Lipase Immobilization onto the Surface of PGMA-b-PDMAEMA-grafted Magnetic Nanoparticles Prepared via Atom Transfer Radical Polymerization

Jingyun Wang,Fangling Ji,Jishuang Xing,Shuang Cui,Yongming Bao,Wenbo Hao

A block copolymer of 2-dimethylaminoethyl methacrylate (DMAEMA) and glycidyl methacrylate (GMA) was grafted onto the surface of magnetic nanoparticles (Fe 3 O 4 ) via atom transfer radical polymerization . The resultant PGMA-b-PDMAEMA-grafted-Fe 3 O 4 magnetic nanoparticles with amino and epoxy groups were characterized by Fourier transform infrared spectroscopy, powder X-ray diffraction, thermo-gravimetric analysis, and scanning electron microscopy. Lipase from Burkholderia cepacia was successfully immobilized onto the magnetic nanoparticles by physical adsorption and covalent bonding. The immobilization capacity of the magnetic particles is 0.5   mg lipase per mg support, with an activity recovery of up to 43.1% under the optimum immobilization condition. Biochemical characterization shows that the immobilized lipase exhibits improved thermal stability, good tolerance to organic solvents with high lg P , and higher pH stability than the free lipase at pH   9.0. After six consecutive cycles, the residual activity of the immobilized lipase is still over 55% of its initial activity.

In situ synthesized rGO–Fe3O4 nanocomposites as enzyme immobilization support for achieving high activity recovery and easy recycling

Dong Yang,Xueyan Wang,Jiafu Shi,Xiaoli Wang,Shaohua Zhang,Pingping Han,Zhongyi Jiang

Herein, the reduced graphene oxide–Fe 3 O 4 (rGO–Fe 3 O 4 ) nanocomposites are synthesized by the simultaneous reduction of graphene oxide (GO) and in situ deposition of Fe 3 O 4 nanoparticles ( ca. 20   nm) enabled by Fe 2+ ions. The rGO–Fe 3 O 4 nanocomposites integrate the magnetic property of Fe 3 O 4 nanoparticles and the large specific surface area of rGO nanosheets. Catalase (CAT), as a commonly used enzyme, can be efficiently immobilized on the rGO–Fe 3 O 4 nanocomposites through physical adsorption. The CAT loading capacity is as high as 312.5   ±   12.6   mg   g −1 , while the activity recovery of CAT can be high up to nearly 98%. The strong magnetic response of immobilized CAT ensures its easy separation from the reaction system when an external magnetic field is applied. Owing to the hydrophobic and hydrogen bonding interactions between enzyme and the support, the immobilized CAT exhibits zero leaching, desirable stability and excellent reusability.

Superparamagnetic Fe3O4 nanoparticles modified by water-soluble and biocompatible polyethylenimine for lipase immobilization with physical and chemical mechanisms

Weiwei Zhu,Yijing Li,Fang Zeng,Hang Yin,Liyuan Wang,Hao Zhu

Superparamagnetic Fe3O4 nanoparticles with diameter about 18 nm were prepared by a solvothermal method. A simple and green method was used to prepare amino-rich magnetic nanoparticles. The magnetic nanoparticles were modified by polyethylenimine (PEI) which is a polycationic polymer when pH < 10. The resulted magnetic nanoparticles were activated by glutaraldehyde to obtain nanoscale support for Candida rugosa Lipase (CRL) immobilization. The CRL was immobilized on the magnetic nanoparticles covalently, as well as via ionic exchange. The structure and magnetic behavior of the magnetic nanoparticles were confirmed by transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR), and vibrating sample magnetometer (VSM). Then the properties of the immobilized CRL were investigated, and the results showed that the obtained immobilized lipase displayed good reusability and applicability. The immobilized Candida rugosa Lipase (ICRL) presented wider pH tolerance (residual activity > 80% in a pH range from 5.5 to 8.0) than free Candida rugosa Lipase (residual activity decreased rapidly when the pH values were away from 7.0). ICRL showed excellent thermal stability (the relative of ICRL > 90%) after keeping in the water bath at 50 °C for 150 min while free Candida rugosa Lipase (FCRL) was absolutely deactivated. After being reused 10 times, ICRL maintained 60% relative activity.

