A sulfur-substituted hemicyanine-based activatable probe for photoacoustic visualization of neutrophil elastase in liver I/R injury

Kai Wang,Yan-Ru Wang,Jun Zhao,Ren-Min Zhou,Xi-Feng Xu,Tong Wang,Zhong-Yuan Cheng

Hepatic ischemia-reperfusion (I/R) injury is a major clinical complication, and the precise monitoring of key inflammatory mediators like neutrophil elastase (NE) in vivo remains a significant challenge. To address this, we developed HDS-NE, a highly specific and activatable molecular probe based on a sulfur-substituted hemicyanine scaffold, engineered for the simultaneous detection of NE via fluorescence and photoacoustic imaging modalities. The probe demonstrated exceptional sensitivity and selectivity toward NE, enabling the real-time visualization of elevated NE activity in a murine model of hepatic I/R injury. Both in vivo and ex vivo imaging results confirmed a markedly enhanced signal in injured livers compared to controls, which was further validated by serum assays and histology. Crucially, this study represents the successful achievement of non-invasive, in vivo imaging of NE activity during hepatic I/R processes, offering a powerful tool for diagnosing and studying inflammatory liver diseases.

Ethylene-independent modulation of root development by ACC via downregulation of WOX5 and group I CLE peptide expression

Wangshu Mou,Ria Khare,Joanna K. Polko,Isaiah Taylor,Juan Xu,Dawei Xue,Philip Benfey,Bram Van de Poel,Caren Chang,Joseph J. Kieber

In seed plants, the canonical role of 1-aminocyclopropane-1-carboxylic acid (ACC) is to serve as the precursor in the biosynthesis of the phytohormone ethylene, and indeed, ACC treatment is often used as a proxy for ethylene treatment. Increasing evidence suggests that ACC can also act independently of ethylene to regulate various aspects of plant growth and development. Here, we explore the effects of ACC on Arabidopsis thaliana root growth and the mechanisms by which it acts. ACC inhibits growth of the primary root in Arabidopsis seedlings when ethylene signaling is blocked, which becomes evident after 36 h of treatment with ACC. This reduced root growth is in part the result of suppressed cell proliferation in the root meristem resulting from altered expression of a key regulator of stem cell niche activity, WOX5. ACC also promotes lateral root (LR) development, in contrast to ethylene, which inhibits LR formation. Transcriptomic analysis of roots revealed no significant changes in gene expression after 45 min or 4 h of ACC treatment, but longer treatment times revealed a large number of differentially expressed genes, including the downregulation of the expression of a small group of phylogenetically related CLE peptides. Reduced expression of these group 1 CLEs in response to ACC leads to the activation of a transcription factor, LBD18, which promotes LR development. These results suggest that ACC acts to modulate multiple aspects of Arabidopsis root growth independently of ethylene via distinct transcriptional effects in the root meristem and LR precursor cells.

Regulatory Effect and Mechanism of Tanshinone I on Cell Apoptosis in Steroid-Induced Osteonecrosis of the Femoral Head

Qing Su,Jia Chen

Steroid-induced osteonecrosis of the femoral head (SIONFH) is a debilitating orthopedic condition. This study investigated the mechanism of Tanshinone I (TsI) in SIONFH modulating apoptosis in SIONFH via the PI3K/AKT/mTOR pathway. A SIONFH rat model was treated with TsI and a PI3K activator. Bone mineral density (BMD), bone volume/total volume (BV/TV), trabecular number (Tb.N), trabecular thickness (Tb.Th), and trabecular separation (Tb.Sp) were determined by microCT. Empty lacunae count, Osteopontin, and apoptosis of the femoral head tissues were assessed. Levels of Bax, cleaved-caspase-3, Bcl-2, and AKT, PI3K, and mTOR phosphorylation in femoral head tissues were determined by Western blot. SIONFH rats exhibited decreased BMD, BV/TV, Tb.N, and Tb.Th, increased Tb.Sp, reduced Osteopontin-positive cells, increased empty lacunae rate, and TUNEL and Osteopontin co-positive cells, elevated Bax and cleaved-caspase-3 protein levels, and diminished Bcl-2 protein expression. TsI promoted osteogenesis, attenuated SIONFH, and reduced apoptosis in SIONFH rats. TsI inhibited AKT, PI3K, and mTOR phosphorylation levels in the femoral head tissues of SIONFH rats, thereby repressing the PI3K/AKT/mTOR pathway activation. Activating the PI3K/AKT/mTOR pathway partially reversed TsI's effect in rats. Collectively, TsI limited the PI3K/AKT/mTOR pathway activation to reduce osteocyte apoptosis in SIONFH rats, which provided potential therapeutic insights for SIONFH treatment.

