Recently, the flowers of Dendrobium catenatum ( D. officinale ) have been approved as new food ingredient. This study aimed to investigate the herb-markers and their antioxidant activities in methanolic extracts of D. catenatum flowers, and to establish the quality evaluation methods for raw materials and their products of flower by HPLC. The methanolic extract of 11 strains of D. catenatum flowers were found to contain a high content of total phenol and flavonoids, and they possessed potential antioxidant capacities based on DPPH radical scavenging assay. A total of 21 phenolic herb-markers were selected according to the similarity and principal component analysis of the chromatographic fingerprinting profiles. Their structures were further elucidated by UV, HPLC-DAD-ESI-QTOF-MS/MS and NMR analyses. The identified compounds included 2 phenylpropanoids, 11 C -glycosylflavones and 6 O -glycosylflavones, which could be employed as the indicators for quantitative evaluation of the quality and authenticity of the flowers. Based on the pre-column DPPH/ABTS + -HPLC analysis, the major compounds contributed to the antioxidative activity were identified as 1- O -caffeoyl- β -D-glucoside, rutin and isoquercitrin, all of which, were also the most abundant constituents in the methanolic extract. The results suggest the potential of D. catenatum flowers as a new antioxidant resources for medicinal and food products.

Hawk tea is a rich and edible resource, traditionally used as a beverage in South China. This drink has many pharmacologic effects, such as acting as an antioxidant and reducing blood sugar and lipids. The objective of this work was to explore the active compound contents, bioactivities and their monthly changes, and optimize the harvest time. In the present study, Hawk tea from each month in 2017 was collected and extracted with 70% (v/v) ethanol. The contents of the total flavonoids and total phenols were determined using the colorimetric method. We determined the contents of seven characteristic active substances—hyperin, isoquercitrin, trifolin, quercitrin, astragalin, quercetin, and kaempferol—using high-performance liquid chromatography. The crude extract was tested for its antioxidant and inhibitory properties on enzymes involved in metabolic syndrome. Specifically, 2,2-diphenyl-1-picrylhydrazyl, 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid), ferric-reducing power assay, and the inhibition capacity test on α-glucosidase and lipase were conducted to determine the antioxidant effect in vitro, as well as the reduction of blood sugar and lipids. Monthly variations in activities and components were determined by numeric analysis and comparison. Correlation analysis revealed that antioxidant effects are significantly correlated with the total flavonoids. The hierarchical cluster analysis of bioactivities and their contents indicates that October and November are the best harvesting months, which differs with the habitual collection of Hawk tea. Keywords:Hawk tea;active components;antioxidant;α-glucosidase and lipase;monthly variation

In this study, synergetic and controlled-release low-density polyethylene (LDPE) antioxidant active films were prepared by cast extrusion. A well-ordered (4 nm) Mobil Composition of Matter No. 41 (MCM-41) was prepared to carry α-tocopherol for reducing the diffusivity ( D ), while improving the D of quercetin. Characterization of MCM-41 included small angle X-ray scattering, scanning electron microscopy, Fourier transform infrared spectroscopy, and nitrogen adsorption/desorption testing. New films added α-tocopherol and α-tocopherol loaded with MCM-41 on the base of a LDPE film containing quercetin. Testing of new films covered the physical properties, release behavior, and 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging assays. The physical properties discussed differences among any additives, which contain mechanical and barrier properties. Release experiments showed the controlled-release of MCM-41 to α-tocopherol, while addition of α-tocopherol promoted the release of quercetin in food simulant (95% ethanol). Free radical scavenging increased obviously after adding α-tocopherol and α-tocopherol loaded with MCM-41. This study demonstrated the feasibility of synergetic and controlled-release antioxidant active packaging films. Practical Application This work provides a method to control the release of α-tocopherol in the packaging films using molecular sieve MCM-41, and promotes the release of quercetin. The preservation of easily oxidized fatty foods is very important. In addition to the usual high-barrier composite packaging, the release-type antioxidant packaging is also a method. Food antioxidant packaging film is an important research direction in the field of intelligent packaging, and has great potential in the field of food flexible packaging. Release-type antioxidants are added to the packaging substrate, and the release rate of antioxidants is adjusted to prevent the oxidation of contents, to extend the shelf life of butter, and other commodities in the process of commodity storage and transportation.

In this work, a facile approach was developed to synthesized alcalase-inorganic hybrid nanocomposite (alcalase@CaHPO 4 ) by immobilizing alcalase with calcium hydrogen phosphate (CaHPO 4 ). The nanocomposite possessed flower-like morphological features with excellent hydrolysis activity on soybean protein isolates (SPI) with 1.57 fold higher compared to free alcalase. The experiment was evident of alcalase@CaHPO 4 hybrid nanoflowers with 90% sustainability after the seven cycles of reusability and 80–100% relative activity at 50–70 °C and with 65% at pH 4 in acidic condition. Soybean protein hydrolysates (SPHs) produced by immobilized alcalase possessed 70% radical-scavenging capacity at 0.8 mg/mL concentration and 20% calcium-binding capacity at pH 6. The solubility of SPHs produced by alcalase@CaHPO 4 hybrid nanoflowers was also improved by 15% compared to free alcalase. The high radical scavenging capability, good calcium binding capacity and improved solubility of SPHs prepared through alcalase@CaHPO 4 hybrid nanoflowers would be highly promising in food industries.

