Background Resistance to androgen receptor signaling inhibitors, including enzalutamide, remains a major obstacle in treating castration-resistant prostate cancer (CRPC). FOXA1, a pioneer transcription factor frequently overexpressed or mutated in prostate cancer, drives disease progression and represents a potential therapeutic target. Methods Through virtual screening, we identified tigecycline, an FDA-approved antibiotic, as a candidate compound that directly targets FOXA1. We assessed its effects on FOXA1 target gene expression and chromatin occupancy using chromatin immunoprecipitation assays in C4‑2 cells. The impact on FOXA1 mutation-driven progression was evaluated using C4‑2 cells stably expressing a FOXA1 point mutant. In vivo efficacy was examined in CRPC xenograft models. Results Tigecycline inhibited FOXA1 target gene expression and reduced FOXA1 chromatin occupancy. In FOXA1-mutant cells, tigecycline repressed mutant-driven gene expression, suppressed AR signaling activity, and decreased PSA levels, reversing the oncogenic effects driven by the mutant. Notably, tigecycline retained efficacy in FOXA1-mutant cell lines, likely through direct binding to FOXA1. In vivo, tigecycline suppressed CRPC tumor growth and reduced tumor PSA levels. Conclusion These findings support tigecycline as a potential therapeutic agent for CRPC by targeting FOXA1.

With the development of advanced electronic packaging technologies, higher requirements have been placed on the service reliability of low-temperature solders. However, Bi-rich phase coarsening and excessive growth of interfacial intermetallic compound (IMC) in Sn58Bi solder restrict its engineering applications. In order to improve the microstructure and mechanical reliability of Sn58Bi solder, Ag-coated ZnO (ZnO@Ag) nanoparticles were successfully fabricated by electroless silver plating and Sn58Bi-xZnO@Ag (x = 0, 0.3, 0.5, 0.7, and 1.0 wt.%) composite solders were prepared by ball milling. The effects of ZnO@Ag addition on the wettability of solder joints, matrix microstructure, interfacial IMC growth, and shear properties were systematically investigated. The results showed that the addition of 0.3 wt% ZnO@Ag exhibited the optimal strengthening effect. Compared with the unreinforced Sn58Bi solder, the wetting angle decreased from 39.18° to 36.32°, the Bi-rich phase became finer with the number fraction of fine Bi-rich phase regions (0.1 < A ≤ 1 μm 2 ) reaching 31.78%. Meanwhile, the average thickness of the interfacial IMC decreased from 0.85 μm to 0.67 μm. Ultimately, the shear strength of the solder joints increased from 51.49 MPa to 69.62 MPa. The performance improvement may be attributed to the synergistic effects of heterogeneous nucleation, grain-boundary pinning, and interfacial diffusion blocking induced by the ZnO@Ag nanoparticles. This study demonstrates that the appropriate addition of ZnO@Ag is an effective approach for improving the reliability of Sn58Bi low-temperature solder, which can provide a reference for the design of high-reliability low-temperature lead-free composite solder.

Natural fiber reinforced polylactic acid (NF/PLA) composites are attractive sustainable materials, yet their broader engineering use is limited by weak interphase load transfer and rapid, dripping combustion. A lignin migration driven interphase phosphorylation strategy was proposed to construct a continuous phosphate-rich interphase on reed fibers, enabling simultaneous reinforcement and flame retardancy in PLA. A mild and efficient solvent activation step induces in- situ lignin deposition and generates a reactive, adhesive fiber surface, followed by aqueous phosphorylation to covalently introduce phosphate functionalities. Crucially, this strategy preserves the intrinsic structural integrity of the macroscopic fibers while simultaneously tailoring the interface for enhanced adhesion and flame retardancy. As a result, the reconstructed interphase strengthens fiber matrix adhesion and promotes condensed phase charring while improving melt resistance to dripping. At a 30 wt% modified fiber loading, the composite reaches a limiting oxygen index of 33.0% and achieves a UL-94 V-0 rating with no cotton ignition. Cone calorimetry shows 38% and 27% reductions in peak heat release rate and total heat release relative to neat PLA. Meanwhile, tensile strength increases by about 15% and notched impact toughness by about 270%, evidencing effective load transfer. The lignin enriched, phosphate functionalized interphase synchronizes rapid melt stabilization with the formation of a compact protective char, thereby reconciling reinforcement with fire safety. This scalable interphase-engineering strategy offers a sustainable route to high performance PLA composites for transportation, electronics, packaging, and related applications.