Facile synthesis of coal tar residue based porous carbon/MnO2 composite for eminent thallium(I) removal from wastewater

Gao Feiyue,Zhou Chuncai,Cao Yachuan,Yang Ke,Li Zhiguo,Li Xin,Liu Guijian

Thallium (Tl) is a typical hypertoxic heavy metal, its accumulation in the water environment can have serious impacts on ecosystems and human health. In this study, a composite material (PC@MnO 2 ) was designed for removing Tl(I) by grafting MnO 2 onto coal tar residue-based porous carbon (PC) via hydrothermal impregnation. δ-MnO 2 with nano flower-like structure was verified to be evenly distributed on PC surface through material characterizations. The adsorption performance was markedly elevated after loading MnO 2 on PC, and the optimal adsorbent (PC@MnO 2 -75) was prepared by hydrothermal process from KMnO 4 (75 mg) using PC (100 mg) as a reducing agent. The adsorption of Tl(I) onto PC@MnO 2 -75 has achieved equilibrium in 90 min, slightly affected by solution pH (4−12), and exhibited an outstanding adsorptive selectivity in the presence of high concentrations of K + , Na + , Ca 2+ , Mg 2+ , Cl - , CO 3 2- , humic acid, and fulvic acid. The pseudo second order kinetic and Freundlich isotherm models were preferably described as the adsorptive characteristics. And the Tl(I) removal was a spontaneous exothermic adsorptive process. Density functional theory (DFT) results manifested that Mn doping enhanced electron transfer rate during adsorption. The highly efficient removal of Tl(I) on PC@MnO 2 -75 was achieved via electrostatic interaction, complexation, and oxidation-adsorption. The porous carbon derived from coal tar residue was engineered into PC@MnO 2 composites for efficient Tl(I) removal, which was in line with the environmental protection concept of sustainable development and green production.

Inhalable DNase I@Au Hybrid Nanoparticles for Radiation Sensitization and Metastasis Inhibition by Elimination of Neutrophil Extracellular Traps

Yuchao Sun,Jian He,Weiyu Chen,Yongfang Wang,Kai Wang,Min Zhou,Yichun Zheng

High-dose radiation therapy is a widely used clinical strategy to inhibit tumor growth. However, the rapid generation of excessive reactive oxygen species (ROS) triggers the formation of neutrophil extracellular traps (NETs), which capture free tumor cells in the bloodstream, promoting metastasis. In this study, we developed a hybrid nanoparticle composed of DNase I and gold (DNase I@Au) to enhance radiotherapy efficacy while mitigating metastasis by precisely eliminating NETs. The DNase I@Au nanoparticles, administered via aerosol inhalation, are efficiently delivered to lung tumor tissue, improving radiosensitization and reducing tumor size. Crucially, the nanoparticles could gradually release DNase I, effectively degrading ROS-induced NETs and preventing the interaction of free malignant cells with tumor sites or vasculature, thereby inhibiting metastasis. Therefore, we provide an enzyme and sensitizer co-loaded strategy that offers a promising approach to improve the therapeutic outcome of radiotherapy and reduce the risk of lung cancer metastasis under ROS stimulation.

I self-grafting enables broad spectrum response and surface localized electric field promoting visible-light photocatalysis of CdBiO2I

Weishan Zheng,Junyi Zhou,Fang Chen,Shuchen Tu,Xinxin Xu,Hongwei Huang

Photocatalysis technology has promising application prospects in the treatment of water pollution. However, at present, the performance of photocatalytic pollutant degradation of photocatalysts is limited by the high recombination rate of photogenerated carriers and the low utilization rate of visible light. In this work, we develop a surface-I ion self-grafted CdBiO 2 I (CBOI-I) as an efficient visible-light photocatalyst via a facile precipitation route under ambient atmosphere. Compared with CdBiO 2 I, the self-grafted I ions allows CdBiO 2 I a grafting-concentration dependent light response range. With the increase in the grafting concentration of I ion, the absorption edge of CBOI-I red-shifts from 580 nm to 650 nm, with a band gap decrease of around 0.2 eV. In-situ Kelvin-probe force microscopy, photoluminescence spectra, photoelectrochemical tests and DFT calculation demonstrate, that the surface self-grafting of I ion on CdBiO 2 I enables the formation of surface localized electric field on the surface of the photocatalyst, which efficiently promotes the separation and transfer of photogenerated holes and electrons, producing more superoxide radicals and holes as reactive species. Thus, the optimal CBOI-I (CBOI-I4) exhibits an excellent photocatalytic performance for degradation of bisphenol A upon visible light illumination, in which the degradation rate reaches 94% within 60 min, beyond 4.9 times that of CdBiO 2 I. Besides, other contaminants, including tetracycline hydrochloride, aureomycin hydrochloride, rhodamine B, and methyl orange are also effectively degraded, demonstrating the photodegradation universality of CBOI-I. This work offers an alternative route for developing high-efficiency and broad-spectrum responsive via surface modification.

