Immobilized lipase post-encapsulated in Cu-based MOFs with promoted stability in biodiesel synthesis

Hongbo Suo,Xinzheng Liu,Shu Chen,Qi Li,Xiangnan Liu,Lili Xu

Biodiesel is more environmentally friendly compared to traditional petroleum diesel. The method of producing biodiesel catalyzed by lipases, as a green chemistry technology, can further reduce its negative impact on the environment. In this work, lipase immobilized on magnetic cellulose was embedded in Cu(PABA), and then the biocomposite (MCL@Cu(PABA)) was used for biodiesel production. The good biocompatibility and flexible skeleton structure of cellulose can buffer the compression effect on enzyme molecules during the formation of Metal-Organic Frameworks (MOFs), and the MOFs protective layer can reduce the damage caused by denaturants to enzyme molecules. The biocatalyst showed enhanced activity and stability with a lower deactivation constant ( K d ). Under the optimized reaction conditions, the biodiesel yield can reach 78.4 %. After reused for 7 times, the biocatalyst can still achieve a biodiesel yield of 58.8 %. Moreover, after stored for 30 days, the biodiesel yield can still reach 70.3 %. For all experimental yields, the standard deviation (SD) ranged from 2.4 % to 4.5 %. Secondary structure analysis showed that the rigid structure and active conformation of lipase was enhanced after Cu(PABA) encapsulation.

Inhibitory effect of luteolin on the metabolism of vandetanib in vivo and in vitro

Yuxin Shen,Fengsheng Hong,Hualu Wu,Xiaohai Chen,Hailun Xia,Ren-ai Xu,Guanyang Lin,Lu Shi

This study aimed to examine the potential drug-drug interaction (DDI) between vandetanib and luteolin in vivo and in vitro, with the objective of establishing a scientific foundation for their appropriate utilization in clinical settings. Sprague-Dawley (SD) rats were randomly divided into two groups: a control group (vandetanib administered by gavage alone) and an experimental group (vandetanib and luteolin administered together). A series of blood samples were collected at different time intervals. The plasma concentrations of vandetanib and its metabolite N-demethyl vandetanib in rats were determined using an ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). Incubation systems were set up with rat liver microsomes (RLM) and human liver microsomes (HLM) to measure the Michaelis-Menten constant (Km) and half-maximum inhibitory concentration (IC50) values. Additionally, the inhibitory mechanism of luteolin on vandetanib was also investigated. Ultimately, the molecular mechanism of inhibition was examined through the utilization of molecular docking techniques. In vivo animal experiment results showed that compared with the control group, the AUC(0-t) and Cmax of vandetanib in the experimental group were significantly increased. The findings from the in vitro experiments revealed that luteolin exhibited a moderate inhibitory effect on the metabolism of vandetanib. The IC50 values for RLM and HLM were determined to be 8.56 μM and 15.84 μM, respectively. The identified inhibition mechanism was classified as mixed. This study utilized molecular docking analysis to provide additional evidence supporting the competitive inhibition of luteolin on vandetanib in CYP3A4. The data presented in our study indicated a potential interaction between vandetanib and luteolin, which may necessitate the need for dose adjustment during their co-administration in clinical settings.

In-situ exsolved Ni nanoparticles for boosting CO2 reduction in solid oxide electrolysis cell

Tao Chen,Fei Shan,Lingting Ye,Kui Xie

Due to their excellent high Faraday efficiency, perovskite solid oxide cells (SOECs) have attracted considerable attention. Nevertheless, they still face significant challenges in terms of stability and electrocatalytic activity during CO₂ electrolysis. In this study, Ni particles in La 0.65 Ba 0.35 Mn 1-x Ni x O 3-δ are successfully separated by a unique in-situ exsolved method of metal nanoparticles, which are uniformly anchored to the electrode surface as Ni nanometal particles. This effectively suppresses the generation of carbon deposits on the cathode surface. Under test conditions of 850 °C, 1.6 V and 50 sccm flow rate, the CO yield of the modified cathode material reached 5.9 mL min −1 cm −2 , which is nearly four times higher than that before doping. The synergistic effect of in-situ exsolution of Ni metal nanoparticles with oxygen defects generated by the perovskite, creating more active locations for CO 2 adsorption and electrolysis, is responsible for the significant improvement in electrochemical performance. This work provides new strategies and ideas for the development of efficient and durable SOEC cathode materials.