Retracted Article: 3D Small molecule microarray with enhanced sensitivity and immobilization capacity monitored by surface plasmon resonance imaging

Vikramjeet Singh,Jinsong Zhu,Amita Nand,Zhiqiang Cheng,Yang Mo

Here we report the 3D photo-cross-linking strategy based on the use of α-cyclodextrin as an alternative surface chemistry for the preparation of small molecule microarray (SMMs). Surface plasmon resonance imaging (SPRi) was used in combination with SMMs as a detection technology. Small molecules of diverse structure containing different functional groups can be covalently captured on a single slide and displayed in various orientations in a given spot. Several known small molecules-protein complexes were selected to validate this platform. Furthermore, a comparison between 2D PEG and 3D α-cyclodextrin photo-cross-linking surface was performed using fluorescence and SPRi techniques. Data produced from both type of surfaces strongly favors the better performance of 3D over 2D surface in terms of loading capacity, spot morphology, signal response, and kinetics values. We believe that this strategy will expand the utility of small molecule microarray for high throughput screening against a variety of target proteins.

Effects of biochar pyrolysis temperature on uranium immobilization in soil remediation: Revealed by 16S rDNA and metabolomic analyses

Huachuan Feng,Yilin Wang,Nan Li,Zishu Qian,Tao Chen,Xiaoming Chen,Qing Wang,Wenkun Zhu

Uranium-stressed soil caused by nuclear industry development and energy acquisition have attracted extensive attentions for a long time. This study investigated the effects of biochar application with different pyrolysis temperatures (300 ℃, 500 ℃ and 700 ℃) on remediation of uranium-stressed soil. The results showed that higher pyrolysis temperature (700 ℃) was benefit for ryegrass growing and caused a lower uranium accumulation in plants. At the same time, uranium immobilization was more effective at higher biochar pyrolysis temperature. Careful investigations indicated that activities of soil urease and sucrase were promoted, and bacterial diversity was strengthened in C700 group, which may contribute to uranium immobilization. The biochar application could activate metabolic of lipids and amino acids , organic acids and derivatives, and organic oxygen compounds. Nicotinate and nicotinamide metabolism, and Benzoxazinoid biosynthesis were unique metabolic pathways in the C700 group, which could enhance the uranium tolerance from different perspectives. Based on these results, we recommend to use biochar with 700 °C pyrolysis temperature when processing remediation of uranium-stressed soil. This study will facilitate the implementation of biochar screening and provide theoretical helps for remediation of uranium-stressed soil.

Effect of sintering temperature on crystal growth and evolution of CaZrTi2O7–CaNdTiNbO7 ceramics: Minor actinides immobilization host materials

Shiyin Ji,Xiuhua Ji,Yanhao Song,Yanxia Wei,Tao Duan

Regulation of microstructure and processing cost of waste form are important for the design and application of high-level radioactive waste immobilization. In this study, CaZrTi 2 O 7 (zirconolite) – CaNdTiNbO 7 (pyrochlore) ceramics were synthesized via conventional sintering method at 1000–1500 °C to investigate the effect of sintering temperature on the microstructure, solidification, and crystal evolution. The powder x-ray diffraction (XRD) and backscattered scanning electron microscopy–energy dispersive spectroscopy (BSEM–EDS) results indicated gradual formation of zirconolite and pyrochlore ceramics, following with their densification when increasing sintering temperature. It was found that sintering temperature should be higher than 1300 °C to form zirconolite-pyrochlore solid solutions. Densified pyrochlore was obtained at 1500 °C. In addition, transformation from relatively low-symmetry lattice to higher-symmetry lattice was found with an increase in sintering temperature. Furthermore, higher temperature can enhance the crystal growth and densification in the ceramic composite. The finding in this study could provide important suggestions to design and process zirconolite-pyrochlore waste forms for industrial application.

Functionalization of biochar using SDS/SAP nanomicelles enhanced its immobilization capacity for dyes and heavy metals in water

