Ultra-broadband emission from Mg7Ga2GeO12:Ni2+,Cr3+ phosphor spanning the NIR I–III regions for spectroscopy applications

Yuan Tang,Pengbo Lyu,Desheng Yin,Zhaoqi Liu,Nian Pan,Kai Li,Ying Ye,Changfu Xu,Lizhong Sun

Ultra-broadband near-infrared (NIR) lighting sources covering NIR Ⅰ to III regions are pivotal components for NIR spectroscopy devices. However, achieving ultra-broadband and efficient emission covering the entire NIR Ⅰ to III regions remains a significant challenge. To address this, we selected Mg 7 Ga 2 GeO 12 as host material to realize ultra-broadband NIR emission. Density functional theory calculations on formation energies confirmed the feasibility of doping both Ni 2+ and Cr 3+ within Mg 7 Ga 2 GeO 12 . Band structure calculations reveal a significantly reduced band gap upon the doping of Ni 2+ . Following that, we synthesized Mg 7 Ga 2 GeO 12 :Ni 2+ through conventional solid-state-reaction method. This phosphor exhibits broadband NIR emission with a full-width-at-half-maximum of 377 nm in the range of NIR II ∼ III (1100–2100 nm). To further enhance the emission efficiency, we co-doped Mg 7 Ga 2 GeO 12 :Ni 2+ phosphor with Cr 3+ . This co-doping strategy not only dramatically enhances the Ni 2+ emission intensity, but also extends the lower limit of emission band to 600 nm. However, a spectral gap around 1150 nm remains between Cr 3+ and Ni 2+ emission centers. To bridge this spectral gap, we employed LiScSiO 4 :Cr 3+ phosphor alongside Mg 7 Ga 2 GeO 12 :Ni 2+ ,Cr 3+ phosphor to fabricate a phosphor converted LED (pc-LED) with ultra-broadband NIR emission covering entire NIR Ⅰ to III regions. By utilizing this pc-LED as the lighting source for NIR spectral detection, multiple organic functional group signals can be identified simultaneously in NIR II and III bands from 1000 to 2100 nm. The performance of such an NIR pc-LED not only confirms the application potential of Mg 7 Ga 2 GeO 12 :Ni 2+ ,Cr 3+ phosphor in non-destructive spectral analysis but also underscores the feasibility of combining two phosphors to achieve ultra-broadband NIR pc-LEDs, rendering them suitable to be lighting sources for portable spectrometers.

Multifunctional MnO2/Ag3SbS3 Nanotheranostic Agent for Single-Laser-Triggered Tumor Synergistic Therapy in the NIR-II Biowindow

Qian Wang,Botao Qu,Juan Li,Yuqin Liu,Jie Dong,Xiaoyang Peng,Ruiping Zhang

Regulating the level of reactive oxygen species (ROS) in a tumor is an efficient and innovative anticancer strategy. However, the therapeutic efficacy of ROS-based therapies, such as chemodynamic therapy (CDT) and photodynamic therapy (PDT), offers finite outcomes due to the oxygen dependence and limited concentration of hydrogen peroxide (H2O2) and overexpression of glutathione (GSH) within the tumor microenvironment (TME), so a single therapeutic strategy is insufficient to completely eliminate tumors. Therefore, we demonstrated an omnipotent nanoplatform MnO2/Ag3SbS3 (abbreviated as MA) with strong optical absorbance in the NIR-II biowindow and oxygen self-sufficient ROS-mediated ability, which not only relieves tumor hypoxia significantly but also enhances the photothermal therapy (PTT)/PDT/CDT efficacy. By 1064 nm laser irradiation, MnO2/Ag3SbS3 nanoparticles (NPs) reveal a favorable photothermal conversion efficiency of 23.15% and achieve a single-laser-triggered NIR-II PTT/PDT effect, resulting in effective tumor elimination. Once internalized into the tumor, MnO2/Ag3SbS3 NPs will be degraded to Mn2+ and Ag3SbS3. The released Ag3SbS3 NPs as a NIR-II phototherapy agent could be utilized for photoacoustic imaging-guided NIR-II PDT/PTT. Mn2+ could be used as a Fenton-like catalyst to continuously catalyze endogenous H2O2 for generating highly virulent hydroxyl radicals (•OH) for CDT and O2 for PDT, enhancing the efficiency of PDT and CDT, respectively. Meanwhile, Mn2+ realizes magnetic resonance imaging-guided accurate tumor therapy. Moreover, the MnO2/Ag3SbS3 NPs could deplete intracellular GSH in TME to promote oxidative stress of the tumor, further strengthening ROS-mediated antitumor treatment efficacy. Overall, this work presents a distinctive paradigm of TME-responsive PDT/CDT/PTT in the second near-infrared biowindow by depleting GSH and decomposing H2O2 for efficient and precise cancer treatment.

