Lanthanide (Ln3+)-doped near-infrared (NIR) luminescent nanocrystals (NCs) have shown great promise in many technological fields. However, because of the parity-forbidden 4f → 4f electronic transitions of Ln3+, these NCs normally suffer from a low absorption and luminescence efficiency. Herein, we report a novel strategy for sensitizing the NIR luminescence of Ln3+ in NaGdS2 NCs based on the ligand-to-metal charge transfer transition from S2− to Yb3+. Through sensitization by the allowed S2−–Yb3+ charge transfer transition, intense NIR luminescence from Yb3+ and Er3+ with quantum yields up to 21.2% and 25.0% is achieved in Yb3+ singly-doped and Yb3+/Er3+ co-doped NaGdS2 NCs, respectively. These findings provide a general approach for the development of highly efficient Ln3+-doped NIR luminescent NCs through charge transfer sensitization, and also lay a foundation for future design of sulfide-based NIR luminescent nanomaterials towards various applications.

Featured with a zero-autofluorescence background, superior signal-to-noise ratio, high sensitivity, and deep penetration ability, near-infrared persistent luminescence nanoparticle (NIR-PLNP)-based multimodal nanoprobes show great potential for full-scale noninvasive cancer diagnosis. However, direct synthesis of NIR-PLNP-based multimodal nanoprobes with high drug loading capacity to meet growing cancer theranostic demands remains a challenge. In this work, multifunctional hybrid mesoporous nanoparticles (HMNPs) that integrate NIR-PLNPs (Ga2O3:Cr3+, Nd3+), magnetic nanoparticles (Gd2O3), and radionuclides (68Ga) are designed and constructed via a large-pore (mesoporous silica nanoparticle) MSN-templated strategy. The ingenious composition design endows HMNPs with rechargeable NIR-PL, superior longitudinal relaxivity, and excellent radioactivity, making these versatile nanoparticles available for long-term in vivo NIR-PL imaging, magnetic resonance imaging (MRI), and positron emission tomography (PET) imaging. More importantly, the application of large-pore MSN templates maintains the mesoporous structure of HMNPs, promising excellent drug loading capacity of these nanoparticles. As a proof-of-concept, HMNPs loaded with a high dose of DOX (chemotherapy agent) and Si-Pc (photosensitizer) are rationally designed for chemotherapy and NIR-PL-sensitized photodynamic therapy (PDT), respectively. Studies with mice tumor models demonstrate that the DOX/Si-Pc-loaded HMNPs possess excellent cancer cell killing ability and an outstanding tumor suppression effect without systemic toxicity. This work shows the great potential of HMNPs as an “all-in-one” nanotheranostic tool for multimodal NIR-PL/MR/PET imaging-guided chemotherapy and NIR-PL-sensitized photodynamic cancer therapy and provides an innovative paradigm for the development of NIR-PLNP-based nanoplatforms in cancer theranostic.

Exploiting two-dimensional nanomaterials as photo-based theranostic agents is promising for the highly efficient ablation of deep-tissue-buried tumors. However, they are limited by their poor absorption in the second near-infrared-light (NIR-II) bio-window (1000–1300 nm) and intrinsic nonbiodegradability. Herein, defect-rich sulfur-doped Ni(OH)2 (S-Ni(OH)2) nanosheets decorated with bovine serum albumin (BSA) as a novel theranostic agent is developed, which can accomplish multimodal-imaging-guided photothermal ablation of mouse cancers in the NIR-II bio-window. Sulfur doping extends the absorption spectra of Ni(OH)2 nanosheets from the visible to NIR-II bio-window, affording highly efficient photothermal conversion (58.20% for 1064 nm), entailing it to become an excellent contrast agent for photoacoustic imaging. Further, because of their intrinsic paramagnetic property, they can be applied for magnetic resonance imaging. Owing to the abundant defective sites in S-Ni(OH)2 nanosheets, they exhibit response to the tumor microenvironment, resulting in effective biodegradation and excretion from the body. In vivo toxicity experiments indicated that S-Ni(OH)2-BSA NSs delivered no appreciable toxicity and good biocompatibility. This work provides an avenue for the rational design of effective theranostics agents.