Designing hollow mesoporous carbon sphere for high-rate supercapacitor in water-in-salt electrolyte

Liping Feng,Huichao Liu,Wenyan Zan,Sidian Li,Yunzhen Chang,Ying Zhang,Wenjing Hou,Yun Zhao,Sheng Zhu,Gaoyi Han

The expansion of the operational voltage in supercapacitors (SCs) through the utilization of water-in-salt electrolytes (WISEs) has garnered considerable attention for achieving elevated energy density. However, the poor rate performance of assembled SCs persists due to the heightened viscosity of WISEs (e.g., 17.0 mol kg −1 NaClO 4 ), so designing the porous structure of electrode materials to improve the rate performance of SC in WISEs has become a research focus. In this study, we present a straightforward approach to fabricate hierarchical porous carbon spheres, designing to expedite ion infilling and transport within WISE-based SCs. The hollow porous carbon is synthesized via one-step pyrolysis process, wherein the decomposition of inner colloid particles yields gases, while the outer resorcinol–formaldehyde resin carbonizes to form a spherical shell. In the WISE of 17.0 mol kg −1 NaClO 4 , the resulting symmetric SCs exhibit an operational voltage of 2.6 V and significantly improved rate capability with a capacitance retention of 47.7 % at 100 A g −1 . Notably, the energy density can reach 31.0 Wh kg −1 at a power density of 2.4 kW kg −1 . Even under high power density conditions of 120.0 kW kg −1 , the energy density of 14.7 Wh kg −1 can remain. Additionally, the SC device manifests an impressive capacitance retention of 99.1 % after 20,000 cycles, showcasing exceptional cyclic stability. This research introduces a novel synthesis strategy for hierarchical porous carbon materials, enabling the development of high-rate SCs with both elevated energy density and power density.

Drawdown and suspension of corn straw in the non-Newtonian fluid in stirred tank

Chao Song,Xuejun Hu,Shuang Yang,Jinxiao Xu,Chang Chen,Guangqing Liu

Due to the low density and hydrophobicity, the corn straw (CS) particles tend to float on the liquid surface in the anaerobic digester, deteriorating the bioconversion efficiency and process stability. Although the mechanical mixing is widely used in digesters, the drawdown and suspension of CS in the non-Newtonian slurry have not been sufficiently studied. This paper investigated the effect of particle size, liquid viscosity, solid phase mass fraction, impeller type and off-bottom distance on the just-drawdown rotational speed ( N jd ), complete drawdown rotational speed ( N cd ) and cloud depth ( CD ) of dry and wet straw particles in the stirred tank. The results showed that the distribution of dry and wet straw in the stirred tank significantly differed due to differences in density and wettability. The models built by dimensional analysis and machine learning were suitable for accurate predictions of specific variables. Based on the model analysis, the N jd of dry straw was mainly affected by the liquid viscosity, impeller type and off-bottom distance, as these factors determined the flow field of liquid phase for its drawdown. N jd of wet straw was related to the liquid viscosity and N cd was influenced by impeller off-bottom distance. The CD of dry and wet straw particles were both affected by rotational speed, impeller type, and liquid viscosity related to the mean flow and turbulence intensity in the tank. The results provide valuable references for the design and operation of mixing configurations for low-density feedstocks in the anaerobic digester.

Direct laser writing photonic crystal hydrogel sensors for in-situ sensing in microfluidic device

Keliang Liu,Haibo Ding,Zhejun Chong,Yi Zeng,Yanfang Niu,Junning Zhang,Youlong Kang,Xin Du,Zhongze Gu

Photonic crystal hydrogels (PCHs) are promising materials for in-situ sensing demands in cell incubation systems such as microfluidic devices. However, the lack of ability to precisely construct and settle PCH sensors limits their future development. In this paper, we developed a versatile and highly controllable strategy to fabricate PCH sensing systems via direct laser writing (DLW) technology. We show that, PCH microsensors with precise geometries and diverse compositions could be exactly generated at desired positions using two-photon lithography (TPL). With such smart sensors, in-situ and spatiotemporal monitor of environmental parameters in liquid microdomains could easily be achieved using a microscope. This is demonstrated by generating PCH sensors for in-situ pH and metal ion strength analysis in microfluidic devices.