Slippery organogel coating based on Ag(I)-imine coordination bonding for enhanced self-healing, antifouling and anticorrosion properties

Yuan Jing,Fandi Meng,Fuhui Wang,Li Liu

Slippery liquid-infused porous surface (SLIPS) has garnered significant attention in marine anticorrosion and antifouling applications due to their slippery attributes. Nevertheless, it remains challenging to fulfill the protection requirements by relying exclusively on the barrier and anti-adhesion mechanisms of surface lubricating layer. In this work, a dual-functional slippery organogel coating was meticulously synthesized by introducing Ag(Ӏ)-imine coordination bonds into organosilicon networks, thereby endowing the coating with self-healing and antibacterial properties. A surface scratch with a width of 50 μm can be effectively repaired within 6 h at 60 °C, and the repair efficiency exceeds 85 %. In addition, the coating exhibits a dual antibacterial mechanism, which functions through both sterilization via the release of Ag + and antiadhesion due to its slippery properties. Following a 7-day cocultivation period with PAO1 bacteria, the coating retained 100 % bactericidal activity and 99.8 % resistance to bacterial adhesion. Moreover, the antialgal efficacy of the AP-TA 0.2 -Ag 1 @Oil coating against Chlorella and P. tricornutum increased by 99.9 % and 99.7 %, respectively. Furthermore, the combination of Ag(Ӏ)-imine coordination bonds and silicone oil effectively preserves the corrosion resistance of the coating. The EIS results demonstrated that the | Z | 0.01 Hz value of the slippery organogel coating was maintained at 1.07 × 10 7  Ω·cm 2 after immersion in 3.5 wt% NaCl solution for 14 days, suggesting excellent anticorrosion performance of the coating. This study provides a novel perspective for the development of marine protective coatings that possess antifouling, self-healing, and anticorrosive properties.

PEA prevented early BBB disruption after cerebral ischaemic/reperfusion (I/R) injury through regulation of ROCK/MLC signaling

Dequan Kong,Baoying Xie,Yuhang Li,Yaping Xu

Palmitoylethanolamide (PEA) offers a strong protection against BBB disruption and neurological deficits after cerebral ischaemic/reperfusion (I/R) injury. To date, these BBB protective effects of PEA are mainly attributed to PPARα-mediated actions. However, whether PEA protects against BBB disruption through direct regulation of cytoskeletal microfilaments remains unknown. Here, we identified PEA as a Rho-associated protein kinase (ROCK2) inhibitor (IC 50  = 38.4 ± 4.8 μM). In vitro data suggested that PEA reduced the activation of ROCK/MLC signaling and stress fiber formation within microvascular endothelial cells (ECs) after oxygen-glucose deprivation (OGD), and consequently attenuated early (0–4 h) EC barrier disruption. These actions of PEA could not be blocked by the PPARα antagonist GW6471. In summary, the present study described a previously unexplored role of PEA as a ROCK2 inhibitor, and propose a PPARα-independent mechanism for pharmacological effects of PEA.

MicroRNA-153 impairs hippocampal synaptic vesicle trafficking via downregulation of synapsin I in rats following chronic cerebral hypoperfusion

Shuai Zhang,Mei-Ling Yan,Lin Yang,Xiao-Bin An,Hong-Mei Zhao,Sheng-Nan Xia,Zhuo Jin,Si-Yu Huang,Yang Qu,Jing Ai

Chronic cerebral hypoperfusion (CCH) promotes the development of Alzheimer's pathology. However, whether and how CCH impairs the synaptic vesicle trafficking is still unclear. In the present study, we found that the hippocampal glutamatergic vesicle trafficking was impaired as indicated by a significant shortened delayed response enhancement (DRE) phase in CA3-CA1 circuit and decreased synapsin I in CCH rats suffering from bilateral common carotid artery occlusion (2VO). Further study showed an upregulated miR-153 in the hippocampus of 2VO rats. In vitro, overexpression of miR-153 downregulated synapsin I by binding the 3′UTRs of SYN1 mRNAs, which was prevented by its antisense AMO-153 and miRNA-masking antisense oligodeoxynucleotides ( SYN1-ODN ). In vivo , the upregulation of miR-153 elicited similar reduced DRE phase and synapsin I deficiency as CCH. Furthermore, miR-153 knockdown rescued the downregulated synapsin I and shortened DRE phase in 2VO rats. Our results demonstrate that CCH impairs hippocampal glutamatergic vesicle trafficking by upregulating miR-153 , which suppresses the expression of synapsin I at the post-transcriptional level. These results will provide important references for drug research and treatment of vascular dementia .

