Green Polymer Poly-l-proline Efficiently Inhibits Formation of Gas Hydrates in Oil–Water System

Huiping Qi,Yanzhen Liu,Shi Shen,Jiafei Zhao

Green polymer poly- l -proline, as a new type of biodegradable polymer material, has a wide application prospect in the field of environmental protection. Recent studies have shown that poly- l -proline can also act as a green inhibitor that is highly effective in inhibiting the production of gas hydrates in oil–water systems. As a natural polymer material, poly- l -proline has good biocompatibility and biodegradability and will not cause pollution to the environment. It is found that adding an appropriate amount of poly- l -proline to the oil–water system can significantly inhibit the formation of gas hydrates. This is because poly- l -proline can reduce the concentration of dissolved gases in water by adsorbing and dispersing ions and molecules in water, thereby reducing the chance of gas forming hydrates with water. In addition, poly- l -proline can also prevent gas molecules from entering water molecules to form hydrates by forming hydrogen bonds with water molecules. This study provides a green, environmentally friendly and efficient solution for controlling the formation of gas hydrates in oil–water mixtures. In the future, the application prospects of poly- l -proline in oil–water separation, marine engineering, natural gas exploitation, and other fields will be broader.

Highly dispersive PEI-modified CDs@ZIF-L dual-emitting fluorescent sensor for detecting metal ions

Xiaoyun Liu,Chunyan Sun,Mingxia Chai,Weijun Song

The leaf-like zeolitic imidazolate framework (ZIF-L) is a promising porous nanomaterial that has attracted increasing attention as an ideal host material to encapsulate functional fluorescent nanoparticles for designing fluorescent sensors. However, owing to the large particle size, gravity readily facilitates the precipitation of the ZIF-L from the aqueous solution, and thus lead to imperfect experimental results. Herein, we report a simple and rapid synthetic method which uses the polyethyleneimine (PEI)-modified ZIF-L as a host to solve the precipitation problem and construct a dual-emitting system that combines its fluorescence with carbon dots (CDs). Furthermore, CDs@ZIF-L/PEI with dual-emitting centres could be utilised as a ratio fluorescence sensor to detect Hg2+ ions. The sensor exhibited excellent dispersibility and good selectivity for sensing Hg2+ ions, with a limit of detection (LOD) of 14.5 nM. Furthermore, experimental results reveal that the CDs@ZIF-L/PEI fluorescent sensor could be effectively dispersed into agarose and less polar organic solvents such as DMF, MeOH, EtOH and CH3CN, expanding the application scope of the fluorescent sensor.

Induced mechanism of phosphatase hormesis by Cd ions and rhizosphere metabolites of Trifolium repens L.

Chenxi Zhang,Haibo Li,Yue Yang,Xulun Zhou,Damiao Zhuang,Wengang Liu,Kaixuan Wang,Pengkai Wang,Wenxin Zhang,Yaran Bai,Haotian Ma,Bingqian Gao,Rui Wang

Rhizosphere phosphatases can exhibit hormetic effects in response to cadmium (Cd) ion stimulation. However, understanding the mechanisms underlying hormesis effects on soil ecosystems is challenging as studies on hormesis are usually specific to an organism, cell, or organ. To comprehensively investigate the mechanism of phosphatase hormesis, this study utilized in situ zymography and metabolomics to analyze the rhizosphere of Trifolium repens L. (white clover). Zymograms showed that rhizosphere phosphatase displayed a hormetic effect in 10 mg kg −1 Cd contaminated soil, with a hotspot area 1.8 times larger than non-Cd contaminated soil and a slight increase in enzyme activity . Nevertheless, the phosphatase activity was substantially suppressed upon elevating the Cd concentration in the soil to 50 mg kg −1 . Differential metabolite identification and KEEG pathway enrichment analysis revealed that both rhizosphere organic acids and amino acid compounds positively affected phosphatase activity, and both were able to stabilize complexation with Cd ions via carboxyl groups . Besides, molecular docking models suggested that Cd ions act as cofactors to induce the formation of hydrogen bonds between amino acids/organic acids and phosphatase residues to form a triplet complex with a more stable structure, thereby improving phosphatase activity. The results indicated that amino acids and organic acids are heavily enriched in the rhizosphere of white clover and form a particular structure with soil Cd ions and phosphatase, which is essential for inducing the phosphatase hormesis as a detoxification mechanism in the rhizosphere micro-ecosystem.