The development of near-infrared (NIR) emission nanoprobes for the ratiometric fluorescent determination of living cells in vitro/vivo is of great analytical importance. In this work, dual-NIR-emissive Zn-doped carbon-based nanosheets (Zn-CNSHs) were prepared with a beneficial and special donor−π–acceptor-conjugated (D−π–A-conjugated) spatial framework, which resulted in not only a much lower HOMO–LUMO energy level but also excellent biocompatibility and physicochemical properties. The Zn-CNSHs were prepared by simple one-pot solvothermal synthesis with zinc gluconate (ZGN) and a strong acid and exhibited two distinctive photoluminescence (PL) peaks at 620 and 720 nm with the 600 nm excitation. The 620 nm peak intensity was dependent on dipicolinic acid (DPA) owing to the aggregation-induced emission enhancement effect via the strong intersheet hydrogen bonds between Zn-CNSHs and DPA, enabling the ratiometric fluorescent determination of DPA, an important clinical anthrax biomarker. An excellent calibration curve showed linear regions over the range of 0.05–500 μM between the ratio of PL intensity (PL620 nm/PL720 nm) and the concentrations of DPA. The detection limit was down to 21.7 nM. Based on the high stability, low cytotoxicity, high selectivity, and outstanding PL-reliant sensitivity for the DPA assay, the nanoprobe has been successfully used to monitor DPA in serum, wastewater, and cells.

Chemodynamic therapy, which produces cytotoxic hydroxyl radicals through intratumoral Fenton reactions, has been extensively explored in nanomedicine for cancer treatment. However, the limited endogenous hydrogen peroxide (H 2 O 2 ) levels, which serve as the reactant, weakens the anticancer effect of the Fenton reaction. In this work, a cascade catalytically reactive nanosystem has been constructed for the treatment of epithelial and embryonal tumors with high synergetic efficacy through the integration of natural glucose oxidase (GOD) and polyethylene glycol (PEG) onto the surface of Fe 3 S 4 nanoplates (abbreviated as Fe 3 S 4 -PEG-GOD NPs). The GOD component consumes glucose in tumor and produces a large amount of H 2 O 2 , which provides rich substrates for the Fenton reaction catalyzed by the Fe catalytic center of the nanoplates. The intriguing photothermal conversion efficiency (45%) of the constructed composite nanocatalysts in the second near infrared (NIR-II) biowindow allows for photothermal treatment to substantially strengthen and synergize the anticancer effects of nanocatalysts both in vitro and in vivo. This synergy occurs through generation of a hyperthermic effect necessary for promoting the sequential catalytic reaction. The synergistic anticancer effect has been verified in both epithelial and embryonal tumor mouse models, and provides justification for the biomedical use of multifunctional nanocatalysts for versatile tumor nanotherapeutics.

A desirable cancer therapeutic strategy is supposed to have effective ability to not only exert maximum anticancer ability but also inspire antitumor immunity for preventing tumor relapse and metastasis. During this research, multifunctional upconversion nanoparticles (UCNPs) coated by ROS-responsive micelles are prepared for tumor targeting and near-infrared (NIR)-triggered photodynamic therapy (PDT)-combined synergistic effect of chemotherapy. Moreover, both PDT and chemotherapy agents could activate antitumor immunity via inducing immunogenic cell death with CD8+ and CD4+ T cells infiltrating in tumors. Through the experiments, intravenous administration of multifunctional nanocarriers with noninvasive NIR irradiation destroys the orthotopic tumors and efficiently suppresses lung metastasis in a metastatic triple-negative breast cancer model by cascade-amplifying chemo-PDT and systemic antitumor immunity. In conclusion, this study provides prospective chemo-PDT with inspired antitumor immunity for metastatic cancer treatment.