Peptide functionalized upconversion/NIR II luminescent nanoparticles for targeted imaging and therapy of oral squamous cell carcinoma

Bi Lin,Jun Wu,Yanxing Wang,Song Sun,Ying Yuan,Xiaofeng Tao,Ruichan Lv

Early diagnosis is critical and challenging for tongue squamous cell carcinoma (TSCC), which is a kind of tumor with high malignancy, poor prognosis, and a high incidence of invasion and metastasis. In this research, dual-modal optical imaging rare earth nanoparticle (RENP) probes with peptide functionalization are designed for targeted TSCC imaging and therapy. RENP@C@Au (UCA) with enhanced red upconversion luminescence (UCL) and near infrared II (NIR II) imaging intensity is designed by metal and codopant modulation, and the two peaks at about 650 nm and 1064 nm of Nd ions are relatively stable with less quenching and suitable for luminescence bioimaging. Then, the cMBP peptide targeted to Cal 27 TSCC cells with highly expressed c-MET proteins is combined with UCA. Compared with human's normal cells of MCF-10A, other tumor cell lines such as A549 and HeLa, together with mice tumor cells of 4T1, the designed probe has targeted imaging and therapy to Cal 27. The final in vivo experiment shows that the probe with high NIR II luminescence signals is not easy to retain when injected into the tail vein, indicating its potential precise clinical application for the diagnosis and therapy of TSCC.

Dual-molecular targeted NIR II probe with enhanced response for head and neck squamous cell carcinoma imaging

Yaqun Ma,Yanxing Wang,Xin Hui,Bi Lin,Ying Yuan,Xiaofeng Tao,Ruichan Lv

In this research, a fluorescent probe of 7-(diethylamine) coumarin derivatives with multiple binding sites to detect biothiols in tumor cell with strong NIR II luminescence in vivo was synthesized. The biothiols include cysteine (Cys) and glutathione (GSH) in tumor cells, and the tumor-response luminescence was proved by the cell experiment. Importantly, the monolayer functional phospholipid (DSPE-PEG) coating and aggregation induced emission (AIE) dye of TPE modification made the probe have good stability and biocompatibility with little luminescence quenching in aqueous phase, which was proved by in vitro and in vivo experiments. The final aqueous NIR II probe combined with bevacizumab (for VEGF recognition in the cancer cells) and Capmatinib (for Met protein recognition in the cancer cells) has stronger targeted imaging on head and neck squamous cell carcinoma (HNSCC) cancer with intravenous injection. This GSH/Cys detection in the tumor cell and strong dual-molecular NIR II bioimaging in vivo may provide new strategy to tumor detection.

In vivo fate of Ag2Te quantum dot and comparison with other NIR-II silver chalcogenide quantum dots

Zhang Jun-Zheng,Tang Huan,Chen Xing-Zhu,Su Qianqian,Xi Wen-Song,Liu Yuan-Yuan,Liu Yuanfang,Cao Aoneng,Wang Haifang

Narrow bandgap Ag 2 Te quantum dot (QDs) hold great promise in the fields of bioimaging, chemo-/bio-detection, and thermoelectric materials, but their biosafety has not been well estimated yet. In this study, we studied the biodistribution and toxicity of polyethylene glycol–coated Ag 2 Te QDs (Ag 2 Te QDs-PEG) in mice after a single intravenous injection. After entering the mice, Ag 2 Te QDs-PEG displayed a short circulation time in the blood (half-life was less than 2 h), and mainly accumulated in the liver and spleen. The stability of Ag 2 Te QDs-PEG in the mice depended on accumulation organ and time. Ag 2 Te QDs-PEG were stable in the spleen within 28 days but decomposed in the liver after 7 days. The decomposed Ag 2 Te QDs-PEG were eliminated from the body via different routes, as Ag was detected only in feces while Te was found only in urine. Meanwhile, Ag 2 Te QDs-PEG did not show obvious adverse effects on the body weights, organ indices, biochemical parameters, the levels of essential metal Cu and Zn in organs, and organ pathological investigations during 28 days. In addition, the important effects of anion to the fates of silver chalcogenide QDs in vivo was demonstrated by comparing the distributions of the Ag 2 Te QDs and other two silver chalcogenide QDs (Ag 2 S and Ag 2 Se). Our findings indicate Ag 2 Te QDs have good biocompatibility in mice, which is a guarantee for their safe applications in the future.