Topoisomerase I Inhibition, DNA Photocleavage Activity, and G-Quadruplex DNA ‘Light Switch’ Based on Nitro-Substituted Ruthenium Complexes

Xue-Wen Liu,Liu Ning-Yi,Deng Yuan-Qing,Wang Shan,Liu Ting

Abstract Two ruthenium complexes containing nitro-substituted ligand, [Ru(bpy) 2 (hnpip)](PF 6 ) 2 ( 1 ) and [Ru(phen) 2 (hnpip)](PF 6 ) 2 ( 2 ) (bpy = 2,2'-bipyridine; phen = 1,10-phenanthroline; hnpip = 2-(3-hydroxyl- 4-nitrophenyl)imidazo[4,5- f ][1,10-phenanthroline] were synthesized and characterized. DNA binding behaviors were studied by luminescent titration, UV-vis titration, FRET (FRET = Fluorescence resonance energy transfer) and molecular docking experiments. Complex 2 exhibited “light switch” effect upon addition of G-quadruplex DNA and displayed fluorescent selectivity towards G-quadruplex DNA over other DNAs in the presence of K 4 [Fe(CN) 6 ]. Fluorescence experiments, FRET experiments and molecular docking results indicated that the selectivity may be attributed to protection of nitro group from water, DNA affinity and the difference in DNA structures. Furthermore, both complexes were found to cleave DNA under irradiation by the formation of singlet oxygen. DNA topoisomerase inhibition experiments indicated that complex 2 exhibited higher topoisomerase I inhibition activity (IC 50 = 18 μM) than complex 1 (IC 50 = 50 μM). Docking studies revealed that complex 2 stabilized Top1cc complex via π–π interaction and the formation of hydrogen bond.

Trehalose against UVB-induced skin photoaging by suppressing MMP expression and enhancing procollagen I synthesis in HaCaT cells

Zhenbang Xiao,Shengtao Yang,Jiali Chen,Chengyong Li,Chunxia Zhou,Pengzhi Hong,Shengli Sun,Zhong-Ji Qian

Ultraviolet radiation B cause photoaging of the skin by enhancing the high expression of matrix metalloproteinases (MMPs) to degrade collagen and inhibit the synthesis of procollagen. Trehalose (TRE), a widely occurring natural disaccharide, has been shown to play a cytoprotective role in many pathological processes. However, few studies have evaluated whether it has anti-photoaging potential. In this study, we investigated the anti-photoaging and mechanism of action of TRE using Human immortalized keratinocytes (HaCaT) cells. The results indicated that TRE pretreatment effectively scavenges reactive oxygen species (ROS) accumulation induced by UVB and increases the content of certain endogenous antioxidant factors in HaCaT cells. TRE suppressed UVB-induced MMP expression by blocking mitogen-activated protein kinases (MAPKs), activator protein 1 (AP-1) and NF-κB signaling. TRE treatment increased procollagen I synthesis via activating transforming growth factor-β (TGF-β)/Smad pathway and demonstrated that TRE may be a potential natural disaccharide for photoaging therapy in the future.

Synthesis and identification of quinoline derivatives as topoisomerase I inhibitors with potent antipsoriasis activity in an animal model

Wen-Jin Zhang,Peng-Hui Li,Min-Cong Zhao,Yao-Hao Gu,Chang-Zhi Dong,Hui-Xiong Chen,Zhi-Yun Du

Psoriasis is a chronic inflammatory and immune-mediated skin disease. Although certain agents have shown clinical success in treating psoriasis, development of safe and effective strategies for the treatment of this condition remains important. Research suggests that DNA topoisomerase I (Topo I) inhibitors may have potent psoriasis-ameliorating effects. Here, 25 quinoline derivatives were synthesized and identified as Topo I inhibitors. These compounds inhibited the 12– O –tetradecanoylphorbol-13-acetate-induced mouse ear inflammation. The most potent analogs, 5i and 5l , suppressed the expression of inflammatory cytokines in lipopolysaccharide-stimulated HaCaT cells. Additionally, the lead compounds significantly improved imiquimod-induced psoriasis-like inflammation in mice. Moreover, the expression levels of cytokines and inflammatory mediators, such as interleukin (IL)-17A, IL-22, IL-23, nuclear factor-κB subunit p65, tumor necrosis factor-α, and interferon-γ, were dramatically inhibited in the dorsal skin of 5i - and 5l -treated mice. These findings indicate that the inhibition of Topo I activity may potentially be an effective strategy for psoriasis treatment.
