In this work, near-infrared (NIR) light driven concave octopod PtCu alloy nanocages (OPtCu-NCs) were fabricated and first developed as photothermal synergetic Fenton catalyst for degradation of pollutant dyes. Moreover, by irradiating with NIR laser (808 nm), the obtained OPtCu-NCs exhibit excellent NIR absorption capacity and thus produce hyperpyrexia over 50 °C. Concomitantly, by the photothermal effect, the OPtCu-NCs nanocatalysts was used for elimination of organic dyes such as Methylene blue (MB) and Rhodamine B (RhB). The NIR-driven photothermal degradation mechanism under NIR irradiation can be attributed to the Fenton-like catalytic performance of Cu ions , then -N C- and/or C-C bonds in the molecular skeleton were fractured and electrons were donated intensively. As confirmed by a typical radical trapping experiment, electrons trigger the formation of miscellaneous reactive species (O 2 - , HO 2 , especially •OH) subsequently . Moreover, the photothermal Fenton-catalytic activity of the as-prepared OPtCu-NCs had maintained at 93.15% (for MB) and 91.87% (for RhB) after ten circulating degradation processes. All acquired results reveal that the OPtCu-NCs have the potential applied value in dyestuff-wastewater treatment under neutral pH.

Graphical Electrospun NaYF 4 :Yb 3+ ,Er 3+ @PLLA nanofibers introduce pH-sensitive functional groups and are load with DOX drug. After the DOX release, the photoluminescence of nanofiberat 550 nm becomes stronger under NIR 980 nm exciatation, which could be used to monitor the release of drug in real time. Stimulus-responsive nanofibers can adjust the speed of drug delivery, and real-time monitoring elucidates the relationship between drug release and therapeutic efficacy. Both of these advantages have great potential in clinical therapy. However, the complex preparation of current stimulus-responsive drug-loaded nanofibers leads to low drug-loading levels, limiting their widespread applications. In this study, we prepared pH-responsive drug-loaded nanofibers with fluorescence monitoring properties using the electrospinning method. A representative drug of DOX was loaded onto the nanofibers. The degradable flexible nanofibers modified by amino groups attained higher drug loading and controlled drug release. Precisely, the rate of drug release varies with different pH. Faster release speed occurs in the tumor microenvironment with faintly acidic conditions than in healthy tissues with neutral conditions. The pH-responsive mechanism was due to hydrazone bonds between the amino groups and DOX molecules, which FTIR confirmed. In addition, these pH-responsive nanofibers with intense upconversion fluorescence enable more sensitive monitoring, whereas weak luminescence cannot achieve.

Herbal medicines and dietary supplements are widely used worldwide. However, reports have revealed that a large number of herbal medicines are implicated in the hepatic damage, which is referred to as Herbal medicines-induced liver injury (HILI). Traditional diagnostic methods for HILI include histological and serological assessments, which fail to offer real-time and unambiguous visualization of hepatotoxicity in vivo . Herein, we constructed a second near-infrared (NIR-II) luminescent nanoprobes to achieve accurate, high-efficiency, and non-invasive detection of HILI by the assembly of lanthanide-based NIR-II luminescence nanoparticles with a chromophore with absorption band in the NIR-II region. Specifically, the NIR-II luminescence of the lanthanide nanoparticles can be effectively quenched by the chromophores on the surface of nanoparticles via an absorption competition-induced quenching effect, and subsequently recovered by the pathophysiological biomarker peroxynitrite (ONOO − ) generated in early HILI. It is demonstrated that the nanoprobes are capable for the screening of liver toxins in a noninvasive pattern, which is more convenient than the conventional diagnostic approaches. Finally, our luminescence detection approach offers an immediate testing technique for the identification of hepatotoxic components of herbal medicine such as colchicine, monocrotaline and stibene glucoside, providing a high-throughput screening strategy for herbal medicine-induced hepatotoxicity.