Multidrug-resistant Staphylococcus aureus (MRSA) seriously endanger human health. The development of efficient methods to eliminate the infections and monitor the treatment process are of great significance. Near-infrared-II (NIR-II) photoacoustic (PA) imaging and photothermal therapy (PTT) are highly integrated theranostic platforms with superior performance including a low imaging background, increased tissue penetration depth, and high photothermal threshold. Herein, we report an activatable near-infrared II (NIR-II) phototheranostic strategy using miniature Au/Ag nanorods (NRs) for the photochemical synergistic therapy of MRSA infections and in situ monitoring of the treatment progress. Au/Ag NRs were efficiently activated by ferricyanide solution and allowed to continuously release free Ag + to eliminate MRSA, triggering NIR-II photothermal and PA performance enhancement. The activated NIR-II photothermal effect in turn accelerated the release of free Ag + from Au/Ag NRs for the synergistic elimination of gram-positive Staphylococcus aureus and promoted wound healing. No photothermal damages or free Ag + -induced side effects were observed in treated mice. After synergistic treatment, a 20-fold NIR-II PA signal increase with a maximum signal-to-noise measurement of 9.5 was observed between the implanted site and normal tissue, enabling sensitive monitoring of Ag + release process. The prepared Au/Ag NRs were stable and biocompatible, showing great potential for activatable NIR-II phototheranostic application.

In this study, the polyacrylamide-gelatin composite hydrogels reinforced by halloysite nanotubes@polydopamine (HNTs@PDA) with NIR triggered shape memory performance and self-healing capacity were prepared through in-situ free radical polymerization of acrylamide in the mixture of Laponite-RD, HNTs@PDA and gelatin. HNTs@PDA was firstly synthesized through oxidative polymerization of dopamine on the surfaces of HNTs. HNTs@PDA was characterized by transmission electron microscopy , scanning electron microscopy, Fourier-transform infrared spectroscopy, X-ray diffraction, thermogravimetric analysis , Zeta potential , particle size analysis , and X-ray photoelectron spectroscopy. The photothermal performance of HNTs@PDA was then determined and the results reveal that HNTs@PDA can be employed as a superior photothermal agent to prepare light responsive hydrogels . The structure, morphology, mechanical properties , NIR triggered shape memory performance and self-healing capacity of the composite hydrogels were studied. The modified nanotubes act as cross-linking agent of polyacrylamide to form a rigid network in the hydrogel matrix, which leads to a significant increase in the mechanical properties. Moreover, the NIR triggered shape recovery process of the hydrogel is quite rapid, for example, it is only 63 s for recovering 720° shape change for the hydrogel of HNTs@PDA 40. The maximum NIR triggered healing efficiency of the hydrogels can reach 76%. The HNTs@PDA reinforced hydrogels with superior mechanical properties, NIR triggered shape memory, and self-healing ability exhibit promising applications in biomedical materials and smart engineering materials .

Hypothermal photothermal therapy (HPTT) employs hyperthermia (<45 °C) to destroy tumor cells with negligible side effects to the surrounding normal tissues. Despite extensive studies, the clinical translation of HPTT is severely hindered, owing to the discounted therapeutic effect and maximum permissible power of near-infrared (NIR) laser by food and drug administration (FDA). Herein, we report a rational design of red blood cell membranes (RBCs) coated FeS 2 (FeS 2 @RBCs) with strong absorption at NIR-Ⅱ window for effective HPTT augmented chemodynamic therapy (CDT). FeS 2 @RBCs exhibits prolonged blood circulation and negligible immune response, leading to improved tumor accumulation for enhanced HPTT. Furthermore, the CDT effect of FeS 2 @RBCs is significantly augmented by the temperature elevation in the tumor region, which leads to the synergetic HPTT and CDT. Lipidomics analysis reveals that the damage of tumor cells by CDT is via the lipid peroxidation. In addition, FeS 2 @RBCs exhibits self-enhanced magnetic resonance imaging after reacting with H 2 O 2 in tumor region for imaging-guided laser irradiation. Thus, FeS 2 @RBCs achieves remarkable inhibition of subcutaneous 4T1 breast tumor growth without obvious side effects by a 1064 nm laser irradiation of 1.0 W/cm 2 (FDA approved power density). Overall, this work provides a HPTT augmented CDT strategy for effective cancer therapy with a clinical approved laser power, which may pave the way for the clinical application of HPTT augmented CDT in the future.

