(1->6)-β- d -glucopyranoside, 6-C-β- d -glucopyranosyl-8-C-α- l -arabinopyranosylchrysin, narcissin), some organic acids, iridoid glucosides, and neolignan were also detected for the first time from bamboo shoot shell extract. The present study revealed that bamboo shoot shell extract contains a larger number of nutraceutically active compounds which can be further utilized by food and nutraceutical industries.

Isolation and identification of an antioxidant collagen peptide from skipjack tuna (Katsuwonus pelamis) bone

Ding Ding,Bowei Du,Chao Zhang,Fakhar Zaman,Yaqin Huang

To date, many researchers have developed active components that are derived from seafood processing for the purposes of healthcare. Here, an antioxidant collagen peptide was obtained from skipjack tuna (Katsuwonus pelamis) bone by using a combination of trypsin and chymotrypsin as the catalyst. The amino acid sequence of the peptide was identified as Ser–Ser–Gly–Pro–Pro–Val–Pro–Gly–Pro–Met–Gly–Pro–Met–Gly–Pro–Arg (SSGPPVPGPMGPMGPR) by liquid chromatography-electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF-MS) analysis. We found that the as-prepared collagen peptide can efficiently scavenge DPPH radical (IC50 3.149 mM), superoxide anion radical (IC50 3.803 mM) and ABTS radical (IC50 9.489 mM). In addition, it has been found that the methionine (Met) residue in the collagen peptide could provide a precise active site during the scavenging of DPPH radicals by Fourier transform infrared spectroscopy (FTIR) analysis and matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry analysis. These results suggest that the peptide can find wide uses in the food, cosmetic and pharmaceutical industries.

Ziyan Chen,Yan Zhao,Mengke Zhang,Xiufang Yang,Pengxiang Yue,Dikang Tang,Xinlin Wei

A new polysaccharide (pFSP) was first isolated from the originally discarded fibrous roots part of Chinese traditional herb, Bletilla striata. pFSP was composed of D-glucose, D-galactose and D-mannose in a molar ratio of 1: 2.03: 3.45 with molecular weight of 9.1 × 10 4 Da. It could effectively scavenge DPPH and superoxide radicals with inhibition rate of 64.47% and 72.27% at 5.0 mg/ml, higher than that of polysaccharide from Bletilla striata tuber. Structural investigations of the periodate oxidation studies and Smith-degradation as well as the FT-IR spectroscopy were performed, and combined with 1D and 2D NMR spectroscopy, the repeating unit of pFSP contained (1→4)-linked-α-D-Glcp, (1→4)-linked-β-D-Manp and (1→3,6)-linked-β-D-Manp units, together with the branches of (1→6)-linked-β-D-Galp and terminated with (1→)-linked-β-D-Manp residue.

Huang Guojie,Lin Yaqi,Zhang Linxiu,Yan Zhihong,Wang Yudong,Liu Yi

The sulfur-selenium doped carbon quantum dots (S,Se-CQDs) were synthesized by one-step through hydrothermal method in this study, which have high fluorescence quantum yield (43%) and advanced ability to scavenge reactive oxygen species (ROS). They were characterized by transmission electron microscope (TEM), nuclear magnetic resonance (NMR), X-ray photoelectron spectroscopy (XPS), fourier transform infrared spectroscopy (FTIR). The results showed that the clearance rate of free radical reached to 40% with 200 μg/mL of S,Se-CQDs. The antioxidant activity of S,Se-CQDs is related to -SH and Se-SH on carbon quantum dots. S,Se-CQDs were able to access to cells which is beneficial to enhance the removal efficiency to ROS. In the biocompatibility experiment, the cell survival rate exceeded 95%, there was little effect on hatching rate, survival rate and heart rate of zebrafish which demonstrated that S,Se-CQDs have an excellent biocompatibility. It prompts that S,Se-CQDs will has proud application prospects in the field of biomedicine.