Photocatalysts driven by near infrared light (NIR) are of great importance for making full use of solar energy and wide applications in biomedicine, biosensor, photocatalysis, and etc. In this work, we demonstrate a facile template assisted hydrothermal approach to fabricate upconversion nanoparticles (UCNPs) and Zn x Cd 1-x S yolk-shell nanoparticles, although the yolk-shell nanoparticles were made from hexagonal phase of β-NaYF 4 (NaYF 4 :Yb/Tm@NaYF 4 :Yb/Er) and II–VI semiconductor materials with a large lattice mismatch of more than 10%. The growth mechanism of the formation of yolk-shell nanostructures has been extensive investigated and the shape evolution with the time has been demonstrated. Steady and dynamic fluorescence spectra show that the as-prepared yolk-shell nanoparticles of UCNPs/Zn x Cd 1-x S can be used as efficient nanotransducer for NIR light owing to enhanced energy transfer efficiency. Interesting, the as-prepared yolk-shell nanoparticles exhibit excellent photocatalytic performance on degradation towards RhB dyes, excellent biocompatible properties and high production ability for hydroxyl radical ( OH) and single oxygen ( 1 O 2 ), which might be employed to wide applications in solar cell, hydrogen production, and photodynamic therapy related nano-biotechnology.

Optogenetics using light-sensitive proteins such as calcium transport channel rhodopsin (CatCh) opens up new possibilities for non-invasive remote manipulation of neural function. However, current optogenetic approaches for neurological disorder therapies rely on visible light excitation and are rarely applied to neurogenesis and nerve regeneration. Herein, we propose a new strategy for tissue engineering which combines optogenetic technology and biomimetic nerve scaffolds. Upconversion nanoparticles (UCNPs) were synthesized and integrated with oriented fibrillar PCL membranes with a collagen coating to establish neuro-matrix interfaces. Benefiting from the excellent bioactivity, oriented fibrillation and NIR-photoresponsivity, the CatCh-transfected PC12 cells on these interfaces exhibited enhanced cell elongation and neurite extension, as well as upregulated neurogenesis upon NIR excitation. Furthermore, a UCNP-integrated scaffold as an optogenetic actuator allowed NIR to penetrate dermal tissues to mediate neural activation, with an efficiency comparable to that of a 470 nm blue light. Compared with current visible light-excited optogenetics, our composite scaffold-mediated NIR stimulation addresses the problem of tissue penetration and will enable less-invasive neurofunctional manipulation, with the potential for remote therapy.

Polydopamine (PDA) as a melanin-like biomimetic material with excellent biocompatibility, full spectrum light absorption capacity and antioxidation property has been extensively applied in the biomedical field. Based on the high reactivity of dopamine (DA), exploiting new strategies to fabricate novel PDA-based nano-biomaterials with controllable size and improved performance is valuable and desirable. Herein, we reported a facile way to synthesize pyrrole-doped polydopamine-pyrrole nanoparticles (PDA-nPY NPs) with tunable size and enhanced near-infrared (NIR) absorption capacity through self-oxidative polymerization of DA with PY in an alkaline ethanol/H2O/NH4OH solution. The PDA-nPY NPs maintain excellent biocompatibility and surface reactivity as PDA. By regulating the volume of added PY, PDA-150PY NPs with a smaller size (<100 nm) and four-fold higher absorption intensity at 808 nm than that of PDA can be successfully fabricated. In vitro and in vivo experiments effectively further demonstrate that PDA-150PY NPs can effectively inhibit tumor growth and completely thermally ablate a tumor. It is believed that these PY doped PDA-nPY NPs can be a potential photothermal (PT) agent in biomedical application.Keywords:polydopamine;pyrrole;self-oxidative polymerization;photothermal treatment;cancer therapy

