Antioxidant activity and α-amylase and α-glucosidase inhibitory activity of a fermented tannic acid product: Trigalloylglucose

Ru-yi Li,Shaoman Wang,David Julian McClements,Yin Wan,Cheng-mei Liu,Gui-ming Fu

Small molecules with beneficial biological activities can be generated by controlled fermentation of natural food products. In this study, the fermentation products of tannic acid by Aspergillus carbonarius FCYN212 were isolated and identified. Two major chemical structures were obtained: gallic acid (GA) and 3,4,6-tri- O -galloyl- d -glucose (TGG). GA and TGG exhibited stronger free radical scavenging ability and ferric-reducing power than butylated hydroxyanisole (BHA). Additionally, TGG demonstrated significant dose-dependent enzyme inhibitory activities against α-amylase and α-glucosidase with IC 50 values of 334.6 ± 8.9 and 46.5 ± 1.6 μM, respectively. Notably, TGG was a mixed type inhibitor of α-amylase and α-glucosidase according to a Dixon plot. This study revealed the antioxidant and enzyme inhibitory potential of TGG isolated from a fermented extract of tannic acid. TGG may therefore serve as an antioxidant and antidiabetic ingredient in functional or medical foods.

The study was designed to investigate the chemical composition and antioxidant activities of polysaccharides from Yingshan Cloud Mist Tea. The chemical composition of green tea polysaccharides (GTPS) was analyzed by Fourier-transform infrared (FT-IR) spectroscopy, scanning electron microscope (SEM), thermogravimetric (TGA), gas chromatograph (GC), and high-performance gel-permeation chromatography (HPGPC). Then, the antioxidant activities in vitro of GTPS, effects of GTPS on body weight, and the antioxidant activities in chickens were studied. The results showed that GTPS were composed of rhamnose (Rha), arabinose (Ara), xylose (Xyl), mannose (Man), glucose (Glu), and galactose (Gal) in a molar ratio of 11.4 : 26.1 : 1.9 : 3.0 : 30.7 : 26.8 and the average molecular weight was . Furthermore, GTPS exhibited obvious capacity of scavenging DPPH radical, hydroxyl radical, and superoxide radical and enhanced the ferric-reducing power in vitro. Last, GTPS significantly increased the body weight of chickens, enhanced the T-AOC, SOD, and GSH-Px level, and decreased the content of MDA in chickens. The results indicated that GTPS might be a kind of natural antioxidant, which had the potential application in feed industry.

In vitro preparation and characterization of amorphous calcium carbonate nanoparticles for applications in curcumin delivery

Rao Chaohui,Guo Xia,Li Min,Sun Xiaoqing,Lian Xiaojie,Wang Huifang,Gao Xianghua,Niu Baolong,Li Wenfeng

The development of high drug loading and sustained release nanoparticles by simple and economical methods is significant for the treatment of diseases. Amorphous calcium carbonate (ACC) is an ideal drug delivery system because of its excellent pH responsiveness, biocompatibility and biodegradability. In this work, ACC nanoparticles were successfully precipitated in ethanol–calcium solutions using ammonium carbonate as the internal source of CO 2 , which exhibited a relatively narrow size distribution in range of 10–200 nm. The microscopic morphology, amorphous characters, porous structure and thermal behavior of resultants were investigated by electron microscopy, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller and thermogravimetric analysis, respectively. In vitro drug release assay showed that ACC nanoparticles had high loading capacity of curcumin (Cur) and favorable drug release properties. Furthermore, ACC-Cur showed excellent abilities to scavenge free radicals, protect Cur stability and damage A549 cells, providing a broadened space for the applications of this material in the fields of biomedical or food.

Quantification of Neoagaro-Oligosaccharide Production through Enzymatic Hydrolysis and Its Anti-Oxidant Activities

Shu-Ying Xu,Jie Kan,Zhong Hu,Yang Liu,Hong Du,Guang-Chang Pang,Kit-Leong Cheong

Neoagaro-oligosaccharides (NAOS) have health benefits that are related to their amount and degree of polymerization (DP). However, the current methods that are used to quantify enzymatically released NAOS are un-specific and time-consuming. Agar has been extracted fromGelidium amansiiand has been degraded by AgaXa (a recombinant β-agarase). Polysaccharide analysis using carbohydrate gel electrophoresis (PACE) has been adapted in order to quantify NAOS. In addition, the anti-oxidant activity of the degraded samples has been assessed. We have found that the PACE method provided sensitive, precise, and accurate quantification for each of the six NAOS samples. PACE has revealed that the DP of the enzymatic products from the AgaXa digestion were mainly neoagaro-octaose and neoagaro-decaose. The degraded samples exhibited increased radical-scavenging activity towards 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis(3-ethylbenzothiazoline sulfonic acid) radicals. While the anti-oxidant activity may have been from NAOS activity and contributions from neoagaro-octaose and neoagaro-decaose. The adapted PACE method that has been presented here is promising for large sample analysis during quality control and for characterizing novel β-agarase degradation mechanisms. Keywords:Gelidium amansiiagar;neoagaro-oligosaccharides;β-agarase;anti-oxidant