Tian Kun,Li Chunping,Liu Huiming,Wang Lianchun

To enhance the adsorption capacity of biochar (BC), herein a novel multifunction modified biochar (SDMBC) was prepared by directly crosslinking of the nanomicelle of sodium dodecyl sulfate/sapindus-saponin (SDS/SAP) composite system onto the BC through a simple, environmental friendly approach. Result showed that the adsorption performance of SDMBC has been greatly improved, compared with BC or using alone SDS and SAP, adsorption ability increased by 48.83%, 29.50%, 36.44%, respectively, the best modified effect was appeared when the concentration of SAP to SDS was 0.8 and 0.8 CMC. SDMBC exhibited high adsorption abilities of 130.23, 108.43, 277.09 125.27, 112.78 mg/g for heavy metal ions lead Pb(II), Cadmium Cd(II) and organic pollutants with different chemical properties bisphenol A(BPA), Methylene blue (MB), P-nitrophenol (PNP), respectively, higher than most previously reported adsorbents, importantly, SDMBC can still efficient removal capabilities even in the binary competition. Subsequently, the SDMBC and BC was characterized by Fourier Transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), Zeta potential (Zeta), it found SDMBC has a more layered structure, richer functional groups and more amorphous structure compared with BC, which are closely related with improving its adsorption capacity. The adsorption behavior of SDMBC for MB show that process was found to be spontaneous, propitious, endothermic, the adsorption isotherms fitted Freundlich models well, pseudo-second-order best describes kinetics adsorption, suggesting that the process is multi-layer chemical adsorption. The little affected by ionic strength and coexisting substances, could remained removal rate over a wide pH range, SDMBC still keep high removal rates even after 5 reuses. Based on FT-IR analysis, plausible adsorption mechanism proposed, including hydrogen bond, electrostatic attraction and π-π bonding. Cost analysis manifests that the SDMBC are high efficiency and cheap eco-adsorbents compared with commercial activated carbon, and the SDMBC dosage required for the removal of 99% of a fixed amount of MB in different volumes of effluent was predicted. Seven machine learning (ML) models were used to predict the MB (60 mg/L) adsorption of the SDMBC, using Shapley Additive Explanations (SHAP) for model interpretation. Finding Extreme Gradient Boosting (XGBoost) exhibited best performance, the order of feature importance as time> Ratio> pH> concentration> temperature. Thus, SDMBC as a new cheap and eco-adsorbents, can be used to effectively remove various types of pollutants, has a great application potential in sewage treatment, while the accurate ML prediction model presented a valuable advice for designing efficient adsorbents and optimization operating conditions in the future.

Immobilization nitrogen-doped sodium alginate-derived carbon quantum dots on sodium alginate hydrogel for enhanced Pb(II) removal

Yu Li,Yuxin Xu,Jiayu Zhu,Xianghui Meng,Shuang Ai,Meiling Zhang,Fan Yang,Huabing Yin,Kui Cheng

Heavy metal contamination, particularly from Pb(II), poses severe environmental and health risks due to its toxicity and persistence in ecosystems. This study presents an innovative and sustainable adsorbent composed of nitrogen doped carbon quantum dots (NCDs), derived from sodium alginate and immobilized on a sodium alginate hydrogel (HL-NCDs), for the efficient extraction of Pb(II) from water. The experimental results indicate that Pb N and Pb C interactions, particularly involving the N H moieties within the carbon quantum dots, are the predominant mechanisms facilitating Pb(II) removal, as confirmed by density functional theory (DFT) calculations. These interactions are substantiated by thermodynamic analyses and charge distribution studies, which highlight the enhanced affinity of HL-NCDs towards Pb(II) ions. The novel HL-NCDs composite material not only demonstrates excellent Pb(II) adsorption efficiency but also suggests broad applicability in water treatment technologies, marking a significant advancement in the use of biomass-derived materials for environmental remediation.

Removal of chromium in electroplating wastewater by nanoscale zero-valent metal with synergistic effect of reduction and immobilization

Zhanqiang Fang,Xiuqi Qiu,Ruixiong Huang,Xinhong Qiu,Mingyu Li

Reductive immobilization of chromium in wastewater by nanoscale zero-valent metal (nZVM) prepared from steel pickling waste liquor was investigated. A series of characterization techniques were conducted to characterize the properties of the synthesized nanoparticles . Effects of pH, buffer substances, natural organic matter (NOM) and hardness were evaluated to probe the impact of environmental factors. Removal of Cr(VI) benefited from a decrease in pH value and the presences of buffer substances and Ca 2+ . NOM was found to be suppressive on the activity of ZVM, and coupled effect of Ca 2+ and NOM was additive effect due to their non-interference when operating under acidic condition. The nZVM was proved to be more effective (up to 40.6-fold) than zero-valent iron and nanoscale zero-valent iron for Cr(VI) removal, and its removal capacity in wastewater was 182   ±   2   mg   g −1 . The removal mechanism included mass transfer on solid–liquid interface, reduction of Cr(VI) and simultaneous co-precipitation as Cr–Fe (oxy)hydroxide on the surface of nanoparticles. The results presented herein indicated nZVM was promising for in-situ remediation of Cr(VI).