CO is a key product of CO 2 electroreduction (CO 2 ER) and an important feedstock for the production of hydrocarbons and alcohols. Herein, we synthesized electrocatalysts by growing Cu nanocluster on manganese oxide (Cu n -Mn 3 O 4 ) for CO 2 ER. In 0.5 M KHCO 3 electrolyte using an H-cell, the optimized Cu 0.4 -Mn 3 O 4 achieved a Faradaic efficiency for CO (FE CO ) of 88.5 ± 2.3% and a current density of 14.3 ± 0.2 mA cm −2 at − 0.6 V vs. RHE, while substantially suppressing the hydrogen evolution reaction (HER). The better performance was due to the electrons transferred from Mn to CO 2 by Cu as a shuttle and anchored Cu nanoclusters shifted the d-band center toward the Fermi level, which was more conducive to the intermediate formation. In situ electrochemical attenuated total internal reflectance (EC-ATR) spectroscopy and density functional theory (DFT) calculations revealed the *COOH intermediate (1399 cm⁻ 1 ) along the CO 2 → *COOH → *CO → CO pathway. Moreover, the DFT calculations further demonstrated the Gibbs free energy barrier (ΔG) of the key steps in the CO 2 ER pathway and the corresponding intermediate configurations, which were used to reveal the relationship between the catalyst structure and CO 2 ER performance. The design of the metal/metal oxide electrocatalyst in this study provides a new perspective for the highly selective reduction of CO 2 to CO. Graphical

Nucleic acid testing is important for infectious-disease diagnosis, but combining high sensitivity with accessible testing remains challenging. Here we show a chip-based visual detection strategy for one or two viral RNA targets that involves only sample loading, without further instrumentation or manual actuation. Target-activated CRISPR-Cas13a alters the wettability of a molecular barrier, causing liquid to cross a preset threshold and generate a dam-break drainage signal. The chip detects a single target at 10 aM within 2 min and two targets at 100 aM within 5 min, with costs of $0.20 and $0.30 per test, respectively. We further establish a practical clinical-sample workflow combining room-temperature lysis with RNase inhibition and validate the platform across 40 nasal-swab samples tested for SARS-CoV-2, influenza A virus and influenza B virus, with results concordant with RT–PCR. This threshold-based fluidic strategy provides a route towards rapid, low-cost and instrument-free molecular testing outside centralized laboratories.

The pyrolysis of energetic materials (EMs) directly determines their combustion performance and energy release efficiency. This study designed a potassium-doped maghemite (KF) catalyst with sea urchin-like structure. Traditional lead-based catalyst could cause serious environmental pollution, whereas KF may offer dual advantages of flame suppression and high catalytic activity. Electron paramagnetic resonance (EPR) spectroscopy demonstrate the increased defect concentration compared with commercial γ-Fe 2 O 3 and α-Fe 2 O 3 . The as-prepared KF catalyst provides more surface active sites, significantly enhancing its catalytic performance for the thermal decomposition of hexanitrohexaazaisowurtzitane (CL-20). Graphene oxide (GO) always acts as a stabilizer for EMs due to the excellent mechanical properties. Hence, K-γ-Fe 2 O 3 /reduced graphene oxide (KFG) was prepared via the one-step solvothermal method. Experimental results show that the ternary KFG composite can lower both the initial decomposition temperature and the apparent activation energy of CL-20. KFG accelerates the decomposition process of CL-20 and narrows the distribution of gaseous products. This study reveals the synergistic mechanism among potassium, γ-Fe 2 O 3 , and graphene, providing new design strategy and theoretical foundation for highly efficient combustion catalysts applied in solid propellants.