To explore whether and how glucuronidation affects pyrogallol-type phytophenols, scutellarein and scutellarin (scutellarein-7-O-glucuronide) were comparatively investigated using a set of antioxidant analyses, including spectrophotometric analysis, UV-vis spectra analysis, and ultra-performance liquid chromatography coupled with electrospray ionization-quadrupole time-of-flight tandem mass spectrometry (UPLC-ESI-Q-TOF-MS/MS) analysis. In spectrophotometric analyses of the scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH•), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+•), and 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-oxide radicals (PTIO•) and the reduction of Cu2+ions, scutellarein showed lower IC50values than scutellarin. However, in•O2−-scavenging spectrophotometric analysis, scutellarein showed higher IC50value than scutellarin. The analysis of UV-Vis spectra obtained after the Fe2+-chelating reaction of scutellarin showed a typical UV-Vis peak (λmax= 611 nm), while scutellarein showed no typical peak. In UPLC-ESI-Q-TOF-MS/MS analysis, mixing of scutellarein with DPPH•yielded MS peaks (m/z678, 632, 615, 450, 420, 381, 329, 300, 288, 227, 196, 182, 161, and 117) corresponding to the scutellarein-DPPH adduct and an MS peak (m/z570) corresponding to the scutellarein-scutellarein dimer. Scutellarin, however, generated no MS peak. On the basis of these findings, it can be concluded that glucuronidation of pyrogallol-type phytophenol antioxidants has a dual effect. On the one hand, glucuronidation can decrease the antioxidant potentials (except for•O2−scavenging) and further lower the possibility of radical adduct formation (RAF), while on the other hand, it can enhance the•O2−-scavenging and Fe2+-chelating potentials.Keywords:scutellarein;scutellarin;pyrogallol-type phytophenol;structure-activity relationship;glucuronidation;antioxidant

Enzyme-assisted aqueous two-phase extraction (EA-ATPE) using ethanol/ammonium sulfate system was investigated for total polyphenol (TP) and lutein from marigold flowers. The important factors were investigated by single factor experiment and response surface methodology combined with Box–Behnken design to optimize the operating parameters of EA-ATPE. The maximum yields of TP and lutein were 83.56 ± 0.69 mg g−1 and 5.59 ± 0.13 mg g−1, respectively. Compared with aqueous two-phase extraction and Soxhlet extraction (SE), the extraction yield of TP by EA-ATPE is 64.91% higher and the extract of EA-ATPE has better antioxidant activity. The pretreatment effect was also researched by scanning electron microscopy. Thus, EA-ATPE is an efficient method for extracting bioactive components from plants.

In this work, we designed polysaccharide-metal complex of dopamine-modified pectin linking ruthenium compound, which exhibited a certain inhibition specificity to human renal cell adenocarcinoma cell line 786-O. The chemical structure and physical properties of the polysaccharide-metal complex were well characterized by multiple analysis methods. The multi-spectral results revealed that pectin-dopamine have been successfully coordinated by ruthenium complex to form nano-conjugates, which self-assembled into relatively regular nanospheres of approximately 200 nm. The polysaccharide-metal complex has more amorphous and less viscosity than pectin, and cannot withstand as much strain as the pectin and pectin-dopamine systems. Pectin-dopamine can decrease the toxicity effect of Ru complex in normal cell line such as human normal renal epithelial cell line 293A.

In the present study, six different polysaccharides (RFPs, MAPs, UWPs, AEPs, HWPs and CEPs) were extracted from Chlorella vulgaris using repeated freeze–thawing, microwave-assisted-, ultrasonic wave-, alkali-, hot water-, and cellulase-based methods; and antioxidant property assays were performed both in vitro and in vivo . Radical-scavenging capacity (using DPPH, superoxide and hydroxyl radicals) and metal chelating ability were assessed in vitro ; Caenorhabditis elegans was used to assess antioxidant effects in vivo . Based on the in vitro screening tests, UWPs exhibited high antioxidant capacity. The UWP yield was 17.1% ± 2.2%; the DPPH-, superoxide-, and hydroxyl radical-scavenging rates were 65.1% ± 2.4%, 61.2% ± 2.7%, and 56.2% ± 2.2%, respectively, and the metal chelating ability was 63.6% ± 2.5% at a concentration of 0.4 mg/mL. UWPs also exhibited high antioxidant activity in vivo . UWPs significantly increased the lifespan of C. elegans under oxidative stress induced by hydrogen peroxide compared with the control group, enhanced stress-resistance-related enzymes, including catalase and superoxide dismutase by 7.29% ± 1.8% and 24.41% ± 4.8%, respectively. The results of the present study indicate that the extraction methods of C. vulgaris polysaccharides were a key factor influencing antioxidant activity.