Bioconversion of cellulose and hemicellulose in corn cob into L-lactic acid and xylo-oligosaccharides

Yiqin Wang,Jian Du,Qiang Li,Yehan Tao,Yi Cheng,Jie Lu,Haisong Wang

With the banning of antibiotic chemical feed additives, multi-functional bioactive feed additives have been extensively sought after by the feed industry. In this study, low-cost and renewable corn cobs were treated with liquid hot water and converted into bioactive xylo-oligosaccharides and L-lactic acid after enzymatic hydrolysis , strain activation, and fermentation under mild conditions, which achieved a full utilization of cellulose and hemicellulose in corn cobs. Simultaneous saccharification fermentation after strain activation with enzymatic hydrolysate delivered the highest conversion rate of glucose to L-lactic acid (93.00 %) and yielded 17.38 g/L L-lactic acid and 2.68 g/L xylo-oligosaccharides. On this basis, batch-feeding fermentation resulted in a 78.03 % conversion rate of glucose to L-lactic acid, 18.99 g/L L-lactic acid, and 2.84 g/L xylo-oligosaccharides. This work not only provided a green and clean bioconversion strategy to produce multi-functional feed additives but can also boost the full utilization of renewable and cheap biomass resources.

Microbial modifications with Lycium barbarum L. oligosaccharides decrease hepatic fibrosis and mitochondrial abnormalities in mice

Zheng Zhang,Wenjia Lu,Pengfei Liu,Mengjie Li,Xinyi Ge,Bin Yu,Zhengzong Wu,Guimei Liu,Nannan Ding,Bo Cui,Xiao Chen

Background Lycium barbarum L. is a typical Chinese herbal and edible plant and are now consumed globally. Low molecular weight L. barbarum L. oligosaccharides (LBO) exhibit better antioxidant activity and gastrointestinal digestibility in vitro than high molecular weight polysaccharides. However, the LBO on the treatment of liver disease is not studied. Purpose Modification of the gut microbial ecosystem by LBO is a promising treatment for liver fibrosis. Study design and methods Herein, LBO were prepared and characterized. CCl 4 -treated mice were orally gavaged with LBO and the effects on hepatic fibrosis and mitochondrial abnormalities were evaluated according to relevant indicators (gut microbiota, faecal metabolites, and physiological and biochemical indices). Results The results revealed that LBO, a potential prebiotic source, is a pyranose cyclic oligosaccharide possessing α-glycosidic and β-glycosidic bonds. Moreover, LBO supplementation restored the configuration of the bacterial community, enhanced the proliferation of beneficial species in the gastrointestinal tract (e.g., Bacillus, Tyzzerella, Fournierella and Coriobacteriaceae UCG-002 ), improved microbial metabolic alterations (i.e., carbohydrate metabolism, vitamin metabolism and entero-hepatic circulation), and increased antioxidants, including doxepin, in mice. Finally, LBO administration reduced serum inflammatory cytokine and hepatic hydroxyproline levels, improved intestinal and hepatic mitochondrial functions, and ameliorated mouse liver fibrosis. Conclusion These findings indicate that LBO can be utilized as a prebiotic and has a remarkable ability to mitigate liver fibrosis.

Electrospun polylactic acid nanofiber membranes containing Capparis spinosa L. extracts for potential wound dressing applications

Peng Zhu,Xingqun Zhang,Yunlong Wang,Changen Li,Xianzhu Wang,Jiancheng Tie,Ying Wang

Capparis spinosa L. (CSL) is a medicinal plant with high antibacterial activity against a variety of pathogens and antioxidation properties. In this paper, for the first time, nanofiber membranes of polylactic acid (PLA) containing 0, 4, 7,and 10 wt% CSL ethyl acetate extract were fabricated by electrospinning. Scanning electron microscopy showed that the fiber diameter decreased after adding CSL to the PLA nanofibers. Fourier transform infrared spectroscopy confirmed that CSL was successfully incorporated in the matrix. The water contact angle test proved that the addition of CSL improved the hydrophilic properties of the material. Moreover, the addition of CSL improved the oxidation resistance of the composite fiber membrane. A burst drug release from the composite nanofibers occurred within the first 12 hr, followed by slow release over a prolonged period of time. As the concentration of CSL increased, the inhibition ability of nanofibers against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) gradually increased. In summary, due to their good mechanical, antioxidant, and antibacterial properties, CSL/PLA nanofiber membranes may possess potential applications as wound dressing materials.

The Great Capacity on Promoting Melanogenesis of Three Compatible Components in Vernonia anthelmintica (L.) Willd.