Paclitaxel/IR1061-Co-Loaded Protein Nanoparticle for Tumor-Targeted and pH/NIR-II-Triggered Synergistic Photothermal-Chemotherapy

Li He,Fangzhen Qing,Maode Li,Daitian Lan

Purpose The aim of this study was to develop an “all-in-one” nanoplatform that integrates at the second near-infrared (NIR-II) region dye IR1061 and anticancer drug paclitaxel (PTX) into an apoferritin (AFN) nanocage (IR-AFN@PTX). Simultaneously, folic acid (FA), tumor target molecule,  was conjugated onto IR-AFN@PTX to be IR-AFN@PTX-FA for tumor-targeted and pH/NIR-II-triggered synergistic photothermal-chemotherapy.Methods IR1061 was firstly reacted with PEG and then conjugated with AFN to be IR-AFN. Then, FA was conjugated onto the surface of IR-AFN to be IR-AFN-FA. At last, PTX was incorporated into IR-AFN-FA to fabricate a nanoplatform IR-AFN@PTX-FA. The NIR-II photothermal properties and pH/NIR-II triggered drug release were evaluated. The ability of IR-AFN@PTX-FA to target tumors was estimated using optical bioluminescence. In vitro and in vivo synergistic therapeutic effects of pH/NIR-II-triggered and tumor-targeted photothermal-chemotherapy were investigated in 4T1 tumor model. Results IR-AFN@PTX-FA showed excellent water solubility and physiological stability, which significantly enhanced the solubility of both IR1061 and PTX. After 5 min of laser irradiation at 1064 nm, IR-AFN@PTX-FA exhibited an effective photothermal effect compared with laser irradiation at 808 nm, even when blocked with 0.6 cm thick chicken breast. Cellular uptake experiments showed IR-AFN@PTX-FA utilized clathrin-mediated and caveolae-mediated endocytosis pathways to enter 4T1 cells, and was then delivered by the endosome to the lysosome. NIR-II laser irradiation and pH could synergistically trigger PTX release, inducing significant tumor inhibition in vitro and in vivo. Conclusion As a novel “all-in-one” nanoplatform, IR-AFN@PTX-FA was found to selectively target tumors and showed very efficient NIR-II photothermal effects and pH/NIR-II triggered drug release effects, showing a remarkable, synergistic photothermal-chemotherapy effect.

The Spectroscopic Properties and Microscopic Imaging of Thulium-Doped Upconversion Nanoparticles Excited at Different NIR-II Light

Tingting Peng,Rui Pu,Baoju Wang,Zhimin Zhu,Kai Liu,Fan Wang,Wei Wei,Haichun Liu,Qiuqiang Zhan

Lanthanide-doped upconversion nanoparticles (UCNPs) are promising bioimaging nanoprobes due to their excellent photostability. As one of the most commonly used lanthanide activators, Tm3+ions have perfect ladder-type electron configuration and can be directly excited by bio-friendly near-infrared-II (NIR-II) wavelengths. Here, the emission characteristics of Tm3+-doped nanoparticles under laser excitations of different near-infrared-II wavelengths were systematically investigated. The 1064 nm, 1150 nm, and 1208 nm lasers are proposed to be three excitation strategies with different response spectra of Tm3+ions. In particular, we found that 1150 nm laser excitation enables intense three-photon 475 nm emission, which is nearly 100 times stronger than that excited by 1064 nm excitation. We further optimized the luminescence brightness after investigating the luminescence quenching mechanism of bare NaYF4: Tm (1.75%) core. After growing an inert shell, a ten-fold increase of emission intensity was achieved. Combining the advantages of NIR-II wavelength and the higher-order nonlinear excitation, a promising facile excitation strategy was developed for the application of thulium-doped upconversion nanoparticles in nanoparticles imaging and cancer cell microscopic imaging. Keywords:upconversion nanoparticles;near-infrared-II;excitation mechanisms;luminescence quenching;microscopic imaging