Diversity in mechanisms of natural humic acid enhanced current production in soil bioelectrochemical systems

Xintong Gao,Kaixuan Liu,Chong Zhang,Xian Cao,Takashi Sakamakic,Xianning Li

The quinoid component of humic acids (HAs) had been studied as exogenous electron mediators (EMs), but the redox-mediating abilities of other functional groups remained unclear. This study evaluated the effects of various HAs functional groups on cellular respiration and extracellular electron transfer. The three EMs increased the current density compared to the control. Current density increased significantly after adding ultraviolet-irradiated HAs (UV-HAs), suggesting that nitrogenous group-mediated redox reactions contributed to high-density current generation. Structural equation model (SEM) results indicated that the contribution of nitrogen-containing groups to electron transfer could exceed 20%. This study proposed a synergistic mechanism: in the soil microbial fuel cells (soil-MFCs), HAs accelerated their component evolution through irreversible redox reactions and promoted extracellular electron transfer. Additionally, HAs-induced high expression of c -Cyts could further enhance high-density current generation. This study demonstrates that humic acids enhance electron transfer and current in bioelectrochemical systems, aiding sustainable energy optimization.

RGD-modified PEG–PAMAM–DOX conjugates: In vitro and in vivo studies for glioma

Lihong Zhang,Saijie Zhu,Lili Qian,Yuanying Pei,Yongming Qiu,Yanyan Jiang

This work was based on our recent studies that a promising conjugate, RGD-modified PEGylated polyamidoamine (PAMAM) dendrimer with doxorubicin (DOX) conjugated by acid-sensitive cis -aconityl linkage (RGD-PPCD), could increase tumor targeting by binding with the integrin receptors overexpressed on tumor cells and control release of free DOX in weakly acidic lysosomes. To explore the application of RGD-PPCD to glioma therapy, the effects of the conjugate were further evaluated in glioma model. For comparative studies, DOX was also conjugated to PEG–PAMAM by acid-insensitive succinic linkage to produce the PPSD conjugates, which was further modified by RGD to form RGD-PPSD. In vitro cytotoxicity of the acid-sensitive conjugates against C6 cells was higher than that of the acid-insensitive ones, and further the modification of RGD enhanced the cytotoxicity of the DOX-polymer conjugates as a result of the increased cellular uptake of the RGD-modified conjugates by C6 cells. In vivo pharmacokinetics, biodistribution and antitumor activity were investigated in an orthotopic murine model of C6 glioma by i.v. administration of DOX-polymer conjugates. In comparison with DOX solution, all the conjugates showed significantly prolonged half-life and increased AUC and exhibited higher accumulation in brain tumor than normal brain tissue. Although RGD-PPCD was more than 2-fold lower tumor accumulation than RGD-PPSD, it exhibited the longest survival times among all treatment groups, and therefore, RGD-PPCD conjugate provide a desirable candidate for targeted therapy of glioma.

Changes in Novel Biomarkers for Protein Oxidation in Pork Patties under Different Cooking Methods

Chuanyu Guo,Shouyin Wang,Xiaolei Jia,Jinfeng Pan,Xiuping Dong,Shengjie Li

The aim of this study was to assess the effectiveness of different biomarkers to identify the levels of protein oxidation in pork patties induced by assorted cooking methods. To achieve this purpose, pork patties prepared fromlongissimus dorsiwere cooked using three methods (frying, steaming, and roasting) at different internal temperatures (60, 70, 80, and 90 °C). Traditional biomarkers including total carbonyl and total thiol and novel biomarkers including α-aminoadipic semialdehyde (AAS) and lysinonorleucine (LNL) were determined. Results demonstrated that total thiol and AAS were the most successful biomarkers in distinguishing the three cooking methods in relation to protein oxidation, with AAS being the most sensitive. Moreover, as indicated by the biomarkers of total thiol and AAS, frying caused the highest level of protein oxidation, while steaming resulted in the lowest level when pork patties were cooked to the internal temperatures of 70 or 80 °C.