Sequential Co-immobilization of Enzymes in Metal-Organic Frameworks for Efficient Biocatalytic Conversion of Adsorbed CO2 to Formate

Li Yan,Wen Liyin,Tan Tianwei,Lv Yongqin

The main challenges in multienzymatic cascade reactions for CO2 reduction are the low CO2 solubility in water, the adjustment of substrate channeling, and the regeneration of co-factor. In this study, metal-organic frameworks (MOFs) were prepared as adsorbents for the storage of CO2 and at the same time as solid supports for the sequential co-immobilization of multienzymes via a layer-by-layer self-assembly approach. Amine-functionalized MIL-101(Cr) was synthesized for the adsorption of CO2. Using amine-MIL-101(Cr) as the core, two HKUST-1 layers were then fabricated for the immobilization of three enzymes chosen for the reduction of CO2 to formate. Carbonic anhydrase was encapsulated in the inner HKUST-1 layer and hydrated the released CO2 to HCO3-. Bicarbonate ions then migrated directly to the outer HKUST-1 shell containing formate dehydrogenase and were converted to formate. Glutamate dehydrogenase on the outer MOF layer achieved the regeneration of co-factor. Compared with free enzymes in solution using the bubbled CO2 as substrate, the immobilized enzymes using stored CO2 as substrate exhibited 13.1-times higher of formate production due to the enhanced substrate concentration. The sequential immobilization of enzymes also facilitated the channeling of substrate and eventually enabled higher catalytic efficiency with a co-factor-based formate yield of 179.8%. The immobilized enzymes showed good operational stability and reusability with a cofactor cumulative formate yield of 1077.7% after 10 cycles of reusing.

PDA-assisted immobilization of nZVI on cotton fabric for the removal of rhodamine B and chromium (VI) ion

Liu Lei,Hu Yufan,Yang Yuhui,Cheng Hongbin,Xiao Shili

Printing and dyeing wastewater can cause serious environmental pollutant and must be treated to meet the emission standards before discharge. In this study, we report a method to prepare nano zero-valent iron (nZVI) particle-immobilized cotton (nZVI@cotton) fabrics assisted by polydopamine (PDA) deposition using cotton fabrics as the substrate. The mechanical property tests of cotton fabric revealed a significant improvement in the breaking strength after deposition of PDA. The pollutant removal capability of nZVI@cotton fabric was evaluated using rhodamine B and Cr(VI) as models. The decolorization efficiency of rhodamine B could be up to 79.45% and Cr(VI) removal efficiency was 70.74% within 60 min, respectively. In addition, the iron content, different nZVI@cotton fabric system, the effect of pollutant composition and reusability of nZVI@cotton fabric were systematically investigated. Results showed that nZVI@cotton fabric prepared with the assistance of PDA was a kind of comparable wastewater purification material with the advantages of high removal efficiency to rhodamine B and Cr(VI) ions and easy to be recycled.

Pancreatic lipase immobilization on cellulose filter paper for inhibitors screening and network pharmacology study of anti-obesity mechanism

Guangxuan Chen,Huicong Yuan,Lumei Zhang,Jingran Zhang,Kefeng Li,Xu Wang

The discovery of pancreatic lipase (PL) inhibitors is an essential route to develop new anti-obesity drugs. In this experiment, chitosan was used to add amino groups to cellulose filter paper (CFP) and then glutaraldehyde was used to covalently combine PL with amino-modified CFP through the Schiff base reaction. Under optimal immobilization conditions, CFP immobilized PL has a wide range of pH and temperature tolerance, as well as excellent reproducibility, reusability and storage stability. Subsequently, 26 natural products (NPs) were screened by immobilized PL with black tea extract having the highest inhibition rate. Three compounds with binding effects on PL (epigallocatechin gallate, theaflavin-3-gallate and theaflavin-3,3′-digallate) were captured. Molecular docking proved that these three compounds have a strong binding affinity for PL. Fluorescence spectra further revealed that theaflavin-3,3′-digallate could statically quench the intrinsic fluorescence of pancreatic lipase. The molecular docking and thermodynamic parameters indicated that electrostatic interaction was considered as the main interaction force between PL and theaflavin-3,3′-digallate. Finally, the potential anti-obesity targets and pathways of the three compounds were discussed through network pharmacology. This study not only proposes a simple and efficient method for screening PL inhibitors, but also sheds light on the anti-obesity mechanism of active compounds in black tea.