Structural characterization, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta

Zheng-Nan Cai,Wan Li,Shomaila Mehmood,Wen-Juan Pan,Ya Wang,Fan-Ju Meng,Xiao-Fang Wang,Yong-Ming Lu,Yan Chen

This study aimed to investigate the structural features, in vitro and in vivo antioxidant activities of a heteropolysaccharide from the fruiting bodies of Morchella esculenta (FMP-1). FMP-1 had an average molecular weight of 4.7 × 10 3  Da and consisted of mannose, glucose and galactose. By methylation and NMR analysis, the backbone of FMP-1 was deduced to be made up of 1,4-linked Glc p and 1,6-linked Gal p . Hydroxyl, DPPH and superoxide radicals could be efficiently scavenged by FMP-1, with IC 50 values of 74.26, 119.32 and 161.49 μg/mL, respectively. Furthermore, FMP-1 could significantly protect zebrafish embryos against AAPH-induced oxidative damage. Decrease in malformations and mortalities was observed along with the reduction of ROS production, NO production and cell death. The protective effects were by decreasing MDA content and increasing SOD, CAT and GSH-Px levels. The current work provided a good suggestion of the potential utilization of FMP-1 as an attractive natural antioxidant.

Highly luminescent N-doped carbon dots from black soya beans for free radical scavenging, Fe3+ sensing and cellular imaging

Jing Jia,Bo Lin,Yifang Gao,Yuan Jiao,Lei Li,Chuan Dong,Shaomin Shuang

The novel N doped carbon dots (N-CDs) were readily fabricated through one-step pyrolysis method from black soya beans. The obtained N-CDs possessed excellent photoluminescence properties with quantum yield of 38.7 ± 0.64% and dual responsive properties towards free radical and Fe 3+ . The N-CDs exhibited favorable radical scavenging activity (RSA) against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide anion radicals . Fe 3+ could induce photoluminescence quenching of N-CDs through static quenching characteristics. The possible quenching mechanism was presented. Combining with negligible toxicity and preferable biocompatibility, the N-CDs were extended to imaging intracellular Fe 3+ . As an attractive candidate, the N-CDs demonstrated a great potential in free radical scavenging, sensing and cellular imaging.

Fermentation and complex enzyme hydrolysis for improving the total soluble phenolic contents, flavonoid aglycones contents and bio-activities of guava leaves tea

Lu Wang,You Luo,Yanan Wu,Yan Liu,Zhenqiang Wu

There are both soluble and insoluble-bound forms of phenolics in tea-leaf products. In order to increase total soluble phenolics contents, guava leaves tea (GLT) was first fermented with Monascus anka and Saccharomyces cerevisiae , and then hydrolyzed with complex enzymes . The changes in phenolics profiles, antioxidant activities and inhibitory effect on α -glucosidase in processed GLT were investigated. Compared with the un-fermented GLT, fermentation and complex enzymatic processing (FE) significantly increased the total phenolics, total flavonoids, quercetin and kaempferol contents by 2.1, 2.0, 13.0 and 6.8 times, respectively. After the FE, a major proportion of phenolics existed in the soluble form. Quercetin was released in the highest amount among different phenolics. In addition, soluble phenolic extracts from GLT following FE exhibited a highest antioxidant activity and inhibitory effect on α -glucosidase. The paper suggested an improved method for processing GLT into high-value products rich in phenolics and flavonoids aglycones with enhanced health benefits.

Quantification of polyphenol composition and multiple statistical analyses of biological activity in Portuguese red wines

Bai Shuang,Cui Can,Liu Jing,Li Pei,Li Qing,Bi Kaishun

The objective of the research was to evaluate potential biological activities of Portuguese red wines with different grape types on the level of composition characterization. An ultrafast liquid chromatography coupled with triple quadrupole mass spectrometry method was validated and used for qualitative analysis of commercial red wine samples through simultaneously determining 31 major active components. Meanwhile, the antioxidant activities of red wines were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl), ABTS [2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid)] and CUPRAC (cupric ion reducing antioxidant capacity) assay. The relations between polyphenols and antioxidant values were further considered by multivariate statistical analysis. Results of correlation analysis and stepwise multiple regression analysis showed that antioxidant activities were highly correlated to polyphenolic profiles, with significant contributions of procyanidins and phenolic acids. Consequently, the assessment method we established appeared to be an effective tool for appraising the nutritional value of polyphenols in wines, and the research strategy could be applied to the evaluation of other beverages.