Yifan Lai,Qingyuan Feng,Rui Zhang,Jing Shang,Hui Zhong

To investigate a possible methodology of exploiting herbal medicine and design polytherapy for the treatment of skin depigmentation disorder, we have made use ofVernonia anthelmintica (L.) Willd., a traditional Chinese herbal medicine that has been proven to be effective in treating vitiligo. Here, we report that the extract ofVernonia anthelmintica (L.) Willd. effectively enhances melanogenesis responses in B16F10. In its compound library, we found three ingredients (butin, caffeic acid and luteolin) also have the activity of promoting melanogenesis in vivo and in vitro. They can reduce the accumulation of ROS induced by hydrogen peroxide and inflammatory response induced by sublethal concentrations of copper sulfate in wild type and green fluorescent protein (GFP)-labeled leukocytes zebrafish larvae. The overall objective of the present study aims to identify which compatibility proportions of the medicines may be more effective in promoting pigmentation. We utilized the D-optimal response surface methodology to optimize the ratio among three molecules. Combining three indicators of promoting melanogenesis, anti-inflammatory and antioxidant capacities, we get the best effect of butin, caffeic acid and luteolin at the ratio (butin:caffeic acid:luteolin = 7.38:28.30:64.32) on zebrafish. Moreover, the effect of melanin content recovery in the best combination is stronger than that of the monomer, which suggests that the three compounds have a synergistic effect on inducing melanogenesis. After simply verifying the result, we performed in situ hybridization on whole-mount zebrafish embryos to further explore the effects of multi-drugs combination on the proliferation and differentiation of melanocytes and the expression of genes (tyr, mitfa, dct, kit) related to melanin synthesis. In conclusion, the above three compatible compounds can significantly enhance melanogenesis and improve depigmentation in vivo. Keywords:melanogenesis;ROS;inflammatory;zebrafish;butin;caffeic acid;luteolin;synergistic effect

Identification and Characterization of the Seed Storage Proteins and Related Genes of Cannabis sativa L.

Sun Xin,Sun Yao,Li Yao,Wu Qiong,Wang Lei

Hemp (Cannabis sativa L.) seed is emerging as a novel source of plant protein owing to its rich protein content and reasonable nutritional structure. In the current study, the storage proteins of hemp seed were extracted using different methods. The modified Osborne method yielded maximum extraction of the hemp seed storage proteins, while degreasing had little effect on the hemp seed protein (HSP) extraction. Protein identification results revealed that 11S globulin (edestin) was the most abundant protein in hemp seed, and the molecular weights of the two subunits of this protein were ~35 and 20 kDa, respectively. The second most abundant protein was 2S albumin (Cs2S), with a molecular weight of ~14–15 kDa. The least abundant protein was 7S vicilin-like protein (Cs7S), with a molecular weight of ~47 kDa. Subsequently, gene families encoding these three storage protein classes, including three genes for edestin, two for Cs2S, and one for Cs7S, were cloned and then analyzed for amino acid composition and structure. The three edestins were different in their amino acid sequences and calculated molecular weights. The analysis of coding sequences revealed a higher percentage of similarity (62.7%) between Edestin1 and Edestin3, while the similarity decreased significantly to ~57% between Edestin1 and Edestin2, and 58% between Edestin2 and Edestin3. The calculated protein molecular weight was the highest for the protein encoded by Edestin1 and the smallest for the protein encoded by Edestin2. All three edestins were rich in arginine, while Edestin3 had a higher methionine content relative to that in the other two, which proved that Edestin3 had a better nutritional value. Cs2S and Cs7S were different from those reported in previous studies. Therefore, it could be inferred that amino acid composition varies with different hemp cultivars. The current research brought significant theoretical advance in illuminating the understanding of hemp seed storage protein and would have significance for future research on improving the nutritional quality of hemp seed and developing bioactive peptides.

Micro-distribution of arsenic and polycyclic aromatic hydrocarbons and their interaction in Pteris vittata L.

Xiao-Yong Liao,Xue-Gang Gong,Li-Li Zhang,Daniel P. Cassidy

Interactive effects of inorganic arsenic (As) species and polycyclic aromatic hydrocarbons (PAHs) on their uptake, accumulation and translocation in the hyperaccumulator Pteris vittata L. ( P. vittata ) were studied hydroponically. The presence of PAHs hindered As uptake and acropetal translocation by P. vittata , decreasing As concentrations by 29.8%–54.5% in pinnae, regardless of the initial As speciation. The inhibitive effect of PAHs was 1.6–8.7 times greater for arsenite [As(III)] than for arsenate [As(V)]. Similarly, inorganic As inhibited the uptake of fluorene (FLU) and benzo[a]pyrene (BaP) by P. vittata roots by 0.4%–21.7% and by 33.1%–69.7%, respectively. Interestingly, coexposure to As and PAHs slightly enhanced the translocation of PAHs by P. vittata with their concentrations increased 0.3 to 0.8 times in shoots, except for the As(III)+BaP treatment. The antagonistic interaction between As and PAHs uptake is likely caused by competitive inhibition or oxidative stress injury. By using synchrotron radiation micro X-ray fluorescence imaging, high concentrations of As were found distributed throughout the microstructures far from main vein of the pinnae when coexposed with PAHs, the opposite of what was observed with exposure to As only. PAHs could also significantly inhibit the accumulation and distribution of As in vascular bundles in rachis treated with As(III). The results of two-photon laser scanning confocal microscopy revealed that PAHs were mainly distributed in the vascular cylinder, epidermal cells, vascular bundles, epidermis and vein tissues, and this was independent of As speciation and treatment. This work offers new positive evidence for the interaction between As and PAHs in P. vittata , presents new information on the underlying mechanisms for interactions of As and PAHs affecting their uptake and translocation within P. vittata L., and provides direction for future research on the mechanisms of PAHs uptake by plants.