An S-scheme Ag/CdS/Fe 2 O 3 (Ag/CS/FO) heterojunction was fabricated via a sequential solvothermal and photodeposition method. The microstructure and optoelectronic properties of the synthesized ternary composite were verified by a series of characterization methods. The transfer of photogenerated electron–hole pairs (e − /h + pairs) follows an S-scheme pathway, driven synergistically by the interfacial electric field (IEF) and band bending. That directs electrons (e − ) to the conduction band (CB) of CdS (CS) and holes (h + ) to the valence band (VB) of Fe 2 O 3 (FO), achieving efficient e − /h + pairs separation while maintaining high redox capabilities. Additionally, the incorporated Ag nanoparticles enhance visible-light harvesting via the surface plasmon resonance (SPR) effect and function as electron-transfer bridges, collectively promoting charge migration. Consequently, the composite exhibits high photocatalytic activity, degrading 98.68% of amoxicillin (AMX) under visible light. It also maintains 91.6% efficiency after four cycles, confirming sound stability. Notably, the material demonstrates superior performance in real-life water matrices (tap and river water), highlighting its strong application potential. Graphical abstract

A set of phosphomolybdic acid catalysts was synthesized by employing ligand-functionalized UiO-66 as the support. The structural properties of these catalysts were characterized via XRD, FT-IR, BET, TEM, and XPS techniques. To explore the influence of catalyst structure on its catalytic performance, the desulfurization of dibenzothiophene (DBT) from simulated fuel was utilized as a model reaction. Under optimized reaction parameters, the Co-POM-UiO-66-NO 2 -F catalyst exhibited the highest DBT conversion efficiency, achieving 99.3%. After five consecutive desulfurization cycles, the catalyst retained a desulfurization activity of 91.8%. This excellent stability was attributed to the incorporation of the –F group, which enhanced the catalyst’s lipophilicity and hydrophobicity. Additionally, the incorporation of –F groups increased the content of the Mo active species (Mo 5+ ), regulated the electronic structure of the catalyst, and optimized the microenvironment surrounding the active centers, which collectively contributed to the enhanced catalytic performance. Sensitive and specific detection of luteinizing hormone (LH) is critical for the early diagnosis of diseases and cancers, offering valuable insights for clinical strategies aimed at regulating physiological functions to maintain health. To this end, we developed a novel sensing platform for the ultrasensitive detection of LH, harnessing a mutually orthogonal cascade amplification composed of DNAzyme and self-replicating catalytical hairpin assembly (SR-CHA). Upon binding of LH to its aptamer, an LH/aptamer complex is formed, leading to the release of an initiator strand (“strand A”) that triggers the assembly of Y-shaped DNA structures. Each Y-shaped structure comprises a modular DNAzyme-based amplification unit, which generates a one-to-many fluorescence signal, and an initiator mimic sequence capable of self-replicating to produce numerous new initiators for accelerating the formation of additional Y-shaped structures, thereby amplifying the detection signal. This synergistic design resulted in outstanding analytical performance, achieving a detection limit as low as 0.00038 mIU/mL in buffer and 0.0006 mIU/mL in real samples, with a broad dynamic range from 0.001 to 100 mIU/mL. In addition, the proposed method also demonstrated excellent selectivity, and a strong correlation with the standard ELISA method, which reveals its promising potential as a universal simple and sensitive methodology for the construction of various aptamer-based bioassays. Objective Primary sclerosing cholangitis (PSC) is a progressive cholestatic liver disease lacking FDA-approved therapy. Calculus Bovis (CB), a traditional medicine derived from animal gallstones, has been historically used for treating hepatobiliary diseases, but its therapeutic potential and mechanisms in PSC remain unexplored. This study aimed to investigate the efficacy of CB in an experimental PSC model and elucidate its underlying mechanisms. Methods A PSC mouse model was induced by a 0.1% 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC) diet for 4 weeks. Mice were treated with CB (50,100, 150 mg/kg/day) or ursodeoxycholic acid (UDCA, 100 mg/kg/day). Liver injury, fibrosis, intestinal barrier integrity, bile acid (BA) profiles, and lipid levels were assessed. Hepatic and intestinal gene/protein expression related to BA and lipid metabolism was analyzed. Integrated transcriptomics, network pharmacology, and in vitro serum pharmacology were employed to elucidate the underlying mechanisms. Results CB administration significantly alleviated liver injury, fibrosis, and intestinal barrier damage in DDC-induced mice. It restored BA homeostasis across the gut-liver axis, normalizing aberrant BA profiles in serum and liver while increasing BA excretion in feces. CB also ameliorated dyslipidemia, reducing hepatic and serum lipid levels. Mechanistically, CB and its bioactive BA components exerted their effects through a dual-pronged mechanism: (1) activation of the SIRT1-PGC-1α axis to transcriptionally upregulate the expression of nuclear receptors FXR and PPARα in the liver and intestine, and (2) direct ligand-dependent activation of FXR and PPARα protein functions. This concerted activation enhanced the transcription of genes involved in BA detoxification, transport, and fatty acid β-oxidation. Inhibition of SIRT1 or antagonism of FXR/PPARα attenuated these protective effects in vitro . Conclusion CB attenuates experimental PSC by modulating BA and lipid homeostasis via the gut-liver axis, mediated through a novel dual mechanism involving SIRT1-PGC-1α pathway activation and direct receptor agonism. These findings not only highlight CB as a promising multi-target agent for PSC treatment, but also provide novel insights into the therapeutic modulation of metabolism in the gut-liver axis.