Nanocarriers boost non-systemic fluazinam transportation in plants and microbial community enrichment in soil

Xiao Wenhui,Du Jiang,Zheng Yuxin,Chen Shanshan,Liu Zhanyun,Chen Fengping,Liu Feng,Li Beixing,Liu Xili,Zhang Can

Utilizing nanotechnology for pesticide delivery can enhance their uptake and distribution in plants, thereby boosting pesticide efficiency. Fluazinam, a highly effective broad-spectrum fungicide with unique mode of action, is hindered by its poor systemic conductivity, limiting its field control efficacy. In this study, fluazinam-loaded nanocapsules were developed, which exhibited significantly higher inhibitory activity against various Phytophthora species compared to the commercial suspension concentrate. Notably, the nanocapsules demonstrated pronounced effects on reducing ATP content in Phytophthora capsici and effectively controlled pepper blight and cucumber downy mildew. Tracking the distribution of fluorescein isothiocyanate (FITC) fluorescein-labeled fluazinam nanocapsules revealed their presence in roots seven days after treatment. High-performance liquid chromatography (HPLC) analysis confirmed the easy absorption and transport of fluazinam nanocapsules by pepper plants. Moreover, treatment with fluazinam nanocapsules significantly increased the expression of two exocytosis genes, FACP and RABF2a , in pepper tissues. Lastly, fluazinam nanocapsules were found to enhance the relative abundance of beneficial bacteria in the soil microbial community. This study presents a promising approach to enhance the upward transport capability of non-systemic pesticides, offering novel insights into their application and improved efficacy.

Proanthocyanidins in seed coat tegmen and endospermic cap inhibit seed germination in Sapium sebiferum

Faheem Afzal Shah,Jun Ni,Jing Chen,Qiaojian Wang,Wenbo Liu,Xue Chen,Caiguo Tang,Songling Fu,Lifang Wu

Sapium sebiferum , an ornamental and bio-energetic plant, is propagated by seed. Its seed coat contains germination inhibitors and takes a long time to stratify for germination. In this study, we discovered that the S. sebiferum seed coat (especially the tegmen) and endospermic cap (ESC) contained high levels of proanthocyanidins (PAs). Seed coat and ESC removal induced seed germination, whereas exogenous application with seed coat extract (SCE) or PAs significantly inhibited this process, suggesting that PAs in the seed coat played a major role in regulating seed germination in S. sebiferum . We further investigated how SCE affected the expression of the seed-germination-related genes. The results showed that treatment with SCE upregulated the transcription level of the dormancy-related gene, gibberellins (GAs) suppressing genes, abscisic acid (ABA) biosynthesis and signalling genes. SCE decreased the transcript levels of ABA catabolic genes, GAs biosynthesis genes, reactive oxygen species genes and nitrates-signalling genes. Exogenous application of nordihydroguaiaretic acid, gibberellic acid, hydrogen peroxide and potassium nitrate recovered seed germination in seed-coat-extract supplemented medium. In this study, we highlighted the role of PAs, and their interactions with the other germination regulators, in the regulation of seed dormancy in S. sebiferum .

Anti-osteoclastogenic effect of epigallocatechin gallate-functionalized gold nanoparticles in vitro and in vivo

Shenting Zhu,Lingxin Zhu,Jingjing Yu,Yanqing Wang,Bin Peng

Background: Epigallocatechin gallate (EGCG), the major anti-inflammatory compound in green tea, has been shown to suppress osteoclast (OC) differentiation. However, the low aqueous solubility of EGCG always leads to poor bioavailability, adverse effects, and several drawbacks for clinical applications.Purpose: In this study, we synthesized EGCG-capped gold nanoparticles (EGCG-GNPs) to solve the drawbacks for clinical uses of EGCG in bone destruction disorders by direct reduction of HAuCl4 in EGCG aqueous solution. Methods and Results: The obtained EGCG-GNPs were negatively charged and spherical. Theoretical calculation results suggested that EGCG was released from GNPs in an acidic environment. Cellular uptake study showed an obviously large amount of intracellular EGCG-GNPs without cytotoxicity. EGCG-GNPs exhibited better effects in reducing intracellular reactive oxygen species levels than free EGCG. A more dramatic anti-osteoclastogenic effect induced by EGCG-GNPs than free EGCG was observed in lipopolysaccharide (LPS)-stimulated bone marrow macrophages, including decreased formation of TRAP-positive multinuclear cells and actin rings. Meanwhile, EGCG-GNPs not only suppressed the mRNA expression of genetic markers of OC differentiation but also inhibited MAPK signaling pathways. Furthermore, we confirmed that EGCG-GNPs greatly reversed bone resorption in the LPS-induced calvarial bone erosion model in vivo, which was more effective than applying free EGCG, specifically in inhibiting the number of OCs, improving bone density, and preventing bone loss. Conclusion: EGCG-GNPs showed better anti-osteoclastogenic effect than free EGCG in vitro and in vivo, indicating their potential in anti-bone resorption treatment strategy.