Effect of different planting areas on the chemical compositions and hypoglycemic and antioxidant activities of mulberry leaf extracts in Southern China

Jing-Yi Hao,Yi Wan,Xiao-Hui Yao,Wei-Guo Zhao,Run-Ze Hu,Cong Chen,Long Li,Dong-Yang Zhang,Guo-Hua Wu

Guangdong, Guangxi and Chongqing are emerging sericulture areas in China where the production of mulberry leaves is huge. In order to identity high quality mulberry leaves that are suitable for healthy products to expand planting, 24 samples from three regions (Guangdong, Guangxi, Chongqing) in the south of China were quantified for two alkaloids (1-deoxynojirimycin and fagomine) and five phenols (chlorogenic acid, rutin, isoquercitrin, etc.) using high-performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). Additionally, the total phenolic and total flavonoid contents, antioxidant and glycosidase inhibitory activities (hypoglycemic activity) were tested using different assays (DPPH, ABTS, FRAP) to comprehensively evaluate the quality of the mulberry leaves. The contents of DNJ and fagomine ranged from 0.401±0.003 to 5.309±0.036 mg/g and from 0.279±0.031 to 2.300±0.060 mg/g, respectively. The main phenolic constituents were chlorogenic acid, rutin and isoquercitrin, with chlorogenic acid present in the highest concentrations, ranging from 3.104±0.191 to 10.050±0.143 mg/g. The antioxidant activity exhibited a tendency as follows: Guangxi > Guangdong > Chongqing, except for two samples from Chongqing, which showed the highest antioxidant activity. Based on our study, mulberry leaves from Guangdong and Guangxi could be future sources of natural hypoglycemic and antioxidant products.

Dual Effect of Glucuronidation of a Pyrogallol-Type Phytophenol Antioxidant: A Comparison between Scutellarein and Scutellarin

Qianru Liu,Xican Li,Xiaojian Ouyang,Dongfeng Chen

To explore whether and how glucuronidation affects pyrogallol-type phytophenols, scutellarein and scutellarin (scutellarein-7-O-glucuronide) were comparatively investigated using a set of antioxidant analyses, including spectrophotometric analysis, UV-vis spectra analysis, and ultra-performance liquid chromatography coupled with electrospray ionization-quadrupole time-of-flight tandem mass spectrometry (UPLC-ESI-Q-TOF-MS/MS) analysis. In spectrophotometric analyses of the scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH•), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+•), and 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl 3-oxide radicals (PTIO•) and the reduction of Cu2+ions, scutellarein showed lower IC50values than scutellarin. However, in•O2−-scavenging spectrophotometric analysis, scutellarein showed higher IC50value than scutellarin. The analysis of UV-Vis spectra obtained after the Fe2+-chelating reaction of scutellarin showed a typical UV-Vis peak (λmax= 611 nm), while scutellarein showed no typical peak. In UPLC-ESI-Q-TOF-MS/MS analysis, mixing of scutellarein with DPPH•yielded MS peaks (m/z678, 632, 615, 450, 420, 381, 329, 300, 288, 227, 196, 182, 161, and 117) corresponding to the scutellarein-DPPH adduct and an MS peak (m/z570) corresponding to the scutellarein-scutellarein dimer. Scutellarin, however, generated no MS peak. On the basis of these findings, it can be concluded that glucuronidation of pyrogallol-type phytophenol antioxidants has a dual effect. On the one hand, glucuronidation can decrease the antioxidant potentials (except for•O2−scavenging) and further lower the possibility of radical adduct formation (RAF), while on the other hand, it can enhance the•O2−-scavenging and Fe2+-chelating potentials.Keywords:scutellarein;scutellarin;pyrogallol-type phytophenol;structure-activity relationship;glucuronidation;antioxidant

Simultaneous extraction and enrichment of polyphenol and lutein from marigold (Tagetes erecta L.) flower by an enzyme-assisted ethanol/ammonium sulfate system

Xiao-Qian Fu,Gen-Lin Zhang,Long Deng,Yan-Yan Dang

Enzyme-assisted aqueous two-phase extraction (EA-ATPE) using ethanol/ammonium sulfate system was investigated for total polyphenol (TP) and lutein from marigold flowers. The important factors were investigated by single factor experiment and response surface methodology combined with Box–Behnken design to optimize the operating parameters of EA-ATPE. The maximum yields of TP and lutein were 83.56 ± 0.69 mg g−1 and 5.59 ± 0.13 mg g−1, respectively. Compared with aqueous two-phase extraction and Soxhlet extraction (SE), the extraction yield of TP by EA-ATPE is 64.91% higher and the extract of EA-ATPE has better antioxidant activity. The pretreatment effect was also researched by scanning electron microscopy. Thus, EA-ATPE is an efficient method for extracting bioactive components from plants.