Nonionic and Water-Soluble Poly(d/l-serine) as a Promising Biomedical Polymer for Cryopreservation

Yuling Sun,Jie Liu,Zhibo Li,Jianjun Wang,Yanbin Huang

Water-soluble, biodegradable, nonionic, and biocompatible polymers with multiple functional groups are highly desired for biomedical applications. Here, we report that water-soluble nonionic poly(d/l-serine) is chirality-controllable, biodegradation-controllable, and non-cytotoxic. Hence, it can be a highly sought-after alternative to the widely used poly(ethylene glycol), with an additional advantage of having multiple hydroxyl groups for further functionalization. As one example of its biomedical applications, poly(d/l-serine) demonstrated an obvious cryoprotective effect on the red blood cells. The usage of poly(d/l-serine) in the cryopreservation field would be of great promise to resolve the difficulties in separating cryoprotectants due to toxicity.

Nanoencapsulation of zeaxanthin extracted from Lycium barbarum L. by complex coacervation with gelatin and CMC

Jiaying Zhang,Guoliang Jia,Zhao Wanbin,Jiang Minghao,Yulong Wei,Jingyi Hao,Xiaolin Liu,Zhilin Gan,Aidong Sun

This study developed nanocapsules by complex coacervation between gelatin (G) and sodium carboxymethyl cellulose (CMC) for the encapsulation of zeaxanthin extracted from Lycium barbarum L. The optimum pH and G-CMC mass mixing ratio were determined by analysis of the zeta potential , turbidity, morphology, particle size distribution, complex coacervate yield, emulsification stability index (ESI) and emulsification activity index (EAI). The formation mechanism of the G-CMC coacervates was examined by fourier transform infrared spectroscopy (FTIR) analysis. Moreover, the morphology, particle size distribution, thermal properties and in vitro simulated gastrointestinal digestion of zeaxanthin nanocapsules were investigated. The results showed that the optimum mass mixing ratio of G-CMC was 9:1 (w/w) with an optimum pH of 4.50. FTIR analysis confirmed the electrostatic interaction between the –NH 3 + of G and the -COO - of CMC in the formation of G-CMC complex coacervates. Thermal gravimetric analysis (TGA) showed that nanoencapsulation could enhance the thermal stability of zeaxanthin. In vitro simulated gastrointestinal digestion experiments showed that zeaxanthin had good sustained release performance in simulated gastric fluid (SGF) and large amounts of zeaxanthin were released in simulated intestinal fluid (SIF).

Highly Efficient Enzymatic Conversion of Rutin to Isoquercitrin and l-Rhamnose Using Deep Eutectic Solvents

Deqing Wang,Pu Zheng,Pengcheng Chen,Dan Wu

Deep eutectic solvents (DESs), considered to be “green media”, have recently been employed in multiple applications including biotransformations. Here, a DES was used in the production of isoquercitrin and l-rhamnose from rutin by enzymatic transformation with recombinant α-l-rhamnosidase from Aspergillus niger. A solution synthesized from choline chloride and urea (ChCl-U) performed best in the conversion of rutin among 24 kinds of DESs. When 130 g/L rutin was used as the substrate, the conversion rate was 100% and the productivity of l-rhamnose and isoquercitrin was 208.68 mM/h under optimized conditions in ChCl-U solution, which was 1.12 times higher than that in buffer (186.92 mM/h). The recycling of ChCl-U solution was also investigated by aqueous two-phase system extraction of l-rhamnose, and ChCl-U/K2HPO4 was screened as the suitable extraction solvent. When the ChCl-U content was 34%, the extraction efficiency of l-rhamnose was 79.06% with a purity and primary crystallization yield of 96.28 and 66.49%, respectively. After five cycles of ChCl-U solution, the average enzymatic conversion rate of recycling was 95.63% with an average purity of 94.22% of the isoquercitrin prepared. After 10 cycles of reuse, the residual conversion rate was 61.14%. The results indicate that the DES has obvious advantages in biocatalytic processes involving flavonoid conversion.