Effect of rosemary extract on lipid oxidation of cooked pork during simulated gastric digestion

Shengnan Liu,Rong Zhang,Liumin Fan,Yunfang Ma,Qisen Xiang

BACKGROUND Oxidation of food lipids occurs in the gastrointestinal tract, resulting in potential adverse health effects. Rosemary extract (RE), as one of the most popular naturally sourced antioxidants, is widely used in the food industry. However, the effect of RE on lipid oxidation during gastrointestinal digestion has not been well investigated. Therefore, this study aimed to evaluate the effect of RE on lipid oxidation of cooked pork during simulated gastric digestion. RESULTS Results showed that RE at 12.5, 25, 50, and 100 mg kg −1 pork effectively decreased the formation of malondialdehyde during simulated gastric digestion of cooked pork. RE also effectively mitigated the decline of fatty acids during the simulated gastric digestion of pork. The total phenolic content in RE was calculated to be 170.67 mg gallic acid equivalent (GAE) g −1 . RE dissolved in distilled water (pH 6.5) or potassium hydrogen phthalate–hydrochloric acid buffer solution (0.2 mol L −1 , pH 3.0) both exhibited strong 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activities as well as ferric reducing capacity. The inhibitory effects of RE on lipid oxidation of cooked pork during simulated gastric digestion may be attributed to the phenolic compounds with antioxidant properties. CONCLUSION The results lend support to the possible application of rosemary or RE as a rich source of natural antioxidants to inhibit the oxidation of food lipids during gastrointestinal digestion. © 2019 Society of Chemical Industry

Phloretin as both a substrate and inhibitor of tyrosinase: Inhibitory activity and mechanism

Jianmin Chen,Qinglian Li,Yaling Ye,Ziyao Huang,Zhipeng Ruan,Nan Jin

Tyrosinase is the rate-limiting enzyme for controlling the production of melanin in the human body, and overproduction of melanin can lead to a variety of skin disorders. In this paper, the inhibitory kinetics of phloretin on tyrosinase and their binding mechanism were determined using spectroscopy, molecular docking, antioxidant assays and chromatography. The spectroscopic results indicate that phloretin reversibly inhibits tyrosinase in a mix-type manner through a multiphase kinetic process with the IC 50 of 169.36 μmol/L. It is shown that phloretin has a strong ability to quench the intrinsic fluorescence of tyrosinase mainly through a static quenching procedure, suggesting that a stable phloretin-tyrosinase complex is generated. Molecular docking results suggest that the dominant conformation of phloretin binds to the gate of the active site of tyrosinase. Moreover, the antioxidant assays demonstrate that phloretin has powerful antioxidant capacity and has the ability to reduce o -dopaquinone to l -dopa just like ascorbic acid . Interestingly, the results of spectroscopy and chromatography indicate that phloretin is a substrate of tyrosinase but also an inhibitor. The possible inhibitory mechanism is proposed, which will be helpful to design and search for tyrosinase inhibitors.

Construction of cellulose-based Pickering stabilizer as a novel interfacial antioxidant: A bioinspired oxygen protection strategy

Qi Li,Pan Chen,Yan Li,Bin Li,Shilin Liu

Oxygen protection/isolation is imperative to prevent the lipid oxidation since oxygen molecule is an ultimate quencher in photon conversion process. Inspired by the structural buildup of seeds from oil crops, a sustainable solid particle stabilizer with novel antioxidant activity was prepared by using cellulose and polyphenol. In this work, bacterial cellulose (BC) nanofibrils modified by tea polyphenols (TPs) was prepared and used as Pickering emulsifier for the O/W emulsion. BC nanofibirls exhibited excellent adsorption capacity up to 55 μg/mg, and the adsorption kinetics between BC and TPs were further investigated. After modification, the interfacial diffusion rate constant of BC was significantly increased to from 0.43 to 1.21 mN m −1  s −0.5 . Moreover, the obtained O/W interfacial modulus of the dilatational elasticity was increased from 58 to 130 mN/m. Furthermore, the emulsions exhibited excellent free-radical scavenging activity at oil-water interface, suggesting a potential application in usage to extend the lifespan of the food containing polyunsaturated fats.

Structural characterization and antioxidant activity of polysaccharide from four auriculariales

Yue Su,Liang Li

The structural characterization and antioxidant activity of four Auricularia polysaccharides (A.cornea ACP)、A.auricula(AAP)、A.polytricha (APP) and M.fungus(MFP)) were studied in this paper. The results shown: polysaccharides of four Auricularia were mainly composed of mannose and galactose, all polysaccharides contained uronic acid and pyran ring structure with spectroscopy and NMR analysis. There was a significant difference in the total antioxidant capacity and APP was significantly higher than the other polysaccharides. The ability of APP to scavenge DPPH radicals and hydroxyl radicals was significantly higher than that of other polysaccharides, respectively. The molecular weight was significantly positively correlated with DPPH radicals, superoxide anion radicals and hydroxyl radicals. Total antioxidant capacity was significantly negatively correlated with fucose and galactose. The result indicated that fucose and galactose jointly determine total antioxidant capacity. The polysaccharide from four Auricularia had good oxidation resistance and could be used as natural antioxidants.