Photocatalysts have offered a successful strategy for degrading organic pollutants, such as various dyes and antibiotics, in wastewater. In this study, the flower-like Bi 2 MoO 6 balls were impregnated on the zirconium-based MOFs (MOF-801) to construct the Bi 2 MoO 6 /MOF-801 heterojunction composite photocatalytic materials through a simple one-pot hydrothermal method. The systematically characterization of the as-synthesized composite materials was performed using various analytical techniques, including XRD, FTIR, SEM-EDS, N 2 adsorption⁃desorption testing, TG, XPS, UV-Vis DRS, PL, transient photocurrent, and EIS techniques, thereby confirming the successful impregnation of Bi 2 MoO 6 into MOF-801. The degradation effect of the Bi 2 MoO 6 /MOF-801 composites on rhodamine B (RhB) dye was investigated, along with the photocatalytic degradation mechanism. Results showed that 0.1-Bi 2 MoO 6 /MOF-801 has better degradation efficiency for RhB dye than that of pure Bi 2 MoO 6 and MOF-801 under visible light conditions, with a complete degradation efficiency of 98.1% achieved within 40 min. The boosted photocatalytic performance of the 0.1-Bi 2 MoO 6 /MOF-801 composite is attributed to their high specific surface area (529.2 m 2 /g) and mesoscale pore size (6.9 nm), wide visible spectral absorption, well-optimized band gap (2.81 eV), and the close contact of Bi 2 MoO 6 and MOF-801 ensuring efficient charge separation and suppressing the recombination of photogenerated electron-hole pairs. Visible-light driven photocatalysis of 0.1-Bi 2 MoO 6 /MOF-801 composite further confirmed superior degradation of MB, AO, CR, MO, and ARS dyes, reaching degradation efficiencies up to 99.7%. Moreover, the results of the free radical capture experiments confirmed that the holes ( h + ) and superoxide radicals (•O 2 − ), and electrons ( e − ) were the primary active substances during the degradation process, supporting the Z-scheme heterojunction charge transfer mechanism. This research study will point the direction for designing potential and affordable MOF-based heterojunction materials for the treatment of dye-contaminated wastewater. Graphical

As popular food packaging materials in the biodegradable plastic market, plant-fiber/PLA composites (PFPCs) require comprehensive evaluation to ensure their safety. In this work, semi-volatile organic compounds migrated from three batches of PFPC were characterized via gas chromatography coupled with quadrupole time-of-flight mass spectrometry. A total of 44 compounds were identified in the 4% (v/v) acetic acid food simulant and 95% (v/v) ethanol food simulant. Ten of them were authorized, but the eugenol migration exceeded the limit of detection in GB 9685–2016. The risk of the other 34 compounds were assessed according to the threshold of toxicological concern. Among them, 21 compounds were assessed as Cramer rule I, and the rest were assessed as Cramer rule III. Ethyl 5,6-dimethoxyindole-2-carboxylate, furfural, tributyl citrate, and caffeine have the greatest potential risks among Cramer class III substances. To systematically investigate the cumulative migration patterns of potentially hazardous compounds, 20 migration tests were conducted to evaluate the risk associated with repeated use of PFPC. As a result, the concentrations of Cramer class III substances in 95% (v/v) ethanol were greater than those in 4% (v/v) acetic acid after repeated migration tests, indicating that the samples presented greater health risks when they were in contact with food containing alcohol or grease than when they were in contact with acidic food.