Multiphonon Relaxation Inhibition and Photon Cutting Multiplication in Cs2NaYCl6: Ho3+/Er3+ for Information Encryption and NIR Bioimaging

Wei Wang,Jiaxin Yang,Xuewei Zhao,Hai Lin

The perovskite-based multifunctional integrated light-emitting platform has garnered significant attention due to the ultralow phonon energy and flexible lattice plasticity. Achieving both high-efficiency multiband light emission and excellent thermal stability for extreme conditions remains a critical challenge in system design and performance regulation. Herein, Ho 3+ and Er 3+ co-doped Cs 2 NaYCl 6 crystals suppress multiphonon relaxation, enabling precise control of the cross-relaxation between ions, thereby achieving mutual sensitization and multicolor visible fluorescence. Simultaneously, a parallel near-infrared (NIR) channel is activated through quantum cutting, where a high-energy photon is converted into two NIR photons via successive resonant energy transfer from Ho 3+ to Er 3+ , achieving a high internal quantum yield of 147.6%. Notably, the material exhibits high thermal sensitivity and exceptional anti-thermal quenching, with a luminescence intensity of 145.0% at 433 K relative to that at 313 K, showcasing tremendous potential for high-temperature optical sensing. The dynamic multicolor luminescence and highly efficient NIR emission are successfully integrated through a dual-channel luminescence mechanism, which establishes a multidimensional platform for optical encryption and bioimaging based on multifunctional luminescent system and high thermal stability, providing an innovative strategy for developing high-security anti-counterfeiting and deep tissue imaging.

Smart Asymmetric Hydrogel with Integrated Multi-Functions of NIR-Triggered Tunable Adhesion, Self-Deformation, and Bacterial Eradication

Lan Feng,Wenbin Shi,Qin Chen,Huitong Cheng,Jianxu Bao,Chunji Jiang,Weifeng Zhao,Changsheng Zhao

Multifunctional hydrogels acting as wound dressing have received extensive attention in soft tissue repair; however, it is still a challenge to develop a non-antibiotic-dependent antibacterial hydrogel that has tunable adhesion and deformation to achieve on-demand removal. Herein, an asymmetric adhesive hydrogel with near-infrared (NIR)-triggered tunable adhesion, self-deformation, and bacterial eradication is designed. The hydrogel is prepared by the crosslinking polymerization of N -isopropylacrylamide and acrylic acid, during the sedimentation of conductive PPy-PDA nanoparticles based on the polymerization of pyrrole (Py) and dopamine (DA). Due to the conversion capacity from NIR light into heat for PPy-PDA NPs, the formed temperature-sensitive hydrogel exhibits tissue adhesive as well as NIR-triggered tunable adhesion and self-deformation property, which can achieve an on-demand dressing refreshing. Systematically in vitro / in vivo antibacterial experiments indicate that the hydrogel shows excellent disinfection capability to both Gram-negative and Gram-positive bacteria. The in vivo experiments in a full-layer cutaneous wound model demonstrate that the hydrogel has a good treatment effect to promote wound healing. Overall, the asymmetric hydrogel with tunable adhesion, self-deformation, conductive, and photothermal antibacterial activity may be a promising candidate to fulfill the functions of adhesion on skin tissue, easy removing on-demand, and accelerating the wound healing process.

Coumarin derivative dye sensitized NaYGdF4:Yb,Er nanoparticles with enhanced NIR II luminescence for bio-vascular imaging

Yaqun Ma,Zhan Wang,Yanxing Wang,Zhenghao Liu,Yukun Wang,Ruichan Lv

Lanthanide-doped biocompatible nanoparticles have promising applications in near-infrared second region imaging due to their high chemical stability, enhanced photostability and sharp emission bandwidth. However, the weak light absorption capacity limits the application of rare-earth nanoparticles (RENPs) for bioimaging. We prepared a coumarin-derived dye sensitized NaYGdF 4 :Yb,Er nanoparticle probe, in which the organic dye enhances photon absorption through the sensitization process, improving the luminescence efficiency of the rare earth particles near 1000 and 1500 nm. In addition, good water solubility and stability of the probe are imparted by coating the particles with amphiphilic polymers distearoyl phosphatidylethanolamine-polyethylene glycol (DSPE-PEG) and polyacrylic acid. This composite probe with good biocompatibility and NIR II luminescence properties can be used for vascular imaging, providing a tool for the detection of hematologic-related diseases.