Background Melanoma is a highly aggressive form of skin cancer with increasing incidence and mortality rates. Chemotherapy, the primary treatment for melanoma, is limited by hypoxia-induced drug resistance and suppressed immune response at the tumor site. Modulating the tumor microenvironment (TME) to alleviate hypoxia and enhance immune response has shown promise in improving chemotherapy outcomes. Methods In this study, a novel injectable and in situ forming hydrogel named MD@SA was developed using manganese dioxide (MnO2) nanosheets pre-loaded with the chemotherapy drug doxorubicin (DOX) and mixed with sodium alginate (SA). The sustainable drug delivery, oxygen generation ability, and photothermal property of MD@SA hydrogel were characterized. The therapeutic efficacy of hydrogel was studied in B16F10 in vitro and B16F10 tumor-bearing mice in vivo. The immune effects on macrophages were analyzed by flow cytometry, real-time quantitative reverse transcription PCR, and immunofluorescence analyses. Results The MD@SA hydrogel catalyzed the tumoral hydrogen peroxide (H2O2) into oxygen, reducing the hypoxic TME, down-regulating hypoxia-inducible factor-1 alpha (HIF-1α) and drug efflux pump P-glycoprotein (P-gp). The improved TME conditions enhanced the uptake of DOX by melanoma cells, enhancing its efficacy and facilitating the release of tumor antigens. Upon NIR irradiation, the photothermal effect of the hydrogel induced tumor apoptosis to expose more tumor antigens, thus re-educating the M2 type macrophage into the M1 phenotype. Consequently, the MD@SA hydrogel proposes an ability to constantly reverse the hypoxic and immune-inhibited TME, which eventually restrains cancer proliferation. Conclusion The injectable and in situ forming MD@SA hydrogel represents a promising strategy for reshaping the TME in melanoma treatment. By elevating oxygen levels and activating the immune response, this hydrogel offers a synergistic approach for TME regulation nanomedicine.

Timely detection of hepatic oxidative stress is effective in preventing the onset and progression of liver disease. However, real-time and dynamic monitoring of oxidative stress in vivo remains challenging. Herein, we report a H 2 O 2 -responsive dual-peak ratiometric NIR-II fluorescent probe (Er@Ag 2 S:Pb), made by combining H 2 O 2 -responsive fluorescent Pb-doped Ag 2 S quantum dots (Ag 2 S:Pb QDs) with a fluorescence-stabilized reference substrate NaYF 4 :Yb/Er @NaYF 4 . By monitoring the change in emitted I 1550 /I 1220 intensities the probe can be used to sense and quantify H 2 O 2 concentrations ranging from 5 to 100 μM with excellent specificity. Experimental studies using mice models have shown that this probe can be used to monitor of hepatic H 2 O 2 -related oxidative stress in real time and non-invasively. Thus, the developed probe provides a powerful ratiometric NIR-II fluorescence imaging tool for timely monitoring of oxidative stress in vivo .

Radiative cooling provides a low-energy consumption remedy for the urban heat island effect. However, most radiative cooling materials use light-colored surfaces to achieve higher reflectivity, which not only causes light pollution problems but also degrades performance after surface contamination. Here, we through the design of the polymer structure used perylene diimide with coloration ability to prepare the crimson polymer perylene poly(aryl ether) (P-FAPE) by simple condensation reaction and prepared the superhydrophobic film with a porous structure by the method of nonsolvent-induced phase transformation. The polymer film has high near-infrared reflectance, with a maximum reflectance of 93% in the near-infrared region and emissivity reaches 95–98% in the atmospheric transmission window. In three days of outdoor testing, the P-FAPE6 film showed an average temperature drop of 5.72–8.52 °C compared to ambient temperature. Importantly, the water contact angle of the P-FAPE6 film is 160°, and the self-cleaning effect prevents the film surface from dirt contamination or wetting in long-term outdoor use. The P-FAPE6 film also demonstrated long-term stability, a V-0 rating for fire resistance in the UL-94 combustion test, which is promising in applications for cooling building facades, outdoor tents, and vehicle sunshades.