Prebiotic xylooligosaccharides (XOS) are derived from xylan-rich agricultural residues and have important applications in the food, pharmaceutical, and cosmetic fields. The high hygroscopicity and low glass transition temperature (T g ) of XOS could result in operating problems and deterioration of product quality during the drying process and storage, so it is important to enhance physicochemical properties of dried product for improve XOS quality. The objectives of this work were spray drying XOS and comprehensive studying their rheological properties, physicochemical, and morphological characterization, when carried by gum Arabic (GA). The antioxidant effects of the products were evaluated by 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2′-Azinobis (3-ethylbenzothiazoline-6-sulfonic acid ammonium salt) (ABTS + ), ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The intuitive perturbation plot was applied to analyse the effects of the independent variables on the responses. A polynomial equation and the Gordon-Taylor equation were used to model the experimental data, and the fitted equations revealed the powder quality and storage stability well ( R 2 > 0. 95). The molecular weight of the carbohydrate affected the rheological properties and T g . Increasing the solid content of the initial solution tended to increase the apparent viscosity and median diameter. The EC 50 values of representative products were 1126.00 and 651.69 μg/mL by DPPH and ABTS + assays, and antioxidant activity values of representative products were 49.68 and 64.95 μmol TE/g by FRAP and ORAC assays, which shown the remarkable antioxidant effects. The drying yield, hygroscopicity, T g , microencapsulating efficiency, and colour attributes (L*, a*, and b*) of representative high GA concentration (GAC) products were 76.10%, 13.00 g H 2 O/100 g dry weight, 71.10°C, 99.69%, 96.93, 1.13, and 3.66, respectively, which shown better quality properties than low GAC products. According to the X-ray diffraction and scanning electron microscopy analysis, the microparticles were amorphous, and well-separated under an appropriate GAC. The Fourier transform infrared spectra results shown that the XOS and GA preserved their structural integrity during the spray drying process.

This study aimed to investigate the antioxidant activity and release behavior of anthocyanin (ANC) loaded within FA-g-MD wall (ANC-FA-g-MD microcapsule) in vitro. The microencapsulation of ANC was prepared by spray drying and displayed a biphasic release profile. The combination of ANC and FA-g-MD (0.0625–1 mg/mL) showed a higher antioxidant activity than that of both individuals. A possible intermolecular interaction between ANC and FA-g-MD was studied by UV-vis spectra. Intracellular reactive oxygen species (ROS), 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test, and protein expression of quinone oxidoreductase 1(NQO1), glutathione reductase (GSR) and γ-glutamate cysteine ligase catalytic subunit (γ-GCLC) were measured through human colon cancer cells (HT-29). After a 24-hour incubation of the HT-29, the combinations (0–60 μg/mL) exhibited a high potential to diminish the ROS level. And the distinct upregulated expressions of GCLC and NQO1 of HT-29 were detected after treatment with combinations compared to those of single ones. These results suggested that the ANC-FA-g-MD microcapsules exerts enhanced antioxidant effect with capability of the modulation of GCLC and NQO1.Keywords:antioxidant;oxidative stress;anthocyanin;HT-29

A free-of-dust pollution extraction method combined-homogenate and ultrasonic cavitation system, namely, homogenate-combined ultrasonic cavitation synergistic extraction (HUCSE), was proposed for the efficient extraction of flavonoids from Cinnamomum camphora leaves. Response surface methodology of Box–Behnken design was employed to optimize the HUCSE process, and the optimum operation conditions attained with an extraction yield of 7.95 ± 0.27 mg/g were ethanol concentration 76%, homogenate/ultrasonic time 25 min, solvent-to-solid ratio 22 mL/g, and ultrasonic power 240 W. A second-order kinetic mathematical methodology was performed to depict the behaviors of HUCSE and heat reflux extraction method. The results suggested that the developed HUCSE is an efficient and green method for the extraction of C. camphora flavonoids or other plant natural products, where the obvious higher parameters of extraction capacity at saturation, second-order extraction rate constant, and original extraction rate were obtained when compared to the heat reflux method. The antioxidant activity assays in vitro showed that the C. camphora flavonoids possessed strong antioxidant activity and are promising to be applied as a natural alternative antioxidant.

The roots of Rehmannia glutinosa (RG) have been widely used for medicinal purposes in Asia. The traditional processing of RG involves repetitive steaming and drying, and 9-time-steamed RG (NSRG) is the most commonly consumed form. For a development of a convenient processing method, RG was puffed at various pressures resulting in significantly increased solid extraction yield by up to 14%. The amount of the Maillard reaction product 5-hydroxymethylfurfural and the antioxidant capacities determined by the ABTS and DPPH radical scavenging assays were enhanced at increasing puffing pressure. Treatment of lipopolysaccharide-stimulated RAW 264.7 macrophages with RG extracts revealed that puffing of RG enhanced its suppression of the pro-inflammatory cytokine IL-6 by up to 37%. The 5-hydroxymethylfurfural contents, ABTS/DPPH radical scavenging capacities, and IL-6 regulatory effects of puffed RG samples were greater than those of the NSRG control, indicating that puffing is a desirable processing technique for development of nutraceuticals using RG.