Endoplasmic reticulum-targeted NIR-II phototherapy combined with inflammatory vascular suppression elicits a synergistic effect against TNBC

Guoyun Wan,Xuheng Chen,Jiayu Chen,Ruiling Gou,Haijiao Wang,Shuhao Liu,Mingyang Zhang,Hongli Chen,Dan Wang,Qiqing Zhang

Recurrence and metastasis are the main reasons for failure in the treatment of triple-negative breast cancer (TNBC). Phototherapy, one of the most well-known potent cancer treatment models is highlighted by ablating primitive tumors with immunogenic cell death (ICD) and is associated with endoplasmic reticulum (ER) stress to elicit long-lasting anti-tumor immunity. However, the provoked inflammatory response after phototherapy will stimulate angiogenesis, which provides nutrition for tumor recurrence. Here, an ER-targeted nanoplatform was constructed based on hollow mesoporous Cu2−XS (HMCu2−XS) nanoparticles to suppress recurrence and metastasis of TNBC by combining photo-ablation and microenvironment remodeling. Profiting from the metal ion coordination and large hollow space, HMCu2−XS can be easily modified with p-toluenesulfonamide for ER-targeting and quantitatively loaded celecoxib (CXB) as a vascular inhibitor, thus obtaining ER-HMCu2−XS/CXB. ER-HMCu2−XS showed great photothermal and photodynamic efficiency for ablating 4T1 tumors and inducing ICD under NIR-II laser irradiation. Compared with non-ER-targeted nanosystems, the ER-targeted nanosystem elicited stronger ICDs and recruited more immune cells. Moreover, the thermal-responsively released CXB successfully inhibited angiogenesis after photothermal therapy. The data showed that the ER-HMCu2−XS/CXB mediated the triplicate therapeutic effect of photo-ablation, immune response activation, and vascular suppression effectively, preventing the recurrence and metastasis of TNBC. In conclusion, this work provides a synergistic strategy to enhance therapeutic outcomes in TNBC.

Dextran 40 hybrid biomimetic bismuth-nanoflower designed for NIR II-triggered hypoxic tumor thermoradiotherapy via macrophage escape

Jing Wen,Chang Liu,Ji Liu,Lu Wang,Si Miao,Daquan Chen,Qiyue Wang,Meirong Huo,Yan Shen

At present, NIR-II-triggered photothermal biomedical applications are limited by complex synthesis reactions , mediocre photothermal conversion efficiency, and difficult degradation. Herein, we prepared biodegradable Bi flower-like nanoparticles (phospholipid-modified Bi nanoflowers , BNFs) with high photothermal conversion efficiency (∼33.52 %) in NIR-II by a simple method and then modified them with the red blood cell membrane and dextran 40 (DRBCM) to improve their in vitro stability, to escape macrophages clearance and to enhance tumor accumulation. Dextran coating onto the surface of particles as a dispersant shell stabilizes inorganic particles by maintaining the surface charges and creating steric repulsions upon compression of neighboring polymer chains. In vitro and in vivo experiments proved that combined thermoradiotherapy of DRBCM-BNFs exhibited significantly enhanced tumor inhibition efficacy than monotherapy with good biocompatibility and low toxicity due to its biodegradability . Furthermore, the mechanism studies demonstrated that DRBCM-BNFs could serve as a nano sensitizer to promote the thermoradiotherapy under NIR-II illumination and X-ray irradiation, by downregulating heat shock protein 70 (HSP70) and phosphorylated-p65 (p-p65) to reduce the thermal resistance and radioresistance of tumor cells and increasing the expression of apoptosis-related protein cleaved caspase-3. In conclusion, DRBCM-BNFs could be a promising green delivery platform for the sensitization of synergistic thermoradiotherapy.