Multiplexed fluorescence imaging is mainly applied to cellular imaging. However, the application of an analogous technique to mammals remains challenging due to the presence of substantial scattering and autofluorescence in biological tissues in the visible (350–700 nm) and near-infrared I region (NIR-I, 700–900 nm). Herein, we report multiplexing excitation and single-channel detection in the near-infrared II region (NIR-II) with two fluorescent probes for dual-color imaging in vivo. The first one, FNIR-1090 was constructed on Pr-1099 derivative by attaching with 1-Ethylpiperazine through the SRN1 displacement reaction. The second one, IR-765 was synthesized by a one-step reaction between the Vilsmeier–Haack reagent with 2-(3,5,5-trimethylcyclohex-2-enylidene) malononitrile (TEMO). It is noteworthy that the absorption spectrum of FNIR-1090 is almost completely separated from that of IR-765, which facilitates dual-color in vivo imaging. The two dyes can be used for high-contrast dual-channel vivo imaging of different tissues. All these prominent performances show the great promise of FNIR-1090 and IR-765 in non-invasive, real-time, and dual-color NIR-II imaging (>900 nm) in vivo.

Graphical BP/MoS 2 heterojunction is prepared by solvothermal method using NMP, BP nanosheets and (NH 4 ) 2 MoS 4 . Under NIR irradiation, BP/MoS 2 can be heated up to 51.8 °C within 5 minutes by photothermal conversion. In addition, the existence of ⋅O 2 − and 1 O 2 in BP/MoS 2 -NIR system is verified to sterilize efficiently. With a loading content of 10 wt % MoS 2 , bacterial cells, especially drug-resistant bacteria (10 6 cfu ⋅ mL −1 ) completely can lose the viability within 10 min of NIR irradiation. In this research, a high-efficiency nano-fungicide with synergistic effect was obtained by combining polycrystalline MoS 2 with layered nano-BP to form heterojunction, which had a new photothermal-photocatalytic antibacterial mechanism to Gram-positive bacteria, Gram-negative and drug-resistance bacteria under NIR. BP/MoS 2 whose lethality of bacteria could reach nearly 100 % at 400 μg/L, exhibited the highest photothermal lysis efficiency against E. coli, B. pumilus and A. baumannii. It was found that the heterojunction could be heated up to 51.8 °C within 5 minutes by photothermal conversion, and it also had an excellent performance of photothermal cycle. High temperature and stability of the material endowed it with an outstanding and long-lasting bactericidal effect. In addition, a series of in vitro free radical experiments was designed, the existence of ⋅O 2 − and 1 O 2 in BP/MoS 2 -NIR system was verified. And the Way of ROS production and the mechanism of action were discussed by EPR test. Subsequently, two mechanisms of synergistic antibacterial action were summarized, that is, abundant ROS and high temperature could destroy cell wall damage and lysis of bacteria effectively. This study provides a theoretical basis for the synergistic antibacterial action of NIR photothermal and photocatalysis of BP system.

Background Therapeutic tumor vaccine (TTV) that induces tumor-specific immunity has enormous potentials in tumor treatment, but high heterogeneity and poor immunogenicity of tumor seriously impair its clinical efficacy. Herein, a novel NIR responsive tumor vaccine in situ (HA-PDA@IQ/DOX HG) was prepared by integrating hyaluronic acid functionalized polydopamine nanoparticles (HA-PDA NPs) with immune adjuvants (Imiquimod, IQ) and doxorubicin (DOX) into thermal-sensitive hydrogel. Results HA-PDA@IQ NPs with high photothermal conversion efficiency (41.2%) and T 1 -relaxation efficiency were using HA as stabilizer by the one-pot oxidative polymerization. Then, HA-PDA@IQ loaded DOX via π-π stacking and mixed with thermal-sensitive hydrogel to form the HA-PDA@IQ/DOX HG. The hydrogel-confined delivery mode endowed HA-PDA@IQ/DOX NPs with multiple photothermal ablation performance once injection upon NIR irradiation due to the prolonged retention in tumor site. More importantly, this mode enabled HA-PDA@IQ/DOX NPs to promote the DC maturation, memory T cells in lymphatic node as well as cytotoxic T lymphocytes in spleen. Conclusion Taken together, the HA-PDA@IQ/DOX HG could be served as a theranostic tumor vaccine for complete photothermal ablation to trigger robust antitumor immune responses.