A β-NaYF4:Yb,Er,Gd nanorods@1T/2H-MoS2 nanocomposite was fabricated and used for photocatalytic sterilization under illumination from single near-infrared (NIR) light. Approximately 99.3% of Escherichia coli were killed effectively by 15 min irradiation of NIR light at a dose of 40 μg mL−1. The effective bactericidal activity is ascribed to the high upconversion luminescence efficiency of β-NaYF4:Yb,Er,Gd and the layered two-dimensional nanosheet structure of 1T/2H-MoS2. Under 980 nm light irradiation, β-NaYF4:Yb,Er,Gd emits visible light, which is absorbed by MoS2, producing strongly oxidative holes and reductive electrons. Photogenerated electron–hole pairs were able to react with H2O, O2 and OH− to produce reactive species in aqueous solution. These reactive species are strong nonselective oxidizing agents for sterilization.

Accurate and sensitive fluorescence imaging of intracellular miRNA is essential for understanding the mechanism underlying some physiological and pathological events, as well as the prevention and diagnosis of diseases. Herein, a highly sensitive ratiometric fluorescent nanoprobe for intracellular miRNA imaging was fabricated by integrating a Ru-SiO2@polydopamine (Ru-SiO2@PDA) nanoplatform with a near-infrared light (NIR)-assisted DNA strand displacement signal amplification strategy. The Ru-SiO2@PDA spheres have excellent biosafety, high photothermal effect, and unique photophysical properties that can both emit a stable red fluorescence and well quench the fluorophores getting closer to them. So, when the fuel DNA and carboxyfluorescein (FAM)-labeled signal DNA are co-assembled on their outer surfaces, the FAM’s green fluorescence is quenched, and a low ratiometric signal is obtained. However, in the presence of miRNA, the target displaces the signal DNA from the capture DNA, releasing the signal DNA far away from the Ru-SiO2@PDA. Then, the green fluorescence recovers and leads to an enhanced Igreen/Ired value. Under NIR light irradiation, the Ru-SiO2@PDA increases the local temperature around the probe and triggers the release of fuel DNA, which thus recycles the target miRNA and effectively amplifies the ratiometric signal. Using A549 cells as a model, the nanoprobe realizes the highly sensitive ratiometric fluorescence imaging of miRNA let-7a, as well as its in vivo up- and down-regulation expressions. It provides a facile tool for highly sensitive and accurate intracellular miRNA detection through one-step incubation and may pave a new avenue for single-cell analysis.

The lysosome is known to be an acidic organelle that is ubiquitous in cells. Acidic pH is a characteristic feature of lysosomes, and aberrant lysosomal pH values have been manifested to be associated with diverse diseases. In order to monitor the lysosomal pH changes, we developed a ratiometric emission NIR-fluorescent probe (CzQl) for the quantitative analysis of the lysosomal pH in living cells and in vivo. CzQl was constructed via the ethylene bridging of carbazole derivatives and quinoline. The protonation of the probe resulted in a marked green-to-red emission color change. The probe exhibited an ideal pKa value of 4.60 with a linear relationship in the pH range of 3.9–5.3, which covered most of the pH ranges of lysosomes, and was useful for the quantitative detection and imaging of lysosomal pH changes. Moreover, CzQl exhibited easy loading capability, high selectivity, a large Stokes shift, specific lysosome-targeting ability and low cytotoxicity; thereby, it was successfully used for monitoring the lysosomal pH changes at a cellular level as well as the LPS-mediated inflammation in vivo.
